Analysis of Gizzerosine in Foodstuffs by HPLC Involving Pre-column Derivatization with o-Phthaldialdehyde.

Gizzerosine [2-amino-9-(4-imidazolyl)-7-azanonanoic acid] is a toxic amino acid formed from histamine and lysine at high temperatures, and may be present in foodstuffs (e.g., fishmeal and meat-bone meal) for animals including cats and dogs. Here we developed a simple, rapid, sensitive, specific, and automated method for the analysis of gizzerosine in foodstuffs by high-performance liquid chromatography (HPLC) involving pre-column derivatization with o-phthaldialdehyde (OPA) in the presence of N-acetylcysteine (instead of the usual 2-mercaptoethanol or ethanethiol reagent). OPA reacted immediately (within 1 min) with gizzerosine in an autosampler at room temperatures (e.g., 20-25°C), and their derivative was directly injected into the HPLC column. The highly fluorescent gizzerosine-OPA derivative was well separated from the OPA derivatives of all natural amino acids known to be present in physiological fluids (e.g., plasma), proteins and foodstuffs, and was detected at an excitation wavelength of 340 nm and an emission wavelength of 450 nm. The total time for chromatographic separation (including column regeneration) was 20 min per sample rather than 40 min and longer in previous HPLC methods. The detection limit for gizzerosine was at least 6 pmol/ml in an assay solution (HPLC vial) or at least0.09pmol per injection into the HPLC column. The analysis of gizzerosine was linear between 1 and 100pmol per injection. When gizzerosine was extracted from foodstuffs, its detection limit was at least 875pmol/g foodstuff or at least0.21 mg/kg foodstuff. Our routine HPLC technique does not require any cleanup of samples or the OPA derivatization products (including the OPA-gizzerosine adduct), and is applicable for the analysis of gizzerosine in bothfoodstuffs and animal tissues.

Differential Expression of Amanitin Biosynthetic Genes and Novel Cyclic Peptides in Amanita molliuscula.

Amanita molliuscula is a basal species of lethal Amanita and intrigues the field because it does not produce discernable α-amanitin when inspected by High Performance Liquid Chromatography (HPLC), which sets it apart from all known amanitin-producing (lethal) Amanita species. In order to study the underlining genetic basis of the phenotype, we sequenced this species through PacBio and Illumina RNA-Seq platforms. In total, 17 genes of the “MSDIN” family (named after the first five amino acid residues of the precursor peptides) were found in the genome and 11 of them were expressed at the transcription level. The expression pattern was not even but in a differential fashion: two of the MSDINs were highly expressed (FPKM value > 100), while the majority were expressed at low levels (FPKM value < 1). Prolyl oligopeptidease B (POPB) is the key enzyme in the amanitin biosynthetic pathway, and high expression of this enzyme was also discovered (FPKM value > 100). The two MSDINs with highest transcription further translated into two novel cyclic peptides, the structure of which is distinctive from all known cyclic peptides. The result illustrates the correlation between the expression and the final peptide products. In contrast to previous HPLC result, the genome of A. molliuscula harbors α-amanitin genes (three copies), but the product was in trace amount indicated by MS. Overall, transcription of MSDINs encoding major toxins (α-amanitin, β-amanitin, phallacidin and phalloidin) were low, showing that these toxins were not actively synthesized at the stage. Collectively, our results indicate that the amanitin biosynthetic pathway is highly active at the mature fruiting body stage in A. molliuscula, and due to the differential expression of MSDIN genes, the pathway produces only a few cyclic peptides at the time.

Detection of residual formaldehyde in N-butyl-2-cyanoacrylate by high-performance liquid chromatography with rhodamine B hydrazide

Rhodamine B hydrazide (RBH) was prepared as a new derivatizing reagent for residual formaldehyde in a drug such as N-butyl-2-cyanoacrylate (NBCA). RBH reacted with formaldehyde rapidly at room temperature, and the resulting derivative can be detected sensitively at 238 nm. After optimizing the derivatization and chromatographic separation conditions, RBH as a pre-column derivatizing reagent has been successfully used for the analysis of formaldehyde in real NBCA samples by high-performance liquid chromatography (HPLC). The detection limit of the method is 0.004 μg/mL for formaldehyde, and the recovery is 94.0%-96.6%. Most notably, compared with the previous HPLC methods, the developed method has the advantages of mild derivatization temperature, short derivatization time and high sensitivity.

Development and validation of a novel high performance liquid chromatography-coupled with Corona charged aerosol detector method for quantification of glucosamine in dietary supplements.

