Abstract PO2-24-08: GD3 Synthase is a Master Regulator of Wild-Type p53–Mediated Apoptosis and Mutant p53–Mediated Tumorigenesis

Abstract The tumor suppressor protein p53 is essential for maintaining genomic stability and regulating cellular responses to stress. TP53 mutations are common in breast cancer (BC) and other malignancies. We previously showed that ganglioside GD2 identifies BC stem-like cells (BCSCs) and that GD3 synthase (GD3S/ST8SIA1) is upregulated in breast tumors with TP53 mutations. Here, we demonstrate the direct involvement of p53 in the transcriptional regulation of GD3S in the GD2 biosynthesis pathway. Our results uncover a novel role of GD3S in inhibiting p53-mediated apoptosis and promoting mutant p53–induced tumor initiation. To investigate the p53-mediated regulation of GD3S expression, we used the TCGA and METABRIC datasets and examined GD3S mRNA expression in breast tumors with different p53 mutation status. GD3S expression was significantly upregulated in BC patients with hotspot (HS) p53 mutations compared to those with wild-type (WT) p53 or non-HS p53 mutations (P< 0.0001). Immuno-histochemical analysis of GD3S and p53 in FFPE primary tumor tissues from BC patients (n=84) with varying p53 expression status demonstrated that patients with HS p53 mutations had higher histological scores for GD3S than did patients with WT p53 (P< 0.0001) or non-HS p53 mutations (P=0.009). GD3S and p53 expression were significantly positively correlated in patients with HS p53 mutations (r=0.735; P=0.006) but not in patients with WT p53 (r=–0.09) or non-HS p53 mutations (r=0.14). Consistent with these patient data, BC cells harboring HS p53 mutations had significantly upregulated and positively correlated expressions of GD3S and p53 (n=8; r=0.85). Moreover, compared to those with WT p53, the cells with HS p53 mutations had substantially greater populations of GD2+ BCSCs (P=0.02) and GD3+ BCSCs (P=0.01). Subsequently, we investigated the regulatory role of p53 in the GD2 biosynthetic pathway by stabilizing WT p53 expression by nutlin-3a treatment in 4 BC cell lines and suppressing the expression of HS p53 mutants by p53 knockdown in their respective cell lines (n=4). In cell lines with WT p53, nutlin-3a significantly decreased GD3S expression and GD2 levels (P< 0.05 and P< 0.0001, respectively), suggesting that WT p53 inhibits GD3S expression. In cell lines with p53 mutations, p53 knockdown significantly inhibited GD3S expression. In both cells with WT p53 (MCF7 and ZR751) and cells with HS p53 mutations (Hs578T and BT549), the stable overexpression of GD3S effectively suppressed apoptosis even when WT p53 was stabilized or HS p53 mutant expression was depleted. Consistent with these results, in mice bearing MCF7 cell–derived xenografts, the overexpression of GD3S counteracted the effect of stabilized WT p53–mediated apoptosis and facilitated tumor growth (P< 0.0001). The results of a promoter-luciferase reporter assay demonstrated that nutlin-3a reduced GD3S promoter activity in a dose-dependent manner in MCF7 and ZR751 cells (P< 0.0001), whereas doxycycline (1µM) significantly reduced GD3S promoter activity in Hs578T and BT549 cells with inducible p53 knockdown (P< 0.001). ChIP-qPCR analysis confirmed specific enrichment of WT p53 at the 300-bp upstream regions and additional enrichment of HS p53 mutants at the downstream regions of the GD3S promoter, indicating that WT p53 and HS p53 mutations have distinct promoter regulation patterns. In summary, our study reveals that GD3S has a previously unknown anti-apoptotic function in BC, in that it helps attenuate p53-mediated apoptosis while activating tumor-promoting pathways that operate independently of p53. Abstract 1476: OSBPL3 facilitates endoplasmic reticulum stress and stemness in triple negative breast cancer

