Multiplex immunofluorescence staining of coverslip-mounted paraffin-embedded tissue sections.

Animal and human tissues are used extensively in physiological and pathophysiological research. Due to both ethical considerations and low availability, it is essential to maximize the use of these tissues. Therefore, the aim was to develop a new method allowing for multiplex immunofluorescence (IF) staining of kidney sections in order to reuse the same tissue section multiple times. The paraffin-embedded kidney sections were placed onto coated coverslips and multiplex IF staining was performed. Five rounds of staining were performed where each round consisted of indirect antibody labelling, imaging on a widefield epifluorescence microscope, removal of the antibodies using a stripping buffer, and then re-staining. In the final round, the tissue was stained with hematoxylin/eosin. Using this method, tubular segments in the nephron, blood vessels, and interstitial cells were labeled. Furthermore, by placing the tissue on coverslips, confocal-like resolution was obtained using a conventional widefield epifluorescence microscope and a 60x oil objective. Thus, using standard reagents and equipment, paraffin-embedded tissue was used for multiplex IF staining with increased Z-resolution. In summary, this method offers time-saving multiplex IF staining and allows for the retrieval of both quantitative and spatial expressional information of multiple proteins and subsequently for an assessment of the tissue morphology. Due to the simplicity and integrated effectivity of this multiplex IF protocol, it holds the potential to supplement standard IF staining protocols and maximize use of tissue.

Artificial intelligence and digital biomarker in precision pathology guiding immune therapy selection and precision oncology.

The currently available immunotherapies already changed the strategy how many cancers are treated from first to last line. Understanding even the most complex heterogeneity in tumor tissue and mapping the spatial cartography of the tumor immunity allows the best and optimized selection of immune modulating agents to (re-)activate the patient's immune system and direct it against the individual cancer in the most effective way. Primary cancer and metastases maintain a high degree of plasticity to escape any immune surveillance and continue to evolve depending on many intrinsic and extrinsic factors In the field of immune-oncology (IO) immune modulating agents are recognized as practice changing therapeutic modalities. Recent studies have shown that an optimal and lasting efficacy of IO therapeutics depends on the understanding of the spatial communication network and functional context of immune and cancer cells within the tumor microenvironment. Artificial intelligence (AI) provides an insight into the immune-cancer-network through the visualization of very complex tumor and immune interactions in cancer tissue specimens and allows the computer-assisted development and clinical validation of such digital biomarker. The successful implementation of AI-supported digital biomarker solutions guides the clinical selection of effective immune therapeutics based on the retrieval and visualization of spatial and contextual information from cancer tissue images and standardized data. As such, computational pathology (CP) turns into "precision pathology" delivering individual therapy response prediction. Precision Pathology does not only include digital and computational solutions but also high levels of standardized processes in the routine histopathology workflow and the use of mathematical tools to support clinical and diagnostic decisions as the basic principle of a "precision oncology".

Inhibition of the ventral midline thalamus does not alter encoding, short-term holding or retrieval of spatial information in rats performing a water-escape working memory task

Working memory (WM) is a function operating in three successive phases: encoding (sample trial), holding (delay), and retrieval (test trial) of information. Studies point to a possible implication of the thalamic reuniens nucleus (Re) in spatial WM (SWM). In which of the aforementioned 3 phases the Re has a function is largely unknown. Recently, in a delayed SWM water-escape task, we found that performance during the retrieval trial correlated positively with c-Fos expression in the Re nucleus, suggesting participation in retrieval. Here, we used the same task and muscimol (MUSC) inhibition or DREADD(hM4Di)-mediated inhibition of the Re during information encoding, right thereafter (thereby affecting the holding phase), or during the retrieval trial. A 6-hour delay separated encoding from retrieval. Concerning SWM, MUSC in the Re nucleus did not alter performance, be it during or after encoding, or during evaluation. CNO administered before encoding in DREADD-expressing rats was also ineffective, although CNO-induced inhibition disrupted set shifting performance, as found previously (Quet et al., Brain Struct Function 225, 2020), thereby validating DREADD efficiency. These findings are the first that do not support an implication of the Re nucleus in SWM. As most previous studies used T-maze alternation tasks, which carry high proactive interference risks, an important question to resolve now is whether the Re nucleus is required in (T-maze alternation) tasks using very short information-holding delays (seconds to minutes), and less so in other short-term spatial memory tasks with longer information holding intervals (hours) and therefore reduced interference risks.

