Continuous Theta-Burst Stimulation on the Left Posterior Inferior Frontal Gyrus Perturbs Complex Syntactic Processing Stability in Mandarin Chinese

Abstract The structure of human language is inherently hierarchical. The left posterior inferior frontal gyrus (LpIFG) is proposed to be a core region for constructing syntactic hierarchies. However, it remains unclear whether LpIFG plays a causal role in syntactic processing in Mandarin Chinese and whether its contribution depends on syntactic complexity, working memory, or both. We addressed these questions by applying inhibitory continuous theta-burst stimulation (cTBS) over LpIFG. Thirty-two participants processed sentences containing embedded relative clauses (i.e., complex syntactic processing), syntactically simpler coordinated sentences (i.e., simple syntactic processing), and non-hierarchical word lists (i.e., word list processing) after receiving real or sham cTBS. We found that cTBS significantly increased the coefficient of variation, a representative index of processing stability, in complex syntactic processing (esp., when subject relative clause was embedded) but not in the other two conditions. No significant changes in d′ and reaction time were detected in these conditions. The findings suggest that (a) inhibitory effect of cTBS on the LpIFG might be prominent in perturbing the complex syntactic processing stability but subtle in altering the processing quality; and (b) the causal role of the LpIFG seems to be specific for syntactic processing rather than working memory capacity, further evidencing their separability in LpIFG. Collectively, these results support the notion of the LpIFG as a core region for complex syntactic processing across languages.

355 Connecting computational models of reading to the brain in post-stroke alexia

OBJECTIVES/GOALS: Many left hemisphere stroke survivors have a reading disorder (alexia), which is experienced as decreasing well-being. Therapies produce inconsistent results, demonstrating a need for treatment response predictors. We are investigating neural correlates of reading computational models to identify biomarkers to improve therapeutic outcomes. METHODS/STUDY POPULATION: Artificial neural network models of reading, mapping between orthography (visual word form), phonology (auditory word form), and semantics (word meaning), are trained to read single words at a healthy, adult capacity. The models are independently damaged to reflect the individual orthography-to-semantics, semantics-to-phonology, and orthography-to-phonology deficits of a sample of left hemisphere stroke survivors (n = 85). These deficits are measured with cognitive tests assessing the intactness of mappings between representations. Model damage is enacted by removing percentages of the connections between representations. For each type of deficit, the percentages of links removed are entered into a voxel-based lesion symptom mapping analysis to identify areas of cortex associated with that mapping. RESULTS/ANTICIPATED RESULTS: We anticipate that the neural correlates of model layers will be localized to a mostly left-lateralized network. Increased damage to the links between semantics and phonology in the model will likely be related to lesions involving the left posterior superior temporal sulcus and inferior frontal gyrus (IFG). Damaged orthography-to-semantic links will be related to the left fusiform gyrus (FG) and IFG. Finally, damage to the orthography-to-phonology links will be related to the left FG and superior temporal gyrus. DISCUSSION/SIGNIFICANCE: Mapping components of language models onto the brain will improve our understanding of the neural networks supporting language processing. Identifying these neural correlates may also produce biomarkers that can be used in predicting reading impairment at the acute stage or optimizing therapy in the chronic stage of stroke.

Speech and lexico-semantic errors during direct cortical stimulation mapping of the language-dominant hemisphere: effects of object and action naming.

