Contamination levels, influencing factors, and risk assessment of polybrominated diphenyl ethers in house dust of northern Serbia.

Polybrominated diphenyl ethers (PBDEs) are a group of compounds that, due to their applications, are considered mainly indoor contaminants. To obtain the first information about the presence of PBDEs in Serbia, dust samples (n = 50) were collected in settlements in the northern Serbian province of Vojvodina. The selected/target congeners (BDE-28, 47, 99, 100, 153, 154, and 183) were extracted from house dust by microwave-assisted extraction technique, and purified extracts were analyzed on a dual-column gas chromatograph with micro-electron capture detectors. A wide range of ΣPBDEs was detected (0.295 to 394 ng g-1 dust), which reflects large differences in contamination among the examined homes. For the majority of samples (72%), ΣPBDEs were lower than 5 ng g-1 indicating that people living in Vojvodina province are exposed to low concentrations of PBDEs present in their households. Based on principal component analysis (PCA), balcony areas and age of the house positively correlate with the PBDE congeners with higher detection frequencies (≥ 50%), namely, with BDE-99, BDE-153, and BDE-183. Statistically significant positive correlation (p < 0.01) was obtained for BDE-99 and the number of household's members. Estimated daily intakes (EDItot) were calculated for ingestion and dermal absorption of dust for two age groups-adults and toddlers. These arethe first data on PBDE status in the area of the Western Balkan, and the health risk assessment indicates that PBDE levels obtained in household dust do not pose a risk for human health.

BDE-209-induced genotoxicity, intestinal damage and intestinal microbiota dysbiosis in zebrafish (Danio Rerio)

The environmental presence of polybrominated diphenyl ethers (PBDEs) is ubiquitous due to their wide use as brominated flame retardants in industrial products. As a common congener of PBDEs, decabromodiphenyl ether (BDE-209) can pose a health risk to animals as well as humans. However, to date, few studies have explored BDE-209's toxic effects on the intestinal tract, and its relevant mechanism of toxicity has not been elucidated. In this study, adult male zebrafish were exposed to BDE-209 at 6 μg/L, 60 μg/L and 600 μg/L for 28 days, and intestinal tissue and microbial samples were collected for analysis to reveal the underlying toxic mechanisms. Transcriptome sequencing results demonstrated a dose-dependent pattern of substantial gene differential expression in the group exposed to BDE-209, and the differentially expressed genes were mainly concentrated in pathways related to protein synthesis and processing, redox reaction, and steroid and lipid metabolism. In addition, BDE-209 exposure caused damage to intestinal structure and barrier function, and promoted intestinal oxidative stress, inflammatory response, apoptosis and steroid and lipid metabolism disorders. Mechanistically, BDE-209 induced intestinal inflammation by increasing the levels of TNF-α and IL-1β and activating the NFκB signaling pathway, and might induce apoptosis through the p53-Bax/Bcl2-Caspase3 pathway. BDE-209 also significantly inhibited the gene expression of rate-limiting enzymes such as Sqle and 3βhsd (p < 0.05) to inhibit cholesterol synthesis. In addition, BDE-209 induced lipid metabolism disorders through the mTOR/PPARγ/RXRα pathway. 16S rRNA sequencing results showed that BDE-209 stress reduced the richness and diversity of intestinal microbiota, and reduced the abundance of probiotics (e.g., Bifidobacterium and Faecalibacterium). Overall, the results of this study help to clarify the intestinal response mechanism of BDE-209 exposure, and provide a basis for evaluating the health risks of BDE-209 in animals.

Atmospheric concentrations of polybrominated diphenyl ethers (PBDEs) in passive air sampler from Batam Island, Indonesia

Air contamination by semi volatile organic compounds including toxic polybrominated diphenyl ethers (PBDEs) has been a matter of global concern due to the important atmospheric transportation in contribution to their widely distribution in environment. This study aims at elucidating the levels and spatial distribution of PBDEs in the atmosphere derived from polyurethane foam passive air sampler (PUF-PAS) and identify potential sources in Batam Island. Fifteen PUF-PAS were deployed in various locations representing different background activities. After deployment around 2.5 months, PUFs were analyzed for forty-two PBDEs congeners from mono- to deca-BDE congeners using gas chromatography with mass spectrometry. PBDEs were widely detected ranging 7.2 – 2900 pg/m3. Significant high concentrations of PBDEs were found at public service areas than offices, residential and remote areas (p<0.05). In particular, high concentration was found in Marina Tanjung Riau (2900 pg/m3), whereas other locations were much lower including electronic shop (50 pg/m3), dumping site area (7.2-15 pg/m3), which indicating specific emission sources according to location. The PBDEs profiles are predominated by BDE-209 congener on average 81% (36-100) while other congeners were lower. This suggest that the presence of PBDEs in Batam Island is mainly due to the commercial formulation of Deca-BDE, of which consistent with its high consumption in Asia.

