Toxicological analysis and anti-inflammatory and antioxidant evaluations of extract, fractions and secoxyloganin obtained from Guettarda viburnoides Cham. & Schltdl. in mice

Ethnopharmacological relevanceGuettarda viburnoides, "veludinho do campo," is traditionally used for the treatment of pain and inflammatory conditions in humans; however, only one scientific study has reported this effect in an ear inflammatory model. Therefore, it is necessary to explore other in vivo models and the chemical composition of this medicinal plant. Aim of the studyA chemical investigation of methanolic extract of G. viburnoides (MEGV) (leaves) led to the isolation of secoxyloganin (GV-1). In addition, the preclinical safety of MEGV (in acute and subacute toxicological models, gavage = p.o.), antioxidants of MEGV, ethyl acetate (EAFGV) and hydromethanolic (HMFGV) fractions were tested using free radical scavenging and lipid peroxidation methodologies, and the anti-inflammatory effects of MEGV, HMFGV and GV-1 (p.o.) were evaluated on carrageenan and complete Freund's adjuvant (CFA) models of inflammation in mice. Materials and MethodsMEGV was obtained from air-dried leaves by maceration with methanol at room temperature. MEGV was then purified by liquid-liquid partitioning, to obtain the EAFGV and HMFGV fractions. Purification of HMFGV afforded GV-1. The quantification of total phenols, flavonoids, flavonols, and condensed tannins was subsequently performed for MEGV. The antioxidant activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2, 2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and oxidation of β-carotene were evaluated in MEGV and its fractions. The anti-inflammatory activity of MEGV (3, 30, and 100 mg/kg, p.o.) was assayed in carrageenan-induced models, followed by assessments of MEGV (100 mg/kg, p.o.), HMFGV (3 and 30 mg/kg, p.o.) and GV-1 (3 mg/kg, p.o.) in CFA- induced models in mice (including paw oedema, mechanical allodynia and cold sensitivity). Acute (14 days of MEGV, 2000 mg/kg, p.o.) and subacute (28 days, MEGV 30, 100, and 300 mg/kg, p.o.) toxicity was assessed in female Swiss mice. ResultsThe major compound was secoxyloganin (GV-1). The oral acute toxicity test of MEGV revealed no evidence of toxicity, indicating low toxicity according to the Organization for Economic Cooperation and Development (OECD) guidelines. In the subacute toxicity group, no clinical signs of toxicity or changes in body weight, water consumption, food consumption, or organ weight or morphology were observed after 28 days of gavage with MEGV (30, 100, and 300 mg/kg) compared with those in the control group. MEGV, EAFGV, and HMFGV showed significant free-radical scavenging and lipid peroxidation activities, with IC50 values ≤ 26.38 ± 4.56 μg/mL. In in vivo anti-inflammatory assays, MEGV (3, 30 and 100 mg/kg) reduced carrageenan-induced oedema (2 and 4 h) and hyperalgesia (3 and 4 h). In the CFA model, MEGV (100 mg/kg), HMFGV (30 mg/kg) and GV-1 (3 mg/kg) reduced inflammation (at 3, 4 and 24 h) in all parameters (oedema, mechanical allodynia and cold sensitivity). ConclusionThis study revealed that G. viburnoides has antioxidant and anti-inflammatory properties, and no toxicity was detected after acute or subacute gavage with MEGV, validating its traditional use in the treatment of inflammatory conditions.

Antimicrobial and Anti-Inflammatory Potential of Euphorbia paralias (L.): a bioprospecting study with phytoconstituents analysis.

