Involvement of Cytokine-Induced Neutrophil Chemoattractant in Hypothalamic Thermoregulation of Luteinizing Hormone-Releasing Hormone

We demonstrated in a previous study that serum IL-8 concentrations were significantly higher in women with hot flashes than without hot flashes. To clarify the role of IL-8 in the pathoetiology of menopausal hot flashes, we examined the effect of rat cytokine-induced neutrophil chemoattractant (CINC), a member of the IL-8 family, on thermoregulation using ovariectomized (OVX) rats treated with intracerebroventricular (i.c.v.) injection of LHRH agonist (LHRHa) as a model of hot flashes. We found that: 1) expression of CINC mRNA was increased around the periventricular area in the hypothalamus at 1 h, and the serum CINC concentration was increased at 2 h after i.c.v. injection of LHRHa; 2) the increase in serum CINC concentration in hypophysectomized rats was significantly lower than that in sham-operated rats; 3) i.c.v. but not iv injection of CINC elevated the rectal temperature of OVX rats; 4) i.c.v. injection of LHRHa into OVX rats produced a rapid rise (maximal increase: 10-25 min) in tail skin temperature, and the elevation was augmented by injection of an anti-CINC antibody; and 5) changes in serum CINC concentration and skin temperature after i.c.v. injection of LHRHa were reversed by replacement of estradiol. In conclusion, the production of CINC in the hypothalamus due to LHRHa injection in OVX rats was increased after elevation of skin temperature, suggesting that CINC plays a key role in the homeostasis of body temperature. Disturbance of the thermoregulatory mechanism involving LHRH and CINC may be related to the pathoetiology of hot flashes.

Hepatocyte Growth Factor Stimulates the Induction of Cytokine-Induced Neutrophil Chemoattractant Through the Activation of NF-κB in Rat Hepatocytes 1

Recent evidence indicates that CXC chemokines such as human interleukine-8 (IL-8) and rat cytokine-induced neutrophil chemoattractant (CINC) have mitogenic and anti-apoptotic functions in addition to their chemotactic activity toward neutrophils. In the liver, hepatocyte growth factor (HGF) is implicated as a mitogen in the regeneration of liver. However, little is known the interaction between HGF and CXC chemokines during liver injury, repair and regeneration. We hypothesized that HGF may stimulate the expression of such chemokines, which contributes to mitogenic action in liver regeneration. Primary cultured rat hepatocytes were treated with recombinant human HGF (rhHGF), in the absence and presence of calpain inhibitor-1 (CI-1), genistein or anti-CINC-1 antibody. Levels of CINC-1 and its mRNA, tyrosine phosphorylation of HGF receptor (c-Met), the activation of transcription factor, nuclear factor-kappaB (NF-kappaB), and activities of DNA synthesis were measured. rhHGF enhanced the production of CINC-1 time- and dose-dependently, which followed the increased levels of CINC-1 mRNA. Under the same conditions, rhHGF increased the tyrosine phosphorylation of c-Met. The electrophoretic mobility shift assay revealed that rhHGF activated the nuclear translocation of NF-kappaB and its DNA binding. Proteasome inhibitor (CI-1) blocked the NF-kappaB activation and the CINC-1 production. The tyrosine kinase inhibitor (genistein) inhibited the activities of CINC-1 production and the DNA synthesis stimulated by rhHGF. However, the treatment of anti-CINC-1 antibody had no effect on the DNA synthesis. These results demonstrate that rhHGF can stimulate the induction of CINC-1 gene expression through the activation of NF-kappaB via its receptor c-Met in hepatocytes. Although CINC-1 seems to be not associated with the enhancement of DNA synthesis by rhHGF, it cannot negate the possibility that CINC-1 may contribute to liver repair and regeneration.

Effect of hepatocyte growth factor on induction of cytokine-induced neutrophil chemoattractant in rat hepatocytes

