A low-cost, easy-to-use prototype bioreactor model for the investigation of human gut microbiota: validation using a prebiotic treatment

In vitro gut models allow for the study of the impact of molecules on human gut microbiota composition and function without the implication of the host. However, current models, such as the Simulator of Human Intestinal Microbial Ecosystem (SHIME®), are expensive, time-consuming, and require specialized personnel. Homemade in vitro models that lessen these issues have limited evidence of their humanlike functionality. In this study, we present the development of a low-cost and easy-to-use bioreactor with the proven functionality of human microbiota. In our model, we evaluated the capability of replicating human gut microbiota growth and the response of the human bacterial community to a prebiotic, resistant starch, particularly resistant starch type 2 (RS2). Our bioreactor produced an environment that was stable for pH, temperature, and anaerobic conditions. The bioreactor consistently cultivated bacterial communities over a 48 h time period, replicating the composition of the gut microbiota and the associated metabolite production response to RS2, in line with prior human studies. In response to the RS2 prebiotic, we observed an increase in Bifidobacterium adolescentis and Bifidobacterium faecale and an increase in the production of the short-chain fatty acids such as acetate, propionate, and isobutyrate. Taken together, these data demonstrate that our low-cost and user-friendly prototype bioreactor model provides a favorable environment for the growth of human gut microbiota and can mimic its response to a prebiotic.

Effects of soil conditioners on vineyard saline soil physicochemical properties and bacterial community in arid areas

AbstractSoil salinization adversely affects soil quality and ecosystem. Many researches have tried to ameliorate saline soils by soil conditioners. However, little is known about the differences in the responses of soil bacterial communities to natural and artificial conditioners applied to saline soils. Therefore, in this study, the effects of natural humic acid (IK), synthetic polymer (IP), and composite material (IF) (mixture of IK and IP (1:1)) on bacterial community structure and functional genes in saline soil were evaluated to clarify their differences. The results showed that the application of the three soil conditioners significantly reduced soil pH and Na+ content but increased soil alkaline phosphatase, urease, invertase and catalase activities, bacterial diversity, and nutrients, compared to the control (no conditioner). IK application increased bacterial relative abundance (e.g., Subgroup_6, RB41, MND1, and KD4‐96) and metabolic functions (e.g., Two‐component system and Biosynthesis of amino acids) by increasing soil nitrogen and maintaining K+ and Na+ balance. IP application increased soil alkaline phosphatase and urease activities as well as bacterial relative abundance (e.g., Subgroup_6, RB41, MND1, Gemmatimonadaceae, and KD4‐96) and metabolic functions (e.g., Quorum sensing and carbon metabolism) by increasing soil organic carbon/nitrogen content. IF application increased the bacterial relative abundance of Subgroup_6, RB41, and MND1 by increasing soil available nitrogen and regulated their metabolic pathways (e.g., ABC transporters and microbial metabolism). On the whole, IK, IP, and IF could regulate the structure and function of soil bacterial community in saline soils. This study clarifies difference in the effects of different soil conditioners on the amelioration of saline soils from the perspective of soil microbiology, and provides a reference for the amelioration of saline soils in arid areas.

CeO2 nanoparticle dose and exposure modulate soybean development and plant-mediated responses in root-associated bacterial communities

Agricultural soils are increasingly undergoing inadvertent and purposeful exposures to engineered CeO2 nanoparticles (NPs), which can impact crops and root-associated microbial communities. However, interactions between NP concentration and exposure duration on plant-mediated responses of root-associated bacterial communities are not well understood. Soybeans seedlings were grown in soil with uncoated NPs added at concentrations of 0, 1 or 100 mg kg−1. Total soil exposure durations were either 190 days, starting 106 days before planting or 84 days with NP amendments coinciding with planting. We assessed plant development, bacterial diversity, differential abundance and inferred functional changes across rhizosphere, rhizoplane, and root tissue compartments. Plant non-monotonic dose responses were mirrored in bacterial communities. Most notably, effects were magnified in the rhizoplane under low-dose, short-exposures. Enriched metabolic pathways were primarily related to biosynthesis and degradation/utilization/assimilation, rather than responses to metals or oxidative stress. Our results indicate that plant-mediated bacterial responses were greater than direct NP impacts. Also, we identify needs for modeling non-monotonic legume stress responses that account for coinfection with mutualistic and parasitic bacteroids. Our findings provide new insights regarding effects of applications of soil amendments such as biosolids containing NPs or nano-enabled formulations used in cultivation of legumes and other crops.

