Area-Efficient Mixed-Signal Time-to-Digital Converter Integration for Time-Resolved Photon Counting.

Digital histogram generation for time-resolved measurements with single-photon avalanche diode (SPAD) sensors requires the storage of many timestamp signals. This work presents a mixed-signal time-to-digital converter (TDC) that uses analog storage to achieve an area-efficient design that can be integrated in large SPAD arrays. Fabricated using a 150 nm CMOS process, the prototype occupies an area of only 18.3 µm × 36.5 µm, a notable size reduction compared to conventional designs. The experimental results demonstrated high performance, with an integral nonlinearity (INL) of 0.35/0.14 least significant bit (LSB) and a differential nonlinearity (DNL) of 0.14/-0.12 LSB. In addition, the proposed TDC can support the construction of histograms comprising up to 512 bins, making it an effective solution to accommodate a wide range of resolution requirements. Validated in a point-of-care (PoC) device for fluorescence lifetime measurements, it distinguished between lifetimes of approximately 4.1 ns, 3.6 ns and 80 ns with the Alexa Fluor (AF) 546 and 568 dyes and Quantum Dot (QD) 705, respectively. The analog storage design and area-efficient architecture offer a novel approach to integrating TDCs in SPAD-based systems, with potential applications in medical diagnostics and beyond.

Label-Free DNA Hybridization Detection Using a Highly Sensitive Fiber Microcavity Biosensor.

A novel label-free optical fiber biosensor, based on a microcavity fiber Mach-Zehnder interferometer, was developed and practically demonstrated for DNA detection. The biosensor was fabricated using offset splicing standard communication single-mode fibers (SMFs). The light path of the sensor was influenced by the liquid sample in the offset open cavity. In the experiment, a high sensitivity of -17,905 nm/RIU was achieved in the refractive index (RI) measurement. On this basis, the probe DNA (pDNA) was immobilized onto the sensor's surface using APTES, enabling real-time monitoring of captured complementary DNA (cDNA) samples. The experimental results demonstrate that the biosensor exhibited a high sensitivity of 0.32 nm/fM and a limit of detection of 48.9 aM. Meanwhile, the sensor has highly repeatable and specific performance. This work reports an easy-to-manufacture, ultrasensitive, and label-free DNA biosensor, which has significant potential applications in medical diagnostics, bioengineering, gene identification, environmental science, and other biological fields.

A Study on the Relation Between Voltage and Bilirubin Concentration in Artificial Blood Solution by Bilirubin Detector

This research project focuses on investigating the correlation between electric potential difference and bilirubin concentration within an artificial blood solution, utilizing the principles of light absorption. The bilirubin detector's design and construction incorporate a blue light source with a 460 nm wavelength and a green light source with a 560 nm wavelength, enabling interaction with bilirubin concentrations ranging from 2 to 14 mg/dl. Analysis of the project's outcomes revealed a direct relationship between the bilirubin detector's output voltage and the concentration of the bilirubin solution. Notably, an interaction with bilirubin was observed at the 460 nm wavelength, influencing light absorption. In cases of higher bilirubin concentration, light transmittance was diminished, leading to a heightened voltage difference. Conversely, lower bilirubin concentrations facilitated increased light transmission, resulting in a reduced voltage difference. This study contributes to a comprehensive understanding of bilirubin detection methodologies and their potential applications in medical diagnostics.

Non-invasive detection of haemoglobin, platelets, and total bilirubin using hyperspectral cameras

Hyperspectral imaging has emerged as a promising high-resolution and real-time imaging technology with potential applications in medical diagnostics and surgical guidance. In this study, we developed a high-speed hyperspectral camera by integrating a Fabry-Perot cavity filter on each CMOS pixel. We used it to non-invasively detect three blood components (haemoglobin, platelet, and total bilirubin). Specifically, we acquired transmission images of the subject's fingers, extracted spectral signals at each wavelength, and used dynamic spectroscopy to obtain non-invasive blood absorption spectra. The prediction models were established using the PLSR method and were modelled and validated based on the standard clinical-biochemical test values. The experimental results demonstrated excellent performance. The best predictions were obtained for haemoglobin, with a high related coefficient (R) of 0.85 or more in both the calibration and prediction sets and a mean absolute percentage error (MAPE) of only 5.7%. The results for total bilirubin were also ideal, with R values exceeding 0.8 in both sets and a MAPE of 10.6%. Although the prediction results for platelets were slightly less satisfactory, the error was still less than 15%, indicating that the results were also acceptable. Overall, our study highlights the potential of hyperspectral imaging technology for the development of portable and affordable devices for blood analysis, which can be used in various settings.

Lab-on-a-chip based mechanical actuators and sensors for single-cell and organoid culture studies.

