Fish-derived lactic acid bacteria supplementation enhanced the immunity and resistance in Crucian carp (Carassius auratus)

This study attempted to evaluate the potential for biofilm formation and the probiotic properties of lactic acid bacteria (LAB) isolated from the Carassius auratus intestine and to determine their antibacterial activity against crucial injurious bacteria of fish. The biological features test was carried out to obtain LAB with well-resistant pH, trypsin, bile and antibiotic challenge, and biofilm formation capacity. The most promising LAB isolates, y11 and y78 were identified as Lactobacillus johnsonii and Weissella confusa using 16S ribosomal RNA gene sequencing. We discovered that oral LAB additives improved immunity in the C. auratus by increasing immune-related expression of immunoglobulin M (IgM), superoxide dismutase (SOD), alkaline phosphatase (AKP), and lysozyme (LYS) in serum and interleukins (IL-10 and IL-1β), interferon-γ (IFN-γ), and tumor necrosis factor-α (TNF-α) in organs. After the introduction of Aeromonas veronii TH0426 for 15 days, the survival rate of C. auratus was determined as follows: the protection rate of group y11 was 45 %, and group y78 was 65 % when challenged with 109 CFU/mL density. This experiment successfully selected two LAB strains as the bioprotective agents and provided theoretical support for further LAB application in aquaculture.

Probiotics for Aquaculture: Hope, Truth, and Reality.

The use of microorganisms as beneficial crops for human and animal health has been studied for decades, and these microorganisms have been in practical use for quite some time. Nowadays, in addition to well-known examples of beneficial properties of lactic acid bacteria, bifidobacteria, selected Bacillus spp., and yeasts, there are several other bacteria considered next-generation probiotics that have been proposed to improve host health. Aquaculture is a rapidly growing area that provides sustainable proteins for consumption by humans and other animals. Thus, there is a need to develop new technologies for the production practices associated with cleaner and environment-friendly approaches. It is a well-known fact that proper selection of the optimal probiotics for use in aquaculture is an essential step to ensure effectiveness and safety. In this critical review, we discuss the evaluation of host-specific probiotics in aquaculture, challenges in using probiotics in aquaculture, methods to improve the survival of probiotics under different environmental conditions, technological approach to improving storage, and delivery along with possible negative consequences of using probiotics in aquaculture. A critical analysis of the identified challenges for the use of beneficial microbes in aquaculture will help in sustainable aquafarming, leading to improved agricultural practices with a clear aim to increase protein production.

Anti-obesity effects of potential probiotic Lactobacillus strains isolated from Mongolian fermented dairy products in high-fat diet-induced obese rodent model.

This study aims to investigate the anti-obesity properties of lactic acid bacteria (LAB) isolated from fermented dairy products such as "Airag" and "Khoormog" in Mongolia. These traditional dairy products are widely used in Mongolia and believe in having potential probiotic, anti-diabetes, anti-cancer, and anti-tuberculosis properties and are made from unheated two-humped camel milk and mare milk, respectively. We chose three LAB strains based on their probiotic characteristics, including tolerance of gastric and bile acids. Then we checked the anti-obesity activity of probiotic strains in vivo. An animal model was evaluated in twenty male C57BL/6J mice by inducing obesity with a high-fat diet (HFD), which was divided into five groups: regular diet group (Negative control), HFD group (Positive control), HFD with Lacticaseibacillus paracasei X-1 (X-1), Lacticaseibacillus paracasei X-17 (X-17), and Limosilactobacillus fermentum BM-325 (BM-325). For six weeks, 5 × 109 colony-forming units (CFU) of bacteria were given orally to the LAB-fed groups. Fasting blood glucose (FBG), lipid profiles, organ index, and organ morphology were all measured. The probiotic strains suppressed growth in adipose cell volume, stabilized FBG, reduced liver cell degeneration, and slowed HFD-induced body weight gain. The results suggest that some strains increase general metabolism while lowering body weight.

