Prevalence of antibiotic residues in milk samples of small-scale dairy households in Bengaluru, India

This study aimed to estimate the burden of subclinical mastitis in community setting, animal handlers’ knowledge, attitude and practices on mastitis and antibiotic use, and to detect antibiotic residues in milk samples. A total of 211 households belonging to four clusters of Karnataka state in Southern province of India were selected through 30-cluster approach. A total of 380 milk samples were analyzed for Subclinical Mastitis (SCM) by California Mastitis Test (CMT) and antibiotic residues by spore-based and Charm Rosa tests. Animal handler’s basic knowledge, attitude and practices (KAP) on antibiotics use were recorded. SCM prevalence was found 42% in the study site; antibiotic residues were detected in 21% (79/380) of milk samples; 7.1% and 5.52% of samples had maximum residual limits (MRL) and Multi Drug Residues (MDR), respectively. Antibiotic residues were detected in 32.22% of the households. Majority of animal handlers heard about antibiotics (83.31%), 51% of handlers treated the animals on their own and 23% of them bought antibiotics without prescription. High prevalence of SCM in the study setting and antibiotic load in milk samples raise food safety concern. Regulatory measures for prudent antibiotic use and knowledge dissemination among animal handlers for clean milk production are the measures required to tackle the problem.

Antibiotic Resistance and Food Safety: Perspectives on New Technologies and Molecules for Microbial Control in the Food Industry

Antibiotic resistance (ABR) has direct and indirect repercussions on public health and threatens to decrease the therapeutic effect of antibiotic treatments and lead to more infection-related deaths. There are several mechanisms by which ABR can be transferred from one microorganism to another. The risk of transfer is often related to environmental factors. The food supply chain offers conditions where ABR gene transfer can occur by multiple pathways, which generates concerns regarding food safety. This work reviews mechanisms involved in ABR gene transfer, potential transmission routes in the food supply chain, the prevalence of antibiotic residues in food and ABR organisms in processing lines and final products, and implications for public health. Finally, the paper will elaborate on the application of antimicrobial peptides as new alternatives to antibiotics that might countermeasure ABR and is compatible with current food trends.

Prevalence of Antibiotic Residues in Pork in Kenya and the Potential of Using Gross Pathological Lesions as a Risk-Based Approach to Predict Residues in Meat.

The human population is growing and urbanising. These factors are driving the demand for animal-sourced proteins. The rising demand is favouring livestock intensification, a process that frequently relies on antibiotics for growth promotion, treatment and prevention of diseases. Antibiotic use in livestock production requires strict adherence to the recommended withdrawal periods. In Kenya, the risk of residues in meat is particularly high due to lack of legislation requiring testing for antibiotic residues in meat destined for the local market. We examined pig carcasses for gross pathological lesions and collected pork samples for antibiotic residue testing. Our aim was to determine if a risk-based approach to residue surveillance may be adopted by looking for an association between lesions and presence of residues. In total, 387 pork samples were tested for antibiotic residues using the Premi®Test micro-inhibition kit. The prevalence of antibiotic residues was 41.26% (95% CI, 34.53-48.45%). A logistic regression model found no significant associations between gross pathological lesions and the presence of antibiotic residues. We recommend that the regulating authorities strongly consider routine testing of carcasses for antibiotic residues to protect meat consumers. Future studies should research on farming practices contributing to the high prevalence of residues.

Antibiotic residues in raw and pasteurized milk in Iran: A systematic review and meta-analysis

<abstract> <p>Improper use of antibiotics to treat or prevent infections, and as a stimulant for livestock growth, can affect public health and the dairy industry due to the spread of antibiotic residues in milk. This systematic review and meta-analysis aimed to investigate antibiotic residues in raw and pasteurized milk in Iran. Data were collected through searching the databases, including Scopus, PubMed, Science Direct, Web of Science, Google Scholar, SID, and Magiran using the following keywords: "pasteurized milk", "raw milk", "milk", "antibiotic residues", " antibiotic", and "Iran". Finally, 40 eligible studies were selected for the systematic review and meta-analysis. According to the reviewed studies, the prevalence of antibiotic residues in raw and pasteurized milk was 26% (95% CI: 20–33%) and 21% (95% CI: 15-27%), respectively. It seems that the control measures are inefficient in dairy industry and milk collection centers for the presence of antibiotic residues and the time of antibiotic withdrawal during milk delivery. Permanent control of milk in the collection centers by the responsible organizations along with implementation of Hazard Analysis Critical Control Point system in milk factories can be very effective in reducing antibiotic residues.</p> </abstract>

