Strategies for managing fungicide resistance in the Brazilian tropical agroecosystem: Safeguarding food safety, health, and the environmental quality

AbstractFungicide resistance is an alarming challenge for the Brazilian tropical agricultural systems, with major implications for food safety, human and animal health, as well as for the environment. This review explores strategies to address fungicide resistance within the Brazilian agroecosystem context. We examined historical and current scenarios of fungicide resistance in the Brazilian agroecosystems and the approaches to delay the emergence and mitigate the selection of resistant variants. Our review indicates that the prevalence of resistance in field populations of key plant pathogens in Brazil was due to failures in the implementation of preventive measures. To address this issue, alternative evolutionary-smart strategies against fungicide resistance are proposed, emphasizing institutional actions and public policies. Crucial steps involve strengthening national networks for large-scale foliar and seed fungicide efficacy testing and resistance monitoring, as well as imposing tighter restrictions on the labeling of high-risk single-active formulations. Additionally, the integration of non-chemical disease management strategies and the establishment of a centralized database and information system on fungicide resistance in Brazil are identified as essential for effective resistance monitoring and informed decision-making. To enhance fungicide resistance management, the adoption of a warning system (e.g., based on aerobiology- or on weather-monitoring) for predicting disease epidemics and minimizing fungicide applications is recommended. Increased funding, collaboration, mandatory reporting, and capacity building are required to overcome these challenges. In addition, promoting integrated disease management approaches is vital. By implementing these tailored strategies, Brazil can actively contribute to safeguarding its food safety, protecting human and animal health, and preserving the delicate balance of its unique agroecosystem. The adoption of evolutionary-smart strategies against fungicide resistance will prolong fungicide efficacy, reduce economic costs, and minimize environmental impacts, ensuring sustainable and resilient agriculture in Brazil.

First Report of Benzimidazole Resistance in Field Population of Haemonchus contortus from Sheep, Goats and Cattle in Bosnia and Herzegovina.

Haemonchus contortus is a globally significant parasitic nematode in ruminants, with widespread resistance to benzimidazole due to its excessive and prolonged use. Given the extensive use of benzimidazole anthelmintics in Bosnia and Herzegovina, we hypothesized that resistance is prevalent. The aim of this study was to identify the presence of anthelmintic resistance to benzimidazole in H. contortus from naturally infected sheep, goats and cattle in Bosnia and Herzegovina through the detection of the Phe/Tyr polymorphism in the amino acid at position 200 of the β-tubulin protein. From 19 locations in Bosnia and Herzegovina, a total of 83 adult H. contortus were collected from the abomasum of ruminants. Among these, 45 H. contortus specimens were isolated from sheep, 19 from goats and 19 from cattle. Results showed that 77.8% of H. contortus in sheep exhibited homozygous resistant genotypes at position 200 of the β-tubulin gene, with 15.5% being heterozygous. In goats, all tested H. contortus (100%) were homozygous resistant, and no heterozygous resistant or homozygous sensitive genotypes were found. Cattle had 94.7% homozygous resistant H. contortus, with no heterozygous resistant genotypes detected. In H. contortus from sheep and cattle, 6.7% and 5.3%, respectively, displayed homozygous sensitive genotypes. This study, for the first time, highlights the presence of a resistant population of H. contortus in sheep, goats and cattle in Bosnia and Herzegovina, using the rt-qPCR method. The resistance likely spread from sheep or goats to cattle, facilitated by shared pastures and the practice of transhumance, indicating a widespread and growing issue of anthelmintic resistance.

