New high-density fermentation method for producing high molecular weight polysialic acid based on the combination fermentation strategy.

Polysialic acid (PSA) is a long-chain linear amino polysaccharide with broad application prospects; however, its relatively low molecular weight limits its application range. This study aims to explore a new fermentation method of combining the three-phase pH control strategy, three-phase mixing speed control strategy, and exogenous substance to produce high molecular weight PSA. In brief, Escherichia coli K235 6E61 (CCTCC M208088) was used as a fermentation strain. 3g·L-1 Na5P3O10 was added to the initial medium. At 0-12h, the mixing speed was controlled to 250 r·min-1, and the pH was maintained at 7.2. At 12-20h, the mixing speed was increased to 400 r·min-1, the pH was changed to 6.8, and 0.75% n-hexadecane was added at hour 16. After 20h, the mixing speed was adjusted to 250 r·min-1; the pH was restored to 7.2. Air flow was regulated to 1.2 vvm throughout the experiment. The combination fermentation strategy greatly improved the molecular weight of PSA up to 498kDa at 32h, which is currently the maximum molecular weight of PSA produced through microbial fermentation. The yield of PSA reached 6.27g·L-1 at the end of fermentation (36h), which is also currently the highest yield of PSA produced by natural bacteria. Therefore, the proposed strategy could simultaneously increase the molecular weight and yield of PSA and is of great importance to the industrial production of high molecular weight PSA. Bioproduction, purification, and application of polysialic acid.

Polysialic acid (PSA) is a negatively charged linear homopolymer linked by N-acetylneuraminic acid and widely present in vertebrates and some pathogens. PSA, commonly found on cell surfaces as glycoproteins and glycolipids, plays important roles in intercellular adhesion, cell migration, and formation and remodeling of the neural system by regulating the adhesive property of nerve cell adhesion molecules. PSA with a molecular weight that can reach as high as 260kDa also belongs to the group II capsule polysaccharide of neonatal meningitis-causing Escherichia coli K1. To date, much effort has been devoted to developing the biotechnological production of PSA. As a non-glycosaminoglycan, PSA is a non-immunogenic and biodegradable polysaccharide that can be used as a biomaterial in protein polysialylation, tissue engineering, and drug delivery. PSA can also combine with other macromolecules to form multifunctional composites. In this mini-review, the production, purification, and application of PSA are summarized to provide a basis for further PSA applications.

Polysialic acid biosynthesis and production in Escherichia coli: current state and perspectives.

Polysialic acid (PSA) is a unique polysaccharide that plays critical roles in many bioprocesses, which makes it useful in a wide range of biomedical applications. The increased demand for PSA has led to considerable efforts to improve its production using bacteria, such as Escherichia coli. Bioprocess optimization and metabolic engineering have allowed the efficient production of PSA. This review aims to summarize the metabolism of PSA with an emphasis on the importance of the key pathway components. In addition, this review provides an update on state of the art PSA production using E. coli with a special emphasis on strategies of strain engineering and process development for the enhanced production of PSA.

Enhanced production of polysialic acid by metabolic engineering of Escherichia coli.

A number of reports have described the production of polysialic acid (PSA), focusing on the fermentation and purification of PSA. However, little work has been done to strengthen the synthetic pathway of PSA to improve PSA production. In this study, an efficient process for enhanced production of PSA using a recombinant Escherichia coli strain was developed. To improve the PSA production efficiency, the key enzymes of PSA synthetic pathway were overexpressed separately or in combination, including N-acetylneuraminate (Neu5Ac) 7-O(or 9-O)-acetyltransferase (NeuD), CMP-Neu5Ac synthetase (NeuA), and alpha-Neu5Ac alpha-2,8-sialyltransferase (NeuS). The PSA production was significantly improved by coexpression of NeuD and NeuA. In terms of the efficiency, NeuD was considered as the most important factor. Secondly, the competing pathway of intermediate Neu5Ac was blocked by nanA deletion. The efficient PSA-producing strain E. coli SA9ΔnanA/pDB1S-DA was constructed, and 16.15 ± 1.45 g/L PSA was obtained in the fed-batch culture. The production of PSA by engineered strain was increased by 85 % compared to the original strain. These results provide evidence for improvement of PSA production by regulation of the PSA biosynthetic pathway. The high productivity of our process should make it a promising cost-effective resource for PSA.