The phytochemicals in the aerial parts of Euphorbia paralias (also known as Sea Spurge) and their anti-inflammatory and antimicrobial activities were investigated. The methanolic extract was characterized using GC-MS and HPLC techniques. The anti-inflammatory feature was estimated through a Human Red Blood Cell (HRBC) membrane stabilization technique, while the antimicrobial feature was evaluated by the disc diffusion agar technique, minimum bactericidal concentration, and minimum inhibitory concentration (MIC) via micro-broth dilution method. The GC/MS results demonstrated the existence of various phytochemicals, such as n-hexadecenoic acid, cis-11-eicosenoic acid, and methyl stearate, recognized for their anti-inflammatory and antibacterial features. The similarity of the phytochemical composition with other Euphorbia species emphasizes the genus-wide similarity. The anti-inflammatory activity exhibited a noteworthy inhibitory effect comparable to the reference drug indomethacin. The extract's antimicrobial potential was tested against a range of microorganisms, demonstrating significant action against Gram-positive bacteria and Candida albicans. The quantification of total phenolics and flavonoids further supported the therapeutic potential of the extract. The methanolic extract from E. paralias emerges as a successful natural source of important active constituents with potential applications as anti-inflammatory and antimicrobial agents. This research provides a first step to valorize Euphorbia paralias insights as a source of worthwhile phytochemicals that have potential applications in the pharmaceutical industry.

Changes in the fruits and seeds morphometric, germination, phytochemicals content, and antioxidant capacity in seed ripening of Echinocereus stramineus

Within the cacti family, most studies have focused on the physicochemical characterization of the stems, fruits, and seeds from Mammillaria, Opuntia, Hylocereus, and Stenocereus genera. However, few information is focused on the morphological and physicochemical characterization of the stems, fruits, and seeds of other genera widely distributed in the arid and semiarid zones of the Chihuahuan desert, such as the genus Echinocereus. The objective of this study was to analyze the fruit morphology, morphometry, germination process, and phytochemical content in four stages of seed in fruit ripening from E. stramineus. Morphometric parameters were measured by picking ten fruits and sixty seeds for each ripening stage. The characterization of the germination process included the germination percentage, mean germination time, germination speed, and mean germination speed which were computed 21 days after germination. The quantification of total phenols, flavonoids, tannins, reducing sugars, protein, and antioxidant activity of seeds was determined using colorimetric approaches under basal conditions. The morphometric results revealed a negative correlation between the fruit ripening stage and total mass (r= -0.980, p = 0.020), shell mass (r= -0.986, p = 0.014), pulp mass (r = -0.979, p = 0.021), fruit length (r = -0.978, p = 0.022), fruit width (r = -0.968, p = 0.032) and fruit area (r = -0.960, p = 0.04). The germination characterization process showed a negative association between the fruit ripening stage and the seed germination percentage (r = -0.979, p = 0.021) and between the seed mean germination time and the mean germination speed (r= -0.986, p = 0.014). The content of flavonoids, reducing sugars, proteins, and antioxidant capacity showed significant differences among the four stages of fruit ripening; however, no association was found between seed phytochemical content and the ripening stage. This study provides the first data on seed phytochemicals and information on the germination process of E. stramineus seeds.

Comparative analysis of non-thermal technologies and solvent systems for the extraction and characterization of phytochemicals and antioxidants in Pandanus amaryllifolius

Pandanus amaryllifolius leaves (PAL) are known for their aroma and are also a rich source of natural phytochemicals. The purity, yield, and stability of phytochemicals depend upon the efficiency and selectiveness of the extraction method and the solvent used. In this study, the phytochemical and antioxidant activity of PAL were evaluated by using non-thermal extraction techniques, i.e., ultrasound (US) and cold plasma (CP). The extraction was evaluated using two different solvents: petroleum ether and 30 % ethanol. Face-centered central composite design was used to design the experimental parameters. The process parameters used were amplitude (30, 45, and 60 %) and treatment time (15, 30, and 45 min) for US, and voltage (10, 20, and 30 kV) and time (10, 20, and 30 min) for CP. The extraction efficiency of both the treatment methods and solvents was evaluated based on the quantification of total phenolics, flavonoids, terpenoids, chlorophyll content, and antioxidant activity. The damaged cell structure as observed from SEM images, confirmed the extraction of phytochemicals. The presence of phenolic and flavonoid compounds in the extract of PAL was confirmed from the FTIR analysis, revealing its nutritional and medicinal properties. Antioxidant activity was higher in case of 30 % ethanol as compared to petroleum ether. In the case of phenolic compounds, CP along with ethanol, had higher extraction efficiency. The use of non-thermal technology along with a suitable solvent can extract phytochemicals and antioxidants from PAL that can be further utilized for value-added product development.