Knock-in mutagenesis in Drosophila Rdl underscores the critical role of the conserved M3 glycine in mediating the actions of broflanilide and isocycloseram on GABA receptors

γ-Aminobutyric acid receptors (GABARs) are crucial targets for pest control chemicals, including meta-diamide and isoxazoline insecticides, which act as negative allosteric modulators of insect GABARs. Previous cell-based assays have indicated that amino acid residues in the transmembrane cavity between adjacent subunits of Drosophila RDL GABAR (i.e., Ile276, Leu280, and Gly335) are involved in mediating the action of meta-diamides. In this study, to confirm this result at the organismal level, we employed CRISPR/Cas9-mediated genome editing, generated six transgenic Drosophila strains carrying substitutions in these amino acid residues, and investigated their sensitivity to broflanilide and isocycloseram. Flies homozygous for the I276F mutation did not exhibit any change in sensitivity to the tested insecticides compared to the control flies. Conversely, I276C homozygosity was lethal, and heterozygous flies exhibited ∼2-fold lower sensitivity to broflanilide than the control flies. Flies homozygous for the L280C mutation survived into adulthood but exhibited infertility. Both heterozygous and homozygous L280C flies exhibited ∼3- and ∼20-fold lower sensitivities to broflanilide and isocycloseram, respectively, than the control flies. The reduction in sensitivity to isocycloseram in L280C flies diminished to ∼3-fold when treated with piperonyl butoxide. Flies homozygous for the G335A mutation reached the adult stage. However, they were sterile, had small bodies, and exhibited reduced locomotion, indicating the critical role of Gly335 in RDL function. These flies exhibited markedly increased tolerance to topically applied broflanilide and isocycloseram, demonstrating that the conserved Gly335 is the target of the insecticidal actions of broflanilide and isocycloseram. Considering the significant fitness costs, the Gly335 mutation may not pose a serious risk for the development of resistance in field populations of insect pests. However, more careful studies using insect pests are needed to investigate whether our perspective applies to resistance development under field conditions.

Voltage-gated sodium channel gene mutation and P450 gene expression are associated with the resistance of Aphis spiraecola Patch (Hemiptera: Aphididae) to lambda-cyhalothrin.

Aphis spiraecola Patch is one of the most economically important tree fruit pests worldwide. The pyrethroid insecticide lambda-cyhalothrin is commonly used to control A. spiraecola. In this 2-year study, we quantified the resistance level of A. spiraecola to lambda-cyhalothrin in different regions of the Shaanxi province, China. The results showed that A. spiraecola had reached extremely high resistance levels with a 174-fold resistance ratio (RR) found in the Xunyi region. In addition, we compared the enzymatic activity and expression level of P450 genes among eight A. spiraecola populations. The P450 activity of A. spiraecola was significantly increased in five regions (Xunyi, Liquan, Fengxiang, Luochuan, and Xinping) compared to susceptible strain (SS). The expression levels of CYP6CY7, CYP6CY14, CYP6CY22, P4504C1-like, P4506a13, CYP4CZ1, CYP380C47, and CYP4CJ2 genes were significantly increased under lambda-cyhalothrin treatment and in the resistant field populations. A L1014F mutation in the sodium channel gene was found and the mutation rate was positively correlated with the LC50 of lambda-cyhalothrin. In conclusion, the levels of lambda-cyhalothrin resistance of A. spiraecola field populations were associated with P450s and L1014F mutations. Our combined findings provide evidence on the resistance mechanism of A. spiraecola to lambda-cyhalothrin and give a theoretical basis for rational and effective control of this pest species.

The evolving story of sulfoxaflor resistance in the green peach aphid, Myzus persicae (Sulzer).

The green peach aphid, Myzus persicae (Sulzer), is one of the most economically important crop pests worldwide. Insecticide resistance in this pest was first detected over 60 years ago, with resistance in M. persicae now spanning over 80 active ingredients. Sulfoxaflor is a relatively new insecticide that is primarily used to control sap-feeding insects. In 2018 resistance to sulfoxaflor was discovered in field populations of M. persicae in Australia. This study aimed to determine the current distribution and phenotypic levels of sulfoxaflor resistance in Australian clones of M. persicae and to investigate how these patterns relate to clonal type. For the first time, we show there is low-level resistance (8-26-fold) distributed across Australia, with resistance being detected in aphids collected from approximately 20% of all M. persicae collected and screened. Furthermore, this study shows sulfoxaflor resistance is found in twoM. persicae haplotypes, providing evidence that there have been multiple independent evolutionary events which have given rise to sulfoxaflor resistance in this species. These findings have important implications for the chemical control of M. persicae in Australia, especially when considering the broader genetic background of these aphids which are known to harbour a number of other insecticide resistance mechanisms. We recommend continuous monitoring of sulfoxaflor resistance in field populations of M. persicae (in Australia and elsewhere) and further research into the underlying genetic mechanisms conferring resistance to sulfoxaflor in both clonal haplotypes. © 2023 The Authors. Establishment and application of bioassay- and molecular marker-based methods for monitoring fluvalinate resistance of Varroa mites

