The association between selective dry cow therapy, milking routine and dry cow management practices with somatic cell count in early lactation cows from 21 commercial grazing dairy herds

This observational study aimed to explore the association of farmer-driven selective dry cow therapy (DCT), milking routine and dry cow management practices with somatic cell count (SCC) in early lactation cows from 21 commercial dairy herds. Milking routine practices evaluated referred to cow preparation for milking, in-lactation mastitis management and recording. Dry cow management practices related to dry cow environment and cleaning, dry-off procedure, milk cessation strategy and calving environment. Data from 2,016 multiparous cows in 21 commercial spring-calving grazing dairy herds were available for the study. Herd owners self-reported DCT (the assignment and administration of DCT was at the discretion of the herd owners with no involvement from the research team), management practices during milking and the dry period. Cow-level last test-day SCC records in 2020 [range = 105 to 285 d in milk (DIM)] and first test-day records in 2021 (range = 5 to 60 DIM) were obtained from milk recording databases. Quarter-level milk sampling was carried out on all cows in late lactation of 2020 (range = 240 to 261 DIM) for bacterial culturing. Bacteriological results were used to define cows with intramammary infection (IMI) when ≥ 1 quarter sample resulted in bacterial growth and there were no contaminated samples from that cow. Mixed model analyses were used to evaluate the association of selective DCT, milking routine and dry cow management practices with cows' first test-day log 10 SCC (logSCC) in 2021. At dry-off in 2020, 47.6% of the cows were administered an internal teat sealant alone (ITS) while 52.4% were administered an antibiotic plus an internal teat sealant (AB+ITS). The mean herd-level percentage of cows with IMI was 19.7% (range = 9.8% to 39.5%); Staphylococcus aureus accounted for the majority of cow-level IMI (89.9%, 357/397). Between herds, the proportion of cows administered ITS ranged from 17.7% (14/79; in a herd with an IMI prevalence of 10.1%) to 86.8% (66/76; in a herd with an IMI prevalence of 27.6%). In total, 11.8% (105/889) and 29.8% (292/980) of cows that were administered ITS or AB+ITS had an IMI in late lactation 2020, respectively. The mean untransformed SCC at the last test-day in 2020 of cows administered ITS and AB+ITS was 55,000 and 197,200 cells/mL, respectively. The statistical analysis showed a significant interaction between selective DCT and milk yield at last test-day in 2020; cows with a milk yield of 15 kg and administered ITS had a 0.1 higher (untransformed SCC of 19,000 cells/mL higher) first test-day logSCC compared with cows administered AB+ITS. Additionally, greater parity, IMI in late lactation, higher log SCC at the last test-day in 2020 and longer dry periods were associated with higher logSCC at the first test-day in 2021. The current study identified cow- and herd-level management practices that could aid dairy farmers in improving the outcome of selective DCT and decrease early lactation SCC.

Pathogen group-specific risk factors for intramammary infection in water buffalo.

A cross-sectional study was conducted to estimate the prevalence of intramammary infection (IMI) associated bacteria and to identify risk factors for pathogen group-specific IMI in water buffalo in Bangladesh. A California Mastitis Test (CMT) and bacteriological cultures were performed on 1,374 quarter milk samples collected from 763 water buffalo from 244 buffalo farms in nine districts in Bangladesh. Quarter, buffalo, and farm-related data were obtained through questionnaires and visual observations. A total of 618 quarter samples were found to be culture positive. Non-aureus staphylococci were the predominant IMI-associated bacterial species, and Staphylococcus (S.) chromogenes, S. hyicus, and S. epidermidis were the most common bacteria found. The proportion of non-aureus staphylococci or Mammaliicoccus sciuri (NASM), S. aureus, and other bacterial species identified in the buffalo quarter samples varied between buffalo farms. Therefore, different management practices, buffalo breeding factors, and nutrition were considered and further analyzed when estimating the IMI odds ratio (OR). The odds of IMI by any pathogen (OR: 1.8) or by NASM (OR: 2.2) was high in buffalo herds with poor milking hygiene. Poor cleanliness of the hind quarters had a high odds of IMI caused by any pathogen (OR: 2.0) or NASM (OR: 1.9). Twice daily milking (OR: 3.1) and farms with buffalo purchased from another herd (OR: 2.0) were associated with IMI by any pathogen. Asymmetrical udders were associated with IMI-caused by any bacteria (OR: 1.7). A poor body condition score showed higher odds of IMI by any pathogen (OR: 1.4) or by NASM (OR: 1.7). This study shows that the prevalence of IMI in water buffalo was high and varied between farms. In accordance with the literature, our data highlight that IMI can be partly controlled through better farm management, primarily by improving hygiene, milking management, breeding, and nutrition.

