A consortium of different Saccharomyces species enhances the content of bioactive tryptophan-derived compounds in wine fermentations

In recent years, the presence of molecules derived from aromatic amino acids in wines has been increasingly demonstrated to have a significant influence on wine quality and stability. In addition, interactions between different yeast species have been observed to influence these final properties. In this study, a screening of 81 yeast strains from different environments was carried out to establish a consortium that would promote the improvement of indolic compound levels in wine. Two strains, Saccharomyces uvarum and Saccharomyces eubayanus, with robust fermentative capacity were selected to be combined with a Saccharomyces cerevisiae strain with a predisposition towards the production of indolic compounds. Fermentation dynamics were studied in pure cultures, co-inoculations and sequential inoculations, analysing strain interactions and end-of-fermentation characteristics. Fermentations showing significant interactions were further analyzed for the resulting indolic compounds and aroma profile, with the aim of observing potential interactions and synergies resulting from the combination of different strains in the final wine. Sequential inoculation of S. cerevisiae after S. uvarum or S. eubayanus was observed to increase indolic compound levels, particularly serotonin and 3-indoleacetic acid. This study is the first to demonstrate how the formation of microbial consortia can serve as a useful strategy to enhance compounds with interesting properties in wine, paving the way for future studies and combinations.

Isolation and selection of microalgae capable of stimulating rice plant development and seed production

The growing global demand for food, in addition to the environmental impacts of agricultural intensification and climate change, reinforce the need for sustainable alternatives to maintain and increase agricultural productivity. Microalgae are widely available renewable resources for application in agricultural environments. There is a vast biodiversity of microalgae species, and their versatile metabolism produces a great diversity of compounds that favor the capture and supply of nutrients, improve soil characteristics, and stimulate plant growth. In this study, we evaluated the production of indole compounds, exopolysaccharides, siderophores, and the development capacity of microalgae strains of the genera Chlamydomonas sp., Chlorella sp. and Desmodesmus sp. isolated from different Brazilian States. Microalgae that produced a greater range of compounds and developed more quickly were selected for the production of aqueous extracts from their biomass in order to evaluate their potential as a biostimulant for rice (Oryza sativa L.) plants. Seed inoculation and foliar application of extracts stimulated plant development. Shoot length in seedlings 7 days after germination was increased after the application of Syn 16 aqueous extract at concentration of 0.5 g/L. Shoots of 30-day-old plants increased in length with the application of Syn 16 and Syn 90 strains, regardless of the tested concentration, while root length was increased by Syn 16 (at both concentrations - 0.1 and 0.5 g/L) and Syn 90 (0.1 g/L). Regarding shoot dry weight, increases of up to 36 % were detected with the Syn 16 strain at concentration of 0.5 g/L. With the Syn 90 strain at a concentration of 0.5 g/L, the increase was 27 %. Furthermore, root dry weight was 43 % and 35 % higher than control plants after the application of Syn 16 and Syn 90, respectively (both at 0.5 g/L concentration). At the maturity stage, application of Syn 46 strain (0.5 g/L concentration) increased 11 % the number of full seeds per panicle when compared to control condition. Our results demonstrate the ability of microalgae extracts in stimulating the development and seed production of rice plants.

Growth Promotion of Guava “Pear” (Psidium guajava cv.) by Sinorhizobium mexicanum in Southern Mexican Agricultural Fields

Biofertilizers formulated with nitrogen-fixing bacteria represent an alternative to chemical fertilizers because they increase soil fertility and protect the environment. Therefore, the objective of this study was to analyze the effects on the growth of guava “pear” (Psidium guajava cv.) after inoculation with a nitrogen fixing bacterium Sinorhizobium mexicanum ITTG-R7T. The study was carried out in an agricultural rural area of Chiapas, Mexico, where farmers do not have programs of regenerative agriculture. First, the agricultural soil was subjected to physicochemical and metagenomic analysis in order to determine the soil quality and its bacterial community composition. Likewise, multifunctional biochemical tests and plant inoculation assays were evaluated to determine the potential of S. mexicanum ITTG-R7T as plant-growth-promoting bacteria (PGPB). The site was rain fed and had silty clay loam soil with abundant Bacillaceae. S. mexicanum ITTG-R7T showed different properties as PGPB such as the production of indole compounds, synthesis of extracellular enzymes, phosphate solubilization, synthesis of siderophores, ACC (1-aminocyclopropane-1-carboxylate) deaminase, and nitrogenase activity (ARA). When the strain ITTG-R7 T was combined with chemical nutrients, it had the highest positive effect on the growth and development of guava plants. Guava biofertilization with ITTG-R7T had a significant influence (p < 0.05) mainly on the total plant height (368.83 cm), number of flowers (36.0) and the amount of chlorophyll (2.81 mg mL−1) in comparison with the other treatments evaluated. ITTG-R7T is a promising strain for improving the guava crop yield.

