Freeze casting of hydroxyapatite-titania composites for bone substitutes.

Hydroxyapatite (HA) is commonly used as a bone substitute material, but it lacks mechanical strength when compared to native bone tissues. To improve the efficacy of HA as a bone substitute by improving the mechanical strength and cell growth attributes, porous composite scaffolds of HA and titania (HA-TiO2 ) were fabricated through a freeze-casting process. Three different compositions by weight percent, 25-75 HA-TiO2 , 50-50 HA-TiO2 , and 75-25 HA-TiO2 , were custom-made for testing. After sintering at 1250°C, these composite scaffolds exhibited improved mechanical properties compared to porous HA scaffolds. Substrate mixing was observed, which helped reduce crystal size and introduced new phases such as β-TCP and CaTiO3 , which also led to improved mechanical properties. The composition of 50-50 HA-TiO2 had the highest ultimate compressive strength of 3.12 ± 0.36 MPa and elastic modulus 63.29 ± 28.75 MPa. Human osteoblast cell proliferation assay also increased on all three different compositions when compared to porous HA at 14 days. These results highlight the potential of freeze casting composites for the fabrication of bone substitutes, which provide enhanced mechanical strength and biocompatibility while maintaining porosity.

Strontium/zinc phytate-based self-assembled monolayers on titanium surfaces enhance osteogenesis and antibacterial performance in vitro

The accumulation of bacteria over implant surfaces is still the first cause of failure, and the development of antimicrobial surfaces constitutes a first line in implant research. Besides, the durability and mechanical performance of implants, in special in the dental area, are mainly determined by their osseointegration capacity into the maxillofacial bone and the appearance of infections. Consequently, implant osseointegration and infection prophylaxis remain as big challenges to attain so a huge investigation is being developed on the production of bioactive surfaces to achieve improvements in these aspects. In this work we propose the functionalization of titanium surfaces (Ti Cp) with self-assembled monolayers (SAMs) of bioactive organophosphate compounds: phytic acid (Ti-PA) and its metallic phytate derivatives bearing Sr2+ and/or Zn2+ (Ti-SrPhy, Ti-ZnPhy and Ti-SrPhy/ZnPhy) which exhibited tunable in vitro osteogenic, antimicrobial and antioxidant properties in a previous work. Thus, phytate compounds are chemically anchored onto Ti discs through a simple procedure consisting of a condensation reaction promoted by heat treatment. EDS and XPS spectroscopies confirm the obtaining of the modified surfaces and the topographic properties and wettability analysed by SEM, AFM, profilometry and contact angle measurements, respectively, are explored. Additionally, phytate-SAMs do not release any cytotoxic compound after 14 days and stimulate in vitro adhesion and proliferation of human osteoblast cells after 14 days of culture. The osteogenic ability of the modified surfaces evaluated by the quantification of ALP activity and matrix mineralization degree shows a significant improvement with respect to unmodified surfaces. Furthermore, the antimicrobial activity of phytate-SAMs against Streptococcus mutans cultures is evaluated. The count of viable cells and the quantification of produced biofilm are significantly reduced by all phytate-SAMs groups (p < 0.001). Cell membrane integrity studies by LIVE/DEAD staining and SEM imaging confirm a decreased viability of adhered bacteria when phytate-based surfaces are tested, due to a disruption in the function and permeability of the cell membrane. Therefore, phytate-SAMs exhibit suitable in vitro features suggesting their promising potential as bioactive coatings of dental implants.

