Erythrocyte nano-ghosts with dual optical and magnetic resonance characteristics.

Fluorescent organic dyes provide imaging capabilities at cellular and sub-cellular levels. However, a common problem associated with some of the existing dyes such as the US FDA-approved indocyanine green (ICG) is their weak fluorescence emission. Alternative dyes with greater emission characteristics would be useful in various imaging applications. Complementing optical imaging, magnetic resonance (MR) imaging enables deep tissue imaging. Nano-sized delivery systems containing dyes with greater fluorescence emission as well as MR contrast agents present a promising dual-mode platform with high optical sensitivity and deep tissue imaging for image-guided surgical applications. We have engineered a nano-sized platform, derived from erythrocyte ghosts (EGs), with dual near-infrared fluorescence and MR characteristics by co-encapsulation of a brominated carbocyanine dye and gadobenate dimeglumine (Gd-BOPTA). We have investigated the use of three brominated carbocyanine dyes (referred to as BrCy106, BrCy111, and BrCy112) with various degrees of bromination, structural symmetry, and acidic modifications for encapsulation by nano-sized EGs (nEGs) and compared their resulting optical characteristics with nEGs containing ICG. We find that asymmetric dyes (BrCy106 and BrCy112) with one dibromobenzene ring offer greater fluorescence emission characteristics. For example, the relative fluorescence quantum yield ( ) for nEGs fabricated using of BrCy112 is -fold higher than nEGs fabricated using the same concentrations of ICG. The dual-mode nEGs containing BrCy112 and Gd-BOPTA show a nearly twofold increase in their as compared with their single optical mode counterpart. Cytotoxicity is not observed upon incubation of SKOV3 cells with nEGs containing BrCy112. Erythrocyte nano-ghosts with dual optical and MR characteristics may ultimately prove useful in various biomedical imaging applications such as image-guided tumor surgery where MR imaging can be used for tumor staging and mapping, and fluorescence imaging can help visualize small tumor nodules for resection.

Efficient turn-on fluorescent probe cooperated by cascade response for disclosing the fluctuation of cysteine in cells

BackgroundThe research on cysteine (Cys) determination is deemed as a hot topic, since it has been reported to be connected with various physiological processes and disease prediction. However, existing Cys-responding probes may expose some defects such as long reaction time, disappointing photostability, and suboptimal sensitivity. Under such a circumstance, our team has proposed an efficient fluorescent probe with novel sensing mechanism to perfectly cope with the above-mentioned drawbacks. ResultsA novel cascade reaction-based probe 9-(2,2-dicyanovinyl)-2,3,6,7-tetrahydro-1H,5H-pyrido[3,2,1-ij]quinolin-8-yl acrylate (DPQA) has been synthesized for the first time. Undergoing addition-cleavage and cyclization-rearrangement processes, DPQA reacts with Cys to generate an iminocoumarin product with relucent green fluorescence, namely 11-imino-2,3,6,7-tetrahydro-1H,5H,11H-pyrano[2,3-f]pyrido[3,2,1-ij]quinoline-10-carbonitrile (IMC-J), and the relative fluorescence quantum yield (Φf) soars from 0.007 to 0.793. Utilizing such a mechanism, DPQA shows a superb turn-on signal (172-fold), low detection limit (4.1 nM), and wide detection range (5–6000 nM) toward Cys detection. Encouraged by the admirable sensing performance of DPQA, bioimaging of endogenous Cys has been attempted in HeLa cells with satisfactory results. Moreover, cell model of H2O2-induced oxidative stress has been established and the Cys fluctuation during this process has been inspected, elucidating how living cells confront with the eruption of reactive oxygen species (ROS) storm. SignificanceThe probe DPQA with such an intriguing cascade responding process for Cys detection has been endowed with many merits, such as fast reaction and superior sensitivity, conducive to improving responsiveness and rendering it more suitable for further applications. Thereby, we expect that the DPQA would be an efficient tool for detecting Cys fluctuation in living cells of different physiological processes.

