Impact of Benzo (a) Pyrene on Thyroid Glands and their Associated Hormones of Male Mice Mus musculus

Benzo [a] pyrene is the main representative of polycyclic aromatic hydrocarbons, and has been repeatedly found in the air, surface water, soil, and sediments. Human exposure to Benzo [a] pyrene is therefore common. Benzo (a) pyrane (BaP) which is mainly presented in newspaper ink and inhibits the function of the thyroid and other endocrine glands of the human system, due to several disorders that occur in the human body. Healthy mature male mice Mus musculus having weight of 125 ± 5 gm were used for the experiments. The effects of a Benzo (a) pyrene (0.65mg/25g body weight/twice in a week) during experimental periods of 60 and 90 days in male mice Mus musculus showed significant difference in body weights compared to control. We assessed the hormonal estimation of thyroid treated with Benzo (a) pyrene by using an appropriate ELISA assay kit and histopathological changes were observed under a light microscope (magnifications 100X) in the thyroid tissue of male mice Mus musculus exposed to BaP after 60 and 90 days. During the study, we observed that in treated mice thyroid hormone concertation was affected the structure of the thyroid follicles was disturbed and epithelial cells were necrotic compared with the control mice which indicates that the BaP may lead to changes in the thyroid morphology as well as secreted hormone concentration in exposed treated male mice.

Formation of nitrated naphthalene in the sulfate radical oxidation process in the presence of nitrite

Polycyclic aromatic hydrocarbons (PAHs) have become a global environmental concern due to their potential hazardous implication for human health. In this study, we found that sulfate radical (SO4•−) could effectively degrade naphthalene (NAP), a representative PAH in groundwaters, generating 1-naphthol. This intermediate underwent further degradation, yielding ring-opening products including phthalic acid and salicylic acid. However, the presence of nitrite (NO2−), a prevalent ion in subsurface environments, was observed to compete with NAP for SO4•−, thus slowing down the NAP degradation. The reaction between NO2− and SO4•− generated a nitrogen dioxide radical (NO2•). Concurrently, in-situ formed 1-naphthol underwent further oxidization to the 1-naphthoxyl radical by SO4•−. The coupling of 1-naphthoxyl radicals with NO2• gave rise to a series of nitrated NAP, namely 2-nitro-1-naphthol, 4-nitro-1-naphthol, and 2,4-dinitro-1-naphthol. In addition, the in-situ formed phthalic acid and salicylic acid also underwent nitration, generating nitrophenolic products, although this pathway appeared less prominent than the nitration of 1-naphthol. When 10 μΜ NAP was subjected to heat activated peroxydisulfate oxidation in the presence of 10 μΜ NO2−, the total yield of nitrated products reached 0.730 μΜ in 120 min. Overall, the presence of NO2− dramatically altered the behavior of NAP degradation by SO4•− oxidation and contributed to the formation of toxic nitrated products. These findings raise awareness of the potential environmental risks associated with the application of SO4•−-based oxidation processes for the remediation of PAHs-polluted sites in presence of NO2−.

The unique biodegradation pathway of benzo[a]pyrene in moderately halophilic Pontibacillus chungwhensis HN14

Benzo[a]pyrene (BaP), as the typical representative of polycyclic aromatic hydrocarbons (PAHs), is a serious hazard to human health and natural environments. Though the study of microbial degradation of PAHs has persisted for decades, the degradation pathway of BaP is still unclear. Previously, Pontibacillus chungwhensis HN14 was isolated from high salinity environment exhibiting a high BaP degradation ability. Here, based on the intermediates identified, BaP was found to be transformed to 4,5-epoxide-BaP, BaP-trans-4,5-dihydrodiol, 1,2-dihydroxy-phenanthrene, 2-carboxy-1-naphthol, and 4,5-dimethoxybenzo[a]pyrene by the strain HN14. Furthermore, functional genes involved in degradation of BaP were identified using genome and transcriptome data. Heterogeneous co-expression of monooxygenase CYP102(HN14) and epoxide hydrolase EH(HN14) suggested that CYP102(HN14) could transform BaP to 4,5-epoxide-BaP, which was further transformed to BaP-trans-4,5-dihydrodiol by EH(HN14). Moreover, gene cyp102(HN14) knockout was performed using CRISPR/Cas9 gene-editing system which confirmed that CYP102(HN14) play a key role in the initial conversion of BaP. Finally, a novel BaP degradation pathway was constructed in bacteria, which showed BaP could be converted into chrysene, phenanthrene, naphthalene pathways for the first time. These findings enhanced our understanding of microbial degradation process for BaP and suggested the potential of using P. chungwhensis HN14 for bioremediation in PAH-contaminated environments.