Determination of antibiotic residues in milk and assessment of human health risk in Bangladesh

Consumption of milk contaminated with antibiotic residues above the maximum residue limit (MRL) causes toxicity to humans and the development of superbugs that leads to the failure of antibiotic therapy and threatens human life. Moreover, long-duration exposure might alter the nature of gut microflora, resulting in the enhancement of many diseases. Therefore, our study aims to find out the residues level of selected antibiotics in milk and assessments of humans health risks. We examined 300 raw and processed milk samples using thin-layer chromatography (TLC) and ultra-high-performance liquid chromatography (UHPLC) methods against five veterinary antibiotics and assessed the health risk for consumers in Chattogram, Bangladesh. Risk analysis was done by using a hazard quotient based on 165 ml per capita milk consumption. We found a total of 7 % prevalence of antibiotic residues in raw milk, which were higher (8 %) in individual milk samples than the pooled samples (4 %). However, we did not find any antibiotic residues in processed milk. The mean concentration of oxytetracycline residue was detected at 61.29 μg/l, and amoxicillin was 124 μg/l in individual milk samples. Risk analysis showed that, the hazard quotient values are 0.0056 for oxytetracycline and 0.0017 for amoxicillin residues. This result implied no significant health risks associated with the consumption of milk produced and marketed in the study area. Our study might fill up the gaps of knowledge in measuring the safety status of milk regarding public health issues.

Alteration of antibiotic-resistant phenotypes and minimal inhibitory concentration of Escherichia coli in pig farming: Comparison between closed and open farming systems

The prevalence of antibiotic residues and antibiotic-resistant Escherichia coli (ARE) in closed (CSF) and open (OSF) pig farming systems was analysed. Results showed that a high level of E. coli populations, antibiotic contamination in supplied water, and additional antibiotics used, such as neomycin (NEO) or colistin (CLT), were observed in OSF. Similar ARE resistance to five antibiotics, including NEO, was observed in the water source, dung, and wastewater/sludge but were different from those of CLT. An increased occurrence of ARE was found in the sludge of anaerobic digestion (AD) and the waste stabilisation pond (WSP), but they were not well correlated with their residual antibiotic concentrations. CLT administration yielded higher ARE prevalence in pig excreta and wastewater in OSF, but its absence in CSF also resulted in ARE occurrence with increased minimal inhibitory concentration (MIC) levels in the anaerobic digester/waste stabilisation ponds. This study revealed that ARE prevalence and MIC levels of CLT could be developed during the wastewater treatment process in the pig farm, although none of the AREs were found in the original excreta/wastewater.

Occurrence of Antibiotic Residues and Antibiotic-Resistant Bacteria in Nile Tilapia Sold in Some Markets in Accra, Ghana: Public Health Implication

In Ghana there are concerns that antibiotics may be used inappropriately to boost fish production, though no study has investigated this problem. To provide preliminary insights into public health aspects of the problem, we investigated the occurrence of antibiotic residues and antibiotic-resistant bacteria in Nile tilapia (Oreochromis niloticus), a fish commonly cultivated and consumed in Ghana. Two hundred Nile Tilapia fish were randomly sampled from four major markets in Accra, the capital city of Ghana. One hundred samples were screened for antibiotic residues using a microbial inhibition plate test that detects sixteen different antibiotics commonly used in animal husbandry and aquaculture. The other 100 samples were cultured for bacteria using direct culture methods, and the isolates were tested against seven antibiotics by the Kirby Bauer method. The overall prevalence of antibiotic residues in the fish samples was 7%. Bacteria that were isolated from the fish samples were Shigella sonnei (10%), Enterobacter cloacae (7%), Escherichia coli (6%), Salmonella Typhi (3%) and Klebsiella pneumoniae and Proteus mirabilis (2%). All bacteria isolated were susceptible to gentamicin and ciprofloxacin but resistant to ampicillin. Multi-drug resistance (ie resistance to three or more different classes of antibiotics) occurred in 86.7% of the isolates. Nile Tilapia sold in Accra is a source of multi-drug resistant bacteria. Consumption of the fish can also lead to significant exposure to antibiotic residues.

