High-Dose Linoleic Acid Activated JAK2-STAT3 Signaling Pathway Involved in Cytokine Production and Lipogenesis in Pancreatic Exocrine Cells.

Linoleic acid has been implicated in the pathogenesis of acute pancreatitis. However, molecular mechanisms underlying adverse effects of large-dose linoleic acid remain unclear. Current study aimed to explore the impact of high-dose linoleic acid on the activation of Janus kinase 2 (JAK2)-signal transducers and activators of transcription 3 (STAT3) pathway, cytokine production, and lipogenesis in pancreatic exocrine cells. MTT assay was used to detect the viability of AR42J rat pancreatic exocrine cells, and lactate dehydrogenase assay was utilized to detect cytotoxicity. Concentrations of interleukin (IL)-6 and tumor necrosis factor (TNF)-α were detected by ELISA, and protein expression of JAK2, p-JAK2, STAT3, p- STAT3, and fatty acid synthase (FAS) was examined by Western Blot. The impact of highdose linoleic acid on JAK2-STAT3 pathway was also examined when JAK2 was inhibited by AG490, and STAT3 expression was interrupted by siRNA. The cell viability of AR42J rat pancreatic exocrine cells was inhibited, and cytotoxicity was increased by high-dose linoleic acid. JAK2 and STAT3 proteins in pancreatic exocrine cells were activated by high-dose linoleic acid via phosphorylation and nuclear localization of phosphorylated STAT3. Moreover, the expression of downstream proteins in JAK2-STAT3 pathway (IL-6, TNF-α and FAS) was up-regulated by high-dose linoleic acid. The increased levels of IL-6 and TNF-α caused by high-dose linoleic acid were attenuated by JAK2 inhibitor AG490. p-JAK2 protein was up-regulated, whereas p-STAT3, STAT3 and FAS proteins were down-regulated by high-dose linoleic acid in the presence of STAT3-siRNA. The cytotoxicity was increased and JAK2-STAT3 signaling pathway was activated by high-dose linoleic acid through cytokine production and lipogenesis in rat pancreatic exocrine cells.

Immunocytochemistry of GLUT2, uptake of fluorescent desnitroso-streptozotocin analogs and phosphorylation of D-glucose in INS-1E cells

The non-invasive imaging of GLUT2-expressing cells remains a challenge. As streptozotocin, and similarly alloxan, may be transported into cells by GLUT2, the major aim of the present study was to assess the possible use of fluorescent desnitroso-streptozotocin analogs for in vitro labeling of GLUT2-expressing cells. INS-1E cells, human embryonic kidney (HEK) cells, rat isolated pancreatic islets, rat hepatic cells, rat exocrine pancreatic cells and tumoral insulin-producing BRIN-BD11 cells were incubated in the presence of two distinct fluorescent desnitroso-streptozotocin analogs, probes A and B. The immunocytochemistry of GLUT2 in INS-1E cells and the phosphorylation of D-glucose by INS-1E cell homogenates were also examined. The uptake of probes A and B (12.0 µM) by INS-1E cells yielded apparent intracellular concentrations approximately one order of magnitude higher than the extracellular concentration. The two probes differed from one another by the absolute values for their respective uptake and time course, but not so by the pattern of their concentration dependency. Comparable results were recorded in HEK cells, rat isolated pancreatic islets and hepatocytes. Vastly different findings were recorded, however, in rat exocrine pancreatic cells, which do not express GLUT2. Moreover, an unusual concentration dependency for the uptake of each probe was observed in tumoral BRIN-BD11 cells. It is proposed that suitable fluorescent desnitroso-streptozotocin analogs may be used to label GLUT2-expressing cells.

