Reactions of tetrasulfur tetranitride with aryl dibromomethyl ketones: one-pot synthesis of 3-aroylformamido-4-aryl-1,2,5-thiadiazoles and their reactions

Heating of dibromomethyl aryl ketones with tetrasulfur tetranitride (S4N4) at 115 °C without a solvent gave 3-aroylformamido-4-aryl-1,2-5-thiadiazoles 8 as major products (12–71%) and 3,5-diaroyl-1,2,4-thiadiazoles 2 as minor products in certain cases. The structures of compounds 8 were determined based on the X-ray analysis of 3-benzoylformamido-4-phenyl-1,2,5-thiadiazole 8a and comparison with an authentic sample of comound 8a, as well as all the spectroscopic and analytical data of compounds 8. Oxidation of compounds 8 with m-chloroperbenzoic acid in chloroform at room temperature gave compounds 2(0–66%), whereas reduction of compounds 8 with sodium boranuide in a mixture of chloroform–ethanol at room temperature gave 3-amino-4-aryl-1,2,5-thiadiazoles 10(71–93%). Treatment of 3-(3-nitrobenzoylformamido)-4-(3-nitrophenyl)-1,2,5-thiadiazole 8d with either sodium hydroxide in aqueous p-dioxane at reflux or sodium hydride in chloroform at room temperature gave 3-amino-4-(3-nitrophenyl)-1,2,5-thiadiazole 10d in 56 and 80% yield, respectively.

Reactions of tetrasulfur tetranitride with alkyl aryl ketoximes: synthesis of 3-aryl- and 3-alkyl-4-aryl-1,2,5-thiadiazoles

Tetrasulfur tetranitride (S4N4) was treated with various alkyl aryl ketoximes having two hydrogens at the α-carbon atom to the oxime functionality in p-dioxane at reflux to give 3-substituted and 3,4-disubstituted 1,2,5-thiadiazoles in moderate yields. Reaction with isobutyrophenone oxime under the same conditions did not give the 1,2,5-thiadiazole derivative. On the other hand, the reaction with cyclohexanone oxime in p-dioxane and toluene afforded 4,5-dihydrobenzo[1,2-c:3,4-c′]bis[1,2,5]thiadiazole in 6 and 7% yield, respectively.

Reactions of tetrasulfur tetranitride with bromomethyl ketones. One pot synthesis of 3,5‐diaroyl‐ and 3,5‐diacyl‐1,2,4‐thiadiazoles

AbstractReactions of tetrasulfur tetranitride (S4N4) with aryl and alkyl bromomethyl ketones 1 in chlorobenzene at reflux temperature gave 3,5‐diaroyl‐ and 3,5‐diacyl‐1,2,4‐thiadiazoles 2 in 17‐60% yields. No 1,2,5‐thiadiazoles were detected. By heating of the two reactants at 115° without the solvent were also obtained 2 in 5‐13% yields. Hydrolysis of 2 with sodium hydroxide in a mixture of ethanol, ethyl acetate, and water (v:v, 4:2:1) at 75° to 85° afforded the heretofore inaccessible 3‐aroyl‐ and 3‐acyl‐1,2,4‐thiadiazoles 3 in 17‐79% yields.

Abstract Tetrasulfur tetranitride (N4S4) reacted with 2-naphthols to afford naphtho [1,2-c][1,2,5] thiadiazoles (13) in high yields, while 1-naphthaols gave naphtho[1,2-c:3,4-c′]bis[1,2,5]thiadiazoles as a major product, together with a small amount of 13. In the reactions with naphthalenediols such as 1,5-, 2,6-, 2,7-, 1,6-, and 1,7-diol, naphtho[1,2-c:5,6-c′]- or naphtho[1,2-c:7,8-c′]bis[1,2,5]thiadiazoles were obtained in varying yields. In some cases, naphtho[1,2-c:3,4-c′:5,6-c″]tris[1,2,5]thiadiazole was formed as a by-product. In reactions with 5-isoquinolinol, 8-quinolinoles, 9H-carbazol-2-ol, and dibenzofuran-2-ol, one and/or two 1,2,5-thiadiazole-ring-fused heteroaromatic compounds were obtained.

Abstract Benzotris- (3) and benzobis[1,2,5]thiadiazole (4) were prepared in a moderate yield, respectively, by the reaction of tetrasulfur tetranitride (5) with halocatechols (6) and -resorcinols (7). The reduction of 3 and 4 with Sn powder in a mixture of concentrated hydrochloric acid and dioxane gave benzenehexamine (1) and 1,2,3,4-benzenetetramine (2a) and its 5-methyl derivative (2b) in good yields, as a stable tri- (1·3HCl) and dihydrochloride (2·2HCl), respectively.

The scope and limitations of the above reaction were investigated and the evidences suggesting the radical anion mechanism are presented.