Pixel super-resolution with spatially entangled photons

Pixelation occurs in many imaging systems and limits the spatial resolution of the acquired images. This effect is notably present in quantum imaging experiments with correlated photons in which the number of pixels used to detect coincidences is often limited by the sensor technology or the acquisition speed. Here, we introduce a pixel super-resolution technique based on measuring the full spatially-resolved joint probability distribution (JPD) of spatially-entangled photons. Without shifting optical elements or using prior information, our technique increases the pixel resolution of the imaging system by a factor two and enables retrieval of spatial information lost due to undersampling. We demonstrate its use in various quantum imaging protocols using photon pairs, including quantum illumination, entanglement-enabled quantum holography, and in a full-field version of N00N-state quantum holography. The JPD pixel super-resolution technique can benefit any full-field imaging system limited by the sensor spatial resolution, including all already established and future photon-correlation-based quantum imaging schemes, bringing these techniques closer to real-world applications.

Cognitive characteristics in firefighter wayfinding Tasks: An Eye-Tracking analysis

During search and rescue, firefighters need to find paths in an unfamiliar space with minimum time and information available. The effective memorization and retrieval of critical spatial information can help reduce risks and increase mission efficiency. Although evidence has shown that different formats of wayfinding information, including landmarks, routes, and surveys, can impact search and rescue performance in different manners, a deeper understanding of the characteristics of firefighters’ cognitive processes related to the varying wayfinding information formats is less explored. To evaluate firefighters’ performance and cognitive characteristics in search and rescue, a firefighter experiment in Virtual Reality (VR) was conducted. Firefighters (n = 40) were recruited to participate in the simulated rescue task. After reviewing the spatial information in different formats, firefighters were requested to find three victims inside a VR maze as quickly as possible. Task performance was evaluated by the number of victims found and the time spent. Firefighters’ gaze patterns were analyzed to evaluate their cognitive status. The result showed that although the cognitive load under the survey and route conditions was significantly higher than under the landmark condition (p < 0.001), the decision-making involved a more effective cognitive process related to choosing the right path at critical waypoints such as where a turning decision must be made. Thus, the perceived workload and fatigue levels of the two conditions were lower, and the wayfinding performance was better. In contrast, with landmark information, the cognitive load levels were consistently high, along with increased mental fatigue.The findings reveal a series of cognitive features related to a more effective spatial decision-making in search and rescue. In the future, it is expected that these cognitive features can be used to develop real-time monitoring and prediction models for wayfinding performance.

Prefrontal pyramidal neurons are critical for all phases of working memory.

The prefrontal cortex (PFC) is essential for working memory (WM) and has primarily been viewed as being responsible for maintaining information over a delay, but it is unclear whether it also plays a more general role during WM. Using task phase-specific optogenetic silencing of pyramidal neurons in the medial PFC (mPFC) of mice performing a spatial WM task, we find that the mPFC is required not only during the delay phase of the task but also during other phases requiring the encoding and retrieval of spatial information. Imaging of mPFC pyramidal neurons reveals that they are most strongly influenced by the animals' position and running direction, indicating a fundamental role in spatial navigation. Pyramidal neuron ensembles also represent to-be-remembered goal locations in a dynamic manner. Taken together, these results delineate the functional contribution of mPFC pyramidal neurons to WM, extending their role beyond the maintenance of information.