Embryonic exposures to cadmium and PAHs cause long-term and interacting neurobehavioral effects in zebrafish

Developmental exposure to either polycyclic aromatic hydrocarbons (PAHs) or heavy metals has been shown to cause persisting and overlapping neurobehavioral effects in animal models. However, interactions between these compounds have not been well characterized, despite their co-occurrence in a variety of environmental media. In two companion studies, we examined the effects of developmental exposure to cadmium (Cd) with or without co-exposure to prototypic PAHs benzo[a]pyrene (BaP, Exp. 1) or fluoranthene (FA, Exp. 2) using a developing zebrafish model. Zebrafish embryos were exposed to Cd (0–0.3 μM), BaP (0–3 μM), FA (0–1.0 μM), or binary Cd-PAH mixtures from 5 to 122 h post fertilization (hpf). In Exp. 1, Cd and BaP produced independent effects on an array of outcomes and interacting effects on specific outcomes. Notably, Cd-induced deficits in dark-induced locomotor stimulation were attenuated by BaP co-exposure in the larval motility test and BaP-induced hyperactivity was attenuated by Cd co-exposure in the adolescent novel tank test. Likewise, in Exp. 2, Cd and FA produced both independent and interacting effects. FA-induced increases on adult post-tap activity in the tap startle test were attenuated by co-exposure with Cd. On the predator avoidance test, FA- and 0.3 μM Cd-induced hyperactivity effects were attenuated by their co-exposure. Taken together, these data indicate that while the effects of Cd and these representative PAHs on zebrafish behavior were largely independent of one another, binary mixtures can produce sub-additive effects for some neurobehavioral outcomes and at certain ages. This research emphasizes the need for detailed risk assessments of mixtures containing contaminants of differing classes, and for clarity on the mechanisms which allow cross-class toxicant interactions to occur.

A new strategy of using periphyton to simultaneously promote remediation of PAHs-contaminated soil and production of safer crops

Contaminated farmland leads to serious problems for human health through biomagnification in the soil-crop-human chain. In this paper, we have established a new soil remediation strategy using periphyton for the production of safer rice. Four representative polycyclic aromatic hydrocarbons (PAHs), including phenanthrene (Phe), pyrene (Pyr), benzo[b]fluoranthene (BbF), and benzo[a]pyrene (BaP), were chosen to generate artificially contaminated soil. Pot experiments demonstrated that in comparison with rice cultivation in polluted soil with ΣPAHs (50 mg kg−1) but without periphyton, adding periphyton decreased ΣPAHs contents in both rice roots and shoots by 98.98% and 99.76%, respectively, and soil ΣPAHs removal reached 94.19%. Subsequently, risk assessment of ΣPAHs based on toxic equivalent concentration (TEQ), pollution load index (PLI), hazard index (HI), toxic unit for PAHs mixture (TUm), and incremental lifetime cancer risk (ILCR) indicated that periphyton lowered the ecological and carcinogenicity risks of PAHs. Besides, the role of periphyton in enhancing the rice productivity was revealed. The results indicated that periphyton alleviated the oxidative stress of PAHs on rice by reducing malondialdehyde (MDA) content and increasing total antioxidant capacity (T-AOC). Periphyton reduced the toxic stress of PAHs on the soil by promoting soil carbon cycling and metabolic activities as well. Periphyton also improved the soil's physicochemical properties, such as the percentage of soil aggregate, the contents of humic substances (HSs) and nutrients, which increased rice biomass. These findings confirmed that periphyton could improve rice productivity by enhancing soil quality and health. This study provides a new eco-friendly strategy for soil remediation and simultaneously enables the production of safe crops on contaminated land.