The Varroa mite, Varroa destructor, is an ectoparasite that infests honey bees. The extensive use of acaricides, including fluvalinate, has led to the emergence of resistance in Varroa mite populations worldwide. This study's objective is to monitor fluvalinate resistance in field populations of Varroa mites in Korea through both bioassay-based and molecular marker-based methods. To achieve this, a residual contact vial (RCV) bioassay was established for on-site resistance monitoring. A diagnostic dose of 200 ppm was determined based on the bioassay using a putative susceptible population. In the RCV bioassay, early mortality evaluation was effective for accurately discriminating mites with the knockdown resistance (kdr) genotype, while late evaluation was useful for distinguishing mites with additional resistance factors. The RCV bioassay of 14 field mite populations collected in 2021 indicated potential resistance development in four populations. As an alternative approach, quantitative sequencing was employed to assess the frequency of the L925I/M mutation in the voltage-gated sodium channel (VGSC), associated with fluvalinate kdr trait. While the mutation was absent in 2020 Varroa mite populations, it emerged in 2021, increased in frequency in 2022, and became nearly widespread across the country by 2023. This recent emergence and rapid spread of fluvalinate resistance within a span of three years demonstrate the Varroa mite's significant potential for developing resistance. This situation further underscores the urgent need to replace fluvalinate with alternative acaricides. A few novel VGSC mutations potentially involved in resistance were identified. Potential factors driving the rapid expansion of resistance were further discussed.

Transcriptional regulation and overexpression of GST cluster enhances pesticide resistance in the cotton bollworm, Helicoverpa armigera (Lepidoptera: Noctuidae)

The rapid evolution of resistance in agricultural pest poses a serious threat to global food security. However, the mechanisms of resistance through metabolic regulation are largely unknown. Here, we found that a GST gene cluster was strongly selected in North China (NTC) population, and it was significantly genetically-linked to lambda-cyhalothrin resistance. Knockout of the GST cluster using CRISPR/Cas9 significantly increased the sensitivity of the knockout strain to lambda-cyhalothrin. Haplotype analysis revealed no non-synonymous mutations or structural variations in the GST cluster, whereas GST_119 and GST_121 were significantly overexpressed in the NTC population. Silencing of GST_119 or co-silencing of GST_119 and GST_121 with RNAi significantly increased larval sensitivity to lambda-cyhalothrin. We also identified additional GATAe transcription factor binding sites in the promoter of NTC_GST_119. Transient expression of GATAe in Hi5 cells activated NTC_GST_119 and Xinjiang (XJ)_GST_119 transcription, but the transcriptional activity of NTC_GST_119 was significantly higher than that of XJ_GST_119. These results demonstrate that variations in the regulatory region result in complex expression changes in the GST cluster, which enhances lambda-cyhalothrin resistance in field-populations. This study deepens our knowledge of the evolutionary mechanism of pest adaptation under environmental stress and provides potential targets for monitoring pest resistance and integrated management.

Evaluation of An Aerial Application of Duet Hd® Against Aedes Dorsalis and Culex Tarsalis in Rural Habitats of the Great Salt Lake, Utah.