The influence of early visual deprivation on audio-spatial working memory.

This study investigates how spatial working memory skills, and the processing and retrieval of distal auditory spatial information are influenced by visual experience. We developed an experimental paradigm using an acoustic simulation. The performance of congenitally blind and sighted participants (n = 9 per group) was compared when recalling sequences of spatialised auditory items in the same or reverse order of presentation. Two experimental conditions based on stimuli features were tested: non-semantic and semantic. Blind participants had a shorter memory span in the backward than the forward order of presentation. In contrast, sighted participants did not, revealing that blindness affects spatial information processing with greater executive source involvement. Furthermore, we found that blind subjects performed worse overall than the sighted group and that the semantic information significantly improved the performance, regardless of the experimental group and the sequences' order of presentation. Lack of early visual experience affects the ability to encode the surrounding space. Congenital blindness influences the processing and retrieval of spatial auditory items, suggesting that visual experience plays a pivotal role in calibrating spatial memory abilities using the remaining sensory modalities. Quantifying the data quality of focal series for inline electron holography

Inline electron holography, the recovery of amplitude and phase of an electron wave function having passed through a thin specimen from a focal series recorded in a transmission electron microscope is being applied in many labs worldwide. At medium range magnification (i.e. typically ≥0.8 nm where the lattice of small unit cell crystals such as silicon is not resolved), where the defocus needs to be varied over a rather large range of several hundred nm or even μm, the retrieval of low spatial frequency information is severely affected by the choice of experimental parameters as well as the way of data normalization. Methods to quantitatively analyze the reliability of phase maps obtained by inline electron holography are presented, and data recorded and processed in different ways are compared. While, even under optimized conditions, the phase reconstructed from an experimental focal series still lacks very low spatial frequency components, regularization schemes exist and are demonstrated to effectively hide artifacts associated with this lack of information.

Prefrontal Pyramidal Neurons are Critical for All Phases of Working Memory

The prefrontal cortex (PFC) has long been known to be essential for working memory (WM), yet exactly how it contributes to this fundamental cognitive process remains incompletely understood. The PFC has primarily been viewed as being responsible for the maintenance of information over a delay, but it is unclear whether it also plays a more general role during WM. To address this, we performed task phase-specific optogenetic silencing of pyramidal neurons in the medial PFC (mPFC) of mice performing a spatial WM task. We found that the mPFC was required not only during the delay phase of the task but also during other phases requiring the encoding and retrieval of spatial information. To gain further insights into how WM is supported by mPFC pyramidal neurons, we imaged their activity during all phases of the task using head-mounted miniscopes. A computational cognitive model of judgments of relative direction

In the past several decades, considerable theoretical progress has been made in understanding the role of reference frames in the encoding and retrieval of spatial information about the environment. Many of these insights have come from participants making judgments of relative direction using their memories of spatial layouts. In this task, participants are asked to imagine standing at a given location and facing a certain direction, and to point to a target location. Although this task has been widely and productively used, a computational cognitive model of judgments of relative direction has yet to be introduced. Computational modeling of judgments of relative direction is a critical next step to formulating and testing hypotheses about the cognitive processes involved in establishing and using spatial reference frames. We present an initial attempt to model judgments of relative direction and fit the model to two datasets exhibiting behavioral patterns commonly observed in the spatial memory literature. The model was able to predict many important features of these data, most notably alignment effects. We discuss directions for future modeling efforts.

Changing Reward Expectation Transforms Spatial Encoding and Retrieval in the Hippocampus

Internal states of reward expectation play a central role in influencing the strength of spatial memories. At the cellular level, spatial memories are represented through the firing dynamics of hippocampal place cells. However, it remains unclear how internal states of reward expectation influence place cell dynamics and exert their effects on spatial memories. Here we show that when reward expectation is altered, the same environment becomes encoded by a distinct ensemble of place cells at all locations. Further, when reward expectation is high versus low, place cells demonstrate enhanced reliability during navigation and greater stability across days at all locations within the environment. These findings reveal that when rewards are expected, neuromodulatory circuits that represent internal reward expectation support and strengthen the encoding and retrieval of spatial information by place cells at all locations that lead to reward. This enhanced spatial memory can be used to guide future decisions about which locations are most likely to lead to rewards that are crucial for survival.