Beneficial traits of grain-residing endophytic communities in wheat with different sensitivity to Pseudomonas syringae

Deep insight into compositional and functional features of endophytic bacterial communities residing in wheat grains opens the way to the use of their plant growth promoting and biocontrol abilities in agricultural biotechnology. The aim of this work was to compare grain-residing endophytes from winter wheat varieties with different sensitivity to Pseudomonas syringae pv. atrofaciens (McCulloch) and to examine their plant-beneficial traits and antagonistic effects. Grain-residing bacteria were isolated from surface-sterilized grains of three wheat varieties sown in Ukraine following a culture-dependent protocol, and were screened for their plant growth promotion (PGP) and antagonistic properties. Bacterial morphotypes were represented by gram-negative rods, endospore-forming bacilli and gram-positive cocci. Different resistance to phytopathogenic pseudomonads was associated with distinctive quantitative and functional features of grain-residing endophytic communities. High resistance to P. syringae was coupled with the prevalence of gram-negative rods in the endophytic community, the highest proportion of endophytic bacteria possessing three PGP activities (phosphate solubilization, nitrogen fixation and production of indolic compounds) simultaneously, and with the most potent antagonistic activity of grain-residing endospore-forming bacilli. In total, five grain-residing isolates, which were obtained from three wheat varieties (two isolates from varieties with medium and high resistance and one – from a low-resistant variety), demonstrated ability to restrain P. syringae pv. atrofaciens (McCulloch) growth. Two isolates (P6 and P10) which were obtained from the high-resistant wheat variety Podolyanka and were assigned to Paenibacillus and Brevibacillus genera according to their biochemical profiling and MS-DS identification, showed the most potent antagonistic effects as indicated by maximum inhibition zone in agar well diffusion assay. These results shed light on the association of the features of grain-residing endophytic bacteria with wheat resistance to phytopathogenic pseudomonads. Isolates from the high-resistant wheat variety can be recommended for grain dressing as plant growth promoting and biocontrol agents for P. syringae pv. atrofaciens (McCulloch).

Streptomyces spp. enhance vegetative growth of maize plants under saline stress.

Saline stress is one of the abiotic stresses that most compromises the yield of crops and can be mitigated by plant growth-promoting rhizobacteria (PGPR). This work characterized rhizobacteria isolates from the genus Streptomyces as PGPR and evaluated their role on growth and alleviation of the effects caused by saline stress in maize (Zea mays L.). Production of indolic compounds (IC), siderophores, ACC deaminase, phenazines, and promotion of plant growth were determined to characterize bacterial isolates. Salinity tolerance was accessed by culturing the Streptomyces isolates under NaCl increasing concentrations (0-300 mM). Four Streptomyces isolates exhibiting PGPR traits and salinity tolerance were selected and their effect on tolerance of maize plants to saline stress was evaluated. Plants obtained from bacterized seeds and submitted to 100 and 300 mM NaCl were used. All Streptomyces spp. produced IC and siderophores, CLV178 being the best producer of these two compounds. ACC deaminase was detected in six of the 10 isolates (CLV95, CLV97, CLV127, CLV179, CLV193, and CLV205), while phenazines were found only in CLV186 and CLV194. All isolates were tolerant to salinity, growing at concentrations up to 300 mM NaCl, with exception of CLV188. Increased concentrations of IC were detected in most of the isolates exposed to salinity. CLV97 and CLV179 significantly promoted growth of roots and leaves of maize plants and attenuated the negative effects of salinity on plant growth. Root colonization by Streptomyces spp. was confirmed in plants cultivated 20 days under saline stress.