Evaluation of functional properties of lactic acid bacteria isolated from traditional cheese

The objective of this research was to study a total of 10 strains of lactic acid bacteria previously isolated from traditional cheese, explore the effects of various in vitro tests, and evaluate their functional and potential probiotic properties. We tested the strains for cholesterol assimilation, antimicrobial activity, enzyme and exopolysaccharides (EPS) production, cell surface traits, antibiofilm, and tolerance to lysozyme and phenol. E. faecalis S135 and E. durans S1121 were characterized by strong antibacterial activities against C. albicans and S. aureus. The strains E. durans S092 and E. faecalis S135 were differentiated by phytase-producing and coagulation activity, respectively. Moreover, E. faecalis S135 had a greater cholesterol reduction percentage (66.6 ± 0.00 %) than the others. Separately, E. durans S104 and L. coryniformis subsp. torquens S232 were found to possess stronger cell surface properties, e.g., auto-aggregation (94.3 ± 0.30 %) and cell surface hydrophobicity (65.1 ± 0.11 %). All LAB 10 strains were able to handle 0.4 % phenol and 2 mg/mL lysozyme well. They also had some antibiofilm activity, but not much EPS production. Industrially important strains with antimicrobial activity, such as E. durans (S1121, S092, and S104) and E. faecalis S135, could serve as bio-protective agents. These four strains can therefore find application in various fields, including the production of fermented and/or functional foods and enzymes.

Harnessing the Health and Techno-Functional Potential of Lactic Acid Bacteria: A Comprehensive Review.

This review examines the techno-functional properties of lactic acid bacteria (LABs) in the food industry, focusing on their potential health benefits. We discuss current findings related to the techno-functionality of LAB, which includes acidification, proteolytic and lipolytic features, and a variety of other biochemical activities. These activities include the production of antimicrobial compounds and the synthesis of exopolysaccharides that improve food safety and consumer sensory experience. LABs are also known for their antioxidant abilities, which help reduce oxidative reactions in foods and improve their functional properties. In addition, LABs' role as probiotics is known for their promising effects on gut health, immune system modulation, cholesterol control, and general wellbeing. Despite these advantages, several challenges hinder the effective production and use of probiotic LABs, such as maintaining strain viability during storage and transport as well as ensuring their efficacy in the gastrointestinal tract. Our review identifies these critical barriers and suggests avenues for future research.

Comparative Genome Analysis and Characterization of the Probiotic Properties of Lactic Acid Bacteria Isolated from the Gastrointestinal Tract of Wild Boars in the Czech Republic.

Probiotics are crucial components for maintaining a healthy gut microbiota in pigs, especially during the weaning period. Lactic acid bacteria (LAB) derived from the gastrointestinal tract of wild boars can serve as an abundant source of beneficial probiotic strains with suitable properties for use in pig husbandry. In this study, we analyzed and characterized 15strains of Limosilactobacillusmucosae obtained from the gut contents of wild boars to assess their safety and suitability as probiotic candidates. The strains were compared using pan-genomic analysis with 49 L.mucosae strains obtained from the NCBI database. All isolated strains demonstrated their safety by showing an absence of transferrable antimicrobial resistance genes and hemolysin activity. Based on the presence of beneficial genes, five candidates with probiotic properties were selected and subjected to phenotypic profiling. These five selected isolates exhibited the ability to survive conditions mimicking passage through the host's digestive tract, such as low pH and the presence of bile salts. Furthermore, five selected strains demonstrated the presence of corresponding carbohydrate-active enzymes and the ability to utilize various carbohydrate substrates. These strains can enhance the digestibility of oligosaccharide or polysaccharide substrates found in food or feed, specifically resistant starch, α-galactosides, cellobiose, gentiobiose, and arabinoxylans. Based on the results obtained, the L.mucosae isolates tested in this study appear to be promising candidates for use as probiotics in pigs.

Enterotype-Dependent Probiotic-Mediated Changes in the Male Rat Intestinal Microbiome In Vivo and In Vitro.

Beneficial properties of lactic acid bacteria have been known long ago, but particular interest in probiotics has arisen in the last two decades due to the understanding of the important role of intestinal microflora in human life. Thus, the ability of probiotics to support healthy homeostasis of gut microbiomes has received particular attention. Here, we evaluated the effect of a probiotic consisting of Bifidobacterium longum and Lacticaseibacillus paracasei on the gut microbiome of male rats, assessed their persistence in the fecal biota, and compared probiotic-mediated changes in vitro and in vivo. As expected, microbiomes of two enterotypes were identified in the feces of 21 animals, and it turned out that even a single dose of the probiotic altered the microbial composition. Upon repeated administration, the E1 biota temporarily acquired properties of the E2 type. Being highly sensitive to the intervention of probiotic bacteria at the phylum and genus levels, the fecal microbiomes retained the identity of their enterotypes when transferred to a medium optimized for gut bacteria. For the E2 biota, even similarities between probiotic-mediated reactions in vitro and in vivo were detected. Therefore, fecal-derived microbial communities are proposed as model consortia to optimize the response of resident bacteria to various agents.