Occurrence, compositional profiles and health risks assessment of polybrominated diphenyl ethers (PBDEs) in groundwater in and around dumpsites in Lagos State, Nigeria

Groundwater from the communities adjoining Abule Egba (ABE), Epe (EPE), Ikorodu (IKR), Olushosun (OSH) and Solous (SOL) dumpsites in Lagos State, Nigeria, were collected and analyzed for polybrominated diphenyl ethers. Water samples were extracted using liquid-liquid extraction followed by Gas Chromatography/Mass Spectroscopy analysis using the already validated standard method. PBDEs concentrations ranged from 7 ± 10 (IKR) to 78 ± 109 µgL−1 (ABE) for BDE-28; 5.4 ± 0.3 (OSH) to 31.5 ± 27.3 µgL−1 (ABE) for BDE-47; 17 ± 24 (ABE) to 183 ± 226 µgL−1 for BDE-100; 11.7 ± 16.5 (ABE) to 174 ± 217 µgL−1(EPE) for BDE-99; 117 ± 166 (ABE) to 2034 ± 2819 µg L−1 (EPE) for BDE −183; 296 ± 392 (IKR) to 4283 ± 1278 µg L−1 (EPE) for BDE-209. BDE- 153 and BDE- 154 were not detected in all the water samples, while BDE- 28 was only detected in ABE and IKR water samples. The BDE- 209 was the most dominant congener in all the water samples. The cumulative Hazard Index (HI) values for children through ingestion route ranged from 4.95 E + 00 (SOL) to 4.53 E + 01 (EPE), while in adults, it ranged from 4.25 E + 00 (SOL) to 3.88 E + 01 (EPE). This study confirmed the presence of PBDEs at an elevated concentration in groundwater. Since there is no regular supply of pipe-borne water in the study area, residents consume the contaminated groundwater and may be exposed to endocrine-disrupting chemicals. Therefore, the government should provide pipe-borne water for the populace and discourage people from living close to the dumpsites.

Insights into the long-term fates and impacts of polybrominated diphenyl ethers in sediment samples in Taiwan: The national project for background monitoring of the environmental distribution of chemical substances (BMECs)

This study aimed to evaluate the impact factors and effectiveness of management policies on the presence of polybrominated diphenyl ethers (PBDEs) in sediment samples in Taiwan from the last 10 years. Twenty-four PBDE congeners were detected in 838 sediment samples collected from 4 stages (2006–2019) in 30 principal rivers, based on the national project for background monitoring of the environmental distribution of chemical substances. The ΣPBDE concentrations in the 4 stages ranged from 30.00 to 147.10 ng/g dw, 6.03–15.30 ng/g dw, 4.99–7.00 ng/g dw, and 1.20–2.10 ng/g dw in the northern, southern, central, and eastern areas, respectively. The concentrations of PBDEs (e.g., penta-BDE and octa-BDE) in sediment samples notably decreased (−6 to −73%) as the Taiwan Environmental Protection Administration implemented policies banning PBDEs (except deca-BDE). The PBDEs levels of the sediment samples collected in the dry season were higher than those collected in the wet season. The levels of ΣPBDEs in sediment samples were affected by season, the amount of general waste present, and nearby PBDE-related factories and e-waste recycling facilities. Reducing the release of PBDEs, especially deca-BDE, through sound waste management and recycling practices is still needed to improve environmental sustainability in Taiwan.

