The Diversity and Community Composition of Three Plants' Rhizosphere Fungi in Kaolin Mining Areas.

Mining activities in the kaolin mining area have led to the disruption of the ecological health of the mining area and nearby soils, but the effects on the fungal communities in the rhizosphere soils of the plants are not clear. Three common plants (Conyza bonariensis, Artemisia annua, and Dodonaea viscosa) in kaolin mining areas were selected and analyzed their rhizosphere soil fungal communities using ITS sequencing. The alpha diversity indices (Chao1, Shannon, Simpson, observed-species, pielou-e) of the fungal communities decreased to different extents in different plants compared to the non-kauri mining area. The β-diversity (PCoA, NMDS) analysis showed that the rhizosphere soil fungal communities of the three plants in the kaolin mine area were significantly differentiated from those of the control plants grown in the non-kaolin mine area, and the extent of this differentiation varied among the plants. The analysis of fungal community composition showed that the dominant fungi in the rhizosphere fungi of C. bonariensis and A. annua changed, with an increase in the proportion of Mycosphaerella (genus) by about 20% in C. bonariensis and A. annua. An increase in the proportion of Didymella (genus) by 40% in D. viscosa was observed. At the same time, three plant rhizosphere soils were affected by kaolin mining activities with the appearance of new fungal genera Ochrocladosporium and Plenodomus. Predictive functional potential analysis of the samples revealed that a significant decrease in the potential of functions such as biosynthesis and glycolysis occurred in the rhizosphere fungal communities of kaolin-mined plants compared to non-kaolin-mined areas. The results show that heavy metals and plant species are the key factors influencing these changes, which suggests that selecting plants that can bring more abundant fungi can adapt to heavy metal contamination to restore soil ecology in the kaolin mining area.

Shift in the rhizosphere soil fungal community associated with root rot infection of Plukenetia volubilis Linneo caused by Fusarium and Rhizopus species.

Plukenetia volubilis Linneo is an oleaginous plant belonging to the family Euphorbiaceae. Due to its seeds containing a high content of edible oil and rich in vitamins, P. volubilis is cultivated as an economical plantworldwide. However, the cultivation and growth of P. volubilis is challenged by phytopathogen invasion leading to production loss. In the current study, we tested the pathogenicity of fungal pathogens isolated from root rot infected P. volubilis plant tissues by inoculating them into healthy P. volubilis seedlings. Metagenomic sequencing was used to assess the shift in the fungal community of P. volubilis rhizosphere soil after root rot infection. Four Fusarium isolates and two Rhizopus isolates were found to be root rot causative agents of P. volubilis as they induced typical root rot symptoms in healthy seedlings. The metagenomic sequencing data showed that root rot infection altered the rhizosphere fungal community. In root rot infected soil, the richness and diversity indices increased or decreased depending on pathogens. The four most abundant phyla across all samples were Ascomycota, Glomeromycota, Basidiomycota, and Mortierellomycota. In infected soil, the relative abundance of each phylum increased or decreased depending on the pathogen and functional taxonomic classification. Based on our results, we concluded that Fusarium and Rhizopus species cause root rot infection of P. volubilis. In root rot infected P. volubilis, the shift in the rhizosphere fungal community was pathogen-dependent. These findings may serve as a key point for a future study on the biocontrol of root rot of P. volubilis.

Differences in rhizosphere soil fungal communities of wild and cultivated Paeonia ludlowii species.