Screening of the Medicinal Plant-Based Metabolites Responsible for Silver Nanoparticle Phytofabrication

In this study, the detailed phytochemical composition of two medicinal plants was investigated and their importance in silver nanoparticles (AgNPs) synthesis was highlighted. Aqueous extracts from the leaves of both plants were used for various analytical techniques, including UV-vis spectroscopy, Gas chromatography-mass spectrometry (GC-MS), Fourier transform infrared spectroscopy (FTIR) and Thin layer chromatography (TLC). Quantification of total phenolics and flavonoids in different fractions of plant extracts revealed significant concentrations. TLC profiling showed a higher abundance of free phenolic and flavonoid compounds compared to bound forms in both plants, suggesting that they contribute significantly to total phenolic and flavonoid content. UV-vis spectra from 200 to 600 nm revealed the presence of aromatic rings and chromophores in leaf extracts. Furthermore, the GC-MS analysis identified several bioactive compounds, some of which were found to be common between these two species. The results demonstrate that various bioactive compounds such as phenols, flavonoids, alkaloids, carotenoids and terpenoids were present in these plant species. These compounds efficiently serve as both reducing and stabilizing agents during phytofabrication of AgNPs, consequently eliminating the need for the use of hazardous chemicals.

Physalis angulata L. (Solanaceae): Antimicrobial and Antioxidant Studies of the Aqueous Methanol Leaves Extract

Physalis angulata has several ethnomedical uses. The study was aimed at investigating the potential antimicrobial and antioxidant of the aqueous-methanol extract of P.angulata leaves. The extract was cold macerated in 80% methanol and tested for phytochemicals, antioxidants, and antimicrobials. The agar well diffusion method was used to determine antimicrobial activities at 6.25–100 mg/mL. The microorganisms tested included Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Proteus vulgaris, Candida albicans, and Aspergillus niger. The quantification of total phenols and flavonoids was conducted using the Folin-Ciocalteu reagent method and the Aluminium Chloride colorimetric method, respectively. The assessment of radical scavenging activity was performed using the DPPH and FRAP models. The plant extract exhibited antibacterial activity, demonstrating concentration-dependent activity against all tested organisms. At 25 mg/mL, S. aureus and A. niger recorded zones of inhibitions of 19.00 and 23.00 mm while B. subtilis, E. coli, P. vulgaris and C. albicans recorded 18.83, 18.67, 14.00 and 4.35 respectively. However, at the maximum concentration of 100 mg/mL, A. niger recorded the highest zone of inhibition of 26.33 mm. The MIC was observed at 12.5 mg/mL against S. aureus, A. niger as well as 25 mg/ml B. substilis, E.coli, P. vulgaris and C. albicans respectively. The total flavonoids and phenolic activities was reported to be of 56.11±0.03 mg QAE/100 g and 96.13±0.07 mg QAE/100 g respectively. The DPPH and FRAP scavenging activities recorded IC50 of 0.05 and 0.07 mg/mL against 0.01 and 0.02 mg/mL of the control. The methanol extract of P. angulata leaves exhibited promising antioxidant and antimicrobial properties, supporting its traditional usage in ethno-medicine for treating many diseases.

Quantification of total phenols in Coleus forskohlii and inhibition of aldose reductase by rosmarinic acid

Many complications of diabetes have been associated with the accumulation of intracellular sorbitol caused by excessive activity of aldose reductase (ALR2). ALR2 is a member of the aldo-keto reductases (AKR) superfamily and has gained significant attention due to its role in the development of diabetic complications. Inhibiting ALR2 could help prevent or delay some of these problems. Coleus forskohlii is a natural medicinal plant that is known for its medicinal effects and is also used as a beverage in several European countries. High-performance liquid chromatography (HPLC) was used to measure the polyphenol content of aqueous leaf extracts of C. forskohlii, and it was found that rosmarinic acid (RA) had the highest concentration (1.25 mg/g) compared to other polyphenols. In vitro bioassays were conducted to evaluate the inhibitory effect of RA on ALR2. RA significantly and dose-dependently reduced the build-up of sorbitol in human erythrocytes under high glucose conditions. Molecular docking studies suggest that RA, which is an essential polyphenol, could be a potential ALR2 inhibitor. This study is the first to show that RA has an IC50 value of 56 % to exhibit an inhibitory effect against ALR2.