Evaluation of the Crosta method for the retrieval of water quality parameters from remote sensing data in the Pearl River estuary

Abstract In recent decades, many algorithms have been developed for the retrieval of water quality parameters using remotely sensed data. However, these algorithms are specific to a certain geographical area and cannot be applied to other areas. In this study, feature-orientated principal component (PC) selection, based on the Crosta method and using Landsat Thematic Mapper (TM) for the retrieval of water quality parameters (i.e., total suspended sediment concentration (TSM) and chlorophyll a (Chla)), was carried out. The results show that feature-orientated PC TSM, based on the Crosta method, obtained a good agreement with the MERIS-based TSM product for eight Landsat TM images. However, the Chla information, selected using the feature-orientated PC, has a poor agreement with the MERIS-based Chla product. The accuracy of the atmospheric correction method and MERIS product may be the main factors influencing the accuracy of the TSM and Chla information identified by the Landsat TM images using the Crosta method. The findings of this study would be helpful in the retrieval of spatial distribution information on TSM from the long-term historical Landsat image archive, without using coincident ground measurements. The influence of encoding and testing directions on retrieval of spatial information in explored and described environments

The verbal descriptions of an environment elicit a spatial mental model, in which the linear disposition of the described objects might be related to the properties of the description. In particular the direction from which the environment is encoded might shape the spatial mental model, as a consequence of a cultural bias in reading and writing direction. The aim of the present study was to examine the influence of the direction in which objects are encoded on the retrieval of spatial information. In two experiments we asked participants to encode an environment through either physical exploration or verbal description, that are encoding modalities which preserve the sequential presentation of spatial information. We manipulated both the encoding and testing directions of the spatial information, and tested participants by using a two-alternative forced choice task. In both experiments, the results did not reveal any significant effect, disconfirming the idea of the left-right cultural bias for western people for this type of task. The lack of effect suggests that encoding an environment through physical movement and verbal descriptions determines the development of a mental representation which is relatively independent from encoding sequential order.

Orientation-dependent spatial memories for scenes viewed on mobile devices.

We examined whether spatial representations for scenes experienced on the screens of mobile devices are orientation dependent and whether the type of movement (physical vs. simulated) during learning affects the encoding and the retrieval of spatial information. Participants studied a spatial layout depicted on a tablet and then carried out perspective-taking trials in which they localized objects from imagined perspectives. Depending on condition, participants either rotated the tablet along with their body or remained stationary and swiped with their finger on the screen to change their viewpoint within the scene. Results showed that participants were faster and more accurate to point to objects from an imagined perspective that was aligned than misaligned to their initial physical orientation during learning, suggesting that they had formed an orientation-dependent representation. Although no differences were found between movement conditions during pointing, participants were faster to encode spatial information with physical than simulated movement.

Spatial Representations in the Human Brain.

