Ultra-stable MOF@MOF nanoplatform for photodynamic therapy sensitized by relieved hypoxia due to mitochondrial respiration inhibition

Metal-organic frameworks (MOFs) with periodically arranged porphyrinic linkers avoiding the self-quenching issue of porphyrins in photodynamic therapy (PDT) have been widely applied. However, the porphyrinic MOFs still face challenges of poor stability under physiological conditions and limited photodynamic efficiency by the hypoxia condition of tumors. Herein, we fabricate the MOF@MOF structure with a protective MOF shell to improve the stability and relieve the hypoxia condition of tumors for sensitized PDT. Under protection of the MOF shell, the MOF@MOF structure can keep intact for 96 h under physiological conditions. Consequently, the tumoral accumulation efficiency is two folds of the MOF core. Furthermore, the MOF shell decomposes under acidic environment, and the loaded inhibitor of mitochondria pyruvate carrier (7-amino carboxycoumarins-2, 7ACC2) will be released. 7ACC2 inhibits the mitochondrial pyruvate influx and simultaneously blocks glucose and lactate from fueling the mitochondrial respiration, thereupon relieving the hypoxia condition of tumors. Under a 5-min laser irradiation, the 7ACC2 carrying MOF@MOF nanoplatforms induced doubled cellular apoptosis and reduced 70% of the tumor growth compared with the cargo-free MOF@MOF. In summary, the design of this stable and hypoxia self-relievable MOF@MOF nanoplatform will enlighten the future development of MOF-based nanomedicines and PDT. Statement of significanceThough widely used for photodynamic therapy (PDT) in previous studies, porphyrinic metal-organic frameworks (MOFs) still face challenges in poor stability under physiological conditions and limited photodynamic efficiency due to the hypoxia condition of tumors. In order to solve these problems, (1) we develop the MOF@MOF strategy to improve the physiological stability; (2) an inhibitor of mitochondria pyruvate carrier, 7-amino carboxycoumarins-2 (7ACC2), is loaded to inhibit the mitochondrial pyruvate influx and simultaneously block glucose and lactate from fueling the mitochondrial respiration, thereupon relieving the hypoxia condition of tumors. In comparison with previous studies, our strategy simultaneously improves stability and overcomes the limited PDT efficiency in the hypoxia tumor tissue, which will enlighten the future development of MOF-based nanomedicines and PDT.

The biological activities of 5,15-diaryl-10,20-dihalogeno porphyrins for photodynamic therapy.

Esophageal cancer is the most common gastrointestinal tumor and is difficult to be eradicated with conventional treatment. Porphyrin-based photosensitizers (PSs) mediated photodynamic therapy (PDT) could kill tumor cells with less damage to normal cells. As the most widely used porphyrin-based photosensitizer in clinics, Photofrin II has excellent anti-tumor effect. However, it has some disadvantages such as weak absorption at near infrared region, the complexity of components and prolonged skin photosensitivity. Here series novel 5,15-diaryl-10,20-dihalogeno porphyrin derivatives were afforded and evaluated to develop more effective and safer photosensitizers for tumor therapy. The photophysical properties and singlet oxygen generation rates of 5,15-diaryl-10,20-dihalogeno porphyrins (I1-6, II1-4) were tested. The cytotoxicity of I1-6 and II1-4 were measured by MTT assay. The pathway of cell death was studied by flow cytometry. In vivo photodynamic efficacy of I3 and II2-4 in Eca-109 tumor-bearing BABL/c nude mice were measured and histopathological analysis were examined. 5,15-Diaryl-10,20-dihalogeno porphyrins I1-6 and II1-4 were synthesized. The longest absorption wavelength of these halogenated porphyrins (λmax = 660nm) displayed a red shift around 30nm compared to the unhalogenated porphyrins PS1 (λmax = 630nm). The singlet oxygen generation rates of I1-6 and II1-4 were significantly higher than PS1 and HMME. All PSs mediated PDT showed obvious cytotoxic effect against Eca-109 cells compared to HMME in vitro and in vivo. Among these PSs, II4 exhibited appropriate absorption in the phototherapeutic window, higher 1O2 generation rate (k = 0.0061s-1), the strongest phototoxicity (IC50 = 0.4μM), lower dark toxicity, high generation of intracellular ROS in Eca-109 cells and excellent photodynamic anti-tumor efficacy in vivo. Besides, cell necrosis was induced by compound II4 mediated PDT. All new compounds have obvious photodynamic anti-esophageal cancer effects. Among them, the photosensitizer II4 showed excellent efficacy in vitro and in vivo, which has the potential to become a photodynamic anti-tumor drug.

