Species-level distribution of microbiota with secondary metabolic potential in Chinese Baijiu fermentation

Chinese Baijiu fermentation is a typical spontaneous fermentation with complex microbial community. However, the species-level distribution of microbiota with secondary metabolic potential in Baijiu fermentation has not been well studied so far. In this study, taking soy sauce aroma type Baijiu and light aroma type Baijiu fermentations as examples, we identified 1575 biosynthetic gene clusters (BGCs) belonging to 874 gene cluster families (GCFs) through metagenomic analysis. Bioactivity prediction analysis revealed 210 BGCs producing potential bioactive secondary metabolites (probabilities of bioactivities ≥70%). We recovered 66 bacterial metagenome-assembled genomes (MAGs) to explore distribution of bioactive secondary metabolites at a higher resolution taxonomic level. A total of 16 MAGs were identified as BGC-rich MAGs (>3 BGCs), and they belonged to 11 genera. Four MAGs, annotated to be Bacillus velezensis, Bacillus licheniformis, Kroppenstedtia eburnea and Kroppenstedtia spp., contained more than 3 BGC-producing potential bioactive secondary metabolites and had more than 1 counts per million (CPM) abundance in fermentations. They were identified as the potential producers of bioactive secondary metabolites. This study contributed to characterising the diversity and distribution of microbiota with secondary metabolic potential in Baijiu fermentation, and mining novel secondary metabolites and their producers.

Biological Control of Pseudomonas syringae in Tomato Using Filtrates and Extracts Produced by Alternaria leptinellae

Endophytic fungi offer promising alternatives for sustainable plant disease management strategies, often through the production of bioactive secondary metabolites. This study investigated the biocontrol potential of filtrates and extracts, produced under controlled conditions, from Alternaria leptinellae E138 against Pseudomonas syringae in tomato plants under greenhouse conditions. To understand the main mechanisms involved in biocontrol, the direct inhibition of bacterial growth and disruption of quorum sensing activity caused by metabolites were studied in vitro, as well as indirect mechanisms, such as their capacity to produce phytohormone-like substances, nutrient mobilization, and antioxidant activity, which can enhance plant growth and fitness. Moreover, a mass spectrometry analysis was used to tentatively identify the secondary metabolites present in the extract with antimicrobial properties, which could explain the biocontrol effects observed. Mycopriming assays, involving the direct treatment of tomato seeds with the fungal A. leptinellae E138 extracts, produced increased germination rates and seedling vigor in tomato seeds. As another treatment, postemergence application of the extracts in greenhouse conditions significantly improved plant health and resulted in a 41% decrease in disease severity. Overall, this study underscores the potential of A. leptinellae E138 extract as a plant growth promoter with biocontrol capabilities, offering promising avenues for sustainable plant disease management.

Induction, Proliferation and Differentiation of Callus in Paris polyphylla Sm. through Leaf Culture

Paris polyphylla Sm. is a vulnerable medicinal plant employed in the treatment of various ailments. This study seeks to establish a protocol for callus induction, proliferation, and differentiation of P. polyphylla. Immature leaf explants were cultured on MS medium with varying concentrations of plant growth regulators (PGRs), including 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin (Kn), 6-benzylaminopurine (BAP), Thidiazuron (TDZ), α-Naphthalene acetic acid (NAA), and Gibberellic acid (GA3), along with 10% coconut water. After 12 weeks of primary culture, the optimal callus induction was observed in MS medium supplemented with 0.25 mg/l 2,4-D + 0.5 mg/l Kn. In the secondary culture at 8 weeks, the best callus proliferation, as determined by callus weight or growth index, occurred in MS medium supplemented with 2.0 mg/l BAP alone, 2.0 mg/l Kn alone, 1.0 mg/l TDZ alone, combinations of 2.0 mg/l Kn + 1.0 mg/l BAP + 2.0 mg/l GA3, and combinations of 0.5 mg/l NAA + 2.0 mg/l BAP + 2.0 mg/l GA3, as well as 10% coconut water. Furthermore, callus differentiation into mini rhizomes with root primordia was successfully achieved in MS media containing 2.5 mg/l Kn and 10% coconut water. This study reports, for the first time, the formation and differentiation of callus from leaf explants in P. polyphylla. Large-scale callus generation from leaf explants has the potential to enhance the production of bioactive secondary metabolites for therapeutic purposes and facilitate the development of plantlets through organogenesis.

