Three-Dimensional Cardiomyocyte-Nanobiosensing System for Specific Recognition of Drug Subgroups.

Abnormal cardiac electrophysiological activities significantly contribute to the incidence of cardiovascular diseases. Therefore, it is crucial to recognize effective drugs, which require an accurate, stable, and sensitive platform. Although conventional extracellular recordings offer a non-invasive and label-free manner to monitor the electrophysiological state of cardiomyocytes, the misrepresented and low-quality extracellular action potentials are difficult to provide accurate and high-content information for drug screening. This study presents the development of a three-dimensional cardiomyocyte-nanobiosensing system that can specifically recognize drug subgroups. The nanopillar-based electrode is manufactured by template synthesis and standard microfabrication technology on a porous polyethylene terephthalate membrane. Based on the cardiomyocyte-nanopillar interface, high-quality intracellular action potentials can be recorded by the minimally invasive electroporation. We validate the performance of a cardiomyocyte-nanopillar-based intracellular electrophysiological biosensing platform by two subclasses of sodium channel blockers, quinidine and lidocaine. The recorded intracellular action potentials accurately reveal the subtle differences between these drugs. Our study indicates that high-content intracellular recordings utilizing nanopillar-based biosensing can provide a promising platform for the electrophysiological and pharmacological investigation of cardiovascular diseases.

Porous Polyethylene Terephthalate Nanotemplate Electrodes for Sensitive Intracellular Recording of Action Potentials.

New strategies for intracellular electrophysiology break the spatiotemporal limitation of the action potential and lead a notable advance in the investigation of electrically excitable cells and their network. Although successful applications of intracellular recording have been achieved by 3D micro/nanodevices, complex micro/nanofabrication processes preclude the progress of extensive applications. We address this challenge by introducing porous polyethylene terephthalate (PET) membrane to develop a new type of nanotemplate electrode. This nanotemplate electrode is manufactured following a fabrication process on a porous PET membrane by atomic layer deposition. The 3D nanotemplate electrodes afford intracellular access to cardiomyocytes to report intracellular-like action potentials. These controllable nanotemplate electrodes exhibit sensitive and prolonged intracellular recordings of action potentials compared with free-growing 3D nanoelectrodes. This study indicates that the optimized structure of the nanoelectrode significantly promotes the performance of intracellular recording to assess electrophysiology in the fields of cardiology and neuroscience at an action potential level.

Distribution of lithium in doped nuclear pores of polyethylene terephthalate by neutron depth profiling

In this paper, doping of porous polyethylene terephthalate membrane using Li-based solution is described and analyzed by nuclear analytic methods. In recent years, the polymeric porous membranes have been studied as advanced battery separators in various applications, such as portable electronic devices or electric vehicles. Apart from mechanical stability, the separators also provide electrical separation and ionic transport between the electrodes during charging and discharging processes. For application of the membranes, it is important to follow the Li distribution in the separator material over time. In this work, the distribution of Li in the porous PET foils, adepts for Li ion battery separators, was analyzed by Neutron Depth Profiling and the 3D shape of the etched pores by Ion Transmission Spectroscopy. The data obtained show that Li can be incorporated, in a passive way in the micropores in large quantities, but its amount is still limited and does not depend on time of doping. Here, the first data are presented.

Active block copolymer layer on carboxyl-functionalized PET film for hydrogen separation

To rationalize the energy requirements and environmental complications of the world, supply of pure hydrogen is the most promising as well best possible approach of such issues. Purified hydrogen gas is the necessity factor for the hydrogen-based economy. Hydrogen perm-selective membrane plays a crucial role for producing a large amount of hydrogen. Palladium is one of the best materials because of its excellent affinity to absorb hydrogen. In present work, our aim to improve selectivity as well permeability of the H2 gas compare to N2 and CO2 gases of the block copolymer coated functionalized porous PET membrane. Porous polyethylene terephthalate (PET) membranes having pore size 0.2 μm, functionalized with a carboxyl group. The supramolecular assembly was prepared from PS (35500)-b- P4VP (4400) and 2-(4- Hydroxyphenylazo) benzoic acid (HABA) in 1, 4-dioxane. Chemically synthesized palladium nanoparticles were deposited on carboxylated block copolymer (BC) coated porous PET membrane. It is an appropriate way to use H2 sensitive materials with block copolymer coated functionalized membranes to enhance the selectivity of H2. It has been found that such membranes gain better permeability and selectivity towards H2 as compared with N2 and CO2. Increment with the dipping time of these membranes in the palladium nanoparticle solution, permeability as well selectivity of H2 over N2, CO2 increases as the more attachment of Palladium nanoparticles. A fine active layer of block copolymer on the carboxyl functionalized PET membrane play a crucial role for hydrogen based gas separation. The magnitude of the permeability of such membranes for different gases shows dependency on the pore size of the upper layer (BC coated) of the membrane in addition to the molecule size of the permeating gas. Block copolymer coating of the membranes established an effective responsibility for the selectivity of H2 over CO2 gas as well over N2 gas.