Intramammary infections in lactating Jersey cows: Prevalence of microbial organisms and association with milk somatic cell count and persistence of infection

There are limited data available regarding pathogens causing intramammary infections (IMI) in Jersey cows. The objectives of this study were to characterize the prevalence of IMI caused by different microorganisms in lactating Jersey cattle and evaluate the associations among microbes and somatic cell count (SCC) and persistence of IMI. This prospective, observational, longitudinal study included lactating Jersey cows (n = 753) from 4 farms within a 415 km radius of Columbia, Missouri. Quarter foremilk samples were aseptically collected monthly for 3 consecutive months. Microorganisms were identified using aerobic milk culture and MALDI-TOF mass spectrometry. A commercial laboratory measured SCC using flow cytometry. Milk culture results were used to classify single microorganism infections as persistent (same microorganism species identified at first sampling and one other sampling) or nonpersistent infection. Mixed models were built to evaluate the associations between IMI status and SCC natural logarithm (lnSCC), as well as persistence and lnSCC. Overall, staphylococci were the most commonly isolated microorganisms among the 7,370 quarter-level milk samples collected. Median prevalence (using all 3 samplings) of specific microbes varied among farms; however, Staphylococcus chromogenes was a common species found at all farms. The most common microbial species that persisted were Staph. chromogenes, Staphylococcus aureus, Staphylococcus simulans, and Streptococcus uberis. Streptococcus dysgalactiae and Staph. aureus were the IMI associated with the most inflammation based on lnSCC. The small number of herds included in this study with the large variation in herd type limits the generalizability of the data. However, results of this study seem to be similar to those of previous studies in other breeds, suggesting management factors are more important than breed-specific differences when evaluating causes of IMI and associated subclinical mastitis.

Udder Hygiene and Mastitis Indicators in Contrasting Environmental Conditions during Half-Time Confinement in Pasture-Based Dairy Systems.

This study aimed to compare the association between two types of housing, compost barns (CB) vs. outdoor soil-bedded yard (OD), and udder hygiene and mastitis indicators in Holstein dairy cows calving in autumn (n = 31) and spring (n = 27). After calving, cows were transferred to a pasture-based system with half-time confinement in one of two treatments: CB or OD. The udder hygiene score (UHS) was evaluated monthly and on days after rainfall over the entire lactation period. Individual somatic cell count (SCC) was determined throughout lactation, and the prevalence of intramammary infection (IMI) was estimated. Cows confined in OD presented higher UHS compared to cows in CB (p < 0.05) in both calving seasons. After rains, autumn-calving cows in OD were dirtier than on days without previous rain (OR = 1.85, CI 95%: 1.1-3.1; p < 0.02). However, no differences in IMI and clinical mastitis were found between OD and CB cows in either calving season.

Staphylococcus aureus adlb gene is associated with high prevalence of intramammary infection in dairy herds of northern Italy: A cross-sectional study

Staphylococcus aureus is a major mastitis pathogen in dairy cattle worldwide, responsible for substantial economic losses. Environmental factors, milking routine, and good maintenance of milking equipment have been described as important factors to prevent intramammary infections (IMI). Staphylococcus aureus IMI can be widespread within the farm or the infection can be limited to few animals. Several studies have reported that Staph. aureus genotypes differ in their ability to spread within a herd. In particular, Staph. aureus belonging to ribosomal spacer PCR genotype B (GTB)/clonal complex 8 (CC8) is associated with high within-herd prevalence of IMI, whereas other genotypes are generally associated with individual cow disease. The adlb gene seems to be strictly related to Staph. aureus GTB/CC8, and is a potential marker of contagiousness. We investigated Staph. aureus IMI prevalence in 60 herds in northern Italy. In the same farms, we assessed specific indicators linked to milking management (e.g., teat condition score and udder hygiene score) and additional milking risk factors for IMI spread. Ribosomal spacer-PCR and adlb-targeted PCR were performed on 262 Staph. aureus isolates, of which 77 underwent multilocus sequence typing. In most of the herds (90%), a predominant genotype was identified, especially Staph. aureus CC8 (30%). In 19 of 60 herds, the predominant circulating Staph. aureus was adlb-positive and the observed IMI prevalence was relevant. Moreover, the adlb gene was detected only in genotypes of CC8 and CC97. Statistical analysis showed a strong association between the prevalence of Staph. aureus IMI, the specific CCs, and carriage of adlb, with the predominant circulating CC and presence of the gene alone explaining the total variation. Interestingly, the difference in the odds ratio obtained in the models for CC8 and CC97 suggests that it is carriage of the adlb gene, rather than the circulation of these CCs per se, that leads to higher within-herd prevalence of Staph. aureus. In addition, the model showed that environmental and milking management factors had no or minimal effect on Staph. aureus IMI prevalence. In conclusion, the circulation of adlb-positive Staph. aureus strains within a herd has a strong effect on the prevalence of IMI. Thus, adlb can be proposed as a genetic marker of contagiousness for Staph. aureus IMI in cattle. However, further analyses using whole-genome sequencing are required to understand the role of genes other than adlb that may be involved in the mechanisms of contagiousness of Staph. aureus strains associated with high prevalence of IMI.