4,7-(bis(octyloxy)-3-(quinoline-2-ylmethylene)isoindoline-1-one and its boronflouride complex. synthesis and spectral-luminescent properties

4,7-Bis(octyloxy)isoindoline-1,3-dione was obtained by reacting 3,6-bis(octyloxy)phthalonitrile with sodium butoxide in butanol, followed by treatment with hydrochloric acid. Its condensation with quinaldine in the presence of zinc oxide leads to the formation of ( E , Z )-4,7-bis(octyloxy)-3-(quinolin-2-ylmethylene)isoindolin-1-one, which was treated with BF3۰Et2O in the presence of triethylamine in toluene to give a new unsymmetrical analogue of BODIPY - ( Z )-2-(difluoroboryl)-4,7-bis(octyloxy)-3-(quinolin-2-ylmethylene)isoindolin-1-one. The complex exhibits a Stokes shift of 25 nm and a high relative fluorescence quantum yield (0.68). To support the experimental data, DFT and TD-DFT calculations were performed.

Blue, Yellow, and Red Carbon Dots from Aromatic Precursors for Light-Emitting Diodes.

In this work, multicolor fluorescent carbon dots with red (R-CDs), yellow (Y-CDs), and blue (B-CDs) emissions were prepared by choosing proper aromatic precursors with different amounts of benzene rings through a simple solvothermal method. The characterization showed that the prepared carbon dots were spherical with a size under 10 nm, rich surface functional groups, and good stability. The emission wavelengths were located at 440, 530, and 580 nm under the excitation of 370 nm. The relative fluorescence quantum yield (QY) of R-CDs, Y-CDs, and B-CDs was 11%, 59%, and 33%, respectively. The related characterization demonstrated that the redshift in the photoluminescence was caused by the synergistic effect of the increasing graphitic nitrogen content, quantum size effect and surface oxidation state. By mixing the three prepared CDs into a PVA matrix, the transparent and flexible films produced relucent blue, yellow, and red emissions under 365 nm UV light, and solid-state quenching was effectively avoided. LEDs were fabricated by using B-CDs, Y-CDs, and R-CDs/PVA with a semiconductor chip. These CDs-based LEDs produced bright blue, yellow, and red light with CIE color coordinates of (0.16, 0.02), (0.38, 0.58), and (0.50, 0.49) were successfully manufactured utilizing the prepared blue, yellow and red multicolor carbon dots as the solid luminescent materials. The results showed that the synthesized CDs can be potentially applied in multi-color monitors as a promising candidate for light-emitting diodes (LEDs). This work blazes a novel trail for the controllable preparation of multicolor fluorescent carbon dots.

Synthesis of a fluorene and quinoxaline-based co-polymer for organic electronics

Quinoxaline has recently gained interest as monomer in conjugated copolymers because of its easy synthetic accessibility and successful use in highly efficient organic solar cells. In this contribution, we introduce a quinoxaline–fluorene-co-polymer, PFQ10, synthesized by copolymerization of 5,8-dibromo-6,7-difluoro-2-[(2-hexyldecyl)oxy]quinoxaline and 9,9-dioctyl-9H-9-fluorene-2,7-bis(boronic acid pinacol ester) using the Suzuki–Miyaura reaction. By optimization of the reaction conditions, polymers with molecular weights up to 17.2 kDa and a low dispersity of 1.3 were obtained. PFQ10 showed blue photoluminescence with an emission maximum at 459 nm and a relative fluorescence quantum yield of 0.37. As proof of principle, PFQ10 was employed in organic light-emitting diodes and showed a blue–green electroluminescence.Graphical abstract

Picomolar Detection of Lead Ions (Pb2+) by Functionally Modified Fluorescent Carbon Quantum Dots from Watermelon Juice and Their Imaging in Cancer Cells.

