The NMN Module Conducts Nodule Number Orchestra.

Legumes control nodule number through nodulation and autoregulation of nodulation (AON) pathways. Nodule Inception (NIN) is essential for rhizobial infection and nodule organogenesis in legumes. The GmNINa-miR172c-NNC1 (NMN) module, which consists of two positive regulators, GmNINa and miR172c, and a suppressor, NNC1, integrates both pathways. GmNINa activates miR172c to downregulate NNC1, leading to nodulation, while NNC1 inhibits miR172c expression, forming a negative feedback loop. GmNINa and NNC1 interact with each other and antagonistically fine-tune GmRIC1/RIC2 expression, turning AON on and off. Conversely, activation of AON inhibits GmNINa and miR172c expression, thereby reducing their inhibitory effects on NNC1 to attenuate both nodulation signaling and AON. The NMN module functions not only as an “accelerator” of the nodulation signal to promote nodulation but also as a “brake” on the signal by activating AON to orchestrate nodule number.

Proline-rich protein-like PRPL1 controls elongation of root hairs in Arabidopsis thaliana.

The synthesis and composition of cell walls is dynamically adapted in response to many developmental and environmental signals. In this respect, cell wall proteins involved in controlling cell elongation are critical for cell development. Transcriptome analysis identified a gene in Arabidopsis thaliana, which was named proline-rich protein-like, AtPRPL1, based on sequence similarities from a phylogenetic analysis. The most resemblance was found to AtPRP1 and AtPRP3 from Arabidopsis, which are known to be involved in root hair growth and development. In A. thaliana four proline-rich cell wall protein genes, playing a role in building up the cross-connections between cell wall components, can be distinguished. AtPRPL1 is a small gene that in promoter::GUS (β-glucuronidase) analysis has high expression in trichoblast cells and in the collet. Chemical or mutational interference with root hair formation inhibited this expression. Altered expression levels in knock-out or overexpression lines interfered with normal root hair growth and etiolated hypocotyl development, but Fourier transform-infrared (FT-IR) analysis did not identify consistent changes in cell wall composition of root hairs and hypocotyl. Co-localization analysis of the AtPRPL1-green fluorescent protein (GFP) fusion protein and different red fluorescent protein (RFP)-labelled markers confirmed the presence of AtPRPL1-GFP in small vesicles moving over the endoplasmic reticulum. Together, these data indicate that the AtPRPL1 protein is involved in the cell's elongation process. How exactly this is achieved remains unclear at present.

OakContigDF159.1, a reference library for studying differential gene expression in Quercus robur during controlled biotic interactions: use for quantitative transcriptomic profiling of oak roots in ectomycorrhizal symbiosis

Oaks (Quercus spp.), which are major forest trees in the northern hemisphere, host many biotic interactions, but molecular investigation of these interactions is limited by fragmentary genome data. To date, only 75 oak expressed sequence tags (ESTs) have been characterized in ectomycorrhizal (EM) symbioses. We synthesized seven beneficial and detrimental biotic interactions between microorganisms and animals and a clone (DF159) of Quercus robur. Sixteen 454 and eight Illumina cDNA libraries from leaves and roots were prepared and merged to establish a reference for RNA-Seq transcriptomic analysis of oak EMs with Piloderma croceum. Using the Mimicking Intelligent Read Assembly (MIRA) and Trinity assembler, the OakContigDF159.1 hybrid assembly, containing 65 712 contigs with a mean length of 1003 bp, was constructed, giving broad coverage of metabolic pathways. This allowed us to identify 3018 oak contigs that were differentially expressed in EMs, with genes encoding proline-rich cell wall proteins and ethylene signalling-related transcription factors showing up-regulation while auxin and defence-related genes were down-regulated. In addition to the first report of remorin expression in EMs, the extensive coverage provided by the study permitted detection of differential regulation within large gene families (nitrogen, phosphorus and sugar transporters, aquaporins). This might indicate specific mechanisms of genome regulation in oak EMs compared with other trees.

