Temperature and Preparation Time Intervals on Survival of Opisthorchis viverrini Metacercariae in Pickled Fish (Pla‐som)

This study involved two series of independent trials that investigated the storage temperature and time of raw fish to eliminate Opisthorchis viverrini metacercarial infection in Thai pickled fish. A total of 330 healthy silver barb fishes (30‐day posthatch) were infected with O. viverrini cercariae at an inoculation ratio of 50 cercariae/fish. After infection, the fish was reared for 3 months. The first trial was to evaluate the effect of fish storage temperature in the recovery from O. viverrini metacercariae. The infected fishes were randomly divided into five groups and stored at −20°C, 2°C, 4°C, and 8°C or room temperature (as a control) for 24 h. The results showed that at a storage temperature of −20°C, the fish initially had a reduced recovery rate from O. viverrini metacercariae. The second trial investigated the storage time for the recovery of infected fish stored at −20°C for 18, 24, 36, 48, or 60 h and used room temperature as a control group. The results revealed that storing infected fish at −20°C for 48 and 60 h had the lowest recovery rate from O. viverrini metacercariae at 0.00%. In conclusion, storing infected fish at a temperature of −20°C for a period of 48 h or longer could restrict the O. viverrini metacercaria recovery rate. These results were recommended as suitable conditions for the preparation of Pla‐som to avoid O. viverrini metacercaria contamination.

ITGB1-Expressing Effector Memory CD4 + T-Cell Response Associates with Clinical Recovery in Childhood Immune Thrombocytopenia

Background: The autoimmune bleeding disorder childhood immune thrombocytopenia (ITP) features antibody and T-cell immune responses against platelet self-antigens. This involves activation of B-cells, CD4 + T-cell help, and CD8 + T-cells. Novel prognostic biomarkers are needed to predict recovery and treatment responses. Compared to healthy children, changes in total lymphocytes, CD4 +, regulatory CD4 +, CD8 +T-cells, CD19 + B-cells, and NK cells were described in childhood ITP. Children with transient ITP were also found to have a fewer CD25 hi CD4 + T-cells than those who develop chronic ITP. In contrast, the platelet-stimulated peripheral blood mononuclear cells (PBMC) of children with chronic ITP produced more IL-2 than those of transient ITP. At present, none of these biomarkers have been validated for ITP prognosis, and consequently they are not in clinical use. This emphasizes the need for clinical-grade biomarkers to predict clinical responses in childhood ITP. Aims: To predict spontaneous recovery and IVIg response in newly diagnosed ITP by (1) validating previously suggested changes in immune cell frequencies, and (2) identifying novel immune cell subsets as predictors. Methods: Children with newly diagnosed ITP were randomized 1:1 to observation or IVIg treatment (TIKI trial). Recovery was determined by platelet counts according to IWG criteria 1, 4, 13, 26 and 52 weeks after the diagnosis. For validation, the CD4 +, regulatory CD4 + (CD25 +/CD127 lo), CD8 +, CD19 +, and NK cells were quantified at the time of diagnosis in a centralized laboratory by flow cytometry. For identification of novel predictors, CD4 + central (CD45RO + CD27 +) and effector memory (CD45RO + CD27 -) frequencies were determined. The primary clinical outcome was longitudinal recovery, either spontaneous or after IVIg (adjusted Cox-proportional hazard model). Secondary outcomes included the mean age-adjusted difference between transient/persistent and chronic ITP by multivariate regression, and comparison to age-appropriate healthy control data. Additionally, PBMC (N=6) were analyzed at diagnosis by single-cell RNA sequencing (scRNA-seq) combined with T- and B- cell receptor sequencing (scTCR-seq and scBCR-seq). Results: For validation of previously suggested predictors, the absolute CD3 +, CD4 +, CD8 +, CD19 +, and NK cell counts of newly diagnosed ITP patients were within the age-appropriate healthy reference range (N=158); they were not associated with recovery, and there were no age-adjusted differences between transient/persistent (N=139) and chronic ITP (N=19). The regulatory CD4 + T cell frequency was not reduced compared to age-appropriate reference data; not associated with recovery, and not different between transient/persistent and chronic ITP patients. For identification of novel predictors, we found that high CD4 +effector memory cells numbers were associated with a reduced recovery rate, with an adjusted hazard ratio for complete recovery over a one-year follow-up to be 0.55 (95% CI, 0.35 - 0.85; ≥ median; adjusted for age and treatment; N=150). Chronic ITP patients displayed a mean age-adjusted increase in effector memory CD4 +cells of 1.4 % (95% CI, 0.4 - 2.4; P= 0.005). The association with recovery was independent of a preceding infection, total leukocyte and lymphocyte counts, and the presence of anti-platelet IgG or IgM autoantibodies. ScRNA-seq analysis of 7965 PBMC also showed an effector memory CD4 + T-cell cluster that was expanded in chronic ITP, present in at a frequency of 8.1 ± 1.4 % (mean ± sem) in transient vs 14.8 ± 1.2 % in chronic ITP. This cluster also expressed high levels of fibronectin receptor integrin b1 ( ITGB1) interleukin 32 (IL32) interleukin 7 receptor (IL7R) andlymphotoxin beta (LTB). The effector memory phenotype of CD4 + ITGB1 + T-cells was confirmed by flow cytometry analysis in healthy individuals and ITP patients. Conclusions: Previously suggested changes in T-, B-, or NK cell frequencies could not be validated as predictors of spontaneous recovery or IVIg response. However, we identified the frequency of ITGB1-expressing effector memory CD4 + T cells as an independent predictor of spontaneous recovery and IVIg response. Thus, T cell phenotyping at the time of diagnosis may be suitable to determine prognosis and personalize treatment decisions in newly diagnosed childhood ITP.