Abstract Background: Triple negative breast cancer (TNBC) is a type of aggressive breast cancer with a poor survival. Endoplasmic reticulum (ER) stress, induced by exposed to intrinsic (e.g. oncogenes) and external factors (e.g. chemotherapies), is an important factor in both tumor progression and responses to chemotherapies. Recent studies have demonstrated that the progression of ER stress may result in the emergence of cancer stem cells (CSC), which also arise as a consequence of metastasis. However, the mechanism of targeting ER stress signaling in cancer stem cells remains to be investigated. Aim: (i) To investigate the mechanistic role of Oxysterol binding protein-like 3 (OSBPL3) in TNBC and its links with breast cancer mortality, particularly caused by chemotherapeutic resistance and metastasis; and (ii) to test a hypothesis that OSBPL3 facilitates the abnormal activation of ER stress and cancer stemness, result in the resistance to chemotherapy and metastasis in TNBC. Methods: The expression level of OSBPL3 in TNBC was determined by immunohistochemical staining and the mRNA expression profiles from the TCGA database. Roles of OSBPL3 in cancer cell growth, metastasis, stemness and activation of ER stress were determined by molecular and cell biology methods. Immunoprecipitation was used to examine the binding of proteins and chromatin immunoprecipitation (ChIP), promoter luciferase reporter assay was included for detecting the transcription of OSBPL3 in TNBC. TNBC xenograft nude mice were used to study the inhibition of tumor development. Results: OSBPL3 is a novel TNBC target, which is overexpressed in TNBC tissues and significantly associated with clinical pathology of advanced TNM stage and poor prognosis of tumor patients. Mechanistically, Increased OSBPL3 induced ER stress and ER stress agonist (tunicamycin) treatment results in the translocation of XBP1, an ER stress sensor, into the nucleus to induce OSBPL3 expression through direct binding to the OSBPL3 promoter. In addition, OSBPL3 facilitates the stemness induced by ER stress via phosphorylating GSK3β and transporting accumulated β-catenin to the nucleus to promote the expression of stemness-related transcriptional factors, ultimately resulting in tumor metastasis and resistance to chemotherapy. Furthermore, OSBPL3-specific siRNAs encapsulated by jetPEI nanocarriers prominently inhibit tumor growth and epirubicin-induced resistance in vivo. Conclusion: We identified an important role of OSBPL3-ER stress-XBP1 feedback loop in the stemness and resistance to chemotherapy, representing a future biomarker and therapeutic target of TNBC. Abstract 133: GMDS is a key driver of MYCN-amplified neuroblastoma core fucosylation and tumorigenesis

Abstract Introduction: MYCN-amplification (MYCN-amp) is a genetic hallmark of ~40% of high-risk neuroblastomas (NBs). N-MYC is an oncogenic transcription factor and master regulator of metabolism. Glycosylation is a major post-translational modification that is critical for cancer progression, spread, and immune evasion. How genetic signatures such as MYCN-amp alter glycosylation profiles is unknown. Herein, we utilized matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) to define the N-linked glycome of MYCN-amp NBs in situ identifying enrichment of core fucosylated glycans. GDP-mannose 4,6-dehydratase (GMDS) is the first enzyme responsible for de novo GDP-fucose synthesis. We hypothesized that GMDS is a key mediator of NB core fucosylation and tumorigenesis. Methods: MALDI-MSI was performed on five MYCN-amp primary NBs and six MYCN non-amplified primary NBs using a timsTOF fleX mass spectrometry imager. Significance analysis of metabolites (SAM) was performed with a q-value <0.05 used to define significance. Kaplan-Meier analysis was performed to determine whether GMDS expression correlated with overall survival using the R2 Platform. GMDS blockade was performed using genetic knockdown and chemical inhibition with 2-fluorofucose (2-FF). Core fucosylated glycan abundance was measured by western blotting, ELISA, and flow cytometry using Aleuria aurantia lectin. MYCN tet-inducible NB cells were used to investigate the interplay of N-MYC and GMDS expression. Chromatin immunoprecipitation (ChIP) and luciferase promoter assay were performed to investigate the direct interplay of N-MYC and the GMDS promoter. Subcutaneous tumor formation was our measure of in vivo tumorigenesis. Results: MALDI-MSI revealed increased expression of eleven N-linked glycans within MYCN-amp NBs, and six of these glycans feature core fucosylation. High GMDS expression is associated with poor overall patient survival (p=3.5 × 10−22) and correlates with MYCN-amp in human NB tumors (p=7.4 × 10−27). Genetic and chemical inhibition of GMDS blocks core fucosylated glycan abundance and secretion, while impeding NB cell growth and adhesion in vitro. Induction and blockade of N-MYC directly correlates with GMDS expression. ChIP and promoter luciferase reporter assay demonstrates direct binding and activation of the GMDS promoter by N-MYC. Importantly, GMDS knockdown blocks NB tumor formation (p<0.01), and 2-FF impedes established tumor progression (p<0.05). Conclusions: This multi-institutional study utilized MALDI-MSI to reveal that core fucosylated glycans are enriched in MYCN-amp NBs. GMDS is a key regulator of NB core fucosylation that is driven by N-MYC. GMDS blockade impairs NB growth in vitro and in vivo. These critical findings identify de novo GDP-fucose production as a metabolic vulnerability that may be exploited in designing novel treatment paradigms for high-risk NBs. Pioneering 4,11-Dioxo-4,11-dihydro-1H-anthra[2,3-d]imidazol-3-ium Compounds as Promising Survivin Inhibitors by Targeting ILF3/NF110 for Cancer Therapy.