Proline-rich cell wall proteins accumulate in growing regions and phloem tissue in response to water deficit in common bean seedlings

Plant cell walls undergo dynamic changes in response to different environmental stress conditions. In response to water deficit, two related proline-rich glycoproteins, called p33 and p36, accumulate in the soluble fraction of the cell walls in Phaseolus vulgaris (Covarrubias et al. in Plant Physiol 107:1119-1128, 1995). In this work, we show that p33 and p36 are able to form a 240 kDa oligomer, which is found in the cell wall soluble fraction. We present evidence indicating that the highest accumulation of these proteins in response to water deficit occurs in the growing regions of common bean seedlings, particularly in the phloem tissues. These proteins were detected in P. vulgaris cell suspension cultures, where the p33/p36 ratio was higher under hyperosmotic conditions than in bean seedlings subjected to the same treatment. The results support a role for these proteins during the plant cell response to changes in its water status, and suggest that cell wall modifications are induced in active growing cells of common bean in response to water limitation.

Coating proteins: structure and cross-linking in fp-1 from the green shell mussel Perna canaliculus.

The protein family known as fp-1 provides mussel byssus with a protective outer coating and has drawn much attention for its water resistant bioadhesive properties in vitro. A new fp-l isolated from the green shell mussel Perna canaliculus (pcfp-1) reveals a composition dominated by only four amino acids: 3,4-dihydroxyphenyl-L-alanine (dopa), lysine, proline, and valine at approximately 20 mol % each. SDS-PAGE and MALDI-TOF mass spectrometry detected size variants at 48 and 52 kDa in preparations of purified Pcfp-1. The N-terminal sequence enabled construction of oligonucleotide primers for PCR and RACE-derived cDNAs from which the complete sequence of four variants was deduced. pcfp-1 deviates from all known homologues in other mussels in several notable respects: its mass is half, most of its sequence is represented by 75 tandem repeats of a tetrapeptide, i.e., PY*VK, in which Y* is dopa, prolines are not hydroxylated, and thiolate cysteines are clustered in homologous sequences at both the amino and carboxy termini. Amino acids in the repeat sequence show a striking resemblance to proline-rich cell wall proteins with tandemly repeated PPVYK pentapeptides [Hong, J. C., Nagao, R. T., and Key, J. L. (1987) J. Biol. Chem. 262, 8367-8376]. Cysteine plays a key role in cross-linking pcfp-1 by forming adducts with dopaquinone. Significant 5-S-cysteinyldopa and smaller amounts of 2-S-cysteinyldopa were detected in hydrolysates of the byssal threads of P. canaliculus. The cross-links could also be formed by oxidation of pcfp-1 in vitro using mushroom tyrosinase. Cysteinyldopa cross-links were present in trace amounts only in the byssus of other mussel species.

Accumulation of extracellular proteins bearing unique proline-rich motifs in intercellular spaces of the legume nodule parenchyma.

Nodulins encoding repetitive proline-rich cell wall proteins (PRPs) are induced during early interactions with rhizobia, suggesting a massive restructuring of the plant extracellular matrix during infection and nodulation. However, the proteins corresponding to these gene products have not been isolated or characterized, nor have cell wall localizations been confirmed. Posttranslational modifications, conformation, and interactions with other wall polymers are difficult to predict on the basis of only the deduced amino acid sequence of PRPs. PsENOD2 is expressed in nodule parenchyma tissue during nodule organogenesis and encodes a protein with distinctive PRP motifs that are rich in glutamate and basic amino acids. A database search for the ENOD2 signature motifs indicates that similar proteins may have a limited phylogenetic distribution, as they are presently only known from legumes. To determine the ultrastructural location of the proteins, antibodies were raised against unique motifs from the predicted ENOD2 sequence. The antibodies recognized nodule-specific proteins in pea (Pisum sativum), with a major band detected at 110 kDa, representing a subset of PRPs from nodules. The protein was detected specifically in organelles of the secretory pathway and intercellular spaces in the nodule parenchyma, but it was not abundant in primary walls. Similar proteins with an analogous distribution were detected in soybean (Glycine max). The use of polyclonal antibodies raised against signature motifs of extracellular matrix proteins thus appears to be an effective strategy to identify and isolate specific structural proteins for functional analysis.