Effect of two biocontrols on the accumulation of phenolic compounds and leaf pigments in sorghum [<i>Sorghum bicolor</i> (L.) Moench] leaves in northern Côte d'Ivoire

The aim of this study was to find an alternative to chemical control by improving sorghum's natural defense system. The effect of two biocontrols, Vacciplant and Calliet, on phenolic compound and leaf pigment levels was evaluated. Vacciplant was prepared at concentrations of 0.675; 0.9; 1.125; 1.35 and 1.575 mg/mL, while Calliet was prepared at concentrations of 4 ; 8 ; 12 ; 16 and 20 mg/mL. After preparation, the biocontrol solutions were sprayed onto sorghum leaves 60 days old. Incubation times of 24, 48, 72 and 96 hours were observed after spraying. The sorghum leaves were then harvested and freeze-dried for quantification of total phenols and leaf pigments. Results showed that Vacciplant at a concentration of 1.125 mg/mL after 72 hours of incubation allowed to have the highest contents of total phenols (76.63 mg/g MF), while Calliet at concentration of 4.00 mg/mL after 24 hours of incubation allowed to have the highest contents of total phenols (67.28 mg/g MF). With regard to photosynthetic activity, the results also showed an increase in leaf pigment content following the application of biocontrols. The use of biocontrols could therefore be an interesting alternative to chemical control in sorghum.

Antioxidant Capacity from Ethyl Acetate Fraction of Kenikir (Cosmos Caudatus K)

Antioxidant is an activity that inhibits oxidation reactions due to exposure to free radicals, the search for antioxidant compounds continues to be developed from natural sources. The purpose is to identify new sources of safe and inexpensive agents through antioxidant capacity from ethyl acetate fraction on kenikir leaves was evaluated through enzymatic and non-enzymatic in vitro models. The research began with the method of separating compounds from ethanol extracts and ethyl acetate fractions, quantification of total phenolics and flavonoids, antioxidant capacity tests with DPPH, and tyrosinase enzyme. The results of the study produced isolate E which was observed in a single stain with Rf 0,80. The compound has a typical marker worth 1734.07 cm-1 with an indication of stretching vibration movement on C-O. It has a maximum wavelength of ± 216 nm, with an abundance of 49.75% and a retention time of 27.20 minutes. TPC and TFC test results, the water fraction is the most dominant value with 0.430 mgGAE/g and 0.056 mQE/g. DPPH assay results, isolate E has an IC50 value of 162.03 ppm and a %inhibition of 12.73%. The tyrosinase inhibition test of L-DOPA substrate was resulting an IC50 value of 77.51 ppm compared to kojic acid of 16.48 ppm.

Evaluation of in vitro enzyme inhibitory, anti-inflammatory, antioxidant, and antibacterial activities of Oldenlandia corymbosa L. and Oldenlandia umbellata L. whole plant extracts