Water contamination due to the presence of lead is one of the leading causes of environmental and health hazards because of poor soil and groundwater waste management. Herein we report the synthesis of functionally modified luminescent carbon quantum dots (CQDs) obtained from watermelon juice as potential nanomaterials for the detection of toxic Pb2+ ions in polluted water and cancer cells. By introducing surface passivating ligands such as ethanolamine (EA) and ethylenediamine (ED) in watermelon juice, watermelon-ethanolamine (WMEA)-CQDs and watermelon-ethylenediamine (WMED)-CQDs exhibited a remarkable ~10-fold and ~6-fold increase in fluorescence intensity with respect to non-doped WM-CQDs. The relative fluorescence quantum yields of WMEA-CQDs and WMED-CQDs were found to be 8% and 7%, respectively, in an aqueous medium. Among various functionally-modified CQDs, only WMED-CQDs showed high selectivity towards Pb2+ ions with a remarkably good limit of detection (LoD) of 190 pM, which is less than that of the permissible limit (72 nM) in drinking water. The functionally altered WMED-CQDs detected Pb2+ metal ions in polluted water and in a human cervical cancer cell line (HeLa), thus advocating new vistas for eco-friendly nanomaterials for their use as diagnostic tools in the environment and biomedical research areas.

Diethylenetriamine-β-CD-modified carbon quantum dots for selective fluorescence sensing of Hg2+ and Fe3+ and cellular imaging

Diethylenetriamine-β-cyclodextrin-modified carbon quantum dots (3 N-CQDs) were synthesized via a one-step hydrothermal method using citric acid as the carbon source and diethylenetriamine-β-cyclodextrin (3 N-β-CD) as the nitrogen source. The successful preparation of 3 N-CQDs were revealed by infrared absorption spectroscopy, ultraviolet (UV)-visible absorption spectroscopy, fluorescence spectroscopy, XRD, XPS, TEM, and TG. Further spectroscopic studies showed that the synthesized carbon quantum dots offered good anti-interference capability. The relative fluorescence quantum yield was 67.2 %. The limits of detection for Hg2+ and Fe3+ were 0.25 µM and 0.57 μM, respectively. Cytotoxicity and imaging studies showed that the prepared carbon quantum dots had low cytotoxicity, good biocompatibility, and good cellular imaging capability for HeLa cells. They offered fluorescent sensing of Hg2+ and Fe3+ in live cells. Therefore, 3 N-CQDs were ideal fluorescent probes for the detection of Hg2+ and Fe3+ in water.

Green synthesis of gardenia seeds-based carbon dots for bacterial imaging and antioxidant activity in aqueous and oil samples.

In this work, luminescent carbon dots with gardenia seeds as carbon precursors (GCDs) were synthesized using a one-step mild pyrolysis process and were then used as probes for imaging of bacterial (Escherichia coli). The GCDs showed a strong emission at 430 nm when excited at 370 nm. The relative fluorescence quantum yield of GCDs was found to be 1.13% in an aqueous medium. Rapid internalization of the GCDs by bacteria was confirmed by three colors (blue, green, and yellow) images that were obtained using confocal fluorescence microscopy. In addition, GCDs were noted to exhibit potent scavenging activities against DPPH˙, ˙OH, and ˙O2- free radicals. GCDs were also assayed as antioxidants in an oil sample by volumetric determination of the peroxide value. Thus, GCDs exhibited good antioxidant properties both in aqueous and oil media. In addition, a free fatty acid quantification kit in the presence of GCDs showed enhanced fluorescence detection of palmitic acid with a remarkably good limit of detection of 0.08 μM, which is lower than that in the absence of GCDs (0.76 μM). The proposed fluorescence method was then successfully used to determine the concentration of palmitic acid spiked in milk powder samples, with spiked recoveries of 82.6-109.6% and relative standard deviations of 0.9-4.6%.

The Influence of Medium on Fluorescence Quenching of Colloidal Solutions of the Nd3+: LaF3 Nanoparticles Prepared with HTMW Treatment.