Cannabinoid agonist WIN55,212 in vitro inhibits interleukin-6 (IL-6) and monocyte chemo-attractant protein-1 (MCP-1) release by rat pancreatic acini and in vivo induces dual effects on the course of acute pancreatitis

Cannabinoids (CBs) evoke their effects by activating the cannabinoid receptor subtypes CB1-r and CB2-r and exert anti-inflammatory effects altering chemokine and cytokine expression. Various cytokines and chemokines are produced and released by rodent pancreatic acini in acute pancreatitis. Although CB1-r and CB2-r expressed in rat exocrine pancreatic acinar cells do not modulate digestive enzyme release, whether they modulate inflammatory mediators remains unclear. We investigated the CB-r system role on exocrine pancreas in unstimulated conditions and during acute pancreatitis. We evaluated in vitro and in vivo changes induced by WIN55,212 on the inflammatory variables amylasemia, pancreatic edema and morphology, and on acinar release and content of the cytokine interleukin-6 (IL-6) and chemokine monocyte chemo-attractant protein-1 (MCP-1) in untreated rats and rats with caerulein (CK)-induced pancreatitis. In the in vitro experiments, WIN55,212 (10(-6) mol L(-1)) inhibited IL-6 and MCP-1 release from acinar cells of unstimulated rats and after CK-induced pancreatitis. In vivo, when rats were pretreated with WIN55,212 (2 mg kg(-1), intraperitoneally) before experimentally-induced pancreatitis, serum amylase, pancreatic edema and IL-6 and MCP-1 acinar content diminished and pancreatic morphology improved. Conversely, when rats with experimentally-induced pancreatitis were post-treated with WIN55,212, pancreatitis worsened. These findings provide new evidence showing that the pancreatic CB1-r/CB2-r system modulates pro-inflammatory factor levels in rat exocrine pancreatic acinar cells. The dual, time-dependent WIN55,212-induced changes in the development and course of acute pancreatitis support the idea that the role of the endogenous CB receptor system differs according to the local inflammatory status.

Plasticity in the adult rat pancreas: transdifferentiation of exocrine to hepatocyte-like cells in primary culture.

Under certain experimental conditions, hepatocytes can arise in the pancreas. It has been suggested that the pancreas retains a source of hepatocyte progenitor cells. However, such cells have not been yet identified in the adult pancreas. We describe here the transdifferentiation of primary rat pancreatic exocrine cells into hepatocyte-like cells during 5 days of tissue culture in the presence of dexamethasone (DX). Using reverse-transcription polymerase chain reaction and immunocytochemistry, it was observed that DX treatment induced albumin RNA and protein expression in the cells. Coexpression of albumin and amylase, and the absence of cell proliferation, demonstrated a direct transdifferentiation of acinar cells to hepatocytic cells. CCAAT enhancer-binding protein-ss protein, a liver-enriched transcription factor that is considered to be the master switch in pancreatohepatic transdifferentiation, and alpha-fetoprotein were markedly upregulated in the cells after treatment with DX. We compared transcriptional profiles of freshly isolated exocrine cells and DX-treated cells using oligonucleotide microarrays and found that multiple liver-specific genes are induced along with albumin, and that certain pancreatic genes are downregulated in the DX-treated cells. In conclusion, these observations support the notion of plasticity in the adult pancreas and that exocrine cells can be reprogrammed to transdifferentiate into other cell types such as hepatocytes.

The ER to Golgi interface is the major concentration site of secretory proteins in the exocrine pancreatic cell.

By using quantitative immuno-electron microscopy of two-sided labeled resin sections of rat exocrine pancreatic cells, we have established the relative concentrations of the secretory proteins amylase and chymotrypsinogen in the compartments of the secretory pathway. Their total concentration over the entire pathway was approximately 11 and approximately 460 times, respectively. Both proteins exhibited their largest increase in concentration between the endoplasmic reticulum and cis-Golgi, where they were concentrated 3-4 and 50-70 times, respectively. Over the further pathway, increases in concentration were moderate, albeit two times higher for chymotrypsinogen than for amylase. From trans-Golgi to secretory granules, where the main secretory protein concentration is often thought to occur, relatively small concentration increases were observed. Additional observations on a third secretory protein, procarboxypeptidase A, showed a concentration profile very similar to chymotrypsinogen. The relatively high concentration of amylase in the early compartments of the secretory route is consistent with its exceptionally slow intracellular transport. Our data demonstrate that secretory proteins undergo their main concentration between the endoplasmic reticulum and cis-Golgi, where we have previously found concentration activity associated with vesicular tubular clusters (Martínez-Menárguez JA, Geuze HJ, Slot JW, Klumperman J. Cell 1999; 98: 81-90).