One-Step Aqueous Synthesis of Anionic and Cationic AgInS2 Quantum Dots and Their Utility in Improving the Efficacy of ALA-Based Photodynamic Therapy.

Silver–indium–sulfide quantum dots (AIS QDs) have potential applications in many areas, including biomedicine. Their lack of regulated heavy metals, unlike many commercialized QDs, stands out as an advantage, but the necessity for alloyed or core–shell structures and related costly and sophisticated processes for the production of stable and high quantum yield aqueous AIS QDs are the current challenges. The present study demonstrates the one-step aqueous synthesis of simple AgInS2 QD compositions utilizing for the first time either a polyethyleneimine/2-mercaptopropionic acid (AIS-PEI/2MPA) mixture or only 2-mercaptopropionic acid (AIS-2MPA) as the stabilizing molecules, providing a AgInS2 portfolio consisting of cationic and anionic AIS QDs, respectively, and tuneable emission. Small AIS QDs with long-term stability and high quantum yields (19–23%) were achieved at a molar ratio of Ag/In/S 1/10/10 in water without any dopant or a semiconductor shell. The theranostic potential of these cationic and anionic AIS QDs was also evaluated in vitro. Non-toxic doses were determined, and fluorescence imaging potential was demonstrated. More importantly, these QDs were electrostatically loaded with zwitterionic 5-aminolevulinic acid (ALA) as a prodrug to enhance the tumor availability of ALA and to improve ALA-induced porphyrin photodynamic therapy (PDT). This is the first study investigating the influence of nanoparticle charge on ALA binding, release, and therapeutic efficacy. Surface charge was found to be more critical in cellular internalization and dark toxicity rather than drug loading and release. Both QDs provided enhanced ALA release at acidic pH but protected the prodrug at physiological pH, which is critical for tumor delivery of ALA, which suffers from low bioavailability. The PDT efficacy of the ALA-loaded AIS QDs was tested in 2D monolayers and 3D constructs of HT29 and SW480 human colon adenocarcinoma cancer cell lines. The incorporation of ALA delivery by the AIS QDs, which on their own do not cause phototoxicity, elicited significant cell death due to enhanced light-induced ROS generation and apoptotic/necrotic cell death, reducing the IC50 for ALA dramatically to about 0.1 and 0.01 mM in anionic and cationic AIS QDs, respectively. Combined with simple synthetic methods, the strong intracellular photoluminescence of AIS QDs, good biocompatibility of especially the anionic AIS QDs, and the ability to act as drug carriers for effective PDT signify that the AIS QDs, in particular AIS-2MPA, are highly promising theranostic QDs.

Zinc(II), copper(II) and nickel(II) ions improve the selectivity of tetra-cationic platinum(II) porphyrins in photodynamic therapy and stimulate antioxidant defenses in the metastatic melanoma lineage (A375).

Tetra-cationic porphyrins with peripheral Pt (II) -bipyridyl complexes demonstrated a potential as photosensitizers to be used in photodynamic therapy (PDT). First-line transition metals, such as zinc (II), copper (II) and nickel (II), can be incorporated into the porphyrin nucleus, making this molecule more selective and more effective for this therapy in combating to tumor cells, such as metastatic melanoma. We characterized these derivatives to verify the improvement in selectivity of platinum (II) 4-PtTPyP porphyrins. Receptors such as LDL and endothelin (ERT-B) were investigated, as well as the binding affinity of two antioxidants: catalase model enzymes and superoxide dismutase. Human serum albumin (SAH) HSA binding properties have been verified. In addition, we evaluated the antitumor action of such metalloporphyrins in an in vitro cell viability. Our results demonstrated that porphyrins have significant antitumor potential when exposed to white light conditions. The affinity for the LDL receptor was better when compared to platinum porphyrin 4-PtTPyP without addition of metals and the affinity for the endothelin receptor was higher than the control used in this study. Still, the interaction with the HSA showed the possibility of this connection taking photosensitizers to places of interest, such as the delivery of medicines.

Synthesis and characterization of novel porphyrin-cinnamic acid conjugates.

