Prevalence, genotypes, and infection risk factors of psittacine beak and feather disease virus and budgerigar fledgling disease virus in captive birds in Hong Kong

Psittacine beak and feather disease virus (PBFDV) and budgerigar fledgling disease virus (BFDV) are significant avian pathogens that threaten both captive and wild birds, particularly parrots, which are common hosts. This study involved sampling and testing of 516 captive birds from households, pet shops, and an animal clinic in Hong Kong for PBFDV and BFDV. The results showed that PBFDV and BFDV were present in 7.17% and 0.58% of the samples, respectively. These rates were lower than those reported in most parts of Asia. Notably, the infection rates of PBFDV in pet shops were significantly higher compared to other sources, while no BFDV-positive samples were found in pet shops. Most of the positive samples came from parrots, but PBFDV was also detected in two non-parrot species, including Swinhoe’s white-eyes (Zosterops simplex), which had not been reported previously. The ability of PBFDV to infect both psittacine and passerine birds is concerning, especially in densely populated urban areas such as Hong Kong, where captive flocks come into close contact with wildlife. Phylogenetic analysis of the Cap and Rep genes of PBFDV revealed that the strains found in Hong Kong were closely related to those in Europe and other parts of Asia, including mainland China, Thailand, Taiwan, and Saudi Arabia. These findings indicate the presence of both viruses among captive birds in Hong Kong. We recommend implementing regular surveillance for both viruses and adopting measures to prevent contact between captive and wild birds, thereby reducing the transmission of introduced diseases to native species.

Investigation and Sequence Analysis of Avian Polyomavirus and Psittacine Beak and Feather Disease Virus from Companion Birds in Eastern Turkey.

IntroductionAvian polyomavirus (APV) and psittacine beak and feather disease virus (PBFDV) induce contagious and persistent diseases that affect the beaks, feathers, and immune systems of companion birds. APV causes hepatitis, ascites, hydropericardium, depression, feather disorders, abdominal distension, and potentially death. PBFDV can induce progressive beak deformity, feather dystrophy, and plumage loss. We conducted the first prevalence survey of both APV and PBFDV infections in companion birds in eastern Turkey.Material and MethodsA total of 113 fresh dropping samples from apparently healthy companion birds were collected in a random selection. The dropping samples were analysed for PBFDV and APV by PCR. Positive samples were sequenced with the Sanger method. The sequence was confirmed through alignment and the phylogenetic tree generated through the maximum likelihood method computationally.ResultsPBFDV and APV were detected in a respective 48.7% and 23.0% of samples. Coinfection was found in 12.4% of the samples, these all being from budgerigars (Melopsittacus undulatus). APV and PBFDV were detected in budgerigar and cockatiel (Nymphicus hollandicus) samples.ConclusionThis report provides a foundation for future studies on the influence of these viruses on the health of companion birds. These high positive rates for both pathogens emphasise that healthy M. undulatus and N. hollandicus in eastern Turkey may be prone to the emergence and spread of APV and PBFDV with subclinical potential.

Complete Genome Sequence of Genotype Psittacine Beak and Feather Disease Virus, a Strain Identified from Budgerigars in China.

Psittacine beak and feather disease virus (PBFDV) has been reported in many countries, such as Australia, Poland, the United States, South Africa, etc. In this study, the complete genome of a PBFDV isolate was determined and characterized from budgerigars in China.

Detection and analysis of new psittacine beak and feather disease virus (PBFDv) nucleotide sequences

Psittacine beak and feather disease (PBFD) affects a large number of Psittaciformes species. In this study, five White Cockatoo parrots (Cacatua alba) with clinical signs of PBFD were examined. After euthanasia, a full necropsy of parrots was performed and organs with macroscopic changes were sampled for routine histopathological evaluation. To confirm the presence of psittacine beak and feather disease virus (PBFDv), feather samples were analyzed with the PCR method. Sequence analysis of the obtained PCR products indicated their close relationship (99%) to other PBFDv isolates. Six variable nucleotide sites were discovered, two missense and four silent mutations. This paper presents the evidence of new PBFDv sequence in Cockatoo species.