Enhancing Viability of Human Embryonic Stem Cells during Cryopreservation via RGD-REP-Mediated Activation of FAK/AKT/FoxO3a Signaling Pathway.

Cryopreservation is a crucial method for long-term storage and stable allocation of human pluripotent stem cells (hPSCs), which are increasingly being used in various applications. However, preserving hPSCs in cryogenic conditions is challenging due to reduced recovery rates. To address this issue, the Arginine-Glycine-Aspartate (RGD) motif was incorporated into a recombinant elastin-like peptide (REP). Human embryonic stem cells (hESCs) were treated with REP containing RGD motif (RGD-REP) during suspension and cryopreservation, and the survival rate was analyzed. The underlying mechanisms were also investigated. The addition of RGD-REP to the cryopreservation solution improved cell survival and pluripotency marker expression. The improvement was confirmed to be due to the activation of the FAK-AKT cascade by RGD-REP binding to hESC surface interin protein, and consequent inhibition of FoxO3a. The inactivation of FoxO3a reduced the expression of apoptosis-related genes, such as BIM, leading to increased survival of PSCs in a suspension state. RGD-REP, as a ligand for integrin protein, improves the survival and maintenance of hPSCs during cryopreservation by activating survival signals via the RGD motif. These results have potential implications for improving the efficiency of stem cell usage in both research and therapeutic applications.

Study on the self-recovery performance of SMAF-ECC under cyclic tensile loading

The incorporation of superelastic shape memory alloy (SMA) fibers into engineered cementitious composites (ECC) has led to a novel composite material, SMAF-ECC, which possesses excellent deformation, energy dissipation, and self-recovery capabilities. To understand the self-recovery performance of this new composite material under tensile loading, this study conducted uniaxial cyclic tension tests on SMAF-ECC specimens to investigate stress, strain, crack width, strain recovery rate, and crack recovery rate, while also considering the primary influencing factors such as SMA fiber diameter and content. The results indicate that the addition of SMA fibers improves the ultimate strain and ultimate tensile strength of ECC; increasing fiber content effectively enhances the strain and crack recovery rates of SMAF-ECC specimens, with the maximum strain recovery rate and crack recovery rate of the tested specimens reaching 69% and 77%, respectively. Both too large and too small SMA fiber diameters lead to reduced recovery rates. In this experiment, when the fiber diameter was 0.5 mm, the strain and crack recovery rates of the specimens were the highest, with a crack recovery rate of 53%, approximately 1.7 times that of specimens with 1 mm and 0.2 mm diameter SMA fibers. These findings provide a reliable basis for establishing a theoretical model of the tensile mechanical properties of SMAF-ECC composite materials.

