Protective Effect Of The Antioxidant Butylated Hydroxytoluene (Bht) On Some Physiological And Biochemical Variables Of Male Rats Exposed To Stress By Starvation.

This study investigated the effect of Butylated Hydroxytoluene (BHT) on some physiological and biochemical parameters of rats subjected to stress by starvation, and 56 mature males were divided into eight groups.(Control, Olive Oil, Starvation, BHT Concentration (100 mg/kg), BHT Concentration (50 mg/kg), Starvation + BHT concentration (100 mg/kg), starvation + BHT concentration (50 mg/kg), starvation + olive oil. The results showed that the olive oil group and BHT groups at a concentration (50, 100 mg/kg) There was no significant difference in most of the treatments within the studied variables. In contrast, the starvation group showed a significant decrease in the concentration of (Total protein, Albumin, Lipase Enzyme, Lactate Dehydrogenase Enzyme (LDH), and Total Antioxidant Capacity (TAC). The control group, while the starvation + BHT group, did not show a concentration (100,50 mg/kg) Significant difference compared with the control group in all the variables as mentioned earlier, which shows clearly the protective effect of BHT against the negative effect of starvation by maintaining its concentrations within its normal level. Protective effect of butylated hydroxytoluene (BHT) against the clastogenic acitivity of cadmium chloride and potassium dichromate in hamster ovary cells

The effect of butylated hydroxytoluene (BHT), a widely used food additive, on chromosomal alterations induced by cadmium chloride (CC) and potassium dichromate (PD) in Chinese hamster ovary (CHO) cells was studied both at metaphase and anaphase-telophase. CHO cells were cultured for 15-16 h in the presence of PD (6.0, 9.0 or 12.0 <FONT FACE="Symbol">m</font>M), BHT (1.0 <FONT FACE="Symbol">m</font>g/ml), or PD plus BHT as well as CC (0.5, 1.0 and 2.0 <FONT FACE="Symbol">m</font>M), BHT or CC plus BHT for the analysis of chromosomal aberrations. To perform the anaphase-telophase test, cells were cultured in cover glasses and treated 8 h before fixation with the same chemicals. An extra dose of CC (4 <FONT FACE="Symbol">m</font>M) was used in this test. Both metal salts significantly increased chromosomal aberration frequencies in relation to untreated controls, and to DMSO- and BHT-treated cells. Post-treatment with BHT decreased the yield of chromosomal damage in relation to treatments performed with CC and PD. However, chromosomal aberration frequencies were significantly higher than those of the controls. In the anaphase-telophase test, CC significantly increased the yield of lagging chromosomes with the four doses employed and the frequency of lagging fragments with the highest dose. In combined treatments of CC and BHT, frequencies of the two types of alterations decreased significantly in relation to the cells treated with CC alone. No significant variation was found in the frequencies of chromatin bridges. Significant increases of numbers of chromatin bridges, lagging chromosomes and lagging fragments were found in cells treated with PD. The protective effect of BHT in combined treatments was evidenced by the significant decrease of chromatid bridges and lagging chromosomes in relation to PD-treated cells. Whereas BHT is able to induce chromosomal damage, it can also protect against oxidative damage induced by other genotoxicants.

The effects of butylated hydroxytoluene on radiation and chemically-induced genetic damage in Drosophila melanogaster

This paper reports the results of a study on the effects of the phenolic antioxidant butylated hydroxytoluene (BHT) on the sensitivity of Drosophila germ cells to the X-ray and chemical induction of genetic damage. Prior to irradiation or chemical treatment, the flies were raised in normal, BHT-containing or dimethyl sulphoxide (DMSO)-containing media. In one series of X-ray experiments, male pupae raised in these different media were irradiated. The concentration of BHT in the medium was 0.05% (in 0.5% DMSO) and that of DMSO, 0.5%. In the X-ray experiments, the frequencies of sex-linked recessive lethals and of autosomal translocations in spermatids of males raised in the BHT-containing medium were significantly lower than in those of males raised in the other two media. If however, the spermatids in pupae were irradiated, the amounts of genetic damage in the three groups of males were nearly the same. Likewise, no significant protective effects attributable to BHT treatment could be demonstrated with respect to the induction of dominant lethals in mature spermatozoa, stage-7 and stage-14 oocytes or of autosomal recessive lethals and sex-chromosome losses in stage-7 oocytes. In the dominant lethal experiments with mature spermatozoa and stage-7 oocytes, the yields were generally higher in males of the DMSO group than in the other two, pointing to a radiosensitizing effect of DMSO. In the ‘maternal effect’ experiments (males raised in a normal medium irradiated and mated to females raised in normal, DMSO-containing or BHT-containing media), the frequencies of dominant lethals in spermatozoa were similar in all the three groups, suggesting no DMSO- or BHT-mediated maternal effects. With both chemicals tested (diepoxybutane and diethylnitrosamine), significantly lower yields of sex-linked recessive lethals were recovered in spermatids of males raised in the BHT-containing medium than in those of males raised in the other two media. In some of these experiments, this protective effect of BHT was measurable in more advanced stages of spermatogenesis as well. These results have been interpreted taking into account the radical-scavenging, enzyme-inducing and enzyme-inhibiting properties of BHT.