Control of phosphorus release from sediment by the combined use of Elodea nuttallii, hydrous ferric oxide, zeolite and calcite: Efficiency, mechanism and response of bacterial communities

The effectiveness of Elodea nuttallii combined with a mixture of hydrous ferric oxide, zeolite and calcite (HZC) to reduce the internal loading of phosphorus (P) from sediment and its mechanism were studied, and the effect of Elodea nuttallii combined with HZC on the composition and function of the microbial communities in the surface sediment was also investigated. The results showed that the combination utilization of Elodea nuttallii and HZC can decrease the risk of phosphorus liberation from sediment to the overlying water, and the controlling efficiency of Elodea nuttallii combined with HZC was higher than that of Elodea nuttallii or HZC alone. The passivation of labile P measured by diffusive gradient in thin film device and mobile P played a crucial role in the control of internal P loading from sediment by the combined Elodea nuttallii + HZC treatment. HZC capping had a promoting effect on the growth of Elodea nuttallii. This was beneficial to the absorption of phosphorus from sediment by Elodea nuttallii. The combined application of Elodea nuttallii and HZC not only had a certain negative effect on the diversity of bacteria in the surface sediment, but also changed the microbial compositions of the surface sediment at the levels of phylum and genus. However, the microbial communities in the surface sediment still can perform good ecological function. The above results suggest that the combined application of Elodea nuttallii and HZC has a high potential in the management of internal P loading from sediment.

Competitive relationships due to similar nutrient preferences reshape soil bacterial metacommunities

Paddy soil, as an ecosystem with alternating drained and flooded conditions, microorganisms in it can maintain the stability of the ecosystem by regulating the composition and diversity of its species when disturbed by external biotic or abiotic factors, and the regulatory mechanism in this process is a controversial topic in ecological research. In this study, we investigate the effects of pigeon feces addition on bacterial communities in three textured soils, two conditions (drained and flooded) based on microcosm experiment using high-throughput sequencing techniques. Our results show that pigeon feces addition reduced environmental heterogeneity and community diversity, both under flooded and drained conditions and in all textured soils, thereby decreasing the effectiveness of environmental selection and increasing diffusion limitations among bacterial communities. Bacterial communities are altered by environmental factors including total organic carbon, available nitrogen, total phosphorus, available phosphorus and available potassium, resulting in the formation of new community structures and dominant genera. Bacteria from pigeon feces did not colonize the original soil in large numbers, and the soil bacterial community structure changed, with some species replaced the indigenous ones as new dominant genera. As nutrient diffusion increases the nutrient content of the soil, this does not lead to species extinction; however, nutrient diffusion creates new nutrient preferences of the bacterial community, which causes direct competition between species, and contributes to the extinction and immigration species. Our results suggest that species replacement is an adaptive strategy of soil bacterial community in response to dispersal of pigeon feces, and that bacterial community regulate diversity and abundance of the community by enhancing species extinction and immigration, thereby preventing bacteria in pigeon feces from colonizing paddy soils and maintaining ecosystem stability.

Metagenomics analysis reveals effects of salinity fluctuation on diversity and ecological functions of high and low nucleic acid content bacteria

Salinity is a critical environmental factor in marine ecosystems and has complex and wide-ranging biological effects. However, the effects of changing salinity on diversity and ecological functions of high nucleic acid (HNA) and low nucleic acid (LNA) bacteria are not well understood. In this study, we used 16S rRNA sequencing and metagenomic sequencing analysis to reveal the response of HNA and LNA bacterial communities and their ecological functions to salinity, which was decreased from 26 ‰ to 16 ‰. The results showed that salinity changes had significant effects on the community composition of HNA and LNA bacteria. Among LNA bacteria, 14 classes showed a significant correlation between relative abundance and salinity. Salinity changes can lead to the transfer of some bacteria from HNA bacteria to LNA bacteria. In the network topology relationship, the complexity of the network between HNA and LNA bacterial communities gradually decreased with decreased salinity. The abundance of some carbon and nitrogen cycling genes in HNA and LNA bacteria varied with salinity. Overall, this study demonstrates the effects of salinity on diversity and ecological functions and suggests the importance of salinity in regulating HNA and LNA bacterial communities and functions.