BackgroundOldenlandia corymbosa and Oldenlandia umbellata are well-known medicinal plants in various traditional systems of medicine and used to treat bronchitis, asthma, tuberculosis, constipation, and leprosy. We evaluated the phytochemical content, antioxidant potential, enzyme inhibitory action, anti-inflammatory, and antibacterial activities of the whole plant extracts of O. corymbosa and O. umbellata due to their therapeutic importance. MethodsThe phytochemical analysis includes qualitative analysis and quantification of total phenolics, flavonoids, tannins, flavonols, proanthocyanidins, and vitamin E. Enzyme inhibitory assays were studied using α-amylase and α-glucosidase enzymes and anti-inflammatory activity was determined by egg albumin denaturation assay. Antioxidant potential was estimated using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis (3-ethyl benzothiozoline-6-sulfonic acid) diammonium salt (ABTS), ferric reducing antioxidant power (FRAP), hydroxyl radical, superoxide, nitric oxide, metal chelating, and phosphomolybdenum assays. Minimum inhibitory concentrations of extracts were studied against ten bacterial strains by the microdilution method. ResultsThe aqueous extract of O.umbellata showed a higher extract yield (5.0%). The highest amount of total phenolics and flavonoids were observed in the ethanolic extract of O.umbellata (77.23±0.46 mg GAE/g) and O.corymbosa (19.01±0.26 mg GAE/g). Ethanolic extract of O.umbellata (IC50 of 31.71±0.32 µg/mL) and aqueous extract of O.corymbosa (30.53±1.83 µg/mL) showed notable α-amylase inhibitory effect and petroleum ether extract of both plants observed good inhibition against α-glucosidase enzyme. Likewise, petroleum ether extract of O.umbellata (IC50 of 57.16±1.88 µg/mL) showed a moderate anti-inflammatory effect. In all extracts, considerable antioxidant potential was recorded specially with aqueous extract in the nitric oxide scavenging assay with an IC50 of 40.72±1.25 µg/mL. Ethanolic extract of O.umbellata and petroleum ether extract of O.corymbosa (MIC of 56.25 µg/mL) showed good antibiotic potency in lower concentrations which proves these two valuable herb's potential in herbal preparations. ConclusionsThe results of the study provide a basis for further studies aiming to evaluate the possibilities of using the O. umbellata and O. corymbosa as natural sources of antioxidants, enzyme inhibitory, and antibacterial agents. Further studies could be directed to exploit these plants in experimental animal models as potential pharmaceutical agents.

Rapid and cost-efficient microplate assay for the accurate quantification of total phenolics in seaweeds

Brown seaweeds (Phaeophyceae) are a rich source of polyphenols (up to 20% dry weight) with a structure based on phloroglucinol (1,3,5-trihydroxybenzene). To-date the determination of total phenolics content (TPC) involves a redox reaction with the Folin-Ciocalteu (FC) reagent. However, side reactions with other reducing substances preclude accurate, direct measurement of TPC. This research reports a novel microplate assay involving a coupling reaction between phloroglucinol with Fast Blue BB (FBBB) diazonium salt, at basic pH, to form a stable tri-azo complex with maximum absorbance at 450 nm. Linear regression correlation values (R2) were ≥0.99 with phloroglucinol as standard. Direct quantification of TPCs (phloroglucinol equivalents, PGEs) in crude aqueous and ethanolic extracts from A. nodosum demonstrated that the new FBBB assay is not subject to side-redox interference and provides a more accurate estimate of TPC (1.2–3.9-fold lower than with the FC assay) in a relatively rapid (30 min), cost-effective (0.24€/test) microplate format.

Antioxidant Potential, Anthelmintic Activity and Quantification of Total Phenols, Tannins, Flavonoids of E. milii Leaf and Flower Extracts

Plants possess a diverse biologically active compound that exhibits high antioxidant activity. The purpose of antioxidant studies of plants is to reveal the significance of the plants as a reservoir of new antioxidant substances and to entail them in treatment of diverse medical ailments. The current research investigates the antioxidant prospect of E. milii leaves and flowers. These leaves and flowers of E. milii were excerpted using various solvents of various polarities. The antioxidant action of the E. milii leaf and flower extracts was determined using the DPPH free radical scavenging assay. The results of antioxidant screening revealed that the aerial parts of the plant i.e., leaf and flower have prominent antioxidant properties. In-vitro anthelmintic activity results confirmed the potential bioactivity of E. milii leaf and flower extracts and could be a promising source for the development of effective anthelmintic drugs. The total flavonoid, tannin, and phenolic contents of the plant extracts were evaluated spectrophotometrically and found a positive relationship between these phytochemicals and the antioxidant activity. Ethyl alcohol extract of a leaf and acetone extract of flower contained the highest concentration of flavonoids. The maximum range of phenolics was observed in ethyl alcohol solvent extract of a leaf and methanol extract of the flower. The most elevated concentration of tannins was recorded in ethyl acetate extract of leaves and ethanol extract of flowers. The existence of phytochemical constituents and high antioxidant activity convinces the anthelmintic potential of E. milii aerial parts. The present study justifies the traditional use of E. milii to treat helminthic diseases.