The Salt Lake City Mosquito Abatement District (SLCMAD) has been conducting aerial applications using an organophosphate insecticide against adult mosquitoes for several decades. In order to evaluate a potential rotation product, aerial applications of Duet HD™, a pyrethroid, were conducted under operational conditions against wild populations of Aedes dorsalis and Culex tarsalis and against colony strains of Cx. pipiens and Cx. quinquefasciatus. The erratic wind patterns of the greater Salt Lake area did not prevent sufficient droplet deposition flux at 9 monitoring locations spread across a 5,120-acre (2,072 ha) spray block within rural habitats. Three separate aerial application trials showed great efficacy against Ae. dorsalis. In contrast, Cx. tarsalis exhibited inconsistent treatment-associated mortalities, suggesting the presence of less susceptible or resistant field populations as a result of spillover from agricultural or residential pyrethroid usage. Bottle bioassays to diagnose pyrethroid resistance using field-collected Cx. tarsalis indicated that some populations of this species, especially those closest to urban edges, failed to show adequate mortality in resistance assays. Despite challenging weather conditions, Duet HD worked reasonably well against susceptible mosquito species, and it may provide a crucial role as an alternative for organophosphate applications within specific and sensitive areas. However, its area-wide adoption into control applications by the SLCMAD could be problematic due to reduced impacts on the most important arboviral vector species, Cx. tarsalis, in this area. This study demonstrates the importance of testing mosquito control products under different operational environments and against potentially resistant mosquito populations by municipal mosquito control districts.

Exaptation of I4760M mutation in ryanodine receptor of Spodoptera exigua (Lepidoptera: Noctuidae): Lessons from museum and field samples

Since 2007, diamide insecticides have been widely used in Korea to control various types of lepidopteran pests including Spodoptera exigua. For nearly a decade, diamide resistance in field populations of S. exigua across 18 localities has been monitored using bioassays. Despite their short history of use, resistance to diamide insecticides has emerged. Based on the LC50 values, some field populations showed a higher level of resistance to chlorantraniliprole, a diamide insecticide, compared to that of the susceptible strain, although regional and temporal variations were observed. To investigate resistance at a molecular level, we examined three mutations (Y4701C, I4790M, and G4946E) in the ryanodine receptor (RyR), which is the primary mechanism underlying diamide insecticide resistance. DNA sequencing showed that only the I4790M mutation was found in most field populations. As resistance levels varied significantly despite the uniform presence of the I4790M mutation, we considered the presence of another resistance factor. Further, the I4790M mutation was also found in S. exigua specimens collected prior to the commercialization of diamide insecticides in Korea as well as in other countries, such as the USA. This finding led us to hypothesize that the I4790M mutation were predisposed in field populations owing to selection factors other than diamide use. For further clarification, we conducted whole-genome sequencing of S. exigua (449.83 Mb) and re-sequencing of 18 individual whole genomes. However, no additional non-synonymous mutations were detected in the RyR-coding region. Therefore, we concluded that the high level of diamide insecticide resistance in Korean S. exigua is not caused by mutations at the target site, RyR, but is attributed to other factors that need to be investigated in future studies.

Occurrence of resistance in field populations of Erysiphe necator to QoI, azanaphthalene and aryl-phenyl-ketone fungicides in Serbia

The objective of this study was to investigate whether the sensitivity of E. necator populations to several important fungicide groups has changed in Serbia. Field trials, bioassays, and molecular tests were conducted from 2019 to 2022. Samples were collected from commercial, experimental and extensive vineyards. Low efficacy of QoIs, azanaphthalenes and aryl-phenyl-ketones was observed in the field trials. A total of 110 isolates were analyzed by qPCR for the presence of the G143A mutation. The mutation was positively detected in 61.8% of the isolates, therefore isolates containing >95% of the G143A allele were QoI-resistant in bioassays. Contrary, QoI-sensitive isolates contained <5% of the G143A allele. Of the 54 isolates tested in bioassays, 46 grew significantly at the recommended concentration, whereas 36 were able to grow at a fourfold higher concentration of quinoxyfen. Proquinazid showed more significant inhibition than quinoxyfen, suggesting that there is no cross-resistance between these two members of the azanaphthalene group. Unsatisfactory effect of metrafenone and pyriofenone at recommended concentrations was observed in 29 of 41 isolates. These isolates grew readily at four times higher concentrations of metrafenone and pyriofenone, indicating the cross-resistance between them. Overall, the results of this study suggest that resistance to QoIs has developed and is widespread in commercial vineyards, requiring growers to exclude them completely from powdery mildew management. The performance of proquinazid in the field was not affected, making it suitable for powdery mildew control. Special attention and extensive monitoring should be directed to metrafenone and pyriofenone, as cross-resistance has been found at certain sites where they have been used intensively over the past decade.

