Alterations in neuromuscular junction morphology with ageing and endurance training modulate neuromuscular transmission and myofibre composition.

Both ageing and exercise training affect the neuromuscular junction (NMJ) structure. Morphological alterations in the NMJ have been considered to influence neuromuscular transmission and myofibre properties, but the direct link between the morphology and function has yet to be established. We measured the neuromuscular transmission, myofibre composition and NMJ structure of 5-month-old (young) and 24-month-old untrained (aged control) and trained (aged trained) mice. Aged trained mice were subjected to 2 months of endurance training before the measurement. Neuromuscular transmission was evaluated in vivo as the ratio of ankle plantar flexion torque evoked by the sciatic nerve stimulation to that by direct muscle stimulation. The torque ratio was significantly lower in aged mice than in young and aged trained mice at high-frequency stimulations, showing a significant positive correlation with voluntary grip strength. The degree of pre- to post-synaptic overlap of the NMJ was also significantly lower in aged mice and positively correlated with the torque ratio. We also found that the proportion of fast-twitch fibres in the soleus muscle decreased with age, and that age-related denervation occurred preferentially in fast-twitch fibres. Age-related denervation and a shift in myofibre composition were partially prevented by endurance training. These results suggest that age-related deterioration of the NMJ structure impairs neuromuscular transmission and alters myofibre composition, but these alterations can be prevented by structural amelioration of NMJ with endurance training. Our findings highlight the importance of the NMJ as a major determinant of age-related deterioration of skeletal muscles and the clinical significance of endurance training as a countermeasure. KEY POINTS: The neuromuscular junction (NMJ) plays an essential role in neuromuscular transmission and the maintenance of myofibre properties. We show that neuromuscular transmission is impaired with ageing but recovered by endurance training, which contributes to alterations in voluntary strength. Neuromuscular transmission is associated with the degree of pre- to post-synaptic overlap of the NMJ. Age-related denervation of fast-twitch fibres and a shift in myofibre composition toward a slower phenotype are partially prevented by endurance training. Our study provides substantial evidence that age-related and exercise-induced alterations in neuromuscular transmission and myofibre properties are associated with morphological changes in the NMJ.

"Biqi" Bayberry Extract Promotes Skeletal Muscle Fiber Type Remodeling by Increasing Fast Myofiber Formation via the Akt/FoxO1 Pathway in Mice.

Bayberry is known to be a rich source of natural flavonoids and has been reported to have various health-promoting abilities. However, its function on regulating skeletal muscle fiber type remains unclear. This study examined whether bayberry extract affects skeletal muscle fiber type by promoting fast myofiber formation, as well as the potential molecular mechanism. After eight weeks, the "Biqi" bayberry extract (BBE) groups mice displayed markedly enhanced grip strength and improved metabolic rate compared to the control group mice. BBE also significantly increased myofibers size, LDH activity, MyHC-IIb (fast-twitch mRNA) expression, and the percentage of fast-twitch myofibers, while decreasing SDH activity, MyHC-I (slow-twitch mRNA) expression, and slow-twitch myofibers percentage in the skeletal muscle of the mice. The effect of BBE on regulating skeletal muscle fiber type remodeling is likely attributed to its activation of the Akt-FoxO1 pathway. Our findings indicated that BBE can effectively regulate the expression and proportion of fast-twitch fibers, making it a potential therapy for improving glucose homeostasis of skeletal muscle.

Nitrosative and Oxidative Stress, Reduced Antioxidant Capacity, and Fiber Type Switch in Iron-Deficient COPD Patients: Analysis of Muscle and Systemic Compartments

We hypothesized that a rise in the levels of oxidative/nitrosative stress markers and a decline in antioxidants might take place in systemic and muscle compartments of chronic obstructive pulmonary disease (COPD) patients with non-anemic iron deficiency. In COPD patients with/without iron depletion (n = 20/group), markers of oxidative/nitrosative stress and antioxidants were determined in blood and vastus lateralis (biopsies, muscle fiber phenotype). Iron metabolism, exercise, and limb muscle strength were assessed in all patients. In iron-deficient COPD compared to non-iron deficient patients, oxidative (lipofuscin) and nitrosative stress levels were greater in muscle and blood compartments and proportions of fast-twitch fibers, whereas levels of mitochondrial superoxide dismutase (SOD) and Trolox equivalent antioxidant capacity (TEAC) decreased. In severe COPD, nitrosative stress and reduced antioxidant capacity were demonstrated in vastus lateralis and systemic compartments of iron-deficient patients. The slow- to fast-twitch muscle fiber switch towards a less resistant phenotype was significantly more prominent in muscles of these patients. Iron deficiency is associated with a specific pattern of nitrosative and oxidative stress and reduced antioxidant capacity in severe COPD irrespective of quadriceps muscle function. In clinical settings, parameters of iron metabolism and content should be routinely quantify given its implications in redox balance and exercise tolerance.