Investigation of the antidiabetic and probiotic properties of lactic acid bacteria isolated from some ethnic fermented foods of Darjeeling District

BackgroundIndigenous communities residing in the Darjeeling Himalayan region and its adjacent hilly areas have a deeply rooted cultural tradition of consuming a diverse range of vegetable and milk-based fermented products, believed to confer various health advantages. With this traditional knowledge, lactic acid bacteria (LAB) were isolated from popular fermented foods such as Chhurpi (derived from Bos grunniens milk), Gundruk (made from Brassica juncea leaves), Sinki (derived from Raphanus sativus taproots), and Kinema (produced from Glycine max beans). This study aimed to investigate the probiotic properties of the prevalent LABs, including aggregation properties, bile salt hydrolase activities, survival under gastro-inhibitory conditions, safety evaluations, and their potential health-promoting attributes, with a specific focus on inhibiting α-amylase and α-glucosidase enzymes.ResultsFive of the LAB isolates demonstrated notable viability rates exceeding 85% when exposed to gastro-inhibitory challenges. Based on 16S rRNA gene sequencing, these isolates were identified as Pediococcus pentosaceus (isolate GAD), Lactobacillus plantarum (isolates KAD and CAD), Lactobacillus brevis (isolate SAD), and Lactiplantibacillus plantarum (isolate CMD). These LAB isolates exhibited versatile carbon source utilization, significant auto- and co-aggregation, and bile salt hydrolase (BSH) properties. Auto-aggregation capacity notably increased over time, ranging from 30 to 150 min, with percentage increments from 4.83 ± 1.92% to 67.60 ± 5.93%. L. brevis SAD displayed the highest co-aggregation increment (%) against Staphylococcus aureus, while L. plantarum KAD demonstrated potent antimicrobial activity. In vitro analyses postulated potential health benefits related to antidiabetic properties, particularly inhibiting α-amylase and α-glucosidase enzymes. L. brevis SAD exhibited the highest α-glucosidase inhibitory activity, while L. plantarum KAD displayed the most potent α-amylase inhibitory activity. Comprehensive safety assessments, including antibiotic susceptibility profiling, hemolytic activity evaluation, and in vivo acute toxicity studies, confirmed the suitability of these LAB isolates for human consumption.ConclusionsThe isolates show promising probiotic characteristics and significant potential in addressing metabolic health. These results carry substantial scientific implications, suggesting the pharmaceutical-based applications of these traditional fermented foods. Further in vivo investigation is recommended to fully elucidate and exploit the health benefits of these LAB isolates, opening avenues for potential therapeutic interventions and the development of functional foods.

Research has been conducted to develop an effective method for preserving the probiotic properties of Lactococcus lactis ssp. lactis strain 194, isolated from milk in Buryatia (Russia). The effect of 10 protective media added before freezing was compared: NaCl, skim milk, 5% sucrose+1% gelatin and smectite at concentrations of 5, 10 and 5% as a cryoprotectant. Physiological and probiotic properties: survival rate, rate of milk clot formation, lactic acid formation and antimicrobial activity were studied in lyophilized bacteria immediately after reconstitution, as well as in a number of passages after long-term storage. Optimal concentrations of smectite were selected using a toxicological method, and particle sizes and the morphology of the surface of lactococcal cells in the presence of a cryoprotector were determined using electron microscopy. It was found that the most effective method of preserving the probiotic properties of lactic acid bacteria is lyophilization with the addition of V% smectite, which ensured high cell survival in the first passage and preserved the number of bacteria and their probiotic potential after long-term storage.

Isolation, characterization of Weissella confusa and Lactococcus lactis from different milk sources and determination of probiotic features.

