Photosystem II Tolerance to Excess Zinc Exposure and High Light Stress in Salvia sclarea L.

High light (HL) intensity has a substantial impact on light energy flow and partitioning within photosynthetic apparatus. To realize the impact of HL intensity on zinc (Zn) tolerance mechanisms in clary sage (Salvia sclarea L., Lamiaceae) plants, we examined the effect of the altered chlorophyll and nutrient uptake under excess Zn supply on the response mechanism of photosystem II (PSII) photochemistry. Eight-week-old clary sage plants were treated with 5 μM Zn (control) or 900 μM Zn in Hoagland nutrient solution. Leaf elemental analysis for Zn, Mn, Mg, and Fe was performed by inductively coupled plasma mass spectrometry (ICP-MS), whereas PSII functioning under HL was evaluated by chlorophyll fluorescence imaging analysis. Exposure of S. sclarea plants to 900 μM Zn increased leaf Zn accumulation and decreased leaf Mg and chlorophyll. The decreased non-photochemical quenching (NPQ) provided evidence of the photoprotection offered by the smaller light-harvesting antennae due to the reduced chlorophyll. The increased Mn after Zn exposure corresponded with higher efficiency of the oxygen-evolving complex (OEC) that was significantly correlated with the maximum efficiency of photosystem II (PSII) photochemistry (Fv/Fm). An increased electron transport rate (ETR) coincided with increased leaf Fe, which is known to play a vital role in the enzymes engaged in ETR. The decreased (32%) NPQ after an 8-day exposure to Zn caused an increased (10%) quantum yield of non-regulated energy loss in PSII (ΦNO), indicative of an increased singlet oxygen (1O2) production. It is suggested that the decreased NPQ induced acclimation responses of clary sage plants to HL and excess Zn by increasing 1O2 production. The reduced (18%) excess excitation energy (EXC) at PSII and the increased (24%) quantum yield of PSII photochemistry (ΦPSII) and ETR indicated improved photosynthetic efficiency under excess Zn and HL intensity. Therefore, the exposure of medicinal plants to excess Zn not only boosts their photosynthetic efficiency, enhancing crop yields, but can also improve Fe and Zn content, ameliorating the human health deficiency of these two essential micronutrients.

Mechanistic Approach on Melatonin-Induced Hormesis of Photosystem II Function in the Medicinal Plant Mentha spicata

Melatonin (MT) is considered a new plant hormone having a universal distribution from prokaryotic bacteria to higher plants. It has been characterized as an antistress molecule playing a positive role in the acclimation of plants to stress conditions, but its impact on plants under non-stressed conditions is not well understood. In the current research, we evaluated the impact of MT application (10 and 100 μM) on photosystem II (PSII) function, reactive oxygen species (ROS) generation, and chlorophyll content on mint (Mentha spicata L.) plants in order to elucidate the molecular mechanism of MT action on the photosynthetic electron transport process that under non-stressed conditions is still unclear. Seventy-two hours after the foliar spray of mint plants with 100 μM MT, the improved chlorophyll content imported a higher amount of light energy capture, which caused a 6% increase in the quantum yield of PSII photochemistry (ΦPSII) and electron transport rate (ETR). Nevertheless, the spray with 100 μM MT reduced the efficiency of the oxygen-evolving complex (OEC), causing donor-side photoinhibition, with a simultaneous slight increase in ROS. Even so, the application of 100 μM MT decreased the excess excitation energy at PSII implying superior PSII efficiency. The decreased excitation pressure at PSII, after 100 μM MT foliar spray, suggests that MT induced stomatal closure through ROS production. The response of ΦPSII to MT spray corresponds to a J-shaped hormetic curve, with ΦPSII enhancement by 100 μM MT. It is suggested that the hormetic stimulation of PSII functionality was triggered by the non-photochemical quenching (NPQ) mechanism that stimulated ROS production, which enhanced the photosynthetic function. It is concluded that MT molecules can be used under both stress and non-stressed conditions as photosynthetic biostimulants for enhancing crop yields.

Impact of Coated Zinc Oxide Nanoparticles on Photosystem II of Tomato Plants.