A new polysialic acid production process based on dual-stage pH control and fed-batch fermentation for higher yield and resulting high molecular weight product

To determine the factors influencing the resulting molecular weight of polysialic acid (PSA), batch fermentations by using Escherichia coli were conducted. It was found that temperature and pH were significant factors affecting the PSA production and its resulting molecular weight. When pH was set at 6.4, temperature of 37 °C was suitable for cell growth and PSA production while 33 °C facilitated production of higher molecular weight of PSA. pH 6.4 was favorable for PSA production while pH 7.4 was good for higher molecular weight of PSA at 37 °C. Intramolecular self-cleavage of PSA might lead to relatively low molecular weight under mild acidic condition. Our data suggest that the PSA molecular weight is significantly affected by the pH condition rather than the temperature. It is concluded that the resulting PSA molecular weight not only depends on fermentation conditions but also relates to cell growth rate and PSA production rate. Higher PSA molecular weight was made when its production rate was faster than degradation rate. A novel two-stage pH control fermentation process for production of high molecular weight PSA was developed. At the first stage, pH was set at 6.4 to encourage cell growth and PSA production, whereas pH was set at 7.4 at the second stage to promote the formation of higher molecular weight PSA. PSA yield up to 5.65 g/L and its resulting molecular weight of 260 kDa was attained, the highest level ever reported.

Comparison of polysialic acid production in Escherichia coli K1 during batch cultivation and fed-batch cultivation applying two different control strategies

The polysialic acid (PSA) production in Escherichia coli ( E. coli) K1 was studied using three different cultivation strategies. A batch cultivation, a fed-batch cultivation at a constant specific growth rate of 0.25 h −1 and a fed-batch cultivation at a constant glucose concentration of 50 mg l −1 was performed. PSA formation kinetics under different cultivation strategies were analyzed based on the Monod growth model and the Luedeking–Piret equation. The results revealed that PSA formation in E. coli K1 was completely growth associated, the highest specific PSA formation rate (0.0489 g g −1 h −1) was obtained in the batch cultivation. However, comparing biomass and PSA yields on the glucose consumed, both fed-batch cultivations provided higher yields than that of the batch cultivation and acetate formation was prevented. Moreover, PSA yield on glucose was also correlated to the specific growth rate of the cells. The optimal specific growth rate for PSA production was 0.32 h −1 obtained in the fed-batch cultivation at a constant glucose concentration of 50 mg l −1, with highest conversion efficiency of 43 mg g −1.

Enhancement of polysialic acid yield by reducing initial phosphate and feeding ammonia water to Escherichia coli CCTCC M208088

Polysialic acid (PSA) is a capsular polysaccharide obtained from aerobic fermentation with Escherichia coli. To enhance PSA production and eliminate the influence of phosphate on the PSA purification process, a lower level of initial phosphate was adopted with pH control. The resulting PSA yield reached 4.1 g/L in fed-batch fermentation with 2.5 g/L K2HPO4 and E. coli strain CCTCC M208088. In addition, an ammonia water (NH4OH) feeding strategy to control the pH at 6.4 was developed resulting in PSA production that reached as high as 5.2 g/L. NMR spectra confirmed the purified biopolymer as a α-2,8 linked PSA, identical to the published NMR spectra, with a molecular weight in the range of 16 ∼ 50 kDa.

A new strategy to enhance polysialic acid production by controlling sorbitol concentration in cultivation of Escherichia coli K235.