An ultra-stable and sensitive Ag-HP-β-CD/GO/NCF SERS substrate combine with the coffee-ring effect strategy for trace PAHs detection

This study proposes an ultra-stable and highly sensitive SERS substrate based on Ag nanoparticles/graphene oxide nanosheets hybrids decorated nanocellulose fiber (Ag/GO/NCF), which was further functionalized with hydroxypropyl-β-cyclodextrin (Ag-HP-β-CD/GO/NCF) for the trace detection of polycyclic aromatic hydrocarbons (PAHs). The Ag/GO hybrid structure enables the substrate to exhibit high SERS activity through the synergistic effect of electromagnetic and chemical enhancement. Simultaneously, the three-dimensional skeleton structure of NCF serves as a support matrix, ensuring the exceptional stability of the substrate. Furthermore, this study introduces a SERS measurement strategy utilizing the coffee-ring effect and explores parameters optimization to further enhance the detection sensitivity while maintaining the homogeneity and reproducibility. With the affinity effect of HP-β-CD and the enrichment effect of the delocalized π-electron system in GO, the hydrophobic PAHs could easily approach the SERS hotspots and achieve their trace detection. The limit of detection (LOD) for four representative PAHs, including pyrene, fluoranthene, phenanthrene and fluorene, are found to be 3.32 ug/L, 3.65 ug/L, 40.97 ug/L and 56.24 ug/L, respectively. These results highlight the potential application of this highly efficient SERS platform in environmental monitoring.

Study on reproductive endocrine disturbance and DNA damage mechanism of female Ruditapes philippinarum under Benzo[a]pyrene stress

The reproductive toxicity of polycyclic aromatic hydrocarbons (PAHs) in aquatic organisms has attracted increasing attention from scholars. Currently, research in this field primarily focuses on vertebrates such as zebrafish and other model species. However, there is still a significant knowledge gap in the toxicity of PAHs to invertebrates and its potential mechanisms. Benzo[a]pyrene (B[a]P) is one of the most representative PAHs. In this study, female Ruditapes philippinarum (R. philippinarum) was treated with B[a]P concentrations of 0, 0.8, 4, and 20 μg/L to investigate reproductive indicators in the proliferative, growth, mature, and spawn stages. Transcriptomics was used to investigate the expression of genes associated with the reproductive endocrine system, DNA repair, autophagy, apoptosis, and ovarian development at different reproductive stages. Our results suggested that B[a]P disrupted the endocrine system by interfering with the production of steroid hormones and the transmission of estrogen signals in female R. philippinarum. The structure of the ovarian DNA duplex is severely damaged under the stress of B[a]P, and a series of cellular responses caused by DNA damage are also interfered. Additionally, we observed a reduction in the gonadosomatic index (GSI) and mature oocytes numbers after B[a]P exposed. Tissue section indicated that severe damage to the ovarian structure at mature and spawn stages. In conclusion, this study combined transcriptomic and toxicological to explore the negative effects on ovarian development induced by B[a]P, focusing on reproductive endocrine disturbance and DNA damage.

Marine pollutant Phenanthrene (PHE) exposure causes immunosuppression of hemocytes in crustacean species, Scylla paramamosain

Phenanthrene (PHE), a representative polycyclic aromatic hydrocarbons (PAHs), is widely found in aquatic environments, which exhibits high toxicity to aquatic organisms and has a substantial impact on overall health. In order to investigate the immunosuppressive effects of PHE exposure on marine crustacean species, the Scylla paramamosain was exposure to different concentrations of PHE, which was 0 μg/L (control group), 0.7 μg/L, 7 μg/L, or 70 μg/L PHE with 35 individuals in every group, respectively. The results showed that the color of hemocytes gradually deepened with increasing PHE concentration, and the total hemocyte count (THC) was activated and increased after PHE exposure analyzed by Flow cytometry. Meanwhile, compared with the control group, cryostat sections of hepatopancreas showed cell infiltration, cell steatosis, eosinophilic masses and vacuolization in PHE groups. The superoxide dismutase (SOD) activity was decreased immensely in PHE exposure groups, meanwhile, the acid phosphatase (ACP) activity and glutathione (GSH) activities were increased after PHE exposure compared with control group. Moreover, the expression profile of Crustin, TLR, MCM7, JAK, caspase, Moyosin and P53 were up-regulated significantly after 7th day PHE exposure in all treatment groups by Q-PCR. Those data illustrated that PHE exposure could inhibit the immune function of mud crab by causing hepatopancreas damage, induce the activity of antioxidant enzymes and the expression of immune genes. These data provide a scientific basis for evaluating the impact of PAH pollution on marine organisms.