Zinc oxide nanoparticles (ZnO NPs) have emerged as a prominent tool in agriculture. Since photosynthetic function is a significant measurement of phytotoxicity and an assessment tool prior to large-scale agricultural applications, the impact of engineered irregular-shaped ZnO NPs coated with oleylamine (ZnO@OAm NPs) were tested. The ZnO@OAm NPs (crystalline size 19 nm) were solvothermally prepared in the sole presence of oleylamine (OAm) and evaluated on tomato (Lycopersicon esculentum Mill.) photosystem II (PSII) photochemistry. Foliar-sprayed 15 mg L-1 ZnO@OAm NPs on tomato leaflets increased chlorophyll content that initiated a higher amount of light energy capture, which resulted in about a 20% increased electron transport rate (ETR) and a quantum yield of PSII photochemistry (ΦPSII) at the growth light (GL, 600 μmol photons m-2 s-1). However, the ZnO@OAm NPs caused a malfunction in the oxygen-evolving complex (OEC) of PSII, which resulted in photoinhibition and increased ROS accumulation. The ROS accumulation was due to the decreased photoprotective mechanism of non-photochemical quenching (NPQ) and to the donor-side photoinhibition. Despite ROS accumulation, ZnO@OAm NPs decreased the excess excitation energy of the PSII, indicating improved PSII efficiency. Therefore, synthesized ZnO@OAm NPs can potentially be used as photosynthetic biostimulants for enhancing crop yields after being tested on other plant species.

Hormesis Responses of Photosystem II in Arabidopsis thaliana under Water Deficit Stress.

Since drought stress is one of the key risks for the future of agriculture, exploring the molecular mechanisms of photosynthetic responses to water deficit stress is, therefore, fundamental. By using chlorophyll fluorescence imaging analysis, we evaluated the responses of photosystem II (PSII) photochemistry in young and mature leaves of Arabidopsis thaliana Col-0 (cv Columbia-0) at the onset of water deficit stress (OnWDS) and under mild water deficit stress (MiWDS) and moderate water deficit stress (MoWDS). Moreover, we tried to illuminate the underlying mechanisms in the differential response of PSII in young and mature leaves to water deficit stress in the model plant A. thaliana. Water deficit stress induced a hormetic dose response of PSII function in both leaf types. A U-shaped biphasic response curve of the effective quantum yield of PSII photochemistry (ΦPSII) in A. thaliana young and mature leaves was observed, with an inhibition at MiWDS that was followed by an increase in ΦPSII at MoWDS. Young leaves exhibited lower oxidative stress, evaluated by malondialdehyde (MDA), and higher levels of anthocyanin content compared to mature leaves under both MiWDS (+16%) and MoWDS (+20%). The higher ΦPSII of young leaves resulted in a decreased quantum yield of non-regulated energy loss in PSII (ΦNO), under both MiWDS (-13%) and MoWDS (-19%), compared to mature leaves. Since ΦNO represents singlet-excited oxygen (1O2) generation, this decrease resulted in lower excess excitation energy at PSII, in young leaves under both MiWDS (-10%) and MoWDS (-23%), compared to mature leaves. The hormetic response of PSII function in both young and mature leaves is suggested to be triggered, under MiWDS, by the intensified reactive oxygen species (ROS) generation, which is considered to be beneficial for activating stress defense responses. This stress defense response that was induced at MiWDS triggered an acclimation response in A. thaliana young leaves and provided tolerance to PSII when water deficit stress became more severe (MoWDS). We concluded that the hormesis responses of PSII in A. thaliana under water deficit stress are regulated by the leaf developmental stage that modulates anthocyanin accumulation in a stress-dependent dose.

Effects of exogenous NO on the growth and photosynthetic fluorescence characteristics of ryegrass seedlings under B[a]P stress

Benzoapyrene (B[a]P) pollution poses a threat to the environment and the food chain and consequently to human health. However, the alleviation of the harmful effects of B[a]P pollution in perennial ryegrass (Lolium perenne L.) by the application of exogenous nitric oxide (NO) has been ignored. Thus, in this paper the effects of exogenous sodium nitroprusside (SNP, a NO donor) on the growth, photosynthetic fluorescence characteristics, and antioxidant enzyme activity of ryegrass exposed to B[a]P stress are investigated. B[a]P stress induced the reduction of the aboveground and belowground dry weights, chlorophyll (a, b), the total chlorophyll contents, the carotenoid content, the net photosynthetic rate (Pn), the intercellular carbon dioxide concentration (Ci), the water use efficiency (WUE), the photosystem II (PSII) potential activity (Fv/F0), the maximum quantum yield of PSII photochemistry (Fv/Fm), the steady-state fluorescence yield (Fs), and the non-photochemical quenching (qN), while enhancement was recorded in response to the foliar spray of SNP at 200 and 300 μmol·L-1 under B[a]P stress. Gray correlation and principal component analyses show that 200 μmol·L 1 of SNP more drastically alleviated the damage caused by B[a]P stress than 300 μmol·L 1 of SNP. The exogenous NO-mediated alleviation of B[a]P toxicity in ryegrass was associated with preserved photosynthetic characteristics and activation of antioxidant enzymes.