Immunocytochemical localization of vigilin, a tRNA-binding protein, after cell fractionation and within the exocrine pancreatic cell of the rat

A polyclonal antibody against a fusion protein composed of two vigilin domains and γ-galactosidase was used to localize the tRNA-binding protein vigilin at the ultrastructural level. Immunoreactivity for vigilin was found on membranes bearing ribosomes within the microsome fraction derived from rat livers and on the rough endoplasmic reticulum in rat exocrine pancreatic cells, thus corroborating the involvement of vigilin in protein synthesis. Gold labeling was also found in the nucleus and in the microsome fraction in an osmiophilic substance resembling heterochromatin. Both of these localizations, the rough endoplasmic reticulum and the nucleus, are in line with the observation that a vigilin-containing ribonucleoprotein complex binds tRNA.

Characterization of novel peptoid agonists for the CCK-A receptor

The successful design of peptoid CCK-B receptor antagonists using rational approaches suggested that it might be feasible to develop similar non-peptide small molecule agonists with potential therapeutic applications. We now report the characterization of such a compound with full agonist activity at CCK-A receptors on rat exocrine pancreatic acinar cells. The compound, PD149164, stimulated a similar maximal response to CCK8 from the exocrine pancreas in anaesthetized rats in vivo, and from isolated pancreatic acini in vitro it also generated intracellular Ca 2+ oscillations similar to those evoked by CCK8. These effects were inhibited by the CCK-A antagonist L-364,718. Interestingly, the enantiomer of PD149164, PD151932, was a CCK-A antagonist and blocked PD149164 stimulated effects on the exocrine pancreas. The data indicate that it is possible to develop both agonist and antagonist activities in enantiomers of small non-peptide molecules.

Giant mitochondria induced in rat pancreatic exocrine cells by ethanol and iron.

Mitochondria are sensitive indicators of cellular pathology. Under certain circumstances, these organelles respond to cellular insult by a marked increase in size, resulting in the formation of giant mitochondria (megamitochondria). Ethanol has been implicated in the induction of giant mitochondria in rat hepatocytes. Since ethanol is reported to affect pancreatic mitochondria, we examined this organ for evidence of mitochondrial giantism in rats administered ethanol and a relatively small amount of supplementary iron. Diets were administered via a chronically implanted gastrostomy catheter. Rats were segregated in four groups: 1) basic high-fat diet, 2) ethanol and a high fat diet, 3) carbonyl iron and a high-fat diet, and 4) ethanol and carbonyl iron combined with a high-fat diet. After the animals were on their respective diets for 16 weeks, specimens of pancreas were extirpated and processed for and examined by transmission electron microscopy. Mitochondria in rats on the basic high fat diet resemble those in untreated controls. With ethanol and a high-fat diet, some mitochondria in virtually every exocrine cell are profoundly altered. Such organelles, which are of normal size, have undergone rearrangement of their internal membranes, with three of four parallel cristae residing at one or both poles of spherical to ovate mitochondria. In rats receiving both ethanol and carbonyl iron, giant mitochondria are present in pancreatic exocrine cells. Except for their size, megamitochondria in many cases are virtual likenesses of the altered mitochondria in the alcohol-high-fat rats, having several stacked cristae at either pole and an enhanced matrix compartment. Many of the giant mitochondria have at least one expanded crista, which contains packets of helically coiled filaments. When control rats are fed carbonyl iron without ethanol, the pancreatic mitochondria display a marked propensity for forming clusters of tightly interlocked organelles, formations that may be a prelude to mitochondrial fusion. It appears that iron supplementation sets the stage for organelle fusion, hence enlargement, with the ethanol providing the stimulus for fusion to actually take place, and controlling the final morphology of the resultant megamitochondria.

Inhibitory Effect of Thyrotropin-Releasing Hormone on Enzyme Secretion from Isolated Rat Pancreatic Acinar Cells

This study reports a direct effect of TRH on amylase secretion from isolated rat exocrine pancreatic acinar cells. TRH inhibited carbachol (10(-5) M)-stimulated amylase secretion by a maximum of 24% at a concentration of 10(-11) M (p < 0.05), but did not affect basal amylase release in concentrations from 10(-13) M to 10(-8) M. Ceruletide (3 x 10(-10) M)-stimulated amylase secretion was maximally reduced by 23% at a TRH concentration of 10(-10) M (p < 0.05). Direct stimulation of protein kinase C-mediated secretion by the diacylglycerol analogue 1-oleoyl-2-acetyl-sn-glycerol (OAG) was not altered by TRH. The TRH metabolite cyclo (His-Pro) did not influence basal or stimulated pancreatic secretion in vitro. These findings point to a TRH-mediated modulation of exocrine pancreatic secretion at the receptor site.