AHR-mediated DNA damage contributes to BaP-induced cardiac malformations in zebrafish

Benzo[a]pyrene (BaP) is a representative polycyclic aromatic hydrocarbon widely present in the environment. We previously reported that the aryl hydrocarbon receptor (AHR) mediates BaP-induced apoptosis and cardiac malformations in zebrafish embryos, but the underlying molecular mechanisms were unclear. Since BaP is a mutagenetic compound, we hypothesize that BaP induces apoptosis and heart defects via AHR-mediated DNA damage. In this study, zebrafish embryos were exposed to BaP at a concentration of 0.1 μM from 2 to 72 h post fertilization, either with or without inhibitors/agonists. AHR activity and levels of reactive oxygen species (ROS) were examined under a fluorescence microscope. mRNA expression levels were quantified by qPCR. DNA damage and apoptosis were detected by immunofluorescence. Our findings revealed that BaP exposure significantly increased BPDE-DNA adducts, mitochondrial damage, apoptosis and heart defects in zebrafish embryos. These effects were counteracted by inhibiting AHR/cyp1a1 using pharmaceutical inhibitors or genetic knockdown. Furthermore, we observed that spironolactone, an antagonist of nucleotide excision repair (NER), significantly enhanced BaP-induced BPDE-DNA adducts, mitochondrial damage, apoptosis and heart malformation rates. Conversely, SRT1720, a SIRT1 agonist, reduced the adverse effects of BaP. Supplementation with spironolactone also enhanced γ-H2AX signals in the heart of zebrafish embryos exposed to BaP. Additional experiments demonstrated that BaP suppressed the expression of SIRT1. We further established that AHR, when activated by BaP, directly inhibited SIRT1 transcription, leading to downregulation of XPC and XPA, which are essential NER genes involved in the recognition and verification steps of the NER process. Taken together, our results indicate that AHR mediates BaP-induced DNA damage in the heart of zebrafish embryos by inducing BPDE-DNA adduct formation via the AHR/Cyp1a1 signalling pathway, as well as suppressing NER via AHR-mediated inhibition of SIRT1.

One-step electrodeposition of poly(o-phenylenediamine)-Zn composite on plaswood propeller as an extraction device for polycyclic aromatic hydrocarbons in coffee.

Coffee beans can be contaminated during roasting by polycyclic aromatic hydrocarbons (PAHs), some of which have been classified as carcinogens. An extraction device for PAHs in coffee drinks was designed with six compact DC motors rotating six sorbents. The sorbents were plaswood propellers modified by one-step electrodeposition of a poly(ortho-phenylenediamine) and Zn composite (PoPD-Zn). Benzo(a)anthracene (BaA), chrysene (Chry), benzo(b)fluoranthene (BbF), and benzo(a)pyrene (BaP) were chosen as representative PAHs. Scanning electron micrographs of the PoPD-Zn showed a porous structure. The extracted PAHs were quantified by gas chromatography coupled with a flame ionization detector. Detected concentrations of PAHs in coffee drink samples were as follows: BaA 1.4±0.4 to 16.5±0.8µg L-1; Chry 0.5±0.2 to 2.1±0.5µg L-1; BbF 2.2±0.6µg L-1; and BaP 6.2±1.0µg L-1. Good recoveries ranging from 82.7±1.9% to 99.0±0.5% were obtained.

