Synergistic roles of acyl-CoA binding protein (ACBP1) and sterol carrier protein 2 (SCP2) in Toxoplasma lipid metabolism.

Toxoplasma gondii relies on apicoplast-localised FASII pathway and endoplasmic reticulum-associated fatty acid elongation pathway for the synthesis of fatty acids, which flow through lipid metabolism mainly in the form of long-chain acyl-CoA (LCACoAs) esters. Functions of Toxoplasma acyl-CoA transporters in lipid metabolism remain unclear. Here, we investigated the roles of acyl-CoA-binding protein (TgACBP1) and a sterol carrier protein-2 (TgSCP2) as cytosolic acyl-CoA transporters in lipid metabolism. The fluormetric binding assay and yeast complementation confirmed the acyl-CoA binding activities of TgACBP1 and TgSCP2, respectively. Disruption of either TgACBP1 or TgSCP2 caused no obviously phenotypic changes, whereas double disruption resulted in defects in intracellular growth and virulence to mice. Gas chromatography coupled with mass spectrometry (GC-MS) results showed that TgACBP1 or TgSCP2 disruption alone led to decreased abundance of C18:1, whereas double disruption resulted in reduced abundance of C18:1, C22:1, and C24:1. 13 C labelling assay combined with GC-MS showed that double disruption of TgACBP1 and TgSCP2 led to reduced synthesis rates of C18:0, C22:1, and C24:1. Furthermore, high performance liquid chromatography coupled with high resolution mass spectrometry (HPLC-HRMS) was used for lipidomic analysis of parasites and indicated that loss of TgACBP1 and TgSCP2 caused serious defects in production of glycerides and phospholipids. Collectively, TgACBP1 and TgSCP2 play synergistic roles in lipid metabolism in T.gondii.

The effect of high-fat diet and inhibition of ceramide production on insulin action in liver.

Liver, as one of the most important organs involved in lipids and glucose metabolism, is perceived as a key tissue for pharmacotherapy of insulin resistance (IRes) and type 2 diabetes. Ceramides (Cer) are biologically active lipids, which accumulation is associated with the induction of muscle IRes. We sought to determine the role of intrahepatic bioactive lipids production on insulin action in liver of insulin-resistant rats and after myriocin administration. The experiments were conducted on male Wistar rats divided into three groups: Control, fed high-fat diet (HFD), and fed HFD and treated with myriocin (HFD/Myr). Before sacrifice, the animals were infused with a [U-13 C]palmitate to calculate lipid synthesis rate by means of tracer incorporation technique in particular lipid groups. Liver Cer, diacylglycerols (DAG), acyl-carnitine concentration, and isotopic enrichment were analyzed by LC/MS/MS. Proteins involved in lipid metabolism and insulin pathway were analyzed by western blot analysis. An OGTT and ITT was also performed. HFD-induced IRes and increased both the synthesis rate and the content of DAG and Cer, which was accompanied by inhibition of an insulin pathway. Interestingly, myriocin treatment reduced synthesis rate not only of Cer but also DAG and improved insulin sensitivity. We conclude that the insulin-sensitizing action of myriocin in the liver is a result of the lack of inhibitory effect of lipids on the insulin pathway, due to the reduction of their synthesis rate. This is the first study showing how the synthesis rate of individual lipid groups in liver changes after myriocin administration.

Chicken domestication changes expression of stress-related genes in brain, pituitary and adrenals

Domesticated species have an attenuated behavioral and physiological stress response compared to their wild counterparts, but the genetic mechanisms underlying this change are not fully understood. We investigated gene expression of a panel of stress response-related genes in five tissues known for their involvement in the stress response: hippocampus, hypothalamus, pituitary, adrenal glands and liver of domesticated White Leghorn chickens and compared it with the wild ancestor of all domesticated breeds, the Red Junglefowl. Gene expression was measured both at baseline and after 45 min of restraint stress. Most of the changes in gene expression related to stress were similar to mammals, with an upregulation of genes such as FKBP5, C-FOS and EGR1 in hippocampus and hypothalamus and StAR, MC2R and TH in adrenal glands. We also found a decrease in the expression of CRHR1 in the pituitary of chickens after stress, which could be involved in negative feedback regulation of the stress response. Furthermore, we observed a downregulation of EGR1 and C-FOS in the pituitary following stress, which could be a potential link between stress and its effects on reproduction and growth in chickens.We also found changes in the expression of important genes between breeds such as GR in the hypothalamus, POMC and PC1 in the pituitary and CYP11A1 and HSD3B2 in the adrenal glands. These results suggest that the domesticated White Leghorn may have a higher capacity for negative feedback of the HPA axis, a lower capacity for synthesis of ACTH in the pituitary and a reduced synthesis rate of corticosterone in the adrenal glands compared to Red Junglefowl. All of these findings could explain the attenuated stress response in the domesticated birds.

