Natural genetic variation in dynamic photosynthesis is correlated with stomatal anatomical traits in diverse tomato species across geographical habitats.

Plants grown under field conditions experience fluctuating light. Understanding the natural genetic variations for a similarly dynamic photosynthetic response among untapped germplasm resources, as well as the underlying mechanisms, may offer breeding strategies to improve production using molecular approaches. Here, we measured gas exchange under fluctuating light, along with stomatal density and size, in eight wild tomato species and two tomato cultivars. The photosynthetic induction response showed significant diversity, with some wild species having faster induction rates than the two cultivars. Species with faster photosynthetic induction rates had higher daily integrated photosynthesis, but lower average water use efficiency because of high stomatal conductance under natural fluctuating light. The variation in photosynthetic induction was closely associated with the speed of stomatal responses, highlighting its critical role in maximizing photosynthesis under fluctuating light conditions. Moreover, stomatal size was negatively correlated with stomatal density within a species, and plants with smaller stomata at a higher density had a quicker photosynthetic response than those with larger stomata at lower density. Our findings show that the response of stomatal conductance plays a pivotal role in photosynthetic induction, with smaller stomata at higher density proving advantageous for photosynthesis under fluctuating light in tomato species. The interspecific variation in the rate of stomatal responses could offer an untapped resource for optimizing dynamic photosynthetic responses under field conditions.

Effects of increasing atmospheric CO2 on leaf water δ18O values are small and are attenuated in grasses and amplified in dicotyledonous herbs and legumes when transferred to cellulose δ18O values.

The oxygen isotope composition of cellulose (δ18O values) has been suggested to contain information on stomatal conductance (gs) responses to rising pCO2. The extent by which pCO2 affects leaf water and cellulose δ18O values (δ18OLW and δ18OC) and the isotope processes that determine pCO2 effects on δ18OLW and δ18OC are, however, unknown. We tested the effects of pCO2 on gs, δ18OLW and δ18OC in a glasshouse experiment, where six plant species were grown under pCO2 ranging from 200 to 500 ppm. Increasing pCO2 caused a decline in gs and an increase in δ18OLW, as expected. Importantly, the effects of pCO2 on gs and δ18OLW were small and pCO2 effects on δ18OLW were not directly transferred to δ18OC but were attenuated in grasses and amplified in dicotyledonous herbs and legumes. This is likely because of functional group-specific pCO2 effects on the model parameter pxpex. Our study highlights important uncertainties when using δ18OC as a proxy for gs. Specifically, pCO2-triggered gs effects on δ18OLW and δ18OC are possibly too small to be detected in natural settings and a pCO2 effect on pxpex may render the commonly assumed negative linkage between δ18OC and gs to be incorrect, potentially confounding δ18OC based gs reconstructions.

Non-steady-state stomatal conductance modeling and its implications: from leaf to ecosystem

Abstract. Accurate and efficient modeling of stomatal conductance (gs) has been a key challenge in vegetation models across scales. Current practice of most land surface models (LSMs) assumes steady-state gs and predicts stomatal responses to environmental cues as immediate jumps between stationary regimes. However, the response of stomata can be orders of magnitude slower than that of photosynthesis and often cannot reach a steady state before the next model time step, even on half-hourly timescales. Here, we implemented a simple dynamic gs model in the vegetation module of an LSM developed within the Climate Modeling Alliance and investigated the potential biases caused by the steady-state assumption from leaf to canopy scales. In comparison with steady-state models, the dynamic model better predicted the coupled temporal response of photosynthesis and stomatal conductance to changes in light intensity using leaf measurements. In ecosystem flux simulations, while the impact of gs hysteresis response may not be substantial in terms of monthly integrated fluxes, our results highlight the importance of considering this effect when quantifying fluxes in the mornings and evenings, as well as interpreting diurnal hysteresis patterns observed in ecosystem fluxes. Simulations also indicate that the biases in the integrated fluxes are more significant when stomata exhibit different speeds for opening and closure. Furthermore, prognostic modeling can bypass the A-Ci iterations required for steady-state simulations and can be robustly run with comparable computational costs. Overall, our study demonstrates the implications of dynamic gs modeling for improving the accuracy and efficiency of LSMs and for advancing our understanding of plant–environment interactions.