Characterization of porous fluorohydroxyapatite bone-scaffolds fabricated using freeze casting

With the increasing demand for orthopedic and dental reconstruction surgeries, there comes a shortage of viable bone substitutes. This study was therefore designed to assess the efficacy of porous fluorohydroxyapatite (FHA) as a potential bone substitute. For this, porous FHA scaffolds were fabricated using the freeze casting technique. They were then sintered at 1250, 1350 and, 1450°C, and microstructural, mechanical, and in vitro properties were analyzed. The microstructure analyses revealed the porosity remained constant within the temperature range. However, the pore size decreased with increasing sintering temperature. The greatest compressive strength and elastic modulus were obtained at 1450°C, which were 13.5±4.0MPa and 379±182MPa, respectively. These are comparable values to human trabecular bone and other porous scaffolds made using hydroxyapatite. This analysis has thus helped to attain an understanding of the mechanical and material properties of freeze-cast FHA scaffolds that have not been presented before. In vitro studies revealed an increasing rate of human osteoblast cell proliferation on freeze-cast FHA scaffolds with increasing sintering temperature, suggesting improved osteogenic properties. Additionally, osteoblasts cells were also shown to proliferate into the interior pores of all freeze-cast FHA scaffolds. These results indicate the potential of porous FHA scaffolds fabricated using the freeze-casting technique to be utilized clinically as bone substitutes.

Protective and curative role of vitamin D and hormones on the cadmium-induced inhibition of proliferation of human osteoblast cells

Exposure to cadmium (Cd), which causes environmental and industrial pollution, causes toxicity in many tissues and organs, especially bone, lung and kidney. Hormones, growth factors and other stimuli act on bone tissue through osteoblasts. In this study, it was aimed to determine the effects of Cd on hFOB1.19 osteoblast cells and the protective and healing potentials of estrogen, androgen and vitamin D against the inhibitory effect of Cd on the proliferation. hFOB1.19cells were cultivated in our laboratory using Dulbecco's Modified Eagle's Medium-F12, HEPES medium, containing 10% fetal bovine serum, 1% penicillin/streptomycin in 34.5°C 5%CO2 incubator. To determine its protective potentials for the toxicity of CdCl2, it was previously applied 1,25(OH) 2D vitamin, 17β-estradiol, and 5α-androstane for 72h to cells. To determine their curative potential, osteoblast cells, which were previously exposed to CdCl2 for 72h, were administered 1,25(OH) 2D vitamin, 17β-estradiol, and 5α-androstane. Following these applications were determined proliferation by XTT analysis and, the amounts of androgen receptor, estrogen receptor, vitamin D receptor, alkaline phosphatase, osteocalcin and osteoprotegerin by ELISA analysis. Vitamin D has been both preventive and curative effective to increase cell proliferation, which Cd reduces. Interestingly, estrogen had a preventive effect and androgen had a curative effect. In addition to showing the negative effects of cadmium on the proliferation of osteoblast cells, this study provides an overview of the effects of hormone and vitamin D applications before and after Cd, and these results may serve as a guide for future studies.

An In Vitro Study on the Effect of Five Commercial Calcium Supplements on Human Osteoblast Cell Proliferation and Ca2+ Mineralization

Calcium is an essential mineral that supports bone and joint health. It is used as a supplement, most typically as calcium carbonate, to assist in preserving bone density especially when dietary calcium intake is inadequate. In the present study we examined the effect of five varied calcium sources, three natural and two synthetic, on stimulation, proliferation and mineralization of cultured human osteoblast cells. A MTT assay using cultured human fetal osteoblast cells (hFOB 1.19) was used to determine the effective proliferative dose for salmon collagen bone calcium (0.25 mg/ml). Elemental calcium equivalence was used to select the appropriate doses for algae calcium (0.14 mg/ml), eggshell powder (0.16 mg/ml), calcium citrate (0.22 mg/ml) and calcium carbonate (0.13). Alkaline phosphatase activity, DNA synthesis rates and calcium ion deposition were evaluated after incubation under different conditions and harvesting the cells. Alkaline phosphatase activity, DNA synthesis rates and calcium ion deposition rates were all highest for the natural salmon bone collagen calcium, followed by algae calcium. The eggshell powder, calcium citrate and calcium carbonate did not exhibit significant changes from control in most of these assays. The results of these assays suggest that natural marine collagen calcium such as salmon bone collagen calcium acts as the most effective intervention on osteoblast performance and actual calcium deposition. It should be recommended as a superior supplement for improved bone and joint health function.

