Effect of Feed Supplementation of Shatavari Root Powder (Asparagus racemosus) on Growth Performance of Broiler in Konkan Region

The present study aimed to highlights the effect of feed supplementation of Shatavari root powder (Asparagus racemosus) on growth performance of broiler in Konkan region. A substantial portion of the production expenses is attributed to the cost of feed and in a developing country like India, the supply of quality and affordable feed has been inconsistent. To enhance the overall productivity of poultry birds, it is imperative to maintain a reliable source of low-cost feed. The experimental trial of six weeks was undertaken for total 200-day old broiler chicks. They were randomly distributed into five experimental diets. Each treatment group was replicated into four with 10 birds per replicate. The control (T0) group was fed standard ration and T1, T2, T3, and T4 group were provided same standard ration supplemented with 5.00, 10.00 , 15.00 and 20.00 g /kg Shatavari root powder, respectively. Result of experiment showed that supplementation of Shatavari root powder was significantly highest live body weight in treatment T2 (1852.50 g/bird) as well as average live weight gain (264.64 g/bird). It is concluded that, 10.00 g of Shatavari root powder significantly improved growth performance than other treatments groups.

Assessment of Ethanolic Extraction of Chlorogenic Acid, Cynarin, and Polyphenols from Burdock (Arctium lappa L.) Roots Under Ultrasound

The bioactive compounds in burdock (Arctium lappa L.) roots, including chlorogenic acid (CGA) and cynarin, are valuable for use in nutraceutical foods. The ultrasound-assisted extraction of bioactive substances from dried burdock root powder (DBR) was investigated with 95% ethanol to reduce the impact of polysaccharide inulin on the extraction of free CGA and cynarin. The ethanolic extraction of CGA and cynarin was evaluated under ultrasound (300 W) at 40 kHz (U40) and 120 kHz (U120) with shaking at 120 rpm (S120) for comparison. Using a 1/30 (g/mL-solvent) solid-to-liquid ratio at 30 °C in 120 min, amounts of CGA and cynarin with U40 were as high as 818.74 μg/g-DBR and 173.68 μg/g-DBR, respectively, being much higher than those with U120 and S120. Total phenolic content, total flavonoid content, and antioxidant activity of the extract using U40 were significantly better than using U120 and S120. For U40 and U120, CGA increased with a decreasing solid-to-liquid ratio, while cynarin showed a decrease with a decreasing solid-to-liquid ratio using U120. Moreover, no observable degradations of free CGA and cynarin in ethanol were detected. By combining ultrasound and ethanol, the extracts with high-content CGA and cynarin from burdock roots were effectively achieved for use in health foods.

Cnicus Benedictus roots as a low-cost adsorbent for methylene blue dye removal from aqueous solution: kinetic and equilibrium studies

In this research, Cnicus Benedictus roots powder (CBRP) was utilized as a low-cost adsorbent for removing methylene blue (MB) as a cationic dye from aqueous solution. Characterization of CBRP by Boehm titration method, Fourier transform infrared (FT-IR) and environmental scanning electron microscope (ESEM) indicates the presence of different types of functional groups and a very rough surface filled with cavities, suitable for MB adsorption. The effect of various parameters such as CBRP dose (0.2–2 g/1L), contact time (0–180 min), initial pH (1–10), coexisting anions/cations and the initial MB concentration (100–500 mg L−1) on MB adsorption were examined, the results showed that MB uptake depends on the pH of solution, the electrostatic attraction forces, the formation of H-bonds and π–π interactions are the possible CBRP–MB interaction mechanisms. Adsorption kinetic of MB on CBRP was best represented by linear and non-linear form of the pseudo-second order model. The experimental equilibrium data were best described by linear and non-linear forms of Langmuir and Redlich–Peterson isotherm models (R 2> 0.98) with a maximum MB adsorption capacity of 85.47 mg g−1 at 25 °C, the adsorption efficiency R (%) was greater than 90 (%). MB adsorption-desorption experiments showed the reusability of CBRP for five consecutive cycles. According to the obtained results, CBRP can be used as a natural adsorbent material for cationic dyes removal from aqueous solution.