All living cells constantly experience and respond to mechanical stresses. The molecular networks that activate in cells in response to mechanical stimuli are yet not well-understood. Our limited knowledge stems partially from the lack of available tools that are capable of exerting controlled mechanical stress to individual cells and at the same time observing their responses at subcellular to molecular resolution. Several tools such as rheology setups, micropipetes, and magnetic tweezers have been used in the past. While allowing to quantify short-time viscoelastic responses, these setups are not suitable for long-term observations of cells and most of them have low throughput. In this Perspective, we discuss lab-on-a-chip platforms that have the potential to overcome these limitations. Our focus is on devices that apply shear, compressive, tensile, and confinement derived stresses to single cells and organoid cultures. We compare different design strategies for these devices and highlight their advantages, drawbacks, and future potential. While the majority of these devices are used for fundamental research, some of them have potential applications in medical diagnostics and these applications are also discussed.

Cell immunocapture microfluidic chip based on high-affinity recombinant protein binders

Cell immunocapture microfluidic devices represent a rapidly developing field with many potential applications in medical diagnostics. The core of such approach lies in the cell binding to antibody coated surfaces through their surface receptors. Here we show, that the small recombinant protein binders (PBs) can be used for this purpose as well, with the advantage of their constructional flexibility, possibility of fusion with range of tags and cheap mass production. For this purpose, two different PBs derived from Albumin Binding Domain (ABDwt) of streptococcal protein G, so called REX and ARS ligands with proved high affinity and selectivity to the human interleukin-23 (IL-23R) and IL-17 receptor A were used. Four PBs variants recognizing two different epitopes on two different receptors and two PBs variants binding to the same epitope on one receptor but having different peptide spacer with Avitag sequence necessary for their immobilization on sensor surface were tested for cell-capture efficiency. The glass microfluidic Y-system with planar immunocapture channel working in so-called stop-flow dynamic regime was designed. Up to 60–74% immunocapture efficiency of model THP-1 cells on REX/ARS surfaces and practically no cell binding on control ABDwt surfaces was achieved. Moreover, the specific immunocapture of THP-1 cells from mixture with IL-17RA negative DU-145 cells was demonstrated. We discuss the role of the epitope, affinity and immobilization spacer of PBs as well as the influence of stop-flow dynamic regime on the effectivity of THP-1 cell immunocapture. Results can be further exploited in design of microfluidic devices for rare cells immunocapture.

Paper-Based Acoustofluidics for Separating Particles and Cells.

Paper is emerging as a versatile platform for automated fluid handling with a broad range of applications in medical diagnostics and analytical chemistry. However, selectively controlling analyte transport in paper to achieve concentration or selection has been a challenge for functional analysis. Here, by combining paper-based microfluidics with acoustics, we present a rapid and powerful method to size dependently control movement of microparticles and cells in paper using surface acoustic waves (SAW). We demonstrate the unique capability of the paper-based SAW approach to trap and concentrate microparticles in paper and release them when required, achieving collection efficiency of over 98%. Given the correlation between collection efficiency, size, and applied power, the paper-based SAW approach is applied to isolate a mixture of microparticles (1.1, 3.2, and 5 μm in diameter) into different regions and also to trap and concentrate human prostate cancer PC3 cells at a predetermined site. This paper-based SAW approach provides opportunities to develop powerful and low-cost selection and analysis tools, capable of processing complex multicomponent samples, with potential applications in medical diagnostics.

Ultrasensitive NOX Detection in Simulated Exhaled Air: Enhanced Sensing via Alumina Modification of In‐Situ Grown WO3 Nanoblocks

Seedless growth of vertically aligned nanostructures, which can induce smoother transport and minimize Ohmic contact between substrate and semiconductor, can be fabricated by in situ growth utilizing modified hydrothermal methods. Such devices can be useful in designing non-invasive ultrasensitive hand-held sensors for diagnostic identification of volatile organic compounds (VOCs) in exhaled air, offering pain-free and easier detection of long-term diseases such as asthma. In the present work, WO3 nanoblocks, with a high surface area and porosity, have been grown directly over transparent conducting oxide to minimize Ohmic resistance, facilitating smoother electron transfer and enhanced current response. Further modification with porous alumina (γ-Al2 O3 ), by electrodeposition, resulted in the selective and ultrasensitive detection of NOX in simulated exhaled air. Crystal phase purity of as-fabricated pristine as well modified samples is validated with X-ray diffraction analysis. Morphological and microstructural analyses reveal the successful deposition of porous alumina over the surface of WO3 . Improved surface area and porosity is presented by porous alumina in the modified WO3 device, suggesting more active sites for the gas molecules to get adsorbed and diffuse through the pores. Oxygen vacancies, which are detrimental in the transport phenomenon in the presented sensors, have been studied using X-ray photoelectron spectroscopic (XPS) analysis. Gas sensing studies have been performed by fabricating chemiresistor devices based on bare WO3 and Al2 O3 -modified WO3 . The higher sensitivity for NOX gas in case of γ-Al2 O3 -modified WO3 based devices, as compared to bare WO3 -based devices, is attributed to the better surface area and charge transport kinetics. The presented device strategy offers crucial understanding in the design and development of non-invasive, hand-held devices for NO gas present in the human breath, with potential application in medical diagnostics.