In this retrospective study, the authors aimed to establish the stereotactically defined probability distribution for speech (i.e., anarthria, speech arrest) and lexico-semantic errors (i.e., anomia) through direct cortical stimulation (DCS) by using two tasks: action naming and object naming. They also analyzed the patterns of interindividual variability in the localization of the language sites involved, and investigated whether any patient or lesion location factors were associated with greater variability. Eighty-one Italian-speaking patients who underwent awake surgery between 2010 and 2021 for low- and high-grade gliomas in eloquent areas of the language-dominant hemisphere were entered in the analyses. The intraoperative DCS protocol included automatic speech tasks, object naming, and action naming. The position of the tags, as depicted on the intraoperative video or photograph, was transposed into Montreal Neurological Institute space. Subsequently, a 2D scatterplot and cluster analysis were performed. Associations between various clinical and radiological characteristics and the quantity of positive stimulated sites were determined by univariate analyses using binary logistic regression. Associated variables (p < 0.2) were included in stepwise multivariate logistic regression with backward elimination (p < 0.05). A total of 1380 cortical sites were stimulated, with a positive response in 511 cases (37%). Most anarthric errors were triggered when stimulating the left precentral gyrus, and most speech arrest errors were elicited when stimulating the left posterior inferior frontal gyrus. Anomias were found in the left inferior frontal gyrus and in the posterior part of the left temporal lobe for object naming. DCS to the left dorsal premotor cortex elicited anomic errors for action naming. Anomias were also elicited during DCS to the left posterior temporal lobe, with both object and action naming. The distribution of speech and lexico-semantic errors is in line with the current literature. The action-naming results are new and mostly involve the dorsal premotor cortex. These findings stress the importance of maximizing the use of different language tasks during surgery, because even when looking for the same type of errors, different tasks may be better suited to map specific brain regions. DCS with action and object naming identifies more positive sites than object naming alone.

Cortical, subcortical, and cerebellar contributions to language processing: A meta-analytic review of 403 neuroimaging experiments.

Language is a key human faculty for communication and interaction that provides invaluable insight into the human mind. Previous work has dissected different linguistic operations, but the large-scale brain networks involved in language processing are still not fully uncovered. Particularly, little is known about the subdomain-specific engagement of brain areas during semantic, syntactic, phonological, and prosodic processing and the role of subcortical and cerebellar areas. Here, we present the largest coordinate-based meta-analysis of language processing including 403 experiments. Overall, language processing primarily engaged bilateral fronto-temporal cortices, with the highest activation likelihood in the left posterior inferior frontal gyrus (IFG). Whereas we could not detect any syntax-specific regions, semantics specifically engaged left posterior temporal areas (left fusiform and occipitotemporal cortex) and the left frontal pole. Phonology showed highest subdomain-specificity in bilateral auditory and left postcentral regions, whereas prosody engaged specifically the right amygdala and the right IFG. Across all subdomains and modalities, we found strong bilateral subcortical and cerebellar contributions. Especially the right cerebellum was engaged during various processes, including speech production, visual, and phonological tasks. Collectively, our results emphasize consistent recruitment and high functional modularity for general language processing in bilateral domain-specific (temporo-frontal) and domain-general (medial frontal/anterior cingulate cortex) regions but also a high specialization of different subareas for different linguistic subdomains. Our findings refine current neurobiological models of language by adding novel insight into the general sensitivity of the language network and subdomain-specific functions of different brain areas and highlighting the role of subcortical and cerebellar regions for different language operations. Continuous theta-burst stimulation over the left posterior inferior frontal gyrus induced compensatory plasticity in the language network.

IntroductionContinuous theta-burst stimulation (cTBS) has been used as an effective tool in inducing inhibitory aftereffect within a short time periods in the motor cortex; this has been demonstrated in the language network to a limited degree with controversial effect. In this study, we aimed to delineate the offline effect of cTBS-induced changes to the left posterior inferior frontal gyrus (pIFG) in healthy subjects using functional magnetic resonance imaging (fMRI).MethodsTwenty healthy, normal subjects (mean age: 30.84 years) were recruited. They all were right-handed and had no contra-indications for fMRI or cTBS. They were randomly assigned into the treatment group or the sham control group.ResultsANOVA showed that cTBS had a significant main effect only when the sham treatment was subtracted from the real stimulation in left superior temporal, left inferior frontal gyrus, thalamus, and right insular cortex (uncorrected p < 0.002). The subjects' post-cTBS condition differed significantly from their pre-cTBS condition in the left pIFG (uncorrected p < 0.002). There were interactions in the pIFG, bilateral superior parietal lobules, left superior temporal, left supramarginal, and left cuneus areas. The application of cTBS induced increased BOLD signals in language-related networks by stimulating the left pIFG (BA 44). This implies that inhibiting the pIFG led to increased use of language network resources.ConclusionThis study demonstrated cTBS-induced changes in the language network caused by stimulation of the left pIFG. Based on these findings, future studies on the therapeutic effects of cTBS on the right Broca's homolog area are warranted.