Background Restrictions on graft size are a serious obstacle to the expansion of indications for adult recipients in living donor liver transplantation. Hepatocyte growth factor (HGF) has a crucial role in regeneration following hepatic injury. Rat cytokine-induced neutrophil chemoattractant (CINC), a member of the interleukin-8 superfamily in humans, has been implicated in chronic liver diseases or development of liver ischemia-reperfusion injury. Studies were performed to examine whether HGF influences the induction of CINC in hepatocytes. Methods Primary cultures of rat hepatocytes were treated with or without recombinant human (rh) HGF. The release of CINC into the culture medium and levels of CINC mRNA were measured using an enzyme-linked immunosorbent assay and Northern blot analysis. Transcription of nuclear factor (NF)-kappa B was detected by electrophoretic mobility shift assays. Results rhHGF increased the release of CINC in the medium dose- and time-dependently, showing a maximal effect at 100 ng/mL. Genistein (100 μmol/L) blocked the release of CINC stimulated by rhHGF. Levels of CINC mRNA were also increased, reaching a maximum at 8 hours after addition of rhHGF. Electrophoretic mobility shift assays revealed rhHGF activated transcription factor, NF-kappa B. Conclusion These results suggest that HGF stimulates the induction of CINC gene expression through activation of NF-kappa B. CINC may be involved in the function of HGF during liver regeneration.

Change in location of cytokine-induced neutrophil chemoattractants (CINCs) in pulmonary silicosis

Rat cytokine-induced neutrophil chemoattractants (CINCs), which belong to the interleukin-8 family, are known to be induced by treatment with lipopolysaccharide (LPS). Recently, CINCs were grouped into four subtypes—CINC-1, CINC-2α, CINC-2β, and CINC-3—and CINC-1 was considered to be a major isoform among the four CINCs in LPS-induced acute lung inflammation in rats. The purpose of this study was to investigate the change in location of CINCs with chronic inflammation induced by experimental pulmonary silicosis. Administration of silica particles induced lung granulomas. Immunohistochemical staining for CINCs showed that the number of cells positive for CINC-2α, CINC-2β, and CINC-3 was increased, peaking at 1 day after treatment with silica particles, whereas CINC-1 was almost undetectable. We suggest that CINC-2α, CINC-2β, and CINC-3 are the most important chemoattractants in the formation of granulomas in chronic inflammation.

The role of rat cytokine-induced neutrophil chemoattractants (CINCs) in inflammation

Rat cytokine-induced neutrophil chemoattractants (CINCs) are the members of the CXC chemokine family. Four neutrophil chemokines, CINC-1, CINC-2 alpha, CINC-2 beta and CINC-3, were purified from the conditioned medium of granulation-tissue culture. CINC-2 alpha and CINC-2 beta differ only in the sequence of three carboxy-terminal residues and are produced by alternative splicing. CINC-3 had neutrophil chemotactic activity similar to that of CINC-1 and CINC-2, but induced greater calcium mobilization than CINC-1 and CINC-2. CINC-1, -2 and -3 induced calcium flux in CXCR2-transfected HEK293 cells. In addition, anti-CXCR2 serum inhibited neutrophil chemotactic activities of the three types of CINCs almost completely. These results indicate that rat CXCR2 is a unique receptor for CINC-1, -2 and -3. CINCs induced calcium mobilization through pertussis toxin-insensitive G-protein but induced chemotaxis through pertussis toxin-sensitive G-protein. CINC-1/-2 and CINC-3 may stimulate both G-proteins with distinct efficiency. The concentration of CINC-1 increased transiently in rat air pouch/lipopolysaccharide inflammation, whereas the CINC-2 level increased linearly. The number of infiltrated cells increased up to 8 h. The increase in cell number was correlated with the total concentration of CINC-1 and CINC-2. Northern blot analyses and enzyme-linked immunosorbent assay showed that CINC expression was very low in rat macrophages without stimulation and increased after lipopolysaccharide stimulation. These data suggest that CINCs are expressed by inflammatory cells such as macrophages at a site of inflammation and play important roles in neutrophil infiltration.

Inhibition of the production of rat cytokine-induced neutrophil chemoattractant (CINC)-1, a member of the interleukin-8 family, by adenovirus-mediated overexpression of IkappaBalpha.

Cytokine-induced neutrophil chemoattractant (CINC)-1, a counterpart of the human growth-regulated gene product (GRO) of the interleukin-8 family, has been suggested to play critical roles as a mediator of inflammatory reactions with neutrophil infiltration in rats. NF-kappaB has been speculated to be involved in the production of CINC-1, since the NF-kappaB-binding domain is important for the CINC-1 promoter activity in several of our reporter assays. In the present study, we examined the effects of an overexpression of IkappaBalpha, a specific natural inhibitor of NF-kappaB, on CINC-1 production. For this purpose, we constructed two recombinant adenoviruses, AxCAIkappaBalpha and AxCAmutantIkappaBalpha, which express respectively wild IkBa and a mutated nondegradable IkappaBalpha in which serine residues 32 and 36 are replaced by alanine residues. Transfecting wild and mutant IkBa by these adenovirus vectors inhibited NF-kappaB activation and CINC-1 production, which were both caused by IL-1beta stimulation in the normal rat kidney epithelial cell line NRK-52E. We also showed that the nondegradable mutant IkappaBalpha was approximately 30 times more potent than the wild type in inhibiting CINC-1 production. These findings demonstrate that CINC-1 production with NF-kappaB activation is primarily regulated by non-phosphorylated IkBa in the cytoplasm.