Current global population size, post-whaling trend and historical trajectory of sperm whales

The sperm whale lives in most deep ice-free waters of the globe. It was targeted during two periods of whaling peaking in the 1840’s and 1960’s. Using a habitat suitability model, we extrapolated estimates of abundance from visual and acoustic surveys to give a global estimate of 736,053 sperm whales (CV = 0.218) in 1993. Estimates of trends in the post-whaling era suggest that: whaling, by affecting the sex ratio and/or the social cohesion of females, reduced recovery rates well after whaling ceased; preferentially-targeted adult males show the best evidence of recovery, presumably due to recruitment from breeding populations; several decades post-whaling, sperm whale populations not facing much human impact are recovering slowly, but populations may be declining in areas with substantial anthropogenic footprint. A theta-logistic population model enhanced to simulate spatial structure and the non-removal impacts of whaling indicated a pre-whaling population of 1,949,698 (CV = 0.178) in 1710 being reduced by whaling, and then then recovering a little to about 844,761 (CV = 0.209) in 2022. There is much uncertainty about these numbers and trends. A larger population estimate than produced by a similar analysis in 2002 is principally due to a better assessment of ascertainment bias.

Analysis of ochratoxin A, aflatoxin B1 and its biosynthetic precursors in cheese – Method development and market sample screening

The presence of Penicillium and Aspergillus fungi during cheese production may lead to the formation of ochratoxin A, aflatoxins, as well as their partly mutagenic biosynthetic precursors. However, data about the presence of these toxins in final cheese is scarce. In the present study, a quantification method for the toxicologically highly relevant compounds ochratoxin A (OTA), aflatoxin B1 (AFB1), sterigmatocystin (STC), and versicolorin A (VerA) was developed and applied for a market screening of hard, semi-hard, and soft cheese samples. The developed method was based on a QuEChERs approach with dimethylsulfoxide as keeper reagent. This stable isotope dilution assay for OTA, AFB1 and STC achieves high sensitivity (limits of detection: 0.02 μg/kg for AFB1, 0.01 μg/kg for STC and 0.37 μg/kg for OTA). However, in certain hard cheese types, the toxins showed matrix binding effects, leading to reduced recovery rates. When applying the method to market samples, detectable amount of STC in eight out of ten grated hard cheese samples up to 4.99 μg/kg and OTA in three of these samples up to 26.56 μg/kg were revealed. Only three out of the 53 further samples contained detectable amounts of mycotoxin. Thus, it can be assumed that the state of maturation and the process of grating favours the presence of OTA and STC in cheese, based on the market screening of 62 market samples. More comprehensive data are required to draw more detailed conclusions of the parameters determining the susceptibility of certain cheese types for mycotoxin contamination.

Multi-year, post-harvesting impact assessment in a neotropical secondary Atlantic Forest

Subject to overexploitation in the past centuries, the Atlantic Forest is subject to very rigorous protection rules. However, the law is a controversial issue since landowners are not compensated for the limited choice of land use possibilities. We believe that, alternatively to a general timber harvest ban, sustainable forest management of the Atlantic Forest has the potential to generate income for the landowners while sustaining important ecological functions of the forest. Such choice would require better understanding of the potential effects of timber harvesting on the forest ecosystem and on species composition and succession over time. In this context, we assessed the harvesting impact of a conventional harvesting method (CM) and compared it to an alternative harvesting method (AM) in three different stands (stands A, B and C) of a secondary forest fragment in southern Brazil. Results from three comprehensive forest inventories over a period of two years were used to assess the timber harvesting impact. Measurements of species composition (i.e., number of species, life form, ecological group), forest structure (i.e., density, basal area, DBH, volume) and saplings density formed the basis of the impact assessment. The inventories were carried out before, immediately after and two years after harvesting. Intensities of damage on remnant trees immediately after harvesting and two years after harvesting were also measured. Before harvesting, a total of 114 tree species (trees, tree fern and palm tree) belonging to 49 families were identified in the study site’s three research stands. Palm trees and secondary species, such as Euterpe edulis, represented the majority of recruited individuals (DBH ≥ 5 cm) two years after harvesting. However, new saplings (DBH ≤ 5 cm) after two years were mainly pioneer woody tree species, such as Cecropia glaziovii and Schizolobium parahyba. On average, AM reduced damage to saplings by 5%. Most of the damages caused by CM were moderate to severe, while AM caused light to moderate damages. Binary logistic regression indicated dependency of the mortality rates on the independent variables “stand, stem and leaning damages.” On the other hand, the recovery rates of damaged trees were dependent on crown, stem and leaning damages. Therefore, two years after harvesting a higher mortality rate of low-dimensional trees was observed in stands with high density of smaller trees and high density of improvement felling. Although crown damages were not related to mortality rates, high intensity of crown damages reduced recovery rates over time.