While extensive research on the neurophysiology of spatial memory has been carried out in rodents, memory research in humans had traditionally focused on more abstract, language-based tasks. Recent studies have begun to address this gap using virtual navigation tasks in combination with electrophysiological recordings in humans. These studies suggest that the human medial temporal lobe (MTL) is equipped with a population of place and grid cells similar to that previously observed in the rodent brain. Furthermore, theta oscillations have been linked to spatial navigation and, more specifically, to the encoding and retrieval of spatial information. While some studies suggest a single navigational theta rhythm which is of lower frequency in humans than rodents, other studies advocate for the existence of two functionally distinct delta–theta frequency bands involved in both spatial and episodic memory. Despite the general consensus between rodent and human electrophysiology, behavioral work in humans does not unequivocally support the use of a metric Euclidean map for navigation. Formal models of navigational behavior, which specifically consider the spatial scale of the environment and complementary learning mechanisms, may help to better understand different navigational strategies and their neurophysiological mechanisms. Finally, the functional overlap of spatial and declarative memory in the MTL calls for a unified theory of MTL function. Such a theory will critically rely upon linking task-related phenomena at multiple temporal and spatial scales. Understanding how single cell responses relate to ongoing theta oscillations during both the encoding and retrieval of spatial and non-spatial associations appears to be key toward developing a more mechanistic understanding of memory processes in the MTL.

Optogenetic suppression of the nucleus reuniens selectively impairs encoding during spatial working memory.

The nucleus reuniens (Re) of the ventral midline thalamus is known to be a critical anatomical link between the hippocampus (HPC) and the medial prefrontal cortex (mPFC). Consistent with this anatomical connectivity, the Re has been shown to be crucial for HPC-mPFC oscillatory synchrony. Moreover, Re inhibition consistently results in spatial working memory (SWM) deficits. Together, these results suggest that SWM requires HPC-mPFC synchrony via the Re. In spite of these findings, an understanding of how the Re contributes to the encoding, maintenance, and retrieval of spatial information during a SWM task is lacking. To address this issue, we trained rats to perform a SWM-dependent delayed-non-match-to-position (DNMP) task in a T-maze. Using optogenetic inhibition of Re activity, we demonstrated that Re suppression during the sample phase, but not the delay or choice phase, significantly decreased choice accuracy. We conclude that the Re contributes to the encoding of spatial information during working memory.

Digital sketch maps and eye tracking statistics as instruments to obtain insights into spatial cognition.

This paper explores map users' cognitive processes in learning, acquiring and remembering information presented via screen maps. In this context, we conducted a mixed-methods user experiment employing digital sketch maps and eye tracking. On the one hand, the performance of the participants was assessed based on the order with which the objects were drawn and the influence of visual variables (e.g. presence & location, size, shape, color). On the other hand, trial durations and eye tracking statistics such as average duration of fixations, and number of fixations per seconds were compared. Moreover, selected AoIs (Area of Interests) were explored to gain a deeper insight on visual behavior of map users. Depending on the normality of the data, we used either two-way ANOVA or Mann-Whitney U test to inspect the significance of the results. Based on the evaluation of the drawing order, we observed that experts and males drew roads first whereas; novices and females focused more on hydrographic object. According to the assessment of drawn elements, no significant differences emerged between neither experts and novices, nor females and males for the retrieval of spatial information presented on 2D maps with a simple design and content. The differences in trial durations between novices and experts were not statistically significant while both studying and drawing. Similarly, no significant difference occurred between female and male participants for either studying or drawing. Eye tracking metrics also supported these findings. For average duration of fixation, there was found no significant difference between experts and novices, as well as between females and males. Similarly, no significant differences were found for the mean number of fixation.

Action compatibility in spatial knowledge developed through virtual navigation.

Action-compatibility effects (ACEs) arise due to incongruity between perceptuo-motor traces stored in memory and the perceptuo-motor demands of a retrieval task. Recent research has suggested that ACEs arising during spatial memory retrieval are additionally modulated by individual differences in how experienced participants are with a college campus environment. However, the extent and nature of experience with a real-world environment is difficult to assess and control, and characteristics of the retrieval task itself might modulate ACEs during spatial memory retrieval. The present study provides a more controlled and in-depth examination of how individual differences and task-based factors interact to shape ACEs when participants retrieve spatial memories. In two experiments, participants with varied video game experience learned a virtual environment and then used the computer mouse to verify spatial relationships from different perspectives. Mouse trajectories demonstrated ACEs, differing by retrieval perspective and video game experience. Videogame experts demonstrated the ACE based on learned spatial relationships during egocentric retrieval only, whereas videogame novices showed the ACE based on semantic processing of directional terms only. Specifically, gaming experts invoke perspective-specific perceptuo-motor associations to retrieve spatial knowledge, whereas non-experts are influenced by semantically based associations specific to the retrieval task. Results are discussed in the context of action-compatibility effects, the intentional weighting hypothesis, and the flexible encoding and retrieval of spatial information.