The ratio of ELR+ to ELR- CXC chemokines affects the lung and liver injury following hepatic ischemia/ reperfusion in the rat.

Hepatic ischemia/reperfusion (I/R) results in a neutrophil-dependent lung and liver injury. The process of neutrophil recruitment and activation in this injury is at least partially dependent on the presence of the ELR+ CXC chemokines. Other investigations have shown that ELR- CXC chemokines can block ELR+ CXC chemokine neutrophil recruitment and activation in vitro. To begin to investigate the role of the balance between these 2 types of molecules in vivo in neutrophil recruitment and activation following hepatic I/R, we used our rat model of lobar hepatic I/R and pretreated animals with pharmacologic doses of gamma-interferon (gamma-IFN). gamma-IFN is known to upregulate some of the ELR- CXC chemokines, including gamma-IFN-inducible protein (IP-10) and monokine-induced by gamma-IFN (MIG), as well as down-regulate ELR+ CXC chemokine production. Following hepatic I/R or sham laparotomy, hepatic and pulmonary levels of the ELR- chemokines, IP-10 and MIG, and the ELR+ chemokines, rat cytokine-induced neutrophil chemoattractant (KC), macrophage inflammatory protein-2 (MIP-2), and epithelial neutrophil activating protein (ENA-78) were determined by ELISA, and lung and liver injury were assessed. In response to gamma-IFN, hepatic and pulmonary levels of the ELR- chemokines were increased and the levels of the ELR+ chemokines were decreased. Immunohistochemical staining confirmed the hepatocyte as the source of these molecules, as well as the changes in chemokine levels in response to gamma-IFN. There was an associated significant decrease in liver and lung injury, although there was no significant decrease in neutrophil influx in either tissue. This suggests that the alteration in the balance of ELR+ to ELR- CXC chemokines results in a decrease in tissue injury through a mechanism other than through an alteration in tissue neutrophil levels.

Appearance of cytokine-induced neutrophil chemoattractant isoforms and immunolocalization of them in lipopolysaccharide-induced acute lung inflammation in rats.

Recently, rat cytokine-induced neutrophil chemoattractant (CINC), which belongs to the interleukin-8 family, was grouped into four isoforms, CINC-1, CINC-2a, CINC-2beta, and CINC-3. To determine the major component and the source of CINC in airways, we investigated the change in appearance of CINC isoforms after exposure of rats to lipopolysaccharide. Male Sprague-Dawley rats, 8-10 weeks old, were used in the present study. Bronchoalveolar lavage (BAL) was performed at 1, 2, 4, 6, 12, and 24 h after lipopolysaccharide inhalation (4 mg/ml for 30 min). The concentrations of each specific rat CINC in the BAL supernatant were measured by use of commercially available kits. Furthermore, lung tissue was employed for immunohistochemical staining of CINCs (CINC-1, -2alpha,-2beta, and -3) using the streptavidin-biotin technique. Inhalation of lipopolysaccharide caused increases in CINC-1, CINC-2aalpha, and CINC-3 in BAL fluids, whereas CINC-2beta was not detected. The increases in CINC-2a and CINC-3 were less than the increase in CINC-1. Positive immunohistochemical staining for CINC-1 was detected in bronchial noncilliated cells and in certain neutrophils that had infiltrated into the submucosa. These findings suggest that CINC-1 is the major isoform among the four CINCs in lipopolysaccharide-induced acute lung inflammation in rats. Its sources are likely to be bronchial noncilliated cells and certain infiltrating neutrophils.

Effects of Cytokine-Induced Neutrophil Chemoattractants (CINCs) on Shape Change, Adhesiveness and Phagocytosis of Rat Neutrophils.