Bioprospection of Tenellins Produced by the Entomopathogenic Fungus Beauveria neobassiana.

Fungi are known as prolific producers of bioactive secondary metabolites with applications across various fields, including infectious diseases, as well as in biological control. However, some of the well-known species are still underexplored. Our current study evaluated the production of secondary metabolites by the entomopathogenic fungus Beauveria neobassiana from Thailand. The fermentation of this fungus in a liquid medium, followed by preparative high-performance liquid chromatography (HPLC) purification, resulted in the isolation of a new tenellin congener, namely pretenellin C (1), together with five known derivatives (2-6). Their chemical structures were elucidated by 1D and 2D nuclear magnetic resonance (NMR) spectroscopy in combination with high-resolution electrospray ionization mass spectrometry (HR-ESI-MS). We evaluated the antimicrobial and cytotoxic activities from all isolated compounds, as well as their inhibitory properties on biofilm formation by Staphylococcus aureus. Generally, tenellins displayed varying antibiofilm and cytotoxic effects, allowing us to propose preliminary structure-activity relationships (SARs). Among the tested compounds, prototenellin D (2) exhibited the most prominent antibiofilm activity, while its 2-pyridone congener, tenellin (4), demonstrated potent cytotoxic activity against all tested cell lines. Given the fact that the biological activities of the tenellins have so far been neglected in the past, our study could provide a good starting point to establish more concise structure-activity relationships in the near future.

Callus Induction and Establishing Cell Suspension Culture of Agastache J.Clayton Ex Gronov

The aim of this study was to initiate calli formation and cell suspension cultures from some species of Agastache genus. These plants could be useful for a production of bioactive secondary metabolites in vitro. For the initiation of callogenesis, two explant types were tested: leaf and stem explants from 40–60 days old in vitro seedlings. Percentage of callus formation was used as criterion to evaluate the efficiency of callus induction. Leaf- and stem-derived friable calli of A. foeniculum and A. urticifolia cultivated on MS medium supplemented with 0.5 mg/L 2.4-dichlorophenoxyacetic acid and 0.1 mg/L kinetin were selected for the cell suspension cultures establishing. The cell suspension cultures of A. foeniculum characterized by growth indexes of 1.08 and 8.57 for MS and B5 media respectively. For A. urticifolia suspension cultures growth indexes were 3.01 for MS medium and 1.29 for B5 medium. The period of culturing was 28 days. Viability of cell suspension cultures varied 50–100 during the period of culturing. According to the growth characteristics for establishing A. foeniculum suspension culture is better to use MS medium, and for A. urticifolia – B5.

Optimization of culture condition for growth and production of bioactive secondary metabolites by endophytic fungus Thermothelomyces thermophiles XJF-9

Endophytic fungi living in intracellular without any negative symptoms of their host and synthase natural bioactive products considered as an elicitor for plant secondary metabolites production. The structural diverse and complex of the natural compounds synthesis by endophytic fungi make them an important rich source of secondary metabolites with novel and unique structures that can help in the treatment of various human diseases. The review focused secondary metabolites with their structure and chemical diversity from the plant-associated endophytic fungi. Moreover, reported to exhibit diverse pharmacological properties such as antimicrobial, antioxidant, anti-inflammatory, anticancer and inhibitory activities. This review discus a total 130 novel bioactive natural compounds isolated from the different species of fungal endophytes reported from 2020 to 2022.

