Tissue engineering and regenerative medicine (TERM) have paved a way for treating musculoskeletal diseases in a minimally invasive manner. The regenerative medicine cocktail involves the usage of mesenchymal stem/stromal cells (MSCs), either uncultured or culture-expanded cells along with growth factors, cytokines, exosomes, and secretomes to provide a better regenerative milieu in degenerative diseases. The successful regeneration of cartilage depends on the selection of the appropriate source of MSCs, the quality, quantity, and frequency of MSCs to be injected, and the selection of the patient at an appropriate stage of the disease. However, confirmation on the most favorable source of MSCs remains uncertain to clinicians. The lack of knowledge in the current cellular treatment is uncertain in terms of how beneficial MSCs are in the long-term or short-term (resolution of pain) and improved quality of life. Whether MSCs treatments have any superiority, exists due to sources of MSCs utilized in their potential to objectively regenerate the cartilage at the target area. Many questions on source and condition remain unanswered. Hence, in this review, we discuss the lineage differentiation potentials of various sources of MSCs used in the management of knee osteoarthritis and emphasize the role of tissue engineering in cartilage regeneration.

Learnings from COVID-19 Forced Lockdown on Regional Air Quality and Mitigation Potential for South Asia

South Asia is a hotspot of air pollution with limited resilience and hence, understanding the mitigation potential of different sources is critically important. In this context the country lockdown initiated to combat the COVID-19 pandemic (during March and April 2020 that is the pre-monsoon season) provides an unique opportunity for studying the relative impacts of different emission sources in the region. Here, we analyze changes in levels of air quality species across the region during selected lockdown periods using satellite and in-situ datasets. This analysis compares air quality levels during the lockdown against pre-lockdown conditions as well as against regional long-term mean. Satellite derived AOD, NO2, and CO data indicates an increase of 9.5%, 2%, and 2.6%, respectively, during the 2020 lockdown period compared to pre-lockdown over the South Asia domain. However, individual country statistics, urban site data, and industrial grid analysis within the region indicate a more varied picture. Cities with high traffic loads reported a reduction of 12–39% in columnar NO2 during lockdown, in-situ PM2.5 measurements indicate a 23–56% percent reduction over the country capitals and columnar SO2 has an approximate reduction of 50% over industrial areas. In contrast, pollutant emissions from natural sources e.g., from biomass burning were observed to be adversely affecting the air quality in this period potentially masking expected lockdown related air quality improvements. This study demonstrates the need for a more nuanced and situation specific understanding of sources of air pollutants (anthropogenic and natural) and for these sources to be better understood from the local to the regional scale. Without this deeper understanding, mitigation strategies cannot be effectively targeted, wasting limited resources as well as risking unintended consequences both for the atmosphere and how mitigation action is perceived by the wider public.

FINANCIAL SOURSCES OF INVESTMENT ACTIVISATION IN RUSSIA

The article proves the necessity of the intensification of the investment process as the condition of innovative economics creation in Russia. It is shown that the sharply reduced growth of investments in recent years, poor investment performance in 2018 create severe limitations of the economic growth at least till 2022. We have conducted the analysis of the financial potential of different sources of investment. Based on the analysis of the empirical data, it has been proven that the existing financial resources are sufficient for the sharp intensification of the investment process in Russia.

An Evaluation of Current Status of Renewable Energy Sources in India

Many people in rural society of India does not have access to natural energy resources like LPG , electricity as there is no facilitation of grid erection. Hence it becomes imperative for country like India to go for renewable energy resources as alternative to conventional energy resources. This paper evaluations the potential of different sources of renewable energy in India. It also highlights the trends in the growth of renewable energy sector, although at the same time shows that there is need of a hybrid renewable energy model for rural electrification in India. Paper also shows the scope in using the agriculture waste as bio fuel which burned otherwise and prompt health hazards besides environmental pollution, also describing many technologies which are in the demonstration stage like Tidal, OTEC, Solar thermal power plants. Many constraints put development in the field of solar and wind sector, on hold like availability of solar rays throughout the year . The main aim of the paper is to evaluation all possible options in renewable energy sector so that large segment of rural population can have access to electricity and to meet their basic energy needs.

Periosteum-derived mesenchymal progenitor cells in engineered implants promote fracture healing in a critical-size defect rat model.