An original method was proposed to reduce the quenching of the NIR fluorescence of colloidal solutions of 0.1 at. % Nd3+: LaF3 nanoparticles (NPs) synthesized by aqueous co-precipitation method followed by hydrothermal microwave treatment. For this, an aqueous colloidal solution of NPs was precipitated by centrifugation and dissolved in the same volume of DMSO. The kinetics of static fluorescence quenching of Nd3+ donors of doped NPs dispersed in two solvents was analyzed to determine and to compare the concentrations of OH- quenching acceptors uniformly distributed throughout the volume of the NPs. The dependences of the relative fluorescence quantum yield φ of colloidal solutions on the concentration of OH- groups in the NPs were calculated and were also used to determine concentration of acceptors in the volume of NPs in different solvents. It was found that the concentration of OH- groups in NPs dispersed in DMSO is almost two times lower than in NPs dispersed in water. This gives an almost two-fold increase in the relative fluorescence quantum yield φ for the former. The sizes of synthesized NPs were monitored by common TEM and by applying a rapid procedure based on optical visualization of the trajectories of the Brownian motion of NPs in solution using a laser ultramicroscope. The use of two different methods made it possible to obtain more detailed information about the studied NPs.

Multicomponent Synthesis of Fluorescent Thiazole-Indole Hybrids and Thiazole-Based Novel Polymers.

Herein, we report an efficient multicomponent reaction for the synthesis of trisubstituted thiazoles involving a one-pot C-C, C-N, and C-S bond-forming process from the readily available starting materials. The reaction of arylglyoxal, indole, and aryl thioamides in the acetic acid medium under sealed heating conditions provided 3-(2,4-diarylthiazol-5-yl)-1H-indoles (4) in good to excellent yields. Using a similar reaction strategy, the reaction of arylglyoxal, aryl thioamide, and 2,5-dihydroxy-1,4-benzoquinone provided structurally interesting bis-thiazoles having dihydroxy-1,4-benzoquinone linker (9). All of the products were fully characterized by spectroscopic techniques. We also recorded single-crystal X-ray diffraction (XRD) of compounds 4b and 9a for unambiguous structure determination. Indole-linked trisubstituted thiazoles (4) exhibit prominent fluorescence properties. The relative fluorescence quantum yields of all of the thiazole-linked indoles were measured in the dimethyl sulfoxide (DMSO) medium with respect to quinine sulfate in 0.1 M H2SO4 as reference. The scope of this reaction was further explored by preparing novel polymers 11a and 11b using naphthalene/benzene-1,4-bis(carbothioamide) in multicomponent polymerization.

Base-Stacking Heterogeneity in RNA Resolved by Fluorescence-Detected Circular Dichroism Spectroscopy.

RNA plays a critical role in many biological processes, and the structures it adopts are intimately linked to those functions. Among many factors that contribute to RNA folding, van der Waals interactions between adjacent nucleobases stabilize structures in which the bases are stacked on top of one another. Here, we utilize fluorescence-detected circular dichroism spectroscopy (FDCD) to investigate base-stacking heterogeneity in RNA labeled with the fluorescent adenine analogue 2-aminopurine (2-AP). Comparison of standard (transmission-detected) CD and FDCD spectra reveals that in dinucleotides, 2-AP fluorescence is emitted almost exclusively by unstacked molecules. In a trinucleotide, some fluorescence is emitted by a population of stacked and highly quenched molecules, but more than half originates from a minor ∼10% population of unstacked molecules. The combination of FDCD and standard CD measurements reveals the prevalence of stacked and unstacked conformational subpopulations as well as their relative fluorescence quantum yields.