Paeonia ludlowii is a rare and endangered plant species with a high application value. However, its low cultivation success rate in China has severely limited its protection, development, and utilization. In addition to natural factors, microorganisms in the rhizosphere play an important role in determining its cultivation success. In this study, growth indexes and soil physicochemical properties of both wild (origin: Nyingchi) and cultivated (introduction: Luanchuan) species of P. ludlowii were measured during the flowering, fruiting, and autumn foliage stages. ITS high-throughput sequencing technology was employed to detect rhizosphere soil fungi, and the diversity, community structure, functional prediction, molecular network, and ecological processes of the microbial community assembly were examined by multidirectional analysis. The results indicated that: both wild and cultivated P. ludlowii species were able to flower and fruit normally, although the wild species had a higher number of flowers and fruits and higher soil available phosphorus and available potassium contents than those of the cultivated species. Ascomycota and Basidiomycota were the dominant rhizosphere soil fungal phyla in both P. ludlowii species. However, our network analysis showed that Ascomycota as the key fungal phylum of the wild species, whereas the cultivated species lacked key fungi. The community assembly mechanisms of rhizosphere soil fungi in both wild and cultivated species were primarily stochasticity, with no significant differences between them. Based on the results of FUNGuild and molecular network analyses, cultivated species had a higher proportion of fungi, such as Soil Saprotroph, that can easily cause diseases. Additionally, the network connections among fungi were weaker in the cultivated species than those in the wild species, which increased the cultivated species susceptibility to external environmental interferences. Therefore, from a soil microorganism perspective, this study suggests that, after the introduction and cultivation of P. ludlowii, if rhizosphere soil fungi fail to gradually form a close network relationship and instead promote the growth of pathogenic fungi, the fungal ecosystem would become vulnerable.

Effects of Bacillus velezensis GUAL210 control on edible rose black spot disease and soil fungal community structure.

Rose black spot is an economically important disease that significantly decreases flower yield. Fungicide and biological control are effective approaches for controlling rose black spot. The objective of this study was to evaluate the effect of application of biological and chemical control agents, including Bacillus velezensis (GUAL210), Bacillus sp. (LKW) and fungicide (CP) on the black spot disease and rhizosphere fungal community structure of edible rose. In this study, the R. chinensis 'Crimson Glory' was taken as the research object, and the field experiment was designed by randomized block design. The experiment contained 3 treatments (CP, GUAL210, LKW) and 1 control. The control effect and growth promoting effect of fungicide and biological control on rose black spot were compared. The composition and diversity of rhizosphere soil fungal community of different treatments of rose were studied by high-throughput sequencing method. The fungal community composition, correlation of environmental factors and differences in metabolic pathways related to rose disease were analyzed, and the correlation between rhizosphere soil fungal community of rose and biological control of disease was explored. Both disease incidence and disease index differed significantly among groups (LKW < GUAL210 < CP < CK), and disease control effect exhibited no significant difference between GUAL210 and LKW (60.96% and 63.86%, respectively). Biological control was superior to chemical control in terms of disease prevention effects and duration, and it significantly increased the number of branches and flowers of rose plants. Ascomycota and Basidiomycota accounted for more than 74% of the total fungal abundance, and the abundance of Ascomycota was highest in CK, followed by GUAL210, CP and LKW, which was consistent with the disease occurrence in each group. The analysis of metabolic pathways showed that the HSERMETANA-PWY in each experimental group was significantly lower than that in control group. The Shannon index in each experimental group was significantly lower than that in control group. PCoA analysis showed that the rhizosphere fungal community structure in each experimental group was significantly different from that in control group. Trichoderma, Paraphaeosphaeria, Suillus, Umbelopsis in GUAL210, and Galerina in LKW replaced Mortierella, Pestalotiopsis, Ustilaginoidea, Paraconiothyrium, Fusarium, and Alternaria as dominant flora, and played a nonneglectable role in reducing disease occurrence. The difference in rhizosphere fungal community structure had an important impact on the incidence of rose black spot disease. Biological control is crucial for establishing environment-friendly ecological agriculture. GUAL210 has promising prospects for application and development, and may be a good substitute for chemical control agents.

Rhizosphere fungal community assembly varied across functional guilds in a temperate forest