Proposing Signaling Molecules as Key Optimization Targets for Intensifying the Phytochemical Biosynthesis Induced by Emerging Nonthermal Stress Pretreatments of Plant-Based Foods: A Focus on γ-Aminobutyric Acid.

Emerging evidence has confirmed the role of emerging nonthermal stressors (e.g., electromagnetic fields, ultrasonication, plasma) in accumulating bioactive metabolites in plant-based food. However, the signal decoding mechanisms behind NonTt-driven phytochemical production remain unclear, hindering postharvest bioactive component intensification. This study aims to summarize the association between signaling molecules and bioactive secondary metabolite production under nonthermal conditions, demonstrating the feasibility of enhancing phytochemical accumulation through signaling molecule crosstalk manipulation. Nonthermal elicitors were found to be capable of inducing stress metabolisms and activating various signaling molecules, similar to conventional abiotic stress. A simplified pathway model for nonthermally induced γ-aminobutyric acid accumulation was proposed with reactive oxygen species and calcium signaling being versatile pathways responsive to nonthermal elicitors. Manipulating signal molecules/pathways under nonthermal conditions can intensify phytochemical biosynthesis. Further research is needed to integrate signaling molecule responses and metabolic network shifts in nonthermally stressed plant-based matrices, balancing quality modifications and intensification of food functionality potential.

Optimization of culture condition for growth and production of bioactive secondary metabolites by endophytic fungus Thermothelomyces thermophiles XJF-9

In this work aimed to optimize the culture condition of the fungal strain Thermothelomyces thermophiles XJF-9 to increase the growth and production of pharmacological active secondary metabolites. The potent fungus is subjected to optimization in various culture medium and incubation time. The optimized culture medium was Potato Dextrose Broth (PDB) using dextrose as carbon source and incubation time 14 days for the maximum growth and production of bioactive natural compounds by T. thermophiles XJF-9. Moreover, secondary metabolites of crude ethyl acetate extract were investigation melanin content assay and tyrosinase activity. These secondary metabolites are effectively utilized in medicinal and pharmaceutical industry.

Antioxidant and Enzyme Inhibitory Potential of Streptomyces sp. G-18 Grown in Various Media.

Streptomyces are bacteria well known for producing bioactive secondary metabolites which are commonly found in diverse habitats. The biosynthesis of metabolites from Streptomyces is influenced by various factors such as the growth medium, environmental conditions, and gene regulation. This study aimed to investigate the influence of different growth media on biomass production and the antioxidant and enzyme inhibitory potential of a crude extract obtained from Streptomyces sp. G-18 isolated from high altitudinal soil of Nepal. The highest dry weight growth was observed in R2YE medium (184 mg/L), followed by R5 (144 mg/L), YEME (38 mg/L), and R5M media (30 mg/L). The crude extract showed notable antioxidant activities against free radicals. The highest alpha-amylase inhibition was observed in the R2YE medium, and worthy lipase and tyrosinase inhibition was observed in the YEME medium. However, only the R2YE medium exhibited inhibitory potential against elastase and acetylcholinesterase, while crude extracts from R5, YEME, and R5 modified did not show any such activity. Overall, our findings suggest that the production of bioactive secondary metabolites in Streptomyces sp. G-18 was significantly influenced by the growth medium. This strain may be a promising source of enzyme inhibitors with potential applications in the pharmaceutical and cosmetic industries.

Interaction of calcium responsive proteins and transcriptional factors with the PHO regulon in yeasts and fungi.