HEALTH ASSESSMENT OF CAPTIVE PSITTACINE SPECIES IN PRERELEASE PROGRAMS AT COSTA RICAN RESCUE CENTERS.

With stricter laws regulating the capture and possession of wild animals in Costa Rica, local wildlife-rescue centers have been overwhelmed by an influx of confiscated or relinquished illegal pets, specifically of psittacine species. As part of a nationwide health-assessment program targeting these centers, 122 birds representing five psittacine species ( Ara macao, Amazona autumnalis, Amazona auropalliata, Amazona farinosa, Aratinga finschi) and one hybrid macaw ( Ara macao × Ara ambiguus) were examined and tested between January 2011 and October 2012. Physical examination, hematology, and serum biochemical analyses were performed. Blood and feathers were tested for psittacine beak and feather disease virus (PBFDV) and avian polyomavirus (APV) via PCR. A DNA-based prevalence and sequence analysis characterized the strains of PBFDV and APV isolated. Physical abnormalities observed in 36% of the 122 birds examined were limited to thin body condition and poor feather quality. None of the feather abnormalities were characteristic of disease caused by either PBFDV or APV. Results of hematological and biochemical analyses were within normal limits except for five birds with leukocytosis and heterophilia, three birds with uric acid values above 16 mg/dl, and two additional birds with AST values above 400 IU/L. No hemoparasites were detected during blood smear examination. Overall prevalences of 9.8% (12/122) for PBFDV and 3.3% (4/122) for APV were documented, with only one bird testing positive for both PBFDV and APV. Birds from two of the eight centers were negative for both viruses. Findings from this study constitute the beginning of a standardized surveillance program for Costa Rican rescue centers, targeting the management of avian species enrolled in propagation and reintroduction programs and expanding of the spectrum of pathogen surveillance and husbandry recommendations in prerelease centers.

Occurrence of Bacterial and Viral Pathogens in Common and Noninvasive Diagnostic Sampling from Parrots and Racing Pigeons in Slovenia.

Airborne pathogens can cause infections within parrot (Psittaciformes) and pigeon (Columbiformes) holdings and, in the case of zoonoses, can even spread to humans. Air sampling is a useful, noninvasive method which can enhance the common sampling methods for detection of microorganisms in bird flocks. In this study, fecal and air samples were taken from four parrot holdings. Additionally, cloacal and oropharyngeal swabs as well as air samples were taken from 15 racing pigeon holdings. Parrots were examined for psittacine beak and feather disease virus (PBFDV), proventricular dilatation disease virus (PDDV), adenoviruses (AdVs), avian paramyxovirus type-1 (APMV-1), avian influenza virus (AIV), Chlamydia psittaci (CP), and Mycobacterium avium complex (MAC). MAC and AdVs were detected in three parrot holdings, CP was detected in two parrot holdings, and PBFDV and PDDV were each detected in one parrot holding. Pigeons were examined for the pigeon circovirus (PiCV), AdVs, and CP; PiCV and AdVs were detected in all investigated pigeon holdings and CP was detected in five pigeon holdings.

Abstract A eight-month-old blue and yellow macaw ( Ara ararauna ) with psittacine beak and feather disease (PBFD)-suspected signs, such as, abnormal feather, depression and diarrhea, was presented to Animal Disease Intervention Center, Kyungpook National University in 16 April 2014. The partial ORF V1 gene of PBFD virus (PBFDV) was detected by polymerase chain reaction (PCR) from DNA templates extracted from feather, blood and cloacal swab sample of the bird, but no other viral DNAs that often infected in psittacine birds including avian bornavirus and avian polyomavirus were detected from the samples of the bird, indicating this case is due to single infection of PBFDV. Nucleotide sequence anal-ysis of the amplified partial ORF V1 gene was confirmed to have 96.7% and 93.6% homology with that of previously reported PBFDV strain (Genbank no. HM748924 and FJ685980). This report de-scribes the first detection of PBFDV in PBFD-suspected blue and yellow macaw in Korea. Prevalence of Psittacine Beak and Feather Disease Virus and Avian Polyomavirus in Captivity Psittacines from Costa Rica

