Supramolecular Host-Guest Interaction-Driven Electrochemical Recognition for Pyrophosphate and Alkaline Phosphatase Analysis.

We report an electrochemical biosensor based on the supramolecular host-guest recognition between cucurbit[7]uril (CB[7]) and L-phenylalanine-Cu(II) complex for pyrophosphate (PPi) and alkaline phosphatase (ALP) analysis. First, L-Phe-Cu(II) complex is simply synthesized by the complexation of Cu(II) (metal node) with L-Phe (bioorganic ligand), which can be immobilized onto CB[7] modified electrode via host-guest interaction of CB[7] and L-Phe. In this process, the signal of the complex-triggered electro-catalytic reduction of H2 O2 can be captured. Next, due to the strong chelation between PPi and Cu(II), a biosensing system of the model "PPi and Cu(II) premixing, then adding L-Phe" was designed and the platform was applied to PPi analysis by hampering the formation of L-Phe-Cu(II) complex. Along with ALP introduction, PPi can be hydrolyzed to orthophosphate (Pi), where abundant Cu(II) ions are released to form L-Phe-Cu(II) complex, which gives rise to the catalytic reaction of complex to H2 O2 reduction. The quantitative analysis of H2 O2 , PPi and ALP activity was achieved successfully and the detection of limits are 0.067 μM, 0.42 μM and 0.09 mU/mL (S/N=3), respectively. With its high sensitivity and selectivity, cost-effectiveness, and simplicity, our analytical system has great potential to for use in diagnosis and treatment of ALP-related diseases.

A sensitive non-enzymatic electrochemical sensor based on acicular manganese dioxide modified graphene nanosheets composite for hydrogen peroxide detection.

In this work, a novel manganese dioxide-graphene nanosheets (MnO2-GNSs) composite was synthesized by a facile one-step hydrothermal method, in which manganese dioxide (MnO2) was fabricated by hydrothermal reduction of KMnO4 with GNSs. The structure and morphology of MnO2-GNSs composite were characterized by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS), X-ray diffraction (XRD) analysis and X-ray photoelectron spectroscopy (XPS). A sensitive non-enzymatic electrochemical sensor based on MnO2-GNSs composite for the detection of low concentration hydrogen peroxide (H2O2) was fabricated. The electrochemical properties of MnO2-GNSs composite modified glassy carbon electrode (MnO2-GNSs/GCE) were investigated by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and amperometry. The observations confirmed that the fabricated sensor exhibited high electrocatalytic activity for oxidation of H2O2 owing to the catalytic ability of MnO2 particles and the conductivity of GNSs. Under the optimum conditions, the calibration curve was linear for the amperometric response versus H2O2 concentration over the range 0.5-350μM with a low detection limit of 0.19μM (S/N=3) and high sensitivity of 422.10μAmM-1 cm-2. The determination and quantitative analysis of H2O2 in antiseptic solution on MnO2-GNSs/GCE exhibited percent recovery of 96.50%-101.22% with relative standard deviation (RSD) of 1.48%-4.47%. The developed MnO2-GNSs/GCE might be a promising platform for the practical detection of H2O2 due to its prominent properties including excellent reproducibility, good anti-interference and repeatability.

Influence of Polyoxometalate Protecting Ligands on Catalytic Aerobic Oxidation at the Surfaces of Gold Nanoparticles in Water.

Metal oxide cluster-anion (polyoxometalate, or POM) protecting ligands, [α-PW11O39]7- (1), modify the rates at which 14 nm gold nanoparticles (Au NPs) catalyze an important model reaction, the aerobic (O2) oxidation of CO to CO2 in water. At 20 °C and pH 6.2, the following stoichiometry was observed: CO + O2 + H2O = CO2 + H2O2. After control experiments verified that the H2O2 product was sufficiently stable and did not react with 1 under turnover conditions, quantitative analysis of H2O2 was used to monitor the rates of CO oxidation, which increased linearly with the percent coverage of the Au NPs by 1 (0-64% coverage, with the latter value corresponding to 211 ± 19 surface-bound molecules of 1). X-ray photoelectron spectroscopy of Au NPs protected by a series of POM ligands (K+ salts): 1, the Wells-Dawson ion [α-P2W18O62]6- (2) and the monodefect Keggin anion [α-SiW11O39]8- (3) revealed that binding energies of electrons in the Au 4f7/2 and 4f5/2 atomic orbitals decreased as a linear function of the POM charge and percent coverage of Au NPs, providing a direct correlation between the electronic effects of the POMs bound to the surfaces of the Au NPs and the rates of CO oxidation by O2. Additional data show that this effect is not limited to POMs but occurs, albeit to a lesser extent, when common anions capable of binding to Au-NP surfaces, such as citrate or phosphate, are present.