Genetic analysis of net-like cracking in soybean seed coats

Summary Cracking of seed coats in soybean (Glycine max (L.) Merr.) deteriorates the external appearance of seeds and reduces their commercial value. Two types of cracking have been reported that occur in some cultivars: Type I with irregular cracks and Type II with net-like cracks. This study was conducted to determine the genetic basis of net-like cracking. Genetic analysis was performed using F1 plants produced by crossing Uzuramame, a Japanese landrace with black seed coats having net-like cracking and a Clark mutant with black seed coats, their F2 population and F3 lines. Degree of cracking in individual plants was calculated by averaging cracking index (no cracking: 0 to severe cracking: 4) of total or 100-seed samples (average cracking index, ACI). Uzuramame exhibited intense cracking, whereas the Clark mutant showed slight cracking. Intermediate degree of cracking in F1 plants suggested incomplete dominance. ACI of F2 plants was continuously distributed. Gene number involved was estimated to be 1.4 by Wright’s method. All F3 lines derived from F2 plants with ACI more than 2.8 displayed severe cracking phenotypes. In contrast, F3 lines derived from F2 plants with ACI less than 2.8 segregated from low to high ACI (0.5 to 3.2). When F2 plants were classified as slight (ACI 2.8) cracking, the frequency distribution of the F2 plants fitted to a 3:1 ratio. Genotypes of SSR marker Satt264 that is closely linked to SoyPRP1 locus for proline-rich cell wall protein had a minor effect on ACI. Further, seed weight was positively associated with ACI (r =0 .46 ∗∗ ). Our results suggest that net-like cracking is controlled primarily by a major gene, and SoyPRP1 and gene(s) contributing to seed weight may have minor effects on the intensity of cracking.

Structure and expression of a hybrid proline-rich protein gene in the solanaceous species, Solanum brevidens, Solanum tuberosum, and Lycopersicum esculentum

Summary The full-length cDNA of a previously identified Solanum brevidens gene was isolated and characterised. DNA sequence analysis revealed an open reading frame that encodes a hybrid proline-rich cell wall protein of 407 amino acids. The putative protein was designated SbrPRP. The SbrPRP harbours three parts, an N-terminal signal peptide followed by a repetitive proline-rich domain and a cysteine-rich C-terminus resembling non-specific lipid-transfer proteins. The repetitive proline-rich domain contains two repeated motifs, PPHVKPPSTPK and PTPPIVSPP extended with TPKYP and TPKPPS motifs, respectively, at their N- or C-terminal. The SbrPRP gene of the non-tuberising Solanum species, Solanum brevidens , possesses highly homologous counterparts in the tuberising species, Solanum tuberosum ( StPRP ) and in the related species, Lycopersicum esculentum ( TFM7 ). All three genes are present in single- or low copy number in the corresponding genome. Organ-specific expression of the genes, however, is different in the three solanaceous species.