Phosphorus Nutrition and Growth of Cotton Plants Inoculated With Growth-Promoting Bacteria Under Low Phosphate Availability

The low availability of phosphorus (P) in the soil drastically limits the world productivity of crops such as cotton. In order to contribute sustainably to the solution of this problem, the current study aimed to evaluate the capacity of phosphate-solubilising bacteria to improve plant growth and its relationship with physiological parameters, as well as the shoot P content in cotton plants in a soil with low P availability amended with rock phosphate. The results showed that, of the six plant growth-promoting bacteria strains evaluated under greenhouse conditions, the Rhizobium strain B02 significantly promoted growth, shoot P content and photosynthetic rate. This strain also improved the transpiration rate and the relative content of chlorophyll but without significant differences. Remarkably, Rhizobium sp. B02 had a more significant effect on plant growth compared to the P nutrition. Furthermore, the effect of its inoculation was more pronounced on the roots' growth compared to the shoot. Finally, application of Rhizobium strain B02 showed the capacity to optimize the use of low-solubility fertilizer as the rock phosphate. These findings could be associated with the metabolic activities of plant growth promotion exhibited by phosphate-solubilising strains, such as phosphate solubilisation, production of indole compounds and siderophores synthesis. In conclusion, this research provides evidence of the biotechnological potential of the Rhizobium genus as phosphate-solubilising bacteria with multiple plant growth-promoting activities capable of improving the plant growth and phosphate nutrition of non-leguminous crops such as cotton in soil with low P availability amended with rock phosphate.

Potential of Herbaspirillum and Azospirillum Consortium to Promote Growth of Perennial Ryegrass under Water Deficit.

Plant growth-promoting bacteria (PGPB) can mitigate the effect of abiotic stresses on plant growth and development; however, the degree of plant response is host-specific. The present study aimed to assess the growth-promoting effect of Herbaspirillum (AP21, AP02), Azospirillum (D7), and Pseudomonas (N7) strains (single and co-inoculated) in perennial ryegrass plants subjected to drought. The plants were grown under controlled conditions and subjected to water deficit for 10 days. A significant increase of approximately 30% in dry biomass production was observed using three co-inoculation combinations (p < 0.01). Genomic analysis enabled the detection of representative genes associated with plant colonization and growth promotion. In vitro tests revealed that all the strains could produce indolic compounds and exopolysaccharides and suggested that they could promote plant growth via volatile organic compounds. Co-inoculations mostly decreased the in vitro-tested growth-promoting traits; however, the co-inoculation of Herbaspirillum sp. AP21 and Azospirillum brasilense D7 resulted in the highest indolic compound production (p < 0.05). Although the Azospirillum strain showed the highest potential in the in vitro and in silico tests, the plants responded better when PGPB were co-inoculated, demonstrating the importance of integrating in silico, in vitro, and in vivo assessment results when selecting PGPB to mitigate drought stress.

Variations of Indole Metabolites and NRPS-PKS Loci in Two Different Virulent Strains of Xenorhabdus hominickii.

Xenorhabdus hominickii ANU1 is known to be an entomopathogenic bacterium symbiotic to nematode Steinernema monticolum. Another bacterial strain X. hominickii DY1 was isolated from a local population of S. monticolum. This bacterial strain X. hominickii DY1 was found to exhibit high insecticidal activities against lepidopteran and coleopteran species after hemocoelic injection. However, these two X. hominickii strains exhibited significant variations in insecticidal activities, with ANU1 strain being more potent than DY1 strain. To clarify their virulence difference, bacterial culture broths of these two strains were compared for secondary metabolite compositions. GC-MS analysis revealed that these two strains had different compositions, including pyrrolopyrazines, piperazines, cyclopeptides, and indoles. Some of these compounds exhibited inhibitory activities against phospholipase A2 to block eicosanoid biosynthesis and induce significant immunosuppression. They also exhibited significant insecticidal activities after oral feeding, with indole derivatives being the most potent. More kinds of indole derivatives were detected in the culture broth of ANU1 strain. To investigate variations in regulation of secondary metabolite production, expression level of leucine-responsive regulatory protein (Lrp), a global transcription factor, was compared. ANU1 strain exhibited significantly lower Lrp expression level than DY1 strain. To assess genetic variations associated with secondary metabolite synthesis, bacterial loci encoding non-ribosomal protein synthase and polyketide synthase (NRPS-PKS) were compared. Three NRPS and four PKS loci were predicted from the genome of X. hominickii. The two bacterial strains exhibited genetic variations (0.12∼0.67%) in amino acid sequences of these NRPS-PKS. Most NRPS-PKS genes exhibited high expression peaks at stationary phase of bacterial growth. However, their expression levels were significantly different between the two strains. These results suggest that differential virulence of the two bacterial strains is caused by the difference in Lrp expression level, leading to difference in the production of indole compounds and other NRPS-PKS-associated secondary metabolites.