Survivin is a novel attractive target for cancer therapy; however, it is considered undruggable because it lacks enzymatic activities. Herein, we describe our efforts toward the discovery of a novel series of 4,11-dioxo-4,11-dihydro-1H-anthra[2,3-d]imidazol-3-ium derivatives as survivin inhibitors by targeting ILF3/NF110. Intensive structural modifications led us to identify a lead compound AQIM-I, which remarkably inhibited nonsmall cell lung cancer cells A549 with an IC50 value of 9 nM and solid tumor cell proliferation with more than 700-fold selectivity against human normal cells. Further biological studies revealed that compound AQIM-I significantly inhibited survivin expression and colony formation and induced ROS production, apoptosis, cell cycle arrest, DNA damage, and autophagy. Furthermore, the promoter-luciferase reporter assay showed that AQIM-I attenuated the survivin promoter activity enhanced by the overexpression of ILF3/NF110 in a concentration-dependent manner, and specific binding (KD = 163 nM) of AQIM-I to ILF3/NF110 was detected by surface plasmon resonance.

SNHG25 promotes colorectal cancer metastasis by regulating MMP2.

LncRNA has been shown to play an important role in tumors, but the functions of most lncRNAs in colorectal cancer is not clear. By analyzing the transcriptome data of tumor tissues and adjacent tissues, we identified the lncRNA profiles that were abnormally expressed in colorectal cancer and selected the abnormally highly expressed lncRNA SNHG25 for further study. The functional assays showed that after knocking down SNHG25, the metastatic ability of colorectal cancer cells was significantly reduced. Western blot and immunofluorescence assays showed that inhibiting SNHG25 would affect the expression of Vimentin and E-Cadherin. In terms of mechanism, the results of RNA pull down assays, RNA immunoprecipitation (RIP) assays and dual luciferase reporter assays showed that SNHG25 could promote MMP2 expression by adsorbing miR-296-3p. In addition, chromatin immunoprecipitation (ChIP) assays and promoter luciferase reporter assays revealed that PAX5 could activate the transcription of SNHG25 in colorectal cancer cells. Our study proved that SNHG25 acts a pro-metastasis role in colorectal cancer, enriching the theory of the functions of lncRNA in colorectal cancer.

ETV2 and VEZF1 interaction and regulation of the hematoendothelial lineage during embryogenesis.

Ets variant 2 (Etv2), a member of the Ets factor family, has an essential role in the formation of endothelial and hematopoietic cell lineages during embryonic development. The functional role of ETS transcription factors is, in part, dependent on the interacting proteins. There are relatively few studies exploring the coordinated interplay between ETV2 and its interacting proteins that regulate mesodermal lineage determination. In order to identify novel ETV2 interacting partners, a yeast two-hybrid analysis was performed and the C2H2 zinc finger transcription factor VEZF1 (vascular endothelial zinc finger 1) was identified as a binding factor, which was specifically expressed within the endothelium during vascular development. To confirm this interaction, co-immunoprecipitation and GST pull down assays demonstrated the direct interaction between ETV2 and VEZF1. During embryoid body differentiation, Etv2 achieved its peak expression at day 3.0 followed by rapid downregulation, on the other hand Vezf1 expression increased through day 6 of EB differentiation. We have previously shown that ETV2 potently activated Flt1 gene transcription. Using a Flt1 promoter-luciferase reporter assay, we demonstrated that VEZF1 co-activated the Flt1 promoter. Electrophoretic mobility shift assay and Chromatin immunoprecipitation established VEZF1 binding to the Flt1 promoter. Vezf1 knockout embryonic stem cells had downregulation of hematoendothelial marker genes when undergoing embryoid body mediated mesodermal differentiation whereas overexpression of VEZF1 induced the expression of hematoendothelial genes during differentiation. These current studies provide insight into the co-regulation of the hemato-endothelial lineage development via a co-operative interaction between ETV2 and VEZF1.

Trichostatin D as a Novel KLF2 Activator Attenuates TNFα-Induced Endothelial Inflammation.