Rat cytokine-induced neutrophil chemoattractants (CINC)-1, -2 alpha, -2 beta and -3 belong to alpha-chemokines and are known to be potent chemoattractants for rat neutrophils in vivo and in vitro. In this study, the abilities of CINCs to affect several functions of rat neutrophils are estimated. All CINCs at a concentration of 10(-8) M induced the polarization of rat neutrophils. The adhesion of each CINC-treated neutrophil to fibrinogen significantly increased, reaching a maximal adhesion at 10(-8) M of each CINC. The CINCs enhanced the phagocytosis of heat-killed yeast cells by neutrophils in a dose-dependent manner. In contrast, CINCs did not induce the production of reactive nitrogen intermediates. The results suggest that CINCs play a significant role in neutrophil infiltration and phagocytosis at inflammatory sites in rats.

Glucocorticoid-mediated Gene Suppression of Rat Cytokine-induced Neutrophil Chemoattractant CINC/gro, a Member of the Interleukin-8 Family, through Impairment of NF-κB Activation

The glucocorticoid dexamethasone inhibited the production of the rat cytokine-induced neutrophil chemoattractant CINC/gro, a counterpart of human melanoma growth-stimulating activity that belongs to the interleukin-8 (IL-8) family, in the normal rat kidney epithelial cell line NRK-52E stimulated with interleukin-1 beta (IL-1 beta), lipopolysaccharide, or tumor necrosis factor alpha. The accumulation of CINC/gro mRNA induced by these activators was also decreased comparably by dexamethasone. A nuclear run-on assay revealed that dexamethasone decreased the IL-1 beta-induced transcription of the CINC/gro gene. The half-life of CINC/gro mRNA transcripts did not change significantly after exposure to dexamethasone, suggesting that this glucocorticoid acts mainly at the transcriptional level. Transfection with luciferase expression vectors containing 5'-deleted and mutated CINC/gro gene sequences demonstrated that the 5'-flanking region containing the NF-kappa B binding site is involved in the IL-1 beta- and dexamethasone-induced activation and repression of the CINC/gro gene expression, respectively. Furthermore, a tandem repeat of the NF-kappa B sequence in the CINC/gro gene conferred the inducibility by IL-1 beta and suppression of luciferase activity by dexamethasone. In an electrophoretic mobility shift assay, dexamethasone diminished the IL-1 beta-induced formation of NF-kappa B complexes, which consisted of p65 and p50. Western blotting revealed that dexamethasone inhibited the IL-1 beta-induced translocation of p65 from the cytoplasm into the nucleus, while the nuclear level of NF-kappa B p50 remained almost unchanged. In addition, the degradation of I kappa B-alpha induced by IL-1 beta was not inhibited by dexamethasone. These results indicated that the suppression of the CINC/gro gene transcription by glucocorticoid occurs through the impairment of NF-kappa B activation, possibly by interference with the translocation of NF-kappa B p65 from the cytoplasm into the nucleus, thereby suppressing transactivation of the CINC/gro gene.

Recombinant production and biological properties of rat cytokine-induced neutrophil chemoattractants, GRO/CINC-2 alpha, CINC-2 beta and CINC-3.

Recently we found four cytokine-induced neutrophil chemoattractants, CINC-1, CINC-2 alpha, CINC-2 beta and CINC-3/macrophage inflammatory protein 2 (MIP-2), in conditioned medium of granulation tissue obtained from carrageenin-induced inflammation in rats [Nakagawa, H., Komorita, N., Shibata, F., Ikesue, A., Konishi, K., Fujioka, M. & Kato, H. (1994) Biochem. J. 301, 545-550]. In the present report, we describe recombinant production of CINC-2 alpha, CINC-2 beta and CINC-3 in Escherichia coli, and biological properties of these chemokines. Neutrophil chemotactic activities of CINC-2 alpha and 2 beta in vitro were the same as the activity of CINC-1. CINC-3 had an activity comparable to other CINCs, but showed a decrease at high concentrations. Stimulation of neutrophils with CINCs induced an increase in intracellular [Ca2+] dose-dependently. CINC-3 was more potent than the other CINCs and still induced an increase in intracellular [Ca2+] in rat neutrophils stimulated first with other CINCs. CINC-2 alpha, CINC-2 beta and CINC-3 induced a comparable response to CINC-1 in the release of cathepsin G from rat neutrophils. Injection of CINC-2 alpha, 2 beta and 3 into preformed air-pouch on the back of rat induced infiltration of neutrophils to an extent similar to that caused by the injection of CINC-1. These data indicate CINC-2 alpha, 2 beta and 3 as well as CINC-1 are chemoattractants specific for neutrophil in vivo.