Impaired Bile Acid Homeostasis in Children with Severe Acute Malnutrition.

ObjectiveSevere acute malnutrition (SAM) is a major cause of mortality in children under 5 years and is associated with hepatic steatosis. Bile acids are synthesized in the liver and participate in dietary fat digestion, regulation of energy expenditure, and immune responses. The aim of this work was to investigate whether SAM is associated with clinically relevant changes in bile acid homeostasis.DesignAn initial discovery cohort with 5 healthy controls and 22 SAM-patients was used to identify altered bile acid homeostasis. A follow up cohort of 40 SAM-patients were then studied on admission and 3 days after clinical stabilization to assess recovery in bile acid metabolism. Recruited children were 6–60 months old and admitted for SAM in Malawi. Clinical characteristics, feces and blood were collected on admission and prior to discharge. Bile acids, 7α-hydroxy-4-cholesten-3-one (C4) and FGF-19 were quantified.ResultsOn admission, total serum bile acids were higher in children with SAM than in healthy controls and glycine-conjugates accounted for most of this accumulation with median and interquartile range (IQR) of 24.6 μmol/L [8.6–47.7] compared to 1.9 μmol/L [1.7–3.3] (p = 0.01) in controls. Total serum bile acid concentrations did not decrease prior to discharge. On admission, fecal conjugated bile acids were lower and secondary bile acids higher at admission compared to pre- discharge, suggesting increased bacterial conversion. FGF19 (Fibroblast growth factor 19), a marker of intestinal bile acid signaling, was higher on admission and was associated with decreased C4 concentrations as a marker of bile acid synthesis. Upon recovery, fecal calprotectin, a marker of intestinal inflammation, was lower.ConclusionSAM is associated with increased serum bile acid levels despite reduced synthesis rates. In SAM, there tends to be increased deconjugation of bile acids and conversion from primary to secondary bile acids, which may contribute to the development of liver disease.

Lipolytic activities in developing turbot larvae as influenced by diet

The development of neutral lipase and phospholipase activities was studied in larval turbot fed live prey. Activities of neutral lipase and phospholipase (activity larva−1) increased significantly between days 6 and 24 after hatching in turbot larvae. The specific activities of both enzymes (activity µg protein−1) decreased in older larvae. Feeding of a microdiet for 3 days (days 10–13) affected the lipolytic activity of neutral lipase and phospholipase negatively, compared to the larvae fed on rotifers. Since neutral lipase activities in whole larval homogenates and in the gut were significantly lower, it suggests a reduced synthesis rate and a reduced secretion of the enzyme in larvae fed the microdiet. A correlation between neutral lipase and phospholipase activities was found in larvae fed rotifers, but not in larvae fed the microdiet. This may indicate different regulating and stimulating mechanisms for these enzymes. The contribution of exogenous enzymes from ingested live prey to the total larval enzyme activity was about 6% for neutral lipase and 10% for phospholipase on day 6. The exogenous prey enzymes accounted for only 2% of the total activities in 12-day-old turbot larvae, suggesting that enzymes from prey did not contribute considerably to the digestion of lipids.

Idiopathic hypoalbuminemia explained by reduced synthesis rate and an increased catabolic rate

Objective: To determine the contribution of albumin synthetic and catabolic rates to steady state levels in a patient with idiopathic hypoalbuminemia. Methods: Using L-[1- 13C] valine, both FSR (fractional synthesis rate) as well as FCR (fractional catabolic rate) were studied. Human albumin cDNA analysis and determination of the exact albumin mass by electrospray mass spectrometry were performed. Results: Compared with controls, plasma albumin concentration in the patient was reduced (6.7 vs. 37.0 ± 2.6 g/L). Albumin FSR (= FCR in steady state) was increased compared to controls. The ASR (absolute synthesis rate) of albumin was decreased based on the enrichment in plasma valine and KIV, but estimated to be normal based on VLDL apoB100 at plateau compared to controls. Direct estimation of albumin FCR rejected the latter. No mutation was found in the transcribed region of albumin gene. The exact mass of albumin (66.493 Da) was not different from controls. Conclusion: The hypoalbuminemia was a result of accelerated clearance of albumin from plasma in addition to defective albumin synthesis. This study also shows that the chosen method of the precursor pool could lead to misinterpretation of data in hepatic protein synthesis.