Krüppel-like factor 2 (KLF2) is an atherosclerotic protective transcription factor that maintains endothelial cell homeostasis through its anti-inflammatory, anti-oxidant, and antithrombotic properties. The aim of this study was to discover KLF2 activators from microbial secondary metabolites and explore their potential molecular mechanisms. By using a high-throughput screening model based on a KLF2 promoter luciferase reporter assay, column chromatography, electrospray ionization mass spectrometry (ESI-MS), and nuclear magnetic resonance (NMR) spectra, trichostatin D (TSD) was isolated from the rice fermentation of Streptomyces sp. CPCC203909 and identified as a novel KLF2 activator. Real-time-quantitative polymerase chain reaction (RT-qPCR) results showed that TSD upregulated the mRNA level of KLF2 in endothelial cells. Functional assays showed that TSD attenuated monocyte adhesion to endothelial cells, decreased vascular cell adhesion protein 1 (VCAM-1) and intercellular adhesion molecule 1 (ICAM-1) expression, and exhibited an anti-inflammatory effect in tumor necrosis factor alpha (TNFα)-induced endothelial cells. We further demonstrated through siRNA and western blot assays that the effects of TSD on monocyte adhesion and inflammation in endothelial cells were partly dependent on upregulating KLF2 expression and then inhibiting the NOD-like receptor protein 3 (NLRP3)/Caspase-1/interleukin-1beta (IL-1β) signaling pathway. Furthermore, histone deacetylase (HDAC) overexpression and molecular docking analysis results showed that TSD upregulated KLF2 expression by inhibiting HDAC 4, 5, and 7 activities. Taken together, TSD was isolated from the fermentation of Streptomyces sp. CPCC203909 and first reported as a potential activator of KLF2 in this study. Furthermore, TSD upregulated KLF2 expression by inhibiting HDAC 4, 5, and 7 and attenuated endothelial inflammation via regulation of the KLF2/NLRP3/Caspase-1/IL-1β signaling pathway.

FOSL2 deficiency delays nonalcoholic steatohepatitis progression by regulating LY6D-mediated NLRP3 activation.

Lymphocyte antigen 6 family member D (LY6D) was enhanced specifically in senescent cells, while its effects on pyroptosis, a programmed cell death, remains unknown. The goal of this study was to assess the role of LY6D in the mediation of pyroptosis during nonalcoholic steatohepatitis (NASH). After screening out LY6D as a specific liver fibrosis-associated gene using the GSE55747 dataset from the GEO database, we established a NASH mouse model using methionine and choline deficient-diet feeding and an in vitro model using lipopolysaccharide (LPS)-treated hepatocytes. LY6D was overexpressed in NASH livers as well as in LPS-treated hepatocytes. Silencing of LY6D inhibited NASH-associated hepatocyte pyroptosis. With the aid of bioinformatics analysis, promoter-luciferase reporter and ChIP-qPCR assays, we identified FOSL2 as an upstream transcription factor of LY6D. FOSL2, which was highly expressed in NASH, promoted LY6D transcription by binding to the promoter of LY6D. Depletion of FOSL2 significantly inhibited NASH-associated hepatocyte pyroptosis, which was significantly reversed after overexpression of LY6D. Moreover, the promotion of hepatocyte pyroptosis by the FOSL2/LY6D axis was significantly attenuated by specific inhibition of NLRP3. These findings suggesting that FOSL2/LY6D axis may be a key molecular axis and a potential target for NASH therapeutics.

The lncARSR/PTEN/Akt/nuclear factor-kappa B feedback regulatory loop contributes to doxorubicin resistance in hepatocellular carcinoma.

Chemoresistance is a major obstacle to hepatocellular carcinoma (HCC) chemotherapy. Our previous study found that long noncoding RNA lncARSR (lncRNA Activated in RCC with Sunitinib Resistance) activated Akt signaling via repressing phosphatase and tensin homolog (PTEN) during doxorubicin resistance in HCC. The purpose of this study is to further explore lncARSR-mediated mechanisms and roles during doxorubicin resistance in HCC. The expression of lncARSR was detected by real-time quantitative polymerase chain reaction (qPCR). Nuclear factor-kappa B (NF-κB) activity was detected by NF-κB luciferase reporter assays, western blot, and NF-κB transcription factor assays. The effects of NF-κB on lncARSR were detected by chromatin immunoprecipitation assay, promoter luciferase reporter assay, and real-time qPCR. The effects of lncARSR/Akt/NF-κB on doxorubicin resistance were detected by Cell Counting Kit-8 assay, capsase-3 activity assay, and terminal deoxynucleotidyl transferase-mediated dUTP nick end labelingassay. lncARSR activated NF-κB signaling through activation of Akt. NF-κB transactivated lncARSR through directly binding lncARSR promoter and increasing lncARSR promoter activity. Akt transactivated lncARSR via activating NF-κB signaling. Thus, lncARSR, Akt, and NF-κB formed a positive feedback regulatory loop in HCC. Through this feedback loop, lncARSR auto-regulated its transcription. Drug sensitivity assays showed that the lncARSR/Akt/NF-κB feedback regulatory loop promoted doxorubicin resistance in HCC. These findings identified the lncARSR/Akt/NF-κB feedback regulatory loop in HCC, which represent potential therapeutic targets for improving doxorubicin sensitivity in HCC.

KLF8 is activated by TGF-β1 via Smad2 and contributes to ovarian cancer progression.