Intratracheal administration of endotoxin and cytokines. VI. Antiserum to CINC inhibits acute inflammation

Cytokine-induced neutrophil chemoattractant (CINC), a chemotactic molecule of the interleukin (IL)-8 family, is known to be induced in the rat in response to tumor necrosis factor (TNF), IL-1, and lipopolysaccharide (LPS). Intratracheal injection of endotoxin (LPS) is shown to cause CINC mRNA expression in pulmonary tissue, peaking after 2 h, and CINC protein expression in bronchoalveolar lavage (BAL) fluid, peaking after 2-4 h. Intratracheal injection of synthetic CINC causes acute inflammation that is abrogated by coinjection of antiserum to purified natural rat CINC. Intratracheal injection of antiserum to CINC inhibits intratracheal LPS- and IL-1-induced neutrophil emigration into BAL fluid by approximately 60-70%. Despite the anti-inflammatory activity of anti-CINC antiserum, TNF is elevated in the lavage fluid of rats receiving anti-CINC, suggesting that CINC may act in a negative feedback loop to downregulate TNF expression. Intratracheal injection of antiserum to CINC combined with intravenous injection of anti-E-selectin antibody inhibits intratracheal LPS- and IL-1-induced neutrophil emigration into BAL fluid by approximately 75-85%. CINC-mediated chemotactic activity and E-selectin-mediated adherence of neutrophils to endothelium contribute significantly to the pathogenesis of LPS-initiated acute inflammation.

Inhibitory Action of Indomethacin on Neutrophil Infiltration in Monosodium Urate-Induced Pleurisy in Rats

The effects of indomethacin on the production of cytokines at inflammatory sites were investigated in the monosodium urate (MSU) pleurisy model characterized by both cellular influx and edema. Indomethacin (10 mg/kg) orally administered 0.5 hr prior to MSU injection into the pleural cavity significantly inhibited MSU-induced neutrophil accumulation in the cavity. In addition, the drug slightly enhanced the level of MSU-induced tumor necrosis factor production without affecting interleukin-1 production. Furthermore, indomethacin inhibited both the levels of MSU-induced rat cytokine-induced neutrophil chemoattractant (CINC/gro) and interleukin-6 (IL-6) production by 78.3% at 3 hr and 45.8% at 4 hr post-injection, respectively. Although intrapleural injection of CINC/gro induced neutrophil infiltration in a dose-dependent manner, IL-6 did not affect the action of CINC/gro on neutrophil influx. These findings suggest that the inhibitory action of indomethacin on neutrophil infiltration is, at least, partly mediated by a decrease in the MSU-induced CINC/gro content in this model.

The three dimensional structure of rat cytokine CINC/Gro in solution by homonuclear 3D NMR

The solution conformation of rat cytokine-induced neutrophil chemoattractant (CINC/Gro), a small protein consisting of 72 amino acid residues with proinflammatory activities, and a member of the interleukin 8 family corresponding to a counterpart of human Gro, was investigated with homonuclear 2D and 3D NMR spectroscopy. At each phase of the structural analysis, the homonuclear 3D NOESY-HOHAHA and HOHAHA-NOESY spectra afforded valuable data, removing ambiguities intractable by conventional 2D NMR techniques. CINC/Gro exists as a dimer in solution and contains a triple stranded anti-parallel β-sheet and C-terminal α-helix in the monomer structure, as observed in human IL-8, but non-trivial differences are also observed.

Regulation of Cytokine-Induced Neutrophil Chemoattractant (CINC) mRNA Production in Cultured Rat Cells

Rat cytokine-induced neutrophil chemoattractant (CINC) is an 8-kD polypeptide originally purified from media conditioned by interleukin-1 beta (IL-1 beta)-stimulated 52E, an epithelioid clone derived from the normal rat kidney (NRK) cell line. Using a fibroblastic clone of NRK cells, 49F, we found that lipopolysaccharide (LPS), IL-1 beta, and tumor necrosis factor-alpha (TNF-alpha) each induce synthesis of CINC mRNA and CINC, although in qualitatively and quantitatively different patterns. Through deadenylation experiments and by probing with oligonucleotides, we discovered that the smaller of the two major CINC transcripts appears to arise from the larger as a result of poly(A) tail removal and/or 3' cleavage.

Investigation of possible autocrine functions for rat GRO/CINC (cytokine-induced neutrophil chemoattractant).

