Red ginseng dietary fiber promotes probiotic properties of Lactiplantibacillus plantarum and alters bacterial metabolism.

Korean red ginseng has been widely used as an herbal medicine. Red ginseng dietary fiber (RGDF) is a residue of the processed ginseng product but still contains bioactive constituents that can be applied as prebiotics. In this study, we evaluated changes on fermentation profiles and probiotic properties of strains that belong to family Lactobacillaceae with RGDF supplementation. Metabolomic analyses were performed to understand specific mechanisms on the metabolic alteration by RGDF and to discover novel bioactive compounds secreted by the RGDF-supplemented probiotic strain. RGDF supplementation promoted short-chain fatty acid (SCFA) production, carbon source utilization, and gut epithelial adhesion of Lactiplantibacillus plantarum and inhibited attachment of enteropathogens. Intracellular and extracellular metabolome analyses revealed that RGDF induced metabolic alteration, especially associated with central carbon metabolism, and produced RGDF-specific metabolites secreted by L. plantarum, respectively. Specifically, L. plantarum showed decreases in intracellular metabolites of oleic acid, nicotinic acid, uracil, and glyceric acid, while extracellular secretion of several metabolites including oleic acid, 2-hydroxybutanoic acid, hexanol, and butyl acetate increased. RGDF supplementation had distinct effects on L. plantarum metabolism compared with fructooligosaccharide supplementation. These findings present potential applications of RGDF as prebiotics and bioactive compounds produced by RGDF-supplemented L. plantarum as novel postbiotic metabolites for human disease prevention and treatment.

Study on the safety and excellent probiotic properties of fermentation strains isolated from traditional dry-cured duck for starter development

The indigenous starter cultures isolated from fermented food can compete with spoilage and pathogenic bacteria and improve the flavor and organoleptic properties of products. The purposes of this work were to screen and assess the safety, potential probiotic, and fermentation properties of indigenous starter cultures isolated from dry-cured ducks. The properties of indigenous starter cultures were investigated by antibiotic resistance, virulence genes, in vivo toxicity, simulated gastrointestinal, adhesion capacity to Caco-2 cells, inoculation fermentation, etc. Overall, six strains were isolated and identified, including Lactobacillus sakei D15, Weissella hellenica D20, Lactobacillus curvatus D2, Staphylococcus simulans D12, Staphylococcus equorum D19, and Staphylococcus edaphicus D17. The obtained strains showed high safety. Additionally, most strains displayed excellent probiotic properties, including tolerance to lower pH, lysozyme, bile salt, and a high survival rate of the simulated gastrointestinal phase. The adhesion ability of strain cells to Caco-2 cells was 9.22%–14.21%. And the probiotic properties of strains can be strengthened by the expression of specific enzymes. The inoculation with the isolated strains (LS-D15, LC-D2 and SS-D12) could contribute to the formation of volatile flavor compounds in samples. Overall, results demonstrated that strains possessed excellent safety and probiotic properties, and can be used as starter cultures to enhance the quality and flavor of the dry-cured duck.

New Potentially Probiotic Strains Isolated from Humans - Comparison of Properties with Strains from Probiotic Products and ATCC Collection.

Lactic acid bacteria are used in various types of probiotic products. Due to the constantly growing probiotics market, new strains with pro-health properties are sought. The present study compared 39 strains of Lactobacillus, Lacticaseibacillus, and Lactiplantibacillus, isolated from probiotic products and healthy people. The current research aimed to search for new, potentially probiotic strains. For this purpose the relationship between Lactobacillaceae strains was carried out; moreover, the basic properties of probiotic microorganisms, such as survival at low pH and bile salt environment, antibiotic susceptibility, aggregation and antagonism were estimated. The properties of these isolates were also compared with the properties of probiotic strains from the ATCC collection. In comparing the genetic relationship (PFGE method) between the tested isolates, it was observed that some of them show a high degree of similarity. All tested strains tolerated an environment with a pH value of 3.0, and the addition of 0.3% bile salt; showed auto-aggregation properties and displayed antagonism against pathogenic microorganisms. In the present study, the bacteria were susceptible to tetracycline, chloramphenicol and ampicillin; the resistance to vancomycin depended on the bacteria type. All the properties were strain-depended. Most of the tested strains had properties comparable to the reference strains. Three L. acidophilus strains isolated from cervical swabs seem to be promising candidates for probiotic strains.

The biotechnological studies when developing lozenges with probiotics

Aim. To develop lozenges with probiotic components for the treatment and prevention of ENT diseases and analyze their effectiveness.Materials and methods. The study objects were lozenges based on xylitol and sorbitol of two compositions – based on live probiotic cultures and their metabolites. The following methods of control of the sample efficiency werechosen: determination of the number of viable lactobacilli in the study of their combined use with the drug components and the number of viable bacteria in finished oral probiotics by the Koch method (direct seeding on cups), as well as the method of diffusion into agar with the growth inhibition zone determination.Results and discussion. The studies of the properties of probiotic strains of L. fermentum 90-TC and L plantarum 8P-A3 have shown the prospects for their use in oral probiotics. It has been also proposed to use metabolitesof these strains as active components, which have a rather high antimicrobial potential to opportunistic pathogens that cause infectious ENT diseases. The oral probiotics in the form of lozenges of two compositions – based on live lactobacilli and their metabolites – have shown high efficiency, bacterial survival and antimicrobial properties, respectively.Conclusions. During the complex of the theoretical, technological and biotechnological studies, lozenges with a probiotic of two compositions (based on viable probiotic cultures and their metabolites) have been developed. After further research they can be recommended as a biologically active additive to support the oral microflora, stimulate the own immunity and as a therapeutic and prophylactic agent in infectious diseases of the oral cavity for use in both adults and children.