Psittacine beak and feather disease virus (PBFDV) and avian polyomavirus (APV) are the most common viral diseases in psittacine birds, both affecting feathers and physical appearance of birds. Between 2005 and 2009, a total of 269 samples were collected from birds presented at veterinary clinics, shelters and rescue centers of wildlife in Costa Rica. They belonged to 19 species of psittacine birds. The most representative species in the sample were Ara macao (157), Ara ambigua (37), Amazona autumnalis (24), Amazon ochrocephala (21) and Ara ararauna (8). A prevalence of 19.7% (53/269) for PBFDV and 4.8% (13/269) for APV was determined using Polymerase Chain Reaction (PCR). In 3.3% (9/269) of the birds mixed infections were detected. Statistical analysis determined that psittacines living in shelters and rescue centers had a greater risk to be positive to PBFDV and APV than birds that were presented at veterinary clinics, while only for PBFDV it was determined, that it is more likely to detect it in feathers than in blood. Finally, birds infected with PBFDV had 6.24 times more probability to become infected with APV, than non-infected birds. This is the first report of prevalence of PBFDV and APV in captive psittacines from Costa Rica.

Prevalence and genetic characterization of avian polyomavirus and psittacine beak and feather disease virus isolated from budgerigars in Mainland China

Budgerigar fledgling disease (BFD) and psittacine beak and feather disease (PBFD) are caused by avian polyomavirus (APV) and psittacine beak and feather disease virus (PBFDV), respectively. These diseases frequently infect psittacine birds and result in similar clinical manifestations. In this study, we observed the prevalence of PBFDV infection and a dual infection of APV and PBFDV in a budgerigar (Melopsittacus undulatus) in Mainland China for the first time. One PBFDV isolate and two APV isolates were harvested using chicken embryos. Genetic characterization and phylogenetic analysis of the complete genome of the two APV isolates revealed nucleotide similarity ranging from 99.0% to 99.6% to other sequences in GenBank, and a 14-bp insertion was observed in the genome of one APV isolate. The results of complete genome analysis of the PBFDV isolate showed nucleotide similarity ranging from 83.0% to 95.0% with other PBFDV sequences in GenBank. Genetic characterization and phylogenetic analysis of the APV and PBFDV strains isolated in this study indicated that the isolates from China were closely related to their Japanese counterparts. The results of this study will help to identify molecular determinants and will aid further research on the prevention and control of APV and PBFD infection.

Detection and Sequence Analysis of Avian Polyomavirus and Psittacine Beak and Feather Disease Virus from Psittacine Birds in Taiwan

Avian polyomavirus (APV) and psittacine beak and feather disease virus (PBFDV) are the most common viral diseases of psittacine birds. In Taiwan, however, the existence of these viruses in psittacine birds has not been established. Polymerase chain reaction (PCR) methodology was therefore employed to ascertain whether APV and PBFDV genomes were present in isolates from psittacine birds of Taiwan. A total of 165 psittacine birds belonging to 22 genera were examined between 2002 and 2005. Findings revealed an APV-positive rate of 15.2%, a PBFDV-positive rate of 41.2%, and an APV/PBFDV dual infection rate of 10.3%. After cloning and sequencing, sequences of the PCR products were compared with sequences obtained from GenBank. For APV, the nucleotide identity among VP1 and t/T antigen coding regions ranged from 97.5% to 100% and 97.6% to 100%, respectively. For PBFDV, the nucleotide identity of ORF V1 and ORF C1 sequences ranged from 92.2% to 100% and 83.3% to 100%, respectively. The derived amino acid sequence alignment for PBFDV ORF V1 fragments revealed the conservation of two replication motifs and of the nucleotide binding site motif. In PBFDV, six of 42 deduced positions in the ORF C1 amino acid sequence were considered hypervariable. The established phylogenetic trees based on the four genome fragments examined in this study did not allow the assignment of particular APV or PBFDV nucleotide sequences to distinct avian species.