To seal or not to seal following an antimicrobial infusion at dry-off? A systematic review and multivariate meta-analysis of the incidence and prevalence of intramammary infections post-calving in dairy cows

Teat sealants (TSs) consist of sterile formulations with no antibacterial activity. Alone or in combination with antimicrobial (AM) or non-AM treatments, TSs have been commonly used in dairy cows at dry-off to prevent intra-mammary infections (IMIs) during the dry period. This study aimed to identify and synthesise the available evidence on the efficacy of combining TSs with AM treatments on the incidence and prevalence of IMIs. A comprehensive search of three electronic databases, two relevant conference proceedings, and reference lists of reviews and eligible articles was conducted to retrieve and identify studies that could answer the following question: in dairy cows, how does the efficacy of an AM–TS combination administered at dry-off compare with an AM alone for preventing new IMI? In addition to the general IMIs, bacterial species-specific data were extracted and combined into nine distinct pathogen groups: coagulase-positive and negative staphylococci; S. dysgalactiae; non-dysgalactiae Streptococci; E. coli; non-E. coli Enterobacteriaceae; Corynebacterium spp.; yeast and other frequent mastitis pathogens. The structural relationship between each study's prevalence and incidence, as the new (incidence) and persistent (uncured) infections make up the prevalence, was utilised to approximate a variance-covariance matrix for the within-study correlation between their study-specific log odds ratios (ORs). A bivariate random-effects meta-analysis was employed, utilising the within- and between-study correlations to synthesise both outcomes simultaneously. The risk of bias was assessed using the Cochrane ROBINS-I tool, and the quality of the body of evidence was rated using the GRADE approach. A total of 17 trials (16 studies), providing either IMIs incidence (n = 4), prevalence (n = 3) or both (n = 10), were identified. Overall, quarters infused with AM-TS combinations showed lower odds of new IMIs post-calving (OR=0.70; 95% CI=0.57–0.86; Wald test P < 0.001) than those which received only AMs. Across the pathogen groups, varying levels of reduction of new IMIs were found, where administration of TSs was most effective against S. dysgalactiae (OR=0.47; 95% CI=0.23–0.98), non-dysgalactiae streptococci (OR=0.60; 95% CI=0.49–0.74), E. coli (OR=0.62; 95% CI=0.50–0.77), Corynebacterium spp. (OR=0.68; 95% CI=0.52–0.90) and coagulase-negative staphylococci (OR=0.85; 95% CI=0.76–0.94). However, additional TS infusion did not significantly reduce new IMIs in the remaining pathogen groups. The current meta-analytic evidence supports the efficacy of using TS add-on infusions in dairy cows at dry-off for reducing the incidence and prevalence of IMIs post-calving; however, pathogen group differences should be considered.

Selective dry-cow therapy can be implemented successfully in cows of all milk production levels

Antibiotic stewardship on dairy farms can be heightened through the implementation of selective dry-cow therapy (SDCT). However, some producers are concerned that this practice may be related to poor udder health outcomes in cows with high milk production at the time of dry-off. The objective of this study was to evaluate if the effect of culture-guided SDCT (Cult-SDCT) and algorithm-guided SDCT (Alg-SDCT) on dry-period intramammary infection (IMI) dynamics and postcalving udder health and performance [when compared with blanket dry-cow therapy (BDCT)] varied according to milk production level before dry-off. Data were compiled from clinical trials conducted in the United States and Canada that compared Cult-SDCT and Alg-SDCT to a positive control, i.e., BDCT. In those trials, cows were enrolled 1-2 d before dry-off, randomized to their dry-cow therapy strategy and followed until 120 d in milk of the subsequent lactation. The number of cows and quarters in the final data set were 1,485 and 5,097, respectively. Measured outcomes included quarter-level antibiotic use at dry-off, quarter-level IMI prevalence after calving, quarter-level dry-period IMI cure risk, quarter-level dry-period new IMI risk, cow-level clinical mastitis and removal from the herd during 1-120 d in milk, and somatic cell count and milk yield during 1-120 DIM. The primary objective of analysis was to investigate if the effect of Cult-SDCT and Alg-SDCT on these outcomes, when compared with BDCT, varied according to milk production level before dry-off. To do this, each cow was classified as having low, mid or high production, based on her milk yield tertile group at the most recent herd test before enrollment (low: <23.7 kg/d, mid: 23.7 to 30.4 kg/d, and high >30.4 kg/d). Multivariable generalized estimating equations were used to estimate risk differences and differences in means, and Cox regression was used to estimate hazard ratios. For Cult-SDCT, the proportion of quarters treated with dry-cow antibiotics within each milk production level were 40.7% (low), 41.7% (mid) and 47.2% (high). For Alg-SDCT, the proportions were 60.6% (low), 38.7% (mid), and 35.1% (high). Measures of udder health were not markedly different when comparing Cult-SDCT to BDCT and Alg-SDCT to BDCT. This was consistently observed in low, mid and high producing cows. In conclusion, the findings from this study indicate that Cult-SDCT and Alg-SDCT can be successfully implemented in cows of all milk production levels.