Phosphate and calcium ions are nutrients that play key roles in growth, differentiation and the production of bioactive secondary metabolites in filamentous fungi. Phosphate concentration regulates the biosynthesis of hundreds of fungal metabolites. The central mechanisms of phosphate transport and regulation, mediated by the master Pho4 transcriptional factor are known, but many aspects of the control of gene expression need further research. High ATP concentration in the cells leads to inositol pyrophosphate molecules formation, such as IP3 and IP7, that act as phosphorylation status reporters. Calcium ions are intracellular messengers in eukaryotic organisms and calcium homeostasis follows elaborated patterns in response to different nutritional and environmental factors, including cross-talking with phosphate concentrations. A large part of the intracellular calcium is stored in vacuoles and other organelles forming complexes with polyphosphate. The free cytosolic calcium concentration is maintained by transport from the external medium or by release from the store organelles through calcium permeable transient receptor potential (TRP) ion channels. Calcium ions, particularly the free cytosolic calcium levels, control the biosynthesis of fungal metabolites by two mechanisms, 1) direct interaction of calcium-bound calmodulin with antibiotic synthesizing enzymes, and 2) by the calmodulin-calcineurin signaling cascade. Control of very different secondary metabolites, including pathogenicity determinants, are mediated by calcium through the Crz1 factor. Several interactions between calcium homeostasis and phosphate have been demonstrated in the last decade: 1) The inositol pyrophosphate IP3 triggers the release of calcium ions from internal stores into the cytosol, 2) Expression of the high affinity phosphate transporter Pho89, a Na+/phosphate symporter, is controlled by Crz1. Also, mutants defective in the calcium permeable TRPCa7-like of Saccharomyces cerevisiae shown impaired expression of Pho89. This information suggests that CrzA and Pho89 play key roles in the interaction of phosphate and calcium regulatory pathways, 3) Finally, acidocalcisomes organelles have been found in mycorrhiza and in some melanin producing fungi that show similar characteristics as protozoa calcisomes. In these organelles there is a close interaction between orthophosphate, pyrophosphate and polyphosphate and calcium ions that are absorbed in the polyanionic polyphosphate matrix. These advances open new perspectives for the control of fungal metabolism.

Review of Marine Cyanobacteria and the Aspects Related to Their Roles: Chemical, Biological Properties, Nitrogen Fixation and Climate Change.

Marine cyanobacteria are an ancient group of photosynthetic microbes dating back to 3.5 million years ago. They are prolific producers of bioactive secondary metabolites. Over millions of years, natural selection has optimized their metabolites to possess activities impacting various biological targets. This paper discusses the historical and existential records of cyanobacteria, and their role in understanding the evolution of marine cyanobacteria through the ages. Recent advancements have focused on isolating and screening bioactive compounds and their respective medicinal properties, and we also discuss chemical property space and clinical trials, where compounds with potential pharmacological effects, such as cytotoxicity, anticancer, and antiparasitic properties, are highlighted. The data have shown that about 43% of the compounds investigated have cytotoxic effects, and around 8% have anti-trypanosome activity. We discussed the role of different marine cyanobacteria groups in fixing nitrogen percentages on Earth and their outcomes in fish productivity by entering food webs and enhancing productivity in different agricultural and ecological fields. The role of marine cyanobacteria in the carbon cycle and their outcomes in improving the efficiency of photosynthetic CO2 fixation in the chloroplasts of crop plants, thus enhancing the crop plant's yield, was highlighted. Ultimately, climate changes have a significant impact on marine cyanobacteria where the temperature rises, and CO2 improves the cyanobacterial nitrogen fixation.

Bioactive metabolites of plants and microbes and their role in agricultural sustainability and mitigation of plant stress