Withania coagulans root powder effect on growth, hematology, digestive enzyme activity, antioxidant status, serum immune response, and tolerance against Aeromonas hydrophila in Common Carp

AbstractObjectiveThe use of plant‐derived products in aquaculture has garnered considerable attention due to their potential benefits. This study investigated the impact of supplementing Withania coagulans root powder (WCRP) in the diet of Common Carp Cyprinus carpio on various parameters, including growth performance, digestive enzymes, hematology, antioxidant activity, and immunological aspects.MethodsCommon Carp (mean weight ± standard deviation = 11.69 ± 0.48 g) were divided into four groups in triplicate, receiving different WCRP concentrations (0.0, 1.0, 1.5, and 2.0%; labeled as WCRP0, WCRP1.0, WCRP1.5, and WCRP2.0, respectively) over a 60‐day period, followed by an experimental challenge with Aeromonas hydrophila to assess the relative percentage survival (RPS) over 14 days.ResultResults showed that Common Carp receiving WCRP1.5 and WCRP2.0 demonstrated significantly improved growth performance, with reduced feed conversion ratios (FCRs) being particularly evident in WCRP1.5. Polynomial contrasts indicated significant linear and quadratic effects on weight gain and FCR. Additionally, WCRP1.5 and WCRP2.0 supplementation led to significantly higher activity of digestive enzymes (lipase and protease). Hematological parameters, including white blood cells, red blood cells, hemoglobin, and hematocrit, were significantly elevated in fish that were fed WCRP1.5 and WCRP2.0 compared to the control group. Moreover, serum parameters, such as total protein, albumin, globulin, lysozyme, catalase, superoxide dismutase, and total immunoglobulins, were significantly enhanced in WCRP1.5‐ and WCRP2.0‐treated fish. Notably, the WCRP1.5 group showed the lowest serum cortisol levels. The RPS was highest in WCRP1.5 (73.77%), followed by WCRP2.0 (70.43%), compared to the other groups.ConclusionIn conclusion, supplementation with WCRP1.5 and WCRP2.0 effectively improved both growth and health parameters in Common Carp.

Harnessing Moringa oleifera root powder (MORP) for the sustainable remediation of heavy metal contaminated water.

Heavy metal environmental pollution is rapidly increasing due to the increase in industrialization and urbanization. Industrial processes, such as paint production, mining, and raw materials producing industries release effluents rich in heavy metals, like Pb2+, Cd2+, Cu2+, and Cr3+. These heavy metals are dangerous because they persist in nature, are non-biodegradable and they have high tendency to accumulate in the environment and in living organisms who are exposed to them. This work studied the removal of heavy metals (Cu, Pb, Cr, and Cd) from aqueous solution using Moringa oleifera root powder (MORP) as the adsorbent. The MORP was characterized by SEM, FTIR, BET, and XRD. Batch adsorption experiments carried out investigated the effects of adsorbate concentration, adsorbent dosage, agitation time, pH and temperature on adsorption. The optimum parameters are: contact time (90 min); pH (9); adsorbent dose (0.6); metal ion concentration (30 mg L-1) for Cr and 40 mg L-1 for the rest; and temperature (50 °C) for Cu and Pb, and 70 °C for Cr and Cd. These experimental data were analyzed with 5 isotherm models (Temkin, Flory-Huggins, Langmuir, D-R and Freundlich). The result obtained fitted best to Temkin isotherm in comparison to others. Kinetic studies revealed that the pseudo-second order kinetic model best described the adsorption (with high R2 values ranging from 0.9810-0.9976) compared to pseudo-first order and intra-particle diffusion kinetics model. Results of the thermodynamic study showed that the sorption process was endothermic for Cu and Pb, but exothermic for Cd and Cr. The adsorbent showed good adsorptive tendencies toward the ions studied, and could be applied on an industrial scale for the remediation of metal contaminated water.

Cadmium (Cd) detoxification and activation of plant defense enzymes activation in Wheat (Triticum aestivum) through the use of endophytic Bacillus thuringiensis and Salix alba root powder