Prevalence of Antibiotic Residues and Antibiotic Resistance in Isolates of Chicken Meat in Korea.

The aim of study was to investigate the correlation between the level of 17 antibiotic residues and 6 antibiotic resistances of Escherichia coli isolates in chicken meats. A total of 58 chicken meats were collected from retail grocery stores in five provinces in Korea. The total detection rate of antibiotic residues was 45% (26 out of 58). Ten out of 17 antibiotics were detected in chicken meats. None of the antibiotics exceeded the maximum residue level (MRLs) in chicken established by the Ministry of Food and Drug Safety (MFDS). The most detected antibiotics were amoxicillin (15.5%), followed by enrofloxacin (12.1%) and sulfamethoxazole (10.3%). In a total of 58 chicken meats, 51 E. coli strains were isolated. E. coli isolates showed the highest resistance to ampicillin (75%), followed by tetracycline (69%), ciprofloxacin (65%), trimethoprim/ sulfamethoxazole (41%), ceftiofur (22%), and amoxicillin/clavulanic acid (12%). The results of study showed basic information on relationship between antibiotic residue and resistance for 6 compounds in 13 chicken samples. Further investigation on the antibiotic resistance patterns of various bacteria species is needed to improve food safety.

Investigating antibiotics, antibiotic resistance genes, and microbial contaminants in groundwater in relation to the proximity of urban areas.

Groundwater is an essential public and drinking water supply and its protection is a goal for global policies. Here, we investigated the presence and prevalence of antibiotic residues, antibiotic resistance genes (ARGs), mobile genetic elements (MGEs), and microbial contamination in groundwater environments at various distances from urban areas. Antibiotic concentrations ranged from below detection limit to 917 ng/L, being trimethoprim, macrolide, and sulfonamide the most abundant antibiotic classes. A total of eleven ARGs (aminoglycoside, β-lactam, chloramphenicol, Macrolide-Lincosamide-Streptogramin B - MLSB, sulfonamide, and tetracycline), one antiseptic resistance gene, and two MGEs were detected by qPCR with relative abundances ranging from 6.61 × 10-7 to 2.30 × 10-1 copies/16S rRNA gene copies. ARGs and MGEs were widespread in the investigated groundwater environments, with increased abundances not only in urban, but also in remote areas. Distinct bacterial community profiles were observed, with a higher prevalence of Betaproteobacteria and Bacteroidetes in the less-impacted areas, and that of Firmicutes in the contaminated groundwater. The combined characteristics of increased species diversity, distinct phylogenetic composition, and the possible presence of fecal and/or pathogenic bacteria could indicate different types of contamination. Significant correlations between ARGs, MGEs and specific taxa within the groundwater bacterial community were identified, revealing the potential hosts of resistance types. Although no universal marker gene could be determined, a co-selection of int1, qacEΔ1 and sulI genes, a proxy group for anthropogenic pollution, with the tetC, tetO, tetW resistance genes was identified. As the tet group was observed to follow the pattern of environmental contamination for the groundwater samples investigated in this study, our results strongly support the proposal of this group of genes as an environmental tracer of human impact. Overall, the present study investigated several emerging contaminants in groundwater habitats that may be included in monitoring programs to enable further regulatory and protection measures.