The interplay of short-term mesophyll and stomatal conductance responses under variable environmental conditions.

Understanding the short-term responses of mesophyll conductance (gm ) and stomatal conductance (gsc ) to environmental changes remains a challenging yet central aspect of plant physiology. This review synthesises our current knowledge of these short-term responses, which underpin CO2 diffusion within leaves. Recent methodological advances in measuring gm using online isotopic discrimination and chlorophyll fluorescence have improved our confidence in detecting short-term gm responses, but results need to be carefully evaluated. Environmental factors like vapour pressure deficit and CO2 concentration indirectly impact gm through gsc changes, highlighting some of the complex interactions between the two parameters. Evidence suggests that short-term responses of gm are not, or at least not fully, mechanistically linked to changes in gsc , cautioning against using gsc as a reliable proxy for gm . The overarching challenge lies in unravelling the mechanistic basis of short-term gm responses, which will contribute to the development of accurate models bridging laboratory insights with broader ecological implications. Addressing these gaps in understanding is crucial for refining predictions of gm behaviour under changing environmental conditions.

Autumnal Potassium Induced Modulations in Plant Osmoprotectant Substances, Nutrient Stoichiometry and Precision Sustainable Seedling Cultivation in Parashorea chinensis

Parashorea chinensis, an endemic tree species in China’s tropical rainforests, holds ecological and economic importance. Challenges like low resistance, poor quality, and low survival rates hinder its successful cultivation. This study explores the potential of autumn potassium fertilization on Parashorea seedlings from two provenances (Napo and Tianyang). The treatments included no fertilizer (CK-1), a single application of 160 mg K·plant−1 (CK-2), and various potassium levels K1, K2, K3, K4, K5, and K6 (corresponding to 0, 40, 80, 160, 320, and 640 mg·K·plant−1, respectively) combined with nitrogen (200 mg·plant−1) and phosphorus (80 mg·plant−1) fertilization. The findings indicate that autumn potassium application, in conjunction with nitrogen (N) and phosphorus (P) fertilization, significantly enhances seedling height and biomass in both provenances, resulting in an average increase of 101% and 89% under the K4 treatment compared to CK-1 and CK-2, comparatively. Both Napo and Tianyang provenances exhibited distinct responses in photosynthetic rate (2.70 μmol·m−2·s−1 and 1.97 μmol·m−2·s−1, respectively) and stomatal conductance (0.042 mol·m−2·s−1 and 0.029 mol·m−2·s−1, respectively) to the K4 treatment, which proved most effective. The chlorophyll content was significantly higher for Napo provenance with the K3 treatment (74.31%, 58.99%), while for Tianyang, it was higher with the K4 treatment (41.48%, 17.36%), compared to CK-1 and CK-2, respectively. Antioxidant enzymes activity, osmoregulatory capacity, and malondialdehyde content all exhibited variations with potassium application levels, with the K4 treatment offering significant benefits. In Napo provenance, lignin (199.82 mg·g−1) and cellulose (252.38 mg·g−1) peaked at K4, while Tianyang exhibited variation, higher lignin (184.25 mg·g−1) at K3, and cellulose (257.73 mg·g−1) at K4. Nutrient content analysis demonstrates that the K4 treatment enhances nutrient absorption and storage, increasing total N (21.56 mg·kg−1), P (4.69 mg·kg−1), and K (13.49 mg·kg−1) content. A comprehensive analysis reveals that the K4 treatment yields the highest quality scores (1.87, 1.85) and membership values (0.82, 0.68) for both Napo and Tianyang seedlings, with Napo seedlings outperforming their Tianyang provenance. Thus, treatment K4 underscores the effectiveness of autumn potassium applications for robust seedling cultivation and adaptation, offering valuable insights for sustainable cultivation practices.