Equatorial Atlantic pelagic predators reveal low content of PBDEs in contrast to MeO-BDEs: An analysis of brominated diphenyl ethers in blue shark and yellowfin tuna

Polybrominated diphenyl ethers (PBDEs) and their methoxylated analogues (MeO-BDEs) are widely distributed in the environment. The main concern about the presence of PBDEs and MeO-BDEs in fish is due to their potential endocrine disruption effects in the specimens, and their potential risk to the health of human consumers. Considering these concerns, the goal of this study was to investigate the occurrence of PBDEs and MeO-BDEs in muscle tissues of blue shark (BSH), Prionace glauca, and yellowfin tuna (YFT), Thunnus albacares, caught in the Equatorial Atlantic Ocean (EAO), North-eastern Brazilian waters, and to evaluate the potential risk of human exposure by consumption. Muscle tissues of YFT and BSH were extracted using a Soxhlet apparatus and an Accelerated Solvent Extractor (ASE), respectively. PBDEs and MeO-BDEs were analysed by GC-NCI-MS. Concentrations of PBDEs ranged from not detected (nd) to 10 ng g−1 lipid weight (lw) in YFT muscle samples, while PBDE levels in BSH muscle samples ranged from <LOQ to 34 ng g−1 lw. Regarding MeO-BDEs, the concentration ranged from 55 to 578 ng g−1 lw and from <LOQ to 263 ng g−1 lw in YFT and BSH muscle samples, respectively. MeO-BDE congeners contribution in both YFT and BSH indicated a predominance of 2′-MeO-BDE-68, which is associated to the sponges or sponge-microbiota metabolites. ∑PBDE were statistically similar between YFT and BSH, as well as observed for ∑MeO-BDE. PBDEs and MeO-BDEs in YFT and BSH represent a low potential risk of human exposure through the consumption of edible tissues. Further studies are necessary for a complete assessment of human safety and species conservation.

First insight into polybrominated diphenyl ethers in car dust in Turkey: concentrations and human exposure implications.

The presence of polybrominated diphenyl ethers (PBDEs) in the car is due to their use as a flame retardant additive in various car components such as dashboard, plastic parts, seat and headliner cushion foams, insulated cables, and electronic circuits. Ingestion of dust inadvertently or dermal contact to dust are significant pathways of human exposure to pollutants including PBDEs. There are no studies documenting presence of car dust associated flame retardants in Turkey. In the current study, a total of 13 PBDEs congeners were investigated in 62 car dust samples collected from Bursa province of Turkey using glass-fiber filters and a vacuum cleaner. Results of the study showed that congener concentrations were within the range of <MDL-40198ng/g and PBDE-209, major component of commercial deca-BDE, showed the highest concentration among the targeted congeners. Assessment of exposure to analyzed PBDEs via inadvertent dust ingestion and skin contact showed toddlers are exposed to these chemicals approx. 10 times higher compared to adults. Hazard quotient (HQ) values calculated based on total exposure (ingestion + dermal contact) and were < 1 for both adults and toddler indicated that exposure to car dust-associated PBDEs through ingestion and skin contact does not pose any health risks for human in Bursa.

Polybrominated diphenyl ethers in plastic waste of electrical and electronic equipment: a case study in Belarus.

Plastic waste of electrical and electronic equipment (WEEE) can contain polybrominated diphenyl ethers (PBDEs) that have been used as fire retardants for a long time. PBDEs were listed in the Stockholm Convention as persistent organic pollutants, so PBDE-containing plastic waste should be separated and disposed of in an ecologically sound manner. In the article, the results of bromine and PBDE content in plastic samples of WEEE collected in Belarus are presented. The screening method for bromine identification and HRGC/HRMS for PBDE identification were applied. It is shown that bromine is present in 43% of the 111 studied samples. Most often, Br-containing plastic was found in CRT monitor, CRT TVs and LCD monitor (about 50%), printers (35%), and LCD TVs (25%). PBDEs were revealed in 12 Br-containing samples, representing TVs, monitors, and printers. The sum of ∑10PBDEs varied from 6.6 to 21,000μg/kg. BDE-209 dominated in 9 samples (75% of cases); BDE-183, in two; and BDE-47 and BDE-99, in one sample. Based on the low content of PBDEs in the plastic, year of equipment production, and year of restriction of PBDEs applied in the countries-manufacturers, it was concluded that the presence of PBDEs in plastic is a consequence of contaminated waste recycling. Despite the relatively low concentrations of PBDEs (below the limit values established by the Stockholm and Basel conventions for POP wastes), further research is necessary with an extension of the list of analyzed types of equipment and identification of other brominated flame retardants.