Rat cytokine-induced neutrophil chemoattractant (CINC) is an eight kilodalton polypeptide originally purified from media conditioned by interleukin-1 beta stimulated 52E, an epithelioid clone derived from normal rat kidney (NRK) cells. Using a fibroblastic clone of the NRK cells, 49F, we found expression of the CINC gene to be induced by either serum or cytokines in growth-arrested cultures within 1 hour of stimulation. There was no observable CINC expression in exponentially growing cells in the absence of cytokine stimulation. CINC protein had no significant effect on 3H-thymidine incorporation or growth rate of NRK49F. We have observed that CINC is constitutively produced by some transformed NRK cells, clone RC20, suggesting an association with the expression of a transformed phenotype. Unlike the parent 49F, RC20 cells are capable of growth in soft agar and serum-free media and form highly metastatic tumors in nude mice. We have examined the possible autocrine functions of CINC and its possible links to the expression of the transformed phenotype by these cells. The use of a blocking CINC polyclonal antibody demonstrated that CINC did not function as an autocrine growth factor for RC20. Though CINC is a potent chemoattractant for neutrophils, it did not induce migration of either RC20 or 49F cells. CINC only moderately promoted adhesion of RC20 cells when used as a matrix protein. These data do not support the hypothesis that production of CINC by the RC20 cells provides an obvious advantage for the transformed cells constitutively producing it.

Inhibition of lung inflammatory reactions in rats by an anti-human IL-8 antibody.

IL-8 belongs to the family of chemotactic cytokines and may play an important role in the inflammatory response. In the current studies, a murine mAb (DM/C7) to human rIL-8 was found to have protective effects in inflammatory lung injury in rats. DM/C7 was nonreactive with the rat cytokine-induced neutrophil chemoattractant peptide. In vivo, DM/C7 blocked the glycogen-induced accumulation of neutrophils in rats and was highly protective against lung and dermal vascular injury after deposition of IgG immune complexes. The latter model of injury has recently been shown to be E-selectin dependent. The protective effects of DM/C7 correlated with reduced tissue accumulation of neutrophils, as measured by myeloperoxidase content. DM/C7 reacted with an epitope expressed by TNF-alpha-stimulated rat pulmonary artery endothelial cells and with the pulmonary vascular endothelium after intrapulmonary deposition of IgG immune complexes. In the model of IgG immune complex-induced lung injury, the protective effects of DM/C7 were abolished by prior absorption of the antibody with human rIL-8. Polyclonal antibody to cytokine-induced neutrophil chemoattractant peptide failed to protect against IgG immune complex-induced vascular injury even though this antibody blocked the in vitro chemotactic activity of cytokine-induced neutrophil chemoattractant. In the model of rapidly developing lung injury due to systemic activation of C after infusion of cobra venom factor, DM/C7 was not protective. As well, in the neutrophil-independent model of IgA immune complex-induced lung injury, treatment with DM/C7 was not protective. These data indicate that in inflammatory lung injury that is linked to E-selectin-dependent recruitment of neutrophils in rats, antibody to human IL-8 also blocks recruitment of neutrophils and thereby affords protection against lung injury. The data suggest the presence of an IL-8-like product in this model of lung injury.

Chemoattractants for neutrophils in lipopolysaccharide-induced inflammatory exudate from rats are not interleukin-8 counterparts but gro-gene-product/melanoma-growth-stimulating-activity-related factors.

Potent chemotactic activity for neutrophils was detected in rat inflammatory exudate induced by a subcutaneous injection of lipopolysaccharide in a carboxymethyl-cellulose suspension. We purified and characterized chemoattractants from the exudate by the following procedures: carboxymethyl-Sephadex C-25 ion-exchange chromatography; G3000SW gel-filtration chromatography; preparative reverse-phase high-pressure liquid chromatography; rechromatography on reverse-phase HPLC. Two chemotactic factors were purified and their N-terminal amino acid sequences were determined. One factor was a protein in which the first 20 N-terminal amino acids were identical to those of rat cytokine-induced neutrophil chemoattractant (CINC), a counterpart of human gro/melanoma growth-stimulating activity (MGSA). The other factor was highly similar to mouse macrophage inflammatory protein 2 (MIP-2). Mouse MIP-2, a chemotactic factor for neutrophils, is a member of the interleukin-8 family; however the protein we purified had higher similarity to human gro/MGSA than to human interleukin-8. These results indicate that, in rats, chemotactic factors for neutrophils induced by lipopolysaccharide stimulation are not counterparts of interleukin-8, but are gro/CINC-related peptides.