Hyaluronate – parathyroid hormone peptide conjugate for transdermal treatment of osteoporosis

Human parathyroid hormone 1–34 fragment (PTH1–34) has been used as a FDA-approved therapeutics to treat osteoporosis by daily subcutaneous injection. In this work, we successfully developed PTH1–34 conjugated hyaluronic acid (HA) for the transdermal treatment of osteoporosis with improved patient compliance. HA-PTH1–34 conjugate was synthesized by the coupling reaction between aldehyde group introduced to HA and amine group of PTH1–34. After characterization by gel permeation chromatography (GPC) and ELISA, the biological effect of HA-PTH1–34 conjugate on the proliferation of human osteoblast cells was confirmed by in vitro calcium colorimetric assay and cAMP assay. Two-photon microscopy clearly visualized the effective skin penetration of FITC modified HA-PTH1–34 conjugate. The transdermally delivered HA-PTH1–34 conjugates elevated the blood calcium concentration for more than 2 days, reflecting the feasibility for the treatment of osteoporosis.

Bioactivity, cytocompatibility and thermal properties of experimental Bioglass-reinforced composites as potential root-canal filling materials

To evaluate the bioactivity and the cytocompatibility of experimental Bioglass-reinforced polyethylene-based root-canal filling materials. The thermal properties of the experimental materials were also evaluated using differential scanning calorimetry, while their radiopacity was assessed using a grey-scale value (GSV) aluminium step wedge and a phosphor plate digital system.Bioglass 45S5 (BAG), polyethylene and Strontium oxide (SrO) were used to create tailored composite fibres. The filler distribution within the composites was assessed using SEM, while their bioactivity was evaluated through infrared spectroscopy (FTIR) after storage in simulated body fluid (SBF). The radiopacity of the composite fibres and their thermal properties were determined using differential scanning calorimetry (DSC). The cytocompatibility of the experimental composites used in this study was assessed using human osteoblasts and statistically analysed using the Pairwise t-test (p<0.05).Bioglass and SrO fillers were well distributed within the resin matrix and increased both the thermal properties and the radiopacity of the polyethylene matrix. The FTIR showed a clear formation of calcium-phosphates, while, MTT and AlamrBlue tests demonstrated no deleterious effects on the metabolic activity of the osteoblast-like cells.BAG-reinforced polyethylene composites may be suitable as obturation materials for endodontic treatment. Since their low melting temperature, such innovative composites may be easily removed in case of root canal retreatment. Moreover, their biocompatibility and bioactivity may benefit proliferation of human osteoblast cells at the periapical area of the root.

Phenolic Compounds in Extra Virgin Olive Oil Stimulate Human Osteoblastic Cell Proliferation.

In this study, we aimed to clarify the effects of phenolic compounds and extracts from different extra virgin olive oil (EVOO) varieties obtained from fruits of different ripening stages on osteoblast cells (MG-63) proliferation. Cell proliferation was increased by hydroxytyrosol, luteolin, apigenin, p-coumaric, caffeic, and ferulic acids by approximately 11–16%, as compared with controls that were treated with one vehicle alone, while (+)-pinoresinol, oleuropein, sinapic, vanillic acid and derivative (vanillin) did not affect cell proliferation. All phenolic extracts stimulated MG-63 cell growth, and they induced higher cell proliferation rates than individual compounds. The most effective EVOO phenolic extracts were those obtained from the Picual variety, as they significantly increased cell proliferation by 18–22%. Conversely, Arbequina phenolic extracts increased cell proliferation by 9–13%. A decline in osteoblast proliferation was observed in oils obtained from olive fruits collected at the end of the harvest period, as their total phenolic content decreases at this late stage. Further research on the signaling pathways of olive oil phenolic compounds involved in the processes and their metabolism should be carried out to develop new interventions and adjuvant therapies using EVOO for bone health (i.e.osteoporosis) in adulthood and the elderly.