This study aimed to investigate the probiotic properties of Lactic Acid Bacteria (LAB) isolates derived from various milk sources. These isolates identified based on their morphological characteristics and 16S rRNA gene sequencing. Four strains of Lactococcus lactis and two strains of Weissella confusa were identified with over 96% 16S rRNA gene similarity according to the NCBI-BLAST results. The survival of the isolates was determined in low pH, pepsin, bile salts, and pancreatin, and their adhesion ability was assessed by in vitro cell adhesion assay, hydrophobicity, auto- and co-aggregation, and safety criteria were determined by hemolytic, gelatinase activities, and DNAse production ability tests. The results showed that the LAB isolates had different levels of resistance to various stress factors. L. lactis subsp. cremoris MH31 showed the highest resistance to bile salt, while the highest pH resistance was observed in L. lactis MH31 at pH 3.0. All the isolates survived in pepsin exposure at pH 3.0 for 3h. The auto-aggregation test results showed that all strains exhibited auto-aggregation ranging from 84.9 to 91.4%. Co-aggregation percentage ranged from 19 - 54% and 17 - 57% against Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 29213, respectively. The hydrophobicity capacity of the LAB isolated ranged from 35-61%. These isolates showed different adhesion abilities to Caco-2 cells (81.5% to 92.6%). None of the isolates exhibited DNase, gelatinase and hemolytic activity (γ-hemolysis). All results indicate that these LAB strains have the potential to be used as probiotics.

Some enzymatic properties of lactic acid bacteria isolated from dairy products

This article presents data on the study of physiological and biochemical properties, antagonistic and enzymatic activity of lactic acid bacteria isolated from dairy products. 9 types of lactic acid bacteria were studied: Lactobacillus bulgaricus GM – 08, Lactobacillus bulgaricus KZh – 01, actobacillus bulgaricus GS – 03, Lactococcus cremoris – 6, Lactococcus cremoris – 17, Lactococcus cremoris – 26, Lactococcus lactis – 1, Lactococcus lactis – 15, Lactococcus lactis – 23. These strains were found to have resistance to 2% and 4%-vertical NaCl concentrations, bile and phenol. In addition, the antagonistic activity of Gram-positive and Gram-negative microorganisms in relation to test cultures of Staphylococcus aureus, Salmonella dublin,Escherichia coli, Bacillus subtilis, Sarcina flava was studied. All studied lactic acid bacteria showed activity in Test cultures with different inhibition zones. The Lactococcus lactis – 23 strains showed high activity for all cultures, with an inhibition zone of 17-25 mm. Further, 5 strains were selected from these strains and their aroma-forming properties, the formation of diacetyl and ammonia from arginine, hemolytic and lecithinase activity were studied. Compositions were compiled from these strains to make yeast. The compatibility of strains of lactic acid bacteria was checked with each strain individually and with the duration of milk clotting according to organoleptic indicators compared to the duration of milk clotting. Thus, the most active clot formation was obtained by the Lactococcus lactis – 23 strain of the selected combinations.

Investigating the antidiabetic efficacy of dairy-derived Lacticaseibacillus paracasei probiotic strains: modulating α-amylase and α-glucosidase enzyme functions.

The current study aims to evaluate and characterize the probiotic andantidiabetic properties of lactic acid bacteria (LAB) obtained from milk and other dairy-based products. The strains were tested physiologically, biochemically, and molecularly. Based on biochemical tests and 16S rRNA gene amplification and sequencing, all three isolates RAMULAB18, RAMULAB19, and RAMULAB53 were identified as Lacticaseibacillus paracasei with homology similarity of more than 98%. The inhibitory potential of each isolate against carbohydrate hydrolysis enzymes (α-amylase and α-glucosidase) was assessed using three different preparations of RAMULAB (RL) isolates: the supernatant (RL-CS), intact cells (RL-IC), and cell-free extraction (RL-CE). Additionally, the isolate was evaluated for its antioxidant activity against free radicals (DPPH and ABTS). The strain's RL-CS, RL-CE, and RL-IC inhibited α-amylase (17.25 to 55.42%), α-glucosidase (15.08-59.55%), DPPH (56.42-87.45%), and ABTS (46.35-78.45%) enzymes differently. With the highest survival rate (>98%) toward tolerance to gastrointestinal conditions, hydrophobicity (>42.18%), aggregation (>74.21%), as well as attachment to an individual's colorectal cancer cell line (HT-29) (>64.98%), human buccal and chicken crop epithelial cells, all three isolates exhibited extensive results. All three isolates exhibited high resistance toward antibiotics (methicillin, kanamycin, cefixime, and vancomycin), and other assays such as antibacterial, DNase, hemolytic, and gelatinase were performed for safety assessment. Results suggest that the LAB described are valuable candidates for their significant health benefits and that they can also be utilized as a beginning or bio-preservative tradition in the food, agriculture, and pharmaceutical sectors. The LAB isolates are excellent in vitro probiotic applicants and yet additional in vivo testing is required.