Mechanistic Insights on Salicylic Acid Mediated Enhancement of Photosystem II Function in Oregano Seedlings Subjected to Moderate Drought Stress.

Dramatic climate change has led to an increase in the intensity and frequency of drought episodes and, together with the high light conditions of the Mediterranean area, detrimentally influences crop production. Salicylic acid (SA) has been shown to supress phototoxicity, offering photosystem II (PSII) photoprotection. In the current study, we attempted to reveal the mechanism by which SA is improving PSII efficiency in oregano seedlings under moderate drought stress (MoDS). Foliar application of SA decreased chlorophyll content under normal growth conditions, but under MoDS increased chlorophyll content, compared to H2O-sprayed oregano seedlings. SA improved the PSII efficiency of oregano seedlings under normal growth conditions at high light (HL), and under MoDS, at both low light (LL) and HL. The mechanism by which, under normal growth conditions and HL, SA sprayed oregano seedlings compared to H2O-sprayed exhibited a more efficient PSII photochemistry, was the increased (17%) fraction of open PSII reaction centers (qp), and the increased (7%) efficiency of these open reaction centers (Fv'/Fm'), which resulted in an enhanced (24%) electron transport rate (ETR). SA application under MoDS, by modulating chlorophyll content, resulted in optimized antenna size and enhanced effective quantum yield of PSII photochemistry (ΦPSII) under both LL (7%) and HL (25%), compared to non-SA-sprayed oregano seedlings. This increased effective quantum yield of PSII photochemistry (ΦPSII) was due to the enhanced efficiency of the oxygen evolving complex (OEC), and the increased fraction of open PSII reaction centers (qp), which resulted in an increased electron transport rate (ETR) and a lower amount of singlet oxygen (1O2) production with less excess excitation energy (EXC).

Temporal heterogeneity in photosystem II photochemistry in Artemisia ordosica under a fluctuating desert environment.

Acclimation strategies in xerophytic plants to stressed environmental conditions vary with temporal scales. Our understanding of environmentally-induced variation in photosystem II (PSII) processes as a function of temporal scales is limited, as most studies have thus far been based on short-term, laboratory-controlled experiments. In a study of PSII processes, we acquired near-continuous, field-based measurements of PSII-energy partitioning in a dominant desert-shrub species, namely Artemisia ordosica, over a six-year period from 2012-2017. Continuous-wavelet transformation (CWT) and wavelet coherence analyses (WTC) were employed to examine the role of environmental variables in controlling the variation in the three main PSII-energy allocation pathways, i.e., photochemical efficiency and regulated and non-regulated thermal dissipation, i.e., Φ PSII, Φ NPQ, and Φ NO, respectively, across a time-frequency domain from hours to years. Convergent cross mapping (CCM) was subsequently used to isolate cause-and-effect interactions in PSII-energy partitioning response. The CWT method revealed that the three PSII-energy allocation pathways all had distinct daily periodicities, oscillating abruptly at intermediate timescales from days to weeks. On a diurnal scale, WTC revealed that all three pathways were influenced by photosynthetically active radiation (PAR), air temperature (T a), and vapor pressure deficit (VPD). By comparing associated time lags for the three forms of energy partitioning at diurnal scales, revealed that the sensitivity of response was more acutely influenced by PAR, declining thereafter with the other environmental variables, such that the order of influence was greatest for T a, followed by VPD, and then soil water content (SWC). PSII-energy partitioning on a seasonal scale, in contrast, displayed greater variability among the different environmental variables, e.g., Φ PSII and Φ NO being more predisposed to changes in T a, and Φ NPQ to changes in VPD. CCM confirmed the causal relationship between pairings of PSII-energy allocation pathways, according to shrub phenology. A. ordosica is shown to have an innate ability to (i) repair damaged PSII-photochemical apparatus (maximum quantum yield of PSII photochemistry, with F v/F m > 0.78), and (ii) acclimatize to excessive PAR, dry-air conditions, and prolonged drought. A. ordosica is relatively sensitive to extreme temperature and exhibits photoinhibition.

Paclobutrazol Ameliorates Low-Light-Induced Damage by Improving Photosynthesis, Antioxidant Defense System, and Regulating Hormone Levels in Tall Fescue.