Hairy root transformation of Brassica rapa with bacterial halogenase genes and regeneration to adult plants to modify production of indolic compounds

During the last years halogenated compounds have drawn a lot of attention. Metabolites with one or more halogen atoms are often more active than their non-halogenated derivatives like indole-3-acetic acid (IAA) and 4-Cl-IAA. Within this work, bacterial flavin-dependent tryptophan halogenase genes were inserted into Brassica rapa ssp. pekinensis (Chinese cabbage) with the aim to produce novel halogenated indole compounds. It was investigated which tryptophan-derived indole metabolites, such as indole glucosinolates or potential degradation products can be synthesized by the transgenic root cultures. In vivo and in vitro activity of halogenases heterologously produced was shown and the production of chlorinated tryptophan in transgenic root lines was confirmed. Furthermore, chlorinated indole-3-acetonitrile (Cl-IAN) was detected. Other tryptophan-derived indole metabolites, such as IAA or indole glucosinolates were not found in the transgenic roots in a chlorinated form. The influence of altered growth conditions on the amount of produced chlorinated compounds was evaluated. We found an increase in Cl-IAN production at low temperatures (8 °C), but otherwise no significant changes were observed. Furthermore, we were able to regenerate the wild type and transgenic root cultures to adult plants, of which the latter still produced chlorinated metabolites. Therefore, we conclude that the genetic information had been stably integrated. The transgenic plants showed a slightly altered phenotype compared to plants grown from seeds since they also still expressed the rol genes. By this approach we were able to generate various stably transformed plant materials from which it was possible to isolate chlorinated tryptophan and Cl-IAN.

Plant growth promotion by Pseudomonas putida KT2440 under saline stress: role of eptA.

New strategies to improve crop yield include the incorporation of plant growth-promoting bacteria in agricultural practices. The non-pathogenic bacterium Pseudomonas putida KT2440 is an excellent root colonizer of crops of agronomical importance and has been shown to activate the induced systemic resistance of plants in response to certain foliar pathogens. In this work, we have analyzed additional plant growth promotion features of this strain. We show it can tolerate high NaCl concentrations and determine how salinity influences traits such as the production of indole compounds, siderophore synthesis, and phosphate solubilization. Inoculation with P. putida KT2440 significantly improved seed germination and root and stem length of soybean and corn plants under saline conditions compared to uninoculated plants, whereas the effects were minor under non-saline conditions. Also, random transposon mutagenesis was used for preliminary identification of KT2440 genes involved in bacterial tolerance to saline stress. One of the obtained mutants was analyzed in detail. The disrupted gene encodes a predicted phosphoethanolamine-lipid A transferase (EptA), an enzyme described to be involved in the modification of lipid A during lipopolysaccharide (LPS) biosynthesis. This mutant showed changes in exopolysaccharide (EPS) production, low salinity tolerance, and reduced competitive fitness in the rhizosphere.

Characterization of bacterial endophytes from the roots of native and cultivated Brazil nut trees (Bertholletia excelsa)