Development of a low pollution medium for the cultivation of lactic acid bacteria

Protein rich culture media are employed in the production of lactic acid bacteria (LAB); however, production costs are high. In this work media formulation and evaluation for LAB production were conducted considering physiological properties of lactic acid bacteria. Consumption efficiency (E), yield production (Y) and specific substrate consumption rate (qS) values as response variables were used. Four culture media were used: (1) Man Rogosa Sharp (MRS); (2) cabbage liquor (MC); (3) a new balanced culture medium (MX); and (4) MX supplemented with cabbage liquor (MXC). The culture media were evaluated using two strains: Lactobacillus acidophilus ATCC 4356 and Lactiplantibacillus plantarum ATCC 10241. The EGLU for L. plantarum was 100 % in the three media and YX/S value was 0.02 ± 0.003 in MRS and MX, while YLAC/S was 0.57 ± 0.03 in MRS and 0.51 ± 0.02 in MX. In MXC, the value obtained for YX/S was 0.07 ± 0.002 while YLAC/S was 0.47 ± 0.04. Specific glucose consumption and lactate formation rates for L. plantarum in MRS and MX media did not show significant differences. These results suggest that MX and MXC can be used for efficient production of the LAB at low cost.

Investigation of Lactobacillus spp. bacteria in infants consuming breast milk and formula and determination of some probiotic characters

Objectives: It was aimed to investigate some probiotic properties of Lactic Acid Bacteria (LAB) isolated from stool samples taken from 45 healthy 0-4 month old babies, who did not take antibiotics or probiotic supplements in the last 3 months, and who did not have any health problems. Methods: Six different species were obtained from 21 isolates selected by the method of Mass Spectrometry (MALDI-TOF MS). Results: The most common strain was Lactobacillus rhamnosus with a rate of 59%, followed by Lactobacillus paracasei with a rate of 13.6%. Vancomycin, tetracycline, gentamicin, netilmicin, tobramycin, penicillin, ampicillin, teicoplanin and amikacin antibiotics were used to evaluate the antimicrobial activities of the strains. In our study, while all strains were resistant to the antibiotic amikacin, they were sensitive to tetracycline, penicillin, gentamicin, netilmicin, teicoplanin, vancomycin, ampicillin and tobramycin antibiotics. In the evaluation of the antagonistic activities of LAB, 6 different pathogens (Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 6538, Bacillus subtilis ATCC 6633, S. aureus ATCC 25923, Candida albicans ATCC 10231, Listeria monocytogenes ATCC 19111) were used and it was determined that the strains showed antimicrobial effects on all pathogenic microorganisms. Cholesterol assimilation abilities, T21 and T22 strains achieved the highest cholesterol assimilation rate of 39.1%. Conclusions: It is thought that most of the isolated strains have probiotic potential, and especially Lactobacillus gasseri T21 and L. paracasei T22 strains may be probiotic strains that can be used in the production of preparations alone or together with other Lactobacillus strains.

Assessment of probiotic properties of lactic acid bacteria isolated from an artisanal Colombian cheese

Lactic Acid Bacteria play an important role in the milk fermentation processes of traditional cheeses and have become an important target for the development of novel cheese cultures because of their ability to confer health benefits. This study aimed to evaluate the probiotic potential of 12 Lactic Acid Bacteria (LAB) strains previously isolated and molecularly identified from an artisanal Colombian Double-Cream Cheese. Probiotic properties, including safety (hemolysis and sensibility to antibiotics), pH and bile salt tolerance, auto-aggregation, cell surface hydrophobicity, antibacterial activity, and exopolysaccharide production, were examined. None of the strains were hemolytic, and Pediococcus (16, 18) and Lactobacillus (28, 29) were found to be sensitive to all antibiotics. Moreover, all the strains tolerated pH (3.0, 6.5 and 8.0) and bile salt conditions (0.3, 0.6 and 1.0 % w/v). Pediococcus pentosaceus (16), Leuconostoc citreum (17), Pediococcus acidilactici (18), Enterococcus faecium (21,22), Enterococcus faecalis (24) and Limosilactobacillus fermentum (29) exhibited medium autoaggregation and affinity to chloroform. Six of the strains exhibited a ropy exopolysaccharide phenotype. Antibacterial activity against foodborne pathogens, Salmonella Typhimurium ATCC 14028, Listeria monocytogenes ATCC 19111, Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 25923, was found to be strain dependent, with the strains 16, 18, 21, 26, 28 and 29 presenting a higher inhibition (>4 mm) against all of them. According to Principal Component Analysis, P. pentosaceus (16), Leu. mesenteroides (26), L. casei (28), L. fermentum (29), and E. faecium (21) showed strong probiotic properties. Our findings suggest that five strains out of the 12 sampled strains are potential probiotics that could be used in the processing of traditional dairy products on an industrial scale to improve their quality.