Nonlinear responses of biofilm bacteria to alkyl-chain length of parabens by DFT calculation

Parabens can particularly raise significant concerns regarding the disruption of microbial ecology due to their antimicrobial properties. However, the responses of biofilm bacteria to diverse parabens with different alkyl-chain length remains unclear. Here, theoretical calculations and bioinformatic analysis were performed to decipher the influence of parabens varying alkyl-chain lengths on the biofilm bacteria. Our results showed that the disturbances in bacterial community did not linearly response to the alkyl-chain length of parabens, and propylparaben (PrP), with median chain length, had more severe impact on bacterial community. Despite the fact that paraben lethality linearly increased with chain length, the PrP had a higher chemical reactions potential than parabens with shorter or longer alkyl-chain. The chemical reactions potential was critical in the nonlinear responses of bacterial community to alkyl-chain length of parabens. PrP could impose selective pressure to disturb the bacterial community, because it had a more profound contribution to deterministic assembly process. Furthermore, N-acyl-homoserine lactones was also significantly promoted under PrP exposure, confirming that PrP could affect the bacterial community by influencing the quorum-sensing system. Overall, our study reveals the nonlinear responses of bacterial communities to the alkyl-chain lengths of parabens and provides insightful perspectives for the better regulation of parabens. Environmental ImplicationParabens are recognized as emerging organic pollutants, which specially raise great concerns due to their antimicrobial properties disturbing microbial ecology. However, few study have addressed the relationship between bacterial community responses and the molecular structural features of parabens with different alkyl-chain length. This investigation revealed nonlinear responses of the bacterial community to the alkyl-chain length of parabens through DFT calculation and bioinformatic analysis and identified the critical roles of chemical reactions potential in nonlinear responses of bacterial community. Our results benefit the precise evaluation of ecological hazards posed by parabens and provide useful insights for better regulation of parabens.

Succession of particle-attached and free-living bacterial communities in response to microalgal dynamics induced by the biological cyanocide paucibactin A

Microalgae, including cyanobacteria and eukaryotic algae, are hotspots of primary production and play a critical role in global carbon cycling. However, these species often form blooms that poses a threat to aquatic ecosystems. Although the use of bacteria-derived cyanocides is regarded as an environmentally friendly method for controlling cyanobacterial blooms, only a few studies have examined their potential impact on ecosystems. This study is the first to explore the response of particle-attached (PA) and free-living (FL) bacteria to the dynamics of microalgal communities induced by the biological cyanocide paucibactin A. The microalgal community dynamics were divided into two distinct phases [phase I (days 0–2) and phase II (days 3–7)]. In phase I, paucibactin A caused a sudden decrease in the cyanobacterial concentration. Phase II was characterized by increased growth of eukaryotic microalgae (Scenedesmus, Pediastrum, Selenastrum, and Coelastrum). The stability of the bacterial community and the contribution of stochastic processes to community assembly were more pronounced in phase II than in phase I. The microalgal dynamics triggered by paucibactin A coincided with the succession of the PA and FL bacterial communities. The lysis of cyanobacteria in phase I favored the growth of microbial organic matter degraders in both the PA (e.g., Aeromonas and Rheinheimera) and FL (e.g., Vogesella) bacterial communities. In phase II, Lacibacter, Phycisphaeraceae, and Hydrogenophaga in the PA bacterial community and Lacibacter, Peredibacter, and Prosthecobacter in the FL bacterial community showed increased relative abundances. Overall, the FL bacterial community exhibited greater sensitivity to the two sequential processes compared with the PA bacterial community. These results highlight the need for studies evaluating the impact of biological cyanocides on aquatic ecosystems when used to control natural cyanobacterial blooms.

Roles of Microbial Community and Keystone Taxa in Rice Productivity under Green Manuring in South China

Green manure (GM)–rice–rice rotation is an important management practice for improving soil fertility and rice productivity. The microbiological mechanisms for the increase in grain yield in GM–rice–rice rotation remain unclear. The responses of soil biodiversity, bacterial and fungal communities, and their interactions in the GM–rice–rice rotation were investigated based on two long-term field experiments in Gaoqiao (GQ) and Nanxian (NX) in Hunan Province, south China. Results showed that rice yields were raised by 11.79% and 15.03% under GM in GQ and NX, respectively. GM promoted Shannon diversity and Pielou’s evenness and changed the community structures of bacteria and fungi. The co-occurrence network analysis found that the percentages of negative edges were higher in GM (40.79% and 44.32% in GQ and NX, respectively) than those in the corresponding winter fallow (34.86% and 29.13% in GQ and NX, respectively) in the combined bacterial–fungal networks, suggesting more stable microbial community under GM. Moreover, GM had higher percentages of bacterial–fungal and fungal–fungal edges than winter fallow, indicating that GM increased the interaction between bacteria and fungi and fungi play more essential roles in affecting soil processes under GM. The keystone taxa in GM were positively linked with C metabolism-related enzymes and soil multifunctionality, and were important in improving soil fertility and rice productivity. We concluded that the fungal community was more sensitive to GM application than the bacterial community and that keystone taxa had important influences on soil properties and rice productivity in the GM–double-rice cropping system, which can effectively support the sustainable development of the paddy field ecosystem in southern China.