Micro-polystyrene plastic and benzo[α]pyrene exposure affects the endocrine system and causes physiological stress in Carassius auratus

Microplastics, owing to their hydrophobic properties and the various chemicals used in their production, can act as carriers of persistent organic pollutants, such as polycyclic aromatic hydrocarbons (PAHs). In this study, we exposed the goldfish Carassius auratus to benzo[α]pyrene (BaP, 10 μg/L), a representative PAH, and micro-polystyrene plastic (MP; 10 and 100 beads/L), of size 1.0 μm, as a single or complex environmental stressor, and evaluated the stress response and the resulting DNA damage. The expression of CRH and ACTH mRNA in the pituitary gland and hypothalamus, of the hypothalamus–pituitary–interrenal (HPI) axis, increased significantly after 6 h of exposure. Plasma cortisol levels showed a similar trend to the expression of stress-regulating genes along the HPI axis, and a significant increase was observed in the combined exposure groups (BaP + LMP [low-concentration MP] and BaP + HMP [high-concentration MP]) compared to those in the single exposure group. H2O2 concentration and CYP1A1 and MT mRNA expression levels in the liver were significantly higher in the combined exposure groups compared with in the single exposure groups. In situ hybridization revealed a similar pattern of MT mRNA expression, and many signals were observed in the BaP + HMP group. Furthermore, the BaP + HMP group showed more DNA damage, and the degree of DNA damage increased with exposure time for all experimental groups, except for the control group. Therefore, exposure to BaP and MP alone can induce stress in goldfish; however, when a combination of both substances is provided, their synergistic effect leads to increased stress and DNA damage. MP was confirmed to be a more serious stress-inducing factor in goldfish than BaP, based on the expression levels of stress-regulating genes along the HPI axis.

Aerobic biodegradation of phenanthrene by a newly isolated Klebsiella sp. DS-1 from wastewater

Representative polycyclic aromatic hydrocarbons (PAHs) compounds such as phenanthrene (PHE) derived from multiple anthropogenic activities are contributing to a growing number of concerns about its threats to the global water environment and human health. Microbial degradation is considered to be an increasingly promising approach for PHE removal, while effective PHE biodegraders from in situ environments are yet to be identified and their underlying degradation mechanisms still remain to be understood. Herein, a Klebsiella sp. strain DS-1 was isolated from petrochemical wastewater using PHE as the sole carbon source. The degradation efficiency of PHE was up to 88.4 % at 30 °C and pH 7 within a maximum of 9 days. DS-1 grew actively in wide ranges of environmental factors, such as pH values (5–9), temperature (20 °C–37 °C), and initial concentrations of PHE (20–200 mg/L), with the maintenance of PHE degradability. Genome analysis revealed 14 genes that were involved in PHE biodegradation by DS-1. Detection of metabolic intermediates revealed that strain DS-1 degraded PHE via the phthalic acid pathway. These findings have expanded the knowledge of Klebsiella sp. as an active PHE degrader in wastewater and indicate its potential in the bioremediation of PHE-contaminated aquatic environments.

Mechanism of growth inhibition mediated by disorder of chlorophyll metabolism in rice (Oryza sativa) under the stress of three polycyclic aromatic hydrocarbons

Photosynthesis mediated by chlorophyll metabolism is the basis for plant growth, and also the important regulatory mechanism of carbon pool in cropland ecosystems. Soil organic pollutants induced growth inhibition in crop plants, herein, we conducted an in-depth investigation on the effects of three representative polycyclic aromatic hydrocarbons (PAHs), including phenanthrene (PHE), pyrene (PYR), and benzo[a]pyrene (BaP) on rice (Oryza sativa) growth and photosynthesis. PAHs were absorbed via root uptake and accumulated in leaves, causing the swelling of thylakoids and the increase of osmiophilic granules in chloroplasts. The actual quantum efficiency of PSII was significantly decreased under the stress of PHE, PYR, and BaP by 29.9%, 11.9%, and 24.1% respectively, indicating the inhibition in photon absorption and transfer, which was consistent with the decrease of chlorophyll a (22.3%–32.2% compared to the control) in rice leaves. Twenty-two encoding genes involved in chlorophyll metabolism were determined and the results indicated that the expression of chlorophyll synthetases was downregulated by over 50% whereas the degradation process was promoted. Consequently, the production of carbohydrates and the carbon fixation were inhibited, which revealed by the downregulation of intermediate metabolites in Calvin cycle and the declined carboxylation rate. The disturbed photosynthesis resulted in the decrease of the biomasses of both roots (21.0%–42.7%) and leaves (6.4%–22.1%) under the tested PAH stresses. The findings of this study implied that the photosynthetic inhibition was possibly attributed to the disorder of chlorophyll metabolism, thus providing novel insights into the mechanism of growth inhibition induced by organic pollutants and theoretical basis for the estimation of cropland carbon sequestration potential.