Molecular profiling of resistance alleles in Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae) collected from different locations

BackgroundAfter the commercialization of insect-resistant transgenic Bt cotton Bollgard I & II, India ranks first in the world in cotton production. Cotton insecticide consumption was drastically reduced as nearly 95% of the cotton area was replaced with Bollgard II. However, the benefits of transgenic cotton appear to have been diminished as the pink bollworm, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae) developed field resistance to Cry1Ac expressing Bt cotton in India in 2008. In 2015, there was an unusual survival of P. gossypiella on Bollgard II cotton in some parts of Gujarat and Maharashtra, which is a cause for concern.ResultsIn the present study, PCR analysis and gel visualization of BGII resistant field population of P. gossypiella showed that the presence of r1, r2, r3, r1s, r2s, r3s, and ss mutated cadherin alleles, which produced amplicon sizes of 750 bp, 700 bp, 730 bp, 12,700 bp, 750 bp, 980 bp, 600 bp and 1600 bp, respectively, when seven different types of cadherin allele(s) specific primers were used. The r1 mutant allele was detected in Amaravati, Wardha, Yavatmal, and Nagpur, but not in Akola and Buldhana, using primers r1disfor and Int-540. The r2 mutant allele was detected in Akola, Wardha, and Nagpur, but not in Buldhana, Amaravati, and Yavatmal, using primers r2disback and Cad2366. The r3 mutant allele was detected in all locations using primers r3disback and Cad3221. Amplicons of sizes 750 bp, 700 bp and 730 bp were obtained for r1, r2, and r3 mutant alleles, respectively. The sizes of the amplicons were 1270 bp for r1s, 750 bp for r2s, and 980 bp for r3s.The absence of bands for r1, r2, and r3 cadherin alleles in individuals indicated the possibility of having the ss allele, which was confirmed using primers Cad3324 and Int-651. The presence of ss mutant allele was observed in field-collected P. gossypiella populations from BG II cotton in various locations, with a 600 bp and 1600 bp size amplicons produced using the same primers.ConclusionsPCR analysis revealed the presence of r1, r2, r3, r1s, r2s, r3s, and ss mutated cadherin alleles in field-collected populations of Vidarbha which provide evidence to field-evolved resistance of P. gossypiella to BGII cotton.

Development of Insecticide Resistance in Field Populations of Onion Thrips, Thrips tabaci (Thysanoptera: Thripidae).

The present study evaluated insecticide resistance in field populations of onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae), collected from eight different onion-growing regions of Punjab, Pakistan. These field-collected populations were assessed for resistance development against eight commonly used active ingredients including deltamethrin, lambda-cyhalothrin, imidacloprid, acetamiprid, spinosad, spinetoram, cypermethrin, and abamectin. In leaf dip bioassays, T. tabaci adults showed varied levels of resistance towards different insecticides. Moderate or high levels of resistance to deltamethrin (58-86 fold), lambda-cyhalothrin (20-63 fold), and cypermethrin (22-54 fold) were observed in T. tabaci field populations. There were very low to moderate resistance levels to imidacloprid (10-38 fold), acetamiprid (5-29 fold), and abamectin (10-30 fold). The lowest levels of resistance were detected in thrips exposed to spinosad (3-13 fold) and spinetoram (3-8 fold). Insecticide resistance levels varied among populations collected from various geographic locations, but all populations exhibited elevated levels of resistance to deltamethrin. Thrips tabaci populations with higher resistance levels were most commonly found from the southern part of Punjab, Pakistan. Our findings revealed that spinosyns could be used as alternatives to conventional insecticides for the successful management of T. tabaci in onion fields.

Assays to Measure Insecticide Toxicity and Insecticide Resistance in Mosquitoes.