Duplex Shuttle PCR for Differential Diagnosis of Budgerigar Fledgling Disease and Psittacine Beak and Feather Disease

Two common viral diseases in psittacine birds including budgerigar fledgling disease (BFD), generally called avian polyomavirus (APV) infection, and psittacine beak and feather disease (PBFD) have similar clinical manifestations characterized by feather disorders. A duplex shuttle PCR was developed for detection of APV and PBFD virus (PBFDV). Two pairs of oligonucleotide primers were designed to amplify a 298-bp fragment of the t/T antigen region of APV genome and a 495-bp fragment of the capsid protein region encoded by open reading frame (ORF) C1 of PBFDV genome, respectively. In the present study, APV and PBFDV were detected simultaneously in one tube by duplex shuttle PCR using these two pairs of primers. The detection limits were 2 viral copies of APV and 3 viral copies of PBFDV. In the clinical application, we detected 16 APV-positive, 15 PBFDV-positive, and 3 mixed infected samples in 39 samples examined. Sequences of the amplified products were read. The t/T antigen region was conserved in the APV-positive samples as expected. ORF C1 of PBFDV genome showed diversity. Phylogenic analysis indicated that PBFDV ORF C1 consisted of 6 clusters which were related to subfamilies of psittacine birds. Our duplex shuttle PCR could be a useful method for differential diagnosis and molecular epidemiology of BFD and PBFD.

Psittacine beak and feather disease virus in budgerigars and ring-neck parakeets in South Africa.

Psittacine beak and feather disease (PBFD) is a common disease of the psittacine species and is caused by the psittacine beak and feather disease virus (PBFDV). In this study the occurrence of the disease in ring-neck parakeets and budgerigars in South Africa suffering from feathering problems, using polymerase chain reaction as a diagnostic test was investigated. The genetic variation between viral isolates was also studied. Results indicate that PBFDV can be attributed to being the cause of feathering problems in some of the ring-neck parakeets and budgerigars in South Africa. Genetic variation of isolates occurs between species and individuals. A cheap and easy to use method of blood sample collection on filter paper for diagnostic purposes was also evaluated. It proved to be less stressful to the birds and did not inhibit further processes.

Detection of pigeon circovirus by polymerase chain reaction.

Pigeon circovirus was identified by polymerase chain reaction (PCR) in young pigeons belonging to 12 different lofts. Viral DNA was extracted from formalin-fed, paraffin-imbedded tissues containing primarily bursa and occasionally liver and spleen with a commercial kit. PCR primers were selected from a published sequence for columbid circovirus and evaluated in a PCR assay. The histopathologic examination of various tissues revealed basophilic globular intracytoplasmic inclusions in the mononuclear cells of the bursa of Fabricius and occasionally in the spleen characteristic for a circovirus. Transmission electron microscopy of a few bursas of Fabricius revealed virus particles measuring 18-21 nm. All the samples were negative by PCR for psittacine beak and feather disease (PBFD) virus and chicken infectious anemia virus. The primers for both pigeon circovirus and PBFD virus did not react in PCR with the chicken anemia virus DNA. Most of the circovirus-infected pigeons had concurrent infections of Escherichia coli, Salmonella, Pasteurella, Aspergillus, candidiasis, nematodiasis, or capillariasis.

Psittacine beak and feather disease in three captive sulphur-crested cockatoos (Cacatua galerita) in Thailand.