Krüppel-like factor 8 (KLF8) is a transcription factor expressed abnormally in various cancer types and promotes oncogenic transformation. However, the role of KLF8 in ovarian cancer (OC) progression remains unclear. This study reports that transforming growth factor-β1(TGF-β1)/Smad2/KLF8 axis regulates epithelial-mesenchymal transition (EMT) and contributes to OC progression. We analyzed the KLF8 expression in OC cells and tissues, wherein a significant overexpression of KLF8 was observed. Increased KLF8 expressions were correlated with higher cell proliferation, EMT, migration, and invasion and conferred poor clinical outcomes in OC patients. Overexpressed KLF8 increases F-actin polymerization and induces cytoskeleton remodeling of OC cells. Furthermore, a dissection of the molecular mechanism defined that TGF-β1 triggers KLF8 through the Smad2 pathway and regulates EMT. Pharmacological and genetic inhibition of Smad2 followed by TGF-β1 treatment failed to activate KLF8 expression and induction of EMT. Using promoter-luciferase reporter assays, we defined that upon TGF-β1 activation, phosphorylated Smad2 binds and promotes the KLF8 promoter activity, and knockdown of Smad2 inhibits KLF8 promoter activation. Together, these results demonstrate that TGF-β1 activates KLF8 expression by the Smad2 pathway, and KLF8 contributes to OC progression and may serve as a potential therapeutic strategy for treating OC patients.

Gentamicin Inhibits Ca2+ Channel TRPV5 and Induces Calciuresis Independent of the Calcium-Sensing Receptor-Claudin-14 Pathway.

Treatment with the aminoglycoside antibiotic gentamicin can be associated with severe adverse effects, including renal Ca2+ wasting. The underlying mechanism is unknown but it has been proposed to involve activation of the Ca2+-sensing receptor (CaSR) in the thick ascending limb, which would increase expression of claudin-14 (CLDN14) and limit Ca2+ reabsorption. However, no direct evidence for this hypothesis has been presented. We studied the effect of gentamicin in vivo using mouse models with impaired Ca2+ reabsorption in the proximal tubule and the thick ascending limb. We used a Cldn14 promoter luciferase reporter assay to study CaSR activation and investigated the effect of gentamicin on activity of the distal nephron Ca2+ channel transient receptor potential vanilloid 5 (TRPV5), as determined by patch clamp in HEK293 cells. Gentamicin increased urinary Ca2+ excretion in wild-type mice after acute and chronic administration. This calciuretic effect was unaltered in mice with genetic CaSR overactivation and was present in furosemide-treated animals, whereas the calciuretic effect in Cldn14-/- mice and mice with impaired proximal tubular Ca2+ reabsorption (claudin-2 [CLDN2]-deficient Cldn2-/- mice) was equivalent to that of wild-type mice. In vitro, gentamicin failed to activate the CaSR. In contrast, patch clamp analysis revealed that gentamicin strongly inhibited rabbit and human TRPV5 activity and chronic gentamicin administration downregulated distal nephron Ca2+ transporters. Gentamicin does not cause hypercalciuria via activation of the CaSR-CLDN14 pathway or by interfering with proximal tubular CLDN2-dependent Ca2+ reabsorption. Instead, gentamicin blocks distal Ca2+ reabsorption by direct inhibition of the Ca2+ channel TRPV5. These findings offer new insights into Ca2+ wasting in patients treated with gentamicin.

SOX8 promotes cetuximab resistance via HGF/MET bypass pathway activation in colorectal cancer.

Cetuximab is an essential drug for the treatment of wild-type K-RAS colorectal cancer (CRC). It improves the overall survival of patients. However, acquired resistance prevents its clinical efficacy. Tumor heterogeneity may be a nonnegligible reason for cetuximab resistance. We attempted to explore the corresponding molecular mechanism. Cetuximab-resistant CRC cell RKO and cetuximab-sensitive CRC cell Caco-2 were applied in this study. Cells were centrifuged to determine the concentration in the culture supernatant (CS). MTT, EdU, and colony formation assays were utilized to evaluate cell survival and proliferation. Chromatin immunoprecipitation (ChIP) and promoter-luciferase reporter assays were employed to confirm the direct binding of transcription factors. Western blot and reverse transcription-polymerase chain reaction (RT-PCR) assays were used to detect the expression of molecular markers in the pathway. Hepatocyte growth factor (HGF) was up-regulated in RKO cell culture supernatant and induced cetuximab resistance in Caco-2 cells. SRY-Box Transcription Factor 8 (SOX8) bound to the promoter sequence of HGF. HGF activated the HGF/MET bypass pathway and induced cetuximab resistance in Caco-2 cells. The SOX8/HGF/MET axis played a crucial role in the communication between cetuximab-resistant cells and cetuximab-sensitive cells, inducing treatment resistance.

Association and functional significance of genetic variants present in regulatory elements of SERPINB5 gene in gallbladder cancer.