Soil management shapes bacterial and archaeal communities in soybean rhizosphere: Comparison of no-tillage and integrated crop-livestock systems

Sustainable agricultural systems play a crucial role in improving soil properties and enhancing crop yields. Particularly for soybean, a vital agricultural commodity, no-tillage (NT) and integrated crop-livestock (ICL) systems have been employed in tropical regions. Despite the recognized benefits of using NT and ICL, there is a significant knowledge gap regarding their impact on the rhizosphere microbiome of soybean. Therefore, this field study aimed to explore and compare the responses of the bacterial and archaeal communities within the soybean rhizosphere in both NT and ICL systems. To address this objective, in addition to sampling the soybean rhizosphere, we collected samples from the bulk soil in the NT area and the rhizospheres of grass (Urochloa brizantha) and corn (Zea mays L.) in the ICL system, covering the typical land use in this region. The results revealed distinct bacterial and archaeal communities in the soybean rhizosphere under NT and ICL. Specifically, the ICL system enriched the soybean rhizosphere with KD4_96 (score 3), Vicinamibacteraceae (score 3), Candidatus Nitrocosmicus (score 2.5), and Methylobacterium (score 2.5). In contrast, NT led to an enrichment of Solirubrobacter (score 3), Amycolatopsis (score 2.8), Sphingomonas (score 2.8), and Nitrososphaeraceae (score 2.5). Microbial community interactions exhibited greater complexity in the soybean rhizosphere under NT (676 nodes and 7095 edges). Notably, both bacterial and archaeal communities in the soybean rhizosphere under NT and ICL demonstrated potential functionality in nitrogen fixation. Thus, this study showed that NT and ICL promoted different responses of bacterial and archaeal communities within the soybean rhizosphere which, can influence the plant's performance.

Response of bacterial communities, enzyme activities and dynamic changes of soil organic nitrogen fractions to six-year different application levels of biochar retention in Northeast China

Composition and availability of soil organic nitrogen play a key role in soil N retention and supply. The application of traditional chemical fertilizers can improve soil production and crop yields. However, it also leads to low N utilization efficiency and soil solidification. Previous studies have shown that fertilizers added with biochar can improve soil fertility and facilitate soil nutrient transformation by regulating soil biological indicators. However, whether biochar used with fertilizers can promote nutrient cycling in the long term, which remains to be tested. Therefore, this study aims to explore the long-term effects of biochar with different levels used with fertilizers on soil fertility, N supply potential of organic N, and microbial communities, thus, providing a theoretical reference for the rational management of biochar applications. In this study, a field experiment was carried out from 2013 to 2018 assess dynamic changes in organic N fractions and diversity and richness of microbial communities under the impacts of chemical fertilizers combined with different levels of biochar. Biochar with no amendment (CK), biochar with 15.75 t ha−1 (BC1), biochar with 31.5 t ha−1 (BC2), and biochar with 47.25 t ha−1 (BC3) are four biochar levels that were tested in this study. The results show that compared with CK, those other three biochar levels can increase soil organic matter, C/N, and pH values. Acid-hydrolysable N (AHN) makes a major contribution to organic N in soil. Biochar retention for six years has significantly affected AHN levels, as well as its four fractions, namely, amino acid N (AAN), acid-hydrolysable ammonium N (AN), amino sugar N (ASN), and hydrolysable unknown N (HUN). Some vital biological characteristics, with which each AHN fraction is primarily regulated, can be altered with the application of biochar. That is, AAN is mainly controlled by enriched Verrucomicrobia, Cyanobacteria, Glucosylbacteria, and Nitrospira, and reduced soil urease, while AN is mainly controlled by enriched Verrucomicrobia, Bacteroidetes, Gemmatimonadetes, Acidobacteria, and Proteobacteria. ASN is regulated by enriched Acidobacteria and depleted soil protease, while HUN is regulated by Gemmatimonadetes and enriched soil nitrate reductase. It has been found that microbial and soil enzyme activities are involved in N cycle processes, indicating that the formation of AHN fractions is closely associated with biochar-induced specific microbial and enzyme activities. This study has provided new insights into the influences of biochar with different levels on distribution of soil organic N fractions, and shifts in soil enzyme activities and microbial communities.