Antimicrobial and Mycotoxin Reducing Properties of Lactic Acid Bacteria and Their Influence on Blood and Feces Parameters of Newborn Calves.

The aim of this study was to evaluate the influence of in acid whey (AW) multiplied Lactiplantibacillus plantarum LUHS135 (L.pl135), Lacticaseibacillus paracasei LUHS244 (L.pc244), and their biomass combination on newborn calves' feces and blood parameters. Additionally, the antimicrobial and mycotoxin-reducing properties and the resistance to antibiotics of the tested lactic acid bacteria (LAB) strains were analyzed. In order to ensure effective biomass growth in AW, technological parameters for the supplement preparation were selected. Control calves were fed with a standard milk replacer (SMR) and treated groups (from the 2nd day of life until the 14th day) were supplemented with 50 mL of AWL.pl135, AWL.pc244, and AWL.pl135×L.pc244 (25 mL AWL.pl135 + 25 mL AWL.pc244) in addition to SMR. It was established that L.pl135 and L.pc244 possess broad antimicrobial activities, are non-resistant to the tested antibiotics, and reduce mycotoxin concentrations in vitro. The optimal duration established for biomass growth was 48 h (LAB count higher than 7.00 log10 CFU mL-1 was found after 48 h of AW fermentation). It was established that additional feeding of newborn calves with AWL.pl135, AWL.pc244, and AWL.pl135×L.pc244 increased lactobacilli (on average by 7.4%), and AWL.pl135 and AWL.pc244 reduced the numbers of Enterobacteriaceae in calves' feces. The tested supplements also reduced the lactate concentration (on average, by 42.5%) in calves' blood. Finally, the tested supplements had a positive influence on certain health parameters of newborn calves; however, further research is needed to validate the mechanisms of the beneficial effects.

Biodiversity and biotechnological properties of lactic acid bacteria isolated from traditional Moroccan sourdoughs.

The present study aimed at characterizing lactic acid bacteria (LAB) strains isolated from traditional sourdoughs collected in different regions of Morocco. Isolated strains were firstly identified using Gram staining and catalase reaction test. Presumptive LAB strains were then checked for various phenotypical properties including growth at 45°C, resistance to NaCl, enzyme production, acidification capacity, diacetyl and exopolysaccharide (EPS) production, and antifungal activity. Finally, selected LAB strains were identified using 16S rDNA sequencing. Results showed that 32.1% of the isolates were thermophilic (45°C) and 83.9% were resistant to NaCl (6.5%). Moreover, 51.7 and 37.5% were able to produce diacetyl and EPS, respectively. Regarding enzyme production, 55.3 and 7.1% of the isolates showed lipolytic and proteolytic activities, respectively. Low pH values (3.37-3.76) were obtained after 24h of incubation of LAB strains in de Man, Rogosa and Sharpe (MRS) broth. Antifungal activity test against Aspergillus flavus, Aspergillus niger and Penicillium spp. showed an inhibition rate up to 50%. Bacterial DNA sequencing showed that LAB isolates belong to seven species, chiefly Levilactobacillus brevis, Lentilactobacillus parabuchneri, Lactiplantibacillus plantarum, Pediococcus pentosaceus, Enterococcus hirae, Bifidobacterium pseudocatenulatum, and Companilactobacillus paralimentarius. These findings, for the first time in Moroccan sourdoughs, indicate that the isolated LAB strains have good multifunctional properties and could be suitable as good starters for sourdough bread production under controlled conditions.

