Ligand-to-Metal Charge Transfer (LMCT) Catalysis: Harnessing Simple Cerium Catalysts for Selective Functionalization of Inert C-H and C-C Bonds.

ConspectusChemists have long pursued harnessing light energy and photoexcitation processes for synthetic transformations. Ligand-to-metal charge transfer (LMCT) in high-valent metal complexes often triggers bond homolysis, generating oxidized ligand-centered radicals and reduced metal centers. While photoinduced oxidative activations can be enabled, this process, typically seen as photochemical decomposition, remains underexplored in catalytic applications. To mitigate decomposition during LMCT excitation, we developed a catalytic cycle integrating in situ coordination, LMCT, and ligand homolysis to activate ligated alcohols transiently into alkoxy radicals. This catalytic approach leverages Ce(IV) LMCT excitation and highly reactive alkoxy radical intermediates for selective functionalizations of C(sp3)-H and C(sp3)-C(sp3) bonds under mild conditions. In this Account, we discuss these advancements, highlighting the practical utility of cost-effective cerium salts as catalysts and their potential to develop innovative transformations, addressing long-standing synthetic challenges.Selective functionalization of chemically inert C(sp3)-H bonds has long posed a significant challenge. We first detail our research using LMCT-enabled alkoxy radical-mediated hydrogen atom transfer (HAT) processes for selective C(sp3)-H functionalizations. Using readily available CeCl3, we established a general protocol for employing free alcohols in the Barton reaction. By integrating LMCT and HAT catalysis, we introduced a selective photocatalytic strategy for functionalizing feedstock alkanes, converting gaseous hydrocarbons into valuable products. Employing simple cerium salts like Ce(OTf)3 and CeCl3, we achieved selective C-H amination of methane and ethane at ambient temperature, achieving turnover numbers of 2900 and 9700, respectively. This catalytic manifold has been further exploited to address the site-selectivity challenge in the C-H functionalization of linear alkanes. The use of methanol as a cocatalyst enabled preferential functionalization of the most electron-rich sites, achieving a high intrinsic selectivity over 12:1 of secondary vs primary sites in pentane and hexane.Next, we discuss the catalytic utilization of alkoxy-radical-mediated β-scission, a frequently encountered side reaction in HAT transformations, for selective cleavage and functionalization of C-C bonds. The versatility of the LMCT catalytic platform facilitates the generation of alkoxy radicals from various free alcohols. In our initial demonstration of LMCT-enabled C(sp3)-C(sp3) bond activation, we developed a cerium-catalyzed ring-opening and amination of cycloalkanols, providing an effective protocol for cleaving unstrained C-C bonds. This strategy has been successfully applied to various radical cross-coupling processes, leading to innovative transformations such as ring expansions of cycloalkanols, dehydroxymethylative alkylation, amination, alkenylation, and ring expansions of cyclic ketones. These results highlight the synthetic potential of employing LMCT-mediated β-scission and ubiquitous C-C bonds as unconventional functional handles for generating molecular complexity.Lastly, we delve into our mechanistic investigations. Beyond the catalytic application of Ce(IV) LMCT in various transformations, we have undertaken comprehensive mechanistic studies. These investigations encompass characterization of Ce(IV) alkoxide complexes to elucidate their structures, evaluation of their photoactivity and selectivity in radical generation, and elucidation of kinetic pathways associated with transient LMCT excited states. Our research has revealed ultrafast bond homolysis, back electron transfer, and the selectivity of heteroleptic complexes in homolysis, providing crucial insights for advancing LMCT catalysis.

Merging New and Old Concepts: Tandem Oxidative Radical‐Polar Crossover Ritter Amidation via Multicomponent Photo‐ and Electrochemical Processes

Nitrogen‐containing drug molecules and pharmaceuticals are ubiquitous, rendering the construction of C–N bonds a crucial target in methodology development. The Ritter reaction is a well‐established method for accessing C–N bonds by generating amide functionality through the reaction of carbocations and nitriles. Since its discovery back in 1948, the Ritter reaction has progressively advanced towards milder and more sustainable conditions for carbocation generation. In this regard, notable contributions have been made by means of single electron transfer (SET) chemistry. Nowadays, photo‐ and electrochemistry are established methods of choice for the generation of reactive radical intermediates and their subsequent oxidation via radical‐polar crossover (RPC) mechanism. We review recent examples of tandem RPC and Ritter‐type protocols, demonstrating how photo‐ and electrochemical energies have been effectively harvested to expand the precursor pool for the Ritter reaction and also reimagine the process with novel mechanisms and additives.