Immunogold localization of actin in the testis and exocrine pancreas: spatial relationship with tight junctional strands.

The fracture-label technique was used in conjunction with a monoclonal antibody to actin and the phospholipase A2-colloidal gold (PLA2-CG) method to examine the spatial distribution of actin filaments in relation to the three-dimensional arrangement of tight junctional strands in rat testes and exocrine pancreatic acinar cells. The intimate association of actin filaments with tight junctional strands in the pancreas and testis was also illustrated by a double-labeling experiment in which freeze-fractured pancreas or testis was labeled with monoclonal antibody-protein A-gold (30 nm gold size) followed by incubation with a PLA2-CG complex (11 nm gold size). Freeze-fracture-exposed tight junctional strands in both testicular and exocrine pancreatic cells labeled by PLA2-CG complex, indicated the presence of phospholipids in these cylindrical membranous structures. Immunolabeling of freeze-fractured testes with a monoclonal antibody to actin revealed a narrow band of gold particles juxtaposed to the cytoplasmic aspect of the protoplasmic membrane halves decorated with parallel linear arrays of cylindrical tight junctional strands. Many of the gold particles representing actin antigenic sites were in direct contact with the cross-fractured tight junctional strands. Fracture-label preparations of exocrine pancreas labeled with the monoclonal anti-actin antibody also exhibited a similar labeling pattern at the apex of acinars cells where the tight junction complex is located. Double-labeling experiments revealed the simultaneous labeling of actin and phospholipids in the same fracture-label preparations. Digestion of testicular and pancreatic tissue samples in a free PLA2 solution prior to labeling with the monoclonal antibody or PLA2-CG complex removed not only the gold labeling previously seen over the tight junctional strands but also reduced drastically the immunolabeling for actin that was previously seen associated with the tight junction complex. Taken together, results of the present study showed that actin filaments are structural components of the tight junction strands and are connected to the cytoplasmic aspect of the latter structures. The interaction between this particular cytoskeletal element and the tight junction may be through the binding of a special domain of the actin filament to the phospholipids that partially make up the tight junctional complex.

Glucocorticoid receptor gene expression in the exocrine and endocrine rat pancreas.

Although pleiotropic effects of glucocorticoids on the endocrine and exocrine pancreas are well documented, it remains controversial whether these effects are due to direct interactions of glucocorticoid hormones with their receptors in the respective target cells. We therefore examined gene expression of the glucocorticoid receptor (GR) in various functional compartments of the rat pancreas. Using in situ hybridization by generating a radioactive cRNA probe from a specific cDNA clone for the rat glucocorticoid receptor we found a gradient in the expression of receptor mRNA transcripts from endocrine > acinar > ductal cells. In contrast, no specific hybridization signal was observed in the endothelial cells of vascular structures. Using a monospecific polyclonal antibody against the rat glucocorticoid receptor in Western blot analyses we found significant expression of the 94 kD receptor protein in an enriched pancreatic acinar cell preparation. This data suggest that rat pancreatic exocrine cells might serve a direct target tissue for glucocorticoid action.

Changes in the gap junction of rat exocrine pancreatic cells induced by long-term ethanol ingestion.

Fourteen Male Wistar rats were orally administered 20% ethanol ad libitum in place of water. After 1, 2, 3, 6 and 12 months of ethanol ingestion, the gap junctions of pancreatic acinar and ductal cells were observed by transparent electron microscopy. The result was compared with fifteen control animals. In the control, the ductal cells proved to have only small sized (0.1 micron in length) gap junction located in the apical domain as a part of junctional complex. While, in the acinar cells, not only small but large-sized gap junctions several microns in length were observed in the baso-lateral domain. After 3 months of ethanol ingestion, the gap of these two types of gap junctions was replaced with a continuous, granular structure. In addition, irregularly running gap junctions were observed after 6 months of ethanol ingestion. The functional implication of these morphological changes is not clear.

