Effect of royal jelly on chronıc lıver and kıdney damage ınduced by cadmıum chlorıde ın rats

Forty eight rats were used to evaluate the protective effect of royal jelly (RJ) against chronic liver and kidney damage induced by cadmium chloride (CdCl2). In the histopathology of the CdCl2 group, the findings of inflammation and necrosis in liver sections, inflammation in kidney sections, proximal tubule degeneration and adiposity in the glomerulus were similar to the histopathological findings in the CdCl2 + RJ group. In immunohistochemical staining, it was determined that Caspase-3, Iba-1, KIM-1 and Tnf-a immunoreactivities in the liver and kidney tissue sections of the CdCl2-only group (Group III) were significantly increased compared to the control groups (Groups I, II). In the CdCl2 + RJ applied group (Group IV), Caspase-3, Iba-1, KIM-1 and TNF-α immunoreactivities were observed to be partially reduced compared to the CdCl2 (Group III) group. It was determined that AST, ALT, glucose, triglyceride, cholesterol and creatinine levels increased in the CdCl2 group and decreased in the CdCl2 + RJ group. While MDA levels increased in the group given only CdCl2 (Group III) compared to the control groups (Groups I, II), GSH levels decreased. In the CdCl2 + RJ group (Group IV), it was determined that MDA levels decreased and GSH levels increased compared to the CdCl2 (Group III) group. While the amounts of cadmium, zinc, copper, iron, magnesium and calcium in the liver tissue homogenates increased significantly in the CdCl2 group, it was observed that they decreased numerically in the CdCl2 + RJ group. This result shows that royal jelly application against CdCl2 toxicity does not prevent liver and kidney tissue damage, but can bring serum biochemical parameters, lipid peroxidation and trace element levels closer to control group levels.

Structural changes in testes of Streptozotocin induced diabetic rats and possible protective effect of royal jelly: light and electron microscopic study

ABSTRACT Diabetes mellitus (DM) is one of the most common metabolic diseases causing damage in many organs in the body including the testes. Royal Jelly (RJ) is one of the honey bee products that has antioxidant, anti-inflammatory and antidiabetic properties. This study was performed to evaluate the changes in the microscopic structure of the testes in Streptozotocin (STZ)-induced diabetic rats, and the possible protective role of RJ. 60 adult male albino rats were divided into three groups. Group I Control group, Group II STZ group, and Group III STZ+RJ group. Group II received a single dose of STZ (50 mg/kg) by intraperitoneal injection. Group III received a single dose of STZ as in the second group then received RJ orally by intragastric tube in dose of (100 mg/kg/day) for 4 weeks after confirmation of diabetes. Light and electron microscopic studies were performed. Group II revealed marked structural changes affecting seminiferous tubules with sever reduction in germinal epithelium and loss of mature spermatozoa in their lumina. The interstitial tissue revealed degenerated Leydig cells and congested blood vessels. Mallory trichrome stained section of group II revealed marked increase in the amount of collagen fibers. Group III revealed highly preserved testicular architecture almost near to that appeared in the control group except few tubules that were damaged. In conclusion, RJ protected the testicular structure from the damaging effect of diabetic oxidative stress through its antioxidant effect thus preserving male fertility.

In this study, the effects of Royal Jelly (RJ) on oxidative stress caused by exhaustive swimming exercise in rat tissues were evaluated. Methods: Twenty four male Wistar albino rats were indiscriminately distributed into four experimental groups: Sedentary control (SC); SC with administration of RJ (100 mg kg-1) (SC + RJ); exhaustive swimming exercise (E); Exhaustive swimming exercise with administration of RJ (100 mg kg-1) (E + RJ). 100 mg kg-1 of RJ were dissolved in drinking water. Rats in the SC+RJ and E+RJ groups supplemented with RJ (100 mg kg-1) orally once a day for two weeks. Rats in groups E and E+RJ subjected to acute exhaustive swimming exercise on the 14th day of the study, then some biochemical parameters related to oxidative stress of all groups were measured. Results: The activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), blood urea nitrogen (BUN) and creatinine (CRE) levels significantly raised in the exercised rats compared with the sedentary rats (P < 0.05). The decreased superoxide dismutase (SOD), glutathione peroxidase (GSHPx), and catalase (CAT) activities of muscular and hepatic tissues significantly increased and the high malondialdehyde (MDA) levels of muscular, hepatic and kidney tissues significantly reduced in exercised rats treated with RJ (P < 0.05). Conclucion: Collectively, in this study, protective effects of RJ on tissues against oxidative damage after exhaustive exercise were observed.