Genotoxic carcinogen 7,12-dimethylbenz[a]anthracene inhibits gap junction intercellular communication through post-transcriptional and post-translational processing involved in connexin 43 stability.

Gap junctional intercellular communication (GJIC) is composed of connexin (Cx) and plays an important role in maintaining intracellular homeostasis. Loss of GJIC is involved in the early stages of cancer pathways of non-genotoxic carcinogens; however, the effect of genotoxic carcinogens, including polycyclic aromatic hydrocarbons (PAHs), on GJIC function remains unclear. Therefore, we determined whether and how a representative PAH 7,12-dimethylbenz[a]anthracene (DMBA) suppresses GJIC in WB-F344cells. First, DMBA significantly inhibited GJIC and dose-dependently reduced Cx43 protein and mRNA expression. In contrast, Cx43 promoter activity was upregulated after DMBA treatment via the induction of specificity protein 1 and hepatocyte nuclear factor 3β, indicating that the promoter-independent loss of Cx43 mRNA can be associated with the inhibition of mRNA stability, which was verified by actinomycin D assay. In addition to a decrease in mRNA stability involved in human antigen R, we also observed DMBA-induced acceleration of Cx43 protein degradation, which was closely related to the loss of GJIC through Cx43 phosphorylation via MAPK activation. In conclusion, the genotoxic carcinogen DMBA suppresses GJIC by inhibiting post-transcriptional and post-translational processing of Cx43. Our findings suggest that the GJIC assay is an efficient short-term screening test for predicting the carcinogenic potential of genotoxic carcinogens.

Development of Phase and Seasonally Dependent Land-Use Regression Models to Predict Atmospheric PAH Levels.

Polycyclic aromatic hydrocarbons (PAHs) are an important class of pollutants in China. The land use regression (LUR) model has been used to predict the selected PAH concentrations and screen the key influencing factors. However, most previous studies have focused on particle-associated PAHs, and research on gaseous PAHs was limited. This study measured representative PAHs in both gaseous phases and particle-associated during the windy, non-heating and heating seasons from 25 sampling sites in different areas of Taiyuan City. We established separate prediction models of 15 PAHs. Acenaphthene (Ace), Fluorene (Flo), and benzo [g,h,i] perylene (BghiP) were selected to analyze the relationship between PAH concentration and influencing factors. The stability and accuracy of the LUR models were quantitatively evaluated using leave-one-out cross-validation. We found that Ace and Flo models show good performance in the gaseous phase (Ace: adj. R2 = 0.14-0.82; Flo: adj. R2 = 0.21-0.85), and the model performance of BghiP is better in the particle phase (adj. R2 = 0.20-0.42). Additionally, better model performance was observed in the heating season (adj R2 = 0.68-0.83) than in the non-heating (adj R2 = 0.23-0.76) and windy seasons (adj R2 = 0.37-0.59). Those gaseous PAHs were highly affected by traffic emissions, elevation, and latitude, whereas BghiP was affected by point sources. This study reveals the strong seasonal and phase dependence of PAH concentrations. Building separate LUR models in different phases and seasons improves the prediction accuracy of PAHs.

Benzo(a)pyrene exposure in early life suppresses spermatogenesis in adult male zebrafish and association with the methylation of germ cell-specific genes

Polycyclic aromatic hydrocarbons (PAHs) are environmental contaminants that are widely present in aquatic ecosystems. To assess the impact of early-life exposure to benzo[a]pyrene (BaP), a representative PAH, on reproductive ability in adult male zebrafish (Danio rerio), fertilized embryos were exposed to 0.05, 0.5, 5 and 50 nM of BaP for 96 h, and then the hatched larvae were raised to adulthood in clean water. In one-year-old male fish, the percentage of spermatozoa in testis was significantly reduced in the 0.5, 5 and 50 nM treatments. When the treated fish were mated with untreated fish, significantly decreased rate of egg fertilization and hatching success and significantly elevated malformation rate the F1 larvae were observed in the 0.5, 5 and 50 nM treatments. The transcriptional levels of genes along the brain-pituitary-gonadal axis, involving gnrh3, gnrhr3, fshβ, lhβ, lhγ, lhrγ and ar, were downregulated. In addition, embryonic BaP exposure upregulated the promotor methylation of germ cell-specific genes in the testis of adult fish. The upregulated methylation of ddx4, dnd1, nanos2 in the testis might be associated with the downregulated mRNA levels of these genes, which could be another reason for the inhibition of spermatogenesis. These results indicate that early-life exposure to BaP suppress the reproductive capability of adult male fish, which would cause a decrease in fish population.

