Rhizosphere Bacterial Isolation from Indigenous Plants in Arid and Semi-Arid Algerian Soils: Implications for Plant Growth Enhancement

The Mediterranean area is one of the geographic zones most affected by land degradation and desertification and these conditions appear to be even more exacerbated by climate change. Based on this idea, this work aimed to isolate, identify, characterize, and select bacterial strains able to tolerate salinity and drought, which could possibly be used in agriculture as plant biofertilizers. The sampling of rhizosphere soil was performed in two Algerian regions, Ghardaïa and Djelfa (arid and semi-arid zones, respectively) in six provinces, targeting fourteen native plant species, known for their therapeutic use. A total of 288 bacterial strains were isolated, identified, and characterized for their growth at different temperatures and salt tolerance. Based on these capabilities, 95 isolates were selected. These strains underwent further evaluation for their plant-beneficial traits, including siderophore synthesis, auxin production, and phosphate solubilization. Additionally, we assessed their impact on tomato, cucumber, and sorghum seed germination. In a final screening step, nine bacterial strains were tested for their potential plant growth-promoting activity on tomato plants grown in semi-controlled conditions. Our results demonstrated that three strains (Bacillus simplex AH24, Microbacterium arborescens PU10, and Microbacterium paludicola AEA23) showed plant growth promotion activities on tomato.

Isolation of Endophytic Salt-Tolerant Plant Growth-Promoting Rhizobacteria From Oryza sativa and Evaluation of Their Plant Growth-Promoting Traits Under Salinity Stress Condition

The application of plant growth-promoting rhizobacteria (PGPR) as vital components for plant growth promotion against biotic and abiotic stresses could be a promising strategy to improve crop production in areas vulnerable to increasing salinity. Here, we isolated Seventy-five endophytic bacteria from roots of healthy Oryza sativa grown in a saline environment of the southern coastal region of Bangladesh. The endophytes in a culture of ~108 CFU/ml showed arrays of plant growth-promoting (PGP) activities: phytohormone (Indole acetic acid) production (1.20–60.13 μg/ ml), nutrient (phosphate) solubilization (0.02–1.81 μg/ml) and nitrogen fixation (70.24–198.70 μg/ml). Four genomically diverse groups were identified namely, Enterobacter, Achromobacter, Bacillus, and Stenotrophomonas using amplified ribosomal DNA restriction analysis followed by their respective 16S rDNA sequence analyses with that of the data available in NCBI GenBank. These four specific isolates showed tolerance to NaCl ranging from 1.37 to 2.57 mol/L in the nutrient agar medium. Under a 200 mmol/L salt stress in vitro, the bacteria in a culture of 108 CFU/ml exhibited competitive exopolysaccharide (EPS) production: Stenotrophomonas (65 μg/ml) and Bacillus (28 μg/ml), when compared to the positive control, Pseudomonas spp. (23.65 μg/ml), a phenomenon ably supported by their strong biofilm-producing abilities both in a microtiter plate assay, and in soil condition; and demonstrated by images of the scanning electron microscope (SEM). Overall, the isolated endophytic microorganisms revealed potential PGP activities that could be supported by their biofilm-forming ability under salinity stress, thereby building up a sustainable solution for ensuring food security in coastal agriculture under changing climate conditions.

Quorum Sensing System Affects the Plant Growth Promotion Traits of Serratia fonticola GS2.

Quorum sensing (QS) enables bacteria to organize gene expression programs, thereby coordinating collective behaviors. It involves the production, release, and population-wide detection of extracellular signaling molecules. The cellular processes regulated by QS in bacteria are diverse and may be used in mutualistic coordination or in response to changing environmental conditions. Here, we focused on the influence of the QS-dependent genes of our model bacterial strain Serratia fonticola GS2 on potential plant growth promoting (PGP) activities including indole-3-acetic acid (IAA) production, 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity, and biofilm formation. Based on genomic and phenotypic experimental data we identified and investigated the function of QS genes in the genome of the model strain. Our gene deletion study confirmed the biological functionality of the QS auto-inducer (gloI) and receptor (gloR) on potential PGP activities of GS2. A transcriptomic approach was also undertaken to understand the role of QS genes in regulation of genes primarily involved in PGP activities (IAA, ACC deaminase activity, and biofilm formation). Both transcriptomic and phenotypic data revealed that the QS-deletion mutants had considerably less PGP activities, as compared to the wild type. In addition, in vivo plant experiments showed that plants treated with GS2 had significantly higher growth rates than plants treated with the QS-deletion mutants. Overall, our results showed how QS-dependent genes regulate the potential PGP activities of GS2. This information may be helpful in understanding the relationship between QS-dependent genes and the PGP activity of bacteria, which aid in the production of practical bio-fertilizers for plant growth promotion.

