The infection of grapes by Talaromyces rugulosus O1 and the role of cell wall-degrading enzymes and ochratoxin A in the infection

Talaromyces rugulosus O1 is an ochratoxin A (OTA)-producing pathogen from grapes. The mechanisms involved in the infection of postharvest grapes by T. rugulosus O1 are not yet clear. In this study, the infection of grapes by T. rugulosus O1 and OTA production in grapes were investigated, and the role of cell wall degrading enzymes of T. rugulosus O1 and OTA in the infection of grapes by T. rugulosus O1 was explored. The results indicated that T. rugulosus O1 showed significant infection effect on grapes at 20 °C, and accumulated OTA in the wound of grapes in the infection progress. In comparison with the control, the activities of polygalacturonase (PG), pectin methylgalacturonase (PMG), exo-1,4-β-glucanase (C1) and β-glucosidase (β-G) secreted by T. rugulosus O1 in grapes were all increased in the infection process. Furthermore, the pectinase (PG and PMG) activities were higher than cellulase (C1 and β-G) activities. The expression levels of genes encoding PG and β-G in T. rugulosus O1 infecting grapes increased as the time prolong. Inoculation of T. rugulosus O1 in combination with addition of OTA in the wound of grapes resulted in higher decay incidence and lesion diameter of treated grapes compared with the treatment with T. rugulosus O1 or OTA alone. This indicated that OTA can promote the infection of grapes by T. rugulosus O1. This study provides useful information about the role of cell wall degrading enzymes and OTA in the infection of grapes by T. rugulosus O1.

Physical and Chemical Barriers in Root Tissues Contribute to Quantitative Resistance to Fusarium oxysporum f. sp. pisi in Pea.

Fusarium wilt caused by Fusarium oxysporum f. sp. pisi (Fop) is one of the most destructive diseases of pea worldwide. Control of this disease is difficult and it is mainly based on the use of resistant cultivars. While monogenic resistance has been successfully used in the field, it is at risk of breakdown by the constant evolution of the pathogen. New sources of quantitative resistance have been recently identified from a wild relative Pisum spp. collection. Here, we characterize histologically the resistance mechanisms occurring in these sources of quantitative resistance. Detailed comparison, of the reaction at cellular level, of eight pea accessions with differential responses to Fop race 2, showed that resistant accessions established several barriers at the epidermis, exodermis, cortex, endodermis and vascular stele efficiently impeding fungal progression. The main components of these different barriers were carbohydrates and phenolic compounds including lignin. We found that these barriers were mainly based on three defense mechanisms including cell wall strengthening, formation of papilla-like structures at penetration sites and accumulation of different substances within and between cells. These defense reactions varied in intensity and localization between resistant accessions. Our results also clarify some steps of the infection process of F. oxysporum in plant and support the important role of cell wall-degrading enzymes in F. oxysporum pathogenicity.

Sugars and pH: A clue to the regulation of fungal cell wall-degrading enzymes in plants

In recent years, substantial progress has been made in understanding the role of cell wall-degrading enzymes in virulence of fungal pathogens using genetic tools. A homologous recombination-mediated targeted gene disruption of a single pectolytic gene decreased virulence in multiple fungal pathogens. Furthermore, differential regulation of a gene that encodes a cell wall-degrading enzyme during infection under different host environments has recently been examined, and host pH and sugars are involved in the regulation of the polygalacturonase gene. We will provide a brief overview of the biology of the macerating diseases caused by phytopathogenic Alternaria pathogens in particular hosts, followed by a description of possible mechanisms controlling the differential expression of genes encoding cell wall-degrading enzymes in the infected host.

The Role of Cell Wall-degrading Enzymes in the Formation of Gum Ducts in Almond Fruit

Abstract Cell wall-degrading enzymes were extracted from the cell wall free space of mesocarp tissue from immature almonds [Prunus dulcis(Mill.)D.A. Webb, ‘Nonpareil’]. The activities of several of these enzymes were found to correlate with the development of gum ducts in this tissue. Polygalacturonase (EC 3.2.1.15) and 1,3-β-D-glucanase (EC 3.2.1.39) activities rose sharply at, or just prior to, the early schizogenous stage of duct initiation, while increases in α-galactosidase (EC 3.2.1.22), β-galactosidase (EC 3.2.1.23), α-arabinosidase (EC 3.2.1.55), and α-mannosidase (EC 3.2.1.24) activities were correlated with the later lysigenous stage of duct formation. Cell wall analysis of almond mesocarp tissue sampled the week preceding gum duct formation determined that the predominant noncellulosic sugars present in the mesocarp cell walls are arabinose, galactose, xylose, and glucose, with smaller amounts of rhamnose and mannose also present. The walls also contain a high percentage of galacturonic acid and trace amounts of glucuronic acid. Methylation analysis of the cell walls confirmed that many of the specific glycosidic linkages that are cleaved by the enzymes tested are present in the mesocarp cell walls immediately prior to gum duct formation.