Multidisciplinary approach to determine the effect of polybrominated diphenyl ethers on gut microbiota

Environmental health is increasingly compromised by persistent toxic substances, which may have serious implications in food safety and, thus, in human health. Polybrominated diphenyl ethers (PBDEs) are anthropogenic contaminants with endocrine disruption abilities and are commonly found in seafood, the main route of human exposure. Growing evidence points out that the human gut microbiota interacts with xenobiotics, which may lead to impairment of host homeostasis if functions of microbiota become compromised. The aim of this study was to ascertain if the physiological balance of human gut microbiome is affected by the presence and degree of exposure to PBDEs. Fermentation was performed in a batch closed-system using an inoculum made from fresh human stool. The volatolomic profile was analysed by solid-phase microextraction coupled to gas chromatography-mass spectrometry. Mesophilic, Gram-negative bacteria and coliforms were quantified by classic plating methods. Changes in the gut microbiome were evaluated after DNA extraction followed by deep sequencing of the 16S rDNA region. The exposure to PBDEs resulted in an imbalance in sulfur, short-chain fatty acids and aromatic organic compounds, changing the microbial volatolome in a dose- and time-dependent manner. Slight deviations in the microbial structure of human gut occurred in the presence of PBDEs, especially for high doses of exposure. For the first time, the impact of PBDEs on the microbial homeostasis of human gut microbiota was taken into consideration, revealing noteworthy modifications with serious health implications even at oral exposure doses considered as safe by worldwide regulatory entities.

Cell damage induced by polybrominated diphenyl ethers on Chironomus sancticaroli (Diptera: Chironomidae)

Abstract Histological effects of polybrominated diphenyl ethers (PBDEs) were observed in Chironomus sancticaroli larvae which underwent acute exposure. 2,2′,4-triBDE (BDE-17), 2,2′,4,4′-tetraBDE (BDE-47) and 2,2′,4,4′,5-pentaBDE (BDE-99) were evaluated at 0.5, 2.0 and 20 μg L-1. Cytoplasm vacuolisation of oenocytes was observed in the larvae exposed to BDE-17 and BDE-47. Cuenot cells were disrupted at the brush border as an effect of the three evaluated congeners highlighting BDE-47 at 2.0 μg L-1; 60% of larvae displayed this disruption. The midgut showed changes in the morphology of apex cells located next to the lumen of region I exposed to BDE-17 and BDE-47, while BDE-99 induced a narrowing of the lumen diameter. Significant cytoplasm vacuolisation of the larvae exposed to BDE-47 and BDE-99 was observed in region II of the midgut. Salivary glands showed acidophilic granules in the cytoplasm exposed to BDE-17 and BDE-47. The results showed that the tissues of C. sancticaroli were sensitive to flame retardants; these histopathologies can compromise the health and physiology of this organism, highlighting the concern with the presence of PBDEs in freshwater sediments.

Accumulation of polybrominated diphenyl ethers and microbiome response in the great pond snail Lymnaea stagnalis with exposure to nylon (polyamide) microplastics

Microplastics attract widespread attention, including for their potential to transport toxic chemicals in the form of plasticisers and associated hydrophobic organic chemicals, such as polybrominated diphenyl ethers (PBDEs). The aims of this study were to investigate how nylon (polyamide) microplastics may affect PBDE accumulation in snails, and the acute effects of nylon particles and PBDEs on survival, weight change and inherent microbiome diversity and community composition of the pond snail Lymnaea stagnalis. Snails were exposed for 96 h to BDEs-47, 99, 100 and 153 in the presence and absence of 1% w/w nylon microplastics in quartz sand sediment. No mortality was observed over the exposure period. Snails not exposed to microplastics lost significantly more weight compared to those exposed to microplastics. Increasing PBDE concentration in the sediment resulted in an increased PBDE body burden in the snails, however microplastics did not significantly influence total PBDE uptake. Based on individual congeners, uptake of BDE 47 by snails was significantly reduced in the presence of microplastics. The diversity and composition of the snail microbiome was not significantly altered by the presence of PBDEs nor by the microplastics, singly or combined. Significant effects on a few individual operational taxonomic units (OTUs) occurred when comparing the highest PBDE concentration with the control treatment, but in the absence of microplastics only. Overall within these acute experiments, only subtle effects on weight loss and slight microbiome alterations occurred. These results therefore highlight that L. stagnalis are resilient to acute exposures to microplastics and PBDEs, and that microplastics are unlikely to influence HOC accumulation or the microbiome of this species over short timescales.