Reductive Electrophilic Cross-Coupling for Constructing C(sp3)–C(sp3) Bonds

AbstractThe C(sp3)–C(sp3) bond is one of the most prevalent motifs in organic compounds and holds significant importance in organic synthesis. The utilization of two alkyl electrophiles for cross-coupling stands as a vital strategy in building C(sp3)–C(sp3) bonds. Nowadays, synthetic electrochemistry is undergoing rapid development owing to its exceptional attributes not only in terms of green and economic properties by reducing the large amount of traditional chemical reductants, but also by its capacity to generate highly reactive radical intermediates under mild conditions, thereby opening up new reaction pathways and presenting novel opportunities to constructing C(sp3)–C(sp3) bonds. Advancements in visible-light-induced reactions via alkenyl radical intermediates.

In recent years, visible-light-induced organic transformations have taken a central role driving forward the progress of modern organic synthesis. These processes typically involve the transient generation of highly reactive radical intermediates, facilitating a diverse array of chemical reactions. Despite the abundance of synthetic strategies enabling the access of aryl and alkyl-centered radicals, the exploitation of photochemistry to generate highly reactive alkenyl radicals has remained notably underdeveloped. In this review, we present recent advancements in visible-light-induced transformations that proceed through the generation of alkenyl radicals from alkenyl-containing precursors, predominantly alkenyl halides, showcasing their application in various organic transformations.

Photoenzymatic Asymmetric Hydroamination for Chiral Alkyl Amine Synthesis.

Chiral alkyl amines are common structural motifs in pharmaceuticals, natural products, synthetic intermediates, and bioactive molecules. An attractive method to prepare these molecules is the asymmetric radical hydroamination; however, this approach has not been explored with dialkyl amine-derived nitrogen-centered radicals since designing a catalytic system to generate the aminium radical cation, to suppress deleterious side reactions such as α-deprotonation and H atom abstraction, and to facilitate enantioselective hydrogen atom transfer is a formidable task. Herein, we describe the application of photoenzymatic catalysis to generate and harness the aminium radical cation for asymmetric intermolecular hydroamination. In this reaction, the flavin-dependent ene-reductase photocatalytically generates the aminium radical cation from the corresponding hydroxylamine and catalyzes the asymmetric intermolecular hydroamination to furnish the enantioenriched tertiary amine, whereby enantioinduction occurs through enzyme-mediated hydrogen atom transfer. This work highlights the use of photoenzymatic catalysis to generate and control highly reactive radical intermediates for asymmetric synthesis, addressing a long-standing challenge in chemical synthesis.

Access to All-Carbon Quaternary Centers by Photocatalytic Fluoroalkylation of α-Halo Carbonyl Compounds.

Perfluoroalkyl groups have become significantly important in pharmaceutical and agrochemical applications. In this study, we present a visible light-mediated photoredox neutral strategy for the fluoroalkylation of tertiary alkyl chlorides under transition-metal-free conditions. This method allows for the facile synthesis of fluoroalkylated all-carbon quaternary centers, exhibiting excellent functional group compatibility. Mechanistic studies reveal the involvement of two reactive radical intermediates and the in situ formation of metal enolates in a radical-polar crossover manner. The versatility of this methodology is demonstrated through synthetic transformations based on the carbonyl group, showcasing its potential for the rapid assembly of diverse organic molecules bearing fluoroalkyl all-carbon quaternary centers.

Nickel-Catalyzed Reductive Arylation of Redox Active Esters for the Synthesis of α-Aryl Nitriles: Investigation of a Chlorosilane Additive.

A nickel-catalyzed reductive cross-coupling of redox active N-hydroxyphthalimide (NHP) esters and iodoarenes for the synthesis of α-aryl nitriles is described. The NHP ester substrate is derived from cyanoacetic acid, which allows for a modular synthesis of substituted α-aryl nitriles, an important scaffold in the pharmaceutical sciences. The reaction exhibits a broad scope, and many functional groups are compatible under the reaction conditions, including complex highly functionalized medicinal agents. Mechanistic studies reveal that reduction and decarboxylation of the NHP ester to the reactive radical intermediate are accomplished by a combination of a chlorosilane additive and Zn dust. We demonstrate that stoichiometric chlorosilane is essential for product formation and that chlorosilane plays a role beyond activation of the metal reductant.

Non-Aufbau electronic structure in radical enzymes and control of the highly reactive intermediates.