Molecular biology of grape berry ripening

Grapevines produce non-climacteric fruit that exhibit a double sigmoidal pattern of growth. Ripening occurs during the second growth phase when grapes change colour, start to soften, accumulate reducing sugars, metabolise organic acids and synthesise flavour compounds. Unlike many other fruit, grapes ripen while the berries are still expanding, and as with most non-climacteric fruit, ripening does not appear to be controlled by ethylene. Sugars and amino acids that accumulate in grapes during ripening are imported via the phloem, while many secondary metabolites are synthesised within the berry itself. Grapes import sucrose but accumulate hexoses. Conversion of sucrose to hexoses is most likely catalysed by invertase. cDNAs encoding vacuolar invertases have been isolated from grape berries. Expression of these genes and an increase in invertase activity occur before veraison, so it seems unlikely that synthesis of this enzyme is a controlling factor for sugar accumulation during ripening. Proteins that transport sugars into the berry vacuole may regulate sugar accumulation, and cDNAs encoding both sucrose and hexose transporters have been isolated from ripening grape berries. Determination of the role of these transporters may reveal the pathway of sugar accumulation in grapes. Anthocyanins are only synthesised in the skin of red grapes after veraison. Analysis of the patterns of expression of genes in the flavonoid pathway has shown that there is a dramatic increase in expression of many of these genes in skin cells at veraison. Expression of the gene encoding a glycosyl transferase involved in the lasts steps of anthocyanin synthesis was absolutely correlated with anthocyanin synthesis and may explain the lack of anthocyanin synthesis in white grapes and in the flesh of most red grapes. We infer that the synthesis of anthocyanins is regulated at the transcription level and is likely to be controlled by regulatory genes. Softening of fruit generally results from changes in the properties of cell walls. Analysis of the cell walls of grapes during ripening suggests that there are no dramatic changes in polysaccharide composition but modification of specific components may contribute to softening. A number of proteins are newly synthesised in grapes during ripening and several of these proteins have now been identified. The most abundant are pathogenesis-related (PR) proteins, including chitinases and thaumatin-like proteins. Expression of genes encoding a number of PR proteins increased dramatically in grapes during ripening. It is not clear what role the PR proteins play during ripening but they may provide resistance to pathogens. Differential screening of a post-veraison grape berry cDNA library has also identified ripening-related genes, some of which encode proline-rich cell wall proteins. Other grape ripening-related genes have homologues that are induced by stress in other plants. These studies indicate that a dramatic change in gene expression occurs in grape berries at veraison and suggest that ripening involves a coordinated increase in transcription of a number of different genes.

Expression of AtPRP3, a proline-rich structural cell wall protein from Arabidopsis, is regulated by cell-type-specific developmental pathways involved in root hair formation.

The tightly regulated expression patterns of structural cell wall proteins in several plant species indicate that they play a crucial role in determining the extracellular matrix structure for specific cell types. We demonstrate that AtPRP3, a proline-rich cell wall protein in Arabidopsis, is expressed in root-hair-bearing epidermal cells at the root/shoot junction and within the root differentiation zone of light-grown seedlings. Several lines of evidence support a direct relationship between AtPRP3 expression and root hair development. AtPRP3/beta-glucuronidase (GUS) expression increased in roots of transgenic seedlings treated with either 1-aminocyclopropane-1-carboxylic acid (ACC) or alpha-naphthaleneacetic acid (alpha-NAA), compounds known to promote root hair formation. In the presence of 1-alpha-(2-aminoethoxyvinyl)glycine (AVG), an inhibitor of ethylene biosynthesis, AtPRP3/GUS expression was strongly reduced, but could be rescued by co-addition of ACC or alpha-NAA to the growth medium. In addition, AtPRP3/GUS activity was enhanced in ttg and gl2 mutant backgrounds that exhibit ectopic root hairs, but was reduced in rhd6 and 35S-R root-hair-less mutant seedlings. These results indicate that AtPRP3 is regulated by developmental pathways involved in root hair formation, and are consistent with AtPRP3's contributing to cell wall structure in Arabidopsis root hairs.

Characterization and expression of four proline-rich cell wall protein genes in Arabidopsis encoding two distinct subsets of multiple domain proteins.

We have characterized the molecular organization and expression of four proline-rich protein genes from Arabidopsis (AtPRPs). These genes predict two classes of cell wall proteins based on DNA sequence identity, repetitive motifs, and domain organization. AtPRP1 and AtPRP3 encode proteins containing an N-terminal PRP-like domain followed by a C-terminal domain that is biased toward P, T, Y, and K. AtPRP2 and AtPRP4 represent a second, novel group of PRP genes that encode two-domain proteins containing a non-repetitive N-terminal domain followed by a PRP-like region rich in P, V, K, and C. Northern hybridization analysis indicated that AtPRP1 and AtPRP3 are exclusively expressed in roots, while transcripts encoding AtPRP2 and AtPRP4 were most abundant in aerial organs of the plant. Histochemical analyses of promoter/beta-glucuronidase fusions localized AtPRP3 expression to regions of the root containing root hairs. AtPRP2 and AtPRP4 expression was detected in expanding leaves, stems, flowers, and siliques. In addition, AtPRP4 expression was detected in stipules and during the early stages of lateral root formation. These studies support a model for involvement of PRPs in specifying cell-type-specific wall structures, and provide the basis for a genetic approach to dissect the function of PRPs during growth and development.