Polysialic acid (PSA) is a new pharmaceutical material used in control release of protein drugs and as scaffold material in biomedical applications. It is also a vital source of sialic acid and its derivatives. In this paper, we demonstrated that the substrate sorbitol has significant effect on bacterial growth and PSA formation in cultivation of Escherichia coli K235. Lower sorbitol concentration favored bacterial growth but retards PSA formation in the fed-batch fermentation. Higher sorbitol concentration stimulated PSA formation but inhibited bacterial growth in the batch fermentation. Sorbitol concentration gradient experiment revealed that PSA formation was seriously limited as sorbitol concentration was lower than 20 g/L, while bacterial growth was obviously inhibited as the sorbitol concentration higher than 40 g/L. A new strategy of controling sorbitol concentration in a range of 20 - 40 g/L in the broth during the fermentation process increased PSA yield by 20%.

YdgG (TqsA) Controls Biofilm Formation inEscherichia coliK-12 through Autoinducer 2 Transport

YdgG is an uncharacterized protein that is induced in Escherichia coli biofilms. Here it is shown that deletion of ydgG decreased extracellular and increased intracellular concentrations of autoinducer 2 (AI-2); hence, YdgG enhances transport of AI-2. Consistent with this hypothesis, deletion of ydgG resulted in a 7,000-fold increase in biofilm thickness and 574-fold increase in biomass in flow cells. Also consistent with the hypothesis, deletion of ydgG increased cell motility by increasing transcription of flagellar genes (genes induced by AI-2). By expressing ydgG in trans, the wild-type phenotypes for extracellular AI-2 activity, motility, and biofilm formation were restored. YdgG is also predicted to be a membrane-spanning protein that is conserved in many bacteria, and it influences resistance to several antimicrobials, including crystal violet and streptomycin (this phenotype could also be complemented). Deletion of ydgG also caused 31% of the bacterial chromosome to be differentially expressed in biofilms, as expected, since AI-2 controls hundreds of genes. YdgG was found to negatively modulate expression of flagellum- and motility-related genes, as well as other known products essential for biofilm formation, including operons for type 1 fimbriae, autotransporter protein Ag43, curli production, colanic acid production, and production of polysaccharide adhesin. Eighty genes not previously related to biofilm formation were also identified, including those that encode transport proteins (yihN and yihP), polysialic acid production (gutM and gutQ), CP4-57 prophage functions (yfjR and alpA), methionine biosynthesis (metR), biotin and thiamine biosynthesis (bioF and thiDFH), anaerobic metabolism (focB, hyfACDR, ttdA, and fumB), and proteins with unknown function (ybfG, yceO, yjhQ, and yjbE); 10 of these genes were verified through mutation to decrease biofilm formation by 40% or more (yfjR, bioF, yccW, yjbE, yceO, ttdA, fumB, yjiP, gutQ, and yihR). Hence, it appears YdgG controls the transport of the quorum-sensing signal AI-2, and so we suggest the gene name tqsA.

Transport of N-acetyl- d-galactosamine in Escherichia coli K92: effect on acetyl-amino sugar metabolism and polysialic acid production

The N-acetyl- d-galactosamine (GalNAc) transport system of Escherichia coli K92 was studied when the bacterium was grown in a chemically defined medium containing GalNAc as a carbon source. Kinetic measurements were carried out in vivo at 37 °C in 25 mM phosphate buffer, pH 7.0. Under these conditions, the uptake rate was linear for at least 3 min and the calculated K m for GalNAc was 3 μM. The transport system was strongly inhibited by sodium arsenate (70%), potassium cyanide (62%) and 2,4-dinitrophenol (75%). Analysis of bacterial GalNAc phosphotransferase activity revealed in vitro GalNAc phosphorylation activity only when phosphoenolpyruvate was present. These results strongly support the notion that GalNAc uptake depends on a specific phosphotransferase system. Study of activity regulation showed that N-acetylglucosamine and mannosamine specifically inhibit the transport of GalNAc in this bacterium. Analysis of expression revealed that the GalNAc transport system is specifically induced by GalNAc but not by N-acetylglucosamine (GlcNAc) or N-acetylmannosamine (ManNAc), two intimately related sugars. Moreover, full induction of GalNAc transport required the presence of both cAMP and GalNAc. Comparative studies revealed that E. coli K92 has developed a regulation mechanism that specifically induces the appropriate permease based on the presence of each respective phospho-amino sugar (GlcNAc, ManNAc and GalNAc). In this regulation system, GlcNAc is the preferred amino sugar as the carbon source. Finally, when E. coli K92 was grown using GalNAc, capsular polysialic acid production was strongly affected. The presence of intracellular phosphoderivative acetylamino sugars, generated by the action of the phosphotransferase transport system, can be responsible for this effect.