SERPINB5 is a mammary serine protease inhibitor, which is involved in various cellular functions. The aberrant expression of SERPINB5 is reported in many cancers along with GBC but limited information is available about its role in genetic predisposition for GBC. We carried out case-control study in 206 cases and 219 controls. Promoter SNPs were genotyped by Sanger's sequencing. In-silico promoter analysis and luciferase reporter assay were done to elucidate the role of promoter variants in regulation of SERPINB5 expression. Out of four SNPs, three SERPINB5 promoter variants showed association with GBC in different models. The 'C' allele of variant rs17071138 was found to be significantly associated with GBC (p=0.017). The 'T' allele of rs3744940 significantly increased the risk for GBC in dominant (p=0.035) and additive models (p=0.005). Also, rs3744941 'T' allele increased the risk for GBC by dominant (p=0.042) as well as additive models (p=0.016). In-silico promoter analysis and luciferase reporter assay revealed the probable regulatory role of the SERPINB5 promoter variant rs17071138 on the expression. Overall, our study reveals the genetic association of SERPINB5 promoter variants with GBC and possible role of rs17071138 in the regulation of expression.

SARS-CoV-2 NSP12 Protein Is Not an Interferon-β Antagonist.

ABSTRACTThe coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is bringing an unprecedented health crisis to the world. To date, our understanding of the interaction between SARS-CoV-2 and host innate immunity is still limited. Previous studies reported that SARS-CoV-2 nonstructural protein 12 (NSP12) was able to suppress interferon-β (IFN-β) activation in IFN-β promoter luciferase reporter assays, which provided insights into the pathogenesis of COVID-19. In this study, we demonstrated that IFN-β promoter-mediated luciferase activity was reduced during coexpression of NSP12. However, we could show NSP12 did not affect IRF3 or NF-κB activation. Moreover, IFN-β production induced by Sendai virus (SeV) infection or other stimulus was not affected by NSP12 at mRNA or protein level. Additionally, the type I IFN signaling pathway was not affected by NSP12, as demonstrated by the expression of interferon-stimulated genes (ISGs). Further experiments revealed that different experiment systems, including protein tags and plasmid backbones, could affect the readouts of IFN-β promoter luciferase assays. In conclusion, unlike as previously reported, our study showed SARS-CoV-2 NSP12 protein is not an IFN-β antagonist. It also rings the alarm on the general usage of luciferase reporter assays in studying SARS-CoV-2.IMPORTANCE Previous studies investigated the interaction between SARS-CoV-2 viral proteins and interferon signaling and proposed that several SARS-CoV-2 viral proteins, including NSP12, could suppress IFN-β activation. However, most of these results were generated from IFN-β promoter luciferase reporter assay and have not been validated functionally. In our study, we found that, although NSP12 could suppress IFN-β promoter luciferase activity, it showed no inhibitory effect on IFN-β production or its downstream signaling. Further study revealed that contradictory results could be generated from different experiment systems. On one hand, we demonstrated that SARS-CoV-2 NSP12 could not suppress IFN-β signaling. On the other hand, our study suggests that caution needs to be taken with the interpretation of SARS-CoV-2-related luciferase assays.

Differential expression patterns of the two paralogous Rec8 from Nile tilapia and their responsiveness to retinoic acid signaling.

REC8 (meiotic recombination protein 8) is an essential component of meiotic cohesion complexes. Interestingly, two paralogous rec8 genes happen to exist in the stra8 (stimulated by retinoic acid gene 8)-absent fishes but not in stra8-existing fishes. Stra8 is usually considered as the prerequirement during RA (retinoic acid)-mediated meiosis initiation in mammals. However, how RA triggers meiosis in the stra8-absent fishes just like Nile tilapia (Oreochromis niloticus) remains elusive. Here we characterized the two paralogous rec8 genes in Nile tilapia (Onrec8a and Onrec8b), and investigated their expression patterns and responsiveness to RA signaling by treatment of ex vivo testicular culture and promoter luciferase reporter assay. OnRec8a and OnRec8b share 36% identity to each other and are true orthologs of REC8. Their expression was predominantly restricted to meiotic germline cells with differential spatiotemporal patterns. During spermatogenesis, OnRec8b predominantly exhibited nuclear expression in spermatocytes from 60 dah (days after hatching), while OnRec8a exhibited cytoplasmic expression from 90 dah. During oogenesis, OnRec8a was expressed from 30 dah, while OnRec8b from 90 dah. Further study shows that RA signaling could upregulate the expression of both Onrec8a and Onrec8b. Collectively, our data implies that OnRec8a and OnRec8b might have differential function during meiosis and be involved in RA-mediated meiosis program.