ABSTRACT Brazil nut is a very important nontimber forest product in the Amazon region. Propagation of this tree still represents a challenge due to slow and uneven seed germination. In this context, plant growth-promoting bacteria can facilitate the process of propagation. The aims of this study were to isolate and characterize endophytic bacteria from the roots of Brazil nut trees in native terra firme forest and cultivation areas in northern Brazil, and to identify mechanisms by which bacteria act in plant growth promotion. Overall, 90 bacterial isolates were obtained from the roots of Brazil nut trees in monoculture, agroforestry and native forest areas by using different semisolid media. The isolates were characterized by sequencing the 16S rRNA gene. Plant growth-promoting characteristics were evaluated by the presence of the nifH gene, aluminum phosphate solubilization and the production of indole compounds. The isolates were affiliated with 18 genera belonging to 5 different classes (α-Proteobacteria, β-Proteobacteria, γ-Proteobacteria, Bacilli and Actinobacteria). The genus Bacillus was predominant in the forest and monoculture areas. Fourteen isolates presented the nifH gene. Most of the bacteria were able to solubilize aluminum phosphate and synthetize indole compounds. The results indicated high diversity of endophytic bacteria present among the roots of Brazil nut trees, mainly in the agroforestry area, which could be related to soil attributes. Among the 90 isolates, the 22 that presented the best results regarding plant growth promotion traits were good candidates for testing in seedling production of Brazil nut trees.

Deciphering the melatonin metabolism in Saccharomyces cerevisiae by the bioconversion of related metabolites.

Melatonin (Mel), originally considered a neurohormone, has been detected in beverages and food-fermented products in which yeast metabolism is highly important. This indolamine is synthesized from serotonin, with L-tryptophan being the initial substrate of both. Regarding Mel metabolism, the biosynthetic pathway in mammals consists in four-step reactions. However, six genes are implicated in the synthesis of Mel in plants, which suggest the presence of many pathways. The aim of this study was to provide new empirical data on the production of Mel and other indole-related compounds in the yeast Saccharomyces cerevisiae (S.cerevisiae). To this end, we performed the addition of the pathway intermediates in S.cerevisiae cells in different growth stages (exponential and arrested cells) to follow the bioconversion and new indolic compound production from them. The different bioconverted indolic compounds tested (L-tryptophan, 5-hydroxytryptophan, tryptamine, serotonin, N-acetylserotonin, 5-methoxytryptamine, and Mel) were analyzed by UHPLC-MS/MS from the extra- and intracellular contents. Our results showed that serotonin, in yeast, was prevalently formed via tryptophan decarboxylation, followed by tryptamine hydroxylation as in plants. Mel production from serotonin can be achieved by either N-acetylation, followed by O-methylation or O-methylation, in turn followed by N-acetylation. Accordingly, the classic pathway of Mel synthesis in vertebrates does not seems prevalent in yeast.

Time course of l -tryptophan metabolites when fermenting natural grape musts: effect of inoculation treatments and cultivar on the occurrence of melatonin and related indolic compounds

Background and Aims Tryptophan is involved in the formation of bioactive compounds, such as melatonin (MEL) and 3-indoleacetic acid (3-IAA), by yeast. Melatonin is a neurohormone whose occurrence in wine has been widely reported in recent years. The occurrence, however, of MEL and other indolic compounds related to tryptophan metabolism by wine yeast strains has been scarcely reported in grape musts. This work examined the occurrence of these compounds during the alcoholic fermentation (AF) of musts from seven grape cultivars, Corredera, Chardonnay, Moscatel, Palomino Fino, Sauvignon Blanc, Tempranillo and Vijiriega. Methods and Results Must was fermented with three Saccharomyces cerevisiae strains and then in two cases an additional sequential inoculation with the non-Saccharomyces yeast Torulaspora delbrueckii was carried out. Fermented must samples were analysed by UHPLC/HRMS to determine the concentration of: l-tryptophan, 5-hydroxytryptophan, 5-hydroxytryptamine, N-acetyl-5-hydroxytryptamine, MEL, 3-IAA, tryptamine, tryptophol and l-tryptophan ethyl ester. The profile of indolic compounds during AF with the Aroma White strain depended on the cultivar. The yeast strain did not influence the profile of indolic compounds; instead, fermentation time was found to be a more influential factor. Conclusions The production of indolic compounds during the AF depends largely on the cultivar used and the day of fermentation on natural grape musts. Significance of the Study This is the first study that quantifies 5-hydroxytryptophan and N-acetyl-5-hydroxytryptamine during the AF of grape must. The occurrence of compounds with bioactive potential, for example 3-IAA and ML, during fermentation with commercial yeast strains is also described.