Stomatal responses to VPD utilize guard cell intracellular signaling components.

Stomatal pores, vital for CO2 uptake and water loss regulation in plants, are formed by two specialized guard cells. Despite their importance, there is limited understanding of how guard cells sense and respond to changes in vapor pressure difference (VPD). This study leverages a selection of CO2 hyposensitive and abscisic acid (ABA) signaling mutants in Arabidopsis, including heterotrimeric G protein mutants and RLK (receptor-like kinase) mutants, along with a variety of canola cultivars to delve into the intracellular signaling mechanisms prompting stomatal closure in response to high VPD. Stomatal conductance response to step changes in VPD was measured using the LI-6800F gas exchange system. Our findings highlight that stomatal responses to VPD utilize intracellular signaling components. VPD hyposensitivity was particularly evident in mutants of the ht1 (HIGH LEAF TEMPERATURE1) gene, which encodes a protein kinase expressed mainly in guard cells, and in gpa1-3, a null mutant of the sole canonical heterotrimeric Gα subunit, previously implicated in stomatal signaling. Consequently, this research identifies a nexus in the intricate relationships between guard cell signal perception, stomatal conductance, environmental humidity, and CO2 levels.

Response of stomatal conductance to plant water stress in buffalograss seed production: Observation with UAV thermal infrared imagery

Stomatal conductance (gs) is an indicator that allows for direct evaluation of plant water status, but it is challenging to achieve rapid monitoring in large-scale fields due to limitations in observation methods. Here this study was conducted to identify the thresholds of gs with different target yields and develop a gs-based water stress diagnostic model for buffalograss (Buchloe dactyloides (Nutt.) Engelm.) using UAV thermal infrared imagery for buffalograss in 2022 and 2023. The results of the field experiment demonstrated that the gs rapidly response to changes in the water stress status of buffalograss. The thresholds of gs were 403 and 385 mmol m−2 s−1 for the vegetative and reproductive growth stages, respectively, with the target seed yield of 1224 kg ha−1. The gs values were classified into three levels for the vegetative growth and four levels for the in reproductive growth stage of buffalograss, respectively. The canopy temperature depression response to water stress is consistent with the gs. Based on this relationship, this study developed a gs-based diagnostic model with a random forest algorithm for buffalograss. Furthermore, a spital map of gs was created using UAV thermal infrared imagery. The modification test results indicated that the model made a good estimation of gs were good with normalized root mean square errors of 15% in the vegetative stage and 11% in the reproductive stage, respectively. Therefore, it is feasible to use thermal infrared imagery for monitoring gs and evaluating the water stress of plants in buffalograss fields.

How are physiological responses to drought modulated by water relations and leaf economics’ traits in woody plants?

Species’ drought resistance is determined by a combination of multiple traits and their plastic response. However, a clear understanding of how these traits are coordinated and modulate plant responses to drought is still lacking.Here we used a water exclusion experiment on 20 Mediterranean woody species to evaluate a new framework to study plant drought responses, in which relatively constant functional traits modulate the physiological responses to a given drought exposure. In particular, we assessed how the response of stomatal conductance (Gs), leaf relative water content (RWC) and leaf water potential difference (ΔΨ) to leaf predawn water potential (Ψpd) were modulated by commonly used functional traits. The latter included hydraulic and water relations attributes such as turgor loss point in leaves (Ψtlp), vulnerability to xylem embolism in stems (Ψ50), hydraulic conductivity per unit sapwood (Ks), hydraulic conductivity per unit leaf area (Kl, hydraulic sufficiency) and Huber value (Hv), as well as specific leaf area (SLA) as a proxy for the leaf economics spectrum.Our results show that functional traits are highly coordinated, defining two main axes: the first related to drought tolerance and resource-use strategies and the second characterising the hydraulic sufficiency of leaves. These two axes, as well as many of the underlying functional traits, showed a significant interaction with Ψpd in explaining physiological responses to drought, thus modulating Gs, RWC, and ΔΨ responses. Drought-tolerant species showed a less stringent regulation of water use and leaf water potential but were more effective at regulating leaf RWC. Also, the leaf water potential at turgor loss (Ψtlp) emerged as an important trait orchestrating plant responses to drought.This study highlights that a clear separation between slow-varying traits (Ψtlp, Ψ50, Ks, Kl, Hv, SLA), physiological responses (Gs, RWC, ΔΨ) and tissue-level exposure to drought (Ψpd) improves our understanding of plant drought resistance strategies.