BackgroundRhizosphere fungi play an important role in plant community dynamics and biogeochemical cycling. While the drivers of fungal community assembly have been studied in varied ecosystems, it is still unclear how these processes function for rhizosphere soil fungi in temperate forests. Furthermore, it is unknown whether the relative contributions of important determinants remain consistent or vary across fungal ecological guilds. This study used high-throughput next-generation sequencing to characterize the fungal communities of 247 rhizosphere soil samples from 19 tree species in a temperate forest within Northeast China. We aimed to investigate how three important determinants in temperate forests (host tree species, neighbouring plant communities, and edaphic properties) influence the community assembly of fungal functional guilds in the rhizosphere soil of trees.ResultsWe found that host tree species contributed more to plant pathogens’ community composition than ectomycorrhizal fungi, and plant pathogens consistently showed higher host specialization than ectomycorrhizal fungi. Saprotrophs also showed high host specialization, which was mediated by the tree species’ effect on rhizosphere soil pH. Although neighboring plant communities contributed remarkably to richness of all fungal guilds, this effect on fungal composition varied across functional guilds, with stronger effect for biotrophic guilds (plant pathogens and ectomycorrhizal fungi) than for non-biotrophic guild (saprotrophs). Neighboring plant communities shaped the ectomycorrhizal community composition strongly in all samples regardless of host trees’ mycorrhizal type, whereas edaphic properties were the most important drivers for this guild in samples from only ectomycorrhizal-associated trees. Edaphic properties played an important role in shaping ectomycorrhizal and saprotrophic fungal compositions, indicating the importance of edaphic properties on the fungal functional guilds associated with the absorption and decomposition of nutrients.ConclusionsThese results demonstrated that rhizosphere soil fungal community assembly determinants varied across fungal guilds, reflecting their different ecological functions in temperate forest ecosystems.

Effects of Tillage and N Applications on the Cassava Rhizosphere Fungal Communities

Cassava (Manihot esculenta Crantz) is mainly cultivated in marginal land in the south of China where seasonal drought stress occurs frequently and the soil becomes more compact year by year. The study aimed to explore the effect of Fenlong tillage (FLT) combined with nitrogen applications on cassava rhizosphere soil particle composition and fungal community diversity. Conventional tillage (CT) was set as the control. The results indicated that the contents of clay and silt of the cassava rhizosphere soil were influenced by the tillage method, nitrogen (N), and their interaction. There was no difference in the richness and diversity of rhizosphere soil fungal communities among all treatments in 2019, while the richness of FLT was lower than that of CT in 2020. FLT caused a stronger influence on the community structure of rhizosphere fungi than N applications in the first year. The differences in the community structure of all treatments were reduced by continuous cropping of cassava in the second year. The top 10 dominant rhizosphere fungi at the class level of cassava found in 2019 and 2020 were Sordariomycetes, Dothideomycetes, Eurotiomycetes, Agaricomycetes, Intramacronucleata, norank_p__Mucoromycota, unclassified_p__Ascomycota, unclassified_k__Fungi, Pezizomycetes, and Glomeromycetes, which had an important relationship with soil pH, activity of urease, available nitrogen, available phosphorus, organic matter, and clay. These results indicated that FLT created a better soil environment for cassava growth than CT, thus promoting the formation of more stable rhizosphere fungal community structures.

Subsurface aeration alters the fungal composition of rhizosphere soil and tomato plant performance in Northwest China

Rhizosphere hypoxia constrains plant growth, and numerous studies have shown that root zone aeration accelerates plant photosynthesis and growth and increases crop yields. Nevertheless, the mechanism by which soil microorganisms are involved in this process is not clear. The purpose of the present study was to examine the effects of aeration and irrigation depth on the composition and structure of rhizosphere soil fungal communities and tomato plant performance. The amount of aeration assayed was equal to 0 (CK), 0.5 (V1), 1 (V2), and 1.5 (V3) times the porosity of the soil. The two depths of subsurface drip irrigation used were 15 (D15) and 40 cm (D40). The results demonstrated that soil aeration not only increased tomato plant performance but also influenced fungal diversity and composition. Compared to the no-aeration treatment, the V3 soil aeration treatment increased the total dry weight and fruit yield by 39.9% and 65.6%, respectively. The results also showed that the abundance of the phylum Ascomycota and the family Lasiosphaeriaceae increased with increasing soil aeration, whereas those of members of the phylum Zygomycota and the order Capnodiales decreased with increasing soil aeration. Moreover, the variation in subsurface irrigation depth altered the rhizosphere soil fungal community. In general, the results of this study demonstrate that root zone aeration can ameliorate hypoxic conditions in Lou soils and is beneficial to soil fungal communities and tomato plant performance. Rhizosphere Soil Fungal Diversity and Soil Physicochemical Properties of Different Vegetations in Tundra of Changbai Mountain