Gene Inactivation with CRISPR/Cas9 to Study DNA Repair Mechanisms in Bdelloid Rotifers

Bdelloid rotifers are aquatic microinvertebrates with an efficient DNA repair system that allows them to recover from severe DNA damage caused by desiccation. While the underlying mechanisms of the DNA repair process in bdelloids are unknown, we have identified several candidate genes that possibly play a role. The objective of this project was to develop a method for using CRISPR/Cas9 genome editing as a tool for investigating the functions of candidate DNA repair genes in the bdelloid Adineta vaga. First, a single guide RNA (sgRNA) that targets the DNA Polymerase β (POLB) gene was synthesized. As a pre-validation step to determine the cleavage efficiency of the sgRNA, the sgRNA was incubated in vitro with the target gene and the Cas9 enzyme. Cleavage products were then analyzed by gel electrophoresis. To use the sgRNA for CRISPR/Cas9 genome editing of POLB in A. vaga, we first designed a single-stranded oligonucleotide donor (ssODN) to be used as the template for homology-directed repair. The 100-nucleotide ssODN was predicted to introduce a stop codon in POLB resulting in a truncated protein if integrated into the genome by homology-directed repair. Next, we carried out in vivo ­genome editing in A. vaga by electroporating embryos with sgRNA/Cas9 ribonucleoprotein complexes and the ssODN with a variety of concentrations of each component. To identify individuals that had undergone genome editing and contain an inactivated POLB gene, we used a combination of the Surveyor nuclease (for detecting mismatches) and PCR with mutation-specific primers. In subsequent assays, individuals with CRISPR-induced POLB mutations will be desiccated and their recovery rates can be quantified relative to a wild-type. The expectation is that a reduced recovery rate in mutants will correspond with the POLB gene being relevant in DNA repair. Understanding this mechanism of DNA repair will allow for a more generalized understanding beyond model organisms and could have implications for biomedical research.

Diet, Nutrition, Obesity, and Their Implications for COVID-19 Mortality: Development of a Marginalized Two-Part Model for Semicontinuous Data

BackgroundNutrition is not a treatment for COVID-19, but it is a modifiable contributor to the development of chronic disease, which is highly associated with COVID-19 severe illness and deaths. A well-balanced diet and healthy patterns of eating strengthen the immune system, improve immunometabolism, and reduce the risk of chronic disease and infectious diseases.ObjectiveThis study aims to assess the effect of diet, nutrition, obesity, and their implications for COVID-19 mortality among 188 countries by using new statistical marginalized two-part models.MethodsWe globally evaluated the distribution of diet and nutrition at the national level while considering the variations between different World Health Organization regions. The effects of food supply categories and obesity on (as well as associations with) the number of deaths and the number of recoveries were reported globally by estimating coefficients and conducting color maps.ResultsThe findings show that a 1% increase in supplementation of pulses reduced the odds of having a zero death by 4-fold (OR 4.12, 95% CI 11.97-1.42). In addition, a 1% increase in supplementation of animal products and meat increased the odds of having a zero death by 1.076-fold (OR 1.076, 95% CI 1.01-1.15) and 1.13-fold (OR 1.13, 95% CI 1.0-1.28), respectively. Tree nuts reduced the odds of having a zero death, and vegetables increased the number of deaths. Globally, the results also showed that populations (countries) who consume more eggs, cereals excluding beer, spices, and stimulants had the greatest impact on the recovery of patients with COVID-19. In addition, populations that consume more meat, vegetal products, sugar and sweeteners, sugar crops, animal fats, and animal products were associated with more death and less recoveries in patients. The effect of consuming sugar products on mortality was considerable, and obesity has affected increased death rates and reduced recovery rates.ConclusionsAlthough there are differences in dietary patterns, overall, unbalanced diets are a health threat across the world and not only affect death rates but also the quality of life. To achieve the best results in preventing nutrition-related pandemic diseases, strategies and policies should fully recognize the essential role of both diet and obesity in determining good nutrition and optimal health. Policies and programs must address the need for change at the individual level and make modifications in society and the environment to make healthier choices accessible and preferable.

