The effect of quaternary ammonium polyethylenimine nanoparticles on bacterial adherence, cytotoxicity, and physical and mechanical properties of experimental dental composites

A significant problem related to the functioning of resin-based composites for dental fillings is secondary or recurrent caries, which is the reason for the need for repeated treatment. The cross-linked quaternary ammonium polyethylenimine nanoparticles (QA-PEI-NPs) have been shown to be a promising antibacterial agent against different bacteria, including cariogenic ones. However, little is known about the properties of dental dimethacrylate polymer-based composites enriched with QA-PEI-NPs. This research was carried out on experimental composites based on bis-GMA/UDMA/TEGDMA matrix enriched with 0.5, 1, 1.5, 2 and 3 (wt%) QA-PEI-NPs and reinforced with two glass fillers. The cured composites were tested for their adherence of Streptococcus Mutans bacteria, cell viability (MTT assay) with 48 h and 10-days extracts , degree of conversion (DC), water sorption (WSO), and solubility (WSL), water contact angle (CA), flexural modulus (E), flexural strength (FS), compressive strength (CS), and Vickers microhardness (HV). The investigated materials have shown a complete reduction in bacteria adherence and satisfactory biocompatibility. The QA-PEI-NPs additive has no effect on the DC, VH, and E values. QA-PEI-NPs increased the CA (a favorable change), the WSO and WSL (unfavorable changes) and decreased flexural strength, and compressive strength (unfavorable changes). The changes mentioned were insignificant and acceptable for most composites, excluding the highest antibacterial filler content. Probably the reason for the deterioration of some properties was low compatibility between filler particles and the matrix; therefore, it is worth extending the research by surface modification of QA-PEI-NPs to achieve the optimum performance characteristics.

Physicochemical and Mechanical Properties of Bis-GMA/TEGDMA Dental Composite Resins Enriched with Quaternary Ammonium Polyethylenimine Nanoparticles

Modification of dental monomer compositions with antimicrobial agents must not cause deterioration of the structure, physicochemical, or mechanical properties of the resulting polymers. In this study, 0.5, 1, and 2 wt.% quaternary ammonium polyethylenimine nanoparticles (QA-PEI-NPs) were obtained and admixed with a Bis-GMA/TEGDMA (60:40) composition. Formulations were then photocured and tested for their degree of conversion (DC), polymerization shrinkage (S), glass transition temperature (Tg), water sorption (WS), solubility (SL), water contact angle (WCA), flexural modulus (E), flexural strength (σ), hardness (HB), and impact resistance (an). We found that the DC, S, Tg, WS, E, and HB were not negatively affected by the addition of QA-PEI-NPs. Changes in these values rarely reached statistical significance. On the other hand, the SL increased upon increasing the QA-PEI-NPs concentration, whereas σ and an decreased. These results were usually statistically significant. The WCA values increased slightly, but they remained within the range corresponding to hydrophilic surfaces. To conclude, the addition of 1 wt.% QA-PEI-NPs is suitable for applications in dental materials, as it ensures sufficient physicochemical and mechanical properties.

Nanoparticles of Quaternary Ammonium Polyethylenimine Derivatives for Application in Dental Materials.

Various quaternary ammonium polyethylenimine (QA-PEI) derivatives have been synthesized in order to obtain nanoparticles. Due to their antibacterial activity and non-toxicity towards mammalian cells, the QA-PEI nanoparticles have been tested extensively regarding potential applications as biocidal additives in various dental composite materials. Their impact has been examined mostly for dimethacrylate-based restorative materials; however, dental cements, root canal pastes, and orthodontic adhesives have also been tested. Results of those studies showed that the addition of small quantities of QA-PEI nanoparticles, from 0.5 to 2 wt.%, led to efficient and long-lasting antibacterial effects. However, it was also discovered that the intensity of the biocidal activity strongly depended on several chemical factors, including the degree of crosslinking, length of alkyl telomeric chains, degree of N-alkylation, degree of N-methylation, counterion type, and pH. Importantly, the presence of QA-PEI nanoparticles in the studied dental composites did not negatively impact the degree of conversion in the composite matrix, nor its mechanical properties. In this review, we summarized these features and functions in order to present QA-PEI nanoparticles as modern and promising additives for dental materials that can impart unique antibacterial characteristics without deteriorating the products’ structures or mechanical properties.