Immunolocalization of phospholipids using freeze-fracture cytochemistry

Phospholipids are found ubiquitously in all mammalian cell membranes. The fracture-label technique offers the opportunity to localize phospholipids in tissues and cells. This technique has recently been used in combination with colloidal-gold-conjugated phospholipase A2 to demonstrate the presence of phospholipids in epithelial tight junctional strands. In the present study, we show the localization of specific phospholipids in the plasma and intracellular membranes in the rat exocrine pancreatic cells using murine monoclonal anti-cardiolipin (CL) antibodies that are reactive with phosphatidylethanolamine (PE) and have lupus anticoagulant activity. These antibodies were produced from BALB/c mice immunized with either ADP-activated murine platelets or CL mixed with apolipoprotein H.Pancreatic tissue was excised from adult male Sprague-Dawley rats under light ether anesthesia and fixed by immersion with 1% glutaraldehde in 0.1 M cacodylate buffer. The tissue was trimmed into 1-mm cubes which were then embedded in 30% bovine serum albumin in 0.01 M PBS, and cross-linked with a final concentration of 1% glutaraldehyde to form a hardened gel.

Cytochemical evidence for the presence of phospholipids in epithelial tight junction strands.

Previous freeze-fracture experiments using either glutaraldehyde-fixed and cryoprotected specimens or unfixed rapid-frozen samples led to the proposal that cylindrical strands of the tight junction (TJ) observed in freeze-fracture preparations are inverted cylindrical micelles made up of membrane lipids and, possibly, membrane proteins. However, no one has yet been able to directly label the structural fibrils of the TJ. To test the hypothesis that TJ strands observed on freeze-fracture preparations are composed at least partially of lipids, we have combined the phospholipase A2-gold and the fracture-label techniques for localization of phospholipids. Phospholipase A2, purified from bee venom, was adsorbed on gold particles and used for specific labeling of its substrate. Phospholipase A2-colloidal gold (PLA2-CG) complex was applied to freeze-fractured preparations of rat exocrine pancreatic cells and testicular Sertoli cells, both of which are known to have extensive TJ complexes on their plasma membranes. Fracture-label replicas of exocrine pancreatic cells revealed specific association of gold particles with TJ fibrils on the protoplasmic fracture-face of the plasma membrane. The majority of these gold particles were observed either directly on the top of the TJ fibrils or adjacent to these cylindrical structures. A high density of PLA2-CG labeling was also observed over the complementary exoplasmic fracture-face of the TJ complex. This intimate association of PLA2-CG labeling with the TJ is particularly evident in the Sertoli cell plasma membrane, where rows of gold particles were observed to be superimposed on parallel arrays of cylindrical strands of the TJ complex. The present findings provide direct cytochemical evidence to support the hypothesis that cylindrical TJ strands observed in freeze-fracture preparations contain phospholipids.

Beta-COP localizes mainly to the cis-Golgi side in exocrine pancreas.