Non-rhizobial Root Nodule Endophytic Yeast, Candida tropicalis VYW1 Impacts Germination, Nodulation behavior and Metabolic flux in Blackgram (Vignamungo L.)

Nodule associated endophytic yeasts have attracted much attention dueto its potential plant growth-promoting activities and protecting legumecrops from both biotic and abiotic stress. The present study evaluated thepotential role of an endophytic yeast Candida tropicalis VYW1 in modulatinghost fitness (blackgram) both as a single inoculant and co-inoculant withRhizobium sp. VRE1. The results showed that nodule associated endophyticyeast C. tropicalis VYW1 when co-inoculated with Rhizobium sp. VRE1significantly increased the germination percent (100%), root (34.6 cm),and shoot length (32.8 cm). The vigor index was also maximum (4250) inthe treatment that received the co-inoculation of Rhizobium sp. VRE1 andC. tropicalis VYW1 besides registering enhanced hydrolytic enzyme,α - amylase, and protein content in seeds. Metabolite profiling of bacterialprimed seed exudates revealed 30 diverse compounds belonging to theclasses viz., sugars, amino acids, organic acids, flavones, prenol lipids,and fatty acids. The blackgram seeds primed with co - inoculants releasedhydroxyl benzoic acid and galactopyranose, which act as potential chemoattractantsand involves in the lectin complement pathway regulatingfructose mannose metabolism. These metabolites lead to a significantincrease in germination and nodulation in blackgram seedlings. Hence,the study suggests the scope of enhancing the fitness of blackgram usinga co-inoculant comprising of C. tropicalis VYW1 and Rhizobium sp. VRE1.

Multifunctionality and diversity of culturable bacterial communities strictly associated with spores of the plant beneficial symbiont Rhizophagus intraradices

Arbuscular Mycorrhizal Fungi (AMF) live in symbiosis with most crop plants and represent essential elements of soil fertility and plant nutrition and productivity, facilitating soil mineral nutrient uptake and protecting plants from biotic and abiotic stresses. These beneficial services may be mediated by the dense and active spore-associated bacterial communities, which sustain diverse functions, such as the promotion of mycorrhizal activity, biological control of soilborne diseases, nitrogen fixation, and the supply of nutrients and growth factors. In this work, we utilised culture-dependent methods to isolate and functionally characterize the microbiota strictly associated to Rhizophagus intraradices spores, and molecularly identified the strains with best potential plant growth promoting (PGP) activities by 16S rDNA sequence analysis. We isolated in pure culture 374 bacterial strains belonging to different functional groups—actinobacteria, spore-forming, chitinolytic and N2-fixing bacteria—and screened 122 strains for their potential PGP activities. The most common PGP trait was represented by P solubilization from phytate (69.7%), followed by siderophore production (65.6%), mineral P solubilization (49.2%) and IAA production (42.6%). About 76% of actinobacteria and 65% of chitinolytic bacteria displayed multiple PGP activities. Nineteen strains with best potential PGP activities, assigned to Sinorhizobium meliloti, Streptomyces spp., Arthrobacter phenanthrenivorans, Nocardiodes albus, Bacillus sp. pumilus group, Fictibacillus barbaricus and Lysinibacillus fusiformis, showed the ability to produce IAA and siderophores and to solubilize P from mineral phosphate and phytate, representing suitable candidates as biocontrol agents, biofertilisers and bioenhancers, in the perspective of targeted management of beneficial symbionts and their associated bacteria in sustainable food production systems.