Brain connectivity during encoding and retrieval of spatial information: individual differences in navigation skills

Emerging evidence suggests that the variations in the ability to navigate through any real or virtual environment are accompanied by distinct underlying cortical activations in multiple regions of the brain. These activations may appear due to the use of different frame of reference (FOR) for representing an environment. The present study investigated the brain dynamics in the good and bad navigators using Graph Theoretical analysis applied to low-density electroencephalography (EEG) data. Individual navigation skills were rated according to the performance in a virtual reality (VR)-based navigation task and the effect of navigator's proclivity towards a particular FOR on the navigation performance was explored. Participants were introduced to a novel virtual environment that they learned from a first-person or an aerial perspective and were subsequently assessed on the basis of efficiency with which they learnt and recalled. The graph theoretical parameters, path length (PL), global efficiency (GE), and clustering coefficient (CC) were computed for the functional connectivity network in the theta and alpha frequency bands. During acquisition of the spatial information, good navigators were distinguished by a lower degree of dispersion in the functional connectivity compared to the bad navigators. Within the groups of good and bad navigators, better performers were characterised by the formation of multiple hubs at various sites and the percentage of connectivity or small world index. The proclivity towards a specific FOR during exploration of a new environment was not found to have any bearing on the spatial learning. These findings may have wider implications for how the functional connectivity in the good and bad navigators differs during spatial information acquisition and retrieval in the domains of rescue operations and defence systems.

Effect of a hallucinogenic serotonin 5-HT2A receptor agonist on visually guided, hippocampal-dependent spatial cognition in C57BL/6J mice.

By acting on serotonin 5-HT2A receptors (5-HT2A Rs), serotonergic psychedelic drugs induce perceptual and visual hallucinations by increasing neuronal excitability and altering visual-evoked neuronal responses. The present study was designed to examine whether the perceptual alterations induced by a serotonergic psychedelic drug would affect the integrity of hippocampal-dependent, visually guided spatial cognition. phenylalkylamine hallucinogen TCB-2 is a selective agonist of 5-HT2A Rs. Mice received TCB-2 (1.0 mgkg-1 , i.p.), and spatial behaviors and hippocampal electrophysiological responses were measured with water maze tasks and in vivo single-unit recording, respectively. TCB-2 did not affect visual cue approach behavior in the visible platform water maze, but increased the latency of trained mice to initiate goal-directed swimming during a probe test in the hidden platform Morris water maze, which could be prevented by 5-HT2A R antagonist MDL 11,939. Interestingly, TCB-2 did not affect the efficiency of the swim path or the proper use of distal visual cues during the probe test. Hippocampal place cell activity is considered to represent spatial and context-specific episodic memory. Systemic TCB-2 did not affect previously established place fields of CA1 neurons in mice exploring a familiar environment, or the remapping of place cells when the mice explored a novel environment. However, TCB-2 impaired the long-term stability of place fields for the novel environment initially encoded under the influence of TCB-2, which could be prevented by 5-HT2A R antagonist MDL 11,939. Our data indicate that hallucinogenic 5-HT2A R agonist delays the initiation of spatial search behavior, but does not impair the use of visual cues to guide goal-directed spatial behavior. Moreover, activation of 5-HT2A Rs does not impair the coding and retrieval of spatial information, but impairs the long-term stability of new formed place fields of CA1 neurons. © 2017 Wiley Periodicals, Inc.