Sialyltransferase ST8Sia-II Assembles a Subset of Polysialic Acid That Directs Hippocampal Axonal Targeting and Promotes Fear Behavior

Polysialic acid (PSA) is a post-translational protein modification that is widely expressed among neural cell types during development. Found predominantly on the neural cell adhesion molecule (NCAM), PSA becomes restricted to regions of neurogenesis and neuroplasticity in the adult. In the mammalian genome, two polysialyltransferases termed ST8Sia-II and ST8Sia-IV have been hypothesized to be responsible for the production of PSA in vivo. Approaches to discover PSA function have involved the application of endoneuraminidase-N to remove PSA and genetic manipulations in the mouse to deplete either NCAM or ST8Sia-IV. Here we report the production and characterization of mice deficient in the ST8Sia-II polysialyltransferase. We observed alterations in brain PSA expression unlike those observed in mice lacking ST8Sia-IV. This included a PSA deficit in regions of neurogenesis but without changes in the frequency of mitotic neural progenitor cells. In further contrast with ST8Sia-IV deficiency, loss of ST8Sia-II did not impair hippocampal synaptic plasticity but instead resulted in the misguidance of infrapyramidal mossy fibers and the formation of ectopic synapses in the hippocampus. Consistent with studies of animal models bearing these morphological changes, ST8Sia-II-deficient mice exhibited higher exploratory drive and reduced behavioral responses to Pavlovian fear conditioning. PSA produced by the ST8Sia-II polysialyltransferase modifies memory and behavior processes that are distinct from the neural roles reported for ST8Sia-IV. This genetic partitioning of PSA formation engenders discrete neurological processes and reveals that this post-translational modification forms the predominant basis for the multiple functions attributed to the NCAM glycoprotein.

Production of polysialic acid from fed-batch fermentation with pH control

During batch fermentation pH affects the production of biomass and polysialic acid in Escherichia coli K235-WXJ4. When the pH of the medium was maintained at 6.4 during the stationary phase, polysialic acid production was prolonged and the specific rate of polysialic acid biosynthesis was higher than when the fermentation process was not controlled for pH. Controlling the pH during fed-batch fermentation of polysialic acid increased the yield of dry cell weight and polysialic acid, which reached 10.16 and 2606 g/l, respectively.

Transport of N-acetyl- D-mannosamine and N-acetyl- D-glucosamine in Escherichia coli K1: effect on capsular polysialic acid production

N-Acetyl- D-mannosamine (ManNAc) and N-acetyl- D-glucosamine (GlcNAc) are the essential precursors of N-acetylneuraminic acid (NeuAc), the specific monomer of polysialic acid (PA), a bacterial pathogenic determinant. Escherichia coli K1 uses both amino sugars as carbon sources and uptake takes place through the mannose phosphotransferase system transporter, a phosphoenolpyruvate-dependent phosphotransferase system that shows a broad range of specificity. Glucose, mannose, fructose, and glucosamine strongly inhibited the transport of these amino-acetylated sugars and GlcNAc and ManNAc strongly affected ManNAc and GlcNAc uptake, respectively. The ManNAc and the GlcNAc phosphorylation that occurs during uptake affected NeuAc synthesis in vitro. These findings account for the low in vivo PA production observed when E. coli K1 uses ManNAc or GlcNAc as a carbon source for growth.