Ecophysiological adaptations shape distributions of closely related trees along a climatic moisture gradient

Tradeoffs between the energetic benefits and costs of traits can shape species and trait distributions along environmental gradients. Here we test predictions based on such tradeoffs using survival, growth, and 50 photosynthetic, hydraulic, and allocational traits of ten Eucalyptus species grown in four common gardens along an 8-fold gradient in precipitation/pan evaporation (P/Ep) in Victoria, Australia. Phylogenetically structured tests show that most trait-environment relationships accord qualitatively with theory. Most traits appear adaptive across species within gardens (indicating fixed genetic differences) and within species across gardens (indicating plasticity). However, species from moister climates have lower stomatal conductance than others grown under the same conditions. Responses in stomatal conductance and five related traits appear to reflect greater mesophyll photosynthetic sensitivity of mesic species to lower leaf water potential. Our data support adaptive cross-over, with realized height growth of most species exceeding that of others in climates they dominate. Our findings show that pervasive physiological, hydraulic, and allocational adaptations shape the distributions of dominant Eucalyptus species along a subcontinental climatic moisture gradient, driven by rapid divergence in species P/Ep and associated adaptations.

A comparison of stomatal conductance responses to blue and red light between C3 and C4 photosynthetic species in three phylogenetically-controlled experiments.

C4 photosynthesis is an adaptation that has independently evolved at least 66 times in angiosperms. C4 plants, unlike their C3 ancestral, have a carbon concentrating mechanism which suppresses photorespiration, often resulting in faster photosynthetic rates, higher yields, and enhanced water use efficiency. Moreover, the presence of C4 photosynthesis greatly alters the relation between CO2 assimilation and stomatal conductance. Previous papers have suggested that the adjustment involves a decrease in stomatal density. Here, we tested if C4 species also have differing stomatal responses to environmental cues, to accommodate the modified CO2 assimilation patterns compared to C3 species. To test this hypothesis, stomatal responses to blue and red-light were analysed in three phylogenetically linked pairs of C3 and C4 species from the Cleomaceae (Gynandropsis and Tarenaya), Flaveria, and Alloteropsis, that use either C3 or C4 photosynthesis. The results showed strongly decreased stomatal sensitivity to blue light in C4 dicots, compared to their C3 counterparts, which exhibited significant blue light responses. In contrast, in C3 and C4 subspecies of the monocot A. semialata, the blue light response was observed regardless of photosynthetic type. Further, the quantitative red-light response varied across species, but the presence or absence of a significant stomatal red-light response was not directly associated with differences in photosynthetic pathway. Interestingly, stomatal density and morphology patterns observed across the three comparisons were also not consistent with patterns commonly asserted for C3 and C4 species. The strongly diminished blue-light sensitivity of stomatal responses in C4 species across two of the comparisons suggests a common C4 feature that may have functional implications. Altogether, the strong prevalence of species-specific effects clearly emphasizes the importance of phylogenetic controls in comparisons between C3 and C4 photosynthetic pathways.

Functional response and resistance to drought in seedlings of six shrub species with contrasting leaf traits from the Mediterranean Basin and California.