Paclobutrazol (PBZ) is a plant-growth regulator (PGR) in the triazole family that enhances plant tolerance to environmental stresses. Low-light (LL) intensity is a critical factor adversely affecting the growth of tall fescue (Festuca arundinacea Schreb.). Therefore, in this study, tall fescue seedlings were treated with PBZ under control and LL conditions to investigate the effects of PBZ on enhancing LL stress resistance by regulating the growth, photosynthesis, oxidative defense, and hormone levels. Our results reveal that LL stress reduced the total biomass, chlorophyll (Chl) content, photosynthetic capacity, and photochemical efficiency of photosystem II (PSII) but increased the membrane lipid peroxidation level and reactive oxygen species (ROS) generation. However, the application of PBZ increased the photosynthetic pigment contents, net photosynthetic rate (Pn), maximum quantum yield of PSII photochemistry (Fv/Fm), ribulose-1,5-bisphosphate carboxylase (RuBisCO) activity, and starch content. In addition, PBZ treatment activated the antioxidant enzyme activities, antioxidants contents, ascorbate acid-glutathione (AsA-GSH) cycle, and related gene expression, lessening the ROS burst (H2O2 and O2∙−). However, the gibberellic acid (GA) anabolism was remarkably decreased by PBZ treatment under LL stress, downregulating the transcript levels of kaurene oxidase (KO), kaurenoic acid oxidase (KAO), and GA 20-oxidases (GA20ox). At the same time, PBZ treatment up-regulated 9-cis-epoxycarotenoid dioxygenase (NCED) gene expression, significantly increasing the endogenous abscisic acid (ABA) concentration under LL stress. Thus, our study revealed that PBZ improves the antioxidation and photosynthetic capacity, meanwhile increasing the ABA concentration and decreasing GA concentration, which ultimately enhances the LL stress tolerance in tall fescue.

Using the diurnal variation characteristics of effective quantum yield of PSII photochemistry for drought stress detection in maize

The importance of detecting drought stress in crops to alleviate the pressure of the growing population and food demand is well recognized. Although chlorophyll fluorescence (ChlF) is closely related to photosynthesis, it is always disturbed by the intensity of irradiance during the measurement. To detect drought stress more effectively with ChlF, we studied the diurnal relationship of the effective quantum yield of photosystem II (PSII) photochemistry (ΦPSII) and photosynthetically active radiation (PAR). We measured the diurnal ΦPSII, leaf temperature (Tleaf) and PAR of maize leaves (ZhengDan958) by pulse-amplitude modulated (PAM) fluorometry under different drought stresses. We found that in the absence of drought stress, the linearity (indicated by R2) between diurnal ΦPSII and PAR was strong. When drought stress was gradually aggravated, the difference between the ΦPSII values in the morning and those in the afternoon under similar PAR values gradually became significant. Therefore, the R2 values of the linear correlation of the diurnal ΦPSII-PAR regression gradually decreased under increased drought stress. The simulation results of a photosynthesis model showed that the diurnal variation in the response of ΦPSII-PAR during drought may be the result of the fluctuation of the substrate saturated Rubisco capacity (Vcmax) , which was dominated by the dynamics of Tleaf. The results indicated that the linearity of diurnal ΦPSII-PAR regression can be used as an effective and convenient field indicator to monitor drought stress.

A Comparison of Photoprotective Mechanism in Different Light-Demanding Plants Under Dynamic Light Conditions.

Light intensity is highly heterogeneous in nature, and plants have evolved a series of strategies to acclimate to dynamic light due to their immobile lifestyles. However, it is still unknown whether there are differences in photoprotective mechanisms among different light-demanding plants in response to dynamic light, and thus the role of non-photochemical quenching (NPQ), electron transport, and light energy allocation of photosystems in photoprotection needs to be further understood in different light-demanding plants. The activities of photosystem II (PSII) and photosystem I (PSI) in shade-tolerant species Panax notoginseng, intermediate species Polygonatum kingianum, and sun-demanding species Erigeron breviscapus were comparatively measured to elucidate photoprotection mechanisms in different light-demanding plants under dynamic light. The results showed that the NPQ and PSII maximum efficiency (Fv′/Fm′) of E. breviscapus were higher than the other two species under dynamic high light. Meanwhile, cyclic electron flow (CEF) of sun plants is larger under transient high light conditions since the slope of post-illumination, P700 dark reduction rate, and plastoquinone (PQ) pool were greater. NPQ was more active and CEF was initiated more readily in shade plants than the two other species under transient light. Moreover, sun plants processed higher quantum yield of PSII photochemistry (ΦPSII), quantum yield of photochemical energy conversion [Y(I)], and quantum yield of non-photochemical energy dissipation due to acceptor side limitation (Y(NA), while the constitutive thermal dissipation and fluorescence (Φf,d) and quantum yield of non-photochemical energy dissipation due to donor side limitation [Y(ND)] of PSI were higher in shade plants. These results suggest that sun plants had higher NPQ and CEF for photoprotection under transient high light and mainly allocated light energy through ΦPSII and ΦNPQ, while shade plants had a higher Φf,d and a larger heat dissipation efficiency of PSI donor. Overall, it has been demonstrated that the photochemical efficiency and photoprotective capacity are greater in sun plants under transient dynamic light, while shade plants are more sensitive to transient dynamic light.