Pathogenesis of enteric Campylobacter infection.

Campylobacter jejuni and related species are important human pathogens, causing acute human enterocolitis, and they are the most common cause of food-borne diarrhoea in many industrialized countries. Previous infection with certain strains of C. jejuni is also linked with the development of the neurological disorder Guillain-Barre syndrome (GBS). Despite its importance as a human pathogen, relatively little is understood of the mechanisms of C. jejuni-associated disease. The recent release of the complete genome sequence of C. jejuni strain NCTC 11168, together with new strategies for random and directed mutagenesis, have allowed a better insight into some of the genetic determinants of C. jejuni virulence. In this review current knowledge on the pathogenesis of C. jejuni infection is summarized, and possible directions of future research indicated.

Regulation of capsular polysialic acid biosynthesis by N-acetyl-d-mannosamine, an intermediate of sialic acid metabolism

N-Acetyl-d-mannosamine (ManNAc) is a specific substrate for the synthesis of N-acetylneuraminic acid, the essential precursor of bacterial capsular polysialic acid (PA). When Escherichia coli K92 used ManNAc as a carbon source, we observed a dramatic reduction (up to 90%) in in vivo PA production. Experiments in which the carbon source was changed revealed that the maximal inhibitory effect occurred when this sugar was present in the medium before the logarithmic phase of bacterial growth had started. Enzymatic analysis revealed that high concentrations of ManNAc-6-phosphate inhibit NeuAc lyase, the enzyme that synthesizes NeuAc for PA biosynthesis in E. coli. These results indicate that ManNAc-6-phosphate is able to regulate NeuAc lyase activity and modulate the PA synthesis.

Biochemical conditions for the production of polysialic acid by Pasteurella haemolytica A2.

The capsular polysaccharide of Pasteurella haemolytica A2 consists of a linear polymer of N-acetylneuraminic acid (Neu5Ac) with alpha(2-8) linkages. When the bacterium was grown at 37 degrees C for 90 h in 250 ml shake flasks at 200 rpm in Brain heart infusion broth (BHIB), it accumulated, attaining a level of 60 microg/ml. Release of this polymer was strictly regulated by the growth temperature, and above 40 degrees no production was detected. The pathway for the biosynthesis of this sialic acid capsular polymer was also examined in P. haemolytica A2 and was seen to involve the sequential presence of three enzymatic activities: Neu5Ac lyase activity, which synthesizes Neu5Ac by condensation of Nacetyl-D-mannosamine and pyruvate with apparent Km values of 91 mM and 73 mM, respectively; a CMP-Neu5Ac synthetase, which catalyzes the production of CMP-Neu5Ac from Neu5Ac and CTP with apparent Km values of 2 mM and 0.5 mM, respectively, and finally a membrane-associated polysialyltransferase, which catalyzes the incorporation of sialic acid from CMP-Neu5Ac into polymeric products with an apparent CMP-Neu5Ac Km of 250 microM.

High Production of Polysialic Acid [Neu5Acα(2-8)-Neu5Acα(2-9)]nbyEscherichia coliK92 Grown in a Chemically Defined Medium. Regulation by Temperature

The capsular polysaccharide of Escherichia coli K92 consists of a linear polymer of Neu5Ac with alternating alpha(2-8) and alpha(2-9) linkages. It accumulates when the bacterium is grown at 37 degrees C in a defined medium containing D-xylose and L-asparagine as carbon and nitrogen sources. Release of the capsular polymer into the medium was maximal (450 micrograms x ml-1) in the stationary phase of growth (76 h). This medium could be useful for obtaining sufficient polymer to develop effective vaccines. The enzyme, CMP-Neu5Ac synthetase, was not detected in cells grown at 20 degrees C. The lack of this enzyme explains the absence of polymer biosynthesis when the bacterium was grown at 20 degrees C.