557. Impact of Concomitant Hydroxychloroquine Use on Safety and Efficacy of Remdesivir in Moderate COVID-19 Patients

BackgroundRemdesivir (RDV) has been shown to shorten recovery time and was well tolerated in patients with severe COVID-19. Hydroxychloroquine (HQN) is an experimental treatment for COVID-19. Effects of coadministration of HQN with RDV have not been studied and are relevant given the long half-life (~22 days) of HQN. We report the impact of concomitant HQN and RDV use on clinical outcomes and safety in patients with moderate COVID-19.MethodsWe enrolled hospitalized patients with confirmed SARS-CoV-2 infection, oxygen saturation >94% on room air, and radiological evidence of pneumonia. Patients were randomized 1:1:1 to receive 5d or 10d of intravenous RDV once daily plus standard of care (SoC), or SoC only. We compared patients on concomitant HQN (HQNpos) vs not (HQNneg). Clinical recovery was evaluated using Cox proportional hazards. Covariate adjustment included age, sex, race, region, symptom duration, oxygen support status and obesity. Recovery and adverse events (AEs) were assessed through death, discharge, or d14.ResultsOf 584 patients, 199 (34%) received HQN (5d RDV: n=57 [30%]; 10d RDV, n=49 [25%]; SoC: n=93 [47%]). Through median follow-up of 13d (range 1-41d), HQNpos patients on 5d or 10d RDV had a lower recovery rate (adjusted HR [95% CI] 0.78 [0.59, 1.03], p=0.09) with longer median time to recovery (8 vs 6 days) compared to HQNneg. HQNpos compared to HQNneg patients in 5d RDV showed a trend of reduced recovery rate (HR: 0.69 [0.45,1.04], p=0.080); such an effect was not observed in 10d RDV or SoC (Table 1). More HQNpos than HQNneg patients had AEs in RDV (5/10d) or SoC arms evaluated separately, and all arms combined. This difference was significant for AEs and SAEs for all arms combined after covariate adjustment (Table 2).Table 1. Table 2. ConclusionIn moderate COVID-19 patients, concomitant HQN may delay recovery on RDV and showed no impact on recovery with SoC alone. Ultrasound-assisted leaching of spent lithium ion batteries by natural organic acids and H2O2

Ultrasound-assisted bioacid leaching was examined for the extraction of valuable metals from spent lithium ion batteries (LIBs). In this work, organic acids in lemon juice were used as the leaching agent together with H2O2. Three effective factors, namely solid/liquid (S/L) ratio, lemon juice percentage, and H2O2 volume percentage, were optimized using Response Surface Methodology (RSM). The optimal conditions were found to be 0.98% (w/v) S/L ratio, 57.8% (v/v) lemon juice and 8.07% (v/v) H2O2 in the leaching liquor, achieving recovery of 100% Li, 96% Co and 96% Ni. Furthermore, the individual effects of ultrasound, H2O2 and lemon juice on metal recovery were studied and the results showed that without H2O2 or lemon juice, the metal recovery rates decreased greatly while the absence of ultrasound reduced recovery rates to a much smaller extent, indicating that both H2O2 and lemon juice were essential in the leaching process. The effect of time on the metals recoveries was examined and results showed that Li and Co recovery reached 100% with the leaching time of 35 min. The modified shrinking core modeling results suggested that chemical reaction was the rate controlling step.

Psychological networks in clinical populations: investigating the consequences of Berkson's bias.