A defective seed coat pattern (Net) is correlated with the post-transcriptional abundance of soluble proline-rich cell wall proteins.

The pigmented seed coats of several soybean (Glycine max (L.) Merr.) plant introductions and isolines have unusual defects that result in cracking of the mature seed coat exposing the endosperm and cotyledons. It has previously been shown that the T (tawny) locus that controls the color of trichomes on stems and leaves also has an effect on both the structure and pigmentation of the seed coat. Distribution of pigmentation on the seed coat is controlled by alleles of the I (inhibitor) locus. It was also found that total seed coat proteins were difficult to extract from pigmented seed coats with i T genotypes because they have procyanidins that exhibit tannin properties. We report that the inclusion of poly-L-proline in the extraction buffer out-competes proteins for binding to procyanidins. Once this problem was solved, we examined expression of the proline-rich cell wall proteins PRP1 and PRP2 in pigmented genotypes with the dominant T allele. We found that both homozygous i T and i t genotypes have reduced soluble PRP1 levels. The epistatic interaction of the double recessive genotype at both loci is necessary to produce the pigmented, defective seed coat phenotype characteristic of seed coats with the double recessive i and t alleles. This implies a novel effect of an enzyme in the flavonoid pathway on seed coat structure in addition to its effect on flavonoids, anthocyanidins, and proanthocyanidins. No soluble PRP1 polypeptides were detectable in pigmented seed coats (i T genotypes) of isolines that also display a net-like pattern of seed coat cracking, known as the Net defect. PRP2 was also absent in one of the these lines. However, both PRP1 and PRP2 cytoplasmic mRNAs were found in the Net-defective seed coats. Together with in vitro translation studies, these results suggest that the absence of soluble PRP polypeptides in the defective Net lines is post-translational and could be due to a more rapid or premature insolubilization of PRP polypeptides within the cell wall matrix.

Our prior study (Fleck & Fitt 1999; J Exp Mar Biol Ecol 234:83-94) indicated that marine bacteria are involved in the release of at least 1 peptidic compound from such leaves. The goal of our present study was to isolate and purify at least 1 natural peptidic cue originating from deteriorating leaves by means of ultrafiltration, gel filtration and reversed phase HPLC and subsequently obtain characteristic data of this cue. The ultrafiltrate (<=10 kD) of the homogenate of decaying mangrove leaves was subjected to gel filtration on a Sephadex G 25 column, resulting in 3 fractions which were tested for their capacity to induce metamorphosis of planula larvae in bioassays performed in the laboratory. Fraction I (>=5 kD) was most effective in inducing metamorphosis of 75% of planulae at 1 mg freeze-dried material ml-1 seawater within 24 h. Fractions II and III (both <=5 kD) resulted in metamorphosis of only 1% of larvae or less within 72 h when applied at 5 mg ml-1. Isochratic HPLC separation of Fraction I with 24% methanol yielded 2 biologically active fractions. One fraction (A/B), which induced 47% of the larvae to metamorphose at 0.9 mg lyophilized material ml-1 seawater within 24 h, consisted of a mixture of at least 2 subfractions and was not further analyzed. The other fraction (C) effected metamorphosis of 85% of larvae at a concentration of 0.5 mg ml-1 within 24 h. Matrix-assisted laser desorption ionization (MALDI) mass spectrometry of this fraction revealed a molecular weight of approximately 5.8 kD. Automated amino acid analysis showed that Fraction C was rich in proline (ca 44%) and glycine residues (ca 16%), corresponding to characteristic proline-rich cell wall proteins of plants. Automated sequencing of the natural inducer failed due to a blocked amino terminus. The results of our present study suggest that metamorphic inducers for C. xamachana may emerge nonspecifically as a byproduct of bacterial degradation of deteriorating, proteinaceous plant tissue in their habitat. Reconstitution in vitro of the components and conditions required for the oxidative cross-linking of extracellular proteins in French bean ( Phaseolus vulgaris L.)

