Preventive effects of taurine against d-galactose-induced cognitive dysfunction and brain damage.

Oxidative stress arising from life processes or environmental influences and its resultant cellular dysfunctions are major causes of neurodegenerative disorders. The objectives of this study were to investigate whether taurine (Tau) can prevent d-galactose-induced cognitive dysfunction and brain oxidative damage. Mice given with Tau supplementation (100 and 400 mg per kg BW per day) spent shorter (p < 0.05) time in searching target in d-galactose (100 mg per kg BW per day) treated mice in a water maze reference memory experiment. Moreover, Tau supplementation extended (p < 0.05) the searching period around the target quadrant in the probe test of the water maze, and neuronal degeneration and nucleus shrinkage in the hippocampus dentate gyrus area of d-galactose treated mice were observed to be attenuated. Tau also downregulated (p < 0.05) expression of the glial fibrillary acidic protein (Gfap) and of the cluster of differentiation marker Cd11b; meanwhile, it strengthened (p < 0.05) antioxidant capacity and lowered (p < 0.05) the accumulation of advanced glycation end-products (AGEs) in the brain. Therefore, Tau could be effective to ameliorate oxidative damage and inflammation in the brain, and apoptosis of brain cells, which further lessen the cognitive dysfunction.

Preventive effect of taurine on experimental type II diabetic nephropathy

BackgroundIt has been verified that taurine has some preventive effects on diabetes and its complications when used alone or together with other drugs, but there are few reports about taurine on the prevention of diabetic nephropathy, the mechanisms of which are still unknown.MethodsTaurine was administered to type Ⅱ diabetic rats induced by high fat high sugar diet combined with STZ injection. The preventive effect of taurine on diabetic nephropathy was investigated by detecting blood glucose, lipid metabolism, kidney function and glomerular basement membrane metabolism.ResultsTaurine could lower blood glucose, TG, TC, BUN, Scr, NAG, U-PRO, the expression of laminin B1( LBN1) mRNA, and increase HDL-C of diabetic rats.ConclusionsThe results indicated that taurine could prevent the occurrence and development of diabetic nephropathy by decreasing blood glucose, improving lipid metabolism, glomerular basement membrane metabolism, and kidney function.

Modulation of cytosolic and nuclear Ca2+ and Na+ transport by taurine in heart cells.

The effect of taurine on the different types of ionic currents appears to depend on [Ca]o and [Ca]i and may also vary accordingly to tissue or cell type studied. Using microfluorometry and Ca2+ imaging techniques, short-term exposure (5-10 min) of single heart cells to taurine was found to increase total intracellular free Ca2+ in a concentration-dependent manner. However, long-term exposure of heart myocytes to taurine was found to decrease both nuclear and cytosolic Ca2+ without significantly changing either nuclear or cytosolic Na+ levels, as measured by 3-dimensional Ca2+ and Na+ confocal imaging techniques. Long-term exposure to taurine was found to prevent cytosolic and nuclear increases of Ca2+ induced by permanent depolarization of heart cells with high [K]o. This preventive effect of taurine on nuclear Ca2+ overload was associated with an increase of both cytosolic and nuclear free Na+. Thus, the effect of long-term exposure to taurine on intranuclear Ca2+ overload in heart cells seems to be mediated via stimulation of sarcolemmal and nuclear Ca2+ outflow through the Na+-Ca2+ exchanger.

Taurine: a preventive agent of the acute ethanol depletive action on the isolated human amniotic membrane

The preventive effect of taurine towards the acute ethanol reduction action was studied on the ionic transfer through the isolated human amniotic membrane. Taurine increased 3 components of the ionic transfer expressed by the conductance measurements (Na(+) and K(+) paracellular conductances through the intercellular spaces and coupling cell factor between 2 adjacent epithelial cells, expressed by a voltage ratio). These components were decreased by ethanol. Electrophysiological studies (conductance and voltage measurements) indicated that the addition of taurine (0.1-1 mM) before ethanol (0.4 g/l) hindered the decrease action of ethanol on the Na(+) and K(+) paracellular conductances and on the coupling cell factor. These data indicated a common target between taurine and ethanol: the membraneous phospholipids, particularly the distribution of the external fixed charges. The preventive action of taurineversus ethanol, on the human amniotic membrane, was exerted on the polar groups of phospholipids, hindering the incorporation of ethanol molecules.

The excretion of PQ into the urine was unaltered. The infusion of glycine did not have such effects.
