Aflatoxins Contamination in Feed Commodities: From Occurrence and Toxicity to Recent Advances in Analytical Methods and Detoxification.

Synthesized by the secondary metabolic pathway in Aspergilli, aflatoxins (AFs) cause economic and health issues and are culpable for serious harmful health and economic matters affecting consumers and global farmers. Consequently, the detection and quantification of AFs in foods/feeds are paramount from food safety and security angles. Nowadays, incessant attempts to develop sensitive and rapid approaches for AFs identification and quantification have been investigated, worldwide regulations have been established, and the safety of degrading enzymes and reaction products formed in the AF degradation process has been explored. Here, occurrences in feed commodities, innovative methods advanced for AFs detection, regulations, preventive strategies, biological detoxification, removal, and degradation methods were deeply reviewed and presented. This paper showed a state-of-the-art and comprehensive review of the recent progress on AF contamination in feed matrices with the intention of inspiring interests in both academia and industry.

Thiazolidinediones: Recent Development in Analytical Methodologies.

The instrumental analytical methods that have been developed and utilized for the determination of thiazolidinedione in bulk medications, formulations and biological fluids have been reviewed after an in-depth analysis of the literature published in a variety of analytical and pharmaceutical chemistry-related journals. The approaches covered by this research, which covers the years 2001-2022, include complex methods for analysis, chromatographic techniques and spectrometric analytical procedures. The mobile phase, flow rate, sample matrix, wavelength and other factors identified in the literature were just a few of the parameters used to evaluate thiazolidinediones. The present review focuses on the published analytical techniques for thiazolidinedione analysis that have been previously identified in the literature. The specified outcomes followed extensive learning, and the most recent advances in analytical methods for the identification of pioglitazone, pioglitazone HCl, rosiglitazone, rosiglitazone maleate and lobeglitazone were reviewed. Additionally, this article briefly discusses features of analytical discovery on thiazolidinediones, which will enable readers to access all discoveries in one place with precise outcomes.

Deciphering Protein O-GalNAcylation: Method Development and Disease Implication.

Mucin-type O-glycosylation is an important protein post-translational modification that is abundantly expressed on cell surface proteins. Protein O-glycosylation plays a variety of roles in cellular biological functions including protein structure and signal transduction to the immune response. Cell surface mucins are highly O-glycosylated and are the main substance of the mucosal barrier that protects the gastrointestinal or respiratory tract from infection by pathogens or microorganisms. Dysregulation of mucin O-glycosylation may impair mucosal protection against pathogens that can invade cells to trigger infection or immune evasion. Truncated O-glycosylation, also known as Tn antigen or O-GalNAcylation, is highly upregulated in diseases such cancer, autoimmune disorders, neurodegenerative diseases, and IgA nephropathy. Characterization of O-GalNAcylation helps decipher the role of Tn antigen in physiopathology and therapy. However, the analysis of O-glycosylation, specifically the Tn antigen, remains challenging due to the lack of reliable enrichment and identification assays compared to N-glycosylation. Here, we summarize recent advances in analytical methods for O-GalNAcylation enrichment and identification and highlight the biological role of the Tn antigen in various diseases and the clinical implications of identifying aberrant O-GalNAcylation.

In this paper, we will describe recent advances in analytical methods to construct exact solutions of the Skyrme model (and its generalizations) representing inhomogeneous Hadronic condensates living at finite Baryon density. Such novel analytical tools are based on the idea to generalize the well-known spherical hedgehog ansatz to situations (relevant for the analysis of finite density effects) in which there is no spherical symmetry anymore. Besides the intrinsic mathematical interest to find exact solutions with nonvanishing Baryonic charge confined to a finite volume, this framework opens the possibility to compute important physical quantities which would be difficult to compute otherwise.

In recent times SARS-CoV-2 pandemic, public health experts have slated testing, tracking infected people, as well as tracing their contacts as an efficient method to reduce the viral spread. Numerous diagnostic approaches are reported for detecting the coronavirus in public health, clinical and research laboratories. Most tests detect the infection directly by detecting the viral RNA while other test may indirectly detect the host antibodies in the course of detecting the infection. A diagnostic test during the pandemic should aid in making proper clinical decision within a short time frame. In this review, we describe the various techniques involved in sample collection from nasal, throat and sputum specimens, standard and developed methods for diagnosis of covid 19 such as the polymerase chain reaction (RT-PCR), CRISPR-based assays and Serological and immunological assays. General information on selected electroactive antiviral Covid-19 drugs have been highlighted including: Favipiravir (FAV), Remdesivir (REM), Lopinavir + Ritonavir (LOP + RIT), Hydroxychloroquine (HCQ) and Chloroquine (CQ), Ribavirin (RIB) and Sofosbuvir. This review presents the major standard, commercial and designed methods for detecting SARS-CoV-2 and their analytical performance. Also, chromatographic techniques have been highlighted as a rapid examination for SARS-CoV-2 identification, and some recent advances and emerging trends in analytical methods for covid-19 diagnosis have been addressed.