Extreme drought events during post-fire regeneration are becoming increasingly frequent in Mediterranean-type ecosystems. Understanding how plants with different traits and origins respond to such conditions during early life stages is therefore critical for assessing the effect of climate change. Here, seedlings of three Cistus (semi-deciduous malacophylls from the Mediterranean Basin) and three Ceanothus (evergreen sclerophylls from California) species, two post-fire seeder genera with contrasting leaf traits, were subjected to complete water deprivation for three months in a common garden experiment. The leaf and plant structure, as well as plant tissue water relations were characterized before the drought, and the functional response (water availability, gas exchange, and fluorescence) were monitored during the drought. Both genera exhibited contrasting leaf structure and tissue water relations traits, with higher leaf area, specific leaf area, as well as higher osmotic potential at maximum turgor and turgor loss point in Cistus than Ceanothus. During drought, Ceanothus showed a more conservative use of water than Cistus, with a water potential less sensitive to decreasing soil moisture and a strong decline in photosynthesis and stomatal conductance in response to water deficit, but also a level of fluorescence more responsive to drought than Cistus. However, we could not find a different degree of drought resistance between the genera. This was particularly clear between Cistus ladanifer and Ceanothus pauciflorus, the two most functionally contrasting species, but at the same time the two most drought resistant. Our findings demonstrate that species with different leaf traits and functional responses to water stress may not differ in their degree of drought resistance, at least during the seedling stage. This underlines the need to take general categorizations by genus or functional traits with caution and deepen our knowledge about Mediterranean-type species ecophysiology, especially during early life stages, for anticipating their vulnerability to climate change.

While peach tree is very sensitive to waterlogging, there are no works on the effect of the flooding over only part of its root system. The objective of this work was to investigate physiological and growth responses of Nemared rootstock to partial flooding of roots. Treatments were performed for 6-days: control trees, half-flooded, and fully-flooded. Both flooding treatments negatively affected the vegetative growth, stomatal conductance, chlorophyll content and leaf water potential. The response observed in the half-flooded treatment was intermediate between the control and fully-flooded treatment. The fastest response of stomatal conductance to soil flooding, before differences among treatments in leaf water potential were perceived, suggested that there should be a stomatal closure mechanism that responds to a signal that arises from flooded roots. The importance of this work, lies in showing the negative effects of unnoticed and partial floods, frequently occurring in peach trees.

A role for ethylene signaling and biosynthesis in regulating and accelerating CO2 - and abscisic acid-mediated stomatal movements in Arabidopsis.

Little is known about long-distance mesophyll-driven signals that regulate stomatal conductance. Soluble and/or vapor-phase molecules have been proposed. In this study, the involvement of the gaseous signal ethylene in the modulation of stomatal conductance in Arabidopsis thaliana by CO2 /abscisic acid (ABA) was examined. We present a diffusion model which indicates that gaseous signaling molecule/s with a shorter/direct diffusion pathway to guard cells are more probable for rapid mesophyll-dependent stomatal conductance changes. We, therefore, analyzed different Arabidopsis ethylene-signaling and biosynthesis mutants for their ethylene production and kinetics of stomatal responses to ABA/[CO2 ]-shifts. According to our research, higher [CO2 ] causes Arabidopsis rosettes to produce more ethylene. An ACC-synthase octuple mutant with reduced ethylene biosynthesis exhibits dysfunctional CO2 -induced stomatal movements. Ethylene-insensitive receptor (gain-of-function), etr1-1 and etr2-1, and signaling, ein2-5 and ein2-1, mutants showed intact stomatal responses to [CO2 ]-shifts, whereas loss-of-function ethylene receptor mutants, including etr2-3;ein4-4;ers2-3, etr1-6;etr2-3 and etr1-6, showed markedly accelerated stomatal responses to [CO2 ]-shifts. Further investigation revealed a significantly impaired stomatal closure to ABA in the ACC-synthase octuple mutant and accelerated stomatal responses in the etr1-6;etr2-3, and etr1-6, but not in the etr2-3;ein4-4;ers2-3 mutants. These findings suggest essential functions of ethylene biosynthesis and signaling components in tuning/accelerating stomatal conductance responses to CO2 and ABA.