Primary metabolites are ubiquitous in nature and actively involved in growth while secondary metabolites (SMs) are involved in interaction with the environment. In plants terpenes are the largest group of secondary metabolites that play an important role in defense mechanisms against the herbivores and pathogens. Plant secondary metabolites such as alkaloids, phenolics and diterpenes have a great potential against many carcinogens. Carotenoids provide photoprotection and act as a pigment to absorb light during photosynthesis. Carotenoids are also efficient in the interaction with other organisms as they are phagostimulant and allelopathic agent. Flavonoids from the higher plants play a significant role in ultra-violet (UV) filtration, nitrogen fixation, floral pigmentation, and defense mechanisms against many diseases. These secondary metabolites provide protection to plants in a different type of biotic and abiotic stresses. Microorganisms also contain a high number of bioactive metabolites but not all of them. Marine microorganisms are a rich source of secondary metabolites such as alkaloids, terpenoids, polyketides, isoprenoids and non-isoprenoid and quinones amongst others. These metabolites are anti-inflammatory, anti-bacterial and anti-tumor in action. Secondary metabolites from fungi have a great therapeutic value as they have antifungal and antibacterial activity against many pathogens. Plant growth promoting rhizobacteria (PGPR) and Plant growth regulators (PGR) are the key biological entities to enhance the production of bioactive secondary metabolites in plants and can change the plant metabolome in response to various environmental stresses. This review paper will provide a new insight about the role of secondary metabolites in plant defense and the role of microorganisms in modulating the plant metabolome under various environmental stresses and their interactions with other microorganisms and plant growth regulators.

Development of a native-locus dual reporter system for the efficient screening of the hyper-production of natural products in Streptomyces.

Streptomyces is renowned for its abundant production of bioactive secondary metabolites, but most of these natural products are produced in low yields. Traditional rational network refactoring is highly dependent on the comprehensive understanding of regulatory mechanisms and multiple manipulations of genome editing. Though random mutagenesis is fairly straightforward, it lacks a general and effective strategy for high throughput screening of the desired strains. Here in an antibiotic daptomycin producer S. roseosporus, we developed a dual-reporter system at the native locus of the daptomycin gene cluster. After elimination of three enzymes that potentially produce pigments by genome editing, a gene idgS encoding the indigoidine synthetase and a kanamycin resistant gene neo were integrated before and after the non-ribosomal peptidyl synthetase genes for daptomycin biosynthesis, respectively. After condition optimization of UV-induced mutagenesis, strains with hyper-resistance to kanamycin along with over-production of indigoidine were efficiently obtained after one round of mutagenesis and target screening based on the dual selection of the reporter system. Four mutant strains showed increased production of daptomycin from 1.4 to 6.4 folds, and significantly improved expression of the gene cluster. Our native-locus dual reporter system is efficient for targeting screening after random mutagenesis and would be widely applicable for the effective engineering of Streptomyces species and hyper-production of these invaluable natural products for pharmaceutical development.

Fungal Endophytes: Microfactories of Novel Bioactive Compounds with Therapeutic Interventions; A Comprehensive Review on the Biotechnological Developments in the Field of Fungal Endophytic Biology over the Last Decade.

The seminal discovery of paclitaxel from endophytic fungus Taxomyces andreanae was a milestone in recognizing the immense potential of endophytic fungi as prolific producers of bioactive secondary metabolites of use in medicine, agriculture, and food industries. Following the discovery of paclitaxel, the research community has intensified efforts to harness endophytic fungi as putative producers of lead molecules with anticancer, anti-inflammatory, antimicrobial, antioxidant, cardio-protective, and immunomodulatory properties. Endophytic fungi have been a valuable source of bioactive compounds over the last three decades. Compounds such as taxol, podophyllotoxin, huperzine, camptothecin, and resveratrol have been effectively isolated and characterized after extraction from endophytic fungi. These findings have expanded the applications of endophytic fungi in medicine and related fields. In the present review, we systematically compile and analyze several important compounds derived from endophytic fungi, encompassing the period from 2011 to 2022. Our systematic approach focuses on elucidating the origins of endophytic fungi, exploring the structural diversity and biological activities exhibited by these compounds, and giving special emphasis to the pharmacological activities and mechanism of action of certain compounds. We highlight the tremendous potential of endophytic fungi as alternate sources of bioactive metabolites, with implications for combating major global diseases. This underscores the significant role that fungi can play in the discovery and development of novel therapeutic agents that address the challenges posed by prevalent diseases worldwide.

Anti-Insect Properties of Penicillium Secondary Metabolites.