An attractive alternative to bone autografts is the use of autologous mesenchymal progenitor cells (MSCs) in combination with biomaterials. We compared the therapeutic potential of different sources of mesenchymal stem cells in combination with biomaterials in a bone nonunion model. A critical-size defect was created in Sprague-Dawley rats. Animals were divided into six groups, depending on the treatment to be applied: bone defect was left empty (CTL); treated with live bone allograft (LBA); hrBMP-2 in collagen scaffold (CSBMP2 ); acellular polycaprolactone scaffold (PCL group); PCL scaffold containing periosteum-derived MSCs (PCLPMSCs ) and PCL containing bone marrow-derived MSCs (PCLBMSCs ). To facilitate cell tracking, both MSCs and bone graft were isolated from green fluorescent protein (GFP)-transgenic rats. CTL group did not show any signs of healing during the radiological follow-up (n=6). In the LBA group, all the animals showed bone bridging (n=6) whereas in the CSBMP2 group, four out of six animals demonstrated healing. In PCL and PCLPMSCs groups, a reduced number of animals showed radiological healing, whereas no healing was detected in the PCLBMSCs group. Using microcomputed tomography, the bone volume filling the defect was quantified, showing significant new bone formation in the LBA, CSBMP2 , and PCLPMSCs groups when compared with the CTL group. At 10weeks, GFP positive cells were detected only in the LBA group and restricted to the outer cortical bone in close contact with the periosteum. Tracking of cellular implants demonstrated significant survival of the PMSCs when compared with BMSCs. In conclusion, PMSCs improve bone regeneration being suitable for mimetic autograft design.

Morphogenetic potential of different sources of explants for efficient in vitro regeneration of Genipa sp.

Genipa americana L. is a species with high economic potential with considerable promise in the fruit, medicinal and agroindustrial fields. The in vitro cultivation of genipapo is an alternative that may overcome the difficulties imposed by the traditional propagation system, allowing the multiplication of plants on a large scale and using technologies in conservation programs and to improve phytopharmaceutical production. In this context, the aim of this work was to evaluate the morphogenetic potentials of different sources of explants to establish an efficient system for in vitro regeneration of G. americana. For callus induction, shoots differentiation and further plant regeneration, segments of hypocotyl, root and leaf from in vitro established seedlings were used. The explants were inoculated in MS medium supplemented with 6-benzylaminopurine (BAP) at concentrations of 0.0, 1.12, 2.25 and 3.37 mg L−1. The morphogenetic pattern and regeneration capacity showed correlations with the explant source and BAP concentration. MS medium supplemented with 1.12 mg L−1 BAP proved to be optimum for adventitious shoots induction in segments hypocotyl. It was possible to obtain a efficient protocol for the in vitro regeneration of G. americana that allowed high shoot regeneration rates (80%) using hypocotyl segments with low concentrations of BAP (1.12 mg L−1). The regenerated plantlets showed a high capacity for acclimatization, presenting 90% survival rate 30 days after exposure to the ex vitro conditions.

In vitro fermentation pattern and acidification potential of different sources of carbohydrates for ruminants given high concentrate diets

The in vitro fermentation pattern of five sources of carbohydrates of differing nature (maize grain, MZ; sucrose, SU; wheat bran, WB; sugarbeet pulp, BP; and citrus pulp, CT) under conditions of high concentrate diets for ruminants was studied. A first 8 h incubation trial was performed under optimal pH using inoculum from ewes given a fibrous diet, to compare fermentative characteristics of substrates. As planned, incubation pH ranged within 6.3 to 6.6. The gas produced from CT was higher than MZ, SU and BP from 4 and 6 h onwards, and at 8 h, respectively (p<0.05). There were no differences (p>0.05) on total volatile fatty acid (VFA) concentration, nor on acetate or propionate proportions, but butyrate was lowest (p<0.05) with CT and BP. The second incubation trial was performed in a poorly-buffered medium, with inoculum from ewes given a concentrate diet. All substrates showed a gradual drop of pH, being lowest with SU after 4 h (p<0.05). Throughout the incubation, gas production was highest with CT and lowest with MZ and BP (p<0.05). Total 8 h VFA concentration was higher with CT than BP, SU and MZ (p<0.05). Acetate proportion was higher, and that of propionate lower, with BP than WB (p<0.05), butyrate proportion being higher with MZ and WB than with BP and CT (p<0.05). Lactic acid concentration was higher (p<0.05) with SU than WB and BP. Fermentation characteristics and acidification potential of feeds depend on the nature of their carbohydrate fraction, and must be considered for practical applications.