Antibacterial Activity of Orthodontic Cement Containing Quaternary Ammonium Polyethylenimine Nanoparticles Adjacent to Orthodontic Brackets.

Enamel demineralization is a common problem found in patients using orthodontic devices, such as orthodontic braces. It was found that Streptoccocus mutans growth increases adjacent to orthodontic devices, which may result in caries development. Incorporated antibacterial quaternary ammonium polyethylenimine (QPEI) nanoparticles were previously shown to be highly efficacious against various bacteria. Combining antibacterial materials in orthodontic cement may be advantageous to prevent bacterial outgrowth adjacent to orthodontic brackets. The aim was to evaluate the efficiency of orthodontic cement containing QPEI nanoparticles in reducing S. mutans and Lactobacillus casei outgrowth adjacent to orthodontic brackets. Orthodontic brackets were bonded to the buccal surfaces of extracted lower incisors. The antibacterial effect on S. mutans and L. casei outgrowth of Neobond bracket adhesive orthodontic cement with and without QPEI nanoparticles was compared. The antibacterial effect was evaluated using crystal violet staining and bacterial count (CFU/mL). The teeth in the experimental group, with the QPEI nanoparticles cement, showed significantly lower optical density (OD) values and CFU counts of S. mutans and L. casei than the teeth in the control group (p < 0.05). Based on the results, it can be concluded that orthodontic cement containing QPEI nanoparticles significantly inhibits S. mutans and L. casei growth around orthodontic brackets.

Antibiofilm Effects of Endodontic Sealers Containing Quaternary Ammonium Polyethylenimine Nanoparticles

IntroductionThis study evaluated the antibiofilm effects of 2 endodontic sealers incorporated with quaternary ammonium polyethylenimine (QPEI) nanoparticles at a 2% concentration (w/w). MethodsThe materials tested were AH Plus and Pulp Canal Sealer EWT (PCS) in the commercial unmodified form or containing 2% QPEI. Antibiofilm assays were conducted by using direct-contact and membrane-restricted tests for evaluation of bacterial viability in biofilms grown onto membranes or paper disks and the crystal violet microtiter-plate assay to evaluate the effects of sealer extracts on the biofilm biomass. Two Enterococcus faecalis strains (ATCC and an endodontic isolate) were used. ResultsDirect contact and membrane-restricted antibiofilm tests revealed that PCS 2% was the only material to promote total killing of E. faecalis ATCC biofilms. All the materials significantly reduced bacterial counts in E. faecalis ATCC biofilms when compared with the positive control in both tests (P < .05). In the direct test against E. faecalis RW35, PCS 2% was significantly more effective than the other materials and was the only one that showed significantly lower counts than the positive control (P < .05). In the crystal violet assay, only AH Plus 2% presented optical density readings significantly lower than the positive control of the ATCC strain (P < .05). No other significant effects on the biofilm biomass of the 2 E. faecalis strains were observed for any of the sealers tested (P > .05). ConclusionsAddition of QPEI nanoparticles improved the killing ability of PCS against biofilms of both E. faecalis strains and the effects of AH Plus on the biomass of biofilms from the ATCC strain.

Antibacterial, physicochemical and mechanical properties of endodontic sealers containing quaternary ammonium polyethylenimine nanoparticles

To evaluate the antibacterial, physicochemical and mechanical properties of two endodontic sealers incorporating quaternary ammonium polyethylenimine (QPEI) nanoparticles at concentrations of 1% and 2% (w/w). The sealers tested were AH Plus and Pulp Canal Sealer EWT in the commercial unmodified form or containing 1% or 2% QPEI. Antibacterial activity was evaluated using a direct contact test (DCT) against Enterococcus faecalis ATCC 29212 and two endodontic isolates (RW35 and RN44). Sealers freshly mixed or set for 7days were exposed to bacterial suspensions for 10, 30 and 60min. Setting time, flow test, solubility, apparent porosity, dimensional change following setting, wettability, zeta potential and compressive strength were assessed according to the International Standard Organization 6876:2001 (ISO 6876). DCT results revealed that both freshly prepared sealers had antibacterial effects unaffected by QPEI incorporation. Both unmodified sealers had lost much of their antibacterial effects after 7days. However, for Pulp Canal Sealer EWT incorporated with 1% and 2% QPEI nanoparticles, the antibacterial effects against all test E.faecalis strains within 30 and 60min of contact were significantly greater than the unmodified formula. Addition of QPEI resulted in no significant increase in the antibacterial effects of AH Plus after ageing. As for physicochemical and mechanical tests, setting time, wettability and zeta potential were influenced by the presence of QPEI nanoparticles. Incorporation of QPEI nanoparticles can improve the long-term antibacterial activity of Pulp Canal Sealer EWT without relevant changes in physicochemical and mechanical properties.