Three sulphur-crested cockatoos (Cacatua galerita) were diagnosed as psittacine beak and feather disease (PBFD). Histopathology of the feather pulp and follicles showed intracytoplasmic botryoid clusters or granular inclusion bodies in epithelial cells and macrophages. Electron microscopy revealed multiple cytoplasmic clusters of electron dense viral particles corresponding to the inclusions. PBFD virus (circovirus) DNA-specific product was detected from formalin-fixed paraffin-embedded feathers by nested polymerase chain reaction (PCR) method.

The sensitivities of various erythrocytes in a haemagglutination assay for the detection of psittacine beak and feather disease virus.

The erythrocytes of various species were tested in psittacine beak and feather disease (PBFD) virus haemagglutination (HA) and haemagglutination inhibition assays to determine which are suitable for use in these assays. HA activity was observed for erythrocytes of the salmon-crested cockatoo, the sulphur-crested cockatoo, the umbrella cockatoo, the goffin's cockatoo and the cockatiel, with differences amongst individuals within species, but not for erythrocytes of humans, the pig, the guinea pig, the chicken, the goose, the rose-ringed parakeet or the budgerigar. Anti-PBFD virus rabbit sera inhibited the virus-induced agglutination of erythrocytes, confirming the specificity of HA activity. This suggests that selection of suitable psittacine species as well as suitable individuals within a species is necessary when obtaining erythrocytes for the PBFD virus HA assay.

Severe Leukopenia and Liver Necrosis in Young African Grey Parrots (Psittacus erithacus erithacus) Infected with Psittacine Circovirus

This paper describes the signs, clinical pathology, and postmortem findings in 14 young African grey parrots (Psittacus erithacus erithacus) that were naturally infected with psittacine beak and feather disease (PBFD) virus (psittacine circovirus). All but two of the parrots had severe leukopenia at clinical presentation. Two other parrots also had severe anemia. All birds died within 3 wk after presentation. Postmortem examination documented liver necrosis in 11 of 14 birds and secondary bacterial or fungal infections in 9 of 14 birds. Tests for Chlamydia psittaci, polyomavirus, and Salmonella sp. were negative. PBFD viral infection could be demonstrated in all birds by polymerase chain reaction. Supporting evidence of PBFD viral infection was gathered by histologic examination of the bursa of Fabricius, electron microscopy, and DNA in situ hybridization. Electron microscopic examination of both the bursa of Fabricius and liver revealed virus particles resembling circovirus. DNA in situ hybridization of six liver tissue samples confirmed the presence of PBFD virus and excluded the presence of avian polyomavirus. Our findings suggest that a specific presentation of peracute PBFD viral infection, characterized by severe leukopenia, anemia, or pancytopenia and liver necrosis in the absence of feather and beak abnormalities, may occur in young African grey parrots.

A review of the most common viruses affecting Psittaciformes

Four of the most common viral infections affecting psittacine birds have been studied in detail: (1) avian polyomavirus, (2) proventricular dilatation disease (PDD), (3) Pacheco's disease and (4) psittacine beak and feather disease (PBFD). The Psittacine Disease Research Group, University of Georgia, has been actively involved in this research. Proventricular dilatation disease has been reported in more than 50 psittacine species and in species in other families, while PBFD affects over 40 psittacine species. The acute and chronic clinical characteristics of the diseases, pathological features, diagnosis, and transmission prevention and control, including vaccination, are described. The vaccine for avian polyomavirus is safe and effective in captive birds, and, when used in high-risk situations, vaccination against Pacheco's disease (a psittacine herpesvirus) can prevent high mortality. A safe vaccine for PBFD is being evaluated. Specific tests for viral nucleic acid (DNA probe tests) for avian polyomavirus and PBFD virus, and anti-virus antibody assays for avian polyomavirus and Pacheco's disease have been developed which will facilitate the diagnosis and monitoring of disease. The benefits and limitations of screening psittacines for disease are also discussed.