Activation of AMPK Entails the Protective Effect of Royal Jelly against High-Fat-Diet-Induced Hyperglycemia, Hyperlipidemia, and Non-Alcoholic Fatty Liver Disease in Rats.

This study examined the mechanism underlying the protective effect of royal jelly (RJ) against high-fat-diet (HFD)-mediated non-alcoholic liver disease (NAFLD) in rats. Adult male rats were divided into five groups (n = 8 each): control fed a standard diet, control + RJ (300 mg/kg), HFD, HFD + RJ (300 mg/kg), and HFD + RJ + CC (0.2 mg/kg). The treatment with RJ reduced weight gain, increased fat pads, and attenuated fasting hyperglycemia, hyperinsulinemia, and glucose tolerance in the HFD-fed rats. It also reduced the serum levels of liver function enzymes, interleukin 6 (IL-6), tumor necrosis factor-α (TNF-α), and leptin but significantly increased the serum levels of adiponectin. In addition, and with no effect on lipid excretion in stool, RJ significantly decreased the hepatic mRNA expression of SREBP1, serum, hepatic cholesterol, and triglycerides but increased hepatic mRNA levels of PPARα. Furthermore, RJ reduced the hepatic levels of TNF-α, IL-6, and malondialdehyde (MDA) in the livers of these rats. Of note, with no effect on the mRNA levels of AMPK, RJ stimulated the phosphorylation of AMPK and increased the levels of superoxide dismutase (SOD) and total glutathione (GSH) in the livers of the control and HFD-fed rats. In conclusion, RJ attenuates NAFLD via its antioxidant potential and adiponectin-independent activation of liver AMPK.

Royal Jelly and Chlorella vulgaris Mitigate Gibberellic Acid-Induced Cytogenotoxicity and Hepatotoxicity in Rats via Modulation of the PPARα/AP-1 Signaling Pathway and Suppression of Oxidative Stress and Inflammation.

Gibberellic acid (GA3) is a well-known plant growth regulator used in several countries, but its widespread use has negative effects on both animal and human health. The current study assesses the protective effect of royal jelly (RJ) and Chlorella vulgaris (CV) on the genotoxicity and hepatic injury induced by GA3 in rats. Daily oral administration of 55 mg/kg GA3 to rats for 6 constitutive weeks induced biochemical and histopathological changes in the liver via oxidative stress and inflammation. Co-administration of 300 mg/kg RJ or 500 mg/kg CV with GA3 considerably ameliorated the serum levels of AST (aspartate aminotransferase), ALT (alanine aminotransferase), ALP (alkaline phosphatase), γGT (gamma-glutamyl transferase), total bilirubin, and albumin. Lowered malondialdehyde, tumor necrosis factor α (TNF-α), and nuclear factor κB (NF-κB) levels along with elevated SOD (superoxide dismutase), CAT (catalase), and GPx (glutathione peroxidase) enzyme activities indicated the antioxidant and anti-inflammatory properties of both RJ and CV. Also, they improved the histological structure and reduced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expressions along with up-regulating peroxisome proliferator activated receptor α (PPARα) and down-regulating activator protein 1 (AP-1) gene expression. Additionally, chromosomal abnormalities and mitotic index were nearly normalized after treatment with RJ and CV. In conclusion, RJ and CV can protect against GA3-induced genotoxicity and liver toxicity by diminishing oxidative stress and inflammation, and modulating the PPARα/AP-1 signaling pathway.

Royal jelly improves learning and memory deficits in an amyloid β-induced model of Alzheimer's disease in male rats: Involvement of oxidative stress.