Protective Effects of Salicylic Acid and Calcium Chloride on Sage Plants (Salvia officinalis L. and Salviaelegans Vahl) under High-Temperature Stress.

High-temperature stress is a major risk to fresh-market Salvia production, and heat intolerance is a major constraint in sage cultivation, particularly during the hot summer season. Previously, we investigated heat tolerance in five common-market cultivars of sage plants using leaf relative injury (RI) values and found that S. elegans Vahl (SE) and S. officinalis L. (SO) were the most and least heat-tolerant species, respectively. The exogenous applications of salicylic acid (SA) and calcium chloride (CaCl2) to alleviate heat stress in various species have been extensively studied, but reports of the effects of SA and CaCl2 treatments on the heat tolerance of sage plants are scarce. The objective of this study was to investigate how SA and CaCl2 affect the physiology and morphology of SE and SO plants under high-temperature conditions. Potted plants were pretreated with SA (0, 100, 200, 400, and 800 μM) and CaCl2 (0, 5, 10, and 15 mM), alone and combined, exposed to 55 °C and 80% humidity for 30 min, then placed in an environment-controlled chamber at 30 °C for three days and evaluated for changes in phenotypic appearance, RI, spectral reflectance, and chlorophyll fluorescence indices at different time intervals. Plants watered without chemical solutions were used as controls. Our results show that the growth of SO plants pretreated with SA and CaCl2 was more robust, compared with control plants, which were considerably affected by heat stress, resulting in brown, withered leaves and defoliation. The effects of the combined applications of SA (100 μM) and CaCl2 (5 mM) to SO plants were superior to control plants in increasing values of soil-plant analysis development (SPAD), normalized difference vegetation index (NDVI), and the maximal quantum yield of photosystemII photochemistry (Fv/Fm), while reducing RI%. Furthermore, SO plants exhibited higher SPAD and Fv/Fm values and lower RI% than SE plants in combined treatments at all time intervals after heat stress, implying that different genotypes displayed variations in their SPAD, Fv/Fm, and RI%. Thus, a combined treatment of 100 μM of SA and 5 mM of CaCl2 is effective and beneficial to plant appearance and ability to ameliorate heat stress. These indices can be used as indicators to characterize the physiology of these plants and applied on a commercial scale for informing the development of rapid and precise management practices on bedded sage plants grown in plant factories to achieve maximum market benefit.

Physiological Mechanism of Drought-Resistant Rice Coping With Drought Stress

Drought stress is one of the major threats to rice production. The weakening of leaf photosynthesis due to drought is the main reason for the reduction of grain yield, but its mechanism is still obscure. The objectives of this study were to reveal the physiological mechanism of drought stress affecting photosynthetic capacity and grain yield. Pot experiments were conducted with drought-tolerant cultivars Hanyou113 (HY113) and Zhonghan3 (ZH3) and drought-sensitive cultivar Huanghuazhan (HHZ) under four water management treatments (traditional flooding (CK), mild drought stress (LD), moderate drought stress (MD) and severe drought stress (HD)) at heading stage in 2013 and 2014. Compared with CK, grain yield was significantly reduced by 14.9%, 30.8%, and 12.8% in HY113, HHZ, and ZH3 under LD, 32.9%, 33.7%, and 22.9% in HY113, HHZ and ZH3 under MD and 53.6%, 45.6%, and 30.7% in HY113, HHZ, and ZH3 under HD, respectively. The photosynthetic rate (Pn) decreased by 49.0% from 20.0 to 10.2 µmol m−2 s−1 in HY113, and 67.6% from 23.4 to 7.58 µmol m−2 s−1 in HHZ, and 39.3% from 23.4 to 14.2 µmol m−2 s−1 in ZH3 under HD. The Pn of HHZ was similar to that of ZH3 under CK conditions. During the drought periods from LD to HD at heading stage, the leaf water potential (LWP) reduced 31.9%, 54.8%, and 15.7% in HY113, HHZ, and ZH3, respectively. The non-photochemical quenching (NPQ) of HY113, HHZ, and ZH3 flag leaves increased by 150%, 97.6%, and 218%, respectively. The effective quantum yield of PSII photochemistry (ΦPSII) of flag leaves reduced by 20.3%, 11.9%, and 22.1% in HY113, HHZ, and ZH3, respectively. The enzymatic activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) increased by 11.4%, 18.0%, and 21.8% in HY113, and 13.2%, 14.3%, and 30.9% in HHZ, and 13.4% 21.7%, and 17.6% in ZH3 under MD on average across two seasons. The yield reduction of drought-resistant cultivars (HY113, ZH3) was smaller than that of conventional cultivars (HHZ). Maintaining leaf water potential (LWP), Pn, photosystem II (PSII) original light energy conversion efficiency, non–photochemical quenching coefficient (NPQ), and increasing in the ratio of photochemical reaction energy in fluorescence and antioxidant enzyme activity is the physiological basis to achieve a relatively high photosynthesis. These traits could be the target for breeder to develop drought-tolerant varieties.