Hypoxia-induced Tie1 drives stemness and cisplatin resistance in non-small cell lung carcinoma cells

BackgroundDrug resistance and metastasis involving hypoxic tumor environments and persistent stem cell populations are detrimental to the survival of patients with non-small cell lung carcinoma (NSCLC). Tie1 is upregulated in hypoxia and is believed to counteract the effectiveness of platinum agents by promoting the stemness properties in cells. We have investigated the association of Tie1 with HIF-1α and cisplatin resistance in NSCLC cell lines.MethodsThe expression of Tie1 in a pulmonary microvascular endothelial cell line (HPMEC) and NSCLC cell lines was detected using qRT-PCR and western blotting. The effect of Tie1 on cell stemness and migration was examined by sphere-forming and transwell assays in NSCLC cells with Tie1 silenced. The regulation of Tie1 by HIF-1α was evaluated by a dual-luciferase reporter assay and chromatin immunoprecipitation.ResultsWe found that hypoxia could induce stemness and cisplatin resistance in vitro. Tie1 was expressed at low levels in NSCLC cells when compared with human pulmonary microvascular endothelial cells, however, its expression was increased by hypoxia. Additionally, Tie1 knockdown could reduce stemness properties and increase sensitivity to cisplatin in vitro and in a xenograft mouse model. The promoter of Tie1 contains two predicted hypoxia-response elements (HREs). We mutated both HRE sites and conducted chromatin immune-precipitation and promoter luciferase reporter assays and were able to conclude that the induction of Tie1 by hypoxia was HIF-1α-dependent.ConclusionsOur findings indicated that Tie1 is upregulated in a hypoxic environment by HIF-1α and contributes to tumorigenesis and cisplatin resistance through the promotion of stemness in NSCLC cells.

Genetics and Genomics of SOST: Functional Analysis of Variants and Genomic Regulation in Osteoblasts.

SOST encodes the sclerostin protein, which acts as a key extracellular inhibitor of the canonical Wnt pathway in bone, playing a crucial role in skeletal development and bone homeostasis. The objective of this work was to assess the functionality of two variants previously identified (the rare variant rs570754792 and the missense variant p.Val10Ile) and to investigate the physical interactors of the SOST proximal promoter region in bone cells. Through a promoter luciferase reporter assay we show that the minor allele of rs570754792, a variant located in the extended TATA box motif, displays a significant decrease in promoter activity. Likewise, through western blot studies of extracellular and intracellular sclerostin, we observe a reduced expression of the p.Val10Ile mutant protein. Finally, using a circular chromosome conformation capture assay (4C-seq) in 3 bone cell types (MSC, hFOB, Saos-2), we have detected physical interactions between the SOST proximal promoter and the ECR5 enhancer, several additional enhancers located between EVT4 and MEOX1 and a distant region containing exon 18 of DHX8. In conclusion, SOST presents functional regulatory and missense variants that affect its expression and displays physical contacts with far reaching genomic sequences, which may play a role in its regulation within bone cells.

Abstract 1279: SUMOylation upregulates EZH2 expression by modifying transcriptional factor E2F1

Abstract Enhancer of Zeste Homolog 2 (EZH2) is the enzymatic subunit of Polycomb Repressive Complex 2 (PRC2), which modifies histone H3 lysine 27 trimethylation (H3K27me3) at target promoters for gene silencing. High level of EZH2 expression is often observed in cancers, however the regulation of EZH2 expression is not well understood. We conducted genome wide RNA profiling in colorectal cancer cell line HCT116 with inducible knockdown of SUMO activating enzyme subunit 2 (SAE2). RNA-sequencing results showed EZH2 mRNA decreased and GSEA analysis indicated suppression of downstream genes of H3K27me3 upon SAE2 knockdown. Real-time PCR and western blot confirmed that knockdown of SAE2 decreased EZH2 mRNA and protein level as well as H3K27me3. The same effects were observed in breast cancer cell lines MB-MDA-231 and HCC1937. EZH2 promoter reporter assay demonstrated that SUMOylation enhanced EZH2 promoter transcriptional activity. SiRNA knockdown of SUMO E1 enzyme SAE2 or SUMO E2 enzyme UBC9 resulted in induction of CDH1, an target gene silenced by EZH2. EZH2 promoter luciferase reporter assay identified E2F transcription factor 1 (E2F1) as a major transcriptional activator of EZH2. The enhancement of EZH2 promoter transcriptional activity of E2F1 was induced by co-expression of SAE2 or SUMO1 but suppressed by expression of deSUMOylation enzyme SENP1. E2F1 SUMOylation site mutation (K266R) significantly decreased its transcriptional activation as measured by the EZH2 promoter reporter assay. SAE2 knockdown decreased the occupancy of E2F1 on EZH2 promoter as measured by Chromatin immunoprecipitation (ChIP) assay. The binding of E2F1 to EZH2 promoter region was significantly decreased when E2F1 SUMOyaltion site was mutated, which suggests that SUMOylation is required for E2F1 binding to EZH2 promoter. Western blot of tumor tissues from xenograft mouse model generated by colorectal cancer cell line HCT116 showed consistent results that knockdown of SAE2 decreased EZH2 mRNA and protein level in vivo. EZH2 protein level was also reduced in patient-derived xenograft (PDX) primary colorectal cancer cells when SAE2 was knockdown by siRNA transfection. Patient specimen data analysis showed SAE2 expression is associated with human colon and breast cancer patient survival and there is a strong positive correlation between EZH2 and SAE2 expression in tumor specimen from colon cancer and breast cancer patients. This data from patient samples supports our findings made in cellular and animal models. Of note, EZH2 and its partner SUZ12 are substrates of SUMOylation. However, their SUMOylation does not appear to impact the functions of PRC2. In conclusion, our results demonstrate that SUMOylation up-regulates EZH2 expression through regulating its gene expression via E2F1's transcriptional activity. Our findings reveal a novel mechanism of EZH2 regulation, indicating that SUMO E1 inhibitor could be synergistic with EZH2 inhibitor in colon and breast cancer therapy. Shrimp miR-965 induced the human melanoma stem-like cell apoptosis and inhibited their stemness by disrupting the MCL-1-ER stress-XBP1 feedback loop in a cross-species manner