ABSTRACT The cow somatic cell count (CSCC) is an indicator of intramammary infection (IMI), and it has been used to make decisions at dry-off about selective dry cow therapy (SDCT). This study assessed ex-ante the economic impact of implementing SDCT under milk production conditions in Argentina, using CSCC to identify cows with IMI at dry-off. Eighty six cows were sampled at dry-off and considered being infected if at least one quarter was infected with major mastitis pathogens. The CSCC sensitivity and specificity were estimated using the CSCC recorded from the test-day prior to dry-off. A simulation model was then developed to look into the economic benefits of applying SDCT under two herd level IMI prevalence (low: 5-15%, and high: 16-25%) at dry-off. The input variables were obtained from the field study, scientific literature, and real-world prices. The output was the net economic difference (NED) between SDCT and blanket dry cow therapy at the herd level. The sensitivity and specificity estimated for identifying cows infected with major pathogens for a CSCC threshold of 200,000 cells/mL were 31.4 (95% CI, 14.6-48.2) and 64.7 (95% CI, 50.6-78.8), respectively; the value for NED was maximized to -634 US$ and -455 US$ in the low and high IMI prevalence, respectively. In general, CSCC specificity was the variable with the most impact on NED and, in high IMI prevalence, milk price had a similar relevance. Thus, SDCT based on CSCC is a cost-effective option, under the milk production conditions in Argentina.

Association of Staphylococcus aureus genotypes with milk or colonization of extramammary sites in Dutch dairy cattle indicates strain variation in reservoirs for intramammary infections

Staphylococcus aureus (S. aureus) is a major mastitis pathogen with a detrimental impact on udder health in dairy cattle. Although S. aureus is generally classified as a contagious mastitis pathogen, control measures aimed at preventing contagious transmission are not always effective. Previous studies showed that various extramammary sites can be colonized with S. aureus and could be a reservoir for S. aureus intramammary infections (IMI). The aim of this study was to determine the prevalence of S. aureus extramammary site colonization in Dutch dairy cattle and to compare the spa genotypes of S. aureus isolates from IMI to spa genotypes of isolates from extramammary sites. Six herds were visited and from cows with a composite milk somatic cell count ≥200,000 cells/mL quarter milk samples and swabs from various extramammary sites (hock, groin, udder cleft, nares, and feces) were taken. Extramammary site samples were processed by a two-step high salt selective culture and presence of S. aureus was confirmed by femA PCR. S. aureus isolates from milk and extramammary sites were compared by spa typing. The cow level colonization varied from 0% to 73%, and the prevalence of IMI in the sampled cows varied from 0% to 61% between herds. The extramammary site with the highest prevalence of colonization was the hock (23%) and the lowest prevalence of colonization was found for the nares (5%) and feces (5%). Spa typing of S. aureus isolates from either extramammary sites or milk showed that in most herds there were one or two predominant S. aureus spa genotypes present. Different S. aureus spa genotypes could be categorized into three groups based on the distribution between milk or extramammary sites: i) predominantly milk associated, ii) associated with both milk and extramammary sites, and iii) associated with extramammary sites. In conclusion, we showed that the prevalence of extramammary site colonization differed significantly between herds and extramammary sites and that specific S. aureus spa genotypes were associated with milk (IMI) or extramammary site colonization. Extramammary S. aureus reservoirs could be a source for IMI that cannot be eradicated by intervention measures aimed at contagious mastitis pathogens.

Characteristics of mammary secretions collected from infected and uninfected primigravid dairy heifer mammary glands.

