Combining ability and heterosis in plant improvement

Information on combining ability and heterosis of parents and crossings is crucial in breeding efforts. Genetic variety is crucial to the effectiveness of yield improvement efforts because it helps to broaden gene pools in any given crop population. The genotype's ability to pass the intended character to the offspring is referred to as combining ability. As a result, information on combining ability is required to determine the crossing pairs in the production of hybrid varieties. Heterosis is the expression of an F1 hybrid's dominance over its parents in a given feature, as measured not by the trait's absolute value, but by its practical use. To put it another way, heterosis is defined as an increase in the character value of F1 hybrids when compared to the average value of both parents. A plant breeder's ultimate goal is to achieve desirable heterosis (hybrid vigor). In a variety of crop species, heterosis has been widely employed to boost output and extend the adaptability of hybrid types. A crucial requirement for discovering crosses with significant levels of exploitable heterosis is knowledge of the quantity of heterosis in different cross combinations. Any crop improvement program's success is contingent on the presence of a significant level of genetic diversity and heritability. The lack of a broad genetic foundation is the most significant constraint to crop improvement and a major bottleneck in breeding operations. Heterosis is a critical factor in hybrid generation, particularly for traits driven by non-additive gene activity. To get the most out of heterosis for hybrid cultivar production, germplasm must be divided into distinct heterotic groups. Similarly, knowledge on genetic diversity is critical for hybrid breeding and population improvement initiatives because it allows them to analyze genetic diversity, characterize germplasm, and categorize it into different heterotic groupings. In general, general combining ability is used to detect a line's average performance in a hybrid combination, whereas specific combining ability is used to find circumstances where definite combinations perform better or worse than expected based on the mean performance of the lines involved.

Gynogenesis in Cucurbita Species.

The development of F1 hybrid vegetable varieties emerges as a result of a great effort, long time, investment, knowledge, and advanced technology. The first stage of hybrid vegetable breeding is obtaining pure lines. It is possible to obtain homozygous parent lines used in the production of hybrid varieties with traditional breeding methods. This period takes 8-10years, especially in some vegetables which are highly open-pollinated, such as Cucurbita spp. Androgenetic- and/or gynogenetic-based dihaploidization methods provide 100% homozygous pure haploid lines in 1-2years and save time and effort.The DH frequency by irradiated pollen technique and anther culture strongly depends on the genotypic response, whereby their practical use in a breeding program is still limited. As a possible alternative technique, gynogenesis (unfertilized ovule/ovarium cultures) switches on to produce haploid plants in some Cucurbita species. In the Cucurbita genus, gynogenesis has been one of the most studied and popular DH techniques and presented remarkable results in recent years.

Barnase-barstar system: an indelible technique to produce hybrid seeds in self-pollinated crops

Self-pollinated crops are sensitive to exploit high level of heterosis. The improvement of crop plants through the production of hybrid varieties is a major goal of plant breeding. The genetically engineered techniques have a pivotal role in developing male sterility and restorer systems. Chimaeric ribonuclease gene (barnase) expression within the anther selectively destroys the tapetal cell layer that surrounds the pollen sac, prevents pollen formation and leads to male sterility. A chimaeric tapetal cell-specific ribonuclease inhibitor gene (barstar) was used in male fertile plants to restore male fertility in F1 progeny. Cytotoxic ribonuclease activity in the anther was suppressed to restore male fertility in F1 hybrid by the formation of cell-specific RNase/RNase inhibitor complexes.

Comparative Transcriptome Analysis of Recessive Male Sterility (RGMS) in Sterile and Fertile Brassica napus Lines.

The recessive genetic male sterility (RGMS) system plays a key role in the production of hybrid varieties in self-pollinating B. napus plants, and prevents negative cytoplasmic effects. However, the complete molecular mechanism of the male sterility during male-gametogenesis in RGMS remains to be determined. To identify transcriptomic changes that occur during the transition to male sterility in RGMS, we examined the male sterile line WSLA and male fertile line WSLB, which are near-isogenic lines (NILs) differing only in the fertility trait. We evaluated the phenotypic features and sterility stage using anatomical analysis. Comparative RNA sequencing analysis revealed that 3,199 genes were differentially expressed between WSLA and WSLB. Many of these genes are mainly involved in biological processes related to flowering, including pollen tube development and growth, pollen wall assembly and modification, and pollen exine formation and pollination. The transcript profiles of 93 genes associated with pollen wall and anther development were determined by quantitative RT-PCR in different flower parts, and classified into the following three major clades: 1) up-regulated in WSLA plants; 2) down-regulated in WSLA plants; and 3) down-regulated in buds, but have a higher expression in stigmas of WSLA than in WSLB. A subset of genes associated with sporopollenin accumulation were all up-regulated in WSLA. An excess of sporopollenin results in defective pollen wall formation, which leads to male sterility in WSLA. Some of the genes identified in this study are candidates for future research, as they could provide important insight into the molecular mechanisms underlying RGMS in WSLA.

Applications of Genie Male Sterility in Plant Breeding

AbstractThe possible applications of genie male sterility (GMS) in plant breeding arc reviewed and discussed. The basic contribution of GMS is that it provides a means of genetic emasculation which can be applied for the massive production of hybrids. There are two main fields of application: (1) the production of hybrid varieties and (2) inter‐ and intraspecific hybridization and back‐crossing programmes for the introduction of genetic variation into crop varieties. Several schemes have been proposed for using GMS in hybrid breeding in different crops. These are discussed under four headings: genetic methods, cy‐togenetic methods, temporary restoration of fertility and manual pollination. The application of GMS in hybridization intended for the introduction of alien genes is discussed as well as factors affecting the pollination of genie male steriles.

Haploidy and plant breeding

A haploid is an organism that looks like a sporophyte, but has the chromosome complement of a reduced gamete. There are several ways in which haploids can occur or be induced in vivo : spontaneously, mostly associated with polyembryony, and through abnormal processes after crosses, like pseudogamy, semigamy, preferential elimination of the chromosomes of one parental species, and androgenesis. In the crops described, haploids are or are near to being used in basic research and plant breeding. The application of haploids in breeding self-pollinated crops is based on their potential for producing fully homozygous lines in one generation, which can be assessed directly in the field. Early generation testing of segregating populations is possible through haploids, because doubled haploids (DH) possess additive variance only. Haploids can also be applied in classical breeding programmes to make these more efficient through improved reliability of selection. The application of haploids in cross-pollinated crops is also based on a rapid production of DH-lines, which can be used as inbred lines for the production of hybrid varieties. By means of haploids all natural barriers to repeated selfing are bypassed. In autotetraploid crops there are two types of haploid. One cycle of haploidization leads to dihaploids; a second cycle produces monohaploids. The significance of dihaploids is in their greatly simplified genetics and breeding and in the possibility of estimation of the breeding value of tetraploid cultivars by assessing their dihaploids. The main drawback of dihaploids is their restriction to two alleles per locus. Also, after doubling, it is impossible to achieve tetra-allelism at many loci, the requirement for maximal performance of autotetraploid cultivars. Tetra-allelism can be obtained when improved dihaploids have a genetically controlled mechanism of forming highly heterozygous restitution gametes with the unreduced number of chromosomes. Monohaploids, after doubling or twice doubling, may lead to fully homozygous diploids and tetraploids. These are important for basic research, but not yet for practical application. Meiotic data of potato homozygotes at three ploidy levels are presented.