The physiology of ice hockey performance: An update.

Ice hockey is an intense team sport characterized by repeated bursts of fast-paced skating, rapid changes in speed and direction and frequent physical encounters. These are performed in on-ice shifts of ~30-80 s interspersed with longer sequences of passive recovery, resulting in about 15-25 min on-ice time per player. Nearly 50% of the distance is covered at high-intensity skating speeds and with an accentuated intense activity pattern in forwards compared to defensemen. During ice hockey match-play, both aerobic and anaerobic energy systems are significantly challenged, with the heart rate increasing toward maximum levels during each shift, and with great reliance on both glycolytic and phosphagen ATP provision. The high-intensity activity pattern favors muscle glycogen as fuel, leading to pronounced reductions despite the relatively brief playing time, including severe depletion of a substantial proportion of individual fast- and slow-twitch fibers. Player-tracking suggests that the ability to perform high-intensity skating is compromised in the final stages of a game, which is supported by post-game reductions in repeated-sprint ability. Muscle glycogen degradation, in particular in individual fibers, as well as potential dehydration and hyperthermia, may be prime candidates implicated in exacerbated fatigue during the final stages of a game, whereas multiple factors likely interact to impair exercise tolerance during each shift. This includes pronounced PCr degradation, with potential inadequate resynthesis in a proportion of fast-twitch fibers in situations of repeated intense actions. Finally, the recovery pattern is inadequately described, but seems less long-lasting than in other team sports.

Changes in masseter muscle fibers by liquid diet rearing in rabbits and recovery by chewing of solid diet

ObjectiveTo investigate the effects of liquid diet on the development of masseter muscle fibers and whether the changes in the masseter muscle can be recovered by chewing of solid diet. DesignMasseter muscles from 40 rabbits (solid- and liquid-diet groups, n = 30; unweaned group, n = 5; recovery group, n = 5) were histochemically examined at 4, 12, 18, and 33 weeks after birth. Six fiber types (I, IC, IIC, IIA, IIAB, and IIB) were distinguished via mATPase staining. Muscle fiber diameter and fiber type composition were measured and compared between groups. ResultsIn the liquid diet group, the diameter of types IIAB (solid group: 81.7 μm, liquid group: 60.9 μm) and IIB (solid group: 89.3 μm, liquid group: 68.8 μm) and the fiber type composition of type I (solid group: 18.4%, liquid group: 9.6%) decreased significantly at 33 weeks of age. In the recovery group, the fiber type composition of type I fibers recovered to 16.5%, while no recovery of type IIAB (56.6 μm) and IIB (64.6 μm) fiber diameter was observed. ConclusionsLiquid diet caused atrophy of muscle fibers and an increase in the proportion of fast-twitch fibers. Although the diameter and ratio of slow-twitch fibers were recovered by chewing of solid diet, recovery was not observed for fast-twitch fibers. Our findings are relevant for dental medicine as it explored the possibility of masticatory muscle function recovery by hard food.

Muscle fibre types in the reduced forelimb and enlarged hindlimb of the quokka (Setonix brachyurus, Macropodidae)

The quokka (Setonyx brachyurus) is restricted to two offshore islands and small isolates on the mainland of south-western Australia. It displays a tendency to saltatorial locomotion and moves at speed by bipedal hopping, although it also uses its forelimbs at low speed. Its bipedal adaptation involves enlarged hind limbs, with elongated feet. The fibre type distribution of the elbow and knee extensors, and the ankle plantar flexors, in comparison with two eutherians, the quadrupedal rhesus monkey, as a locomotor generalist, and the jerboa, a small eutherian hopping species morphologically similar to the quokka, were studied. The quokka’s forelimb showed the same characteristics as that of the jerboa, lacking the fatigue-resistant Type I fibres that are used to sustain posture. As in the jerboa, the gastrocnemius lateralis was the muscle head with the highest proportion of fast twitch fibres. Muscular fibre pattern is not identical in the quokka and the jerboa hindlimb, but it appears that both species have similar anatomical adaptations to saltatorial locomotion. Differences in muscle fibre proportions could be due to several factors including, resting posture, body size and the propensity for elastic energy storage, the burrowing behaviour of the jerboa, but also to phylogenetic constraints where the adaptation to hop on the hindlimbs is a shared behaviour of the Macropodoidea (jerboas are the only Dipodidae to have elongated hindlimbs).