By studying the diversity and community structure of rhizosphere soil fungi of different plants in the tundra on the northern slope of Changbai Mountain, it provides theoretical support for the restoration of environmental degradation and in-depth study of fungal diversity in the tundra of Changbai Mountain. High-throughput sequencing technology was used to determine the ITS1 region of fungal amplicons, so as to analyze the diversity of fungal communities in the rhizosphere soil of six plants in the tundra of Changbai Mountain, and to analyze the correlation between the environment and the diversity and richness of fungal communities in combination with relevant soil physical and chemical factors. The diversity and richness of fungal community in the rhizosphere soil of six plants in Changbai Mountain tundra were different. The Simpson and Shannon indexes of Saxifraga stolonifera Curt were the highest, and the richness of fungal community in Dryas octopetala was the highest. The analysis of fungal community composition showed that the fungal colonies in plant rhizosphere soil samples mainly belonged to Ascomycota and Basidiomycota, which were the main dominant phyla. Mortierella, Fusarium and Sordariomycetes are common fungal genera in the rhizosphere soil of six plants, but their abundances are different among different plants. Water content was negatively correlated with fungal diversity, and TP was positively correlated with fungal community diversity. There were some differences in the composition and diversity of rhizosphere soil fungal communities of six plants in Changbai Mountain tundra. Ascomycota and Basidiomycota were the main soil fungal phyla in the rhizosphere of six plants in Changbai Mountain tundra. The results could provide theoretical guidance for ecological protection of Changbai Mountain tundra.

Conversion of rainforest to rubber plantations impacts the rhizosphere soil mycobiome and alters soil biological activity

AbstractIn Southeast Asia and Asia, large swathes of rainforest have been converted to rubber plantations, with major consequences for biodiversity and ecosystem services. However, the impact of this land use conversion on rhizosphere soil mycobiome has not yet been addressed. This study aimed to investigate how rhizosphere soil fungal communities and their associated biological activity (soil respiration, soil methane (CH4), and potential soil enzyme production) are impacted by the conversion of rainforest to rubber plantations. Fungal richness and community composition in rhizosphere soils collected from natural rainforests, immature rubber, and mature rubber plantations were analyzed using paired‐end Illumina sequencing. The conversion of natural rainforest to rubber plantations significantly altered fungal community composition of specific functional groups (saprotrophs, pathogens, and mycorrhiza). We observed significant loss of saprotrophic and ectomycorrhizal fungi in natural rainforests, but enrichment of plant pathogenic fungi in immature rubber plantations. The mechanism underlying the effects of forest conversion on changes in fungal communities is related to reductions in soil pH, total nitrogen (N), and ammonium (NH4) in rubber plantations. Conversion to rubber plantation also resulted in decline of soil respiration rates and less potential for cellulase and chitinase productions. The significant negative correlations between fungal richness and soil respiration in mature rubber plantations indicated high competition among fungi and low nutrient availability in this system. We demonstrate the negative consequences of the conversion of rainforest to rubber plantations on soil biological activity and significant changes in fungal community composition that could threaten long‐term ecosystem functions.

Discovery of Fusarium proliferatum f. sp. malus domestica Causing Apple Replant Disease in China.

Apple replant disease (ARD) is the most serious threat facing the apple industry globally. ARD is mainly manifested as decreased plant growth, serious root rot disease, and considerable yield loss. Microbial factors are the dominant factors leading to the occurrence of ARD. Research on soil-borne pathogenic fungi leading to the occurrence of ARD in China is limited. In the present study, we selected 16 replanting orchards from the Northwest Loess region and around the Bohai Gulf. Diseased roots and rhizosphere soil from healthy apple trees and trees showing ARD symptoms were sampled at random. High-throughput sequencing was used to study the fungal communities in the rhizosphere soil, which showed that the composition of the rhizosphere soil fungal community of ARD-symptomatic and healthy apple trees was different. Nectriaceae at the family level and Fusarium at the genus level dominated the rhizosphere soil fungal community in the two regions, while for healthy apple trees, the relative abundance of Mortierella, Minimedusa, Tetracladium, and Chaetomium was higher. Tissue separation and serial dilution were used to separate fungi, and a total of 89 genera and 219 species were obtained, most of which were Fusarium. Fusarium was further confirmed to be the most abundant pathogen species leading to the occurrence of ARD in China through pathogenicity assays. A pathogenicity assay was carried out by the dip-and-cut technique using different host plants. It was found that Fusarium MR5 showed strong aggressiveness to apple rootstocks. Diseased seedlings specifically exhibited chlorosis of the leaves, browning from the edge of the leaf, followed by rolling and yellowing of the leaves, resulting in wilting and eventually death. Strain MR5 was preliminarily identified as F. proliferatum according to the morphological and cultural characteristics. A maximum likelihood analysis of identities based on six gene sequence (ITS, TUB2, IGS, mtSSU, RPB2, and the TEF gene) alignments between the MR5 strain and other strains showed 99 to 100% homology with F. proliferatum. Based on our test results, strain MR5 was identified as F. proliferatum f. sp. malus domestica, which is of great significance for finding new measures to control ARD in China.