MicroRNA162 regulates stomatal conductance in response to low night temperature stress via abscisic acid signaling pathway in tomato

MicroRNAs (miRNAs) mediate the degradation of target mRNA and inhibit mRNA translation to regulate gene expression at the transcriptional and post-transcriptional levels in response to environmental stress in plants. We characterized the post-transcriptional mechanism by deep sequencing small RNA (sRNA) to examine how miRNAs were involved in low night temperature (LNT) stress in tomato and whether the molecular mechanism depended on the abscisic acid (ABA) signaling pathway. We annotated conserved miRNAs and novel miRNAs with four sRNA libraries composed of wild-type (WT) tomato plants and ABA-deficient mutant (sit) plants under normal growth and LNT stress conditions. Reverse genetics analysis suggested that miR162 participated in LNT resistance and the ABA-dependent signaling pathway in tomato. miR162-overexpressing (pRI-miR162) and miR162-silenced (pRNAi-miR162) transgenic tomato plants were generated to evaluate miR162 functions in response to LNT stress. miR162 deficiency exhibited high photosynthetic capacity and regulated stomatal opening, suggesting negative regulation of miR162 in the ABA-dependent signaling pathway in response to LNT stress. As feedback regulation, miR162 positively regulated ABA to maintain homeostasis of tomato under diverse abiotic stresses. The mRNA of DICER-LIKE1 (DCL1) was targeted by miR162, and miR162 inhibited DCL1 cleavage in LNT response, including the regulation of miRNA160/164/171a and their targets. The DCL1-deficient mutants (dcl1) with CRISPR/Cas9 prevented stomatal opening to influence photosynthesis in the ABA signaling pathway under LNT stress. Finally, we established the regulatory mechanism of ABA-miR162-DCL1, which systematically mediated cold tolerance in tomato. This study suggests that post-transcriptional modulators acted as systemic signal responders via the stress hormone signaling pathway, and the model at the post-transcriptional level presents a new direction for research in plant abiotic stress resistance.

Interactive effects of elevated temperature and drought on plant carbon metabolism: A meta-analysis.

Elevated temperature (Te ) and drought often co-occur and interactively affect plant carbon (C) metabolism and thus the ecosystem C cycling; however, the magnitude of their interaction is unclear, making the projection of global change impacts challenging. Here, we compiled 107 journal articles in which temperature and water availability were jointly manipulated, and we performed a meta-analysis of interactive effects of Te and drought on leaf photosynthesis (Agrowth ) and respiration (Rgrowth ) at growth temperature, nonstructural carbohydrates and biomass of plants, and their dependencies on experimental and biological moderators (e.g., treatment intensity, plant functional type). Our results showed that, overall, there was no significant interaction of Te and drought on Agrowth . Te accelerated Rgrowth under well-watered conditions rather than under drought conditions. The Te × drought interaction on leaf soluble sugar and starch concentrations were neutral and negative, respectively. The effect of Te and drought on plant biomass displayed a negative interaction, with Te deteriorating the drought impacts. Drought induced an increase in root to shoot ratio at ambient temperature but not at Te . The magnitudes of Te and drought negatively modulated the Te × drought interactions on Agrowth . Root biomass of woody plants was more vulnerable to drought than that of herbaceous plants at ambient temperature, but this difference diminished at Te . Perennial herbs exhibited a stronger amplifying effect of Te on plant biomass in response to drought than did annual herbs. Te exacerbated the responses of Agrowth and stomatal conductance to drought for evergreen broadleaf trees rather than for deciduous broadleaf and evergreen coniferous trees. A negative Te × drought interaction on plant biomass was observed on species-level rather than on community-level. Collectively, our findings provide a mechanistic understanding of the interactive effects of Te and drought on plant C metabolism, which would improve the prediction of climate change impacts.