Responses of Attached Bacterial Communities to Blooms of the Swimming Shelled Pteropod Creseis acicula in Daya Bay, Southern China.

The shelled pteropod Creseis acicula is a marine pelagic shellfish widely distributed from temperate to tropical seas around the world. From June to July 2020, a C. acicula bloom first happened in the Daya Bay, southern China, and its density reached the highest value (5600 ind. m-3) ever recorded around the world. However, few studies have investigated the responses of bacterial communities to the C. acicula bloom. In the present study, we examined the community profiles of three communities of bacteria including the free-living and particle-attached bacteria in the blooming and reference waters, and bacteria attached to the whole body and shell of C. acicula using a high-throughput sequencing method. The results indicated that the C. acicula bloom had a greater impact on particle-attached bacteria than free-living bacteria. Among the bloom-sensitive particle-attached bacteria, the predominant bacterial phyla were Pseudomonadota, Bacteroidota, and Verrucomicrobiota in the blooming areas, whereas, they were Actinomycetota and Planctomycetota in the reference areas. Specifically, fecal bacteria Haloferula and Halioglobus sp. were significantly enriched in the blooming waters and accumulated on C. acicula shells. Conversely, the significantly lower relative abundance of Nocardioides sp. in the blooming area and accumulated on the whole body of C. acicula indicated their attachment to particles consumed by C. acicula. Overall, our results suggested that the C. acicula bloom influenced marine bacteria, particularly particle-attached bacteria by increasing (e.g. providing shells and feces) or decreasing (e.g. filter-feeding the suspended particles) the abundance of available substances.

Response of bacterial and micro-eukaryotic communities to spatio-temporal fluctuations of wastewater in full scale constructed wetlands

How microbial communities respond to wastewater fluctuations is poorly understood. Full-scale surface flow constructed wetlands (SFCWs) were constructed for investigating microbial communities. Results showed that influent wastewater changed sediment bacterial community composition seasonally, indicating that a single bacterial taxonomic group had low resistance (especially, Actinobacteriota and Gammaproteobacteria). However, copy numbers of 16S rRNA, ammonia oxidizing archaea, ammonia oxidizing bacteria, nirS and nirK in the first stage SFCWs were 2.49 × 1010, 3.48 × 109, 5.76 × 106, 8.77 × 108 and 9.06 × 108 g−1 dry sediment, respectively, which remained stable between seasons. Moreover, decreases in the nitrogen concentration in wastewater, changed microbial system state from heterotrophic to autotrophic. Micro-eukaryotic communities were more sensitive to wastewater fluctuations than bacterial communities. Overall, results revealed that microbial communities responded to spatio-temporal fluctuations in wastewater through state changes and species asynchrony. This highlighted complex processes of wastewater treatment by microbial components in SFCWs.

BackgroundTicks serve as vectors for a diverse array of pathogens, including viruses responsible for both human and livestock diseases. Symbiotic bacteria hold significant potential for controlling tick-borne disease. However, the alteration of tick gut bacterial community in response to pathogen infection has not been analyzed for any tick-borne viruses. Here, the impact of severe fever with thrombocytopenia syndrome virus (SFTSV) infection on bacterial diversity in the gut of Haemaphysalis longicornis is investigated.MethodsUnfed tick females were artificially infected with SFTSV. The gut samples were collected and the genomic DNA was extracted. We then investigated alterations in gut bacterial composition in response to SFTSV infection through 16S rRNA gene sequencing.ResultsThe study found that a reduction in the number of operational taxonomic units (OTUs) in the tick gut following SFTSV infection. However, there were no significant changes in alpha diversity indices upon infection. Four genera, including Corynebacterium, Arthrobacter, Sphingomonas, and Escherichia, were identified as biomarkers for the tick gut without SFTSV infection. Notably, the predicted correlation network indicated that the biomarkers Sphingomonas and Escherichia exhibited positive correlations within the same subcommunity, which was altered upon viral infection.ConclusionsThese findings revealed that the change in tick gut bacterial composition upon SFTSV infection and could facilitate the discovery new target for tick-borne viral disease control.Graphical

Bacterial community shifts occur primarily through rhizosphere expansion in response to subsoil amendments.

