Iron Speciation in Animal Tissues Using AC Magnetic Susceptibility Measurements: Quantification of Magnetic Nanoparticles, Ferritin, and Other Iron-Containing Species.

The simultaneous detection and quantification of several iron-containing species in biological matrices is a challenging issue. Especially in the frame of studies using magnetic nanoparticles for biomedical applications, no gold-standard technique has been described yet and combinations of different techniques are generally used. In this work, AC magnetic susceptibility measurements are used to analyze different organs from an animal model that received a single intratumor administration of magnetic nanoparticles. The protocol used for the quantification of iron associated with the magnetic nanoparticles is carefully described, including the description of the preparation of several calibration standard samples of nanoparticle suspensions with different degrees of dipolar interactions. The details for the quantitative analysis of other endogenous iron-containing species such as ferritin or hemoglobin are also described. Among the advantages of this technique are that tissue sample preparation is minimal and that large amounts of tissue can be characterized each time (up to hundreds of milligrams). In addition, the very high specificity of the magnetic measurements allows for tracking of the nanoparticle transformations. Furthermore, the high sensitivity of the instrumentation results in very low limits of detection for some of the iron-containing species. Therefore, the presented technique is an extremely valuable tool to track iron oxide magnetic nanoparticles in samples of biological origin.

Cellular uptake of magnetic nanoparticles imaged and quantified by magnetic particle imaging

Magnetic particle imaging (MPI) is a non-invasive, non-ionizing imaging technique for the visualization and quantification of magnetic nanoparticles (MNPs). The technique is especially suitable for cell imaging as it offers zero background contribution from the surrounding tissue, high sensitivity, and good spatial and temporal resolutions. Previous studies have demonstrated that the dynamic magnetic behaviour of MNPs changes during cellular binding and internalization. In this study, we demonstrate how this information is encoded in the MPI imaging signal. Through MPI imaging we are able to discriminate between free and cell-bound MNPs in reconstructed images. This technique was used to image and quantify the changes that occur in-vitro when free MNPs come into contact with cells and undergo cellular-uptake over time. The quantitative MPI results were verified by colorimetric measurements of the iron content. The results showed a mean relative difference between the MPI results and the reference method of 23.8% for the quantification of cell-bound MNPs. With this technique, the uptake of MNPs in cells can be imaged and quantified directly from the first MNP cell contact, providing information on the dynamics of cellular uptake.

Quantification of magnetic nanoparticles by compensating for multiple environment changes simultaneously.

The quantification of magnetic nanoparticles is important for many applications, especially for in vivo biosensing. The magnetization harmonics used in spectroscopy of magnetic nanoparticles can be used to estimate nanoparticle number or weight. However, other effects such as temperature or relaxation time change can also influence the nanoparticle magnetization. Therefore, it is necessary to compensate for these factors when estimating the amount of magnetic nanoparticles. This paper shows through simulation that a two-dimensional scaling method can be used to improve the accuracy of nanoparticle quantification, especially when multiple effects are present which can influence the nanoparticle magnetization. Finally, an experiment was performed on a Magnetic Spectroscopy of Brownian motion (MSB) apparatus to demonstrate this method, and nanoparticle weight was determined with a mean error of 1.3 ng (1.81%).

Imaging and quantification of magnetic nanoparticles: Comparison of magnetic resonance imaging and magnetic particle imaging

Abstract Quantification and imaging of magnetic nanoparticles is of vital importance for various novel biomedical applications, like cell tracking, drug targeting or hyperthermia treatments. In this work we studied the performance of magnetic resonance imaging (MRI) and magnetic particle imaging (MPI) for quantitative imaging of magnetic nanoparticles (MNP). This was done by measurements of serial dilutions of MNP (Ferucarbotran) in two different media (water and CuSO4 solution). The concentration range in which quantification was possible was determined for each technique, and the influence of the environment was analyzed and discussed. This revealed a significantly stronger influence of the surrounding medium on MRI performance as compared to MPI. All results were validated by measurements using their respective zero-dimensional (spectroscopic) techniques nuclear magnetic resonance and magnetic particle spectroscopy, showing similar behavior compared to the imaging modalities. Physical explanations of all observed effects are given, and a concentration range is determined in which the advantages of both imaging techniques can be utilized.

Degradation of magnetic nanoparticles mimicking lysosomal conditions followed by AC susceptibility.

A deeper knowledge on the effects of the degradation of magnetic nanoparticles on their magnetic properties is required to develop tools for the identification and quantification of magnetic nanoparticles in biological media by magnetic means. Citric acid and phosphonoacetic acid-coated magnetic nanoparticles have been degraded in a medium that mimics lysosomal conditions. Magnetic measurements and transmission electron microscopy have been used to follow up the degradation process. Particle size is reduced significantly in 24 h at pH 4.5 and body temperature. These transformations affect the magnetic properties of the compounds. A reduction of the interparticle interactions is observed just 4 h after the beginning of the degradation process. A strong paramagnetic contribution coming from the degradation products appears with time. A model for the in vivo degradation of magnetic nanoparticles has been followed to gain insight on the changes of the magnetic properties of iron oxides during their degradation. The degradation kinetics is affected by the particle coating, in our case being the phosphonoacetic acid-coated particles degraded faster than the citric acid-coated ones.

Quantification of magnetic nanoparticles with low frequency magnetic fields: compensating for relaxation effects

Quantifying the number of nanoparticles present in tissue is central to many in vivo and in vitro applications. Magnetic nanoparticles can be detected with high sensitivity both in vivo and in vitro using the harmonics of their magnetization produced in a sinusoidal magnetic field. However, relaxation effects damp the magnetic harmonics rendering them of limited use in quantification. We show that an accurate measure of the number of nanoparticles can be made by correcting for relaxation effects. Correction for relaxation reduced errors of 50% for larger nanoparticles in high relaxation environments to 2%. The result is a method of nanoparticle quantification suitable for in vivo and in vitro applications including histopathology assays, quantitative imaging, drug delivery and thermal therapy preparation.

Quantification of Magnetic Nanoparticles by Magnetorelaxometry and Comparison to Histology After Magnetic Drug Targeting

Magnetic nanoparticles can be used in medicine in vivo as contrast agents and as a drug carrier system for chemotherapeutics. Thus local cancer therapy is performed with Magnetic Drug Targeting (MDT) and allows a specific delivery of therapeutic agents to desired targets, i.e., tumors, by using a chemotherapeutic substance bound to magnetic nanoparticles and focused with an external magnetic field to the tumor after intraarterial application. Important for this therapeutic principle is the distribution of the particles in the whole organism and especially in the tumor. Therefore we used magnetorelaxometry to quantify ferrofluids delivered after MDT. Tissue samples of some mm3 volume of a VX2 squamous cell carcinoma were measured by magnetic relaxation and the amount of iron was determined using the original ferrofluid suspension as a reference. From this the distribution of the magnetic particles within the slice of tumor was reconstructed. Histological cross-sections of the respective tumor offer the opportunity to map quantitatively the particle distribution and the vascularisation in the targeted tumor on a microscopic scale. Our data show that the integral method magnetorelaxometry and microscopic histological methods can complete each other efficiently.