In Vitro and In Silico Evaluation of Indole-Bearing Squaraine Dyes as Potential Human Serum Albumin Fluorescent Probes

The quantitative determination of proteins is an important parameter in biochemistry, biotechnology and immunodiagnostics, and the importance of serum albumin in clinical diagnosis should be highlighted, given that alterations in its concentration are generally associated with certain diseases. As possible probes for this purpose, squaraine dyes have been arousing the interest of many researchers due to their unique properties, such as absorption in the visible spectra, moderate relative fluorescence quantum yields and increased fluorescence intensity after non-covalent binding to specific ligands. In this work, five squaraine dyes, four of which have never been reported in the literature, were characterized and evaluated in vitro and in silico concerning their potential application as fluorescent probes for human serum albumin detection. After interaction with the protein, the fluorescence intensity increased from 12 to 41 times, depending on the dye under study. High sensitivity (1.0 × 105–5.4 × 105 nM), low detection limits (168–352 nM) and moderate quantitation limits (560–1172 nM) were obtained, proving the efficiency of the method. In addition, moderate-to-excellent selectivity was observed compared to γ-globulin proteins. Molecular docking suggests that the dyes interact more effectively with the Sudlow site I, and binding energies have been markedly higher than those of warfarin, a molecule known to bind to this site specifically.

A Comparison of the Photophysical, Electrochemical and Cytotoxic Properties of meso-(2-, 3- and 4-Pyridyl)-BODIPYs and Their Derivatives.

Boron dipyrromethene (BODIPY) dyes bearing a pyridyl moiety have been used as metal ion sensors, pH sensors, fluorescence probes, and as sensitizers for phototherapy. A comparative study of the properties of the three structural isomers of meso-pyridyl-BODIPYs, their 2,6-dichloro derivatives, and their corresponding methylated cationic pyridinium-BODIPYs was conducted using spectroscopic and electrochemical methods, X-ray analyses, and TD-DFT calculations. Among the neutral derivatives, the 3Py and 4Py isomers showed the highest relative fluorescence quantum yields in organic solvents, which were further enhanced 2-4-fold via the introduction of two chlorines at the 2,6-positions. Among the cationic derivatives, the 2catPy showed the highest relative fluorescence quantum yield in organic solvents, which was further enhanced by the use of a bulky counter anion (PF6−). In water, the quantum yields were greatly reduced for all three isomers but were shown to be enhanced upon introduction of 2,6-dichloro groups. Our results indicate that 2,6-dichloro-meso-(2- and 3-pyridinium)-BODIPYs are the most promising for sensing applications. Furthermore, all pyridinium BODIPYs are highly water-soluble and display low cytotoxicity towards human HEp2 cells.

Donor-Acceptor Pyridinium Salts for Photo-Induced Electron-Transfer-Driven Modification of Tryptophan in Peptides, Proteins, and Proteomes Using Visible Light.

Tryptophan (Trp) plays a variety of critical functional roles in protein biochemistry; however, owing to its low natural frequency and poor nucleophilicity, the design of effective methods for both single protein bioconjugation at Trp as well as for in situ chemoproteomic profiling remains a challenge. Here, we report a method for covalent Trp modification that is suitable for both scenarios by invoking photo-induced electron transfer (PET) as a means of driving efficient reactivity. We have engineered biaryl N-carbamoyl pyridinium salts that possess a donor-acceptor relationship that enables optical triggering with visible light whilst simultaneously attenuating the probe's photo-oxidation potential in order to prevent photodegradation. This probe was assayed against a small bank of eight peptides and proteins, where it was found that micromolar concentrations of the probe and short irradiation times (10-60 min) with violet light enabled efficient reactivity toward surface exposed Trp residues. The carbamate transferring group can be used to transfer useful functional groups to proteins including affinity tags and click handles. DFT calculations and other mechanistic analyses reveal correlations between excited state lifetimes, relative fluorescence quantum yields, and chemical reactivity. Biotinylated and azide-functionalized pyridinium salts were used for Trp profiling in HEK293T lysates and in situ in HEK293T cells using 440 nm LED irradiation. Peptide-level enrichment from live cell labeling experiments identified 290 Trp modifications, with 82% selectivity for Trp modification over other π-amino acids, demonstrating the ability of this method to identify and quantify reactive Trp residues from live cells.