We examined the distribution of the non-clathrin-coated vesicle-associated coat protein beta-COP in rat exocrine pancreatic cells by immunogold cytochemistry. Labeling for beta-COP was found in the Golgi region (48%) where it was associated with vesicles and buds of approximately 50 nm, showing a characteristic approximately 10-nm-thick coat. The other half of the label was present in the cytoplasm, not associated with visible coats or membranes, with a minor fraction present on small clusters of tubules and vesicles. Clathrin-coated vesicles were typically located at the trans-side of the Golgi complex, and showed a thicker coat of approximately 18 nm. Of the total beta-COP labeling over the Golgi region, 68% occurred on the cis-side, 6% on the cisternae, 17% on the rims of the cisternae, and only 9% on the trans-side. For clathrin these figures were 16, 2, 4, and 78%, respectively. At the cis-Golgi side beta-COP was present in transitional areas (TA), on so-called peripheral elements (PE), consisting of tubules and vesicles located between the cup-shaped transitional elements (TE) of the RER and the cis-most Golgi cisternae. Label for Sec23p was also present in TA but was located closer to the TE, while beta-COP labeled PE were located near the cis-Golgi cisternae. Upon energy depletion, Golgi associated beta-COP was almost exclusively (86%) in spherical aggregates of 200-500 nm in diameter, whereas the cis-side (6%), the cisternae (1%), the rims (4%) and trans-side (3%) of the Golgi complex, were barely labeled; 50% of the total label remained in the cytoplasm. The aggregates were predominantly located at the cis-side of the Golgi stack, next to, but distinct from the Sec23p positive TA, that were devoid of beta-COP and had only a few recognizable vesicles left. Incubation with aluminum fluoride resulted in fragmentation of the Golgi complex into large clusters of beta-COP positive vesicles, while 50% of the label remained in the cytoplasm, as in control cells. After 10 min of Brefeldin A treatment 91% of beta-COP was cytoplasmic and only 7% associated with membranes of the Golgi complex. The total label for beta-COP over exocrine cells remained unchanged during the incubation with either of the drugs, indicating that the drugs induce reallocation of beta-COP. Our data suggest that beta-COP plays a role in membrane transport at the cis-side of the Golgi complex.

Inhibitors of nonselective cation channels in cells of the blood-brain barrier.

In the antiluminal membrane of isolated capillaries of rat and porcine brain (blood-brain barrier) nonselective cation channels with g = 31 pS were observed in cell-excised membrane patches. The channel inactivated by decreasing cytosolic Ca2+ below 1 microM and was inhibited by 1 mM ATP on the intracellular side. Anions and divalent cations did not pass the channel, but Na+ and K+ were equally permeant. Like the nonselective cation channel of rat exocrine pancreatic cells, the channel in cerebral capillary endothelial cells was inhibited reversibly by derivatives of diphenylamine-2-carboxylate (DPC), like 3',5-dichlorodiphenylamine-2-carboxylic acid (DCDPC, ki = 1 microM), and flufenamic acid (ki = 4.9 microM). 4'-methyldiphenylamine-2-carboxylic acid (4-MDPC), 5-chloro-2(3-trifluormethylphenylamino)-3-nitrobenzoic acid, and 5-nitro-2-(3-phenylpropylamino)-2-carboxylic acid (NPPB), as well as the antiinflammatory drug ((Z)-5-chloro2,3-dihydro-3-(hydroxy-2-thienylmethylene)-2-ox o-1H-indole-1- carboxamide (Tenidap)) had a relatively low blocking potency (ki > 10 microM). Gadolinium (10 microM), a blocker of stretch-activated channels, inhibited the nonselective cation channel potently.

Characterization of a 58 kDa cis-Golgi protein in pancreatic exocrine cells.

We have studied the biochemical characteristics and localization of a 58 kDa cis-Golgi marker protein (p58) in rat pancreatic exocrine cells. The protein remained associated with membranes after extraction at alkaline pH and was largely resistant to proteases, added to intact microsomes. By electrophoresis p58 could be resolved into two bands which in two-dimensional gels separated into several charge variants around pI 5.5. This size and charge heterogeneity of p58 did not appear to be due to acylation, glycosylation or phosphorylation. In non-reduced gels p58 migrated as two kinetically related, high relative molecular mass forms, apparently corresponding to disulfide-linked homo-dimers and -hexamers. Immuno-electron microscopy localized p58 to both the fenestrated cis-Golgi cisternae and small Golgi vesicles or buds as well as large, pleiomorphic structures, scattered throughout the cells and associated with distinct smooth ER (endoplasmic reticulum) clusters. These findings correlated with cell fractionation results showing the concentration of p58 in two microsomal subfractions, banding at intermediate densities between the rough ER and trans-Golgi in sucrose gradients. Our results indicate that p58 is a major component of pre- and cis-Golgi elements and could be part of the transport machinery that operates in these membranes. Together with results obtained with other cell types, these observations suggest that the peripheral smooth ER clusters are involved in the early stages of the secretory pathway in the pancreatic acinar cells.