In connection with their widespread occurrence in diverse environments and ecosystems, fungi in the genus Penicillium are commonly found in association with insects. In addition to some cases possibly implying a mutualistic relationship, this symbiotic interaction has mainly been investigated to verify the entomopathogenic potential in light of its possible exploitation in ecofriendly strategies for pest control. This perspective relies on the assumption that entomopathogenicity is often mediated by fungal products and that Penicillium species are renowned producers of bioactive secondary metabolites. Indeed, a remarkable number of new compounds have been identified and characterized from these fungi in past decades, the properties and possible applications of which in insect pest management are reviewed in this paper.

Endophytic fungi mediates production of bioactive secondary metabolites via modulation of genes involved in key metabolic pathways and their contribution in different biotechnological sector.

Endophytic fungi stimulate the production of an enormous number of bioactive metabolites in medicinal plants and affect the different steps of biosynthetic pathways of these secondary metabolites. Endophytic fungi possess a number of biosynthetic gene clusters that possess genes for various enzymes, transcription factors, etc., in their genome responsible for the production of secondary metabolites. Additionally, endophytic fungi also modulate the expression of various genes responsible for the synthesis of key enzymes involved in metabolic pathways of such as HMGR, DXR, etc. involved in the production of a large number of phenolic compounds as well as regulate the expression of genes involved in the production of alkaloids and terpenoids in different plants. This review aims to provide a comprehensive overview of gene expression related to endophytes and their impact on metabolic pathways. Additionally, this review will emphasize the studies done to isolate these secondary metabolites from endophytic fungi in large quantities and assess their bioactivity. Due to ease in synthesis of secondary metabolites and their huge application in the medical industry, these bioactive metabolites are now being extracted from strains of these endophytic fungi commercially. Apart from their application in the pharmaceutical industry, most of these metabolites extracted from endophytic fungi also possess plant growth-promoting ability, bioremediation potential, novel bio control agents, sources of anti-oxidants, etc. The review will comprehensively shed a light on the biotechnological application of these fungal metabolites at the industrial level.

Identification of Antimicrobial Metabolites from the Egyptian Soil-Derived Amycolatopsis keratiniphila Revealed by Untargeted Metabolomics and Molecular Docking.

Actinomycetes are prolific producers of bioactive secondary metabolites. The prevalence of multidrug-resistant (MDR) pathogens has prompted us to search for potential natural antimicrobial agents. Herein, we report the isolation of rare actinobacteria from Egyptian soil. The strain was identified as Amycolatopsis keratiniphila DPA04 using 16S rRNA gene sequencing. Cultivation profiling, followed by chemical and antimicrobial evaluation of crude extracts, revealed the activity of DPA04 ISP-2 and M1 culture extracts against Gram-positive bacteria. Minimum inhibitory concentrations (MIC) values ranged from 19.5 to 39 µg/mL. Chemical analysis of the crude extracts using ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-QTOF) led to the identification of 45 metabolites of different chemical classes. In addition, ECO-0501 was identified in the cultures with significant antimicrobial activity. Multidrug resistance in Staphylococcus aureus is reported to be related to the multidrug efflux pump (MATE). ECO-0501 and its related metabolites were subjected to molecular docking studies against the MATE receptor as a proposed mechanism of action. ECO-0501 and its derivatives (AK_1 and N-demethyl ECO-0501) had better binding scores (-12.93, -12.24, and -11.92 kcal/mol) than the co-crystallized 4HY inhibitor (-8.99 kcal/mol) making them promising candidates as MATE inhibitors. Finally, our work established that natural products from this strain could be useful therapeutic tools for controlling infectious diseases.

Genome Mining and Screening for Secondary Metabolite Production in the Endophytic Fungus Dactylonectria alcacerensis CT-6.