Proximal nitrogen reduces the fluorescence quantum yield of nitrogen-vacancy centres in diamond

The nitrogen-vacancy colour centre in diamond is emerging as one of the most important solid-state quantum systems. It has applications to fields including high-precision sensing, quantum computing, single photon communication, metrology, nanoscale magnetic imaging and biosensing. For all of these applications, a high quantum yield of emitted photons is desirable. However, diamond samples engineered to have high densities of nitrogen-vacancy centres show levels of brightness varying significantly within single batches, or even within the same sample. Here we show that nearby nitrogen impurities quench emission of nitrogen-vacancy centres via non-radiative transitions, resulting in a reduced fluorescence quantum yield. We monitored the emission properties of nitrogen-vacancy centre ensembles from synthetic diamond samples with different concentrations of nitrogen impurities. All samples were irradiated with high energy electrons to create high densities of nitrogen-vacancy centres relative to the concentration of nitrogen impurities. While at low nitrogen densities of 1.81 ppm we measured a lifetime of 13.9 ns, we observed a strong reduction in lifetime with increasing nitrogen density. We measure a lifetime as low as 4.4 ns at a nitrogen density of 380 ppm. The change in lifetime matches a reduction in relative fluorescence quantum yield from 77.4% to 32% with an increase in nitrogen density from 88 ppm to 380 ppm, respectively. These results will inform the conditions required to optimise the properties of diamond crystals devices based on the fluorescence of nitrogen-vacancy centres. Furthermore, this work provides insights into the origin of inhomogeneities observed in high-density nitrogen-vacancy ensembles within diamonds and nanodiamonds.

Facile and High-yield Synthesis of N-doped Carbon Quantum Dots from Biomass Quinoa Saponin for the Detection of Co2.

Hydrothermal synthesis of carbon quantum dots (CQDs) from natural biomass is a green and sustainable route for CQDs applications in various fields. In this work, the preparation and characterization of CQDs based on quinoa saponin were investigated. The optimum synthetic conditions determined by orthogonal experiments were as follows: 2 g quinoa saponin powder and 0.04 mol ethylenediamine reacted at 200°C for 10 h. The relative fluorescence quantum yield (QY = 22.2%) can be obtained, which is higher than some results reported in the literatures. The prepared CQDs had a small and uniform size (∼2.25 nm) and exhibited excitation wavelength-dependent blue light emission behavior. The CQDs displayed excellent sensitivity for Co2+ detection along with good linear correlation ranging from 20 to 150 µM and the detection limit of 0.49 µM. The CQDs prepared in this experiment were successfully implanted into soybean sprouts for fluorescence imaging. The sprouts could grow healthily even soaked in the CQDs solution for two weeks, demonstrating the low toxicity of the CQDs. The advantages of the CQDs, such as low cost, ease of manufacture, nontoxicity, and stability, have potential applications in many areas such as metal ion detection and biosensing.

Aesculetin Exhibits Strong Fluorescent Photoacid Character.

Coumarins are bioactive molecules that often serve as defenses in plant and animal systems, and understanding their fundamental behavior is essential for understanding their bioactivity. Aesculetin (6,7-dihydroxycoumarin) has recently attracted attention due to its ability to act as an antioxidant, but little is known about its photophysical properties. The fluorescence lifetimes of its neutral and anion form in water are 19 ± 2ps and 2.3 ± 0.1ns, respectively. Assuming the short lifetime of the neutral is determined by ESPT, we estimate kPT ~ 5 × 1010s-1. Using steady-state and time-resolved fluorescence spectroscopy, we determine its ground and excited-state [Formula: see text] to be 7.3 and -1, respectively, making it one of the strongest photoacids of the natural coumarins. Aesculetin exhibits a strong pH dependence of the relative fluorescence quantum yield becoming much more fluorescent above [Formula: see text]. The aesculetin anion [Formula: see text] slightly photobasic character. We also report that aesculetin forms a fluorescent catechol-like complex with boric acid, and this complex has a [Formula: see text] of 5.6.