PBDE serum concentration and preschool maturity of children from Slovakia

Polybrominated diphenylethers (PBDEs) are persistent organic pollutants (POPs), they are considered endocrine disruptors and can bioaccumulate in nature, and in living tissue. Human exposure to and the presence of PBDEs in human samples is of concern due to their potential health risks. Young children are one of the most vulnerable populations to PBDE's potential health effects.Ninety-one serum samples of 6-year-old children, residing in a contaminated location, due to former production of polychlorinated biphenyls (PCBs), were analysed to examine children's exposure to PBDEs in Slovakia. Median serum concentrations found for individual PBDE congeners BDE-28+33, −47, −99, −100, −153, −154 and −183 were 0.015, 0.184, 0.079, 0.046, 0.176, 0.014, and 0.097 ng g−1 lipid weight, respectively.Children's preschool maturity was measured using the Wechsler Preschool and Primary Scale of Intelligence (WPPSI-III) test. In multivariate analyses BDE-153 serum concentrations were significantly inversely associated with WPPSI-III composite score (p = 0.011, β = −23.6), while adjusting for PCB-153 and sex. Significant negative associations were observed for BDE-153 serum concentrations (p = 0.002, β = −29.8) and WPPSI-III composite score, after controlling for PCB-118 and sex. Negative associations were also observed for BDE-47, BDE-100 and BDE-153, with different individual WPPSI subtest scores, after adjustment with PCB-153 and/or PCB-118 and sex. Serum concentrations of PCB-153 and PCB-118 were not statistically significantly associated with WPPSI-III composite score and individual subtest scores. These findings demonstrate adverse effects of PBDE serum exposure on preschool maturity of children, and PBDEs potentially negative impact on child neuropsychological development.

Establishment of pantropic spotted dolphin (Stenella attenuata) fibroblast cell line and potential influence of polybrominated diphenyl ethers (PBDEs) on cytokines response

The presence of polybrominated diphenyl ethers (PBDEs) in the aquatic environment is an issue of major concern which may be a cause of increasing prevalence and severity of diseases in marine mammals. Although, cell culture model development and in vitro investigation approach is a prime need of time to progress immunotoxic research on aquatic mammals. In this study, we stablished fibroblast cell line (pantropic spotted dolphin) to assess the potential effects of PBDEs on cytokines response. Cells were grown in 6 well cell culture plate and complete media (DMEM and Ham's F12 nutrient mixture, fetal bovine serum, antibiotic and essential amino acids) was provided. The primary culture of (PSP-LWH) cells identification was achieved by vimentin (gene and protein) expressions. Karyotyping revealed pantropic spotted dolphin chromosomes 20 pairs with XX. Transfection was achieved by SV40 LT antigen and transfected cells were expended for passages. Stability of cell line was confirmed at various passages intervals using RT-PCR, western blotting and immunofluorescence methods. After confirmation, cell line was exposed to BDE-47 (250 ng/ml), BDE-100 (250 ng/ml) and BDE-209 (1000 ng/ml), with control group (PBS), positive control DMSO (0.1%) and negative control LPS (500 ng/ml) for 24 h. The ELISA results showed significant increase in IL-6 in BDE- 100 and BDE-209 while IL-1β and IL-8 were found higher in BDE-47 and BDE-100. TNFα and IL-10 secretion was noted higher in control and positive control groups. Altogether, these results emphasize importance of transfected (PSP-LWHT) cell line in aquatic research and potential effects of PBDEs on fibroblast provides evident to understand immune modulating effects of PBDEs in marine mammals. The impact of PBDEs on dolphin’s fibroblast cells immune response and altered cytokine response have been presented for the first time.