Elevated nitrate promoted photodegradation of PAHs in aqueous phase: Implications for the increased nutrient discharge.

Polycyclic aromatic hydrocarbons (PAHs) are frequently released in aqueous phase by oil spill or from other sources, and photochemical oxidation is one of their major weathering processes. In this study, the photochemical behavior of phenanthrene (PHE, as a representative PAH) were studied and the effects of nitrogenous compounds were evaluated. The results showed that nitrate was an effective photosensitizer for improving the photodegradation of PHE, but the promoting effect was less effective in seawater due to the presence of halogen ions; the ammonia played a negligible role on PHE degradation. The photochemical ionization was a key process for PHE degradation, it can be retarded due to the quenching of triplet excited state by dissolved oxygen, and the inhibition was most prominent in fresh water. The presence of nitrate increased the steady state concentration of •OH from 2.08×10-15 M to 1.04×10-14 M in fresh water, and from 1.5×10-16 M to 2.08×10-15 M in seawater. The secondary-order reaction rate constant between PHE and •OH (k•OH,PHE) was determined as 5.70 × 109 M-1 s-1. Similar trend was observed for 1O2. The contribution of •OH to PHE removal was more prominent in fresh water than in seawater due to the quenching effects of halogen, and the increasing of nitrate enlarged the contribution of •OH. Two possible PHE degradation pathways were proposed based on GC-MS analysis and DFT calculation. The Quantitative Structure-activity Relationship (QSAR) evaluation showed that some degradation intermediates were more toxic than PHE, but the total environmental risk was still diminished due to the low percentage of toxic intermediates. This study provided theoretical and experimental insights into the influence of nitrogenous compounds on the photodegradation of PHAs in water environment.

Enhanced benzofluoranthrene removal in constructed wetlands with iron- modified biochar: Mediated by dissolved organic matter and microbial response.

Polycyclic aromatic hydrocarbons (PAHs) pose a high risk to ecosystems owing to their adverse environmental effects. The use of biochar in constructed wetlands (CWs) to remove PAH has received increased interest, but is frequently challenging because of saturation adsorption. To enhance the microbial degradation, electron acceptors are provided. This study aimed to remove a representative PAH, benzofluoranthrene (BbF), using iron-modified biochar as a supplement to the CW substrate. Results revealed that iron-mediated biochar based CWs increased the removal of BbF by 20.4 % and ammonium by 25.6 %. The BbF retained in substrate with biochar (36.6 % higher content) and further removed with iron modification (40.6 % lower content). Iron-modified biochar increased dissolved organic carbon content, particularly low-aromaticity, and low-molecular-weight organic matters (25.7 % higher tryptophan-like material), which contributed to PAH degradation by microorganisms. Microbial analysis confirmed that iron-mediated biochar enriched the abundance of microbes (e.g., Cellulomonas, Actinotalea, and Sphingomonas) and key enzymes (e.g., catA, lipV, and sdhA) that are involved in PAH degradation. Higher proportion of iron-reducing bacteria (e. g., Thiobacillus, Rhodobacter) played a significant role in driving microbial iron cycle, which was beneficial for PAHs removal. Based on the results, we confirmed that the use of iron-modified biochar in CWs enhance PAH removal.

BPDE, the Migration and Invasion of Human Trophoblast Cells, and Occurrence of Miscarriage in Humans: Roles of a Novel lncRNA-HZ09.