Stable Aqueous Colloidal Solutions of Nd3+: LaF3 Nanoparticles, Promising for Luminescent Bioimaging in the Near-Infrared Spectral Range.

Two series of stable aqueous colloidal solutions of Nd3+: LaF3 single-phase well-crystallized nanoparticles (NPs), possessing a fluorcerite structure with different activator concentrations in each series, were synthesized. A hydrothermal method involving microwave-assisted heating (HTMW) in two Berghof speedwave devices equipped with one magnetron (type I) or two magnetrons (type II) was used. The average sizes of NPs are 15.4 ± 6 nm (type I) and 21 ± 7 nm (type II). Both types of NPs have a size distribution that is well described by a double Gaussian function. The fluorescence kinetics of the 4F3/2 level of the Nd3+ ion for NPs of both types, in contrast to a similar bulk crystal, demonstrates a luminescence quenching associated not only with Nd–Nd self-quenching, but also with an additional Nd–OH quenching. A method has been developed for determining the spontaneous radiative lifetime of the excited state of a dopant ion, with the significant contribution of the luminescence quenching caused by the presence of the impurity OH– acceptors located in the bulk of NPs. The relative quantum yield of fluorescence and the fluorescence brightness of an aqueous colloidal solution of type II NPs with an optimal concentration of Nd3+ are only 2.5 times lower than those of analogous Nd3+: LaF3 single crystals.

Ratiometric covalent organic framework florescence sensor for detecting hydrazine produced from isoniazid metabolism in cell

Designing luminescent COFs with high fluorescence quantum yield is always a huge challenge. In this work, we synthesized COF TzDha through Schiff base reaction between 4, 4′, 4′'-(1,3,5-triazine-2,4,6-triyl) trianiline (Tz) and 2,5-dihydroxyterephthalaldehyde (Dha), and post-modified COF TzDha-AC was grafted by refluxing in acetic anhydride. TzDha-AC shows higher relative fluorescence quantum yield (Φf) in various organic solvents and water, which is approximately 1.35 to 5.31 times to TzDha. What’s more surprising, TzDha-AC shows excellent ratiometric effect when it is used to detecte hydrazine (N2H4). Detection mechanism is attributed to that hydrazine prevents the intramolecular charge transfer (ICT) from benzene ring to triazine ring of TzDha-AC, thus the emission blue shifted from 601 nm to 526 nm. The detection limit (DL) of N2H4 is calculated to be 0.038 mM in the linear range of 0−10 mM. In addition, we also use TzDha-AC to detecte hydrazine produced from isoniazid metabolism in mouse liver cells and find that TzDha-AC could be a new material employed for simple isoniazid metabolism in cell.

Nitrogen-doped carbon dots for wash-free imaging of nucleolus orientation.

Carbon dots (CDs) are a rising star in the field of cellular imaging, especially cytoplasmic imaging, attributing to the super-stable optical performance and ultra-low biological toxicity. Nucleolus can accurately reflect the expression state of a cell and is strongly linked to the occurrence and development of many diseases, so exploring bran-new CDs for nucleolus-orientation imaging with no-wash technology has important theoretical value and practical significance. Herein, nitrogen-doped carbon dots (N-CDs) with green fluorescence (the relative fluorescence quantum yield of 24.4%) was fabricated by the hydrothermal treatment of m-phenylenediamine and p-aminobenzoic acid. The N-CDs possess small size, bright green fluorescence, abundant surface functional groups, excellent fluorescence stability and good biocompatibility, facilitating that the N-CDs are an excellent imaging reagent for cellular imaging. N-CDs can particularly bind to RNA in nucleoli to enhance their fluorescence, which ensures that the N-CDs can be used in nucleolus-orientation imaging with high specificity and wash-free technique. This study demonstrates that the N-CDs have a significant feasibility to be used for nucleolus-orientation imaging in biomedical analysis and clinical diagnostic applications.