Hfq (HF1) stimulates ompA mRNA decay by interfering with ribosome binding

The adaptation of mRNA stability to environmental changes is a means of cells to adjust the level of gene expression. The Escherichia coli ompA mRNA has served as one of the paradigms for regulated mRNA decay in prokaryotes. The stability of the transcript is known to be correlated inversely with the bacterial growth rate. Thus, the regulation of ompA mRNA stability meets the physiological needs to adjust the level of ompA expression to the rate of cell division. Recently, host factor I (Hfq/HF1) was shown to be involved in the regulation of ompA mRNA stability under slow growth conditions. Here, we present the first direct demonstration that 30S ribosomes bound to the ompA 5'-UTR protect the transcript from RNase E cleavage in vitro. However, the 30S protection was found to be abrogated in the presence of Hfq. Toeprinting and in vitro translation assays revealed that translation of ompA is repressed in the presence of Hfq. These in vitro studies are corroborated by in vivo expression studies demonstrating that the reduced synthesis rate of OmpA effected by Hfq results in functional inactivation of the ompA mRNA. The data are discussed in terms of a model wherein Hfq regulates the stability of ompA mRNA by competing with 30S ribosomes for binding to the ompA 5'-UTR.

The effect of age on protein metabolism.

The mechanisms of senescence remain to be fully defined. This review focuses on recent advances in our understanding of body protein turnover, which is essential for the remodeling of tissues and production of specific proteins in time of need. Recent advances in technology make it possible to measure the synthesis rate of muscle myosin heavy chain, mitochondrial proteins and sarcoplasmic proteins, providing insight into the mechanisms of the sarcopenia of aging. A reduced synthesis rate of myosin heavy chain and mitochondrial protein may explain muscle weakness and fatiguability that occurs with aging. Aging also seems to affect selected liver proteins such as fibrinogen. The potential roles of exercise and hormone replacement in slowing the age-related decline in protein turnover is discussed.

Rate of protein synthesis in the skin and muscle of non-pregnant, pregnant and lactating Merino ewes

Fractional synthesis rates of protein (FSR) in the midside skin and Longissimus dorsi muscle of Merino ewes were measured to assess the partitioning of nutrients during late pregnancy and early lactation in two experiments. Protein synthesis was measured by using a flooding-dose of L −[ 15 N] phenylalanine. The effects of diets (canola meal vs lupin seeds) on nutrient partitioning to skin and muscle in dry, pregnant and lactating ewes were also investigated. The FSR in the skin of non-pregnant ewes ranged between 10.6% d −1 and 12.8% d −1, and in muscle between 1.13% d −1 and 2.06% d −1 at maintenance feed intakes. There were no statistical differences ( P>0.05) in FSR of the skin and muscle between pregnant and non-pregnant ewes though the skin FSR values were slightly lower in pregnant ewes. During lactation, FSR increased in the skin but declined in the muscle in comparison with pregnancy, indicating that muscle protein was being expended while skin and wool follicles continued to use nutrients. The ewes fed a diet containing canola meal, which had a lower degradability of protein in the rumen and a higher content of sulphur-containing amino acids than the lupin seed diet, had a higher rate of protein synthesis in the skin ( P=0.04) but a lower rate of synthesis in the muscle ( P=0.02) than those ewes fed lupins. The differences between the two diets, in skin protein synthesis, were greater in the reproducing than in the dry ewes. In conclusion, FSR in the skin of ewes was not significantly reduced during reproduction but tended to be lower during pregnancy. Reduced wool growth in late pregnancy may be due to a reduction in wool producing tissue alone or to a combination of less tissue and a slightly reduced synthesis rate. Feeding canola meal increased protein synthesis in the skin of reproducing and non-pregnant ewes. Finally FSR in skin and muscle responded differently to pregnancy, lactation and protected protein.

Erythrocyte glutathione deficiency in symptom-free HIV infection is associated with decreased synthesis rate.