Intramammary infections (IMI) in primigravid dairy heifers can affect mammary growth and development, which can reduce first-lactation milk yield. Detection of IMI in heifers most often involves the use of culture-based methods that are not often used in production dairy settings given their labor- and time-consuming nature. The objective of this study was to determine whether mammary secretion somatic cell count (SCC) and viscosity were associated with the infection status of primigravid heifer mammary glands. A total of 270 heifers from a single farm were used, selected based on the farmer's willingness to participate. The study was conducted from June to October 2020. Mammary secretion samples were aseptically collected from a randomly selected quarter of each heifer at 75 d prepartum (75PP), and another quarter of each heifer was sampled at 35 d prepartum (35PP). The remaining 2 quarters of each heifer were not examined. Mammary secretion samples underwent bacteriological examination to determine IMI status and quantitative SCC measurement and were also assessed for secretion viscosity based on visual observation. Prevalence of IMI was 26% (69/270) and 28% (71/255) at 75 and 35 d prepartum, respectively. Uninfected secretion samples had 133.2 [95% confidence interval (CI): 16.8 to >999.9] times greater odds to be thick compared with samples infected with a major pathogen, and 14.4 (95% CI: 8.5 to 24.1) times greater odds to be thick compared with samples infected with non-aureus staphylococci (NAS). The mean secretion SCC of uninfected quarters (6.04 ± 0.03 log10 cells/mL) was significantly lower than that of secretions collected from quarters infected with Staphylococcus chromogenes (6.34 ± 0.04 log10 cells/mL), other NAS species (6.28 ± 0.10 log10 cells/mL), or a major pathogen (6.73 ± 0.08 log10 cells/mL). These results indicate that mammary secretion viscosity and SCC measurement may be useful tools in identifying primigravid heifer quarters with IMI. The ability to evaluate viscosity at time of sampling may be a useful strategy that could be incorporated into interventions designed to diminish the negative effects of prepartum IMI on lactational performance.

Invited review: Selective use of antimicrobials in dairy cattle at drying-off.

Administering intramammary antimicrobials to all mammary quarters of dairy cows at drying-off [i.e., blanket dry cow therapy (BDCT)] has been a mainstay of mastitis prevention and control. However, as udder health has considerably improved over recent decades with reductions in intramammary infection prevalence at drying-off and the introduction of teat sealants, BDCT may no longer be necessary on all dairy farms, thereby supporting antimicrobial stewardship efforts. This narrative review summarizes available literature regarding current dry cow therapy practices and associated impacts of selective dry cow therapy (SDCT) on udder health, milk production, economics, antimicrobial use, and antimicrobial resistance. Various methods to identify infections at drying-off that could benefit from antimicrobial treatment are described for selecting cows or mammary quarters for treatment, including utilizing somatic cell count thresholds, pathogen identification, previous clinical mastitis history, or a combination of criteria. Selection methods may be enacted at the herd, cow, or quarter levels. Producers' and veterinarians' motivations for antimicrobial use are discussed. Based on review findings, SDCT can be adopted without negative consequences for udder health and milk production, and concurrent teat sealant use is recommended, especially in udder quarters receiving no intramammary antimicrobials. Furthermore, herd selection should be considered for SDCT implementation in addition to cow or quarter selection, as BDCT may still be temporarily necessary in some herds for optimal mastitis control. Costs and benefits of SDCT vary among herds, whereas impacts on antimicrobial resistance remain unclear. In summary, SDCT is a viable management option for maintaining udder health and milk production while improving antimicrobial stewardship in the dairy industry.

Potential impacts to antibiotics use around the dry period if selective dry cow therapy is adopted by dairy herds: An example of the western US

Mastitis is a prevalent and expensive disease in dairy herds worldwide. Blanket dry cow therapy (BDCT), in which all quarters of all cows are infused with antimicrobials at the dry-off, is a cornerstone for mastitis control in many countries. An alternative approach is the use of selective dry cow therapy (SDCT), in which only cows with high risk for intramammary infection (IMI) at dry-off receive antimicrobials. Our objectives in this cross-sectional study were to estimate the potential reduction in the use of antimicrobials if SDCT was adopted in the US by using cow-level dairy herd data and to describe the factors associated with cows being classified as high-risk for an IMI at dry-off. Besides, we aimed to describe the seasonality in IMI at dry-off. We used cow-level somatic cell score (SCS) test-day data from herds in the western US (DHIA, Dairy Herd Improvement Association, AgriTech, Visalia, CA) to create five scenarios to classify cows as high risk for IMI at dry-off. Associations between cow-level data and state were also used in logistic regression models. We also calculated the average animal-defined daily dosage of antimicrobials per cow per year around the dry period if a BDCT or SDCT approach is used, adjusting for the risk of cases of clinical mastitis in the next lactation for the SDCT approach. The point prevalence of IMI at the last test-day before dry-off varied between 15.0 % and 54 % for primiparous and 34.0 % and 85 % for multiparous cows, depending on the scenario. By extrapolating our results obtained from using data from dairies enrolled in the DHIA testing program for the western US, we demonstrated that regardless of the criteria used to classify cows as high risk of IMI at dry-off (scenarios 1–5), if selective dry cow therapy is adopted in the US, the dairy industry could reduce the use of antimicrobials around the dry-off between 31 % and 66 %. Multiparous cows had greater odds of being classified as high-risk than primiparous cows. Cows dried off in the spring, summer, and fall had lower odds of being classified as high-risk compared to cows dried off in the winter. Advanced days in milk at dry-off was associated with greater risk of IMI at dry-off. Greater milk yield and higher protein percentage at the last test-day before dry-off were associated with decreased odds of a cow being classified as high-risk at dry-off, cows in small herds had greater odds of being classified as high-risk at dry-off, and a variation among states was observed.