Effect of Aerated Irrigation on the Growth and Rhizosphere Soil Fungal Community Structure of Greenhouse Grape Seedlings

Conventional irrigation methods decrease greenhouse soil aeration, which leads to restricted root growth and reduced soil fungal abundance in greenhouse grapes. In this study, aerated irrigation equipment was used to investigate the effects of aerated irrigation on the biomass accumulation, root growth, and soil fungal community structure of grape seedlings. The results show that aerated irrigation significantly increased the root length, root surface area, root volume, and number of root tips by 38.5%, 32.1%, 62.1%, and 23.4%, respectively, at a soil depth of 20–40 cm (p ≤ 0.05). The chao1 index and ACE index of fungi at different soil depths under aerated irrigation were higher than those without aerated treatment; aerated irrigation changed the relative abundance of dominant fungi in rhizosphere soil. At a soil depth of 20–40 cm, aerated irrigation increased the abundance of Fusarium by 42.2%. Aerated irrigation also contributed to the abundance of the beneficial fungal genera Mortierella, Cladosporium, and Glomus. At a soil depth of 0–20 cm, the abundance of Mortierella in the soil that received aerated treatment was 180.6% higher than in the control treatment. These findings suggest that aerated irrigation is a promising strategy for the promotion of grape root growth and biomass accumulation, and it can also increase the abundance of some beneficial fungi.

Control efficiency of hexaconazole-lentinan against wheat sharp eyespot and wheat crown rot and the associated effects on rhizosphere soil fungal community.

The use of polysaccharides to induce the systemic immune response of plants for disease resistance has become an effective plant protection measure. Sharp eyespot wheat and crown rot wheat are serious diseases of wheat. In this study, the control effects of hexaconazole and lentinan (LNT) seed dressing of the two wheat diseases were evaluated by field experiments, and the effects of the seed dressing on plant growth, soil enzyme activity, and community diversity in the wheat rhizosphere were discussed. The results showed that the combined seed dressing of hexaconazole at 0.5 a.i. g·100 kg−1 and LNT at 4 a.i. g·100 kg−1 could significantly improve the control effect of the two wheat diseases. The combined treatment of hexaconazole and LNT had little effect on wheat soil enzyme activities. Different seed dressing treatments changed the fungal community structure in the wheat rhizosphere soil, and the combination of LNT and hexaconazole reduced the relative abundance of Rhizoctonia, Cladosporium, Fusarium, Bipolaris, and Gibberella in wheat planting soils. These findings suggested that the combined seed dressing of hexaconazole and LNT could effectively control soilborne diseases of wheat, concurrently could change in rhizosphere fungal community, and reduce in potential soilborne pathogens.

Optimal NPK Fertilizer Combination Increases Panax ginseng Yield and Quality and Affects Diversity and Structure of Rhizosphere Fungal Communities.