Oxidative cross linking of three (hydroxy)proline-rich cell wall proteins, known to be immobilised during the elicitor-induced oxidative burst in French bean cells, was modelled using peroxidases with cysteine or H 2O 2. Further reconstitution of the homologous system was achieved with a 46 kDa bean cell-wall peroxidase using conditions known to appertain in cell walls prior to the immobilisation. Thus, cell wall alkalinisation and secretion of a reductant have been reconstituted by addition of apoplastic fluid to the wall proteins and the 46 kDa peroxidase with resultant cross-linking. This is the first demonstration of the oxidative cross-linking of cell wall glycoproteins without the addition of external H 2O 2. © 1997 Federation of European Biochemical Societies.

Transcripts of a gene encoding a putative cell wall-plasma membrane linker protein are specifically cold-induced in Brassica napus.

We have isolated a gene and cDNA from Brassica napus encoding a hybrid-proline-rich protein. The putative protein is modular in structure. The N-terminal domain has properties of a signal peptide which would direct the protein into the ER. Amino acids 27 to 287 comprise three domains which contain high levels of proline and several other amino acids common in proline-rich cell wall proteins. These domains are characterised by repeating amino acid motifs. The C-terminal domain (amino acids 288 to 376) contains three putative membrane-spanning regions and shows a high degree of amino acid similarity to known hybrid-proline-rich proteins from several species. It is likely that the protein is secreted from the cell, located in the cell wall and anchored in the plasma membrane via the C-terminal domain. Transcripts encoding this protein are induced in leaf tissue within 8 h of cold treatment and decrease rapidly when plants are returned to normal temperatures. The transcripts are not induced by heat shock, dehydration, exogenous ABA or wounding, whereas transcripts of a control B. napus gene are induced by dehydration and ABA. The possible function of this protein in cold tolerance is discussed.

Starch synthesis-, and tuber storage protein genes are differently expressed in Solanum tuberosum and in Solanum brevidens

Studying in vitro stem cuttings of Solanum tuberosum induced for tuberization and those of a non-tuberizing Solanum species, differences both in morphology and in gene expression were detected. Stolon formation essentially depended on light while tuberization was triggered by the elevated level of sucrose in the medium. Genes involved in starch synthesis were induced by sucrose in both species, however, starch granules were detected only in potato. A new tuber specific cDNA clone, GM7, encoding a putative metallocarboxypeptidase inhibitor and the cDNA of a proline rich cell wall protein with S. brevidens specific expression were isolated by differential screening. Sucrose mediated transcription of the tuber storage proteins like patatin and proteinase inhibitors (Kunitz-type, winI, GM7) failed in S. brevidens.

Differential chemical allocation and plant adaptation: A Py-MS Study of 24 species differing in relative growth rate

The chemical composition of leaves of 24 wild species differing in potential relative growth rate (RGR) was analysed by pyrolysis-mass spectrometry. The variation in RGR significantly correlated with differences in chemical composition: slow-growing species were richer in glucan-based polysaccharides and in C16:0 fatty acid, whereas fast growing ones contained more protein (other than those incorporated in cell walls) and chlorophyll, sterols and diglycerides. Other, apparently significant correlations, e.g. for pentose-based hemicellulose and for guaiacyl lignin appeared solely based on a group separation between mono- and dicotyledonous species. Considering the eleven monocotyledonous and thirteen dicotyledonous species separately, correlations were found in addition to the previously mentioned general ones. Within the group of the monocotyledons the low-RGR species were significantly enriched in pentose-based hemicellulose, ferulic acid and (hydroxy)proline-rich cell wall protein and nearly significant in guaiacyl and syringyl lignin, fast-growing species contained more potassium. Within the group of the dicotyledons slow-growing species were enriched in triterpenes and aliphatic wax esters. In general, the monocotyledons contained more cell wall material such as pentose-based hemicellulose, ferulic acid, glucans (including cellulose) and guaiacyl-lignin, and also more aliphatic wax esters, than the dicotyledons. The dicotyledons, on the other hand, contained somewhat more protein than the grasses. Per unit weight of cell wall, the amount of (hydroxy)proline- rich protein in low-RGR species was comparatively low. A higher investment of cell wall proteins to explain the low rate of photosynthesis per unit of leaf nitrogen of slow-growing species as suggested by Lambers and Poorter (1992), therefore, seems unlikely.