Although several studies have documented intra- and extracellular glutathione (GSH) deficiency in asymptomatic human immunodeficiency virus (HIV) infection, the mechanisms responsible for the altered GSH homeostasis remain unknown. To determine whether decreased synthesis contributes to this alteration of GSH homeostasis, a primed-constant infusion of [2H2]glycine was used to measure the fractional and absolute rates of synthesis of GSH in five healthy and five symptom-free HIV-infected subjects before and after supplementation for 1 wk with N-acetylcysteine. The erythrocyte GSH concentration of the HIV-infected group was lower (P < 0.01) than that of the control group (1.4 +/- 0.16 vs. 2.4 +/- 0.08 mmol/l). The smaller erythrocyte GSH pool of the HIV-infected group was associated with a significantly slower (P < 0.01) absolute synthesis rate of GSH (1.15 +/- 0.14 vs. 1.71 +/- 0.15 mmol. l-1. day-1) compared with controls. Cysteine supplementation elicited significant increases in both the absolute rate of synthesis and the concentration of erythrocyte GSH. These results suggest that the GSH deficiency of HIV infection is due in part to a reduced synthesis rate secondary to a shortage in cysteine availability.

First steps from a two-dimensional protein index towards a response-regulation map for Bacillus subtilis.

Data on the identification of proteins of Bacillus subtilis on two-dimensional (2-D) gels as well as their regulation are summarized and the identification of 56 protein spots is included. The pattern of proteins synthesized in Bacillus subtilis during exponential growth, during starvation for glucose or phosphate, or after the imposition of stresses like heat shock, salt- and ethanol stress as well as oxidative stress was analyzed. N-terminal sequencing of protein spots allowed the identification of 93 proteins on 2-D gels, which are required for the synthesis of amino acids and nucleotides, the generation of ATP, for glycolyses, the pentose phosphate cycle, the citric acid cycle as well as for adaptation to a variety of stress conditions. A computer-aided analysis of the 2-D gels was used to monitor the synthesis profile of more than 130 protein spots. Proteins performing housekeeping functions during exponential growth displayed a reduced synthesis rate during stress and starvation, whereas spots induced during stress and starvation were classified as specific stress proteins induced by a single stimulus or a group of related stimuli, or as general stress proteins induced by a variety of entirely different stimuli. The analysis of mutants in global regulators was initiated in order to establish a response regulation map for B. subtilis. These investigations demonstrated that the alternative sigma factor sigma B is involved in the regulation of almost all of the general stress proteins and that the phoPR two-component system is required for the induction of a large part but not all of the proteins induced by phosphate starvation.

Synthesis of ventricular mitochondrial proteins in vivo: effect of acute ethanol toxicity.

Most studies on the pathological responses of the heart to ethanol have been conducted in isolated systems. The objectives of this study were to determine (1) the synthesis rate of ventricular mitochondrial proteins in vivo and (2) whether the synthesis rates of these proteins are perturbed by acute ethanol exposure in vivo. Fractional rates of protein synthesis [defined as the percentage of tissue protein renewed each day; i.e., ks (%/day)] were determined in male Wistar rats by in vivo injection of a flooding dose of L-[4-3H] phenylalanine. Subsarcolemmal mitochondria were released by polytron treatment, and the isolation of interfibrillar mitochondria involved treatment of the cardiac homogenate with the proteolytic enzyme Nagarse. In the control rats mean ks values of 22.4%/day were observed for mixed cardiac proteins. The synthesis rates of subsarcolemmal and interfibrillar mitochondrial proteins were lower, i.e., 16.9%/day and 10.9%/day, respectively. Acute ethanol administration (75 mmol/kg body weight ip, 2.5 hr) depressed the fractional rate of protein synthesis in all cardiac fractions, including those pertaining to the mitochondria, as follows: mixed fraction--21%, p < 0.01; subsarcolemmal mitochondria--23%, p < 0.01; interfibrillar mitochondria--26%, p < 0.05; and nuclear fraction--20%, p < 0.05. In conclusion, the reduced synthesis rate of the mitochondrial proteins in response to acute ethanol exposure may in some way be partly connected with the depression in myocardial contractility and associated functional damage of mitochondrial metabolism.