BackgroundMelanoma is a type of aggressive skin cancer with a poor survival rate. The resistance to conventional therapy of this disease is, at least in part, attributed to its cancer stem cell population. However, the mechanism of survival and stemness maintenance of cancer stem cells remains to be investigated.MethodsTumorsphere formation assay was used to study the stem-like property of melanoma stem-like cells (MSLC). Chromatin immunoprecipitation (ChIP), promoter luciferase reporter assay were included for exploring the role of MCL-1 in MSLC and electrophoretic mobility shift assay were used to evaluate the interaction between shrimp miR-965 and human Ago2 protein. Melanoma xenograft nude mice were used to study the inhibition of tumor development.ResultsIn the present study, our results showed that myeloid cell leukemia sequence 1 (MCL-1) knocking down induced ER stress and apoptosis, and the expression reduction of stemness associated genes in MSLC, which implied a significant role of MCL-1 in MSLC. Further study indicated that ER stress agonist (tunicamycin) treatment in MSLC results in the translocation of XBP1, an ER stress sensor, into the nucleus to induce MCL-1 expression through direct binding to the − 313- to − 308-bp region of MCL-1 promoter. In addition, we found that a shrimp-derived miRNA (shrimp miR-965) could interact with the human Ago2 protein and suppressed the human MCL-1 expression by binding to the 3′ UTR of MCL-1 mRNA, thereby inhibiting the MSLC proliferation and stemness in vitro and in vivo in a cross-species manner.ConclusionIn conclusion, we identified an important role of MCL-1-ER stress-XBP1 feedback loop in the stemness and survival maintenance of MSLC, and shrimp miR-965, a natural food derived miRNA, could regulate MSLC stemness and survival by targeting MCL-1 and disrupting the balance of MCL-1-ER stress-XBP1 feedback loop. In conclusion, this study indicated an important mechanism of the regulation of MSLC stemness and survival, otherwise it also demonstrated the significance of cross-species-derived miRNA as promising natural drugs in melanoma therapy.

228 Loss of EPHA2 represses GATA-3 function and causes a terminal differentiation defect

Epidermal morphogenesis and differentiation require the coordination of complex signal transduction networks. These signal relays are often initiated at the plasma membrane and transmitted to the nucleus to control gene expression. Receptor tyrosine kinases (RTKs) are integral in orchestrating communication cascades to induce differentiation. We report loss of EPHA2 RTK causes a terminal differentiation defect in 3D human skin equivalents (3D HSE) resulting in loss of stratification and ablation of the granular and cornified layers. In EPHA2-deficient (shEPHA2) 3D HSE, we show significant loss (P<0.05) of loricrin, filaggrin, and involucrin protein and mRNA, indicating EPHA2 signaling can impact keratinocyte differentiation at the transcriptional level. The transcription factor GATA-3 is a key driver of differentiation-associated gene expression in epidermis, although the upstream signals regulating its activity are unknown. GATA-3 is expressed in the nuclei of suprabasal keratinocytes in mature 3D HSE, mimicking the pattern in normal human skin (R2=0.99). In 2D cultures, GATA-3 accumulates in the nucleus following 24 h exposure to 1.2 mM calcium. However, GATA-3 protein and transcript expression are lost in shEPHA2 3D HSE and 2D cultures (P<0.001). Further, there is a decrease of GATA-driven transcription indicated by a significant loss in GATA binding activity in a promoter luciferase reporter assay (P<0.001). Re-expression of nuclear GATA-3 using a retroviral construct in EPHA2-deficient 3D HSE restored differentiation. Taken together, these results indicate that EPHA2 promotes GATA-3 nuclear accumulation to positively regulate the transcription of terminal differentiation genes in epidermal keratinocytes.