Neural sensitivity to semantic neighbors is stable across the adult lifespan

As we age, language reflects patterns of both stability and change. On the one hand, vocabulary and semantic abilities are largely stable across the adult lifespan, yet lexical retrieval is often slower and less successful (i.e., slower picture naming times, increased tip of the tongue incidents). Although the behavioral bases of these effects have been well established, less is known about the brain regions that support these age-related differences. We used functional Magnetic Resonance Imaging (fMRI) to examine the neural basis of picture naming. Specifically, we were interested in whether older adults would be equally sensitive to semantic characteristics, specifically the number of semantic near neighbors. Near neighbors, defined here as items with a high degree of semantic feature overlap, were of interest as these are thought to elicit competition among potential candidates and increase naming difficulty. Consistent with prior reports, pictures with more semantic near neighbors were named more slowly and less accurately for all adults. Additionally, this interference for naming times was larger as age increased, starting around 30 years old. In contrast to the age-related behavioral slowing, the neural basis of these effects was stable across adulthood. Across all adults, a number of language-relevant regions including left posterior middle temporal gyrus and left inferior frontal gyrus, pars triangularis were sensitive to the number of near neighbors. Our results suggest that although middle-aged and older adults’ picture naming is more slowed by increased semantic competition, the brain regions supporting semantic processes remain stable across the adult lifespan.

348 Resting state fMRI connectivity in individuals with aphasia and the role of the inferior frontal gyrus

OBJECTIVES/GOALS: This poster will explore lesion-symptom mapping and resting state functional magnetic resonance imaging (rsfMRI) in participants with aphasia. This analysis aims to reveal the behavioral impact of distinct lesions to the left inferior frontal gyrus (LIFG) and the potential for rsfMRI to provide evidence of cortical reorganization. METHODS/STUDY POPULATION: The analyses will be conducted on a small sample of approximately six participants with post-stroke language impairment (i.e., aphasia) recruited as part of an ongoing study. Lesion load in the anterior LIFG (aLIFG), consisting of the orbital (BA 47) and triangular (BA 45) parts, and the posterior LIFG (pLIFG), consisting of the opercular part (BA 44) will be correlated with performance on tasks of semantic and phonological processing. Functional connectivity will be determined between regions of interest (i.e., anterior superior temporal, posterior middle temporal, angular, and supramarginal gyri) in the left and/or right hemispheres based on rsfMRI data. RESULTS/ANTICIPATED RESULTS: Lesion load in the aLIFG is expected to correlate to performance on the semantic task, whereas lesion load in the pLIFG is expected to correlate to performance on the phonological task. For participants whose behavioral performance exceeds expectations based on lesion load (e.g., larger aLIFG lesion load, yet better semantic task performance), rsfMRI connectivity is expected to be greater (e.g., between the posterior middle temporal and angular gyri), suggesting cortical reorganization. DISCUSSION/SIGNIFICANCE: If our hypotheses are supported, our results will corroborate evidence of a functional spectrum (i.e., semantic-phonological) in the LIFG. Future work will compare rsfMRI connectivity to neurotypical controls and sites of connectivity may be used as targets in studies of non-invasive brain stimulation as an adjuvant to behavioral therapy.