Aberrant biosynthesis and transport of class I major histocompatibility complex molecules in cells transformed with highly oncogenic human adenoviruses

The expression of class I major histocompatibility complex antigens on the surface of cells transformed by adenovirus 12 (Ad12) is generally very low. The absence of class I antigens correlates with the high oncogenicity of this virus. In primary embryonal fibroblasts from transgenic mice that express both endogenous H-2 genes and a miniature swine class I gene (PD1), Ad12-mediated transformation results in complete suppression of cell surface expression of class I antigens, but only 50% of the cell lines tested demonstrated decreased steady state levels of class I mRNA. The complete absence of cell surface class I antigens is accompanied by decreased levels of newly synthesized class I molecules. Those molecules that are immunoprecipitated by class I-specific antibodies are assembled with beta 2-microglobulin, but their transport through the Golgi is inefficient. The biosynthesis of both the endogenous H-2K, H-2D, and the transgene product, PD1, is similarly altered in the transformed cells. The results suggest that Ad12 transformation is associated with both reduced synthesis rate and inefficient transport of class I molecules to the cell surface. This might be a general mechanism by which virus-infected or -transformed cells escape immune surveillance.

Intoxications with anticholinesterases: effect of different combinations of antidotes on the dynamics of acetylcholine in mouse brain.

Intoxications with organophosphorus compounds are normally treated with a mixture of atropine and an enzyme regenerating oxime. The addition of diazepam to the conventional drug therapy is reported to greatly improve the antidotal effect. The implication of the cholinergic system in such intoxications prompted us to study the effect of different combinations of antidotes on the acetylcholine (ACh) synthesizing system in mouse brain in vivo. The antidotes studied in this paper are diazepam, HI-6 and 1-hyoscyamine, the active enantiomer of atropine. Diazepam decreases the synthesis rate of ACh both when administered separately and in combination with 1-hyoscyamine and HI-6. This is in contrast to 1-hyoscyamine which, in addition to blocking muscarinic receptors, also increases the release and rate of synthesis of ACh, which probably is an unfavourable effect of the antidote. This might at least partly explain the advantage of combining 1-hyoscyamine and an oxime with diazepam in intoxications with anticholinesterases. Mice administered soman (0.75 x LD50), after pretreatment with the three-drug combination of antidotes, show no cholinergic symptoms despite a 50% increase in endogenous ACh. The rate of synthesis of ACh in these mice is in the same range as in animals administered diazepam alone. Mice administered the same dose of soman with no antidotal pretreatment suffer from severe tremor and salivation, and have a strongly reduced synthesis rate of ACh.

Isopropyl-?-d-thiogalactopyranoside influences the metabolism of Escherichia coli

Addition of isopropyl β-D-thiogalactopyranoside (IPTG) to a strain of Escherichia coli with one lac promoter in its chromosome causes reduction in synthesis rate for a set of protiens. One of these proteins, designated H35, is a prominent cellular protein present only during exponential growth. Reduction of H35 synthesis is transient and delayed following an IPTG pulse. Cellular response to an IPTG pulse during exponential growth shares several features with a heat shock response. Significant increases in the specific growth rate of cells in both amino-acid-supplemented minimal medium and complex medium were observed for some IPTG concentrations relative to IPTG-free cultures. Other IPTG concentrations caused a reduction in specific growth rate.

Apolipoprotein gene expression in the rat is regulated in a tissue-specific manner by thyroid hormone

We have studied the regulation of rat intestinal and hepatic apolipoprotein gene expression, in vivo, after alterations in thyroid hormone status. When compared to those of chow-fed controls, rates of synthesis of intestinal apoA-I and apoB-48 decreased 60-66% in hypothyroid animals and increased three- to fourfold after triiodothyronine (T3) administration. These changes were not accompanied by changes in mRNA abundance. By contrast, intestinal apoA-IV synthesis rates and mRNA abundance were both unaltered over the range of thyroid hormone manipulations tested. Hepatic apoA-I and apoA-IV synthesis rates decreased by 70-80% in hypothyroid animals, while synthesis rates and mRNA abundance increased coordinately six- to eightfold when hypothyroid rats were made hyperthyroid. Hepatic apoE synthesis rates increased twofold in hypothyroid rats and decreased sevenfold in hyperthyroid animals. ApoE mRNA abundance, however, was comparable in all groups. Hypothyroid animals had reduced synthesis rates of hepatic apoB-100 and apoB-48. After induction of hyperthyroidism, apoB-100 synthesis (studied from 5 to 60 min) was undetectable (less than 0.01%) without further change in apoB-48 synthesis and without alterations in either apoB mRNA abundance or transcript size. Despite undetectable hepatic apoB-100 synthesis rates in hyperthyroid animals, total plasma triglyceride secretion rates (after Triton WR-1339 injection) were normalized compared to a 50% decrease in hypothyroid rats. Taken together, the data provide evidence for tissue-specific, independent regulation of apolipoprotein gene expression in vivo. Furthermore, the data suggest that aspects of hepatic triglyceride assembly and secretion and apolipoprotein gene expression may be coordinately responsive to alterations in thyroid hormone status.