In clinical research, populations are often selected on the sum-score of diagnostic criteria such as symptoms. Estimating statistical models where a subset of the data is selected based on a function of the analyzed variables introduces Berkson's bias, which presents a potential threat to the validity of findings in the clinical literature. The aim of the present paper is to investigate the effect of Berkson's bias on the performance of the two most commonly used psychological network models: the Gaussian Graphical Model (GGM) for continuous and ordinal data, and the Ising Model for binary data. In two simulation studies, we test how well the two models recover a true network structure when estimation is based on a subset of the data typically seen in clinical studies. The network is based on a dataset of 2807 patients diagnosed with major depression, and nodes in the network are items from the Hamilton Rating Scale for Depression (HRSD). The simulation studies test different scenarios by varying (1) sample size and (2) the cut-off value of the sum-score which governs the selection of participants. The results of both studies indicate that higher cut-off values are associated with worse recovery of the network structure. As expected from the Berkson's bias literature, selection reduced recovery rates by inducing negative connections between the items. Our findings provide evidence that Berkson's bias is a considerable and underappreciated problem in the clinical network literature. Furthermore, we discuss potential solutions to circumvent Berkson's bias and their pitfalls.

Long-term hydrophilization of polydimethylsiloxane (PDMS) for capillary filling microfluidic chips

We present a simple and facile method for long-term preservation of hydrophilicity of oxygen plasma-hydrophilized poly (dimethylsiloxane) (PDMS) by cold storage. We show that storage under temperature of − 80 °C can maintain superhydrophilicity of plasma-exposed PDMS for at least 100 days. Storage at − 15 °C and at 22 °C room temperature (RT) is shown to exhibit, respectively, about half and full recovery of the original hydrophobicity after 100 days in storage. Furthermore, we investigated the implications of the cold storage for microfluidic applications, the capillary filling rate and the ability of the flow to bypass geometrical obstacles in a microfluidic channel. It is shown that the preservation of capillary filling properties of microchannels is in close agreement with the contact angle (CA) measurements and that the colder the storage temperature, the better the capillary filling capability of the channels is preserved. We ascribe the significantly reduced recovery rate to reduced thermally activated relaxation phenomena such as diminished diffusion of low molecular weight species (LMW) in the polymer matrix at colder temperatures. This is supported by ATR-FTIR measurements of the OH vibration band over time for samples stored at different temperatures.

Series of semi-Markov processes to model infrastructure resilience under multihazards

Civil infrastructure systems are subjected to multiple hazards, including natural and anthropogenic, that disrupt their function or the level of service offered. Estimating the function recovery of these systems (or how soon normalcy of operations will be restored) when subjected to repeated hazard events by considering the inter-event dependencies is an important problem in multihazard infrastructure resilience. However, this problem has been less addressed in the field. This paper proposes a series of semi-Markov processes model to capture the inter-event dependencies in infrastructure recovery when subjected to successive hazard events. Recovery after each new hazard event is represented by a unique semi-Markov process that models the reduced recovery rates and the increased recovery times caused by the system’s incomplete recovery from the preceding event. Two novel formulations of the inter-event dependency modeling, namely Maximal Effects Dependency (considers the worst impact of two successive hazard events) and Cumulative Effects Dependency (considers the aggregated impacts of two successive hazard events), are proposed and discussed. The model is demonstrated by considering the following applications: Three-state system subjected to deterministic and random occurrences of identical hazard events; and Multihazard resilience of a building in Charleston, SC, considering earthquake and hurricane hazards. Results indicate that considering inter-event dependencies in recovery modeling can lead to lesser-predicted resilience, thereby affecting resilience-based decision-making.

Terrestrial single-station analog for constraining the martian core and deep interior: Implications for InSight