IntroductionGlucosamine dietary supplements are commonly used for the management of osteoarthritis (OA). However, clinical trials have reported varying outcomes with regard to joint function and disease progression. One of the possible reasons for variability in observed effects of glucosamine could be that, unlike prescription drugs, the quality of manufactured dietary supplements is not closely monitored in many countries. Therefore, there is the possibility that the actual amount of glucosamine present in a dietary supplement is different from that claimed on the label. The quality control of glucosamine supplements is further complicated by the unavailability of a simple and effective analytical method for the analysis of glucosamine. Therefore, the aim of this study was to develop a simple analytical method that could be easily adapted by the pharmaceutical industry for routine analysis of glucosamine.AimsTo develop a novel high-performance liquid chromatography (HPLC) method for the quantification of glucosamine, and determine the amount of glucosamine present in a sample of dietary supplements commercially available in Australia and India.MethodsChromatographic separation of glucosamine was achieved using a zwitter-ionic hydrophilic interaction liquid chromatography column with a mobile phase consisting of 60% acetonitrile and 40% of 85 mM ammonium acetate, at a flow rate of 0.3 mL/min and column temperature 40°C. The developed method was validated for intra- and inter-day linearity, accuracy, precision, and reproducibility. The newly-developed method was subsequently used to analyse 12 glucosamine supplements.ResultsThe developed method was selective for glucosamine, which had a retention time of 5.9 min. The standard curve was linear with a correlation coefficient (r2) exceeding 0.99, over the range of 10–200 μg/mL for glucosamine. The relative standard deviations for intra- and inter-day accuracy, precision and reproducibility were all less than 4%. The amount of glucosamine determined in six Australian and six Indian glucosamine supplements ranged between 98.7–101.7% and 85.9–101.8% of the labelled values, respectively.DiscussionUnlike previous HPLC methods, this newly-developed HPLC technique does not require pre-derivatisation and can separate glucosamine from both hydrochloride and sulphate salts, and from other amino sugars, such as chondroitin sulphate present in dietary supplements. This simple and effective technique can be employed by analytical laboratories for the quality control of glucosamine dietary supplements.ConclusionThe current study has developed a new analytical technique using HPLC-Corona CAD, which can analyse underivatised glucosamine hydrochloride and sulphate within 6 minutes. Using the novel assay, we confirmed that unlike the tested Australian dietary supplements, only half of the tested Indian products had a glucosamine content within ±10% of what was claimed on the label.

Potential pitfalls in the diagnosis of Hb Handsworth in areas with high prevalence of HbS

Hb Handsworth is a rare α-globin structural variant caused by a missense mutation either on the α2 or α1-globin gene (HBA2 or HBA1: c.55G>C, p.Gly18Arg). This variant might be erroneously diagnosed as HbS unless secondary confirmative tests are carried out. We encountered a child with a prominent peak eluting in the 'S' window on high-performance liquid chromatography (HPLC). Sickle solubility test, gel electrophoresis, and selective direct nucleotide sequencing of α1, α2, and β globin genes were performed on the patient's sample. In addition, previous HPLC results on a cord blood sample were retrieved. Sickle solubility test was negative. Gel electrophoresis revealed a band migrating at the S region with an extra faint band seen on acid gel electrophoresis. Molecular analysis of α2 globin gene revealed heterozygous state of Hb Handsworth. Hb Handsworth is a rare variant that can mimic HbS on HPLC. Failure to recognize this rare variant in regions where HbS is highly prevalent may result in serious misdiagnosis and subsequent incorrect genetic counseling.

Sequential injection chromatography with a miniaturized multi-channel fiber optic detector for separation and quantification of propranolol and hydrochlorothiazide.

BackgroundSequential injection chromatography (SIC) is a new alternative separation technology. It has some advantages over high performance liquid chromatography (HPLC) regarding simplicity, inexpensiveness, portability, ease of use, maintenance requirement and operation time. In contrast, SIC has had suffered from some limitations.ResultsThe current work involves four achievements. (a) One of the limitations of SIC has been overcome. A higher pressure resistant selection valve with additional ports was installed in an SIC system. This development allows propelling solution without showing solution leakage. (b) A new inexpensive rapid and green method for the separation and quantification of propranolol (PRP) and hydrochlorothiazide (HTZ) in their formulations was optimized and validated. (c) A miniaturized multi-channel fiber optic detector was coupled with the newly developed SIC system to detect PRP and HTZ at 270 and 290 nm, respectively. This issue enhanced the sensitivity rather than using a single-channel detector. (d) A comparative study on the efficiency of the SIC method with that of previous HPLC methods was conducted.ConclusionsBesides the benefits of the instrumentation of SIC, the proposed method is rapider and more reagent-saving than previous HPLC methods. The total volume of consumed reagents and sample was 4.04 mL. The sample frequency was 22 samples/h. Other such analytical characters of the SIC method as resolution, peak symmetry, numbers of theoretical plates, linearity range, accuracy, precision and limits of detection and quantification recorded comparable results.