Xylem-specific gene expression in loblolly pine

Two genes preferentially expressed in differentiating xylem of loblolly pine (Pinus taeda L.) were cloned from cDNA and genomic libraries and designated PtX3H6 and PtX14A9. Transcripts of PtX3H6 and PtX14A9 are very abundant in differentiating xylem, less abundant in needles, and very low or non-detectable in embryos and megagametophytes. PtX3H6 contains a putative signal peptide, a threonine-rich region, a proline-rich region, and a hydrophobic tail. Repeats of Pro-Pro-Pro-Val-X-X are similar to repeats found in proline-rich cell wall proteins. The amino acid compositions of PtX3H6 and PtX14A9 are similar to those of arabinogalactan proteins (AGPs). PtX14A9 contains an 8 amino acid sequence similar to amino terminal sequences of ryegrass, carrot and rose AGPs. Upstream sequences have been determined from genomic clones encoding PtX3H6 and PtX14A9. A 7 bp sequence found in the 5' flanking regions of both genes has previously been shown to be involved in the vascular-specific expression of GRP 1.8, a glycine-rich protein found in bean. The sequence is also present upstream of another glycine-rich protein from bean, GRP 1.0, and may be partially responsible for the xylem-specific expression of pTx3H6 and PtX14A9.

Post-transcriptional regulation of a salt-inducible alfalfa gene encoding a putative chimeric proline-rich cell wall protein.

A cDNA previously shown to identify a salt-inducible root-specific transcript in Medicago sativa was used to screen an alfalfa library for the corresponding genomic sequence. One positive clone was recovered. The nucleotide sequence of a subclone contained a 329 bp 5' region upstream of the first ATG codon, a 1143 bp coding segment, and a 447 bp 3'-untranslated region interrupted by a single 475 bp intron. Translation of the coding segment, which was designated MsPRP2, suggested it encodes a chimeric 40,569 Da cell wall protein with an amino-terminal signal sequence, a repetitive proline-rich sequence, and a cysteine-rich carboxyl-terminal sequence homologous to nonspecific lipid transfer proteins. The 3'-untranslated region of MsPRP2 contained a sequence similar to one found to destabilize mRNAs transcribed from the elicitor-regulated proline-rich protein gene PvPRP1. Transcription run-on experiments using nuclei from salt-sensitive and salt-tolerant alfalfa callus suggested that the accumulation of MsPRP2 transcripts in salt-tolerant alfalfa cells grown in the presence of salt is due primarily to increased mRNA stability. The MsPRP2 gene thus may be a useful model for studying post-transcriptional salt-regulated expression of cell wall proteins.

Differential Expression of Two Soybean (Glycine max L.) Proline-Rich Protein Genes after Wounding.

We have investigated the wound-induced expression of two members of the soybean (Glycine max L.) proline-rich cell wall protein gene family and show that SbPRP1 and SbPRP2 exhibit unique patterns of expression after physical damage. SbPRP1 mRNA can be detected in the hook of soybean seedlings within 2 h after wounding and is present at high levels in the hook and elongating hypocotyl 20 h after wounding. In contrast, SbPRP2 mRNA increases transiently and rapidly throughout the soybean seedling after wounding. SbPRP2 is also induced by wounding in soybean leaves, but the pattern of mRNA accumulation in leaves is distinct from that seen in seedlings and reaches high levels of expression 20 h after physical damage. SbPRP2 mRNA levels were also found to increase in the mature hypocotyl and roots of seedlings in response to treatment with 10 [mu]M indoleacetic acid and naphthalene-1-acetic acid. These data indicate that the wound-induced expression of PRPs in soybean is tissue specific and that the regulation of these genes after physical damage may operate through different signal transduction pathways.