We used a terrestrial single-station seismometer to quantify the uncertainty of InSight (INterior explorations using Seismic Investigations, Geodesy and Heat Transport) data for determining Martian core size. To mimic Martian seismicity, we formed a catalog using 917 terrestrial earthquakes, from which we randomly selected events. We stacked ScS amplitudes on modeled arrival times and searched for where ScS produced coherent seismic amplitudes. A core detection was defined by a coherent peak with small offset between predicted and user-selected arrival times. Iterating the detection algorithm with varying signal-to-noise (SNR) ranges and quantity of events determined the selection frequency of each model and quantified core depth uncertainty. Increasing the quantity of events reduced core depth uncertainty while increasing the recovery rate, while increasing event SNR had little effect. Including ScS2 multiples increased the recovery rate and reduced core depth uncertainty when we used low quantities of events. The most-frequent core depths varied by back azimuth, suggesting our method is sensitive to the presence of mantle heterogeneities. When we added 1° in source distance errors, core depth uncertainty increased by up to 11 km and recovery rates decreased by <5%. Altering epicentral distances by 25% added ~35 km of uncertainty and reduced recovery rates to <50% in some cases. From these experiments, we estimate that if InSight can detect five events with high location precision (<10% epicentral distance errors), that there is at least an 88% chance of core depth recovery using ScS alone with uncertainty in core depth approaching 18 km and decreasing as more events are located. Plain language summaryWe used a seismometer on Earth to study how well the NASA mission, InSight (INterior explorations using Seismic Investigations, Geodesy and Heat Transport) could detect the Martian core. We created a catalog of earthquakes from which we randomly selected events to mimic the seismic events (marsquakes) InSight is expected to detect. Several models of Earth's interior were tested to see which best predicted the arrival of seismic waves reflecting off the core. We found utilizing more events allowed us to determine the core size better than using a limited number of events. Using an additional seismic phase also helped us recover a more accurate core depth if there were not a lot of events. We were also able to find variations in the mantle by using events within certain location ranges. Since the InSight team may not be able to locate marsquakes as accurately as we locate earthquakes, we changed the location of the earthquakes. When changes in location were small, we were still able to determine the size of the core. Larger changes in location prevented us from finding an accurate core depth. If InSight detects at least five events with high location accuracy, we will be able to determine the Martian core size.

Factors Influencing Outcomes of Children Hospitalized with Acute Severe Malnutrition

Background: Nutrition is still a global health issue, and improving nutrition is among the targets of the Sustainable Developmental Goals (SDGs). One of Indonesia’s SDGs indicators is to reduce prevalence of malnutrition in children. Indonesia, as a developing country, implements a comprehensive malnutrition treatment program for children, including various aspects related to management of children with acute malnutrition. The purpose of this study was to identify factors associated with the outcome of children hospitalized with acute severe malnutrition. Methods: This cross sectional study enrolled 195 children with severe acute malnutrition, admitted to hospital from November 2016 to August 2017. Statistical Analysis was conducted using multivariable logistic regression to identify factors associated with the outcome. Adjusted Ratio with Confident Interval 95% and the value of P <0 .05 was considered significant. Result: A total of 195 children were diagnosed havings acute severe malnutrition. A history of complete immunization status (p value <0.001) was proved to be directly proportional to the recovery rate of patients with acute severe malnutrition. Provision of other types of antibiotic (p value 0.001) showed to increase recovery rates of patients with acute severe malnutrition. Several comorbid diseases reduced recovery rates such as pneumonia (Crude OR 0.619) tuberculosis (Crude OR 0.606) and HIV (p value 0.08). Conclusions: This study shows that although the provided treatments are appropriate with the standard treatment for severe malnutrition, they still encounter high levels of morbidity. Further large-scale studies should be conducted to develop better interventions and management of acute severe malnutrition, in order to achieve better outcome.

Study of the Effect of Clay Particles on Low Salinity Water Injection in Sandstone Reservoirs

The need for optimal recovery of crude oil from sandstone and carbonate reservoirs around the world has never been greater for the petroleum industry. Water-flooding has been applied to the supplement primary depletion process or as a separate secondary recovery method. Low salinity water injection is a relatively new method that involves injecting low salinity brines at high pressure similar to conventional water-flooding techniques, in order to recover crude oil. The effectiveness of low salinity water injection in sandstone reservoirs depends on a number of parameters such as reservoir temperature, pressure, type of clay particle and salinity of injected brine. Clay particles present on reservoir rock surfaces adsorb polar components of oil and modify wettability of sandstone rocks to the oil-wet state, which is accountable for the reduced recovery rates by conventional water-flooding. The extent of wettability alteration caused by three low salinity brines on oil-wet sandstone samples containing varying clay content (15% or 30%) and type of clay (kaolinite/montmorillonite) were analyzed in the laboratory experiment. Contact angles of mica powder and clay mixture (kaolinite/montmorillonite) modified with crude oil were measured before and after injection with three low salinity sodium chloride brines. The effect of temperature was also analyzed for each sample. The results of the experiment indicate that samples with kaolinite clay tend to produce higher contact angles than samples with montmorillonite clay when modified with crude oil. The highest degree or extent of wettability alteration from oil-wet to intermediate-wet state upon injection with low salinity brines was observed for samples injected with brine having salinity concentration of 2000 ppm. The increase in temperature tends to produce contact angles values lying in the higher end of the intermediate-wet range (75°–115°) for samples treated at 50 °C, while their corresponding samples treated at 25 °C produced contact angle values lying in the lower end of intermediate-wet range.