Demystifying racemic natural products in the homochiral world.

Natural products possess structural complexity, diversity and chirality with attractive functions and biological activities that have significantly impacted drug discovery initiatives. Chiral natural products are abundant in nature but rarely occur as racemates. The occurrence of natural products as racemates is very intriguing from a biosynthetic point of view; as enzymes are chiral molecules, enzymatic reactions generating natural products should be stereospecific and lead to single-enantiomer products. Despite several reports in the literature describing racemic mixtures of stereoisomers isolated from natural sources, there has not been a comprehensive review of these intriguing racemic natural products. The discovery of many more natural racemates and their potential enzymatic sources in recent years allows us to describe the distribution and chemical diversity of this ‘class of natural products’ to enrich discussions on biosynthesis. In this Review, we describe the chemical classes, occurrence and distribution of pairs of enantiomers in nature and provide insights about recent advances in analytical methods used for their characterization. Special emphasis is on the biosynthesis, including plausible enzymatic and non-enzymatic formation of natural racemates, and their pharmacological significance.

Recent Advances in Analytical Methods for the Detection of Olive Oil Oxidation Status during Storage along with Chemometrics, Authenticity and Fraud Studies.

Olive oil is considered to be a food of utmost importance, especially in the Mediterranean countries. The quality of olive oil must remain stable regarding authenticity and storage. This review paper emphasizes the detection of olive oil oxidation status or rancidity, the analytical techniques that are usually used, as well as the application and significance of chemometrics in the research of olive oil. The first part presents the effect of the oxidation of olive oil during storage. Then, lipid stability measurements are described in parallel with instrumentation and different analytical techniques that are used for this particular purpose. The next part presents some research publications that combine chemometrics and the study of lipid changes due to storage published in 2005–2021. Parameters such as exposure to light, air and various temperatures as well as different packaging materials were investigated to test olive oil stability during storage. The benefits of each chemometric method are provided as well as the overall significance of combining analytical techniques and chemometrics. Furthermore, the last part reflects on fraud in olive oil, and the most popular analytical techniques in the authenticity field are stated to highlight the importance of the authenticity of olive oil.

Recent Advances in Analytical Methods for Determination of Polyphenols in Tea: A Comprehensive Review.

Polyphenols, the most abundant components in tea, determine the quality and health function of tea. The analysis of polyphenols in tea is a topic of increasing interest. However, the complexity of the tea matrix, the wide variety of teas, and the difference in determination purposes puts forward higher requirements for the detection of tea polyphenols. Many efforts have been made to provide a highly sensitive and selective analytical method for the determination and characterization of tea polyphenols. In order to provide new insight for the further development of polyphenols in tea, in the present review we summarize the recent literature for the detection of tea polyphenols from the perspectives of determining total polyphenols and individual polyphenols in tea. There are a variety of methods for the analysis of total tea polyphenols, which range from the traditional titration method, to the widely used spectrophotometry based on the color reaction of Folin–Ciocalteu, and then to the current electrochemical sensor for rapid on-site detection. Additionally, the application of improved liquid chromatography (LC) and high-resolution mass spectrometry (HRMS) were emphasized for the simultaneous determination of multiple polyphenols and the identification of novel polyphenols. Finally, a brief outline of future development trends are discussed.

Characterization of the Structural Diversity and Structure-Specific Behavior of Oxidized Phospholipids by LC-MS/MS.

Polyunsaturated fatty acids (PUFAs), esterified to phospholipids, are susceptible to oxidation. They form oxidized phospholipids (OxPLs) by oxygenases or reactive oxygen species (ROS), or both. These OxPLs are associated with various diseases, such as atherosclerosis, pulmonary injuries, neurodegenerative diseases, cancer, and diabetes. Since many types of OxPLs seem to be generated in vivo, precise determination of their structural diversity is required to understand their potential structure-specific functions. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) is a powerful method to quantitatively measure the structural diversity of OxPLs present in biological samples. This review outlines recent advances in analytical methods for OxPLs and their physiological relevance in health and diseases.