In vivo maximal fascicle-shortening velocity during plantar flexion in humans.

Interindividual variability in performance of fast movements is commonly explained by a difference in maximal muscle-shortening velocity due to differences in the proportion of fast-twitch fibers. To provide a better understanding of the capacity to generate fast motion, this study aimed to 1) measure for the first time in vivo the maximal fascicle-shortening velocity of human muscle; 2) evaluate the relationship between angular velocity and fascicle-shortening velocity from low to maximal angular velocities; and 3) investigate the influence of musculo-articular features (moment arm, tendinous tissues stiffness, and muscle architecture) on maximal angular velocity. Ultrafast ultrasound images of the gastrocnemius medialis were obtained from 31 participants during maximal isokinetic and light-loaded plantar flexions. A strong linear relationship between fascicle-shortening velocity and angular velocity was reported for all subjects (mean R(2) = 0.97). The maximal shortening velocity (V(Fmax)) obtained during the no-load condition (NLc) ranged between 18.8 and 43.3 cm/s. V(Fmax) values were very close to those of the maximal shortening velocity (V(max)), which was extrapolated from the F-V curve (the Hill model). Angular velocity reached during the NLc was significantly correlated with this V(Fmax) (r = 0.57; P < 0.001). This finding was in agreement with assumptions about the role of muscle fiber type, whereas interindividual comparisons clearly support the fact that other parameters may also contribute to performance during fast movements. Nevertheless, none of the biomechanical features considered in the present study were found to be directly related to the highest angular velocity, highlighting the complexity of the upstream mechanics that lead to maximal-velocity muscle contraction.

The effect of icaritin on skeletal muscle atrophy following mechanical unloading

Skeletal muscle undergoes rapid and profound atrophy in response to decreased mechanical loading, e.g., limb immobilization, bed rest, denervation, hindlimb suspension and spaceflight. This study was to investigate the effect of icaritin (ICT) on skeletal muscle atrophy following hindlimb unloading (HU). Male adult rats were suspended by tail for 28 days to induce HU-related skeletal muscle atrophy. Rats were orally administered daily with ICT at low (80 mg/kg, Low-ICT) and high (120 mg/kg, High-ICT) doses throughout the HU period for a total of 28 days. Age-matched, weight-bearing rats served as the control (n = 8 for each group). The results showed that soleus muscle fiber cross-sectional area in High-ICT group was significantly larger than that in HU only group. The proportion of fast-twitch fibers in High-ICT group was lower than that in HU only group. Functional tests showed that the maximal tetanic force of soleus muscle in High-ICH group was remarkably higher than that in HU only group, and the contraction time and half relaxation time in High-ICT group didn't shorten as HU group did. Current data indicates that high dose of icaritin treatment could prevent skeletal muscle atrophy and delay the slow-to-fast muscle fiber type transformation following mechanical unloading.

Muscle fiber type effects on energetically optimal cadences in cycling

Fast-twitch (FT) and slow-twitch (ST) muscle fibers vary in their mechanical and energetic properties, and it has been suggested that muscle fiber type distribution influences energy expenditure and the energetically optimal cadence during pedaling. However, it is challenging to experimentally isolate the effects of muscle fiber type on pedaling energetics. In the present study, a modeling and computer simulation approach was used to test the dependence of muscle energy expenditure on pedaling rate during submaximal cycling. Simulations were generated using a musculoskeletal model at cadences from 40 to 120 rev min −1, and the dynamic and energetic properties of the model muscles were scaled to represent a range of muscle fiber types. Energy expenditure and the energetically optimal cadence were found to be higher in a model with more FT fibers than a model with more ST fibers, consistent with predictions from the experimental literature. At the muscle level, mechanical efficiency was lower in the model with a greater proportion of FT fibers, but peaked at a higher cadence than in the ST model. Regardless of fiber type distribution, mechanical efficiency was low at 40 rev min −1, increased to a broad plateau between 60 and 100 rev min −1 , and decreased substantially at 120 rev min −1. In conclusion, muscle fiber type distribution was confirmed as an important determinant of the energetics of pedaling.

Protein use and muscle-fiber changes in free-ranging, hibernating black bears.