Cadmium (Cd) is a toxic heavy metal and a threat to the ecosystem therefore the current investigation was designed to use endophytic bacteria from the Salix alba roots and to investigate its plant growth promoting and Cd detoxification ability with and without Salix alba root powder. In a complete randomized design (CRD), the cadmium sulfate was applied at the rate of 20mg/kg and 40mg/kg soil. The Bacillus thuringiensis (Accession # MW979616) was identified from Salix alba roots. The combination of Bacillus thuringiensis inoculated seeds +0.5gm root powder showed significant increase in wheat shoot dry weight, root fresh weight, catalase, and ascorbate peroxidases by 457%, 223%, 105% and 74%, respectively. The application of Bacillus thuringiensis with Salix alba root powder boosted the plant growth and defense at higher concentrations of Cd. In another treatment with Bacillus thuringiensis inoculated seeds + CdSO4 40mg/kg + 0.5gm root powder significantly increased the shoot fresh weight, root fresh weight, root dry weight, proline, sugar, superoxide dismutase, and peroxidase by 456%, 650%, 115%, 91%, 80%, 350%, and 250%, respectively with 80% reduction in plant Cd accumulation and increased bacterial population. Bacillus thuringiensis and Salix alba root powder can be useful for plant growth, Cd toxicity mitigation, accelerating bacterial activity in Cd-contaminated soil and uplifting the plant defense under heavy metal stress.

Comparison of piperine content, antimicrobial and antioxidant activity of Piper chaba root and stem

Piper chaba (locally known as “Choi Jhal”) is used traditionally as spices and folk medicine in different parts of Bangladesh. One of the most important bioactive compounds in this plant is piperine. In this study, the amount of piperine in P. chaba root and stem was investigated and the optimal solvent for piperine extraction at room temperature was also studied. High performance liquid chromatography (HPLC) was operated using a reverse phase column where methanol and water (70:30) were used as mobile phase. The detection was performed using photo diode array (PID) detector at a wavelength of 345 nm. The standard piperine showed linearity between 0.005 % and 0.04 % and the correlation co-efficient found for the linearity was 0.9933. The percentage of relative standard deviation (RSD) for both retention time and peak area were less than 2.0 %. The theoretical plate number (N) > 3000 and a tailing factor (T) < 1.5 were found in the acceptable range. The recovery percentage (%) of standard piperine was 99.16 %. Low value of co-efficient of variation and standard deviation are recognized for high precision of the method. The highest amount of piperine was found in root extracted with methanol (MR) amounting to 1.75 % in the root powder. The maximum amount of piperine in the stem was 1.59 % when extracted with methanol (MS). The piperine quantification in other extract like n-hexane root (HR), ethyl acetate root (ER), n-hexane stem (HS), ethyl acetate stem (ES) were 0.76 %, 1.69 %, 0.33 % and 1.46 % respectively. Methanol has given the highest yield of piperine compared to ethyl acetate and n-hexane for both root and stem. The developed method was simple, rapid, economic and validated in terms precision, accuracy and recovery. This selective method is found to be repeatable, accurate and successfully utilized for the Piper extract in marketed and pharmaceutical samples with well chromatographic conditions. The ethyl acetate extract of root and stem showed promising DPPH (1, 1-diphenyl-2-picrylhydrazyl) free radical scavenging activity with an IC50 value of 39.62 ± 0.95 μg/mL and 43.85 ± 1.50 μg/mL respectively. The study reports potential antibacterial activity and antifungal activity of P. chaba root and stem extracts. These outcomes revealed that different extracts of P. chaba may be used as natural preservatives.

Heating alters the nutritional and antioxidant characteristics of lotus root

To investigate the impact of heating on the nutritional and antioxidant properties of lotus root, a comparative analysis was conducted between its heated and unheated whole powders. The whole powder of heated lotus root (HLRWP) exhibited a higher level of total soluble sugars but a lower level of free phenolic compounds compared to the whole powder of fresh lotus root (FLRWP). Meanwhile, they showed significant differences in metabolite composition encompassing phenolic compounds, flavonoids, amino acids, vitamins and lipids. The replacement of sugar and corn in the medium with 25% HLRWP effectively enhanced the activities of antioxidant enzymes (total superoxide dismutase and catalase), while reducing malonaldehyde content in Drosophila melanogaster males. The average, median and maximum lifespans of the fruit flies were extended by 35.90%, 23.49% and 24.00% respectively, accompanied by an improvement in climbing ability. Additionally, their resistance to oxidative stress induced by hydrogen peroxide or paraquat was enhanced. The antioxidant capacity of HLRWP in vivo was found to be obviously stronger compared to FLRWP, which may have a crucial association with the regulation of L-arginine and adenosine monophosphate levels. The results indicate that heating can enhance the nutritional quality of lotus root products by strengthening their antioxidant capacity.