Simple and Efficient Method for the Analysis of Transesterification Reaction Mixtures for Biodiesel Production by Reversed-Phase High Performance Liquid Chromatography

Isocratic reversed-phase high performance liquid chromatography (HPLC) methods have been developed for the analysis of biodiesel methyl esters and intermediate species associated with transesterification reactions. While most HPLC methods for biodiesel analysis employ gradient elution and ultraviolet (UV) detection, these methods employ isocratic elution with refractive index detection (RID). For the separation of methyl esters, monoglycerides, and free fatty acids, a mobile phase of 85% acetonitrile + 15% deionized water is used. Diglycerides can be separated using 100% methanol, and triglycerides can be quickly analyzed using a mixture of 60% 2-propanol + 40% methanol. While previous HPLC methods for methyl ester analysis have been shown to successfully separate components found in biodiesel mixtures, the use of UV detection is inadequate for the detection of species with saturated carbon chains. Improved method for direct high-performance liquid chromatography assay of angiotensin-converting enzyme-catalyzed reactions

A rapid and sensitive assay was developed for determination of the activity of angiotensin-converting enzyme (ACE) in the presence of inhibitory peptides present in soybean protein hydrolysates. The method utilizes reversed-phase high-performance liquid chromatography (HPLC) to separate and quantify hippuryl-histidyl-leucine (HHL) and hippuric acid (HA). HHL and HA were separated on a Symmetry C 18 column by gradient elution that used mixtures of trifluoroacetic acid (TFA)–acetonitrile and TFA–water as solvents. Analytical time and baseline separation of HA from HHL were improved over previous HPLC methods. In comparison to the standard spectrophotometric method, the new HPLC method obviates the need for ethyl acetate extraction of HA but requires direct injection of the ACE reaction mixture onto the HPLC column.

HPLC analysis of complete BZ systems. Evolution of the chemical composition in cerium and ferroin catalysed batch oscillators: experiments and model calculations.

In the last few years many new reaction routes and intermediates have been discovered in the mechanism of the Belousov-Zhabotinsky (BZ) reaction with the aid of high performance liquid chromatography (HPLC). These previous HPLC studies, however, were limited to the Ce(4+)-organic substrate (malonic or bromomalonic acid) systems only. Very recently some measurements were made on a cerium catalysed full BZ system but only in its induction period. The present work follows the evolution of the main chemical components in a cerium and in a ferroin catalysed full BZ system from the start until the end of the oscillatory regime in a batch reactor. While recording the potential oscillations of a bromide selective electrode we measured from time to time the concentration of the following components: malonic and bromomalonic acids and bromate as main components; malonyl malonate, ethanetetracarboxylic and bromoethenetricarboxylic acids which are recombination products of organic free radicals; oxidized intermediates: tartronic, oxalic (OA) and mesoxalic (MOA) acids, and brominated products: dibromoacetic and tribromoacetic acids. Recombination products are generated in the intervals when the autocatalytic reaction is "switched off". In the course of the autocatalytic periods, however, the organic radicals react with the inorganic bromine dioxide radical mainly which leads to the formation of MOA and OA. Due to a very fast Ce(4+)-MOA reaction, MOA can be detected in the ferroin catalysed BZ system only. Our model calculations deal exclusively with the cerium catalysed system. The suggested new Marburg-Budapest-Missoula (MBM) model includes both negative feedback loops (bromous acid-bromide ion Oregonator type and bromine dioxide-organic free radicals Radicalator type feedback) and the recently discovered radical-radical recombination reactions. Comparison of the experimental data with the model calculations shows a good qualitative agreement but some open problems still remain. To overcome these problems oxygen atom transfer and other redox reactions are proposed.

Identification of by-products from an orthogonal peptide ligation by oxime bonds using mass spectrometry and tandem mass spectrometry.

Synthetic proteins with unusual architecture are obtained through chemoselective ligation, a method based on the condensation of unprotected peptides under mild aqueous conditions. The last step of a new procedure leading to a tri-branched conjugate consists of the chemoselective ligation reaction between an (aminooxy)acetyl peptide and a peptide aldehyde resulting from a first ligation via an oxime bond. In order to optimize the reaction conditions, electrospray ionization mass spectrometry combined with liquid chromatography and tandem mass spectrometry has been used. In addition to the target tri-branched conjugate, two other conjugates were characterized allowing documentation of transoximation reactions in peptide chemistry. A fourth conjugate was identified as a side product appearing after the first ligation. Data obtained by low-energy collision-induced dissociation led to a rapid and reliable identification of impurities observed in the (aminooxy)acetyl peptide despite a previous high performance liquid chromatography (HPLC) purification. This highlights the great reactivity of the aminooxy group towards carbonyl-containing compounds.