Assessment of Antibiotic Residues in the Marketed Meat of Kailali and Kavre of Nepal

This is a cross-sectional study to detect presence of antibiotic residues in meat sample of Kailali and Kavre districts. Each sample out of 55 meat samples (muscles and liver) of poultry-41, goat-12, buffalo-9 and pig-4 from Kailali and Kavre was tested for different groups of antibiotic residues separately. The antibiotics tested were Penicillin, Tetracycline, Aminoglycosides, Macrolides and Sulfonamides. Samples were preserved at -84˚C in the deep freezer after collecting in sterile plastic pouch. Rapid test kit (78.9% sensitivity and 96.7% specificity) of G9 Co. Ltd., Thailand was used to detect residues in the samples. The protocol of kit was followed for the test. The overall prevalence of antibiotic residues in meat was around 22%. Meat samples exceeding the Maximum Residue Limit (MRL) were 6. Liver was found with higher number of residues than muscles. The organ and location wise prevalence of residue was found statistically significant at p<0.05. The samples contained alarmingly high level of antibiotic residues which is because of neglecting the withdrawal period. The prescription of antibiotic is not prudent. Int. J. Appl. Sci. Biotechnol. Antimicrobial Residues in Beef Animals Slaughtered in Abattoir and Non-Abattoir Small Holders Slaughter Houses in Negeri Sembilan, Malaysia

Many researches have suggested a link between antibiotic administration in food animals and increase in bacterial resistance in human populations. In this cross-sectional study, the screening for antibiotic residues in kidneys was done using Microbial Inhibition six (6) plate method to provide baseline data for the prevalence of antibiotic residues in animals slaughtered in Abattoir and non- Abattoir slaughter house in Negeri Sembilan district areas. Basically, the agar diffusion method uses six media which have been separately cultured with different types of bacteria that are relatively susceptible and resistant towards specific antimicrobial at certain pH level. Forty one (41) kidney samples and another fourteen (14) samples were obtained from and non–abattoir and approved (Senawang, Negeri Sembilan Abattoir) slaughter houses respectively. Prevalence of antibiotic residues in commercial milk and its variation by season and thermal processing methods

Aims: In this study, the prevalence of antibiotic residues in pasteurized and sterilized commercial milk available in Shahre-kourd, Iran, was investigated. In addition, the influence of seasonal temperature changes on the prevalence of contamination was studied. Materials and Methods: Commercial milk samples of 187, including 154 pasteurized and 33 sterilized, milk samples were collected from the market between early January 2012 and late July of the same year. The presence of antibiotic residues was detected using the microbiological detection test kit, Eclipse 100, as a semi-quantitative method. Results: The results showed that 37 of the samples (19.8%) have contained antibiotic residues above the European Union Maximum Residues Limits (EU-MRLs), of which 28 samples (14.97%) were found to be contaminated but at the concentrations below the EU-MRLs. There was no significant difference between the contamination rate of pasteurized and Ultra High Temperature (UHT)-sterilized samples. Similarly, variation of weather temperature with seasons had no effect on the contamination prevalence of milk samples ( P > 0.05). Conclusion: Based on the result of this study, antibiotics residues were present in the majority of milk samples. Neither the season nor the type of thermal processing of the commercial milks had noticeable impact on the prevalence level of the milk samples. However, an increasing trend of prevalence level for antibiotic residues was observed with increasing the temperature through the warm season.

Investigation on the prevalence of antimicrobial residues in milk obtained from urban smallholder dairy and non-dairy farming households in Dagoretti Division, Nairobi, Kenya

To assess the prevalence and the level of awareness of the risk posed by antibiotic residues among the urban dairy and non-dairy farming households using a household survey and laboratory analysis of milk samples. A cross sectional study. Urban small holder dairy farming and non-farming households in Dagoretti division, Nairobi. The prevalence of antibiotic residues in dairy household milk samples was 4% (11/259) and in milk samples from non-dairy neighbouring households the prevalence was 0.07% (1/136). One sample was detected to belong to beta lactam and one to the tetracycline group of antibiotics. The remaining 10 were not categorised. Approximately 20% of dairy and non-dairy respondents were unsure of the risk that may be posed by the presence of antibiotic residues in milk. A high proportion of the respondents said they would be able to protect themselves from risk posed by antibiotic residues by following advice given by veterinary officers on the withdrawal periods after treatment of animals. Education of the farmers, continuous surveillance and understanding of the socio-economic incentives that would be traded off at farm level would help to reduce or eliminate the risk posed by the residues in marketed or consumed milk.