Endophytic fungi are a treasure trove of natural products with great chemical diversity that is largely unexploited. As an alternative to the traditional bioactivity-guided screening approach, the genome-mining-based approach provides a new methodology for obtaining novel natural products from endophytes. In our study, the whole genome of an endophyte, Dactylonectria alcacerensis CT-6, was obtained for the first time. Genomic analysis indicated that D. alcacerensis CT-6 has one 61.8 Mb genome with a G+C content of 49.86%. Gene annotation was extensively carried out using various BLAST databases. Genome collinearity analysis revealed that D. alcacerensis CT-6 has high homology with three other strains of the Dactylonectria genus. AntiSMASH analysis displayed 45 secondary metabolite biosynthetic gene clusters (BGCs) in D. alcacerensis CT-6, and most of them were unknown and yet to be unveiled. Furthermore, only six known substances had been isolated from the fermented products of D. alcacerensis CT-6, suggesting that a great number of cryptic BGCs in D. alcacerensis CT-6 are silent and/or expressed at low levels under conventional conditions. Therefore, our study provides an important basis for further chemical study of D. alcacerensis CT-6 using the gene-mining strategy to awaken these cryptic BGCs for the production of bioactive secondary metabolites.

Comparative Genomics Analysis of Ganoderma Orthologs Involved in Plant-Pathogenesis

Ganoderma species are producers of bioactive secondary metabolites and lignin degraders. A few Ganoderma species are known to be plant pathogens that attack economically important trees. In this study, comparative genomics analysis was conducted on the proteome of ten Ganoderma species/strains, focusing on the proteins that have been reported to be involved in plant-pathogenesis in other fungi. Fungal trophic lifestyle prediction of these Ganoderma species/strains supported that G. boninense (a potent pathogen to oil palm) is a hemibiotrophic fungus while the other Ganoderma species/strains analyzed were predicted to be saprophytes or a symbiont based on their Carbohydrate-Active Enzyme (CAZyme) contents. Although these Ganoderma species/strains were demonstrated to share many CAZymes and secondary metabolite core gene clusters, individual species may produce unique CAZymes and secondary metabolite core genes that determine their lifestyles, host-specificity, and potential as a producer of bioactive secondary metabolites. Ortholog groups that are related to fungal virulence from seven Ganoderma species/strains including those involved in lignin degradation, mycotoxin, siderophore and ergosterol biosynthesis, and virulence were summarized. Potential effectors were predicted from the proteome of these Ganoderma species/strains, and putative effectors that were being expressed in G. boninense in oil palm roots but not found in other species were identified. The findings provide a useful resource to further analyze plant-pathogenesis and wood degradation activities of these Ganoderma species.

Antimicrobial and antioxidant potentials of crude extracts of culturally dissimilar endophytic fungi

Endophytes from an unexplored plant such as Spatterdock (SD) Nuphar lutea variegata could be a dependable producer of novel secondary metabolites active against drug resistance organisms. This study explored the diversity of fungal endophytes from mid-rib (MR) and leaf blade (LB) of Nuphar lutea variegata as well as the antimicrobial and antioxidant potentials of their secondary metabolites. Morphological features observed showed four species were isolated. A broad-spectrum antimicrobial activity observed to be concentration-dependent was observed for all the fungal crude extracts 1 mg/mL. Crude extract of SD-LB3 showed the best antibacterial activities against S. aureus and P. aeruginosa with a minimum inhibitory concentration of 0.5 and 0.25 µg/mL respectively. The antioxidant capacities of the fungal crude extracts showed a better neutralizing effect of free radicals compared to the standard Quercitin. Effective inhibition of free radicals was observed for all the fungal extracts from 20 to 100 mg/mL (96.7 – 99.3 %). The highest antioxidant activity was shown by the extracts of fungi isolated from the leaf blade with an average of 98 to 99.2 % at the tested concentrations of 20 to 100 mg/mL. Our study provides evidence of diverse endophytic fungi associated with Nuphar lutea variegata, and the antibacterial and antioxidant activities observed confirm their capacities as producers of bioactive secondary metabolites. This study is the first to isolate endophytic fungi form Nuphar lutea variegata and screen the antimicrobial and antioxidant activities of their secondary metabolites.