Radicals are highly reactive, short-lived chemical species that normally react indiscriminately with biological materials, and yet, nature has evolved thousands of enzymes that employ radicals to catalyze thermodynamically challenging chemistry. How these enzymes harness highly reactive radical intermediates to steer the catalysis to the correct outcome is a topic of intense investigation. Here, the results of detailed QM/MM calculations on archetype radical B12-enzymes are presented that provide new insights into how these enzymes control the reactivity of radicals within their active sites. The catalytic cycle in B12-enzymes is initiated through the formation of the 5'-deoxyadenosyl (Ado˙) moiety, a primary carbon-centred radical, which must migrate up to 8 Å to reach the target substrate to engage in the next step of the catalytic process, a hydrogen atom abstraction. Our calculations reveal that Ado˙ within the protein environment exhibits an unusual non-Aufbau electronic structure in which the singly occupied molecular orbital is lower in energy than the doubly occupied orbitals, an uncommon phenomenon known as SOMO-HOMO inversion (SHI). We find that the magnitude of SHI in the initially formed Ado˙ is larger compared to when the Ado˙ is near the intended substrate, leading to the former being relatively less reactive. The modulation of the SHI originates from Coulombic interactions of a quantum nature between a negative charge on a conserved glutamate residue and the spin on the Ado˙. Our findings support a novel hypothesis that these enzymes utilize this quantum Coulombic effect as a means of maintaining exquisite control over the chemistry of highly reactive radical intermediates in enzyme active sites.

Visible light-induced halogen-atom transfer by N-heterocyclic carbene-ligated boryl radicals for diastereoselective C(sp3)-C(sp2) bond formation.

Photoinduced halogen-atom transfer (XAT) has rapidly emerged as a programmable approach to generate carbon-centered radical intermediates, mainly relying on silyl and α-aminoalkyl radicals as halogen abstractors. More recently, ligated boryl radicals have also been proposed as effective halogen abstractors under visible-light irradiation. In this study, we describe the use of this approach to enable C(sp3)-C(sp2) bond formation via radical addition of carbon-centered radicals generated via XAT onto chloroalkynes. Our mechanistic investigation reveals a complex interplay of highly reactive radical intermediates which, under optimized conditions, delivered the targeted vinyl chlorides in excellent yields and Z : E ratios. Finally, we demonstrated the synthetic value of these products in transition metal-based cross-coupling reactions.

Isolation, Screening, Identification, and Assessment of Laccase-Producing Fungi Isolated From Different Environmental Samples

ABSTRACT: One of the extracellular enzymes involved in ligninolytic is lacasse, secreted by white and brown rot fungus. The class of blue copper proteins that includes laccases consists of N-glycosylated multicopper oxidases. Ascomycetes, Deuteromycetes, and Basidiomycetes are all fungi that contain laccase; many of these are white-rot fungi that break down lignin. Due to their wide range of substrate specificity, laccases have been the focus of extensive research over the past few decades. Their latest applications include anything from the textile pulp and paper industries to culinary applications and bioremediation techniques. Laccases are also used in organic synthesis, where phenols and amines are common substrates. Dimers and oligomers are produced due to the coupling of reactive radical intermediates in these reactions. The current investigation gathered 50 white rot fungi, and the most incredible laccase-producing organisms in submerged fermentation were looked into. Ten of the 21 cultures displayed a reddish-brown color zone. Of these ten isolates (PTD 19, PTD 4, PP2J15, LKT 34, ITC 1, NRL 7, GOJ 7, PTD2, PP2J, and PKT12), only PP2J15 and GOJ 7 displayed the most reddish-brown color zone. The isolation of white rot fungus, their molecular characterization, and testing for laccase production are all covered in this Paper. Talaromyces verruculosus and Cladosporium cladosporioides were identified as the PP2J15 and GOJ 7 strains based on sequence comparison and phylogenetic analysis with reference taxa.

Electrochemistry and Organic Synthesis: New Adventures with Olefin Coupling Reactions and Electrode Surfaces

Electrochemistry has long offered opportunities to explore new modes of reactivity, new approaches for introducing selectivity into synthetic reactions, and new strategies for the synthesis of everything from complex molecules to complex, addressable molecular surfaces. In the talk to be given, two aspects of our efforts along these lines will be detailed. In the first, we will take a look at recent efforts to use anodically-generated reactive intermediates to convert readily available small molecules into functionalized synthetic building blocks and natural product derivatives. Key to these efforts are oxidation reactions that generate reactive radical cation and radical intermediates. In one application, newly available chiral lactols synthesized using methodology developed by the Lin (Cornell) and Miller (Yale) groups are being transformed into C-glycoside and aza-C-glycoside derivatives. The reactions take advantage of oxidation reactions of either enol ether or vinylsulfide groups. The cyclization reactions and the subsequent deprotection strategy needed for converting the cyclization products into a useful synthetic building block will be highlighted. In a second application of an anodic cyclization, the role of thioamides in reactions targeting the chrysosoporazine family of natural products will be discussed. The reactions seek to provide an efficient method for converting lignin derived building blocks into molecules that reduce the efflux of anticancer drugs from cells. Initial cyclization attempts utilizing amide based N-radicals and anodic olefin coupling reactions were not successful. Those reactions consistently led to over-oxidation of the products that contain a highly electron-rich aryl ring. The use of thio-amide derived radicals provides a method to circumvent this problem.In the second topic to be discussed, the question of how one might begin to induce asymmetry into such electrochemical cyclizations will be highlighted. The chemistry focuses on efforts to influence the course of both electrochemical oxidation and reduction reaction and non-redox based transition metal catalyzed reactions with the surface of an electrode.