Aktivitas Antirayap Kulit Batang dan Akar Tumbuhan Kokosan (Lansium domesticum cv Kokossan) Terhadap Rayap Tanah (Coptotermes curvignathus Holmgren)

Kokosan (Lansium domesticum cv Kokossan) is one of the Meliaceae family plants known to produce various compounds with various activities. The Meliaceae family is known to have fruit seeds with a bitter taste that can be utilized as an antifeedant in insects. However, research on the bioactivity and content of secondary metabolites in the roots and bark of kokosan as termiticides against subterranean termites has never been reported. This study aims to determine the content of secondary metabolite compounds and test the anti-termite activity of extracts and fractions of kokosan root and stem bark against Coptotermes curvignathus Holmgren termites. Root and stem bark powder were macerated separately using methanol solvent and then fractionated to obtain n-hexane, dichloromethane, ethyl acetate, and methanol fractions (residue). Each extract and fraction was fed to termites through cellulose paper at a concentration of 5% for 3 days. Based on the results of phytochemical tests on extracts and fractions from the roots and bark of kokosan, it is known to contain alkaloid, flavonoid, terpenoid, and phenolic compound groups. The results of the anti-termite activity test are based on the percentage mortality value obtained by the methanol extract of the most active root with a value of 97.2% and paper weight loss of only 6.92%. In the ethyl acetate fraction of the roots, mortality was 84.4% and paper weight loss was 2.38%, and the n-hexane fraction of the stem bark showed mortality of 87.6% and paper weight loss of 2.20%. The kokosan root obtained has the most active activity against Coptotermes curvignathus Holmgren.

Development of technology for pita bread with increased nutritional value

The majority of the population living in large cities and some regions of the Russian Federation show signs of decreased immunity, most often manifested in colds. One of the ways to strengthen the human body’s immune system is to eliminate the deficiency of vitamins that have immunomodulatory properties. While developing the technology for the pita bakery product, dough has been prepared from a mixture of high-grade rye and wheat flour in equal quantities. As a source of vitamins that have a beneficial effect on the human immune system, a solution of pine needles has been added instead of half the amount of water. As a source of vitamin C, black currant and cranberry puree are used in the ratio of 1 : 1. An additional source of nutrients with immunomodulatory properties is dry ginger root powder added to the stuffing in an amount of 2 % by weight of the stuffing. Apple pectin has been used to stabilize the stuffing. A solution of pine needles is prepared by mixing an alcoholic infusion of pine needles and an aqueous decoction of pine needles in a ratio of 1 : 4. The pine needle solution has been used to replace half the amount of water used to prepare the dough. The developed bakery product "Pita with berry stuffing" is prepared with a ratio of pita to stuffing of 1 : 2 by weight. The developed bakery product can be recommended for stimulating and strengthening the immune system due to the additional inclusion of vitamins (C, K, A, B2, B6, B12) in the diet with beneficial effect on the human immune system, including for residents of the northern regions of Russia.

Adsorption dynamics of chloride removal with activated vetiver root powder: dynamic modeling perspectives and ANN predictions

ABSTRACT This study evaluates activated vetiver root powder as chloride ion adsorbent for saline water treatment. Experimental results demonstrate that activated vetiver root effectively removes chloride ions from saline solutions. Higher bed heights and lower flow rates improve efficiency by providing more adsorption sites and a longer contact time. However, higher concentrations lead to rapid bed exhaustion, highlighting the need for parameter optimization. Dynamic adsorption modeling using Thomas, Yoon–Nelson, and Clark models assesses the impact of bed height, flow rates, and initial concentration on chloride removal efficiency. Breakthrough equations closely fitted experimental data in the following order: Thomas model (R2 = 0.999) &amp;gt; Clark (R2 = 0.996) &amp;gt; Yoon–Nelson (R2 = 0.994). Scanning electron microscopy (SEM) analysis identifies pores and pockets as main surface components. Insights into adsorption mechanisms were gained through infrared and X-ray spectroscopies. A novel predictive algorithm for chloride removal in a continuous mode was developed using neural networks. Artificial neural network (ANN) prediction ability was assessed using MSE and R2 values. Future commercial applications of activated vetiver powder can be investigated using real salinity-induced wastewater. Additionally, assessing the adsorbent's regeneration and reusability is vital for confirming its cost-effectiveness and sustainability in longstanding use. This study offers perceptions for developing efficient water treatment strategies by understanding adsorption dynamics and surface characteristics for optimizing chloride removal processes.