Electrophoretic deposition of calcium silicate–reduced graphene oxide composites on titanium substrate

Calcium silicate (CS)/graphene coatings have been used to improve the biological and mechanical fixation of metallic prosthesis. Among the extraordinary features of graphene is its very high mechanical strength, which makes it an attractive nanoreinforcement material for composites. Calcium silicate–reduced graphene oxide (CS–rGO) composites were synthesized, using an in situ hydrothermal method. CS nanowires were uniformly decorated on the rGO, with an appropriate interfacial bonding. The CS–rGO composites behaved like hybrid composites when deposited on a titanium substrate by cathodic electrophoretic deposition (EPD). Compared to a pure CS coating on Ti, the CS–1wt% rGO coating has improved adhesion by 70%, hardness by 150% and the elastic modulus by 240%. The CS–rGO composite coatings exhibit good apatite-forming ability in simulated body fluid (SBF). Moreover, the effect of addition of rGO on morphology, adhesion and proliferation of human osteoblast cells (hFOB) was investigated in vitro.

Bio-inspired mineralization of hydroxyapatite in 3D silk fibroin hydrogel for bone tissue engineering

To fabricate hard tissue implants with bone-like structure using a biomimetic mineralization method is drawing much more attentions in bone tissue engineering. The present work focuses in designing 3D silk fibroin hydrogel to modulate the nucleation and growth of hydroxyapatite crystals via a simple ion diffusion method. The study indicates that Ca2+ incorporation within the hydrogel provides the nucleation sites for hydroxyapatite crystals and subsequently regulates their oriented growth. The mineralization process is regulated in a Ca2+ concentration- and minerlization time-dependent way. Further, the compressive strength of the mineralized hydrogels is directly proportional with the mineral content in hydrogel. The orchestrated organic/inorganic composite supports well the viability and proliferation of human osteoblast cells; improved cyto-compatibility with increased mineral content. Together, the present investigation reports a simple and biomimetic process to fabricate 3D bone-like biomaterial with desired efficacy to repair bone defects.

Regulation of proliferation and apoptosis in human osteoblastic cells by microRNA-15b

MicroRNAs (miRNAs) play an important role in proliferation and differentiation of osteoblasts. We recently reported that miR-15b acts as a positive regulator of osteoblast differentiation, whereas its functional role in osteoblastic proliferation remains not known. In this study, we found that there was increased proliferation of human osteoblastic cells (MG63) when they were transiently transfected with miR-15b inhibitor. A significant level of cell population was found to be decreased at G0/G1 phase, and increased at S and G2/M phase by miR-15b inhibitor treatment. Cyclin E1 was found to be one of the putative target genes of miR-15b, and miR-15b mimic and miR-15b inhibitor treatments in cells decreased and increased cyclin E1 expression, respectively. We further identified that the cyclin E1 3′UTR is directly targeted by miR-15b using the luciferase reporter gene system. No significant effect was found on apoptosis of MG63 cells with miR-15b inhibitor. Thus, these findings provide new insights in understanding the role of miR-15b expression as negative regulator of osteoblast proliferation.

Mechanical and in vitro biological performance of graphene nanoplatelets reinforced calcium silicate composite.

Calcium silicate (CaSiO3, CS) ceramic composites reinforced with graphene nanoplatelets (GNP) were prepared using hot isostatic pressing (HIP) at 1150°C. Quantitative microstructural analysis suggests that GNP play a role in grain size and is responsible for the improved densification. Raman spectroscopy and scanning electron microscopy showed that GNP survived the harsh processing conditions of the selected HIP processing parameters. The uniform distribution of 1 wt.% GNP in the CS matrix, high densification and fine CS grain size help to improve the fracture toughness by ∼130%, hardness by ∼30% and brittleness index by ∼40% as compared to the CS matrix without GNP. The toughening mechanisms, such as crack bridging, pull-out, branching and deflection induced by GNP are observed and discussed. The GNP/CS composites exhibit good apatite-forming ability in the simulated body fluid (SBF). Our results indicate that the addition of GNP decreased pH value in SBF. Effect of addition of GNP on early adhesion and proliferation of human osteoblast cells (hFOB) was measured in vitro. The GNP/CS composites showed good biocompatibility and promoted cell viability and cell proliferation. The results indicated that the cell viability and proliferation are affected by time and concentration of GNP in the CS matrix.