Asterohyptis stellulata: Phytochemistry and wound healing activity

Wound healing is a fast and efficient process. However, several factors can inhibit healing, causing scars and chronic wounds. This affects patients in their physical integrity, economic and emotional stability. Many plants that are used in wound healing treatment have antioxidant/anti-inflammatory activity through ROS regulation and antibacterial activity. Asterohyptis stellulata is used for wound treatment. Nonetheless, this activity has not been evaluated and its secondary metabolites are unknown. Therefore, the objective of this study was to analyze its phytochemical composition and evaluate the healing activity of A. stellulata. For the identification of secondary metabolites, the following were carried out: phytochemical screening, quantification of total phenols, and HPLC/MS analysis. The antibacterial and antioxidant activities were also determined. Next, to evaluate the healing activity of topical application of A. stellulata, the extract was mixed with the Beeler base to form an emulsion and an irritability test was performed. The emulsion was evaluated on incisional wounds of CD1 et/et mice, with tests of tensile strength, total re-epithelialization time, wound closure speed, and histological analysis. The phytochemical analysis showed that A. stellulata has phenolic compounds, two quercetin derivatives and a glycoside rosmarinic acid as main compounds. The extract exhibited medium antibacterial and antioxidant activities. A. stellulata emulsion was non-irritating, promoted closure speed of wound (1.479 ± 0.043 mm/day), increased tensile strength (980 ± 89.44 g), and histological analysis showed it promoted mice skin regeneration, compared to the controls (Recoveron® and Beeler base). This study confirms the traditional use of the A. stellulata.

Evaluation of Different Extraction Methods on the Phenolic Profile and the Antioxidant Potential of Ceratonia siliqua L. Pods Extracts.

The present work was designed to investigate the effects of different extraction processes, namely ultrasonic-assisted, supercritical fluid, microwave-assisted and Soxhlet applied to carob pods. The total phenolic quantification and the antioxidant activity were assessed by the means of rapid in vitro spectrophotometric assays; the phenolic profile was identified using ultra-high performance liquid chromatography coupled to mass spectrometry. The results revealed that the phenolic compounds and the antioxidant capacity varied significantly with the nature of the extraction process. The content of total phenolic compounds ranged from 11.55 to 34.38 mg GAE/g DW; the content of total flavonoids varied from 3.50 to 10.53 mg QE/g DW, and the content of condensed tannins fluctuated from 3.30 to 6.55 mg CE/ g DW. All extracts performed differently on antioxidant activity when determined by the DPPH assay producing a dose-dependent response, with IC50 extended from 11.33 to 6.07 µg/mL. HPLC analysis enabled the identification of nine compounds. As a function of the studied extraction methods, the phenolic compound contents were positively correlated with antioxidant activity.

Free and Bound Phenolic Compounds Present in Orange Juice By-Product Powder and Their Contribution to Antioxidant Activity.

Interest in fruit/vegetable consumption is not always linked to a particular diet but rather derives from their high antioxidant activity (AOA), with potential health benefits provided, in part, by polyphenols. Although phenols can be found in free form (FP) or bound to plant tissues (BP), the experimental methodology most frequently used for the quantification of total phenols (TP) is based on the extraction of a portion of FP, which may justify the lack of correlation often found between them and AOA. In this context, four successive extractions were performed to obtain FP and BP of powdered orange juice by-product, and their contribution to the AOA was studied. The first extract (MeOH, 30 °C) can be assumed to be one of the conventional methods for the quantification of TP. Re-extraction with MeOH (60 °C) afforded the FP. Two subsequent basic and acidic extractions yielded the BP. Although the FPs were the most abundant, the AOA (DPPH method) of the last fraction of BP was of the same order found in the first fraction of FP. This highlights the interest in extracting the BP from the by-product of orange juice if its antioxidant capacity is to be exploited.