INVESTIGATION OF ANTENNA ARRAY CONFIGURATIONS USING FAR-FIELD HOLOGRAPHIC MICROWAVE IMAGING TECHNIQUE

Biomedical imaging has played an important role in identifying and monitoring the effectiveness of the current state of the art treatments for many diseases. The authors recently proposed a novel single-transmitter-multiple-receiver holographic microwave imaging (HMI) technique for imaging small inclusion embedded in a dielectric object which has potential application in medical diagnostics. HMI image quality depends highly on the antenna baseline difference, in order words, the antenna array configuration. Different antenna arrays produce different quality of dielectric images by using HMI imaging algorithm. This paper investigates the antenna array configurations effect on image quality by using HMI imaging approach. Three configurations including spiral, random and regularly spaced arrays are presented. Both simulated and experimental results are obtained and compared to fully demonstrate the effectiveness of antenna arrays to the HMI technique. The results show that the proposed spiral and random array configurations have an ability to produce high-resolution images at significantly lower costs than regularly spaced arrays.

Tuning nano electric field to affect restrictive membrane area on localized single cell nano-electroporation

Interaction of electric field with biological cells is an important phenomenon for field induced drug delivery system. We demonstrate a selective and localized single cell nano-electroporation (LSCNEP) by applying an intense electric field on a submicron region of the single cell membrane, which can effectively allow high efficient molecular delivery but low cell damage. The delivery rate is controlled by adjusting transmembrane potential and manipulating membrane status. Thermal and ionic influences are deteriorated from the cell membrane by dielectric passivation. Either reversible or irreversible by LSCNEP can fully controlled with potential applications in medical diagnostics and biological studies.

One-Step Solvothermal Synthesis of Targetable Optomagnetic Upconversion Nanoparticles for in Vivo Bimodal Imaging

Bionanoparticles and nanostructures with high biocompatibility and stability, low toxicity, diversification of imaging modality, and specificity of targeting to desired organs or cells are of great interest in nanobiology and medicine. However, integrating all of these desired features into a single bionanoparticle, which can be applied to biomedical applications and eventually in clinical prediagnosis and therapy, is still a challenge. We herein report a facile one-step solvothermal approach to fabricate targetable and biocompatible β-NaYF4:Yb,Gd,Tm upconversion nanoparticles (UCNPs) with bimodal-signals (near-infrared (NIR) fluorescence and magnetic resonance (MR) signals) using hyaluronic acid (HA) as a multifunctional molecule. The prepared UCNPs with low toxicity are successfully applied for in vitro and in vivo targeted tumor imaging. The developed biomimetic surface modification approach for the synthesis of biomolecule-guided multifunctional UCNPs holds great potential applications in medical diagnostics and therapy.

Ultra-localized single cell electroporation using silicon nanowires

Analysis of cell-to-cell variation can further the understanding of intracellular processes and the role of individual cell function within a larger cell population. The ability to precisely lyse single cells can be used to release cellular components to resolve cellular heterogeneity that might be obscured when whole populations are examined. We report a method to position and lyse individual cells on silicon nanowire and nanoribbon biological field effect transistors. In this study, HT-29 cancer cells were positioned on top of transistors by manipulating magnetic beads using external magnetic fields. Ultra-rapid cell lysis was subsequently performed by applying 600-900 mV(pp) at 10 MHz for as little as 2 ms across the transistor channel and the bulk substrate. We show that the fringing electric field at the device surface disrupts the cell membrane, leading to lysis from irreversible electroporation. This methodology allows rapid and simple single cell lysis and analysis with potential applications in medical diagnostics, proteome analysis and developmental biology studies.

Rapid Transformation of Protein-Caged Nanomaterials into Microbubbles As Bimodal Imaging Agents

We present a general method for converting colloidal nanomaterials into microbubbles as ultrasound contrast agents. Protein-caged nanomaterials, made either by self-assembled nanoparticles' protein corona or by fluorescent gold nanoclusters, can be rapidly transformed into microbubbles via a sonochemical route, which promote disulfide cross-linking of cysteine residues between protein-caged nanomaterials and free albumin during acoustic cavitation. The proposed methods yielded microbubbles with multiple functions by adjusting the original nanoparticle/protein mixture. We also showed a new dual-modal imaging agent of fluorescent gold microbubbles in vitro and in vivo, which can hold many potential applications in medical diagnostics and therapy.

Detection and identification of proteins using nanoparticle–fluorescent polymer ‘chemical nose’ sensors

A sensor array containing six non-covalent gold nanoparticle-fluorescent polymer conjugates has been created to detect, identify and quantify protein targets. The polymer fluorescence is quenched by gold nanoparticles; the presence of proteins disrupts the nanoparticle-polymer interaction, producing distinct fluorescence response patterns. These patterns are highly repeatable and are characteristic for individual proteins at nanomolar concentrations, and can be quantitatively differentiated by linear discriminant analysis (LDA). Based on a training matrix generated at protein concentrations of an identical ultraviolet absorbance at 280 nm (A280 = 0.005), LDA, combined with ultraviolet measurements, has been successfully used to identify 52 unknown protein samples (seven different proteins) with an accuracy of 94.2%. This work demonstrates the construction of novel nanomaterial-based protein detector arrays with potential applications in medical diagnostics.