Alzheimer's disease (AD) as the commonest type of dementia is associated with the cognitive function failure. Oxidative stress performs an essential role in the progression of AD. Royal jelly (RJ) is a natural product of bees with antioxidant and anti-inflammatory properties. The present research aimed to investigate the possible protective effect of RJ on learning and memory in a rat model of Aβ-induced AD. Forty male adult Wistar rats were equally distributed into five groups: control, sham-operated, Aβ (receiving intracerebroventricular (ICV) injection of amyloid beta (Aβ1-40)), Aβ + RJ 50mg/kg, and Aβ + RJ 100mg/kg. RJ was administered daily post-surgery by oral gavage for four weeks. Behavioral learning and memory were examined using the novel object recognition (NOR) and passive avoidance learning (PAL) tests. Also, oxidative stress markers, such as malondialdehyde (MDA), total oxidant status (TOS) and total antioxidant capacity (TAC), were assessed in the hippocampus. Aβ reduced step-through latency (STLr) and increased time spent in the dark compartment (TDC) in the PAL task and also decreased discrimination index in the NOR test. Administration of RJ ameliorated the Aβ-related memory impairment in both NOR and PAL tasks. Aβ decreased TAC and increased MDA and TOS levels in the hippocampus, whereas RJ administration reversed these Aβ-induced alterations. Our results indicated that RJ has the potential to ameliorate learning and memory impairment in the Aβ model of AD via attenuating oxidative stress.

Protective effects of royal jelly and Echinacea against moxifloxacin-induced renal and hepatic injury in rats

Antibiotic use, especially fluoroquinolones, has been linked to extensive renal and hepatic injury thus inflicts a considerable health problem. Fifty rats were allocated into five groups (n = 10). Group 1 represented the normal-control group. Group 2 received moxifloxacin only (MOX; 8 mg/kg/day, i.p.) for seven days and represented the MOX-control group. Groups 3, 4, and 5 received MOX for seven days accompanied by royal jelly (RJ; 100 mg/kg/day, p.o.), Echinacea (ECH; 40 mg/kg/day, p.o.), and a combination of both at the aforementioned doses respectively for 30 days. All groups were investigated for renal and hepatic function tests. Renal tissue content of kidney injury molecule-1 (KIM-1) along with renal and hepatic tissue contents of reduced glutathione (GSH) and malondialdehyde (MDA) were assessed for all groups. Histopathological examination was performed followed by immunohistochemical staining for caspase-3 in renal and hepatic tissues. MOX administration resulted in significant renal and hepatic damage. RJ and ECH significantly improved the serum parameters of renal and hepatic functions along with increasing GSH and decreasing MDA in renal and hepatic tissues. Renal contents of KIM-1 were also reduced. Moreover, RJ, ECH, and their combination amended MOX-induced histopathological changes and significantly reduced caspase-3 immunohistochemical staining in both renal and hepatic tissues. The current study is the first to elucidate the effect of RJ, ECH, and their combination against MOX-induced renal and hepatic injury in rats. The study suggests that these protective effects are mainly via the reduction of oxidative stress induced by MOX administration.

Protective effect of royal jelly on fluoride-induced nephrotoxicity in rats via the some protein biomarkers signalling pathways: a new approach for kidney damage

Introduction Protective effect of royal jelly (RJ) on fluoride-induced nephrotoxicity was investigated in this study. Methods 42 healthy male Wistar rats (n = 42, 8 weeks of age) were divided equally into 6 groups with 7 rats in each; (1) Group-1: Controls fed with standard diet; (2) Group-2: RJ [100 mg/kg] bw (body weight), by oral gavage; (3) Group-3: Fluoride [50 mg/kg] bw, in drinking water; (4) Group-4: Fluoride [100 mg/kg] bw, in drinking water; (5) Group-5: RJ [100 mg/kg] bw, by oral gavage + Fluoride [50 mg/kg] bw, in drinking water; (6) Group-6: RJ [100 mg/kg] bw, by oral gavage + Fluoride [100 mg/kg] bw, in drinking water. After 8 weeks, all rats were decapitated and their kidney tissues were removed for further analysis. The protein expression levels of caspase-3, caspase-6, caspase-9, Bcl-2, Bax, VEGF, GSK-3, BDNF, COX-2 and TNF-α proteins in kidney tissue were analysed by western blotting technique Results RJ increased Bcl-2, COX-2, GSK-3, TNF-α and VEGF protein levels and a decreased caspase-3, caspase −6, caspase-9, Bax and BDNF protein levels in fluoride-treated rats. Conclusion RJ application may have a promising therapeutical potential in the treatment of many diseases in the future by reducing kidney damage.