Preliminary examination of bioactive components and bioactivity of Hardwickia binata Roxb. extracts obtained through conventional extraction methods

The phytochemical constituents, quantitative antioxidant, and total phenolic content of Hardwicikia binata Roxb. plant parts were investigated in the present study. Methanolic extracts of H. binata leaves and stems were obtained using the traditional extraction methods, soxhlet and maceration. Preliminary tests for phytochemicals, revealed the presence of flavonoid, steroids, tannin and terpenoids, tannins in the stem and, flavonoids and phenols in the methanolic extracts of the stem and leaves, respectively. Saponin was found in the leaves and stem parts, but no alkaloids were identified in either. The high antioxidant content of the H. binata stem and leaf methanolic extract was proven by the total antioxidant assay. Furthermore, total phenol quantification among the extracts revealed that the plant's stem extract was rich in phenols. DPPH scavenging activity showed the potential antioxidant capability of H. binata parts. Overall, the preliminary findings revealed the bioactive components and antioxidant properties of H. binata leaves and stem parts; further detailed examination and screening of the plant may prove it to be a potential source of medication in the pharmaceutical field.

Phytochemical screening, total phenolics and antimicrobian action from the brazilian cherry, black mulberry and blueberry fruits’ pulps

The demand for functional foods with exotic flavors is emerging among consumers. The fruits brazilian cherry, black mulberry and blueberry have demonstrated considerable potential as inhibitors of the growth of microorganisms. The phytochemicals present in different extracts were obtained from fruits (pulps), employing specific tests to detect classes of compounds besides Infrared. Tests were realized for antimicrobial activity and quantification of total phenolics. In the phytochemical tests, the presence of mainly the flavonoid group was observed, in agreement with the high values of total phenolics found. The 70% ethanol is the most indicated solvent because it presents greater extraction of polar clusters, besides providing signals of absorption of O-H bonds in all pulps studied. The blueberry showed fungistatic action for Candida tropicalis. The diffusion of the extract through the culture medium is difficult, consequently, the formation of a halo, visible as that of the control, too is difficult. Therefore, results obtained in this study are considered screening and not as definitive results.

Antioxidant and Antibacterial Activities of Secondary Metabolites from Microporus xanthopus (Fr.) Kuntze (Polypore) Collected from the Wild in Lagos, Nigeria

The secondary metabolites in medicinal mushrooms are responsible for their activity against infectious diseases, cancer, diabetes, and diseases caused by presence of free radicals in the body. Microporus xanthopus a polypore medicinal mushroom was collected from the wild in Lagos Nigeria and identified using standard manuals and oligosaccharides, polysaccharide and polyphenols extracted from its tissues were investigated for antibacterial and antioxidant activities. M. xanthopus oligosaccharides were extracted with neutral and acid detergents and hydrolysis with concentrated H2SO4. Polysaccharides and polyphenols extracted with hot water and acidified methanol, respectively. Concentrations of oligosaccharides, polysaccharides and polyphenols were determined with the total carbohydrate and total phenolic quantification assay kits, respectively. The antioxidant activities of the extractsnvestigated using the DPPH Radical Scavenging Assay and Trolox Antioxidant Equivalent Capacity (TEAC) Assay in In-Vitro experiments in 96-well microtiter plates. The antibacterial effect of the extracts was determined with broth microdilution assay using human pathogenic bacteria (Escherichia coli (0157:H7) and Staphylococcus aureus ATCC®700698 (MRSA). Oligosaccharides showed the highest DPPH radical scavenging activity (86%) with half maximal effective (EC50) of 16.46µg/mL. The highest TEAC value (1.18µM TE/g) was recorded in the oligosaccharide extract and the least TEAC value (0.39µM TE/g) was in the polyphenol extract. The most potent antimicrobial agent was the oligosaccharide extract with IC50 of 44.64 µg/mL and 40.08 µg/mL for E. coli and S. aureus, respectively. Oligosaccharide extracts were more active than the polyphenol and polysaccharide extracts. M. xanthopus oligosaccharides can be developed as potential new dietary supplements with antioxidant and antibacterial activities.