A series of novel porphyrin-cinnamic acid (porphyrin/CA) conjugates (4a-4c) have been synthesized by condensation of 5-(4-Hydroxyphenyl)-10,15,20-triphenyl- porphyrin (1) with different substituted cinnamic acids (CAs) through an alkyl linker due to the biological activities of CAs and the application of porphyrins in photodynamic therapy (PDT) of cancer. Their related zinc (II) complexes (5a-5c) were also prepared. These novel compounds have been fully characterized by 1H NMR, Infrared (IR), Mass spectra (Ms) and Elemental analysis. The photophysical properties of these target molecules were studied by absorption and Fluorescence spectroscopy. In solution, the effect of pH, ionic strength in acid media and concentration on the aggregation behaviors of 4a has also been investigated by UV-Vis spectra. The broadened and red shifted Soret band indicated the formation of J-aggregates when the pH value was up to 2.0 in THF-aqueous solution. Furthermore, the higher ionic strength of 0.3 M NaCl in acid media resulted in the generation of J-aggregates in THF-aqueous solution. And the significant blue shift of Soret band also demonstrated the formation of H-aggregates of 4a at 1.1 × 10-4 M in THF.

Development of Gd(III) porphyrin-conjugated chitosan nanoparticles as contrast agents for magnetic resonance imaging

A novel magnetic resonance imaging (MRI) contrast agent based on gadolinium meso-tetrakis(4-pyridyl)porphyrin [Gd(TPyP)] conjugated with chitosan nanoparticles has been developed. The chitosan nanoparticles were synthesized following an ionic gelation method and the conditions optimized to generate small nanoparticles (CNs) with a narrow size distribution of 35-65 nm. The gadolinium meso-tetrakis(4-pyridyl)porphyrin [Gd(TPyP)] was loaded into chitosan nanoparticles by passive adsorption. The interaction of chitosan with Gd(TPyP) has been examined by UV-visible, Fourier transform infrared spectroscopies (FT-IR) and inductively coupled plasma mass spectrometry (ICP-MS), which indicate the successful association of Gd(TPyP) without any structural distortion throughout the chitosan nanoparticles. The potential of Gd(TPyP)-CNs as MRI contrast agent has been investigated by magnetic resonance imaging (MRI) in-vitro. Relaxivities of Gd(TPyP)-CNs obtained from T1-weighted images, increased with Gd concentration and attained an optimum r1 of 38.35 mM(-1) s(-1), which is 12-fold higher compared to commercial Gd-DOTA (~4 mM(-1) s(-1) at 3T). The combination of such strong MRI contrast with the known properties of porphyrins in photodynamic therapy and biocompatibility of chitosan, presents a new perspective in using these compounds in cancer theranostics.

Rational design of shear-thinning supramolecular hydrogels with porphyrin for controlled chemotherapeutics release and photodynamic therapy

An amphiphilic porphyrin-cored, star-shaped poly (ε-caprolactone)-b-poly (ethylene glycol) (SPPCL-b-PEG) copolymer was synthesized and used as a building block for constructing supramolecular hydrogels. With the aid of α-cyclodextrin (α-CD), thixotropic and reversible supramolecular hydrogel could be formed by copolymers based on the host–guest inclusion complexation (IC). Due to the biodegradable and biocompatible properties, this IC-based supramolecular hydrogel was used as carrier for doxorubicin (DOX) delivery systems. The release results presented a triphasic drug-release profile and the release of this system lasted more than 3weeks. Also, upon irradiation with light (650nm), cytotoxic singlet oxygen (1O2) could be generated by porphyrin core and easily released from hydrogel matrix. Considering that both sustained-release drug delivery of hydrogels and porphyrin for photodynamic therapy (PDT) were achieved in a combined compound, this shear-thinning supramolecular hydrogel with porphyrin was elaborately designed as a promising replacement therapy to medium or advanced cancer for surgery.

Herein, we report the synthesis of such novel platinum–porphyrin conjugates as well as their photophysical characterization and in vitro light‐induced anticancer properties. These conjugates showed only minor cytotoxicity in the dark, but IC50 values down to 19 nM upon irradiation with light at 420 nm.These values correspond to an excellent phototoxic index (PI=IC50 in the dark/IC50 in light), which reached 5000 in a cisplatin‐resistant cell line. After incubation with HeLa cells, nuclear Pt concentrations were 30 times higher than with cisplatin. All of these favorable characteristics imply that tetraplatinated porphyrin complexes are worthy of exploration as novel PDT anticancer agents in vivo.