Antimicrobial properties of lactic acid bacteria from fermented palm sap against E. coli and S. aureus growth

Palm sap produced from tapping can be a growing place for lactic acid bacteria (LAB). Palm sap has a lot of carbohydrate content, such as sucrose, fructose, and glucose, which can be a source of nutrition for the growth of microorganisms such as LAB). The result of LAB metabolism that is formed can be in the form of organic acid acids that can act as antimicrobial compounds. LAB have been isolated from fermented palm sap. This study aimed to determine the antibacterial properties of LAB from the fermentation of palm sap against E. coli and S. aureus using the well-agar diffusion method and the paper disc diffusion method. The study showed that LAB isolates (Lactobacillus A1, A6, B12, C1, and C4) and cell-free supernatants have antibacterial activity against E. coli and S. aureus. LAB antibacterial properties using cells with a clear zone diameter against E. coli ranges from 8.03 to 0.58 mm, and S. aureus inhibition zone diameter ranges from 9.13 to 11.16 mm. The antimicrobial activity of LAB using cell-free supernatants against E. coli ranges from 4.59 to 5.01 mm, and the diameter of the inhibitory zone against S. aureus ranges from 4.34 to 5.79 mm. From these results, LAB isolated from fermented palm sap can potentially be probiotics.

Bacteriocin and Antioxidant Production, a Beneficial Properties of Lactic Acid Bacteria Isolated from Fermented Vegetables of Northwest Bulgaria.

Lactiplantibacillus plantarum ST01BG, ST07BG, ST10BG, and ST15BG; Latilactobacillus curvatus ST02BG; Lacticaseibacillus paracasei ST04BG; Pediococcus pentosaceus ST05BG; Leuconostoc mesenteroides ST06BG; and Enterococcus faecium ST11BG were isolated from home-made fermented vegetables from Northwest Bulgaria and identified by biochemical, physiological, and biomolecular analyses, including partial 16S rRNA sequencing. The strains were designated as bacteriocin producers and the expressed antimicrobials partially characterized with a focus on their proteinaceous nature, stability to different pH and temperatures. The bacteriocins were effective in inhibiting different strains of Listeria spp., Enterococcus spp. (including vancomycin resistant enterococci) and Staphylococcus spp. These strains can be considered safe, based on the evaluation of hemolytic activity, production of biogenic amines, mucin degradation, antibiotic susceptibility/resistance, and gelatinase enzyme production. Moreover, the strains can be considered potentially beneficial based on their stability and survival under simulated gastrointestinal tract conditions (stomach and duodenum), the production of diacetyl, and specific levels of hydrophobicity. Special attention was given to antioxidant properties (DPPH radical, hydroxyl radical, superoxide anion radical scavenging activity, Fe+2 ion chelating activity, and anti-lipid peroxidation) of the strains. Antioxidant properties were found to be strain specific. The beneficial attributes (antimicrobial and antioxidant) of these cultures to fermented food products may enable the reduction of chemical additives in line with consumers' demand for more natural and chemical-free food commodities.

Probiotic Properties of Lactic Acid Bacteria Newly Isolated from Algerian Raw Cow's Milk.

This study aims to screen new LAB from Algerian cow's milk to assess their probiotic properties. Molecular identification and MALDI-TOF mass spectrometry methods were used to identify the LAB isolates. The probiotic potential of isolates was determined with in vitro tests of survival to gastrointestinal conditions (pH 2, 0.3% pepsin, 0.5% bile salts, 0.1% trypsin, and 0.1% pancreatic amylase) and antimicrobial and antioxidant activities. Eight isolates were identified as Lactiplantibacillus plantarum (100%) and one isolate as Lacticaseibacillus rhamnosus (95.75%). The MALDI-TOF MS analysis of the isolates confirms that the strains belong to the group of lactobacilli bacteria, particularly Lactiplantibacillus plantarum. The high survival rate reflects a good strain tolerance to the in vitro host simulated gastrointestinal conditions. All bacteria exhibit an antibacterial activity strain with inhibition zone diameters ranging from 4.9 mm against Aspergillus niger ATCC 106404 to 17.47 mm against Candida albicans ATCC 10231. The antioxidant activity with the highest DPPH scavenging activity (92.15%) was obtained with the LbN09 strain. In light of these results, some of the strains isolated from raw milk of the local Algerian breed cows show promising probiotic properties, giving them a possible use in preserving food from microbial spoilage and oxidation during storage.