The haemagglutination spectrum of psittacine beak and feather disease virus

A simple method for concentrating psittacine beak and feather disease virus (PBFDV) from crude feather suspensions is described. The addition of 10% polyethylene glycol (MW 6000 to 9000) to feather suspensions facilitated the precipitation and pelleting of PBFDV by low speed centrifugation. Pellets were resuspended in one-twentieth of the original volume with caesium chloride (CsCl) buffer and subjected to isopycnic ultracentrifugation. Peak haemagglutination activity (HA) occurred at 1.35 g/ml in PBFDV CsCl gradients. CsCl purified virus agglutinated galah (Eolophus roseicapillus), eastern long-billed corella (Cacatua tenuirostris), sulphur-crested cockatoo (Cacatua galerita), Major Mitchell's cockatoo (Cacatua lead-beateri) and gang gang cockatoo (Callocephalon fimbriatum) erythrocytes, but not those of 19 other avian or five mammalian species. PBFDV agglutinated galah erythrocytes at 4 degrees C and 37 degrees C over a wide range of pH and no change in HA titre was observed when PBFDV was treated with chloroform. HA persisted in PBFDV suspensions heated to 80 degrees C for 30 min, but was not detected after incubation at higher temperatures. High HA titres were detected in the feathers, serum, liver and kidneys of PBFD-affected birds.

Particles Resembling Circovirus in the Bursa of Fabricius of Pigeons

Histological examination of the bursae from 12 pigeons under 4 months old revealed basophilic globular inclusion bodies, 5 to 25 microns in diameter, in the cytoplasm and the nuclei of the various bursal follicular cells. Electron microscopy of these inclusions revealed large electron-dense areas containing non-enveloped icosahedral viral particles, 14-19 nm in diameter, either loosely arranged or in paracrystalline array. Similar basophilic globular inclusion bodies were seen in the spleen and cecal tonsils of a few pigeons and in the duodenum of one pigeon. There were various degrees of lymphoid depletion in the bursa, spleen, and bone marrow. The morphology of the inclusions in the bursa and size of the viral particles are most consistent with circovirus. Preliminary studies on the bursae of two pigeons were negative for psittacine beak and feather disease (PBFD) viral antigen and nucleic acid by immunoperoxidase staining, DNA in situ hybridization, and polymerase chain reaction techniques, suggesting that this virus differs from PBFD virus. Most of the pigeons had concurrent infections such as paramyxovirus-1, salmonellosis, herpesvirus, and hepatic and cerebral trichomoniasis associated with adenovirus.

Vaccination and challenge studies with psittacine beak and feather disease virus.

Psittacine beak and feather disease virus (PBFDV) was administered to adult galahs (Eolophus roseicapillus) by mouth or by intramuscular injection. Concentration of PBFDV antibodies in serum and excretion of PBFDV were monitored by haemagglutination inhibition (HI) and haemagglutination (HA) respectively. After oral administration, 17 of 18 galahs remained clinically normal and a small rise in antibody titre was detected in 3 of 18 birds. After intramuscular administration, antibody was detected in all birds. PBFDV was not detected in the feather dander of birds in either group. One bird developed diarrhoea and high faecal HA titres within 4 days of oral administration and then died. Adult and nestling cockatoos were vaccinated with an experimental inactivated double-oil emulsion vaccine. PBFDV antibody responses are comparable to those induced by a primary-oil emulsion vaccination regimen using Freund's adjuvants. Both vaccines protected nestlings. Three sibling wild-caught sulphur-crested cockatoos were vaccinated but died of PBFD before experimental challenge despite antibody responses in all birds. Unvaccinated control chicks developed acute PBFD within 4 weeks of challenge, probably from PBFDV-induced hepatitis since high concentrations of PBFDV were detected in their livers.