Neuro-cognitive development of semantic and syntactic bootstrapping in 6- to 7.5-year-old children

The present study examined the longitudinal relations of brain and behavior from ages 6–7.5 years old to test the bootstrapping account of language development. Prior work suggests that children's vocabulary development is foundational for acquiring grammar (e.g., semantic bootstrapping) and that children rely on the syntactic context of sentences to learn the meaning of new words (e.g., syntactic bootstrapping). Yet, little is known about the dynamics underlying semantic and syntactic development as children enter elementary school. In a series of preregistered and exploratory analyses, we tested how semantic and syntactic behavioral skills may influence the development of brain regions implicated in these processes, i.e. left posterior middle temporal gyrus (pMTG) and inferior frontal gyrus (pars opercularis, IFGop), respectively. Vice-a-versa, we tested how these brain regions may influence the development of children's semantic and syntactic behavioral skills. We assessed semantic (N = 26) and syntactic (N = 30) processes behaviorally and in the brain when children were ages 5.5–6.5 years old (Time 1) and again at 7–8 years old (Time 2). All brain-behavior analyses controlled for T1 autoregressive effects and phonological memory. Exploratory hierarchical regression analyses suggested bi-directional influences, but with greater support for syntactic bootstrapping. Across the analyses, there was a small to medium effect of change in variance in models where semantics predicted syntax. Conversely, there was medium to large change in variance in models where syntax predicted semantics. In line with prior literature, results suggest a close relationship between lexical and grammatical development in children ages 6–7.5 years old. However, there was more robust evidence for syntactic bootstrapping, suggesting that acquisition of phrase structure in school age children may allow for more effective learning of word meanings. This complements prior behavioral studies and suggests a potential shift in the early reliance on semantics to later reliance on syntax in development.

Both frontal and temporal cortex exhibit phonological and semantic specialization during spoken language processing in 7- to 8-year-old children.

A previous functional magnetic resonance imaging (fMRI) study by Weiss et al. (Weiss et al., Human Brain Mapping, 2018, 39, 4334–4348) examined brain specialization for phonological and semantic processing of spoken words in young children who were 5 to 6 years old and found evidence for specialization in the temporal but not the frontal lobe. According to a prominent neurocognitive model of language development (Skeide & Friederici, Nature Reviews Neuroscience, 2016, 17, 323–332), the frontal lobe matures later than the temporal lobe. Thus, the current study aimed to examine if brain specialization in the frontal lobe can be observed in a slightly older cohort of children aged 7 to 8 years old using the same experimental and analytical approach as in Weiss et al. (Weiss et al., Human Brain Mapping, 2018, 39, 4334–4348). One hundred and ten typically developing children were recruited and were asked to perform a sound judgment task, tapping into phonological processing, and a meaning judgment task, tapping into semantic processing, while in the MRI scanner. Direct task comparisons showed that these children exhibited language specialization in both the temporal and the frontal lobes, with the left posterior dorsal inferior frontal gyrus (IFG) showing greater activation for the sound than the meaning judgment task, and the left anterior ventral IFG and the left posterior middle temporal gyrus (MTG) showing greater activation for the meaning than the sound judgment task. These findings demonstrate that 7‐ to 8‐year‐old children have already begun to develop a language‐related specialization in the frontal lobe, suggesting that early elementary schoolers rely on both specialized linguistic manipulation and representation mechanisms to perform language tasks.

Online Effects of Beta-tACS Over the Left Prefrontal Cortex on Phonological Decisions

The left posterior inferior frontal gyrus in the prefrontal cortex is a key region for phonological aspects of language processing. A previous study has shown that alpha-tACS over the prefrontal cortex applied before task processing facilitated phonological decision-making and increased task-related theta power. However, it is unclear how alpha-tACS affects phonological processing when applied directly during the task. Moreover, the frequency specificity of this effect is also unclear since the majority of neurostimulation studies tested a single frequency only. The present study addressed the question whether and how 10 Hz online tACS affects phonological decisions. To this end, 24 healthy participants received tACS at 10 Hz or 16.18 Hz (control frequency) or sham stimulation over the left prefrontal cortex during task processing in three sessions. As an unexpected finding, 16.18 Hz significantly impaired task accuracy relative to sham stimulation, without affecting response speed. There was no significant difference in phonological task performance between 10 Hz and 16.18 Hz tACS or between 10 Hz and sham stimulation. Our results support the functional relevance of the left prefrontal cortex for phonological decisions and suggest that online beta-tACS may modulate language comprehension.