Harmful flame retardant found in electronic cigarette aerosol

Studies on the health impact of vaping so far have largely ignored the fact that electronic cigarette (e-cigarette) is essentially an electronic product which is likely to contain a group of endocrine disrupting flame retardants, namely, polybrominated diphenyl ethers (PBDEs) as additives in the combustible components of the product. Thus, the conclusion that e-cigarette is less harmful to health than tobacco smoking may be based on incomplete information. This study reports moderate to elevated levels of PBDEs in 5 out of the 13 samples of e-cigarettes. This finding is suggestive of the continued use of PBDEs in the manufacturing of e-cigarette atomizers and the associated protective casing. This study is unique as it confirms the existence of this developmental neurotoxicant in e-cigarette aerosols. In view of the significant levels of PBDEs and other known carcinogens (polycyclic aromatic hydrocarbons and formaldehydes) in e-cigarette aerosol, there is an urgent need for conducting a thorough review of the health risks of vaping by relevant professionals. A further lesson learnt from this study is that policy makers and relevant product manufacturers should be aware of the possible presence of PBDEs in the aerosol of body care and medical electrical devices such as face steamers, inhalators and nebulizers, especially when PBDEs are utilized in the combustible components of these devices.

PBDEs and Dechloranes in Raccoons in the Midwestern United States.

Raccoons (Procyon lotor) are one of the most widespread and abundant mammals in North America. To evaluate the suitability of using raccoons as bioindicator species, we analyzed liver tissues (n = 32) collected from 2013 to 2015 for the presence of polybrominated diphenyl ethers (PBDEs) and Dechloranes. ∑PBDE concentrations ranged from 19.1 to 2125ng/g lw (median = 98.0ng/glw) and did not differ between gender or age of raccoon. Dechloranes were detected in 38% of raccoons and ranged from 0.15 to 50.4ng/g lw (median = 2.32ng/glw). The comparatively high PBDE concentrations, and presence of Dechloranes in the raccoons in our study suggest that biota in terrestrial habitats are still widely exposed to and susceptible to the bioaccumulation of current and emerging flame retardants.

After the Research Is Done: Legal Obligations for Participants in Household Exposure Studies.

Household chemical exposure studies are essential for assessing human health risks associated with indoor environments. 1 But as in all studies involving human subjects, researchers must strive to ensure that individuals understand the risks incurred by participating in a study. 2 , 3 For household exposure studies, these risks may include legal obligations placed upon participants once they learn of contamination issues in their homes. A new review provides an overview of these potential obligations along with guidance for sharing this information with participants. 1

Environmental pollution of polybrominated diphenyl ethers from industrial plants in China: a preliminary investigation

Although numerous studies have shown the presence of polybrominated diphenyl ethers (PBDEs) in various environmental media, attention to their distribution in the environmental media surrounding industrial facilities is limited. In this study, eight PBDEs congeners (BDE-28, -47, -99, -100, -153, -154, -183, -209) were investigated in surface soils and water samples collected from commercial PBDE manufacturers, flame-retardant plastic modification plants and waste electrical and electronic equipment recycling facilities in China. Analysis of target compounds was performed using the model NCI GC-MS in SIM mode. The concentrations of ∑8PBDEs varied from 193.1 to 22,004.3 ng/L in water samples and from 1209.3 to 226,906 ng/g dry wt in surface soils, respectively. More severe PBDE contamination, when compared with previously reported data, was found in industrial areas in this study. This indicates that these industrial areas are highly polluted with PBDEs. BDE-209 was the predominant congener, accounting for more than 94% in this study, except for a 68.75% portion at one site. Our results show that PBDE manufacturing and flame-retardant plastic modification plants, easily overlooked by the public, are two primary PBDE pollution sources although they affect surrounding areas. Further research is needed, aimed at managing industrial PBDE emissions and eliminating environmental PBDE pollution, to investigate the material flows and environmental fates of PBDEs in all stages of the life cycle.

Polybrominated diphenyl ethers (PBDEs) concentration in soil from San Luis Potosi, Mexico: levels and ecological and human health risk characterization

The aim of this study was to assess the levels of polybrominated diphenyl ethers (PBDEs) in soils from the city of San Luis Potosi in Mexico and perform an ecological and human health risk characterization. In order to confirm the presence of PBDEs, outdoor surface soil samples were collected and the concentrations of PBDEs in urban, industrial, agricultural, and brick kiln industry areas were determined. The mean total PBDEs levels obtained in the study sites were 25.0 ± 39.5 μg/kg (geometric mean ± standard deviation) in the brick kiln industry zone; 34.5 ± 36.0 μg/kg in the urban zone; 8.00 ± 7.10 μg/kg in the industrial zone and 16.6 ± 15.3 μg/kg in the agricultural zone. The ecological and human health risk characterization showed relatively low-hazard quotient values. However, the moderately high PBDEs levels found in soils highlight the necessity to establish a systematic monitoring process for PBDEs in environmental and biological samples.