Quantification of Alpha-Fetoprotein and Beta-HCG in Testis Tumor Patients

Patients with testis tumor were investigated for serum and tissue levels of alpha-fetoprotein and beta-human chorionic gonadotropin (beta-HCG). The tissue immune peroxidase-antiperoxidase staining for the tumor marker was quantitated by computer-assisted immunohistophotometry and immuno-gamma ray histospectrometry. The results supported the general view that mostly polynuclear giant cells produce beta-HCG in 66% of nonseminoma cancer. This finding qualifies beta-HCG as relatively unspecific in the absence of chorioepithelial cells in the tumor. Discrepancies of tissue and serum beta-HCG values may be caused by deglycolysation of beta-HCG while penetrating the perivascular tissues. Alpha-fetoprotein (AFP) appears helpfully to discriminate the true seminoma cancer, which is constantly negative. Histologically pure seminoma which reacts for AFP therefore suggests sclerotic teratoma compartments. A constant finding is the significantly reduced synthesis rate of tumor markers in metastasis compared to primary tumor.

Antagonistic effects of progesterone on estradiol-induced synthesis and degradation of uterine glucose-6-phosphate dehydrogenase.

Estradiol administered daily to ovariectomized mature rats stimulates uterine glucose-6-phosphate dehydrogenase (G6PD) to maximum levels within 72 h, a stimulation that is nearly 14-fold greater than the preinduced levels. Twice daily injections of progesterone given with daily estradiol injections caused the increases in G6PD activity to plateau prematurely after 36 h at levels that were 38% of the maximum estrogeninduced value at 72 h. The mechanisms of this delayed antagonistic effect of progesterone were determined by comparing the rates of uterine G6PD synthesis and degradation at various times during the 72-h response to estradiol in the presence and absence of progesterone. Progesterone first caused a significant reduction in the estradiol-induced rate of synthesis of uterine G6PD after a lag of 24 h, and the inhibition continued until 72 h, at which time rates of synthesis in animals treated with both hormones were only slightly above the rates in untreated controls. Studies on the effect of indomethacin and sulfinpyrazone on the rates of synthesis of albumin and acute-phase α 1-acid glycoprotein by rat liver slices

The rates of synthesis of albumin and α 1-acid glycoprotein were determined in the presence of indomethacin and sulfinpyrazone. There were significant reductions in synthesis rates of both proteins in the presence of concentrations of 0.5 m m or greater in the case of both drugs tested. Incorporation studies showed that decreased synthesis rates were accompanied by inhibition of incorporation of radioactive precursors into albumin and α 1-acid glycoprotein and into total protein isolated from liver and incubation medium. Incorporation of [ 3H]orotic acid into liver RNA was also subject to inhibition by the drugs used in the present work as well as by salicylate and phenylbutazone, two drugs used in an earlier study. Synthesis of RNA followed by orotic acid incorporation was more sensitive to inhibition by the four drugs in question; substantial inhibition was observed in the presence of 0.25 m m or greater of indomethacin, sulfinpyrazone, and phenylbutazone, and in the presence of 2 m m or greater of salicylate.

The role of amino acids in the regulation of protein synthesis in perfused rat liver. I. Reduction in rates of synthesis resulting from amino acid deprivation and recovery during flow-through perfusion.

The role of perfusate amino acid concentrations in regulating rates of protein synthesis was investigated using the perfused rat liver. Livers from fed rats were perfused with a nonrecirculating medium and the incorporation of [3H]leucine into albumin and total protein was determined under conditions where the leucyl-tRNALeu and perfusate leucine specific activities were equal and constant. During perfusions of less than 1 h, rates of total protein synthesis were sensitive to the concentrations of amino acids in the perfusate. When no exogenous amino acids were provided, rates of synthesis of albumin and total protein were 40% of the maximal rates which were achieved when the medium was supplemented with 5 times the normal plasma concentrations of amino acids. However, rates of synthesis in livers perfused with amino acid-deficient medium rose with extension of the duration of perfusion to 95 min. The defect induced by amino acid deficiency did not appear to result from reductions in the charging of tRNA since no change in the quantities of amino acids bound to tRNA occurred in the amino acid-deficient perfusion. The recovery of protein synthesis with time was prevented by inhibitors of proteolysis suggesting a role for protein degradation in this phenomenon.