To comprehensively evaluate the impact of agricultural management practices on soil productivity, it is imperative to conduct a thorough analysis of soil bacterial ecology. Deep-banding nutrient-rich amendments is a soil management practice that aims to improve plant growth and soil structure by addressing the plant-growth constraints posed by dense-clay subsoils. However, the response of bacterial communities to deep-banded amendments has not been thoroughly studied. To address this knowledge gap, we conducted a controlled-environment column experiment to examine the effects of different types of soil amendments (poultry litter, wheat straw + chemical fertiliser and chemical fertiliser alone) on bacterial taxonomic composition in simulated dense-clay subsoils. We evaluated the bacterial taxonomic and ecological group composition in soils beside and below the amendment using 16S rRNA amplicon sequencing and robust statistical methods. Our results indicate that deep-banded amendments alter bacterial communities through direct and indirect mechanisms. All amendments directly facilitated a shift in bacterial communities in the absence of growing wheat. However, a combination of amendments with growing wheat led to a more pronounced bacterial community shift which was distinct from and eclipsed the direct impact of the amendments and plants alone. This indirect mechanism was evidenced to be mediated primarily by plant growth and hypothesised to result from an enhancement in wheat root distribution, density and rhizodeposition changes. Therefore, we propose that subsoil amendments regardless of type facilitated an expansion in the rhizosphere which engineered a substantial plant-mediated bacterial community response within the simulated dense-clay subsoils. Overall, our findings highlight the importance of considering the complex and synergistic interactions between soil physicochemical properties, plant growth and bacterial communities when assessing agricultural management strategies for improving soil and plant productivity.

Evaluation of Soil Total Nitrogen as an Indicator of Soil Bacterial Community Response to Biochar and Plant Growth-Promoting Rhizobacteria Applications

Biochar and plant growth-promoting rhizobacteria (PGPR) are widely used as an amendment for soil physicochemical properties and soil bacterial community diversity. In Guangxi, China, we carried out a study to determine how PGPR and biochar influence the soil’s environmental stability in an Eucalypt plantation. We applied biochar and PGPR in a contrasting application manner to an acidic red loam soil. Thus, three treatments were set up as 5 × 1010 CFU·mL−1 PGPR-only (MB0), 20 t·hm−2 biochar-only (B20), and co-application of 20 t·hm−2 biochar and 5 × 1010 CFU·mL−1 PGPR (MB20), as well as no biochar and no PGPR (M0B0). Our results indicated that MB20 significantly decreased the soil total nitrogen (TN) and increased the soil total phosphorus (Soil TP), soil ammonium nitrogen (NH4+), and soil water content (SWC) when compared with the control. The MB20 also significantly increased the Simpson, ACE, and Chao indices of the soil bacterial community’s diversity relative to the control. We also observed a significant effect of the Soil TN on both the bacterial community and the functional diversity in soil. These findings may indicate that assessing the soil N status is expected to be an essential indicator of the soil microenvironment’s response to biochar and PGPR applications.

Effects of Reduced Seawater pH and Oil Contamination on Bacterial Communities and Biochemical Markers of Estuarine Animal Hosts

Ecosystem functioning depends on complex interactions between microorganisms, hosts, and the environment. Changes in environmental conditions (e.g., ocean acidification) in combination with anthropogenic pollution have been shown to affect the composition and function of free-living microbial communities, but little is known about the effects these stressors on host-associated communities. This study aims to characterize the response of host-associated bacterial communities of the bottom-dwelling polychaete Hediste diversicolor and the epibenthic gastropod Peringia ulvae to oil contamination and reduced seawater pH. The independent and interactive effects of both stressors were simulated under controlled conditions. The response of host-associated bacterial communities was assessed using the high-throughput sequencing of the 16S rRNA gene and several biochemical markers related to host metabolic pathways, e.g., neurotransmission, anaerobic metabolism, biotransformation, oxidative stress, and energy consumption. In H. diversicolor, reduced seawater pH was associated with a high relative abundance of Cyanobacteria, while in P. ulvae oil contamination was associated with a reduction in the relative abundance of Chitinophagales. In P. ulvae, enrichment with oil hydrocarbon-degrading bacteria suggests a possible role of these organisms in the dispersion of oil hydrocarbon degraders. Furthermore, oil supplementation shifted some specific biochemical markers of gastropods related to oxidative stress and energy consumption, which suggests host stress. In general, the bacterial communities and biochemical markers of the gastropod were more affected by stressors than those of the polychaete. Overall, this study contributes to a better understanding of the response of host-associated bacterial communities of benthic macrofauna to anthropogenic contamination and environmental change.