Enhanced functional connectivity between insular subregions correlates with the efficacy of music and instruction-guided relaxation in depression.

Music and instruction-guided relaxation (MIGR) is a complementary therapeutic tool used in the treatment of the major depressive disorder (MDD). However, the neural mechanism that underlies the effect of MIGR on MDD patients is not known. Twenty-three right-handed MDD patients and 23 age-, sex-, handedness-, and educational level-matched healthy controls were enrolled. Resting-state functional MRI data were acquired from patients before and after MIGR and from healthy controls. The relationships between insular subregion-based functional connectivity and Hamilton Depression Rating Scale, Hamilton Anxiety Rating Scale (HAM-A), Automatic Thoughts Questionnaire, and Ruminative Responses Scale scores were examined. One-way analysis of variance exhibited significant differences among the three groups in functional connectivity between the left dorsal anterior insula (dAI) and left superior medial frontal gyrus (SMFG), left dAI and left precuneus, left posterior insula and left gyrus rectus, right ventral anterior insula (vAI) and left posterior cingulate cortex (PCC), right vAI and right inferior frontal gyrus (R-IFG). Further comparisons in regions of interest showed that MDD patients before MIGR showed decreased functional connectivity between the left dAI and left SMFG, left dAI and left precuneus, left posterior insula, and left gyrus rectus, right vAI and left PCC, right vAI and R-IFG relative to those in healthy controls. The strength of functional connectivity between the right dAI and left putamen also exhibited a negative correlation with the HAM-A score in MDD cases before MIGR. MIGR may result in enhanced functional connectivity in insular subregions, thereby potentially increasing the regulatory influence of cognitive reappraisal.

The phonological loop: is speech special?

It has been proposed that the maintenance of phonological information in verbal working memory (vWM) is carried by a domain-specific short-term storage center-the phonological loop-which is composed of a phonological store and an articulatory rehearsal system. Several brain regions including the left posterior inferior frontal gyrus (pIFG) and anterior supramarginal gyri (aSMG) are thought to support these processes. However, recent behavioral evidence suggests that verbal and non-verbal auditory information may be processed as part of a unique domain general short-term storage center instead of through specialized subsystems such as the phonological loop. In the current study, we used a single-pulse transcranial magnetic stimulation (TMS)-delayed priming paradigm with speech (syllables) and acoustically complex non-speech sounds (bird songs) to examine whether the pIFG and aSMG are involved in the processing of verbal information or, alternatively, in the processing of any complex auditory information. Our results demonstrate that TMS delivered to both regions had an effect on performance for speech and non-speech stimuli, but the nature of the effect was different. That is, priming was reduced for the speech sounds because TMS facilitated the detection of different but not identical stimuli, and accuracy was decreased for non-speech sounds. Since TMS interfered with both speech and non-speech sounds, these findings support the existence of an auditory short-term storage center located within the dorsal auditory stream.

Syntax-Sensitive Regions of the Posterior Inferior Frontal Gyrus and the Posterior Temporal Lobe Are Differentially Recruited by Production and Perception.

Matchin and Hickok (2020) proposed that the left posterior inferior frontal gyrus (PIFG) and the left posterior temporal lobe (PTL) both play a role in syntactic processing, broadly construed, attributing distinct functions to these regions with respect to production and perception. Consistent with this hypothesis, functional dissociations between these regions have been demonstrated with respect to lesion–symptom mapping in aphasia. However, neuroimaging studies of syntactic comprehension typically show similar activations in these regions. In order to identify whether these regions show distinct activation patterns with respect to syntactic perception and production, we performed an fMRI study contrasting the subvocal articulation and perception of structured jabberwocky phrases (syntactic), sequences of real words (lexical), and sequences of pseudowords (phonological). We defined two sets of language-selective regions of interest (ROIs) in individual subjects for the PIFG and the PTL using the contrasts [syntactic > lexical] and [syntactic > phonological]. We found robust significant interactions of comprehension and production between these 2 regions at the syntactic level, for both sets of language-selective ROIs. This suggests a core difference in the function of these regions with respect to production and perception, consistent with the lesion literature.