Phenolic content of Sicilian virgin olive oils and their effect on MG-63 human osteoblastic cell proliferation

The aim of this study was, first, to investigate the influence of olive variety and elevation of orchards on the phenolic compound content of Sicilian virgin olive oils (VOOs) and, second, to investigate the effects of VOO phenolic extracts on osteoblast cell growth using the human MG-63 osteosarcoma cell line. Olive oil phenolic content and its effect on human osteosarcoma cell proliferation varied according to the type of cultivar and the grove altitude. This variation was also observed within the same type of cultivar. This observation demonstrates that the cultivar and the grove location can significantly affect the chemical composition and bioactivity of virgin olive oil. Although this study supports the hypothesis that virgin olive oil phenolic fractions exert a beneficial effect on bone health, further studies assessing the in vivo accessibility of virgin olive oil phenolic compounds to osteoblast cells should be carried out.

Hydrothermal preparation and characterization of ultralong strontium-substituted hydroxyapatite whiskers using acetamide as homogeneous precipitation reagent.

The ultralong strontium- (Sr-) substituted hydroxyapatite (SrHAp) whiskers were successfully prepared using acetamide as homogeneous precipitation reagent. The effect of the Sr substitution amount on the lattice constants and proliferation of human osteoblast cells (MG-63) was further investigated. The results showed that the SrHAp whiskers with diameter of 0.2–12 μm and ultralong length up to 200 μm were obtained and the Sr substitution level could be facilely tailored by regulating the initial molar ratio of Sr/(Sr + Ca) in raw materials. The Sr2+ replaced part of Ca2+ and the lattice constants increased apparently with the increase of the Sr substitution amount. Compared with the pure HAp whiskers, the Sr substitution apparently stimulated the proliferation of MG-63 at certain extracted concentrations. Our study suggested that the obtained SrHAp whiskers might be used as bioactive and mechanical reinforcement materials for hard tissue regeneration applications.

Biomimetic mineralization on a macroporous cellulose-based matrix for bone regeneration.

The aim of this study is to investigate the biomimetic mineralization on a cellulose-based porous matrix with an improved biological profile. The cellulose matrix was precalcified using three methods: (i) cellulose samples were treated with a solution of calcium chloride and diammonium hydrogen phosphate; (ii) the carboxymethylated cellulose matrix was stored in a saturated calcium hydroxide solution; (iii) the cellulose matrix was mixed with a calcium silicate solution in order to introduce silanol groups and to combine them with calcium ions. All the methods resulted in a mineralization of the cellulose surfaces after immersion in a simulated body fluid solution. Over a period of 14 days, the matrix was completely covered with hydroxyapatite crystals. Hydroxyapatite formation depended on functional groups on the matrix surface as well as on the precalcification method. The largest hydroxyapatite crystals were obtained on the carboxymethylated cellulose matrix treated with calcium hydroxide solution. The porous cellulose matrix was not cytotoxic, allowing the adhesion and proliferation of human osteoblastic cells. Comparatively, improved cell adhesion and growth rate were achieved on the mineralized cellulose matrices.