Fast elimination of onychomycosis by hematoporphyrin derivative-photodynamic therapy

Onychomycosis is a fungal nail disease and is one of the major onychopathy worldwide. Topical or oral antifungal therapies are used to treat this disease, but often they are inefficient and oral medications can even cause several side effects. Photodynamic therapy (PDT) is a well established technique and hence, may represent an alternative non invasive technique for the treatment of onychomycosis. In this work, we present a case of onychomycosis that was completely cured by using the porphyrin-photodynamic therapy. A 59-year-old patient, who had two nails with onychomycosis (the right and the left hallux, with more than thirty and ten years, respectively) caused by fungi was treated once a week for a period of six weeks. The nails were first treated and prepared by a specialist. An hour after the photosensitization, the nail was illuminated using a light source based on light emitting diodes (LEDs) in the red wavelength (630 nm, at a total dose of 54 J/cm(2)).

Synthesis, Photophysical Studies and Anticancer Activity of a New Halogenated Water‐Soluble Porphyrin

A water-soluble halogenated porphyrin, namely 5,10,15,20-tetrakis(2-chloro-3-sulfophenyl)porphyrin (TCPPSO(3)H), was prepared and evaluated as sensitizer for photodynamic therapy (PDT). Photophysical properties of TCPPSO(3)H, such as high photostability, long triplet lifetime and high singlet oxygen quantum yield suggest high effectiveness of this class of halogenated porphyrins in PDT. TCPPSO(3)H is non-toxic in the dark and causes a significant photodynamic effect examined against MCF7 (human breast carcinoma), SKMEL 188 (human melanoma) and S91(mouse melanoma) cell lines upon red light irradiation (cutoff < 600 nm) at low light doses. Time-dependent cellular uptake of TCPPSO(3)H reached plateau at 120 min and was the highest for S91, 20% lower for MCF7 and 70% lower for SKMEL 188. Our results show that this halogenated water-soluble porphyrin is an efficient photosensitizer and reveal the potential of this class of compounds as PDT agents.

Thermodynamic analysis of porphyrin binding to Momordica charantia (bitter gourd) lectin.

Owing to the use of porphyrins in photodynamic therapy for the treatment of malignant tumors, and the preferential interaction of lectins with tumor cells, studies on lectin-porphyrin interaction are of significant interest. In this study, the interaction of several free-base and metalloporphyrins with Momordica charantia (bitter gourd) lectin (MCL) was investigated by absorption spectroscopy. Difference absorption spectra revealed that significant changes occur in the Soret band region of the porphyrins on binding to MCL. These changes were monitored to obtain association constants (Ka) and stoichiometry of binding. The tetrameric MCL binds four porphyrin molecules, and the stoichiometry was unaffected by the presence of the specific sugar, lactose. In addition, the agglutination activity of MCL was unaffected by the presence of the porphyrins used in this study, clearly indicating that porphyrin and carbohydrate ligands bind at different sites. Both cationic and anionic porphyrins bind to the lectin with comparable affinity (Ka =10(3)-10(5) m(-1)). The thermodynamic parameters associated with the interaction of several porphyrins, obtained from the temperature dependence of the Ka values, were found to be in the range: DeltaH degrees = -98.1 to -54.4 kJ.mol(-1) and DeltaS degrees =-243.9 to -90.8 J.mol(-1).K(-1). These results indicate that porphyrin binding to MCL is governed by enthalpic forces and that the contribution from binding entropy is negative. Enthalpy-entropy compensation was observed in the interaction of different porphyrins with MCL, underscoring the role of water structure in the overall binding process. Analysis of CD spectra of MCL indicates that this protein contains about 13%alpha-helix, 36%beta-sheet, 21%beta-turn, and the rest unordered structures. Binding of porphyrins does not significantly alter the secondary and tertiary structures of MCL.