Plant Growth–Promoting Bacteria (PGPB): Isolation and Screening of PGP Activities

Plant roots are associated with numerous and diverse types of beneficial and pathogenic microorganisms. Plant growth-promoting (rhizo)bacteria (PGPB or PGPR) are isolated from plants crops worldwide, and many of them are used as agricultural inoculants. Agricultural biofertilization and biocontrol of pathogens are eco-friendly alternatives to chemical usage and have less energy, environmental, and economic costs. PGPB isolation and evaluation are essentials steps for determining bacteria that are able to improve plant development and productivity. In this unit, we present protocols to isolate bacteria from soil and plant roots ("putative" diazotrophic and endospore-forming bacteria), as well to evaluate some of their beneficial characteristics for the promotion of plant growth (e.g., nitrogen fixation, production of indolic compounds and siderophores, phosphate solubilization, and 1-aminocyclopropane-1-carboxylate deaminase activity). © 2017 by John Wiley & Sons, Inc.

The genus Gluconacetobacter comprises different species with agricultural and industrial importance. This study aims at determining the presence of Gluconacetobacter diazotrophicus and Gluconacetobacter sacchari in sugarcane and tomato crops considering their potential biotechnological applications. Bacteria were isolated from roots, stems and leaf tissues and identified using phenotypic and biochemical evaluations, and molecular and phylogenetic analyses. Isolates were characterized by the production of indolic compounds, nitrogenase activity and phosphate solubilization. For this, G. diazotrophicus ATCC 49037 was used as the reference strain. The results showed that all isolates of native G. diazotrophicus exhibited equal or better phosphate solubilization index (SI) for CaCO3 and AlPO4 than reference strain. For G. sacchari, GIBI031 isolate displayed better SI for both phosphate sources and GIBI014 isolate had better SI only for AlPO4. G. diazotrophicus GIBI029 had a greater production of indolic compounds than ATCC 49037 strain in the presence of tryptophan. All isolates except G sacchari GIBI031, showed better nitrogenase activity than the control. These results constitute the first report confirming the presence of G. diazotrophicus and G. sacchari associated with sugarcane in Colombia. In addition, this is the first report on the presence of G. sacchari in tomato under natural conditions. Finally, one G. sacchari isolate presented nitrogenase activity despite the fact that this is a differential characteristic between G. diazotrophicus and G. sacchari. These findings have ecological significance and will advance research towards the evaluation of plant-soil interactions involving these bacteria in crops other than sugarcane. The isolates found are potential candidates for the development of novel biotechnological processes for production of new alternative biofertilizers considering the significant plant-growth promotion properties determined in this work. Desarrollo de un sistema de fermentación líquida y de enquistamiento para una bacteria fijadora de nitrógeno con potencial como biofertilizante

The indiscriminate use of chemical fertilizers has contributed to the deterioration of the biological, physical and chemical properties of the soil, resulting in the loss of its productive capacity. For this reason, the use of biofertilizers has emerged as a technological alternative. The objective of this research was to develop a suitable liquid fermentation system and encystment for the multiplication of Azotobacter chroococcum AC1 strain, a bacterium employed in a biofertilizer formulation produced at present by CARPOICA, Colombia. Sequential statistical designs were used to determine the conditions in the fermentation system. The interaction between agitation, aeration and pH was evaluated on the viable biomass (CFU/ml) of AC1. In addition, the encystment ability of the strain was evaluated using two encystment agents and the potential plant growth-promoting rhizobacteria (PGPR) activity was assessed by different techniques, such as nitrogen fixation by ARA, phosphate solubilization by the phospho-molybdenum-blue reaction and indolic compound production by colorimetric reaction using the Salkowski reagent. Results showed significant effects (p<0.05) on the viable biomass in the three conditions (pH, aeration and agitation) tested individually, in one dual interaction and one tripartite interaction, were demonstrated to have a positive effect on the response variable aeration and agitation. The addition of the two encystment agents evaluated, AE01 and AE02, demonstrated the ability of AC1 to form cysts under stress conditions. Likewise, fermentation and encystment conditions did not affect the biological activities tested.