Studies of the metabolic and physiological changes that bears undergo during hibernation have, for the most part, supported the paradigm that bears use only fatty tissues as a metabolic substrate during hibernation. This study was performed to document the extent of protein loss and alteration of muscle-fiber characteristics of selected muscles in black bears during winter dormancy. Muscle biopsies were removed from the gastrocnemius and biceps femoris from seven free-ranging female black bears on the Uncompahgre Plateau in west-central Colorado. Six of the seven bears produced cubs during the hibernating season. Muscle samples were collected from the left hind limb shortly after bears entered their dens (fall), and additional samples were collected from the right hind limb just prior to bears leaving their dens (spring). Protein concentration, fast- and slow-twitch muscle-fiber ratios and muscle-fiber cross-sectional areas, and citrate synthase activity were measured in the laboratory. While protein concentration decreased in both muscles during the hibernation period, it was lower than predicted for lactating females. In addition, muscle-fiber number and cross-sectional area were unchanged in these muscles, suggesting only limited muscle atrophy. In support of these observations, there was a moderate but significant increase in the proportion of fast-twitch fibers only in the biceps femoris, with a concomitant decrease in citrate synthase activity, but no alteration of the fiber ratio in the gastrocnemius during hibernation. These findings suggest that hibernating bears, particularly lactating females, do use some protein, in concert with fat catabolism, as a metabolic substrate and as a source of water. However, the extent of this protein use is moderate and is associated with limited alteration of muscle structure, characteristic of disuse atrophy.

Muscle fibre type and dimension in genetically obese and lean Zucker rats

Skeletal muscle structure and morphology may be altered in obesity. To study this further, muscles from six genetically obese (fa/fa) and six normal male rats were examined at 15 weeks of age. The gluteus medius, vastus lateralis and rectus abdominis muscles were dissected out and stained for histochemical fibre typing. In addition the fibre cross-sectional area was measured on a graphic tablet. The proportion of fast-twitch fibres was larger in the vastus lateralis and rectus abdominis muscles of the obese rats (P < 0.01); no difference was seen for the gluteus medius muscle. For the normal rats the cross-sectional area of the fast-twitch fibres was 2-3 times larger than the area of slow-twitch fibres in the same muscle. The cross-sectional area of the fast-twitch fibres in the obese rats was 40-47% less than in the control animals (P < 0.003), while no difference between the two groups was found for the slow-twitch fibre area. The data thus suggest that in the genetically obese rats the development of fast-twitch fibres was primarily affected. Moreover, in these animals some muscles may be more affected than others.

Effects of age on calcium transport activity of sarcoplasmic reticulum in fast- and slow-twitch rat muscle fibres.

1. The calcium transport activity of the sarcoplasmic reticulum (SR) was measured in chemically skinned single fast- and slow-twitch muscle fibres from young (3 months) and old (23-24 months) rats. Contractile properties, the myosin heavy chain (MHC) composition and enzyme histochemical features were studied in relation to the SR characteristics. 2. In fast-twitch single motor units, the contraction time of the isometric twitch increased (P less than 0.001) from 13 +/- 1 ms in young animals to 18 +/- 2 ms in old ones. In the slow-twitch soleus, the contraction (P less than 0.001) and half-relaxation (P less than 0.05) times increased from 30 +/- 5 and 45 +/- 10 ms, respectively, in the young animals to 43 +/- 3 and 55 +/- 4 ms in the old ones. The proportion of slow-twitch (type I) fibres increased (P less than 0.05) with age in the soleus from 92 +/- 6 to 98 +/- 2% and the proportion of fast-twitch fibres (type IIA) decreased (P less than 0.01) from 6 +/- 5 to 0 +/- 0%. 3. The Ca2+ accumulation capacity (an index of SR volume), the rate of Ca2+ uptake and the fractional rate of SR filling (an estimate of the specific activity of the Ca2+ pump) were decreased by 18 (P less than 0.05), 32 (P less than 0.01) and 32% (P less than 0.001), respectively, in the old fast-twitch muscle fibres. In the slow-twitch muscle fibres, on the other hand, no significant age-related changes were observed in the Ca2+ transport activity of the SR. Thus, ageing exerts a differential influence on SR volume and function in fast- and slow-twitch fibres. 4. It is concluded that an age-related impairment of intrinsic SR function and a decrease in SR volume are probable factors underlying the decreased speed of contraction of fast-twitch muscle fibres in old age. In the slow-twitch soleus, on the other hand, one or more other mechanisms are responsible for the age-related decrease in the speed of contraction. The loss of fast-twitch muscle fibres in old soleus is one mechanism, but not the dominant one.

Protein metabolism in skeletal muscle, diaphragm, and heart of diabetic rats.