An Efficient Aqua-Based Culture Method for the Propagation of High-Quality Arundo donax Seedlings

Arundo donax (family: Poaceae) is a perennial rhizomatous plant with considerable economic and ecological benefits. The plant is characterized by its fast growth, high yield, wide adaptability, and strong disease resistance. However, widespread cultivation is difficult because A. donax can only be asexually propagated. This study tested the efficacy of an aqua-based culture method with short duration and high rooting rate. The aqua-based culture protocol involved treating cane fragments with the commercial broad-spectrum rooting powder ABT-1 (No. 1 of the ABT series) at 100 mg/L for 24 h, followed by culturing in tap water for 19 days. We found that plant materials sampled from September to February of the following year were more suitable for aqua-based culturing than materials sampled from other months. Nodes on different parts of the cane did not vary significantly in the ability to form seedlings. The rooting percentage reached &gt;95% after only 20 days, while the sprouting percentage was &gt;93%. The seedlings obtained were strong, with high survival rates during cultivation and acclimation. No strict requirements for the growing media were necessary to achieve these results. In conclusion, this study provides an empirical foundation for a technique that could facilitate the factory production of high-quality A. donax seedlings.

Proximate Analysis and Techno-Functional Properties of Berberis aristata Root Powder: Implications for Food Industry Applications.

Berberis aristata, commonly known as Indian barberry, has been traditionally used for its medicinal properties. Despite its recognized pharmacological benefits, its potential application in the food industry remains underexplored. This study aims to investigate the proximate analysis and techno-functional properties of Berberis aristata root powder to evaluate its feasibility as a functional food ingredient. The root powder of Berberis aristata was subjected to proximate analysis to determine its moisture, ash, protein, fat, fiber, and carbohydrate content. Techno-functional properties, including water and oil absorption capacity, emulsifying and foaming properties, and bulk density, were evaluated using standardized analytical techniques. The proximate analysis revealed a high fiber content and a significant number of bioactive compounds. The root powder exhibited favorable water and oil absorption capacities, making it suitable for use as a thickening and stabilizing agent. Emulsifying and foaming properties were comparable to conventional food additives, indicating their potential in various food formulations. The findings suggest that Berberis aristata root powder possesses desirable techno-functional properties that could be leveraged in the food industry. Its high fiber content and bioactive compounds offer additional health benefits, making it a promising candidate for functional food applications. Further research on its incorporation into different food matrices and its sensory attributes is recommended to fully establish its utility.

Green synthesis, characterization and biological activity analysis of silver nanoparticles from commercially available root powder of Picrorhiza kurroa Royle ex Benth.

Picrorhiza kurroa Royle ex Benth., a medicinal plant native to the alpine regions of the Himalayas, is renowned for its diverse therapeutic properties. Recently, green-synthesized silver nanoparticles have gained attention for their potential in pharmaceutical and industrial applications. In this study, silver nanoparticles (Pk-AgNPs) were synthesized using commercially available root powder of P. kurroa. The objective was to evaluate their potential biomedical applications by assessing their antioxidant properties and cytotoxicity. The Pk-AgNPs were characterized using Scanning Electron Microscopy (SEM) and Fourier-Transform Infrared Spectroscopy (FT-IR), which revealed their morphology and functional groups. The Total Antioxidant Capacity (TAC) of methanolic root extract of P. kurroa (PkRE) and the Pk-AgNPs was determined using the Phosphomolybdenum method. The results indicated that the antioxidant activity of PkRE was higher than that of the Pk-AgNPs. Moreover, the activity increased with concentration, ranging from 20 µg/mL to 120 µg/mL, demonstrating their capability to eliminate harmful free radicals. The cytotoxicity of PkRE and Pk-AgNPs was evaluated using the MTT assay against the SV40 T-antigen-containing HEK293T cell line, derived from human embryonic kidney 293 cells and the L929 cell line, a mouse fibroblast cell line. The results demonstrated dose-dependent cytotoxicity. L929 cells remained 100 % viable at concentration up to 200 µg/mL for PkRE and 100 µg/mL Pk-AgNPs. In contrast, HEK293T cells didn’t show 100 % viability even at a concentration of 50 µg/mL for both samples. This study highlights the potential use of PkRE and Pk-AgNPs as antioxidants. However, the cytotoxicity findings suggests that they may be harmful to humans if consumed orally, though they could be suitable for use in other organisms. This paves the way for further research into the application of AgNPs synthesized from P. kurroa in biomedical and pharmaceutical domains as well as in veterinary sciences.