Acclimatization of Tropical Palm Species Associated with Leaf Morpho-Physiological Traits to the Understorey Environment of Hevea Rubber Farms

Hevea rubber farm is viable for agroforestry systems since its canopy lessens extreme weather conditions and contributes to the adaptation of shade-tolerant plants. However, some limitations in the availability of soil water and shades vary with the age of rubber trees and affect the understorey plants’ acclimatization. Tropical palms are potentially associated plants for the rubber-based agroforestry systems because they are rainforest species adaptable to understorey environments. Two rubber farms, ages 12 and 25 years, intercropped with tropical palms were selected to investigate the acclimatization of the palms to the seasonal abiotic variations in the mature rubber farms. The studied palm species were Chrysalidocarpus lutescens and Rhapis excelsa in the 12-year-old rubber farm and Livistona speciosa and Licuala spinosa in the 25-year-old rubber farm, respectively. Leaf area, stomatal conductance, photosynthesis pigments, and leaf nitrogen content were identified as the palms’ morpho-physiological traits. The 12-year-old rubber farm had a marked soil water deficit in all soil depths at the beginning of the rainy season, reaching around 200 kPa at the 80 cm soil depth, while the 25-year-old rubber farm received greater light transmissions, ranging between 37 and 46% in the late dry season. All palms adjusted leaf area to balance the photosynthetic capacity. The Rhapis palm had greater acclimatization with significant responses of stomatal conductance. Other than the Licuala palm, all palms exhibited the allocation of chlorophyll pigments and nitrogen content significantly in their leaves in response to the different intensities of abiotic stresses in the understorey of the rubber farms.

A simple system for phenotyping of plant transpiration and stomatal conductance response to drought

Plant breeding for increased crop water use efficiency or drought stress resistance requires methods to quickly assess the transpiration rate (E) and stomatal conductance (gs) of a large number of individual plants. Several methods to measure E and gs exist, each of which has its own advantages and shortcomings. To add to this toolbox, we developed a method that uses whole-plant thermal imaging in a controlled environment, where aerial humidity is changed rapidly to induce changes in E that are reflected in changes in leaf temperature. This approach is based on a simplified energy balance equation, without the need for a reference material or complicated calculations. To test this concept, we built a double-sided, perforated, open-top plexiglass chamber that was supplied with air at a high flow rate (35 L min−1) and whose relative humidity (RH) could be switched rapidly. Measurements included air and leaf temperature as well as RH. Using several well-watered and drought stressed genotypes of Arabidopsis thaliana that were exposed to multiple cycles in RH (30–50 % and back), we showed that leaf temperature as measured in our system correlated well with E and gs measured in a commercial gas exchange system. Our results demonstrate that, at least within a given species, the differences in leaf temperature under several RH can be used as a proxy for E and gs. Given that this method is fairly quick, noninvasive and remote, we envision that it could be upscaled for work within rapid plant phenotyping systems.

Optimal stomatal theory predicts CO2 responses of stomatal conductance in both gymnosperm and angiosperm trees.

Optimal stomatal theory predicts that stomata operate to maximise photosynthesis (Anet ) and minimise transpirational water loss to achieve optimal intrinsic water-use efficiency (iWUE). We tested whether this theory can predict stomatal responses to elevated atmospheric CO2 (eCO2 ), and whether it can capture differences in responsiveness among woody plant functional types (PFTs). We conducted a meta-analysis of tree studies of the effect of eCO2 on iWUE and its components Anet and stomatal conductance (gs ). We compared three PFTs, using the unified stomatal optimisation (USO) model to account for confounding effects of leaf-air vapour pressure difference (D). We expected smaller gs , but greater Anet , responses to eCO2 in gymnosperms compared with angiosperm PFTs. We found that iWUE increased in proportion to increasing eCO2 in all PFTs, and that increases in Anet had stronger effects than reductions in gs . The USO model correctly captured stomatal behaviour with eCO2 across most datasets. The chief difference among PFTs was a lower stomatal slope parameter (g1 ) for the gymnosperm, compared with angiosperm, species. Land surface models can use the USO model to describe stomatal behaviour under changing atmospheric CO2 conditions.