Productive and reproductive performance of crossbred dairy heifers with induced lactation and efficacy of antimicrobial therapy associated with internal teat sealants

This study evaluated (a) the efficacy of an association between injectable antibiotic therapy and sealant (ATBS) on milk yield (MY), somatic cell count (SCC), and prevalence of intramammary infections (IMI); and (b) the efficacy of gonadotropin-releasing hormone (GnRH) on follicular cyst (FCs) resolution (cyclicity at the 45th day in milk; DIM) and cumulative pregnancy rate (CPR) in heifers submitted to a lactation induction protocol (LIP). A total of 114 crossbred (Holstein × Jersey) heifers, with 34.7 ± 4.8 months and 439 ± 56.35 kg were submitted to LIP. On the 5th day of the LIP, the heifers were assigned to (i) ATBS (n = 57) with 7 mg/kg of norfloxacin associated with sealant and (ii) Control 1 (n = 57; CONT1) with no treatments. Lactation began on the 21st day of LIP and the 15th DIM, FCs were diagnosed and 106 heifers were randomized into two treatment groups with 53 heifers each: (i) GnRH (5 mL injectable GnRH) and (ii) Control 2 (CONT2; no treatment). Of the 114 heifers initially induced, 83.33% (n = 95) responded to LIP with an average MY of 15.19 kg/milk/day during 22 weeks of lactation. In the first 14 DIM, the IMI prevalence was 18% and 28% for heifers ATBS and CONT1 treated, respectively. Additionally, coagulase-negative Staphylococcus was the most frequently isolated group of pathogens. Mammary quarters that received ATBS treatment had a lower risk of IMI and SCC than CONT1. The cyclicity at 45 DIM was 68% (ATBS) and 35% (CONT1), and 57% and 46% for animals in the GnRH and CONT2. CPR was 60% in the ATBS group and 89% in CONT1, but GnRH treatment did not affect the CPR. In conclusion, LIP was effective in stimulating MY in heifers, and the IMI prevalence decreased with ATBS treatment. Also, the use of GnRH did not affect the FC regression, cyclicity at 45 DIM, and CPR.

Internal teat sealants alone or in combination with antibiotics at dry-off – the effect on udder health in dairy cows in five commercial herds

In the dairy industry, the dry period has been identified as an area for potential reduction in antibiotic use, as part of a one health approach to preserve antibiotic medicines for human health. The objective of this study was to assess the impact of dry cow treatment on somatic cell count (SCC), intramammary infection (IMI) and milk yield on five commercial Irish dairy herds. A total of 842 cows across five spring calving dairy herds with a monthly bulk tank SCC of < 200 000 cells/mL were recruited for this study. At dry-off, cows which had not exceeded 200 000 cells/mL in the previous lactation were assigned one of two dry-off treatments: internal teat seal (ITS) alone (Lo_TS) or antibiotic plus ITS (Lo_AB + TS). Cows which exceeded 200 000 cells/mL in the previous lactation were treated with antibiotic plus ITS and included in the analysis as a separate group (Hi_AB + TS). Test-day SCC and lactation milk yield records were provided by the herd owners. Quarter milk samples were collected at dry-off, after calving and at mid-lactation for bacteriological culture and quarter SCC analysis. Cow level SCC was available for 789 cows and was log-transformed for the purpose of analysis. Overall, the log SCC of the cows in the Lo_TS group was significantly higher than the cows in Lo_AB + TS group and not statistically different to the cows in the Hi_AB + TS group in the subsequent lactation. However, the response to treatment differed according to the herd studied; the log SCC of the cows in the Lo_TS group in Herds 3, 4 and 5 was not statistically different to the cows in Lo_AB + TS group, whereas in the other two herds, the log SCC was significantly higher in the Lo_TS when compared to the Lo_AB + TS group. There was a significant interaction between dry-off group and herds on SCC and odds of infection in the subsequent lactation. The results of this study suggest that the herd prevalence of IMI may be useful in decision-making regarding the treatment of cows with ITS alone at dry-off to mitigate its impact on udder health.

Investigations on some aerobic bacterial agents isolated from heifers at the beginning ofthe first lactation

In this study, it was aimed to identify the predominant coagulase-negative staphylococci (CNS) species in subclinical, clinical, and persistent infections in Konya province, to determine various virulence and resistance mechanisms of CNS species within the agelactation level. The research has been carried out in a dairy herd that is free from brucellosis and tuberculosis and uses an automated herd management system. Thirty-two heifers at the beginning of lactation with milk yields of more than 20 liters have been observed in terms of udder infections during the first 1-4 months of lactation. The prevalence of intramammary infections (IMI) and of CNS-caused IMI has been 75% and 53%, respectively. The rate of CNS was 61% among all isolates, with the rate of the major pathogen 26%, and the rate of the other pathogen 12%. S. chromogenes has been the most frequently isolated CNS species (45.5%). In conclusion, CNS may cause udder infections at the herd level in heifers during the early lactation period, which may cause persistent infections with different mechanisms, and S. chromogenes may emerge as a new contagious mastitis agent in the future. CNSs should be identified at the species level rather than the group level in order to develop effective control programs. Based on the epidemiological data, the current udder health control programs in heifers in the early lactation period were detected to be insufficient.