Carcass Essential Amino Acids Profile of &lt;i&gt;Clarias gariepinus &lt;/i&gt;(Burchell, 1822) Fingerlings Fed Different Levels of Maca (&lt;i&gt;Lepidium meyenii&lt;/i&gt;, Walp.) Root Powder as Feed Additive

This research was carried out to determine the carcass essential amino acids profile of Clarias gariepinus fingerlings fed different levels of Maca root powder as a feed additive. Five (5) isonitrogenous (40% crude protein) and isocaloric (1,732kcal kg-1 gross energy) diets were formulated, where Maca root powder in the form of feed additive was included at 0.0g/100g (TM0), 0.25g/100g (TM1), 0.5g/100g (TM2), 0.75g/100g (TM3) and 1.0g/100g (TM4) inclusion levels, diet without Maca root powder (TM0), served as the control diet. Formulated diets were fed to Clarias gariepinus fingerlings (n = 300, 10.0±0.00g) in fifteen (15) rectangular plastic tanks (n = 20) at a fixed feeding rate of 3% body weight twice daily between the hours of 8:00 - 9:00 am and 4:00 - 5:00 pm at regular interval and adjusted after every two (2) weeks of sampling for a period of twelve (12) weeks. At the end of the research three (3) Clarias gariepinus from each treatment were sacrificed with the carcass subjected to essential amino acids analysis. Data collected were analysed using one - way analysis of variance (ANOVA) at P = 0.05. All the nine (9) essential amino acids were found in the final carcass profile of the Clarias gariepinus. Carcass histidine value (15.81mg/g) was highest in fish fed diet TM3 and least (10.02mg/g) in the fish fed the control diet (TM0), carcass isoleucine value (25.42mg/g) was highest in fish fed diet TM3 and least (18.20mg/g) in the fish fed control diet (TM0), while the carcass leucine value (14.17mg/g) was highest in fish fed diet TM4 and least (11.98mg/g) in the fish fed control diet (TM0), carcass lysine value (20.13mg/g) was highest in fish fed diet TM3 and least (14.68mg/g) in the fish fed control diet (TM0), carcass methionine value (33.07mg/g) was highest in fish fed diet TM3 and least (26.80mg/g) in the fish fed control diet (TM0), carcass phenylalanine value (22.24mg/g) was highest in fish fed diet TM3 and least (17.69mg/g) in the fish fed control diet (TM0), carcass threonine value (22.23mg/g) was highest in fish fed diet TM3 and least (15.13mg/g) in the fish fed control diet (TM0), carcass tryptophan value (25.74mg/g) was highest in fish fed diet TM3 and least (20.92mg/g) in the fish fed control diet (TM0) and the carcass valine value (39.77mg/g) was highest in fish fed diet TM3 and least (32.95mg/g) in the fish fed control diet (TM0).. The incorporation of Maca root powder as a feed additive into the diet of Clarias gariepinus fingerlings increases the concentration of the carcass's essential amino acids and therefore the Clarias gariepinus can be a very important source of the essential amino acids. The incorporation of Maca root powder as a feed additive into the diet of Clarias gariepinus fingerlings is recommended for improving the Clarias gariepinus carcass essential amino acids profile.

Quality Optimization and Evaluation of New Cookie Product with Celery Root Powder Addition.

Combined drying, an energy-efficient method that includes osmotic pretreatment in molasses and shortened successive lyophilization, was used to obtain celery root powder and incorporate it in the formulation of cookies, with the aim of obtaining a new product. Wheat flour was substituted with combinedly dehydrated celery root powder at levels from 0 to 30%, and optimization of the amount of wheat flour substitution regarding technological, sensory and nutritive characteristics was performed. The optimal level of 20% substitution was determined using Z-score analysis, from the aspect of the best nutritive improvement and the mildest adverse impact on the technological and sensory quality. In the second research phase, comparison of the cookies with the 20% celery root powder substitution, dehydrated by different methods, indicated that combined dehydration showed upgraded results in terms of the overall quality of the final product, for 28.85 percentile points higher than cookies with lyophilized and for 65.24 percentile points higher than cookies with the addition of convectively dried celery root powder. The cookie containing celery powder previously osmodehydrated in molasses had higher contents of analyzed minerals (1.2-3.3 times), total phenols (10.8%) and antioxidant activities (14% for DPPH and 4% for ABTS) compared to the cookie with lyophilized powder.