Analytical Methods for Os Isotope Ratios and Re-PGE Mass Fractions in Geological Samples.

The recent advances in analytical methods of Re-Os and PGE in geological materials including sample dissolution, chemical separation, mass spectrometric determinations, as well as the developments of matrix-matched reference materials for data quality control are thoroughly reviewed. Further, the in-situ measurement methods for Re-PGE mass fractions and 187Os/188Os ratios, as well as the measurement methods for stable isotope ratios of Re and PGE are also briefly reviewed. This review stands as a comprehensive reference for researchers to consider in the development of measurement methods for Re-PGE mass fractions and 187Os/188Os ratios in geological materials.

Recent Advances in Geochemical Paleo-Oxybarometers

Knowledge of how and why oxygenic photosynthesis, eukaryotes, metazoans, and humans evolved on Earth is important to the search for complex life outside our Solar System. Hence, one grand challenge for modern geoscience research is to reconstruct the story of how Earth's environment and life coevolved through time. A critical part of the effort to understand Earth's story is the use of geochemical signatures from the rock record—paleo-oxybarometers—to constrain atmosphere and ocean O2 levels and their spatiotemporal variations. Recent advances in analytical methods and improved knowledge of elemental and isotopic (bio)geochemical cycles have fostered development and refinement of many paleo-oxybarometers. Each offers its unique perspective and challenges toward obtaining robust (semi)quantitative O2 estimates. Overall, these paleo-oxybarometers have provided critical new insights but have also spurred new debates about Earth's oxygenation and its impact on biological evolution (and vice versa). Integrated approaches with multiple paleo-oxybarometers are now more critical than ever. ▪ Paleo-oxybarometers estimate atmosphere or ocean O2 levels, providing insight on how Earth's environment and life coevolved over time. ▪ Recent conceptual, analytical, and modeling advances, aided by studies on modern environments, have improved quantitative O2 estimates. ▪ Atmosphere and ocean paleo-oxybarometers reveal a complex history of dynamic O2 fluctuations since oxygenic photosynthesis evolved. ▪ Further improvements in the accuracy and robustness of atmosphere-ocean O2 estimates will require more integrated approaches.

Composition of Amino Acids in Foodstuffs for Humans and Animals.

Amino acids (AAs) are the building blocks of proteins that have both structural and metabolic functions in humans and other animals. In mammals, birds, fish, and crustaceans, proteinogenic AAs are alanine, arginine, asparagine, aspartate, cysteine, glutamate, glutamine, glycine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, proline, serine, threonine, tryptophan, tyrosine, and valine. All animals can synthesize de novo alanine, asparagine, aspartate, glutamate, glutamine, glycine, proline, and serine, whereas most mammals (including humans and pigs) can synthesize de novo arginine. Results of extensive research over the past three decades have shown that humans and other animals have dietary requirements for AAs that are synthesizable de novo in animal cells. Recent advances in analytical methods have allowed us to determine all proteinogenic AAs in foods consumed by humans, livestock, poultry, fish, and crustaceans. Both plant- and animal-sourced foods contain high amounts of glutamate, glutamine, aspartate, asparagine, and branched-chain AAs. Cysteine, glycine, lysine, methionine, proline, threonine, and tryptophan generally occur in low amounts in plant products but are enriched in animal products. In addition, taurine and creatine (essential for the integrity and function of tissues) are absent from plants but are abundant in meat and present in all animal-sourced foods. A combination of plant- and animal products is desirable for the healthy diets of humans and omnivorous animals. Furthermore, animal-sourced feedstuffs can be included in the diets of farm and companion animals to cost-effectively improve their growth performance, feed efficiency, and productivity, while helping to sustain the global animal agriculture (including aquaculture).

A Partial Charred Wooden Bowl From Aztalan (47JE1), Wisconsin

Abstract Fragments of a charred wooden bowl were recovered from Aztalan during excavations by the State Historical Society of Wisconsin (SHSW) in 1964. Recent advances in analytical methods facilitated a multidimensional study of these fragments. Radiocarbon-dated to cal AD 994–1154 and found in association with Late Woodland, Mississippian, and hybrid forms of ceramics, the bowl augments our understanding of perishable technologies in these cultural contexts. 3-D models of the fragments allow for a virtual reconstruction of a portion of the bowl, which was carved from a solid piece of ash. Strontium isotope analysis of the wood indicates that the bowl was manufactured from wood locally available to the people at Aztalan.