The present study addresses whether adults with Broca’s aphasia who have damage in their Posterior Left Inferior Frontal Gyrus (PLIFG) show similar performance to typically developing children in their processing of complex syntax. We tested comprehension of Turkish relative clauses using a sentence-picture matching task in Broca’s patients and children with typical development (aged: 3;04-4;03). Both groups showed better performance in subject relative clauses compared to object relative clauses. Children's similar performance to Broca's patients might be due to the fact that PLIFG is a late-maturing neural region. Our participants had more errors in object relative clauses despite the first referent was the agent so there was no evidence for the agent-first strategy. Poor performance in object relative clauses was linked to the morphosyntactic complexity in these structures.

Musicians use speech-specific areas when processing tones: The key to their superior linguistic competence?

It is known that musicians compared to non-musicians have some superior speech and language competence, yet the mechanisms how musical training leads to this advantage are not well specified.This event-related fMRI study confirmed that musicians outperformed non-musicians in processing not only of musical tones but also syllables and identified a network differentiating musicians from non-musicians during processing of linguistic sounds.Within this network, the activation of bilateral superior temporal gyrus was shared with all subjects during processing of the acoustically well-matched musical and linguistic sounds, and with the activation distinguishing tones with a complex harmonic spectrum (bowed tone) from a simpler one (plucked tone). These results confirm that better speech processing in musicians relies on improved cross-domain spectral analysis.Activation of left posterior superior temporal sulcus (pSTS), premotor cortex, inferior frontal and fusiform gyrus (FG) also distinguishing musicians from non-musicians during syllable processing overlapped with the activation segregating linguistic from musical sounds in all subjects. Since these brain-regions were not involved during tone processing in non-musicians, they could code for functions which are specialized for speech. Musicians recruited pSTS and FG during tone processing, thus these speech-specialized brain-areas processed musical sounds in the presence of musical training.This study shows that the linguistic advantage of musicians is linked not only to improved cross-domain spectral analysis, but also to the functional adaptation of brain resources that are specialized for speech, but accessible to the domain of music in the presence of musical training.

Anterior and Posterior Left Inferior Frontal Gyrus Contribute to the Implementation of Grammatical Determiners During Language Production

The left inferior frontal gyrus (IFG) is a key region for language comprehension and production. Previous studies point to a preferential involvement of left anterior IFG (aIFG) in lexical and semantic processes, while the posterior IFG (pIFG) has been implicated in supporting syntactic and phonological processes. Here we used focal neuronavigated transcranial magnetic stimulation (TMS) to probe the functional involvement of left IFG in lexical and grammatical processing at the sentence level. We applied 10 Hz TMS effective or sham bursts to left aIFG and pIFG, while healthy volunteers performed an adjective-noun production task contrasting grammatical and lexical determiners. For each trial, we measured the time from the stimulus onset to the moment of articulation (response time) and the time from articulation onset to the end of articulation (duration). Focal TMS of IFG generally delayed response times. The TMS-induced delay in response times was relatively stronger for the grammatical condition compared to the lexical condition, when TMS targeted aIFG. Articulation of the determiner was generally shorter in trials presenting grammatical determiners relative to lexical determiners. The shorter articulation time for grammar determiners was facilitated by effective TMS to pIFG. Together, the effects of TMS on task performance provide novel evidence for a joint involvement of anterior and posterior parts of left IFG in implementing grammatical determiners during language production, suggesting an involvement of aIFG in the initiation and pIFG in the production of grammatically appropriate verbal responses at the sentence level.

Stronger right hemisphere functional connectivity supports executive aspects of language in older adults

Healthy older adults commonly report increased difficulties with language production. This could reflect decline in the language network, or age-related declines in other cognitive abilities that support language production, such as executive function. To examine this possibility, we conducted a whole-brain resting-state functional connectivity (RSFC) analysis in older and younger adults using two seed regions—the left posterior superior temporal gyrus and left inferior frontal gyrus. Whole-brain connectivities were then correlated with Stroop task performance to investigate the relationship between RSFC and executive function. We found that overall, younger adults had stronger RSFC than older adults. Moreover, in older, but not younger, adults stronger RSFC between left IFG and right hemisphere executive function regions correlated with better Stroop performance. This suggests that stronger RSFC among older adults between left IFG and right hemisphere regions may serve a compensatory function.