Mosquitoes' resistance to commonly used insecticides is now widespread, hampering control efforts and leading to substantial increases in human illness and mortality rates in many areas of the world. Insecticide bioassays are quantitative methodologies used to determine the dose-response relationship of insects to insecticides and to evaluate the susceptibility or resistance of mosquitoes to specific insecticides. They are frequently used to monitor the development of insecticide resistance in mosquitoes for both field resistance diagnoses (surveillance assays), in which the ability of mosquitoes to survive exposure to a standard dose or concentration of an insecticide is measured, and laboratory bioassays, in which responses to insecticides are tested in parallel populations of resistant (field) populations and laboratory susceptible strains using serial doses or concentrations. Metabolic detoxification, in which insecticides are metabolized by enzymes, including cytochrome P450s, hydrolases, and glutathione-S-transferases (GSTs), to become more polar and less toxic, is one resistance mechanism. Piperonyl butoxide (PBO), S,S,S-tributyl phosphorotrithioate (DEF), and diethyl maleate (DEM) are the inhibitors of P450s, hydrolases, and GSTs, respectively, and act as synergists for rapidly testing the involvement of these enzymes in insecticide resistance. Such synergistic assays are used to identify the detoxification enzyme that leads to resistance to a specific insecticide. This introduction and its associated protocols present a detailed discussion of appropriate methodologies and procedures for laboratory larval, adult, and synergistic bioassays and introduces the field surveillance tests used to monitor insecticide resistance as recommended by the latest World Health Organization (WHO) and U.S. Centers for Disease Control (CDC) guidelines.

Mosquito Larval Bioassays.

Insecticide bioassays are frequently used to measure levels of insecticide resistance in mosquito populations, examining the ability of mosquitoes to survive exposure to insecticides. Laboratory bioassays measure insects' responses to insecticides in resistant (field) populations and laboratory susceptible strains using serial doses or concentrations across the range of >0 and <100% mortality. This protocol measures the toxicity of insecticides to mosquito larvae and determines the level of insecticide resistance. Generally, laboratory-reared mosquito larvae of known age or instar are exposed to H2O containing various concentrations of an insecticide, and the response (mortality) is recorded 24 h after the test. Larval bioassay tests can (1) identify the lethal concentrations of larvicide that cause 50% and 90% mortality (LC50 and LC90, respectively); (2) determine the diagnostic concentration needed to monitor susceptibility in mosquito larvae in the field; and (3) investigate the resistance status and the mechanisms governing insecticide resistance to a specific insecticide.

CRISPR/Cas9-mediated knockout of NlCYP6CS1 gene reveals its role in detoxification of insecticides in Nilaparvata lugens (Hemiptera: Delphacidae).

The brown planthopper (Nilaparvata lugens) is one of the major rice insect pests in Asia. Recently, high levels of insecticide resistance have been frequently reported and cytochrome P450 monooxygenase (P450)-mediated metabolic detoxification is a common resistance mechanism in N. lugens. However, there has been no persuasive genetic method to prove the role of P450s in insecticide resistance in N. lugens. Here, CRISPR/Cas9 system was used to disrupt the P450 gene NlCYP6CS1 to elucidate its role in insecticide resistance in field populations of N. lugens. We successfully constructed a homozygous strain (Nl6CS1-KO) with a 5-bp deletion and 1-bp insertion mutation of NlCYP6CS1. Compared with a background resistant strain (Nl-R), the susceptibility of knockout strain Nl6CS1-KO to imidacloprid, nitenpyram, thiamethoxam, dinotefuran, and pymetrozine was increased by 2.3-, 3.4-, 7.0-, 4.2- and 3.9-fold, respectively, but not significantly changed to triflumezopyrim, chlorpyrifos and buprofezin. Life table analysis demonstrated that the Nl6CS1-KO strain resembled the Nl-R strain in terms of egg and nymph developmental duration and adult lifespan, but differed from the Nl-R strain in the survival rate of eggs and nymphs, reproduction, and body weight. Our study demonstrates the effect of functional deletion of NlCYP6CS1 on multiple insecticide resistance in N. lugens. For the first time, we applied CRISPR/Cas9 system to reveal the mechanism of insecticide resistance in N. lugens, which may shed light on similar studies in other hemipteran insects. © 2023 Society of Chemical Industry.