The effects of dark treatment on photosynthesis and growth in young and mature leaves of Syzygium rehderianum

Light initiates plant photosynthesis, and plant growth depends on light. In order to explore the effects of long-term dark treatment on photosynthesis and leaf growth of plants, this study compared and analyzed the growth state, pigment content, antioxidant capacity, photosynthetic capacity and chlorophyll fluorescence of the leaves of Syzygium rehderianum Merr. and L.M. Perry under full sunlight (natural full sunlight) and a dark treatment. The results showed that the photosynthesis and development of leaves were significantly inhibited after the dark treatment day 3. After the dark treatment day 6, the length and width of the leaves and the synthesis of photosynthetic pigments were significantly lower than those in leaves under full sunlight. Compared with those under full sunlight, the net photosynthetic rate (Pn), maximal quantum yield of PSII photochemistry (Fv/Fm), nonphotochemical quenching and actual photochemical efficiency (Yield) of young and mature leaves were significantly reduced after the dark treatment day 6. These results indicated that the timing of long-term dark treatment caused stress to the leaves. It directly affected the photosynthesis of leaves by affecting the absorption of light energy, inhibiting the synthesis of photosynthetic pigments and destroying the activity of the photosystem II (PSII) reaction center, which ultimately affected the growth and development of plant leaves.

CO2 uptake and chlorophyll a fluorescence of Suaeda fruticosa grown under diurnal rhythm and after transfer to continuous dark

Although only 2-4% of absorbed light is emitted as chlorophyll (Chl) a fluorescence, its measurement provides valuable information on photosynthesis of the plant, particularly of Photosystem II (PSII) and Photosystem I (PSI). In this paper, we have examined photosynthetic parameters of Suaeda fruticosa L. (family: Amaranthaceae), surviving under extreme xerohalophytic conditions, as influenced by diurnal rhythm or continuous dark condition. We report here CO2 gas exchange and the kinetics of Chl a fluorescence of S. fruticosa, made every 3hours (hrs) for 3days, using a portable infra-red gas analyzer and a Handy PEA fluorimeter. Our measurements on CO2 gas exchange show the maximum rate of photosynthesis to be at 08:00hrs under diurnal condition and at 05:00hrs under continuous dark. From the OJIP phase of Chl a fluorescence transient, we have inferred that the maximum quantum yield of PSII photochemistry must have increased during the night under diurnal rhythm, and between 11:00 and 17:00hrs under constant dark. Overall, our study has revealed novel insights into how photosynthetic reactions are affected by the photoperiodic cycles in S. fruticosa under high salinity. This study has further revealed a unique strategy operating in this xero-halophyte where the repair mechanism for damaged PSII operates during the dark, which, we suggest, contributes to its ecological adaptation and ability to survive and reproduce under extreme saline, high light, and drought conditions. We expect these investigations to help in identifying key genes and pathways for raising crops for saline and dry areas.