Soil microorganisms affect crop rhizospheres via the transformation and transport of nutrients, which has important influences on soil fertility, carbon sequestration, and plant yield and health in agroecosystems. There are few reports on the effects of fertilizer application on the growth of Panax ginseng (C. A. Mey.) or the structure of its rhizosphere microbial communities. In this study, an orthogonal experimental design was used to explore the effects of nine different combinations of nitrogen (N), phosphorus (P), and potassium (K) fertilizers with different amounts and proportions on ginseng growth and accumulation of ginsenosides and the structure of rhizosphere soil fungal communities. Soil without fertilization was the control. With the combined application of NPK, ginseng growth and development increased. The fertilization scheme N3P1K3, with N fertilizer at 50 g·m−2, P fertilizer at 15 g·m−2, and K fertilizer at 60 g·m−2, had the most comprehensive benefit and significantly increased ginseng rhizome biomass and ginsenoside contents (Rg1, Re, Rf, Rg2, Rb1, Ro, Rc, Rb2, Rb3, and Rd). Amplicon sequencing showed that NPK application increased the diversity of fungal communities in ginseng rhizospheres, whereas richness was bidirectionally regulated by proportions and amounts of NPK. Ascomycota was the dominant fungal phylum in ginseng rhizosphere soil, and relative abundances decreased with combined NPK application. Combined NPK application increased the relative abundance of potential beneficial fungi, such as Mortierella, but decreased that of potentially pathogenic fungi, such as Fusarium. Correlation analysis showed that potential beneficial fungi were significantly positively correlated with ginseng rhizome yield and ginsenoside contents, whereas the opposite relation was observed with potential pathogenic fungi. Thus, in addition to directly increasing crop growth, precise NPK application can also increase crop adaptability to the environment by shaping specific microbial communities. The results of this study suggest that the combined effects of biotic and abiotic processes on agricultural production determine crop yield and quality.

Influence of different irrigation methods on the alfalfa rhizosphere soil fungal communities in an arid region.

Xinjiang is the largest arid and saline agricultural region in China. The common irrigation methods in this area are traditional flood irrigation and drip irrigation. In this study, we investigated the effects of these two irrigation methods on the fungal diversity, community structures, and functions in alfalfa rhizosphere soil as well as the associated environmental factors in northern Tianshan Mountain (Xinjiang, China). Soil enzyme activities (urease and neutral phosphatase) were significantly higher in the drip-irrigated alfalfa rhizosphere soil than in the flood-irrigated alfalfa rhizosphere soil, whereas the fungal alpha diversity in the drip-irrigated alfalfa rhizosphere soil was significantly lower than that in the flood-irrigated alfalfa rhizosphere soil. Six dominant fungal phyla were identified (>0.1%), with Ascomycota being the most abundant in all soils, followed by Basidiomycota (5.47%), Mortierellomycota (1.07%), Glomeromycota (0.55%), Rozellomycota (0.27%), and Chytridiomycota (0.14%). Ascomycota and Glomeromycota species were significantly less abundant in drip-irrigated alfalfa rhizosphere soil than in flood-irrigated alfalfa rhizosphere soil. A LEFSe analysis identified Cladosporiaceae (20.8%) species as the most abundant marker fungi in drip-irrigated alfalfa rhizosphere soil. Of the 13 fungal functional groups identified on the basis of the functional annotation using the FUNGuild database, Ectomycorrhizal (22.29%) was the primary functional group. Compared with flood irrigation, drip irrigation significantly decreased the relative abundance of Ectomycorrhizal and Arbuscular_Mycorrhizal, while increasing the relative abundance of Plant_Pathogen, although not significantly (P = 0.19). Available potassium was revealed to be the main environmental factor influencing soil enzyme activities, fungal alpha diversity, fungal community structures, and fungal functions in response to the different irrigation methods. In conclusion, drip irrigation may be more appropriate than flood irrigation in the Tianshan dryland agricultural area for enhancing soil enzyme activities, but it may also increase the abundance of plant pathogenic fungi in the soil.

Changes in Rhizosphere Soil Fungal Communities of Pinus tabuliformis Plantations at Different Development Stages on the Loess Plateau.