One-pot preparation of SBP-PANI-PAA-ethylene glycol diglycidyl ether sensor for electrochemical detection of H2O2

Soybean seed coat peroxidase (SBP) was used in combination with aniline (ANI), acrylic acid (AA), ethylene glycol diglycidyl ether (EGDE) and H2O2, to prepare a biosensor in one-pot synthesis strategy. SBP catalyzed the chemical polymerization of ANI initiated by H2O2, to give the conductive polymer (PANI) in acidic medium where acrylate acted as dopant. Further reactions between free radicals, AA and the diepoxy EGDE determined the entrapment of the SBP together with PANI in a hydrogel on the electrode surface, becoming wired for the amperometric detection of H2O2 at neutral pH and −100mV vs Ag/AgCl. On the other hand, the absence of AA and EGDE in the reaction mixture led to an unstable sensor modifier and an erratic response to H2O2. The apparent Km for immobilized SBP in the flow system was 0.0763mM, lower than the parameter for the free enzyme at pH 7.0. Quantitative analysis of H2O2 was carried out in a flow injection system (FIA); the calibration plot was linear in the range of 5.0–50μM, with a limit of detection of 2.2μM. A dental bleaching sample was successfully analyzed and the catalase test results demonstrated the specificity of the response.

Preparation of a Superhydrophobic and Peroxidase-like Activity Array Chip for H2O2 Sensing by Surface-Enhanced Raman Scattering.

In this paper, we propose a novel and simple method for preparing a dual-biomimetic functional array possessing both superhydrophobic and peroxidase-like activity that can be used for hydrogen peroxide (H2O2) sensing. The proposed method is an integration innovation that combines the above two properties and surface-enhanced Raman scattering (SERS). We integrated a series of well-ordered arrays of Au points (d = 1 mm) onto a superhydrophobic copper (Cu)/silver (Ag) surface by replicating an arrayed molybdenum template. Instead of using photoresists and the traditional lithography method, we utilized a chemical etching method (a substitution reaction between Cu and HAuCl4) with a Cu/Ag superhydrophobic surface as the barrier layer, which has the benefit of water repellency. The as-prepared Au points were observed to possess peroxidase-like activity, allowing for catalytic oxidation of the chromogenic molecule o-phenylenediamine dihydrochloride (OPD). Oxidation was evidenced by a color change in the presence of H2O2, which allows the array chip to act as an H2O2 sensor. In this study, the water repellency of the superhydrophobic surface was used to fabricate the array chip and increase the local reactant concentration during the catalytic reaction. As a result, the catalytic reaction occurred when only 2 μL of an aqueous sample (OPD/H2O2) was placed onto the Au point, and the enzymatic product, 2,3-diaminophenazine, showed a SERS signal distinguishable from that of OPD after mixing with 2 μL of colloidal Au. Using the dual-biomimetic functional array chip, quantitative analysis of H2O2 was performed by observing the change in the SERS spectra, which showed a concentration-dependent behavior for H2O2. This method allows for the detection of H2O2 at concentrations as low as 3 pmol per 2 μL of sample, which is a considerable advantage in H2O2 analysis. The as-prepared substrate was convenient for H2O2 detection because only a small amount of sample was required in each analysis. Highly sensitive detection was realized using SERS. Therefore, this chip was shown to exhibit significant potential for applications in bioanalysis.

Preliminary study on flg22-induced defense responses in female gametophytes of Saccharina japonica (Phaeophyta)

Flg22, which is the most conserved 22-amino acid peptide in the N-terminal part of flagellin, functions as an effective elicitor in higher plants. Marine algae and higher plants share some conserved characteristics on defense response pathways. Flg22-induced defense responses were investigated in female gametophytes of Saccharina japonica. Condensation of chloroplasts and thickened cell walls, as well as relatively stable structures of mitochondria and nucleus indicated that there were hypersensitive programmed cell death occurred after induction by flg22. By using luminol-dependent luminescence detection method, rapid release of H2O2 was detected in the induced female gametophytes and reached a peak concentration of about 46 μM at 2 h. Reactive oxygen species production was also observed histologically using the fluorescent dye 2′,7′-dichlorofluorescein diacetate, showing a consistent result with quantitative analysis of H2O2. Furthermore, results of antioxidant enzyme activities indicated that there was a trend in the order of catalase > superoxide dismutase > glutathione peroxidase. Finally, high level phenol content of cell-free extracts was found after flg22 induction. According to our results, flg22 could be an effective elicitor which could induce defense responses in female gametophytes of S. japonica.
