KLF5 protects the intestinal epithelium against Th17 immune response in a murine colitis model.

Inflammatory bowel disease (IBD) is a chronic illness characterized by dysregulated immune cascades in the intestines, in which the Th17 immune response plays an important role. We demonstrated that mice with intestinal epithelium–specific deletion of Krüppel-like factor 5 (Klf5) developed Th17-dependent colonic inflammation. In the absence of KLF5, there was aberrant cellular localization of phosphorylated STAT3, an essential mediator of the Th17-associated cytokine, IL-22, which is required for epithelial tissue regeneration. In contrast, mitigation of IL-17A with anti–IL-17A neutralizing antibody attenuated colitis in Klf5-deficient mice. There was also a considerable shift in the colonic microbiota of Klf5-deficient mice that phenocopied human IBD. Notably, the inflammatory response due to Klf5 deletion was alleviated by antibiotic treatment, implicating the role of microbiota in pathogenesis. Finally, human colitic tissues had reduced KLF5 levels when compared with healthy tissues. Together, these findings demonstrated the importance of KLF5 in protecting the intestinal epithelium against Th17-mediated immune and inflammatory responses. The mice described herein may serve as a potential model for human IBD.

The role of microbiota in pathogenesis and development of viral infections

Introduction and purpose
 The microbiota plays an important role in human metabolism, immune system, and development of many chronic diseases, cancers, and infectious diseases. The aim of the study is to present the role of gut microbiota in viral infections, including HBV, Herpesviridae, HIV, and SARS-CoV-2. The newest publications from the last 5 years available on the PubMed database were taken into account.
 A brief description of the state of knowledge
 The mechanism by which bacterias have an impact on viral infection is based on a synthesis of specific short-chain fatty acids (SCFAs) and modulation of cytokine release and immune system function by bacterias. Domination of Gardnerella vaginalis and lack of Lactobacillus in the vaginal microbiome increased the risk of HSV-2 infection in women. Inflammation of the genital tract can influence susceptibility to HIV infection, but probiotics via enhancement of the gut barrier integrity, change TH17/Treg ratio, can restore microbiome composition. LPS - component of the structure of Gram-negative bacteria can be a marker of HBV infection. In the airway microbiome of patients with COVID-19 opportunistic microorganisms were identified.
 Conclusions 
 Diet, intake of probiotics, fecal microbiota transplantation (FMT) are interventions that might be efficient methods in prophylaxis and treatment of viral diseases. Further studies are needed to evaluate the mechanism of action of microbiome in pathogenesis of infectious diseases.

Role of Microbiota in Pathogenesis and Management of Viral Hepatitis.

Hepatitis is a condition that can be self-limiting or can progress to fibrosis (scarring), cirrhosis, or liver cancer. These days, gut microbiota becomes an important part of our immune system, which is important for disease progression or recovery. Translocation of gut microbial and metabolic products causes intestinal inflammation by modulating immune cells leading to impairment of the primary barrier. But there are limited studies discussing pathogenesis and management of hepatitis with gut microbiota. In this review, we have discussed the role of gut microbiota in pathogenesis and management of various hepatitis, especially hepatitis B and C. We have discussed the role of bacterial products, LPS-TLR4 pathway, and unmethylated CpG DNA, which ultimately affects downstream NF-kB signaling in hepatitis. Finally, we have discussed the role of fecal microbiota transplantation in the management of hepatitis.

The recent revival of old theories and setting them on modern scientific rails to a large extent are also relevant to mitochondrial science. Given the widespread belief that mitochondria are symbionts of ancient bacterial origin, the processes inherent to mitochondrial physiology can be revised based on their comparative analysis with possible involvement of bacteria. Such comparison combined with discussion of the role of microbiota in pathogenesis allows discussion of the role of "mitobiota" (we introduce this term) as the combination of different phenotypic manifestations of mitochondria in the organism reflecting pathological changes in the mitochondrial genome. When putting an equal sign between mitochondria and bacteria, we find similarity between the mitochondrial and bacterial theories of cancer. The presence of the term "bacterial infection" suggests "mitochondrial infection", and mitochondrial (oxidative) theory of aging can in some way be transformed into a "bacterial theory of aging". The possible existence of such processes and the data confirming their presence are discussed in this review. If such a comparison has the right to exist, the homeostasis of "mitobiota" is of not lesser physiological importance than homeostasis of microbiota, which has been so intensively discussed recently.