Rates of protein synthesis were measured in several muscle types in young streptozotocin-diabetic rats at different times after withdrawal of insulin therapy. Protein synthesis decreased in all types of muscle studied. In muscles with a substantial proportion of fast-twitch fibers (gastrocnemius and extensor digitorum longus), this decline was associated with falls in both protein synthesis-to-RNA and RNA-to-protein ratio, but in soleus and heart the effect could be explained entirely in terms of decreased ribosome concentration. Rates of protein degradation were calculated for heart, diaphragm, and gastrocnemius muscle from the changes in protein synthesis and protein content and were found in all cases to be elevated by 4 days of insulin withdrawal. However, measurements of protein synthesis and protein mass in the gastrocnemius muscle of a group of chronically diabetic rats indicated that in this condition, unlike in acute diabetes, the rate of protein degradation was depressed.

Daily in vivo neuromuscular stimulation effects on immobilized rat hindlimb muscles.

The purpose of the investigation was to determine the effects of a daily regimen of near-maximal contractions, produced via in vivo electrical stimulation of peripheral nerve, on functional and histochemical properties of rat hindlimb muscles immobilized for 28 days in a plaster cast. Rats had knee and ankle joints of one hindlimb immobilized; then while anesthetized, half of the group was subjected to a daily regimen of 480 semifused tetanic contractions (50 Hz) via fine-wire electrodes chronically implanted around the sciatic nerve. Immobilization caused significant decreases in soleus and gastrocnemius muscle weights, fiber cross-sectional areas, and twitch and tetanic strength measured in situ. In addition, immobilized soleus muscles had faster time to peak tension (TPT) and higher proportions of fast-twitch fibers, whereas immobilized gastrocnemius muscles demonstrated faster half-relaxation times (RT1/2) and total twitch durations (TPT plus RT1/2). The only significant effects of the imposed contractions were evident in the gastrocnemius in which stimulation prevented the shortening of RT1/2 and total twitch duration and resulted in significantly higher relative tensions at 50 Hz and higher fatigue resistance. Muscle activity of this type imposed on immobilized muscle is ineffective in attenuating atrophy but can, in fast muscle such as gastrocnemius, prevent changes in twitch characteristics resulting from immobilization, as well as augment contractile responses during semifused and fatiguing contractions.

Variation among normal persons in short-term ventilatory capacity.

Among individuals of comparable size with normal lungs, short-term breathing capacity varies widely for unknown reasons. If the velocity of ventilatory muscle contraction limits airflow, then persons with a high proportion of fast twitch fibers in the ventilatory muscles should have large ventilatory capacities. As one test of this hypothesis, we correlated ventilatory capacity with vertical jump, an index of leg and thus of diaphragmatic muscle fiber composition. We found significant positive correlations between vertical jump and the 12-second maximal voluntary ventilation in both women (n = 19, r = 0.6; p less than 0.01) and men (n = 25; r = + 0.4; p = 0.06). Both correlations were independent of body weight or vital capacity. These results are consistent with the hypothesis that short-term ventilatory capacity is influenced by the contractile characteristics of the ventilatory muscles.

Central and peripheral circulation in relation to muscle-fibre composition in normo- and hyper-tensive man.

1. Fibre composition in the vastus lateralis muscle, and blood pressure, were determined in age-matched normotensive (n = 22) subjects and previously untreated patients (n = 19) with essential hypertension. 2. In all subjects the muscle fibres were classified, as fast-twitch (FT) and slow-twitch (ST), and blood pressure was recorded. Eleven patients and seven of the normotensive subjects participated in an extended haemodynamic study with intra-arterial pressure measurements and determinations of leg blood flow. 3. In both normo- and hyper-tensive subjects, ambulant and intra-arterial blood pressure and leg vascular resistance were negatively correlated to the percentage of ST fibres; no significant correlation was found to total leg blood flow. 4. The findings show that muscles with a high proportion of FT fibres have a higher resistance than muscles rich in ST fibres and suggest that the type of fibre in skeletal muscle might be of importance for the development of the hypertensive disease.

Muscle strength and fiber composition in athletes and sedentary men

Members of Swedish national teams in track and field events (sprinting and jumping), downhill skiing, race walking, orienteering, and a group of sedentary men were studied to examine the relationship between muslce fiber characteristics in needle biopsy samples form m. vastus lateralis and muscle strength measured as peak torque during isokinectic knee extensions. In comparison with the sedentary group the following differences were found: a) percentage fast twitch fibers was lower in the endurance athetes, b) fast to slow twitch muscle fiber area ratio was higher in the track athletes, c)track athletes and downhill skier attained higher peak torque values at all angular velocities examined. The track athletes had, however, higher torque values at the fastest angular velocity as compared to the downhill skiers, whereas there was no differnce under isometric conditions. The proportion of fast twitch fibers was related to torque produced, especially at high motion velocity. The training also appeared to affect the force-velocity relationship.