Differential Activities of Antioxidant Enzymes, Superoxide Dismutase, Peroxidase, and Catalase vis-à-vis Phosphine Resistance in Field Populations of Lesser Grain Borer (Rhyzopertha dominica) from India.

Susceptibility to phosphine was compared in 15 populations of lesser grain borer (Rhyzopertha dominica) collected from grain storage godowns across India. A high level of resistance to phosphine was noticed in R. dominica collected from northern India compared to those collected from northeastern regions of India. The median lethal concentration values varied from 0.024 mg/L to 1.991 mg/L, with 1.63 to 82.96-fold resistance compared to laboratory susceptible checks. Antioxidant enzymes have been reported to negate the reactive oxygen species generated upon encountering the fumigant phosphine. Distinct differences in the activity of antioxidant enzymes were noticed in the field populations exposed to phosphine. Peroxidase activity varied between 1.28 and 336.8 nmol H2O2 reduced/min/mg protein. The superoxide dismutase inhibition rate was between 81.29 and 99.66%, and catalase activity varied between 6.28 and 320.13 nmol H2O2 reduced/min/mg protein. The findings of our investigation show that the activities of peroxidase and superoxide dismutase are positively linked (p < 0.01) with an increase in resistance ratios, whereas catalase was found to have a negative association with resistance to phosphine. The reported results elucidate the differential activities of principal antioxidant enzymes in scavenging the oxyradicals (O2•-, H2O2,•OH) associated with tolerance to phosphine in R. dominica.

Prevalence of Carbendazin Resistance in Field Populations of the Rice False Smut Pathogen Ustilaginoidea virens from Jiangsu, China, Molecular Mechanisms, and Fitness Stability.

Rice false smut (RFS), caused by Ustilaginoidea virens, is an important fungal disease of rice. In China, Methyl Benzimidazole Carbamates (MBCs), including carbendazim, are common fungicides used to control RFS and other rice diseases. In this study, resistance of U. virens to carbendazim was monitored for three consecutive years during 2018 to 2020. A total of 321 U. virens isolates collected from Jiangsu Province of China were tested for their sensitivity to carbendazim on PSA. The concentration at which mycelial growth is inhibited by 50% (EC50) of the carbendazim-sensitive isolates was 0.11 to 1.38 µg/mL, with a mean EC50 value of 0.66 μg/mL. High level of resistance to carbendazim was detected in 14 out of 321 isolates. The resistance was stable but associated with a fitness penalty. There was a statistically significant and moderate negative correlation (r= -0.74, p &lt; 0.001) in sensitivity between carbendazim and diethofencarb. Analysis of the U. virens genome revealed two potential MBC targets, Uvβ1Tub and Uvβ2Tub, that putatively encode β-tubulin gene. The two β-tubulin genes in U. virens share 78% amino acid sequence identity, but their function in MBC sensitivity has been unclear. Both genes were identified and sequenced from U. virens sensitive and resistant isolates. It is known that mutations in the β2-tubulin gene have been shown to confer resistance to carbendazim in other fungi. However, no mutation was found in the Uvβ2Tub gene in either resistant or sensitive isolates. Variations including point mutations, non-sense mutations, codon mutations, and frameshift mutations were found in the Uvβ1Tub gene from the 14 carbendazim-resistant isolates, which have not been reported in other fungi before. Thus, these results indicated that variations of Uvβ1Tub result in the resistance to carbendazim in field isolates of Ustilaginoidea virens.

Status of the Resistance of Aphis gossypii Glover, 1877 (Hemiptera: Aphididae) to Afidopyropen Originating from Microbial Secondary Metabolites in China.