Short-term modulation of the lesioned language network.

Language is sustained by large-scale networks in the human brain. Stroke often severely affects function and network dynamics. However, the adaptive potential of the brain to compensate for lesions is poorly understood. A key question is whether upregulation of the right hemisphere is adaptive for language recovery. Targeting the potential for short-term reorganization in the lesioned brain, we applied 'virtual lesions' over left anterior or posterior inferior frontal gyrus (IFG) in post-stroke patients with left temporo-parietal lesions prior to functional neuroimaging. Perturbation of the posterior IFG selectively delayed phonological decisions and decreased phonological activity. The individual response delay was correlated with the upregulation of the lesion homologue, likely reflecting compensation. Moreover, stronger individual tract integrity of the right superior longitudinal fascicle was associated with lesser disruption. Our results provide evidence for functional and structural underpinnings of plasticity in the lesioned language network, and a compensatory role of the right hemisphere.

Functional Dissociations of the Left Anterior and Posterior Occipitotemporal Cortex for Semantic and Non-semantic Phonological Access

Previous studies have identified the ventral and dorsal brain regions that respectively support semantic and non-semantic phonological access. Nevertheless, the specific role of the left occipitotemporal cortex (lOTC) in the two pathways of phonological access is ambiguous. To address that question, the present study compared word reading in Chinese (presumably relying on the semantic pathway) with that in English (presumably relying on the non-semantic pathway). Results revealed a clear dissociation in the involvement of the anterior and posterior lOTC in semantic and non-semantic phonological access. Specifically, the anterior lOTC showed greater activation for Chinese than for English, whereas the posterior lOTC showed greater activation for English than for Chinese. More importantly, both psychophysiological interaction analysis and resting-state functional connectivity analysis showed that the anterior lOTC was functionally connected to the ventral brain regions (e.g., left anterior fusiform gyrus, anterior temporal lobe, and ventral inferior frontal gyrus), whereas the posterior lOTC was functionally connected to the dorsal brain regions (e.g., left posterior superior temporal gyrus, supramarginal gyrus, and dorsal inferior frontal gyrus). These results suggest that the anterior and posterior lOTC are involved in semantic and non-semantic phonological access, respectively.

After-effects of 10 Hz tACS over the prefrontal cortex on phonological word decisions

IntroductionPrevious work in the language domain has shown that 10 Hz rTMS of the left or right posterior inferior frontal gyrus (pIFG) in the prefrontal cortex impaired phonological decision-making, arguing for a causal contribution of the bilateral pIFG to phonological processing. However, the neurophysiological correlates of these effects are unclear. The present study addressed the question whether neural activity in the prefrontal cortex could be modulated by 10 Hz tACS and how this would affect phonological decisions. MethodsIn three sessions, 24 healthy participants received tACS at 10 Hz or 16.18 Hz (control frequency) or sham stimulation over the bilateral prefrontal cortex before task processing. Resting state EEG was recorded before and after tACS. We also recorded EEG during task processing. ResultsRelative to sham stimulation, 10 Hz tACS significantly facilitated phonological response speed. This effect was task-specific as tACS did not affect a simple control task. Moreover, 10 Hz tACS significantly increased theta power during phonological decisions. The individual increase in theta power was positively correlated with the behavioral facilitation after 10 Hz tACS. ConclusionOur results show a facilitation of phonological decisions after 10 Hz tACS over the bilateral prefrontal cortex. This might indicate that 10 Hz tACS increased task-related activity in the stimulated area to a level that was optimal for phonological performance. The significant correlation with the individual increase in theta power suggests that the behavioral facilitation might be related to increased theta power during language processing.