Amphiphilic porphyrins in reverse micelles: the influence of the molar ratio of water to surfactant and side-chain length on their triplet-state lifetimes. Using bis(2-ethylhexyl) sodium sulfosuccinate (AOT) as surfactant, a series of amphiphilic porphyrins terminated with imidazole were studied in AOT/isooctane/water reverse micelles, intending to mimic the relationship between microenvironments in organisms and the amphiphilic properties of porphyrins for photodynamic therapy (PDT) applications. Laser flash photolysis experiments and fluorescence quenching of porphyrins by phenothiazine show that longer side-chain lengths and higher molar ratio of water to AOT (wo) of reverse micelles promotes firm embedding of porphyrin molecules in the interfacial region of reverse micelles. Such an embedding may shift porphyrins from aggregated to monomeric form in reverse micelles and, accordingly, the slower excited-state decay may increase the efficiency of the photosensitizer in PDT. The variation in the ability of singlet oxygen generation detected by ESR also supports such a conclusion. The effect of amphiphilic properties on the status of porphyrins in microenvironments provides light on the synthesis of other amphiphilic porphyrins for PDT applications.

Porphyrins in photodynamic therapy - a search for ideal photosensitizers.

The utility of light as a therapeutic agent can be traced back over thousands of years when it was used in Ancient Egypt, India and China to treat a variety of skin diseases like psoriasis, vitiligo, rickets, cancer and psychosis. The isolation of porphyrins and their inherent tumor localizing properties coupled with its ability to generate reactive singlet oxygen when activated by light of particular wavelength which in turn results in cytotoxicity led to the emergence of a new modality namely, photodynamic therapy (PDT) as a therapeutic tool. The higher degree of selectivity offered by this modality and fewer side effects when compared to chemotherapy and radiotherapy has prompted the researchers around the globe to generate new photosensitizers. Porphyrins and expanded porphyrins are one class of molecules under intense investigation due to their photosensitizing ability for PDT application. Expanded porphyrins result from the expansion of the phi electron conjugation by increasing the number of heterocyclic rings or bridging carbons of the existing porphyrin framework. These chromophores show strong absorptions in the red region (650-800 nm) compared to that of normal 18phi porphyrins. The strong absorption of light by a water soluble nontoxic photosensitizing molecule in the therapeutic window resulting in maximum penetration of light into the tissues coupled with high singlet oxygen production will conceptualize an ideal photosensitizer. This review highlights various porphyrinoid sensitizers reported till date and their photosensitizing ability both in vitro and in vivo studies. Furthermore, the urgent need for developing ideal photosensitizer for PDT will also be highlighted.

The impact of stability and solubility of photodynamic therapies on method development

A well characterised and fully validated bioanalytical method used for measuring a novel porphyrin photodynamic therapy (CompoundI) in human plasma was modified in order to enable the measurement of a structurally and metabolically related compound (CompoundII) in rat plasma.

On the mitochondrial aspect of reactive oxygen species action in external magnetic fields

Some retinoids or porphyrins can form radical pairs, alter transfer of electrons, and increase synthesis of reactive oxygen species in the mitochondrial respiratory chain. This leads to cell damage and apoptosis. We propose that co-application of an external static magnetic field of several to several hundreds miliTesla (mT) can enhance those effects by further alteration of electron flow in the mitochondrial respiratory chain, facilitation of forbidden transitions from the triplet to the singlet state of retinoid or porphyrin radicals, and modification of radical pair amount. Since electron flow in the mitochondrial respiratory chain seems to possess fractal dynamics, the magnetic field can initiate self- organization of the flow into more regular patterns, and create optimal conditions for damage of cancer cells without any detriment for the normal counterparts. External low magnetic field should improve effectiveness and selectivity of retinoid chemoprevention or porphyrin photodynamic therapy.

Photodynamic Tumor Therapy: Mitochondrial Benzodiazepine Receptors as a Therapeutic Target

Photodynamic therapy employs photosensitive agents such as porphyrins to treat a variety of tumors accessible to light-emitting probes. This approach capitalizes on the selective retention of porphyrins by cancer cells. Cancer cells also have elevated levels of mitochondrial benzodiazepine receptors which bind porphyrins with high affinity. Cultured cancer cell lines were exposed to porphyrin and porphyrin-like compounds and then irradiated with light. Cytotoxicity of this treatment was measured via clonogenic assays. Mitochondrial benzodiazepine receptor pharmacology was studied using [3H] PK11195 binding to cancer cell homogenates and isolated kidney mitochondrial membranes. We show that therapeutic potencies of porphyrins correlate closely with affinities for mitochondrial benzodiazepine receptors. Sensitivities of tumor cell lines to photodynamic therapy parallel their densities of these receptors. We propose that porphyrin photodynamic therapy is mediated by mitochondrial benzodiazepine receptors.