The resistance of cotton aphids to various forms of commonly used pesticides has seriously threatened the safety of the cotton production. Afidopyropen is a derivative of microbial metabolites with pyropene insecticide, which has been shown to be effective in the management of Aphis gossypii. Several field populations of Aphis gossypii were collected from the major cotton-producing regions of China from 2019 to 2021. The resistance of these populations to afidopyropen was estimated using the leaf-dipping method. The LC50 values of these field populations ranged from 0.005 to 0.591 mg a.i. L-1 in 2019, from 0.174 to 4.963 mg a.i. L-1 in 2020 and from 0.517 to 14.16 mg a.i. L-1 in 2021. The resistance ratios for all A. gossypii populations ranged from 0.03 to 3.97 in 2019, from 1.17 to 33.3 in 2020 and from 3.47 to 95.06 in 2021. The afidopyropen resistance exhibited an increasing trend in the field populations of Cangzhou, Binzhou, Yuncheng, Kuerle, Kuitun, Changji and Shawan from 2019 to 2021. This suggests that the resistance development of the cotton aphid to afidopyropen is inevitable. Therefore, it is necessary to rotate or mix afidopyropen with other insecticides in order to inhibit the development of afidopyropen resistance in field populations.

Impact of Imidacloprid Resistance on the Demographic Traits and Expressions of Associated Genes in Aphis gossypii Glover.

Imidacloprid is one of the most widely used neonicotinoid insecticides to control sap-sucking insect pests, including Aphis gossypii. The intensive application of chemical insecticides to A. gossypii led to the development of resistance against several insecticides, including imidacloprid. Therefore, it is crucial to understand the association between imidacloprid resistance and the fitness of A. gossypii to limit the spread of the resistant population under field contexts. In this study, we used the age-stage, two-sex life table method to comprehensively investigate the fitness of imidacloprid resistant (ImR) and susceptible strains (SS) of melon aphids. Results showed that ImR aphids have prolonged developmental stages and decreased longevity, fecundity, and reproductive days. The key demographic parameters (r, λ, and R0) were significantly reduced in ImR strain compared to SS aphids. Additionally, the molecular mechanism for fitness costs was investigated by comparing the expression profile of juvenile hormone-binding protein (JHBP), juvenile hormone epoxide hydrolase (JHEH), juvenile hormone acid O-methyltransferase (JHAMT), Vitellogenin (Vg), ecdysone receptor (EcR), and ultraspiracle protein (USP) supposed to be associated with development and reproduction in insects. The results of RT-qPCR showed that EcR, JHBP, JHAMT, JHEH, and Vg genes were downregulated, while USP was statistically the same in ImR A. gossypii compared to the SS strain. Together, these results provide in-depth information about the occurrence and magnitude of fitness costs against imidacloprid resistance that could help manage the evolution and spread of A. gossypii resistance in field populations.

Lack of fitness cost and inheritance of resistance to abamectin based on the establishment of a near-isogenic strain of Tetranychus urticae

Many populations of the two-spotted spider mite, Tetranychus urticae Koch, have developed high levels of resistance to the pesticide abamectin in China and other countries. This study developed a near-isogenic line to understand better the inheritance, cross-resistance, and fitness costs associated with abamectin resistance in the field population of T. urticae in China. We introduced the trait that confers extremely high abamectin resistance in a field-collected population of T. urticae into a susceptible laboratory strain (IPP-SS) to generate an abamectin-resistant near-isogenic line (NIL-Aba). This process was carried out through multiple backcrossing to IPP-SS and via parthenogenesis and abamectin screening. Compared with IPP-SS, the NIL-Aba strain had a 25 147-fold resistance to abamectin and a high level of cross-resistance to bifenthrin (288.17-fold), an intermediate level to emamectin benzoate (42.57-fold), and low levels to bifenazate, chlorfenapyr, cyflumetofen, cyenopyrafen, and cyetpyrafen with resistance ranging from 3.18- to 9.31-fold. But it had no cross-resistance to profenofos. The resistance to abamectin in NIL-Aba was autosomal, incompletely dominant, and polygenic. Based on two sex life table parameters, no fitness cost was found in NIL-Aba. Establishing the NIL-Aba strain provides a reliable basis for an in-depth study of abamectin resistance in T. urticae. New information on toxicological characteristics and fitness cost should facilitate the management of abamectin resistance in field populations of T. urticae.