Effect of selective dry cow therapy on dry period intramammary infection dynamics and their association with management factors in Japan.

Selective dry cow therapy (SDCT) allocates antibiotics in the dry period to cows or quarters with a high risk of intramammary infection (IMI), potentially improving antibiotic stewardship. However, SDCT has not been used in Japan, possibly due to concerns of negative impacts on udder health. This research aims to evaluate how the SDCT use affected dry period IMI dynamics in Japan. Additionally, the effects of management factors were also considered. At dry-off, 44 cows received antibiotics plus external teat sealant or external sealant in isolation based on their IMI risk, which was assessed using milk culture, a modified California mastitis test (CMT), and mastitis records. The SDCT approach allowed antibiotic use to be reduced by 33.7%. However, quarters with a low risk of infection who received no antibiotics had a numerically higher prevalence of IMI before calving than those who received antibiotics (28.6% vs. 19.2%). In addition, an increased risk of IMI in quarters without antibiotics was also attributed to poor herd hygiene and a shorter duration of external teat sealant adherence. The result suggests that these factors influence the outcome of an SDCT program. Therefore, a uniform recommendation for dry cow herd management may not be optimal.

Effects of 2 intramammary antimicrobial formulations on control of mastitis during the dry-period and performance of dairy cows

Our objective was to evaluate the effects of benzathine cloxacillin (BC) and ceftiofur hydrochloride (CH) treatment at dry-off on prevalence of intramammary infections at calving, incidence of clinical mastitis and SCC and milk yield up to 100 DIM, and removal from the herd and reproductive responses within 100 DIM. At dry-off, cows from 2 herds were assigned randomly to the BC (n = 1,160; Orbenin-DC, Merck Animal Health) and the CH (n = 1,183; Spectramast DC, Zoetis) treatments. Composite milk samples collected at dry-off and at calving were submitted for bacterial culture. Monthly milk samples were used to determine SCC. Days in milk at first insemination and pregnancy outcomes to first and second inseminations were compared between the 2 treatments. Treatments did not differ regarding prevalence of intramammary infections at dry-off (BC = 45.2 ± 1.8% vs. CH = 42.6 ± 1.7%). Treatment did not affect the prevalence of intramammary infections at calving (BC = 39.1 ± 2.3% vs. CH = 40.4 ± 2.3%), incidence of clinical mastitis within 100 DIM (BC = 6.6 ± 1.0% vs. CH = 7.7 ± 1.1%), and milk yield up to 100 DIM (BC = 3.6 ± 0.03 kg × 10 3 vs. CH = 3.6 ± 0.03 kg × 10 3 ). Treatment did not affect reproductive outcomes. Producers may choose between BC and CH at dry-off without being concerned about differences in prevalence of IMI at calving and lactational performance.

Administration of internal teat sealant in primigravid dairy heifers at different times of gestation to prevent intramammary infections at calving

Intramammary infections (IMI) are common in primigravid dairy heifers and can negatively affect future milk production. Bismuth subnitrate-based internal teat sealants (ITS) have been used to prevent prepartum IMI in dairy heifers by creating a physical barrier within the teat, preventing pathogens from entering the gland, though determination of when to administer ITS in heifers has yet to be investigated. The objectives of this study were to determine if administration of ITS in primigravid heifers reduced the odds of IMI at calving and if administration of ITS at different stages of gestation (75 vs. 35 d prepartum) affected the odds of IMI at calving. A total of 270 heifers were used at a single farm. One quarter of each heifer was randomly chosen to be aseptically sampled and administered ITS 75 d prepartum (ITS75), another quarter of each heifer was sampled and received ITS 35 d prepartum (ITS35), whereas the remaining 2 quarters of each heifer served as control quarters (CON) and were not sampled before calving. Within 12 h of calving, aseptic colostrum samples were collected from all quarters to determine quarter infection status. When an IMI was caused by mastitis pathogens other than non-aureus staphylococci (NAS), CON quarters were 3 times [95% confidence interval (CI): 1.4-6.3] and 2.5 times (95% CI: 1.2-4.9) more likely to be infected at calving than ITS75 and ITS35 quarters, respectively. For IMI with NAS, CON quarters were 5.8 (95% CI: 3.2-10.5) and 6.4 (95% CI: 3.4-12.0) times more likely to be infected than ITS75 and ITS35 quarters, respectively. Odds of IMI at calving was similar between ITS75 and ITS35 quarters for both NAS (odds ratio = 0.9) and other pathogens (odds ratio = 1.2). Results indicate that ITS administration at either 75 and 35 d prepartum reduced IMI prevalence at calving in primigravid dairy heifers. Farm specific factors may influence prevalence and timing of heifer IMI and earlier administration of ITS provides an extended period of protection for the developing gland.