The soil fungal community is an important factor in the forest ecosystems, and a better understanding of its composition and dynamic changes will contribute to the maintenance, preservation, and sustainable development of the forest ecosystems. Pinus tabuliformis has been widely planted for local ecological restoration on the Loess Plateau in China in recent decades. However, these plantations have been degraded to different degrees with increasing stand age. Hence, we tried to find the possible causes for the plantation degradation by analyzing soil environmental changes and soil fungal community composition at different stand ages. We collected rhizosphere soil samples from young (10-year-old), middle-aged (20-year-old), and near-mature (30-year-old) P. tabuliformis plantations in this region and characterized their soil properties and soil fungal community diversity and composition. Our results showed that with increasing stand age, the contents of organic carbon, ammonium nitrogen (AN) and nitrate nitrogen (NN) in the soil increased significantly, while the content of available phosphorus (AP) decreased significantly. The main factors affecting the composition of the soil fungal community were the contents of AP, AN, and NN in the soil. In addition, the genus Suillus was the dominant ectomycorrhizal (ECM) fungus in all periods of P. tabuliformis plantations in this region. The results of structural equation modeling showed that the community composition of ECM fungi was significantly correlated with stand age, soil NN, and AP contents, and that of pathogenic (PAG) fungi was significantly correlated with soil AN and AP contents. The decrease in the relative abundance of ECM fungi and the increase in the relative abundance of PAG fungi would exacerbate the degradation of P. tabulaeformis plantation. Our results illustrated that the content of soil AP is not only an important factor limiting the development of plantations, but it also significantly affects the community composition of soil fungi in the rhizosphere of the P. tabuliformis plantation. This study provides a novel insight into the degradation of P. tabuliformis plantations and builds a solid foundation for their subsequent management, restoration, and sustainable development on the Loess Plateau of China.

High-throughput sequencing reveals rhizosphere fungal community composition and diversity at different growth stages of Populus euphratica in the lower reaches of the Tarim River.

BackgroundPopulus euphratica is one of the most ancient and primitive tree species of Populus spp and plays an important role in maintaining the ecological balance in desert areas. To decipher the diversity, community structure, and relationship between rhizosphere fungi and environmental factors at different growth stages of P. euphratica demands an in-depth investigation.MethodsIn this study, P. euphratica at different growth stages (young, medium, overripe, and decline periods) was selected as the research object, based on the determination of the physicochemical properties of its rhizosphere soil, the fungal community structure and diversity of P. euphratica and their correlation with soil physicochemical properties were comprehensively analyzed through high-throughput sequencing technology (internal transcribed spacer (ITS)) and bioinformatics analysis methods.ResultsAccording to the analysis of OTU annotation results, the rhizosphere soil fungal communities identified in Populus euphratica were categorized into10 phyla, 36 classes, 77 orders, 165 families, 275 genera and 353 species. The alpha diversity analysis showed that there was no obvious change between the different growth stages, while beta diversity analysis showed that there were significantly differences in the composition of rhizosphere soil fungal communities between mature and overripe trees (R2 = 0.31, P = 0.001), mature and deadwood (R2 = 0.28, P = 0.001). Ascomycota and Basidiomycota were dominant phyla in the rhizosphere fungal community and the dominant genera were Geopora, Chondrostereum and unidentified_Sordariales_sp. The relative abundance of the top ten fungi at each classification level differed greatly in different stages. Canonical correspondence analysis (CCA) and Spearman’s correlation analysis showed that conductivity (EC) was the main soil factor affecting the composition of Populus euphratica rhizosphere soil fungal community (P < 0.01), followed by total dissolvable salts (TDS) and available potassium (AK) (P < 0.05).ConclusionsOur data revealed that the rhizosphere fungal communities at the different growth stages of P. euphratica have differences, conductivity (EC) was the key factor driving rhizosphere fungi diversity and community structure, followed by total dissolvable salts (TDS) and available potassium (AK).

Diversity and spatiotemporal dynamics of fungal communities in the rhizosphere soil of Lycium barbarum L.: a new insight into the mechanism of geoherb formation