Growth inhibitory potential of quinone-rich fraction of Plumbago zeylanica L. Against MG-63 osteosarcoma cells via Bax/Bcl-2 modulation

ObjectiveThe objectives of this study were to explore the cytotoxic potential of different fractions (Hexane, chloroform, ethyl acetate and methanol) of Plumbago zeylanica L. root powder. MethodologyIn this study, PzMH, PzMC, PzME, and PzMM fractions from the root powder of P. zeylanica were obtained through the cold maceration technique. The cytotoxic properties of these fractions on MG-63 cells were assessed using the MTT assay. The most potent fraction was further analyzed to understand its cytotoxic mechanism. To explore its apoptosis-inducing potential, microscopic and flow cytometric studies were conducted. Additionally, western blot analysis was used to examine the expression levels of proteins associated with apoptotic pathways. ResultsPzMH showed the highest cytotoxicity against MG-63 osteosarcoma cells, with a GI50 value of 53.07 µg/ml. Flow cytometric studies revealed an upsurge in reactive oxygen species and disruption of mitochondrial membrane potential. The highest concentration of PzMH fraction (183.48 µg/ml) led to 62 % cell population arrest at G0/G1 stage. The anti-apoptotic protein Bcl2 was downregulated, with p53, cleaved-caspase 9, cleaved-caspase3, Bad and Bax expression upregulated. HPLC analysis revealed the presence of lawsone, p-coumaric acid, plumbagin, and 4-hydroxy benzaldehyde, indicating that the quinone-rich fraction of Plumbago zeylanica L. attenuates the proliferation of MG-63 osteosarcoma cells through the Bax/Bcl-2 pathway. ConclusionTo the best of our current knowledge, this marks the first documented case where the hexane fraction enriched with quinones extracted from the roots of P. zeylanica demonstrates its ability to induce apoptosis by influencing the Bax/Bcl2 pathway.

Phytochemical and Pharmacological Profile of Desmodium gangeticum (L.) DC.: A Comprehensive Review

Abstract: Desmodium gangeticum (L.) DC., commonly known as Shalparni in Ayurveda, has been used for treating various diseases. The plant is one among ten ingredients of classical Ayurvedic formulations Dasamula. Decoctions of root and leaf, root powder and leaf juice of D. gangeticum are used as tonics, febrifuge, digestive, antiemetic, astringent, anti-asthmatic, anti-diarrhoeal and anti-inflammatory agents in traditional medicinal practices of India and its neighbouring countries. Phytochemical investigations of this plant revealed the presence of many bioactive phytoconstituents. The present review was prepared with the objective of documenting the phytochemical and pharmacological profiles of D. gangeticum. The data presented here were collected from various sources like books, journals, various online databases, and monographs and internet search engines Google Scholar, PubMed, Science Direct, etc. Studies reported from phytochemical screening revealed that D. Gangeticum leaves and roots are rich in flavonoids, alkaloids, terpenoids, tannins, phenols, carbohydrates, steroids, phenylpropanoids, pterocarpans, coumarins, and volatile oil. Among the isolated compounds, major bio-active constituents are alkaloids, flavonoids and pterocarpans. All these phytochemicals are widely distributed in various parts of D. gangeticum. The other active constituents like desmodin, hordenine and gangetin are largely responsible for its broad spectrum of therapeutic potentiality. Desmodin belongs to the class of pterocarpans and is reported to have antifungal and antibacterial activities. Gangetin, which also belongs to the pterocarpans, is reported to have anti-inflammatory and analgesic effects. FTIR spectroscopy showed that D. gangeticum extracts are mainly rich in phenolic derivatives. Scientific experiments carried out in vitro as well as preclinical animal experimentations, showed that the plant and its bioactive chemicals exhibit various therapeutic activities. It is reported that D. gangeticum extracts showed potent antioxidant, antibacterial, hypoglycaemic, analgesic, hepatoprotective, anti-inflammatory and antiarthritic activities. Studies reported that D. gangeticum or its formulations are safe when used in appropriate therapeutic dosage.