Oxidant production from source-oriented particulate matter – Part 1: Oxidative potential using the dithiothreitol (DTT) assay

Abstract. Recent epidemiological evidence supports the hypothesis that health effects from inhalation of ambient particulate matter (PM) are governed by more than just the mass of PM inhaled. Both specific chemical components and sources have been identified as important contributors to mortality and hospital admissions, even when these end points are unrelated to PM mass. Sources may cause adverse health effects via their ability to produce reactive oxygen species in the body, possibly due to the transition metal content of the PM. Our goal is to quantify the oxidative potential of ambient particle sources collected during two seasons in Fresno, CA, using the dithiothreitol (DTT) assay. We collected PM from different sources or source combinations into different ChemVol (CV) samplers in real time using a novel source-oriented sampling technique based on single-particle mass spectrometry. We segregated the particles from each source-oriented mixture into two size fractions – ultrafine Dp ≤ 0.17 μm) and submicron fine (0.17 μm ≤ Dp ≤ 1.0 μm) – and measured metals and the rate of DTT loss in each PM extract. We find that the mass-normalized oxidative potential of different sources varies by up to a factor of 8 and that submicron fine PM typically has a larger mass-normalized oxidative potential than ultrafine PM from the same source. Vehicular emissions, regional source mix, commute hours, daytime mixed layer, and nighttime inversion sources exhibit the highest mass-normalized oxidative potential. When we apportion DTT activity for total PM sampled to specific chemical compounds, soluble copper accounts for roughly 50% of total air-volume-normalized oxidative potential, soluble manganese accounts for 20%, and other unknown species, likely including quinones and other organics, account for 30%. During nighttime, soluble copper and manganese largely explain the oxidative potential of PM, while daytime has a larger contribution from unknown (likely organic) species.

Intake fraction variability between air pollution emission sources inside an urban area.

The cost-effective mitigation of adverse health effects caused by air pollution requires information on the contribution of different emission sources to exposure. In urban areas the exposure potential of different sources may vary significantly depending on emission height, population density, and other factors. In this study, we quantified this intraurban variability by predicting intake fraction (iF) for 3,066 emission sources in Warsaw, Poland. iF describes the fraction of the pollutant that is inhaled by people in the study area. We considered the following seven pollutants: particulate matter (PM), nitrogen oxides (NOx), sulfur dioxide (SO2), benzo[a] pyrene (BaP), nickel (Ni), cadmium (Cd), and lead (Pb). Emissions for these pollutants were grouped into four emission source categories (Mobile, Area, High Point, and Other Point sources). The dispersion of the pollutants was predicted with the CALPUFF dispersion model using the year 2005 emission rate data and meteorological records. The resulting annual average concentrations were combined with population data to predict the contribution of each individual source to population exposure. The iFs for different pollutant-source category combinations varied between 51 per million (PM from Mobile sources) and 0.013 per million (sulfate PM from High Point sources). The intraurban iF variability for Mobile sources primary PM emission was from 4 per million to 100 per million with the emission-weighted iF of 44 per million. These results propose that exposure due to intraurban air pollution emissions could be decreased more effectively by specifically targeting sources with high exposure potency rather than all sources.

Human endometrial adult stem cells can be differentiated into hepatocyte cells

Human endometrial adult stem cells have great potentiality to differentiated into other cell type. In the paper we hypothesize that these cells have remarkable capacity for differentiation into hepatocyte cells. • Human endometrial adult stem cells a new reliable stem cell source for differentiation into hepatocyte cells. • Human endometrial adult stem cells can provide the attractive alternative resources for liver cell-based therapies. Liver transplantation is the only choice for patients with end-stage liver diseases. Hepatocyte transplantation is a promising alternative for treatment of these groups. However, the major challenge is insufficiency of donor organs that can provide good-quality cells. Therefore, numerous experimental and clinical studies have evaluated the potential of different sources of adult stem cells, which differentiate into hepatocytes, for cell therapy. Endometrial (stem) stromal cells are readily isolated and expandable; moreover, these cells have more clonogenicity and pose less technical problems, so they are considered to possess great autologous therapeutic potential. We propose that endometrial stem (stromal) cells could be an invaluable and realistic source in this regard.