Optimization of mobile phase in the separation of β-blockers by HPLC

β-blockers are generally determined using high-performance liquid chromatography (HPLC). Previous HPLC separations of β-blockers have often required a mobile phase containing three components; acetonitrile or methanol to control the retention; buffer to control the ionic strength and pH of the mobile phase; ion-pairing reagent to provide adequate retention of β-blockers or organic amines as masking agent to reduce peak tailing. Due to the complexity of the mobile phases employed, development of these assays can be a laborious process. Additionally, alkyl sulphonates and organic amines dramatically reduces the life-time reduction of silica based C 18 columns. The results of this study demonstrated that the addition of tested alkyl sulphonates and organic amines is not essential for an adequate separation of β-blockers. In this study, we developed a simple HPLC method for the simultaneous separation of model β-blockers, atenolol, practolol, metoprolol, oxprenolol and propranolol. Atenolol, practolol, metoprolol, oxprenolol and propranolol adequately separated with high peak symmetries using a mobile phase consisted of methanol/acetonitrile/phosphate buffer (10 mM, pH 3.0) (15:15:70, v/v/v). By altering only the fraction of methanol with respect to acetonitrile, method development becomes a more efficient separation. Furthermore, atenolol, practolol, metoprolol, oxprenolol and propranolol can be detected up to 0.25, 5, 10, 50 and 10 ng ml −1. In this publication, we present the simultaneous separation of β-blockers having a wide range of polarity. It is proposed that this new mobile phase, consisting only acetonitrile, methanol and phosphate buffer can be used for the analysis of the several β-blockers presently in doping control analysis as well as others.

Simple and versatile high-performance liquid chromatographic method for the simultaneous quantitation of the lactone and carboxylate forms of camptothecin anticancer drugs.

The well documented hydrolysis of the alpha-hydroxy-delta-lactone ring moiety in camptothecin and related analogues is routinely monitored using high-performance liquid chromatography (HPLC). Previous HPLC separations of the lactone and carboxylate forms of camptothecins have often required mobile phases containing three to four components; ion-pairing reagent to provide adequate retention of the carboxylate form of the drug; buffer to control the ionic strength and pH of the mobile phase; acetonitrile to control the retention of the lactone form and, in some instances, sodium dodecyl sulfate to reduce peak tailing. Because of the complexity of the mobile phases employed, development of these assays can be a laborious process, requiring re-optimization for each new analogue. In this study, we have developed a simple HPLC methodology for the simultaneous separation of the lactone and carboxylate forms of numerous camptothecin analogues. The mobile phase employed includes only triethylamine acetate (TEAA) buffer and acetonitrile. In this application, triethylamine serves multiple roles; as the ion-pairing reagent, as a masking agent for underivatized silanols and as the major buffer component. By altering only the composition of TEAA buffer with respect to acetonitrile, method development becomes a more streamlined and time efficient process. In this publication, we present the simultaneous separation of the lactone and carboxylate forms of camptothecin and four related analogues, namely, topotecan, GI147211, 10-aminocamptothecin and the CPT-11-SN-38 drug-metabolite pair. It is proposed that this new mobile phase, consisting of only triethylamine acetate buffer and acetonitrile, can be used for the analysis of the several camptothecin derivatives presently in clinical trials as well as the numerous other analogues in preclinical development.

A Non—Ion-Pairing HPLC Method for Measuring New Forms of Ascorbate and Ascorbic Acid

Abstract A new high-performance liquid-chromatography (HPLC) method separates vitamins L-ascorbic acid (C1) and L-ascorbyl-2-sulfate (C2). Previous HPLC reverse phase methods utilized ion-pairing reagents and resulted in co-elution with other substances. Vitamin C1 preceded C2 in retention time. In this new method, which deletes the ion-pairing reagent, vitamin C2 precedes the elution of vitamin C1. This new direct method allows for a better separation from other conflicting metabolites and constituents of extraction mixtures. In addition, this method provides a way of identifying ascorbyl-2-monophosphate (C3). a form of vitamin C that may be found in animal diets. Examples will be shown of HPLC profiles of extractives from tissue and diets known to contain these new forms of vitamin C.

Semi-preparative purification and characterization of peptides from complex haemoglobin hydrolysate by HPLC-mass spectrometry

An important goal of previous high-performance liquid chromatography (HPLC) studies was the development of a simple procedure for the purification of peptides from very complex haemoglobin enzymatic hydrolysates. This report demonstrates that the separation system described can be used for large sample loadings at laboratory preparative scale (more than 50 mg) without loss of resolution. The efficiency of peptide purification was shown by fast atom bombardment-mass spectrometry (FAB-MS) and tandem mass spectrometry (tandem M S). The procedure described will be of interest in the biotechnology area for the extensive preparation of peptides for fine applications from complex enzymatic hydrolysates.