Engineering multiple defect sites on ultrathin graphitic carbon nitride for efficiently photocatalytic conversion of lignin into monomeric aromatics via selective C–C bond scission

Lignin depolymerisation via photocatalytic cleavage of the selective interunit linkage in lignin could be a sustainable approach to produce monomeric aromatic chemicals. However, the insufficient investigation of interunit C–C bond fragmentation has obstructed the rational design of efficient photocatalytic system and further limit the yields of aromatic monomers from lignin depolymerisation. Herein, this work developed the ultrathin g-C3N4 with multiple defective sites by simple self-assembly process and in-situ thermal gas-shocking/etching process to catalyse the cleavage of lignin C–C bonds under visible light irradiation. Compared with the pristine g-C3N4, the developed g-C3N4 photocatalyst exhibited a superior catalytic activity (improved 102 %) and selectivity (∼90 %) in the cleavage of C–C bonds in lignin. This study demonstrated that the defects construction and ultrathin structure can optimise the electronic structures of g-C3N4 for better separation and transfer of photoinduced charges. And the control experiments and DFT calculation indicated that the created defect sites can promote the generation of essential reactive radicals (e.g., the activation of O2) and radical intermediates (C–H activation). The present work provides useful insights for the rational use of defect engineering in designing the efficient photocatalytic system for the conversion of lignin into aromatic monomers via the C–C bond cleavage.

Ketyl Radical Coupling Enabled by Polycyclic Aromatic Hydrocarbon Electrophotocatalysts.

Herein, we report a new class of electrophotocatalysts, polycyclic aromatic hydrocarbons, that promote the reduction of unactivated carbonyl compounds to generate versatile ketyl radical intermediates. This catalytic platform enables previously challenging intermolecular ketyl radical coupling reactions, including those that classic reductants (e.g., SmI2/HMPA) have failed to promote. More broadly, this study outlines an approach to fundamentally expand the array of reactive radical intermediates that can be generated via electrophotocatalysis by obviating the need for rapid mesolytic cleavage following substrate reduction.

Elucidation of Underlying Reactivities of Alternating Current Electrosynthesis by Time-resolved Mapping of Short-lived Reactive Intermediates.

Alternating current (AC) electrolysis is an emerging field in synthetic chemistry, while its mechanistic studies are challenged by the effective characterization of the elusive intermediate processes. Herein, we develop an operando electrochemical mass spectrometry platform that allows time-resolved mapping of stepwise electrosynthetic reactive intermediates in both direct current and alternating current modes. By dissecting the key intermediate processes of electrochemical functionalization of arylamines, the unique reactivities of AC electrosynthesis, including minimizing the over-oxidation/reduction through the inverse process, and enabling effective reaction of short-lived intermediates generated by oxidation and reduction in paired electrolysis, were evidenced and verified. Notably, the controlled kinetics of reactive N-centered radical intermediates in multi-step sequential AC electrosynthesis to minimize the competing reactions was discovered. Overall, this work provides direct evidence for the mechanism of AC electrolysis, and clarifies the underlying reasons for its high efficiency, which will benefit the rational design of AC electrosynthetic reactions.

Iodine(III)-Mediated Oxidation of Anilines to Construct Dibenzazepines.

The development of an efficient process that produces bioactive medium-sized N-heterocyclic scaffolds from 2-substituted anilines using either iodosobenzene or (bis(trifluoroacetoxy)iodo)-benzene is reported. The tether between the sulfonamide and the aryl group can be varied to access dihydroacridine-, dibenzazepine-, or dibenzazocine scaffolds. While substitution on the aniline portion is limited to electron-neutral- or electron-poor groups, a broader range of functional groups are tolerated on the ortho-aryl substituent and site selective C-NAr bond formation can be achieved. Preliminary mechanistic investigations suggest that medium-ring formation occurs via radical reactive intermediates.