Lycium barbarum L. is a well-known traditional geoherb in Ningxia, China. The fruits of L. barbarum contain several dietary constituents, and thus, they exert many beneficial effects on human health. However, a few studies have been conducted on the geoherb L. barbarum and its rhizosphere soil fungal community. In this study, we determined the physicochemical properties and fungal community structure of rhizosphere soil of L. barbarum from three regions of China, namely Ningxia (NX), Qinghai (QH), and Xinjiang (XJ), during three development stages of L. barbarum. Soil pH varied between 7.56 and 8.60 across the three regions, indicating that alkaline soil is conducive to the growth of L. barbarum. The majority of soil properties in NX, an authentic geoherb-producing area, were substantially inferior to those in XJ and QH during all three developmental stages. Total sugar, polysaccharide (LBP), and flavonoid contents were the highest in wolfberry fruits from NX. High-throughput sequencing showed that the abundance of the soil fungal population in NX was higher than that in QH and XJ during the flowering and fruiting stage and summer dormant stage. Moreover, the soil fungal diversity increased with the development of wolfberry. Ascomycota and Mortierellomycota were the predominant phyla in the rhizosphere fungal communities in all samples. Redundancy analysis showed a significant correlation of the soil-available phosphorus and LBP of wolfberry fruits with the fungal community composition. The characteristics of rhizosphere fungal communities determined in the present study provide insights into the mechanism of geoherb formation in NX wolfberry.

Co-inoculation of Trichoderma gamsii A5MH and Trichoderma harzianum Tr906 in wheat suppresses in planta abundance of the crown rot pathogen Fusarium pseudograminearum and impacts the rhizosphere soil fungal microbiome

Wheat root endophytic strains Trichoderma gamsii A5MH and T. harzianum Tr906 were assessed for their abilities to suppress in planta abundance of the crown rot pathogen Fusarium pseudograminearum and increase growth of durum wheat. Fusarium crown rot suppressive assays were conducted in natural Kandosol and Chromosol wheat cropping soils with greater disease expression observed in the Chromosol. Rhizosphere abundance of T. gamsii A5MH, T. harzianum Tr906 and F. pseudograminearum were quantified by species-specific qPCR at plant maturity. Strains A5MH and Tr906 actively colonized the wheat rhizosphere and suppressed in planta pathogen abundance in both soil types, with both inoculants persisting in roots and soil to 84 days postemergence. Strain A5MH increased wheat biomass in both soils, whereas strain Tr906 only increased biomass of Chromosol-grown wheat. Metabolites of strains A5MH and Tr906 decreased in vitro growth of wheat pathogenic F. pseudograminearum, Rhizoctonia solani and Pythium irregulare, but did not inhibit either inoculant. Compared with individually inoculated strains, co-inoculation of A5MH and Tr906 did not affect rhizosphere niche abundance, in planta pathogen suppressive efficacies or wheat growth-benefits of either inoculant in Chromosol-grown wheat. Fusarium crown rot symptoms however, only decreased in the A5MH and co-inoculated treatments. Each Trichoderma treatment had distinct impacts on the structure (beta diversity) of the indigenous rhizosphere soil fungal community of Chromosol-grown wheat. Strain A5MH and co-inoculated treatments had the greatest overall effects on soil fungal community (alpha) diversity and structure. Community differentiation was primarily attributed to increased abundance of Trichoderma spp. in the inoculated treatments and associated decreases in plant pathogenic and predominantly saprophytic fungal genera.

Effects of Flurochloridone Application on Rhizosphere Soil Fungal Community and Composition in Potato Growing Areas of the Qinghai-Tibet Plateau.

The application of herbicides to arable land is still the most effective and accepted method to protect plants from weeds. Extensive use of chemicals in conventional agricultural practices has resulted in continuous and serious environmental pollution. Flurochloridone (FLC) is a monophenyl pyrrolidinone selective herbicide that is commonly used to inhibit weeds that occur during the growth of potatoes. In recent years, research on the toxicity of FLC has gradually increased. However, it is relatively rare to analyze the role of FLC by studying the composition of soil microorganisms. Therefore, we used NGS methods to identify the fungal community structure of the low content soil (LS) and high content soil (HS) samples in this study. Subsequently, we identified the fungal community and composition differences of these two group samples using the statistical analysis. Despite the variances of fungal community and composition across the different samples within the group, the fungal composition of the LS samples and the HS samples. LS samples were predominated by Ascomycota, while the HS samples were predominated by Mortierellomycota and Basidiomycota. The major species in the LS samples were Plectosphaerella cucumerina and Trichocladium asperum, whereas the dominant species in the HS samples were Epicoccum nigrum and Cladosporium chasmanthicola. These results suggested that the LS samples and the HS samples had different rhizosphere soil fungal community and composition changes resulting from implementation of FLC in potato growing areas.