Osteoblast viability on hydroxyapatite with well-adjusted submicron and micron surface roughness as monitored by the proliferation reagent WST-1

The impact of the cell surface roughness on titanium alloys used for biomedical implants has been extensively studied, whereas the dependency of human osteoblast viability on hydroxyapatite (HA) submicron and micron surface roughness has hitherto not yet been investigated in detail. Therefore, we investigate in this study the effect of HA substrates with different well-adjusted surface roughness on human osteoblast proliferation using the standard colorimetric reagent WST-1. By grinding, we obtained HA surfaces with six levels of well-defined surface roughness ranging from Sa = 3.36 µm down to 0.13 µm, resulting in hydrophilic contact angles from 11° to 27°. Energy dispersive X-ray spectroscopy, X-ray diffraction, and X-ray fluorescence measurements confirmed that neither grinding paper residues nor changes of the crystal structure were introduced to the HA substrates by the grinding process. By applying this simple surface treatment, we were able to isolate other effects from surface chemistry, crystal structure, and relative density. The changes of the osteoblast proliferation (WST-1 assay) on these different roughened HA surfaces after 7 days were found to be insignificant (p > 0.05), evaluated by one-way analysis of variance and Tuckey's Multiple Comparison Method. The results of this study show that all roughened HA surfaces, regardless of the microtopography, are biocompatible and allow osteoblast attachment, proliferation, and collagen type I production. The comparison with surface roughness used for standard Ti-based implants yielded that for HA no finishing process is necessary to ensure a sound human osteoblast cell proliferation in vitro.

Comparison of platelet rich fibrin and collagen as osteoblast‐seeded scaffolds for bone tissue engineering applications

The loss of jaw bone caused by different kinds of pathologies leads to dysfunction and reduced quality of life in affected patients. Thus, the pivotal goal in bone tissue engineering is to reconstruct these defects. The essential precondition for new tissue generation is an extracellular matrix which acts as a scaffold so that cells can migrate, differentiate, and proliferate. Fibrin, a biopolymer responsible for blood clot formation, has been shown to be suitable for tissue engineering applications. The aim of the present study is a comparison of platelet rich fibrin (PRF) with the commonly used collagen membrane BioGide(®) as a scaffold for human osteoblast cell seeding for bone tissue engineering. Human osteoblasts were cultured with eluates from PRF (n=7) and BioGide(®) (n=8) membranes incubated in serum-free cell culture medium. Vitality of these cells was assessed by fluorescein diacetate and propidium iodide staining, biocompatibility with the lactate dehydrogenase test and proliferation levels with the MTT ([3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium-bromide]), and BrdU (5-bromo-2-deoxyuridine) tests. In addition, human osteoblasts were seeded on both membrane systems and cell growth was compared by the water soluble tetrazolium (WST-1) (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulfonate) test and scanning electron microscopy (SEM). Osteoblastic differentiation was assessed by alkaline phosphatase activity measured by ELISA in the supernatant of osteoblasts cultivated on PRF membranes (n=10), PRF clots (n=10), and BioGide(®) membranes (n=10). Lactate dehydrogenase test values were higher for PRF compared to BioGide(®) . The BrdU test showed superior cell growth after cultivation in eluate from PRF than in eluate from BioGide(®) . The WST-1 assay demonstrated superior cell proliferation on PRF than on BioGide(®) . SEM revealed osteoblast colonization of both membranes. Cultivation of osteoblasts on PRF membranes and PRF clots showed significantly higher alkaline phosphatase activity than on BioGide(®) membranes. Metabolic activity and proliferation of human osteoblast cells in vitro were supported to a significant higher extent by eluates from PRF membranes. Both membranes are suitable as scaffolds for cultivation of human osteoblast cells in vitro; proliferation was significant higher on PRF membranes and on PRF clot than on BioGide(®) membranes.

Electrohydrodynamic deposition of nanotitanium doped hydroxyapatite coating for medical and dental applications