Enantioselective Dearomatization of Indoles via SmI2-Mediated Intermolecular Reductive Coupling with Ketones

Samarium diiodide (SmI2) mediated reductive coupling reactions are powerful methods for the construction of carbon-carbon bond in organic synthesis. Despite the extensive development in recent decades, successful examples of the corresponding asymmetric reactions remained scarce, probably due to the involvement of highly reactive radical intermediates. In this Article, we report an enantioselective dearomatization of indoles via SmI2-mediated intermolecular reductive coupling with ketones. The utilization of samarium reductant supported by chiral tridentate aminodiol ligands allows the facile synthesis of indoline molecules bearing two contiguous stereogenic centers in high yields (up to 99%) and stereoselectivity (up to 99:1 er and >20:1 dr). Combined experimental and computational investigations suggested that parallel single-electron transfer to each substrate from the chiral samarium reductant allows the radical-radical recombination in an enantioselective manner, which is a unique mechanistic scenario in SmI2-mediated reductive coupling reactions.

Fragile intermediate identification and reactivity elucidation in electrochemical oxidative α-C(sp3)-H functionalization of tertiary amines.

The direct α-C(sp3)-H functionalization of widely available tertiary amines holds promise for the rapid construction of complex amine architectures. The activation of C(sp3)-H bonds through electron transfer and proton transfer by oxidants, photoredox catalysis and electrochemical oxidation have received wide attention recently. In these reactions, the direct capture and identification of the key reactive radical intermediates are technically difficult due to their short life-time. Herein, an online electrochemical mass spectrometry (MS) methodology was utilized to probe the short-lived intermediates in the electrochemical oxidative α-C(sp3)-H functionalization of tertiary amines. The resulting electrochemical oxidation intermediates, α-amino radical cation and iminium cation were successfully detected. Further, the α-amino C(sp3) radical added to the double bond of a phenyl trans-styryl sulfone, yielding another C(sp3) radical that leads to the final vinylation. Based on the mass spectrometric elucidation of the reactivity of the α-amino radical, a scale-up electrochemical radical vinylation methodology was established, with which a large variety of allylic amines with broad functional group tolerance were synthesized.

Recent Developments in Copper(I)‐Catalyzed Enantioselective Alkynylation Reactions via a Radical Process

Comprehensive SummaryAlkynes are one of the most significant functional groups in organic chemistry and great efforts have been made to explore efficient approach for the construction of chiral alkynes. The asymmetric C(sp3)–C(sp) cross‐coupling provides a significant complementary strategy through radical‐initiated process. However, the stereocontrol of highly reactive and unstable radical intermediate has been a challenge for decades. To address this problem, a variety of chiral ligands are developed for initiating the reaction and achieving enantiocontrol of alkyl radicals. This review summarizes recent developments in copper‐catalyzed enantioselective alkynylation of prochiral alkyl radicals and their brief mechanistic studies.

Metal‐ and Oxidant‐Free Electrosynthesis of Heterocycles from 1,2‐Diarylalkene Derivatives

AbstractSynthesis of heterocycles from 1,2‐diarylalkene derivatives through electrosynthesis under metal‐ and oxidant‐free conditions has been discovered. Cathodic reduction of 2‐alkenylbenzoic acid or anodic oxidation of 2‐alkenylbenzamide, 2‐alkenylphenol and 2‐alkenylaniline leads to the formation of reactive radical intermediates which afford corresponding phthalide, isochroman‐1‐one, isoindolin‐1‐one, benzofuran, and indole in satisfying yields with good functional group tolerance. Interestingly, different chemoselectivities were found in different reaction solvents. Several mechanistic investigations including cyclic voltammetry studies and control experiments were carried out to elucidate the reaction mechanisms.magnified image

Divergent Auto‐oxidative Alkylation and Alkanoacylation of Quinoxalin‐2(1H)‐ones with Aliphatic Aldehydes

AbstractAuto‐oxidation of aliphatic aldehydes generates alkyl radicals and alkanoyl radicals, which can be intercepted to generate carbon‐carbon bonds. However, the highly reactive and unstable radical intermediates make the controlling selectivity alkylation and alkanoacylation especially challenging. Herein, we report a sustainable and controllable protocol for the auto‐oxidative alkylation and alkanoacylation of quinoxalin‐2(1H)‐ones with good reactivity and selectivity. This protocol was presented in a green and sustainable manner, using oxygen as the only oxidant, avoiding the use of catalysts and explosive oxidants. It offers a new paradigm for a convenient, green synthesis of alkyl and alkanoyl compounds from abundant aliphatic aldehydes.