“One-shot” analysis of polybrominated diphenyl ethers and their hydroxylated and methoxylated analogs in human breast milk and serum using gas chromatography-tandem mass spectrometry

The presence of polybrominated diphenyl ethers (PBDEs) and their hydroxylated (OH-BDE) and methoxylated (MeO-BDE) analogs in humans is an area of high interest to scientists and the public due to their neurotoxic and endocrine disrupting effects. Consequently, there is a rise in the investigation of the occurrence of these three classes of compounds together in environmental matrices and in humans in order to understand their bioaccumulation patterns. Analysis of PBDEs, OH-BDEs, and MeO-BDEs using liquid chromatography-mass spectrometry (LC-MS) can be accomplished simultaneously, but detection limits for PBDEs and MeO-BDEs in LC-MS is insufficient for trace level quantification. Therefore, fractionation steps of the phenolic (OH-BDEs) and neutral (PBDEs and MeO-BDEs) compounds during sample preparation are typically performed so that different analytical techniques can be used to achieve the needed sensitivities. However, this approach involves multiple injections, ultimately increasing analysis time. In this study, an analytical method was developed for a “one-shot” analysis of 12 PBDEs, 12 OH-BDEs, and 13 MeO-BDEs using gas chromatography with tandem mass spectrometry (GC-MS/MS). This overall method includes simultaneous extraction of all analytes via pressurized liquid extraction followed by lipid removal steps to reduce matrix interferences. The OH-BDEs were derivatized using N-(t-butyldimethylsilyl)-N-methyltrifluoroacetamide (TBDMS-MTFA), producing OH-TBDMS derivatives that can be analyzed together with PBDEs and MeO-BDEs by GC-MS/MS in “one shot” within a 25-min run time. The overall recoveries were generally higher than 65%, and the limits of detection ranged from 2 to 14 pg in both breast milk and serum matrices. The applicability of the method was successfully validated on four paired human breast milk and serum samples. The mean concentrations of total PBDEs, OH-BDEs, and MeO-BDEs in breast milk were 59, 2.2, and 0.57 ng g−1 lipid, respectively. In serum, the mean total concentrations were 79, 38, and 0.96 ng g−1 lipid, respectively, exhibiting different distribution profiles from the levels detected in breast milk. This “one-shot” GC-MS/MS method will prove useful and cost-effective in large-scale studies needed to further understand the partitioning behavior, and ultimately the adverse health effects, of these important classes of brominated flame retardants in humans.

An assessment of polybrominated diphenyl ethers and polychlorinated biphenyls in the indoor dust of e-waste recycling facilities in South Africa: implications for occupational exposure.

Workplace exposure to persistent organic pollutants is a concern for human health. This study examined the presence of polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) in the indoor dust from two major e-waste recycling sites and a university electronic equipment repair workshop in Durban, South Africa, in order to evaluate the implication of dust for occupational exposure. The mean ∑(n = 8)PBDEs and ∑(n = 3)PCBs were 20,094 and 235ngg(-1), respectively. The levels of PBDEs and PCBs obtained in one of the recycling sites (123-27,530 and 161-593ngg(-1)) were significantly higher than the levels obtained (91-7686 and <DL-42ngg(-1), respectively) in the same site after site cleanup/maintenance. Occupational exposure was assessed for different exposure scenarios by using the 5th and 95th percentile, and the median and mean concentrations measured at the sites. By assuming a mean and a high dust intake rate, the average and 95th percentile daily exposure doses (∑DED/ngkg(-1)bwday(-1)) of PBDEs were 3.98, 8.52 and 7.58, 16.19, respectively, and of PCBs were 0.047, 0.094 and 0.089, 0.179, respectively. The ∑DEDs of PBDEs and PCBs were lower than the reference (RfD) values for BDE 47, BDE 99, BDE 153 and BDE 209, and Aroclor 1254 and 1016. Nevertheless, continual exposure to high levels of these pollutants is a concern; but as shown, the exposure can be significantly reduced if the e-waste facilities are cleaned thoroughly regularly.