Nitrous oxide and methane fluxes in south Brazilian gleysol as affected by nitrogen fertilizers

Nitrogen fertilizers increase the nitrous oxide (N2O) emission and can reduce the methane (CH4) oxidation from agricultural soils. However, the magnitude of this effect is unknown in Southern Brazilian edaphoclimatic conditions, as well as the potential of different sources of mineral N fertilizers in such an effect. The aim of this study was to investigate the effects of different mineral N sources (urea, ammonium sulphate, calcium nitrate, ammonium nitrate, Uran, controlled- release N fertilizer, and urea with urease inhibitor) on N2O and CH4 fluxes from Gleysol in the South of Brazil (Porto Alegre, RS), in comparison to a control treatment without a N application. The experiment was arranged in a randomized block with three replications, and the N fertilizer was applied to corn at the V5 growth stage. Air samples were collected from a static chambers for 15 days after the N application and the N2O and CH4 concentration were determined by gas chromatography. The topmost emissions occurred three days after the N fertilizer application and ranged from 187.8 to 8587.4 µg m-2 h-1 N. The greatest emissions were observed for N-nitric based fertilizers, while N sources with a urease inhibitor and controlled release N presented the smallest values and the N-ammonium and amidic were intermediate. This peak of N2O emissions was related to soil NO3--N (R² = 0.56, p < 0.08) when the soil water-filled pore space was up to 70 % and it indicated that N2O was predominantly produced by a denitrification process in the soil. Soil CH4 fluxes ranged from -30.1 µg m-2 h-1 C (absorption) to +32.5 µg m-2 h-1 C (emission), and the accumulated emission in the period was related to the soil NH4+-N concentration (R² = 0.82, p < 0.001), probably due to enzymatic competition between nitrification and metanotrophy processes. Despite both of the gas fluxes being affected by N fertilizers, in the average of the treatments, the impact on CH4 emission (0.2 kg ha-1 equivalent CO2-C ) was a hundredfold minor than for N2O (132.8 kg ha-1 equivalent CO2-C). Accounting for the N2O and CH4 emissions plus energetic costs of N fertilizers of 1.3 kg CO2-C kg-1 N regarding the manufacture, transport and application, we estimated an environmental impact of N sources ranging from 220.4 to 664.5 kg ha-1 CO2 -C , which can only be partially offset by C sequestration in the soil, as no study in South Brazil reported an annual net soil C accumulation rate larger than 160 kg ha-1 C due to N fertilization. The N2O mitigation can be obtained by the replacement of N-nitric sources by ammonium and amidic fertilizers. Controlled release N fertilizers and urea with urease inhibitor are also potential alternatives to N2O emission mitigation to atmospheric and systematic studies are necessary to quantify their potential in Brazilian agroecosystems.

EVALUATION OF ENGRAFTMENT AND DIFFERENTIATION OF DIFFERENT SOURCES OF HUMAN STEM CELLS IN FETAL SHEEP BRAIN

It is known that the brain can be affected by many diseases/disorders for which therapies are not yet available. Recently, different investigators have reported on the ability of stem cells to differentiate into different neural cell types in vitro and in vivo. This knowledge opened new perspectives in stem cell therapy for neurological disorders/diseases. Hematopoietic (HSC), mesenchymal (MSC) and neural (NSC) stem cells are the most promising source of stem cells given either their easiness of harvest or their differentiative ability. Upon grafting into the brain, stem cells could give rise directly to new neurons or could generate other cells of the central nervous system, such oligodendrocytes, or glial cells, that may contribute to neural growth. Furthermore these cells could be therapeutic by secreting missing substances or enzymes that are needed for the normal function of the brain. Since mesenchymal stem cells (MSCs) have been shown to suppress T-cell activation in vitro and in vivo, transplantation of these cells into the brain could led not only to a source of new neural cells but also could provide an anti-inflammatory response. In order to investigate the therapeutic potential of different sources of stem cells, pre-immune fetal sheep were transplanted by intraperitoneal injection, at 60 days of gestation, with HSC, NSC or MSC. Animals were euthanized at 60-80 days post transplantation and tissues were collected from different regions of the brain and analyzed by flow cytometry and immunohistochemistry using different antibodies for known neural markers. Levels of donor cell engraftment and types of neural cells obtained were then correlated to the source of the transplanted cells. We hope that these studies will be able to discern what is/are the best source(s) of stem cells to use for neural cell replacement therapies.

Creation of long‐lasting blood vessels in vivo

A major challenge in tissue engineering and regenerative medicine is the creation of stable vasculature. One approach in vascularizing an engineered tissue is to add the cellular components of blood vessels (endothelial and perivascular cells) directly in the tissue‐engineered construct. We have shown the formation of long‐lasting vascular networks in vivo by co‐implanting human umbilical cord vein endothelial cells (HUVECs) and 10T1/2 mesenchymal precursor cells in a three‐dimensional fibronectin‐type I collagen scaffold. Because mesenchymal precursor cells differentiate into perivascular cells and fortify blood vessels in vivo, the resulting vessels are stable and functional for one year. In contrast, the HUVEC‐alone constructs showed minimal perfusion and disappeared by two months. These findings indicate that fostering endothelial‐perivascular cell interactions in vivo offers a promising approach for engineering blood vessels. Furthermore, the tissue‐engineered blood vessel model provides an ideal platform to study cellular and molecular mechanisms of vessel formation and maturation. The potential of different sources of vascular cells in the creation of engineered vessels in vivo can also be tested with this model.Supported in part by NIH grants (P01CA80124, R01CA96915).