Effect of Nicotine on the Rat’s Tongue and the Possible Protective Role of Royal Jelly Light and Scanning Electron Microscopic Study

Background: nicotine (NIC) is an active substance in tobacco, the excessive use of tobacco products has been associated with various lesions in the oral cavity. Royal jelly (RJ) believed by many authors to be an anti-oxidant that protects different organs. Aim of the work: this study aimed to detect the effect of nicotine on the structure of the tongue and the possible protective effect of royal jelly. Materials and methods: 30 adult male rats were equally categorized into 3 groups. Group I: 10 rats used as the control group; Group II: 10 rats received nicotine sulphate 2 mg/kg subcutaneously daily for 4 weeks; Group III: 10 rats received nicotine as the previous group and royal jelly (100 mg/kg by intra-gastric tube daily) for 4 weeks. After 24 hrs of the last dose, the animals were dissected. Tongues were processed for the histological and scanning electron microscopic study.Results: group II revealed that the lingual dorsal surface was formedof irregularly and destructed papillae. Most of these papillae were thin with blunted or absent tips. Fungiform papillae showed destruction of their epithelium and ill-defined pores. Lamina propria showed depletion of the collagen fibers, skeletal muscle fibers revealed separations and destruction. Group III revealed restoration of the normal appearance at some regions and presence of nicotine effects at the others within light microscope and apparent restoration of the normal shape as seen within the electron microscope. Conclusion nicotine has harmful effects on the tongue and the addition of RJ can decrease these harmful effects, but not prevent it.

Protective effects of royal jelly on testicular torsion induced ischaemia reperfusion injury in rats.

This study was performed to investigate the protective effects of royal jelly (RJ) on a testicular torsion-induced ischaemia/reperfusion (I/R) injury in adult rats. A total of 40 male Wistar rats were divided into four groups, including 10 rats in each group: Group 1 (sham), Group 2 (Control), group 3 (I/R rats treated with 100mg/kg RJ for 50days after torsion) and group 4( I/R rats treated with 20mg/kg vitamin C for 50days after torsion). Testicular torsion was created by rotating the right testes 720° a clockwise direction for 90min. The levels of testosterone were measured by ELISA. Pathological evaluation, mean maturity and quality of the seminiferous tubules were used. Results showed that the testicular histopathology standards and testosterone levels changes were statistically significant in groups 3 and 4. The results obtained in this study may suggest that RJ like vitamin C had protective effects on a testicular ischaemia/reperfusion-induced injury in rats.

Involvement of Mfn2, Bcl2/Bax signaling and mitochondrial viability in the potential protective effect of Royal jelly against mitochondria-mediated ovarian apoptosis by cisplatin in rats.

Objective(s):The current study aimed to assess cisplatin-mediated ovarian apoptosis in a rat model by Royal jelly (RJ). Materials and Methods:Thirty female adult albino rats (180-200 g) were divided into three groups (n=10): saline (0.9% NaCl, IP) was given to the control group, the cisplatin group: received (5 mg/kg/once a week IP) for 5 successive weeks, the RJ+Cis. group: received RJ (100 mg/kg/ day PO daily), and Cisplatin (5 mg/kg/once per week IP) for 5 successive weeks. At the end of the experiment, rats were sacrificed and their ovaries were isolated and used for biochemical analysis, molecular investigations and morphometric assessment as well as histological study. Moreover, blood samples were collected for determination of follicle-stimulating hormone (FSH), luteinizing hormone (LH), Estradiol, progesterone and anti-mullerian hormone (AMH).Results:The current study clarified that RJ given to rats prior to cisplatin significantly increased the ovarian and uterine weights, in addition to follicular count at P˂0.05 compared to rats injected only with cisplatin. Moreover, it restored normal ovarian histological structure with a concurrent reduction in FSH, and LH levels, and increased AMH and ovarian hormone concentrations at P˂0.05 compared to cisplatin group. Also, RJ decreased the ovarian antioxidant/oxidative imbalance harmonized with significant suppression of inducible nitric oxide synthase and increase of quinone oxidoreductase 1 mRNA expression at P˂0.05 compared to cisplatin group.Conclusion:We concluded that RJ could alleviate mitochondrial-induced ovarian apoptosis caused by cisplatin via increasing anti-apoptotic Bcl2, and diminishing pro-apoptotic Bax with a concomitant increase of Mfn2 mRNA and protein expressions.