Recurrent miscarriage (RM) affects 1%-3% of pregnancies. However, in almost 50% of cases, the cause is unknown. Increasing evidence have shown that benzo(a)pyrene [B(a)P], a representative of polycyclic aromatic hydrocarbons (PAHs), is correlated with miscarriage. However, the underlying mechanisms of B(a)P/benzo(a)pyrene-7,8-dihydrodiol-9,10-epoxide (BPDE)-induced trophoblast cell dysfunctions and miscarriage remain largely unknown. The objective was to discover the role(s) of a novel lncRNA, lnc-HZ09, in the regulation of BPDE-inhibited migration and invasion of trophoblast cells and the occurrence of miscarriage. Human trophoblast cells were treated with 0, 0.25, 0.5, BPDE with or without corresponding lnc-HZ09 silencing or overexpression. Using these cells, we evaluated cell migration and invasion, the mRNA and protein levels of members of the PLD1/RAC1/CDC42 pathway, the regulatory roles of lnc-HZ09 in PLD1 transcription and mRNA stability, and lnc-HZ09 transcription and stability. Human villous tissues were collected from RM () group and their matched healthy control (HC, ) group. We evaluated the levels of BPDE-DNA adducts, lnc-HZ09, and the mRNA and protein expression of members of the PLD1/RAC1/CDC42 pathway, and correlated their relative expression levels. We further constructed 0, 0.05 or B(a)P-induced mouse miscarriage model (each ), in which the mRNA and protein expression of members of the Pld1/Rac1/Cdc42 pathway were measured. We identified a novel lnc-HZ09. Human trophoblast cells treated with lnc-HZ09 exhibited less cell migration and invasion. In addition, the levels of this lncRNA were higher in villous tissues from women with recurrent miscarriage than those from healthy individuals. SP1-mediated PLD1 mRNA levels were lower, and HuR-mediated PLD1 mRNA stability was less in trophoblast cells overexpressing lnc-HZ09. However, trophoblast cells treated with MSX1 had higher levels of lnc-HZ09, and METTL3-mediated m6A methylation on lnc-HZ09 resulted in greater lnc-HZ09 RNA stability. In BPDE-treated human trophoblast cells and in RM villous tissues, MSX1-mediated lnc-HZ09 transcription and METTL3-mediated lnc-HZ09 stability were both greater. In our mouse miscarriage model, B(a)P-treated mice had lower mRNA and protein levels of members of the Pld1/Rac1/Cdc42 pathway. These results suggest that in human trophoblast cells, BPDE exposure up-regulated lnc-HZ09 level, suppressed PLD1/RAC1/CDC42 pathway, and inhibited migration and invasion, providing new insights in understanding the causes and mechanisms of unexplained miscarriage. https://doi.org/10.1289/EHP10477.

A comparative investigation of the sorption of polycyclic aromatic hydrocarbons to various polydisperse micro- and nanoplastics using a novel third-phase partition method

Evidence for direct adverse effects of micro- and nanoplastic particles (MNPs) on human health is scarce, but it has been hypothesized that MNPs act as carriers for environmental pollutants such as polycyclic aromatic hydrocarbons (PAHs). Many studies have already investigated the sorption of PAHs to microplastics, typically using the batch-equilibrium method. Here we established a novel third-phase partition (TPP) method utilizing thermo-extractable polydimethylsiloxane-coated stir-bars as re-usable passive samplers to compare the sorption of PAHs to 17 different MNPs. This method facilitates the quantification of MNP-sorbed pollutants, including those with poor water-solubility without requiring laborious filtration and solvent-extraction steps. Using benzo[a]pyrene (B[a]P) as a representative PAH, sorption kinetics and isotherms for MNPs were evaluated. B[a]P sorption was generally strong but differed by over two-orders of magnitude, clustering according to polymer types in the order of polyamides > polyethylenes ≫ Tire Rubber > polyurethanes > polymethyl methacrylate. B[a]P sorption was diminished for photo-aged MNPs. Within given polymer types, properties including particle size, polarity/hydrophobicity and chain mobility notably influenced B[a]P sorption. When comparing different PAHs, their sorption to selected MNPs increased over five-orders of magnitude with hydrophobicity: anthracene < B[a]P < dibenzo[a,l]pyrene. Our data is an important contribution to the understanding of the sorption behaviors of MNPs. The novel TPP-method represents a universally-applicable approach for the reliable evaluation of sorption characteristics of contaminants and MNPs, and can be easily adapted to desorption studies.Graphical