Toxic Effects of 2,2′,4,4′-Tetrabromodiphenyl Ether on Chinese Cabbage

2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) is one of the most toxic polybrominated diphenyl ethers (PBDEs). The toxic effects of BDE-47 on Chinese cabbage seedlings were analyzed in this study. After a 30-day hydroponic exposure to BDE-47 at different concentrations (25, 50, 75, and 100 µg·L−1), the fresh weight of Chinese cabbage seedlings was significantly decreased, whereas their root:shoot ratio was increased, indicating that BDE-47 inhibited the growth of the plant, especially the overground parts. The water content, chlorophyll content, and protein content of Chinese cabbage leaves also markedly decreased with the increase of the BDE-47 concentration. In addition, BDE-47 weakened the photosynthetic capacity of the leaves, which was supported by the decreased photosynthetic parameters [net photosynthetic rate (Pn) and stomatal conductance (gS)]. Although the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) in the leaves were enhanced after exposure to BDE-47, the increased malondialdehyde (MDA) content attested to the existence of membrane lipid peroxidation. The increased plasma membrane permeability and the decreased chlorophyll fluorescence parameters [the maximum quantum yield of PSII photochemistry at t = 0 (Fv/Fm), photosystem II (PSII) reaction centers (RCs) per cross section (CS) (RC/CS), absorption energy flux per CS (ABS/CS), trapped energy flux per CS (TRo/CS), electron transport flux per CS (ETo/CS), performance index on the absorption basis (PIabs), and driving force for photosynthesis (DF)] further proved that the plasma membrane and photosynthetic membrane were damaged by BDE-47. Our study demonstrated the phytotoxicities of BDE-47 to Chinese cabbage, which can provide valuable information for understanding the toxicity of BDE-47 on vegetables.

Chlorophyll Composition, Chlorophyll Fluorescence, and Grain Yield Change in esl Mutant Rice.

To evaluate the effect of changes in chlorophyll (Chl) composition and fluorescence on final yield formation, early senescence leaf (esl) mutant rice and its wild-type cultivar were employed to investigate the genotype-dependent differences in Chl composition, Chl fluorescence, and yield characteristics during the grain-filling stage. However, the temporal expression patterns of key genes involved in the photosystem II (PSII) reaction center in the leaves of two rice genotypes were analyzed by quantitative real-time polymerase chain reaction (qRT-PCR). Results showed that the seed-setting rate, 1000-grain weight, and yield per plant remarkably decreased, and the increase in the 1000-grain weight during the grain-filling stage was retarded in esl mutant rice. Chl composition, maximal fluorescence yield (Fm), variable fluorescence (Fv), a maximal quantum yield of PSII photochemistry (Fv/Fm), and net photosynthetic rate (Pn) in esl mutant rice considerably decreased, thereby indicating the weakened abilities of light energy harvesting and transferring in senescent leaves. The esl mutant rice showed an increase in the minimal fluorescence yield (F0) and 1 − Fv/Fm and decreases in the expression levels of light-harvesting Chl a/b binding protein (Cab) and photosystem II binding protein A (PsbA), PsbB, PsbC, and PsbD encoding for the reaction center of the PSII complex during the grain-filling stage. These results indicated the PSII reaction centers were severely damaged in the mesophyll cells of senescent leaves, which resulted in the weakened harvesting quantum photon and transferring light energy to PSI and PSII for carbon dioxide assimilation, leading to enhanced heat dissipation of light energy and a decrease in Pn.

Partitioning of absorbed light energy within photosystem II in barley can be affected by chloroplast movement

Plants have developed many ways to protect reaction centres of photosystems against overexcitation. One of the mechanisms involves reduction of the leaf absorption cross-section by light-induced chloroplast avoidance reaction. Decrease in the probability of photon absorption by the pigments bound within photosystem II (PSII) complexes leads to the increase in quantum yield of PSII photochemistry (ΦPSII). On the other hand, the decrease of PSII excitation probability causes reduction of chlorophyll a fluorescence intensity which is manifested as the apparent increase of determined quantum yield of regulated light-induced non-photochemical quenching (ΦNPQ). Absorption of different light intensity by phototropins led to the different chloroplast distribution within barley leaves, estimated by measurement of the leaf transmittance. Due to a weak blue light used for transmittance measurements, leaves exposed to actinic light with wavelengths longer than 520 nm undergo chloroplast accumulation reaction, in contrast with leaves exposed to light with shorter wavelengths, that showed a different extent of chloroplast avoidance reaction. Based on the ΦNPQ action spectra measured simultaneously with the transmittance, the influence of different chloroplast distribution on ΦNPQ was assessed. The analysis of results showed that decrease in the leaf absorption cross-section due to increasing part of chloroplasts reaching profile position significantly affected the partitioning of excitation energy within PSII and such rearrangement also distorted measured ΦNPQ and cannot be neglected in its interpretation. When the majority of chloroplasts reached profile position, the photoprotective effect appeared to be the most prominent for strong blue light that has the highest absorption in the upper leaf layers in comparison with green or red ones.