Control methods for reduction of mastitis in ewes caused by bacterial pathogens

Controlling the risk factors and eliminating the causative agents of mastitis is crucial for modern dairy sheep farms. The aim of this study was to analyze during two milking seasons the effect of proposed anti-mastitis measures focused on the reduction of mammary gland infection caused by bacterial pathogens in ewes. During the study, aseptic samples of foremilk were collected from 400 ewes three times: at the beginning (in May), in the middle (in July), and at the end (in October) of the lactation season. Totally, 2211 samples were examined over two years. Cytological and bacteriological analyses of samples were performed to evaluate the relationship between the implemented anti-mastitis measures and intramammary infections (IMI). In the 682 positive samples, the most prevalent mastitis agents detected during all examinations were coagulase-negative Staphylococci (CNS), 47.5% (n = 324 udder samples) with S. chromogenes, S. epidermidis, S. warneri and S. xylosus being the most-prevalent species. Other detected species were Staphylococcus aureus (27.4%) and Streptococcus sanguinis (15.1%), isolated mainly from clinical forms of mastitis. In the first season, due to application of antibiotic treatment to positive sheep, exclusion of chronically ill individuals from the herd and consistent implementation of the hygiene rules at milking, the prevalence of IMI was reduced by 45.2% to 30.5% in the period from May to October. In the second season, after the introduction of a complex of preventive measures consisting in examination of ewes after lambing and weaning, regular maintenance of hooves, replacement of bedding, ensuring adequate nutrition, adherence to a hygienic milking programme and rejection of positive ewes that did not respond to antibiotic treatment, the incidence of mastitis was reduced from 28.0% (in May) to 17.8% (in October). A comprehensive approach is inevitable for achieving an appropriate control and prevention of mastitis in dairy farming and for the production of high quality milk with maintenance of udder health.

Randomized controlled non-inferiority trial investigating the effect of 2 selective dry-cow therapy protocols on antibiotic use at dry-off and dry period intramammary infection dynamics

Selective dry-cow therapy (SDCT) could be used to reduce antibiotic use on commercial dairy farms in the United States but is not yet widely adopted, possibly due to concerns about the potential for negative effects on cow health. The objective of this study was to compare culture- and algorithm-guided SDCT programs with blanket dry-cow therapy (BDCT) in a multi-site, randomized, natural exposure, non-inferiority trial for the following quarter-level outcomes: antibiotic use at dry-off, dry period intramammary infection (IMI) cure risk, dry period new IMI risk, and IMI risk at 1 to 13 d in milk (DIM). Two days before planned dry-off, cows in each of 7 herds were randomly allocated to BDCT, culture-guided SDCT (cult-SDCT), or algorithm-guided SDCT (alg-SDCT). At dry-off, BDCT cows received an intramammary antibiotic (500 mg of ceftiofur hydrochloride) in all 4 quarters. Antibiotic treatments were selectively allocated to quarters of cult-SDCT cows by treating only quarters from which aseptically collected milk samples tested positive on the Minnesota Easy 4Cast plate (University of Minnesota, St. Paul, MN) after 30 to 40 h of incubation. For alg-SDCT cows, antibiotic treatments were selectively allocated at the cow level, with all quarters receiving antibiotic treatment if the cow had either a Dairy Herd Improvement Association test somatic cell count >200,000 cells/mL during the current lactation or 2 or more clinical mastitis cases during the current lactation. All quarters of all cows were treated with an internal teat sealant. Intramammary infection status at enrollment and at 1 to 13 DIM was determined using standard bacteriological methods. The effect of treatment group on dry period IMI cure, dry period new IMI, and IMI risk at 1 to 13 DIM was determined using generalized linear mixed models (logistic), with marginal standardization to derive risk difference (RD) estimates. Quarter-level antibiotic use at dry-off for each group was BDCT (100%), cult-SDCT (45%), and alg-SDCT (45%). The crude dry period IMI cure risk for all quarters was 87.5% (818/935), the crude dry period new IMI risk was 20.1% (764/3,794), and the prevalence of IMI at 1 to 13 DIM was 23% (961/4,173). Non-inferiority analysis indicated that culture- and algorithm-guided SDCT approaches performed at least as well as BDCT for dry period IMI cure risk. In addition, the final models indicated that the risks for each of the 3 IMI measures were similar between all 3 treatment groups (i.e., RD estimates and 95% confidence intervals all close to 0). These findings indicate that under the conditions of this trial, culture- and algorithm-guided SDCT can substantially reduce antibiotic use at dry-off without negatively affecting IMI dynamics.