Improved Coral Population Estimation Reveals Trends at Multiple Scales on Australia’s Great Barrier Reef

The global erosion of biodiversity presents unique challenges for identifying major changes in population dynamics, establishing their causes, and managing and conserving affected ecosystems at broad spatial scales. Adaptive learning approaches connecting different spatial scales through the transfer of hierarchical information are powerful tools to address such challenges. Here, we use a Semi-Parametric Bayesian Hierarchical (SPa-BaH) model to estimate coral cover trajectories using 16 years of a broad-scale survey on Australia’s Great Barrier Reef (GBR). The spatiotemporal variability of coral populations has been considered by separating three-tiered spatial scales and allowing for alternating phases of increasing and decreasing in the estimation of their trajectories. Model estimates revealed coral cover trajectories that were highly variable according to location but that fairly consistently declined at a regional spatial scale. Notwithstanding this general trend, individual reefs within subregions in the central part of the GBR often displayed different trajectory types between sites separated by only a few hundred meters. These coral dynamics were also associated with reduced recovery rates in the Cairns and Swain subregions. Our study highlights the importance of accounting for local variation in coral cover when estimating the spatiotemporal trends in coral cover trajectories, in this case, at the GBR scale. By retaining information at different hierarchical spatial scales, our SPa-BaH model supports better estimation of large-scale coral cover trajectories. The quantitative approaches developed here can also be applied to other species with complex dynamics thereby enhancing estimations of their trajectories at local- and larger-scales and options for their management.

Effect of neglecting geothermal gradient on calculated oil recovery

Reduced recovery rate with time is a common challenge for most of the oil producing reservoirs. Water flooding is one of the most common methods used for enhanced oil recovery. Simulating water-flooding process is sometimes carried out without considering the effect of geothermal gradient, and an average temperature is assumed for all the grid blocks. However, the gradient plays a significant role on the reservoir fluid properties. So neglecting its effect might result in a large error in the calculated oil recovery results, especially for the thick reservoirs, which in theory can show significant variations in temperature with depth. In this paper, first, advancing the waterfront during injection into a geothermal oil reservoir is discussed. Then, the performance of considering either an average temperature or gradient temperature, are considered and compared with each other. The results suggest that assuming a fixed average reservoir temperature with no geothermal gradient, can lead to a pronounced error for calculated oil recovery.

The impact of comorbid personality difficulties on response to IAPT treatment for depression and anxiety

The UK's Improving Access to Psychological Therapies (IAPT) initiative provides evidence-based psychological interventions for mild to moderate common mental health problems in a primary care setting. Predictors of treatment response are unclear. This study examined the impact of personality disorder status on outcome in a large IAPT service. We hypothesised that the presence of probable personality disorder would adversely affect treatment response. MethodWe used a prospective cohort design to study a consecutive sample of individuals (n = 1249). ResultsHigher scores on a screening measure for personality disorder were associated with poorer outcome on measures of depression, anxiety and social functioning, and reduced recovery rates at the end of treatment. These associations were not confounded by demographic status, initial symptom severity nor number of treatment sessions. The presence of personality difficulties independently predicted reduced absolute change on all outcome measures. ConclusionsThe presence of co-morbid personality difficulties adversely affects treatment outcome among individuals attending for treatment in an IAPT service. There is a need to routinely assess for the presence of personality difficulties on all individuals referred to IAPT services. This information will provide important prognostic data and could lead to the provision of more effective, personalised treatment in IAPT.