Heavy chain binding protein (BiP/GRP78) and endoplasmin are exported from the endoplasmic reticulum in rat exocrine pancreatic cells, similar to protein disulfide-isomerase

Previously we found that in rat exocrine pancreatic cells, protein disulfide-isomerase (PDI), one of the major resident proteins in the lumen of the endoplasmic reticulum (ER) of many cells, is localized not only in the ER but also in the Golgi apparatus, secretory granules, plasma membranes, and even in the glandular lumens, despite possessing the ER retention signal KDEL (LysAspGluLeu) at the carboxyl terminus. In this report, we examined whether other ER luminal proteins bearing the KDEL signal at their C-termini, such as BiP/GRP78 and endoplasmin/GRP94 are also exported from the ER. We prepared two kinds of affinity-purified polyclonal antibodies; one against a synthetic peptide with 12 amino acids which is identical to the carboxyl terminus of BiP and another against purified endoplasmin. Immunoblot analysis using these two antibodies showed that BiP and endoplasmin exist in both the plasma membrane and the microsomal fractions, similar to the intracellular distribution of PDI in rat exocrine pancreas. The ratios of the amount of the three proteins in the two fractions, however, were variable, suggesting that the KDEL-bearing proteins such as PDI, BiP, and endoplasmin are exported from the ER with different efficiencies. Postembedding protein A-immunogold electron microscopy revealed that endoplasmin was exported from the ER and secreted to the extracellular space. The secretion of PDI in rat pancreatic lobules was inhibited by Brefeldin A (BFA) and by guanidino acid esters (FOY-305), which are known to be the inhibitors of the intracellular transport. Taken together with the previous immunogold electron microscopic analyses by Akagi et al. (1988), it is strongly suggested that in rat exocrine pancreatic cells PDI and the other KDEL-bearing proteins found in the extracellular space were not artificially released by cell damage during incubation but were secreted via the normal secretory pathway.

Effect of colchicine on the three-dimensional organization of elongated lysosomes(Nematolysosomes) in rat pancreatic exocrine cells.

The effect of colchicine on the organization of lysosomal system in rat pancreatic exocrine cells was three-dimensionally examined by 2μm semithin section electron microscopy combined with acid phosphatase (ACPase) cytochemistry. In control rat pancreatic exocrine cells, ACPase activity was localized in trans Golgi cisternae, usual spherical lysosomes, and elongated lysosomes (nematolysosomes). The distribution of lysosomes was geographically characteristic. Most spherical lysosomes were located around the Golgi complex. The nematolysosomes elongated from near basolateral plasma membrane to the Golgi area. In colchicine-treated rats which were intraperitoneally injected with 5 mg/kg B.W. of colchicine 4 hr before fixation, the normal distribution of lysosomes was strikingly disorganized. The spherical lysosomes increased in number and size. The nematolysosomes appeared to be fragmented or shrunk, and change their form to spherical. The normal appearance of lysosomes was recovered at 12-24 hr after colchicine injection. The changes in distribution and shape of lysosomes by colchicine seemed to be correlated with the integrity of microtubules. The morphological association of microtubules with nematolysosomes was also revealed by electron microscopy. These results suggest that the microtubule is essential for nematolysosomes in pancreatic exocrine cells.

Effect of GTP and Ca 2+ on inositol 1,4,5-trisphosphate induced Ca 2+ release from permeabilized rat exocrine pancreatic acinar cells

The effects of Ca 2+ and GTP on the release of Ca 2+ from the inositol 1,4,5-trisphosphate (IP 3) sensitive Ca 2+ compartment were investigated with digitonin permeabilized rat pancreatic acinar cells. The amount of Ca 2+ released due to IP 3 directly correlated with the amount of stored Ca 2+ and was found to be inversely proportional to the medium free Ca 2+ concentration. Ca 2+ release induced by 0.18 μM IP 3 was half maximally inhibited at 0.5 μM free Ca 2+, i.e. at concentrations observed in the cytosol of pancreatic acinar cells. GTP did not cause Ca 2+ release on its own, but a single addition of GTP (20 μM) abolished the apparent desensitization of the Ca 2+ release which was observed during repeated IP 3 applications. This effect of GTP was reversible. GTPγS could not replace GTP. Desensitization still occurred when GTPγS was added prior to GTP. The reported data indicate that GTP, stored Ca 2+ and cytosolic free Ca 2+ modulate the IP 3 induced Ca 2+ release.