Complete genome sequence of Serratia fonticola DSM 4576T, a potential plant growth promoting bacterium

Here, we present the first complete genome sequence of Serratia fonticola DSM 4576T, a potential plant growth promoting (PGP) bacterium which confers solubilization of inorganic phosphate, indole-3-acetic acid production, hydrogen cyanideproduction, siderophore production and assimilation of ammonia through the glutamate synthase (GS/GOGAT) pathway. This genome sequence is valuable for functional genomics and ecological studies which are related to PGP and biocontrol activities.

Genome Sequence of Enterobacter radicincitans DSM16656 T , a Plant Growth-Promoting Endophyte

Enterobacter radicincitans sp. nov. DSM16656(T) represents a new species of the genus Enterobacter which is a biological nitrogen-fixing endophytic bacterium with growth-promoting effects on a variety of crop and model plant species. The presence of genes for nitrogen fixation, phosphorous mobilization, and phytohormone production reflects this microbe's potential plant growth-promoting activity.

Genetic diversity and antifungal activity of native Pseudomonas isolated from maize plants grown in a central region of Argentina

Pseudomonas strains producing antimicrobial secondary metabolites play an important role in the biocontrol of phytopathogenic fungi. In this study, native Pseudomonas spp. isolates were obtained from the rhizosphere, endorhizosphere and bulk soil of maize fields in Córdoba (Argentina) during both the vegetative and reproductive stages of plant growth. However, the diversity based on repetitive-element PCR (rep-PCR) and amplified ribosomal DNA restriction analysis (ARDRA) fingerprinting was not associated with the stage of plant growth.Moreover, the antagonistic activity of the native isolates against phytopathogenic fungi was evaluated in vitro. Several strains inhibited members of the genera Fusarium, Sclerotinia or Sclerotium and this antagonism was related to their ability to produce secondary metabolites. A phylogenetic analysis based on rpoB or 16S rRNA gene sequences confirmed that the isolates DGR22, MGR4 and MGR39 with high biocontrol potential belonged to the genus Pseudomonas. Some native strains of Pseudomonas were also able to synthesise indole acetic acid and to solubilise phosphate, thus possessing potential plant growth-promoting (PGPR) traits, in addition to their antifungal activity. It was possible to establish a relationship between PGPR or biocontrol activity and the phylogeny of the strains.The study allowed the creation of a local collection of indigenous Pseudomonas which could be applied in agriculture to minimise the utilisation of chemical pesticides and fertilisers.

Bacillus strains isolated from rhizosphere showed plant growth promoting and antagonistic activity against phytopathogens

Seven bacterial isolates screened from rhizosphere of common bean growing at Uttarakhand Himalaya showed potential plant growth promoting (PGP) and antagonistic activities. Based on 16S rRNA gene sequence the isolate BPR7 was identified as Bacillus sp. BPR7. The strain BPR7 produced IAA, siderophore, phytase, organic acid, ACC deaminase, cyanogens, lytic enzymes, oxalate oxidase, and solubilized various sources of organic and inorganic phosphates as well as potassium and zinc. Strain BPR7 strongly inhibited the growth of several phytopathogens such as Macrophomina phaseolina, Fusarium oxysporum, F. solani, Sclerotinia sclerotiorum, Rhizoctonia solani and Colletotricum sp. in vitro. Cell-free culture filtrate of strain BPR7 also caused colony growth inhibition of all test pathogens. PGP and antifungal activities of Bacillus sp. BPR7 suggest that it may be exploited as a potential bioinoculant agent for P. vulgaris.

Potential Plant Growth-Promoting Activity of Pseudomonas sp Isolated from Paddy Soil in Malaysia as Biocontrol Agent

Alstonia scholaris R. Br. is an elegant evergreen tree of family Apocynaceae, generally bears leaf galls caused by an insect Pauropsylla tuberculata Crawf. In the present investigation the electrophoretic protein analysis technique was used, which revealed that some of the protein bands are varied and shown their presence and absence in gel at different stages of gall formation. The amount of total protein increased during early development and young stages of gall formation and falls down in older stages. It is due to a rapid enzymatic activity in gall tissue during early and mature stages as a response to insect interaction. Pathogens inject some elicitors and lead to the synthesis of different type of enzymes and some specific proteins at high amount in the plant. The insect switches the defense mechanism of the plant which results increased amount of some specific proteins in gall visible as dark bands in the gel. The degeneration of proteins during older stages shows the exit of insect and death of gall tissue.