Comparison of culture media indicates a role for autologous serum in enhancing phenotypic preservation of rabbit limbal stem cells in explant culture.

In this study, we aimed to compare the effects of six different cell culture media and autologous serum (AS) on the phenotypic characteristics of rabbit limbal epithelial stem cells (LESC) cultivated on porous polyethylene terephthalate (PET) membranes. Limbal explants from rabbit corneas were grown on PET membrane inserts in five different media: DMEM-F12 with fetal bovine serum (FBS) (DMEM-F12-FBS), with pluripotin (DMEM-F12-pluripotin) and with autologous serum (DMEM-F12-AS), Epilife, Keratinocyte Serum Free Medium (KSFM) and Defined-Keratinocyte Serum Free Medium. The effects of different media were evaluated by total cell yield from explants, measuring the expression of proteins by immunofluorescence and gene expression by Real Time PCR. In all five media tested, most of the limbal epithelial cells (LEC) which proliferated from explants were positive for cytokeratin (CK) 14 (85-90%), indicating that all five media support the growth of LESC from explants. The expression of differentiation markers; CK 3 and 12 was highest in DMEM-F12-FBS (56%), was lower in Epilife and KSFM (26 and 19%, respectively), with the lowest values (13%) obtained in DMEM-F12-AS. Gene expression of limbal cultures on PET membrane inserts was compared to fresh limbal tissue. In DMEM-F12-FBS, DMEM-F12-pluripotin, and DMEM-F12-AS, expression of potential LESC markers CXCR4 and polycomb complex protein BMI-1 were similar to limbal tissue. DMEM-F12 with 10% AS maintained a higher percentage of potential stem cell marker genes and lower expression of genes involved in differentiation compared to Epilife or KSFM. Our study shows that rabbit LEC can be cultivated on PET inserts using DMEM-F12 with autologous serum without a requirement for amniotic membrane or feeder cells.

Development of an oxygenation culture method for activating the liver-specific functions of HepG2 cells utilizing a collagen vitrigel membrane chamber.

We recently developed a collagen vitrigel membrane (CVM) chamber possessing a scaffold composed of high-density collagen fibrils. In this study, we first confirmed that the advantage of CVM chamber in comparison to the traditional culture chamber with porous polyethylene terephthalate membrane is to preserve a culture medium poured in its inside even though the under side is not a liquid phase but solid and gas phases. Subsequently, we designed three different culture systems to grow HepG2 cells in a culture medium (liquid phase) on the CVM which the under side is a culture medium, a plastic surface (solid phase) or 5 % CO2 in air (gas phase) and aimed to develop a brief culture method useful for activating the liver-specific functions and analyzing the pharmacokinetics of fluorescein diacetate. HepG2 cells cultured for 2 days on the liquid–solid interface and subsequently for 1 day on the liquid–gas interface represented excellent cell viability and morphology in comparison to the others, and remarkably improved albumin secretion and urea synthesis to almost the same level of freshly isolated human hepatocytes and CYP3A4 activity to about half the level of differentiated HepaRG cells. Also, the cells rapidly absorbed fluorescein diacetate, distributed it in cytosol, metabolized it into fluorescein, and speedily excreted fluorescein into both bile canaliculus-like networks and extracellular solution. These data suggest that hepatic structure and functions of monolayered HepG2 cells can be induced within a day after the oxygenation from beneath the CVM.