Water-stress induced downsizing of light-harvesting antenna complex protects developing rice seedlings from photo-oxidative damage

The impact of water-stress on chloroplast development was studied by applying polyethylene glycol 6000 to the roots of 5-day-old etiolated rice (Oryza sativa) seedlings that were subsequently illuminated up to 72 h. Chloroplast development in drought environment led to down-regulation of light-harvesting Chl-proteins. Photosynthetic proteins of Photosystem II (PSII) and oxygen evolving complex i.e., Cytb559, OEC16, OEC23 and OEC33 as well as those of PSI such as PSI-III, PSI-V, and PSI-VI, decreased in abundance. Consequently, due to reduced light absorption by antennae, the electron transport rates of PSII and PSI decreased by 55% and 25% respectively. Further, seedling development in stress condition led to a decline in the ratio of variable (Fv) to maximum (Fm) Chl a fluorescence, as well in the quantum yield of PSII photochemistry. Addition of Mg2+ to the thylakoid membranes suggested that Mg2+-induced grana stacking was not affected by water deficit. Proteomic analysis revealed the down-regulation of proteins involved in electron transport and in carbon reduction reactions, and up-regulation of antioxidative enzymes. Our results demonstrate that developing seedlings under water deficit could downsize their light-harvesting capacity and components of photosynthetic apparatus to prevent photo-oxidative stress, excess ROS generation and membrane lipid peroxidation.

Aggravation of photoinhibition during variegated leaf development in Actinidia kolomikta (Rupr. & Maxim.) Maxim.

Variegated leaves of Actinidia kolomikta (Rupr. & Maxim.) Maxim. are white, pink and pale green according to the order of leaf development, respectively. However, the pale-green leaves are the most sensitive to high-light stress. To clarify the photoinhibitory mechanism in pale-green leaves, we investigated leaf pigment, spectral properties, gas exchange and chlorophyll fluorescence. During variegated leaf development, the chlorophyll content and light absorption increased rapidly, but only pink leaves contained higher anthocyanin contents. With increasing light absorption, the photosynthetic capacity and the actual quantum yield of photosystem II (PSII) in variegated leaves increased slightly, while the non-photochemical quenching (NPQ) significantly decreased. Under high-light conditions, the maximum quantum yield of PSII photochemistry in white and pink leaves decreased less, while in pale-green leaves it decreased significantly together with a substantial increase in malonaldehyde (MDA) and hydrogen peroxide (H2O2) levels. Therefore, during variegated leaf development, although the light absorption increased significantly in pale-green leaves, the photosynthetic capacity and thermal dissipation were not significantly enhanced, which may result in serious photoinhibition under high-light conditions.

Silicon deposition in roots minimizes the cadmium accumulation and oxidative stress in leaves of cowpea plants.

Silicon (Si) frequently accumulates in plants tissues, mainly in roots of dicotyledons, such as cowpea. By contrast, Cadmium (Cd) is a metal that is extremely toxic to plant metabolism. This research aims to investigate if the deposition of Si in root can reduce Cd contents and minimize its negative effects on leaves, measuring gas exchange, chlorophyll fluorescence, antioxidant metabolism, photosynthetic pigments and growth, which may explain the possible role of Si in the attenuation of Cd toxicity incowpea. This study had a factorial design, with all factors completely randomized and two Cd concentrations (0 and 500µM Cd, termed as -Cd and +Cd, respectively) and three Si concentrations (0, 1.25 and 2.50mM Si). Si reduced Cd contents in the roots and in other plant organs, such as stems and leaves. The Si contents were highest in roots, followed by stems and leaves, which was explained by the passive absorption of Si. The application of Si promoted increase in both the macro- and micronutrient contents in all tissues, suggesting that Si mitigates the effect of Cd on nutrient uptake. Si attenuated Cd-mediated effects on light absorption of photosystem II (PSII), increasing the effective quantum yield of PSII photochemistry and the electron transport rate. Additionally, toxic effects induced by Cd on gas exchange were mitigated by the action of Si. Plants treated with Cd+Si showed increase in the activities of antioxidant enzymes and reductions in oxidant compounds; these modifications were promoted by Si via detoxification mechanisms. Increases in the photosynthetic pigments and growth of plants treated with Si and exposed to Cd stress were detected and were due to the reduced deterioration of cell membranes and maintenance of chloroplasts, which had positive repercussions on growth and development. This study validated the hypothesis that the accumulation of Si in roots induces benefits on metabolism and alleviates the toxic effects caused by Cd in leaves of cowpea.