Protective effect of royal jelly against diclofenac-induced hepato-renal damage and gastrointestinal ulcerations in rats

ObjectiveEvaluation of traditionally used royal jelly (RJ) for the management of hepato-renal damage and gastrointestinal ulcerations caused by diclofenac. MethodsForty adult male Wistar rats were allocated into four groups. Rats of the 1st group received only saline and served as normal group. The remaining 3 groups received diclofenac (50 mg/kg/day, I.P.) for 7 days. Group 2 served as diclofenac-control group. Groups 3 and 4 received RJ (150 and 300 mg/kg/day, P.O.) respectively for 30 days. Twenty-four hours after the last treatment, blood samples were collected, rats were sacrificed, and livers, kidneys, stomachs & intestines were harvested. Stomachs and intestines were tested for ulcer counts. Serum levels of AST, ALT, creatinine and urea were investigated. Hepatic, renal, gastric and intestinal tissue contents of myeloperoxidase (MPO) and prostaglandin-E2 (PGE2) were measured. Histopathological examinations were also performed followed by immunohistochemical determination of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expression. ResultsDiclofenac administration caused significant deterioration of all the above mentioned parameters. RJ improved hepatic and renal functions. Gastric and intestinal ulcer counts were significantly ameliorated. Hepatic, renal, gastric and intestinal tissue PGE-2 contents and COX-2 expression were significantly elevated. RJ also significantly reduced MPO content and iNOS expression as compared to diclofenac-control group. Improvements of the histopathological pictures of hepatic, renal, gastric and intestinal tissues were also apparent. ConclusionThe study demonstrates promising protective effects of RJ against diclofenac-induced hepato-renal damage and gastrointestinal ulceration in rats.

Protective Role of Royal Jelly Against Gamma Radiation Induced Oxidative Stress, Cardio-Toxicity and Organ Dysfunctions in Male Rats

Background: Oxidative stress is known to be a key factor in several diseases and was reported as a result of radiation exposure in human and experimental animals. Royal jelly contains free amino acids, proteins, sugars, fatty acids, minerals, and vitamins, which are reported as a preventive treatment of a wide range of disorders. Aim: The present study was designed to determine the possible protective effects of royal jelly against gamma (γ-) radiation-induced oxidative stress in male rats. Materials and Methods: Thirty two rats were divided into four groups (n=8), control, treated (rats were administrated by RJ (250 mg/kg/day) by stomach tube for 15 days, irradiated (rats were exposed to (10 Gy) whole body gamma radiations as a fractionated dose (2Gy X 5 times), treated irradiated, (rats were administrated by RJ (250 mg/kg/day) by stomach tube for 15 days before exposure to whole body gamma irradiation (10 Gy) as a fractionated dose (2X5). Serum reduced glutathione (GSH) content, superoxide dismutase (SOD) activity and lipid peroxide [malondialdehyde (MDA)] levels were assessed as well as serum total cholesterol (TC), triglyceride (TG), High density lipoprotein-cholesterol (HDL-C), low density lipoprotein-cholesterol (LDL-C), creatinine kinase-MB (CK-MB) and cardiac troponin I (cTnI). Also, histopathologic and histochemical changes were assessed. Results: The results showed that, GSH content, SOD activity and HDL-C were highly significantly decreased, while there was marked highly significant increase in MDA, TC, TG, LDL-C, CK-MB and cTnI due to irradiation. Numerous histopathologic changes were detected in the heart tissue of rats of the irradiated group with altered cardiac muscle fibres of the radiation-exposed group. These changes manifested good amelioration in the exposed groups that supplemented with RJ (250 mg/kg/day). Conclusion: Royal jelly has a beneficial role in reducing oxidative stress and cardio-toxicity induced by radiation exposure.