Native microorganisms as potent bioinoculants for plant growth promotion in shifting agriculture (Jhum) systems

Microorganisms native to shifting cultivation were isolated and screened for plant growth promotion (PGP) properties. The bacterial and fungal isolates that exhibed production of indole compounds, siderophores, and ammonia; phosphate solubilization; and catalase activity, were selected and tested for their ability to promote the early growth of upland rice plants. Potential isolates from a preliminary seed germination assay carried out in plates were further tested in pots under greenhouse conditions. Observations after two and four weeks of growth indicated that all treated rice plants had higher shoot and root length than those in the control group. Of the isolates used in the experiment, the bacterial isolate SB5 and the fungal isolate SF4 exhibit higher growth activity, with growth increases of 26.20% and 15.87% respectively, over the control group. These isolates are being explored for consortium development as bioinoculants, in an attempt to restore the fertility of jhum fallows

Bacillus effect on the germination and growth of tomato seedlings (Solanum lycopersicum L).

La capacidad para solubilizar fosfatos y producir compuestos indólicos del tipo Ácido Indol Acético, se evaluó en 15 cepas aisladas de residuos lignocelulósicos de higuerilla (Ricinus communis). Para determinar la actividad solubilizadora de fosfatos se realizó una prueba cualitativa utilizando medio de cultivo Pikovskaya y para evaluar la producción de compuestos indólicos (AIA) se empleó la técnica colorimétrica de Salkowsky. Entre los microorganismos evaluados, Bacillus pumilus GIBI 206 mostró tener la capacidad para solubilizar fosfatos y Bacillus subtilis GIBI 200 evidenció capacidad para solubilizar fosfatos y producir Ácido Indolacético (AIA). Para determinar el efecto de la cepa Bacillus subtilis sobre la germinación y promoción de crecimiento, se inoculó en semillas de tomate (Solanum lycopersicum cultivar Santa Clara); la inoculación de las semillas con el microorganismono mostró diferencias estadísticamente significativas en el tiempo de germinación en comparación con el tratamiento control, sin embargo mostró influir positivamente en el desarrollo de las plantas de tomate generando un aumento significativo sobre la masa y longitud del tallo y de la raíz. Los resultados de esta investigación ofrecen la posibilidad de utilizar a Bacillus subtilis como promotora de crecimiento en plántulas de tomate y en la formulación de bioinsumos

The isolation and characterization of resident yeasts from the phylloplane of Arabidopsis thaliana.

The genetic model plant Arabidopsis thaliana (arabidopsis) has been instrumental to recent advances in our understanding of the molecular function of the plant immune system. However, this work has not yet included plant associated and phytopathogenic yeasts largely due to a lack of yeast species known to interact with arabidopsis. The plant phylloplane is a significant habitat for neutral-residents, plant-growth and health-promoting species, and latent-pathogenic species. However, yeast phylloplane residents of arabidopsis remain underexplored. To address this, resident yeasts from the phyllosphere of wild arabidopsis collected in field conditions have been isolated and characterized. A total of 95 yeast strains representing 23 species in 9 genera were discovered, including potentially psychrophilic and pathogenic strains. Physiological characterization revealed thermotolerance profiles, sensitivity to the arabidopsis phytoalexin camalexin, the production of indolic compounds, and the ability to activate auxin responses in planta. These results indicate a rich diversity of yeasts present in the arabidopsis phylloplane and have created culture resources and information useful in the development of model systems for arabidopsis-yeast interactions.

Detecting and Monitoring the Production of Melatonin and Other Related Indole Compounds in Different Saccharomyces Strains by Solid-State Electrochemical Techniques

Recently, melatonin (Mel) has been detected in wine, and as its quantity increases during alcoholic fermentation, yeasts are primarily responsible for its production during the process. L-tryptophan (L-Trp) is an indolic molecule that plays an important role as a precursor of a large amount of tryptamine molecules, including Mel. We propose a novel method for detecting Mel and other derived indole compounds directly on yeast cells based on the voltammetry of immobilized particles methodology. This will abridge and facilitate the detection of these highly conserved and electroactive molecules. Tafel and modified Tafel analyses have been used to distinguish among Mel, tryptophol, and indole-3-acetic acid. We obtained different voltammetry responses in various Saccharomyces strains treated with L-Trp, 5-hydroxytryptophan, and serotonin and without them. The different responses allowed us to assess the production of indole compounds from L-Trp and other precursors. The voltammetric changes in mutant strains with deleted genes of tryptophan metabolism were also tested.