Antibacterial Activity of Dental Cements Containing Quaternary Ammonium Polyethylenimine Nanoparticles

Glass ionomer cements (GICs) are commonly used for cementing full cast crown restorations. Regrettably, although the dental cements fill the gap between the tooth and the crown, bacterial microleakage may occur, resulting in secondary caries. As microleakage cannot be completely prevented, GICs possessing antibacterial properties are in demand. In the present study the antibacterial activity of insoluble, cross‐linked quaternary ammonium polyethylenimine (QPEI) nanoparticles incorporated at 1% w/w in two clinically available GICs were studied. The antibacterial activity was tested against Streptococcus mutans and Lactobacillus casei using the direct contact test (DCT) and the agar diffusion test (ADT). Using the direct contact test, antibacterial activity (P &lt; 0.05) was found in both tested GICs with incorporated QPEI nanoparticles, the effect lasting for at least one month. However, the ADT showed no inhibition halo in the test bacteria, indicating that the antimicrobial nanoparticles do not diffuse into the agar. The results show that the incorporation of QPEI nanoparticles in glass ionomer cements has a long‐lasting antibacterial effect against Streptococcus mutans and Lactobacillus casei. Changing the antibacterial properties of glass ionomer cements by incorporating QPEI antibacterial nanoparticles may prolong the clinical performance of dental crowns.

Polyethyleneimine nanoparticles incorporated into resin composite cause cell death and trigger biofilm stress in vivo

Incorporation of cross-linked quaternary ammonium polyethylenimine (QPEI) nanoparticles in dental resin composite has a long-lasting and wide antimicrobial effect with no measured impact on biocompatibility in vitro. We hypothesized that QPEI nanoparticles incorporated into a resin composite have a potent antibacterial effect in vivo and that this stress condition triggers a suicide module in the bacterial biofilm. Ten volunteers wore a removable acrylic appliance, in which two control resin composite specimens and two resin composite specimens incorporating 1% wt/wt QPEI nanoparticles were inserted to allow the buildup of intraoral biofilms. After 4 h, the specimens were removed and tested for bacterial vitality and biofilm thickness, using confocal laser scanning microscopy. The vitality rate in specimens incorporating QPEI was reduced by > 50% (p < 0.00001), whereas biofilm thickness was increased (p < 0.05). The ability of the biofilm supernatant to restore bacterial death was tested in vitro. The in vitro tests showed a 70% decrease in viable bacteria (p < 0.05). Biofilm morphological differences were also observed in the scanning electron microscope micrographs of the resin composite versus the resin composite incorporating QPEI. These results strongly suggest that QPEI nanoparticles incorporated at a low concentration in resin composite exert a significant in vivo antibiofilm activity and exhibit a potent broad spectrum antibacterial activity against salivary bacteria.

Antibacterial activity of dental composites containing quaternary ammonium polyethylenimine nanoparticles against Streptococcus mutans

The antibacterial activity of quaternary ammonium polyethylenimine (PEI) nanoparticles embedded at 1% w/w with clinically used bonding, flowable and hybrid dental composite resins and cured by light polymerization was studied. The antibacterial activity was tested with Streptoccocus mutans by: (i) the agar diffusion test (ADT); (ii) the direct contact test; (iii) bacterial growth in the materials elute; (iv) and scanning electron microscope (SEM). Using the direct contact test, antibacterial activity ( p<0.001) was found in all three types of composite resins incorporated with the synthesized nanoparticles. The effect lasted for at least 1 month. SEM demonstrated bacterial debris and no streptococcal chains at 24 h of bacterial contact. The addition of 1% w/w of nanoparticles did not affect the flexural modulus and the flexural strength of the dental composite materials.The results indicate that quaternary ammonium PEI nanoparticles immobilized in resin-based materials have a strong antibacterial activity upon contact without leach-out of the nanoparticles and without compromise in mechanical properties.