Engraftment Potential of Different Sources of Human Hematopoietic Progenitor Cells in BNX Mice

Human hematopoietic progenitor cells (HPCs) from mobilized peripheral blood mononuclear cells (PBMCs), adult bone marrow (ABM), and fetal bone marrow (FBM) were evaluated for their ability to produce multilineage human hematopoietic engraftment in vivo. Sublethally irradiated BNX (beige/nude/xid) mice were injected with either unfractionated cells or CD34+ cells purified from these sources. The presence of human cells in the mouse PB, BM, and spleen was evaluated by flow cytometry at either 6 to 8 weeks or 6 months postinjection. Recipients with > or = 1% human cells in any of these tissues were considered chimeric. Of 26 mice injected with FBM, 4 showed up to 73% human cells in the BM or spleen at 6 months. The phenotypes of these cells included CD13/33+ myelomonocytic cells (38%), CD19+ B cells (67%), and CD34+ progenitor cells (28%). In contrast, ABM gave rise to a mean of 5% human cells in the PB in 2 of 42 (4%) recipients at 6 to 8 weeks. These circulating human cells were predominantly CD3+, whereas CD13/33+ and CD34+ cells were detected in the BM for up to 6 months. A total of 18% of mice injected with PBMCs showed a mean of 36% human cells in the PB. Both the BM and spleens of PBMC-injected mice contained CD3+ cells in a proportion similar to that observed in the PB. These CD3+ cells were phenotypically mature CD4+,CD8- or CD4-,CD8+ T cells and coexpressed a variety of Vbeta T-cell receptor (TCR) genes. The percentage of CD3+ cells in the circulation of chimeric recipients injected with either FBM, ABM, or PBMCs correlated well with the input CD3+ cell dose for each of these HPC sources (r = .99). The high levels of engraftment of CD3+ cells in recipients of PBMCs and the long-term multilineage engraftment of FBM recipients have important implications for developing strategies to study the regulation of these human cells in vivo.

Greenhouse evaluation of agronomic potential of different sources of phosphate fertilizer in a typical concretionary soil of Northern Ghana

The concretionary soils of Northern Ghana, which are near neutral with respect to pH and which comprise mostly lateritic ferruginous nodules are known to sorb significant amounts of phosphate. Instead of imported superphosphate, the use of less expensive indigenous Togo rock phosphate (PR) or partially acidulated (50%) Togo rock phosphate (PAPR-50), are possible alternative phosphate fertilizer options for these soils. The objective of this research was to evaluate the effectiveness of freshly-applied SSP, PR and PAPR-50, and the effectiveness of the residues of these fertilizers in a glasshouse pot study. Laboratory studies were also undertaken to study the transformation of these fertilizers after their application to the concretionary ferruginous soils. In the greenhouse study, yield of dried tops and the P uptake by the tops of maize var. Dobidi (Zea mays) was used to measure fertilizer effectiveness. One level of P was applied for each fertilizer (26.4 kg P ha−1). Plants were grown for 28 days. After harvesting the first crop, subsequent cropping was carried out to evaluate the effects of the residual P in the pots. The results showed that increases in dry matter yield of shoot and total P uptake followed the trend SSP > PAPR-50 > PR > control. The relative agronomic effciency (RAE) of PAPR-50 was 58% that of commercial SSP in increasing growth of the crop, while that of PR was only 23%. The residual effect of either PAPR-50 or PR on dry matter yield and total P uptake was found to be negligible compared with SSP, suggesting that apatitic P was poorly effective relative to SSP in the used soils. The P fractionation results confirmed that PR and PAPR-50 did not significantly increase any of the P fractions in either the soil fines or nodules after the first crop. By contrast, application of SSP increased all extractable Pi fractions, most of the P added being recovered from the nodules in forms associated with Fe (hydroxide and residual Pi). It is concluded that, relative to SSP, the P from residues of PAPR-50 and PR are poorly effective in the soils studied for sustainable plant production.