Recent advances in analytical methods for the determination of citrinin in food matrices.

Citrinin is a toxic small organic molecule produced as a secondary metabolite by fungi types Penicillium, Monascus and Aspergillus and is known to contaminate various food commodities during postharvest stages of food production. During the last 10 years, most reported methods for citrinin analysis employed enzyme-linked immunosorbent assays or high-performance liquid chromatography. Over this same time period, liquid extraction, solid-phase extraction, dispersive liquid-liquid microextraction and QuEChERS were the most cited sample preparation and clean-up methods. In this review the advantages and disadvantages of the various sample preparation, separation and detection methods for citrinin analysis over the last decade are evaluated. Furthermore, current trends, emerging technologies and the future prospects of these methods are discussed.

LIGNOCELLULOSIC BIOMASS CHARACTERISTICS FOR BIOENERGY APPLICATION: AN OVERVIEW

Considering world energy demand and its security, much attention has been focused on identifying promising lignocellulosic substrates to generate a variety of downstream fuels and chemicals. In response, a recent key research area has centered on hydrolysis of cell wall polysaccharides by various physical, chemical and biological means. Due to their complex chemical and molecular structure, there is a substantial knowledge gap in understanding of biomass recalcitrance and its relationship with cell wall structure. It has delayed the response in meeting the target to achieve economically feasible large-scale conversion of lignocellulosic biomass to biofuel and market entrance. Biomass of wheat straw, rice straw, corn stover, sugarcane bagasse and switchgrass has been characterized to assess their potential for bioenergy application. The mechanism involved in digestibility of lignocellulosic biomass is complex since it is hindered by many physico-chemical, structural and compositional factors. This paper highlights recent advances in analytical methods employed for physico-chemical characterization of lignocellulosic biomass and its deconvolution to maximize the benefit of biomass as a material and energy feedstock.

Advances, challenges and opportunities for point-of-need screening of mycotoxins in foods and feeds.

The assurance of food and feed safety, including the identification and effective monitoring of multiple biological and chemical hazards, is a major societal challenge, given the increasing pace at which food commodities are demanded, produced and traded across the globe. Within this context, mycotoxins are globally widespread secondary fungal metabolites, which can contaminate crops either in the field or during storage and have serious human and animal health impacts such as carcinogenic, teratogenic and hepatotoxic effects. Therefore, their presence in a wide range of foods and feeds is strictly regulated, particularly in the European Union. In order to perform effective and routine monitoring of mycotoxin levels in the field prior to further processing, during transport or during processing, rapid, simple, portable and sensitive means of screening of regulated mycotoxins are in high demand. This review focuses on (1) discussing the relevance of mycotoxins and the standard approaches for their sampling and monitoring; and (2) compiling and discussing recent advances in miniaturized analytical tools for mycotoxin detection. This provides insights into current research efforts and opportunities to develop a truly integrated and fit-for-purpose analytical tool, suitable for use at critical points of the food, feed and raw material processing and distribution chains.

Recent advances in analytical methods for the therapeutic drug monitoring of immunosuppressive drugs.

Therapeutic drug monitoring (TDM) of immunosuppressive drugs (ISDs) with a narrow therapeutic index is an increasingly popular tool for minimizing drug toxicity while maximizing the prevention of graft loss and organ rejection. This review focuses on trends regarding analytical methods for the TDM of ISDs since 2011. The five most commonly prescribed immunosuppressive medications are critically reviewed: cyclosporine A, tacrolimus, sirolimus (rapamycin), everolimus, and mycophenolic acid. This review introduces the general background of TDM and ISDs and presents the recent developments in using liquid chromatography-tandem mass spectrometry (LC-MS/MS) and immunoassays for the TDM of ISDs. Finally, a future perspective for these analytical methods is briefly discussed.

Many aspects of the way plants protect themselves against pathogen attack, or react upon such an attack, are realized by metabolites. The ambitious aim of metabolomics, namely the identification and annotation of the entire cellular metabolome, still poses a considerable challenge due to the high diversity of the metabolites in the cell. Recent advances in analytical methods and data analysis have resulted in improved sensitivity, accuracy, and capacity, allowing the analysis of several hundreds or even thousands of compounds within one sample. Investigators have only recently begun to acknowledge and harness the power of metabolomics to elucidate key questions in the study of plant biotic interactions; we review trends and developments in the field.