Nano-sized titanium containing hydroxyaptite has been prepared, the particle size of nanoTiHA was shown to be 12-20 nm in width and 30-40 nm in length, smaller than that of nanoHA. X-ray diffraction analysis revealed the phase purity of nanoTiHA produced. Antimicrobical assays demonstrated that nanoTiHA has excellent growth inhibitory properties, and is able to inhibit the growth of all bacterial strains tested, both Gram-negative and Gram-positive species, including multi-antibiotic resistant EMRSA 15 and EMRSA 16 'superbugs'. Biocidal activity against all four Staphylococcus spp was also shown at the concentration tested. Nanostuctured TiHA coating was successfully deposited onto Ti surfaces using EHDA spraying under optimized processing conditions with the thickness of the coating being further controlled by the spraying time. All of the nanoTiHA coated Ti surfaces were able to support human osteoblast (HOB) cell attachment and growth. The coating thickness did not significantly influence the proliferation of HOB cells on nanoTiHA coatings, while the ability of nanoTiHA coating to support HOB cell differentiation was demonstrated from the alkaline phosphatase activity. Our study showed that nanoTiHA has excellent anti-bacterial properties and the thin nanoTiHA coating was also able to support the attachment, growth and differentiation of HOB cells. Therefore, nanoTiHA coating could pave the way for the development of the next generation of dental and orthopedic implants by offering anti-infection potential in addition to osteoconductivity.

Electrospinning, characterization and in vitro biological evaluation of nanocomposite fibers containing carbonated hydroxyapatite nanoparticles

Poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) fibers containing carbonated hydroxyapatite (CHA) nanoparticles with different CHA amounts (5, 10 and 15 wt%) were electrospun with the aid of ultrasonic power for dispersing the nanoparticles. Scanning electron microscopy and energy-dispersive x-ray spectroscopy results showed that the distribution of CHA within the CHA/PHBV nanocomposite fibers was homogeneous when the CHA content was 10 wt%. Slight particle agglomeration occurred when the CHA content was 15 wt%. The diameters of the electrospun CHA/PHBV nanocomposite fibers and PHBV polymer fibers were around 3 µm. Fourier transform infrared spectroscopic analysis further confirmed the presence of CHA in CHA/PHBV nanocomposite fibers. Both PHBV and CHA/PHBV fibrous membranes exhibited similar tensile properties. Compared with PHBV solvent-cast film, the PHBV fibrous membrane was hydrophobic but the incorporation of CHA nanoparticles dramatically enhanced its wettability. In vitro studies revealed that both types of electrospun fibrous membranes (PHBV and CHA/PHBV) supported the proliferation of human osteoblastic cells (SaOS-2). The alkaline phosphatase activity of SaOS-2 cells seeded on the CHA/PHBV fibrous membranes was higher than that of the cells seeded on the PHBV fibrous membranes after 14 days of cell culture. The electrospun CHA/PHBV nanocomposite fibrous membranes show promises for bone tissue engineering applications.

Preparation, microstructures, mechanical properties, and cytocompatibility of TiMn alloys for biomedical applications

The titanium-manganese (TiMn) alloys have been extensively used in aerospace and hydrogen storage. In this study, the TiMn alloys with various manganese contents ranging from 2 to 12 wt % were prepared by using mechanical alloying and spark plasma sintering (SPS) techniques. The microstructures, mechanical properties including hardness, elastic modulus and ductility, cytotoxicity and cell proliferation properties of the TiMn alloys were investigated to explore their biomedical applications. The addition of manganese to the titanium reduced the alpha to beta transformation temperature and was confirmed as a beta stabilizer element. The manganese increased the relative density of the alloy and thus high density TiMn alloys with alpha+beta structure were prepared by using SPS at 700 degrees C. The hardness increased significantly ranging from 2.4 GPa (Ti2Mn) to 5.28 GPa (Ti12Mn) and the elastic modulus ranging from 83.3 GPa (Ti2Mn) to 122 GPa (Ti12Mn), the ductility decreased ranging from 21.3% (Ti2Mn) to 11.7% (Ti12Mn) with increasing manganese content in the Ti. Concentrations of Mn below 8 wt % in titanium reveal negligible effects on the metabolic activity and the cell proliferation of human osteoblasts. The Mn could be used in lower concentrations as an alloying element for biomedical titanium. The Ti2Mn, Ti5Mn, and Ti8Mn alloys with supervisor mechanical properties and acceptable cytocompatibility have a potential for use as bone substitutes and dental implants.
