Predicted essential fatty acid intakes for a group of dairy cows also apply at individual animal level

The ruminant requirements for essential fatty acids (EFAs), particularly linoleic acid (LA) and alpha-linolenic acid (ALA), have not been fully determined, although evidence suggests that an adequate supply of polyunsaturated fatty acids (FAs) could improve immunity and reproduction in transition cows. In previous studies, we predicted EFA intake for a group of cows based on animal characteristics and milk EFA secretions. However, to support precision livestock feeding, we need to match the nutrient requirements and intakes of each cow as closely as possible. Our group-level predictions may not be accurate enough to estimate the EFA intake of an individual cow, due to inter-individual variations in EFA digestion and metabolism related to differences in feed intake, intake patterns, and the composition and functioning of the rumen microbiota. To address this issue, here we set out to establish specific equations that predict EFA intake for an individual cow based on the difference (i.e. the residuals) between observed EFA intake and the predicted EFA intake based on our group-level equations. We studied a database of individual dairy cows (26 experiments; 503 datapoints from three research teams) and we predicted the residuals from (1) dietary and animal-related factors (i.e. full predictions) and (2) animal-related factors only (i.e. field predictions), which are considered more field-amenable. The variance of predicted LA and log ALA intake was explained to 68% by observed LA intake and 66% by observed log ALA intake, respectively. The residuals of LA intake were predicted by dietary ALA content, total FA intake, BW, milk yield and fat content in full predictions, and by BW, feeding level, milk yield and fat content, and sum of milk C4:0 to C14:0 FA in field predictions. The log residuals of ALA intake were predicted by dietary NDF and total FA contents, NDF intake, BW, milk protein, LA and ALA contents, and fat yield in full predictions, and by BW, DM intake, milk LA and ALA contents, and fat yield in field predictions. The field predictions showed a moderate loss of accuracy compared to full predictions based on RMSE of prediction (from 38 to 54 g/d for LA and from 0.090 to 0.12 log (g/d) for ALA). This work is the first to predict the EFA intake of an individual cow based on previously established group-level predictions of EFA intake adjusted for dietary and animal-related factors.

A review of polyunsaturated fatty acids (PUFA) of marine zoosporic microorganisms and their commercialization

Abstract Long-chain polyunsaturated fatty acids (LC-PUFA) are nutritionally and pharmacologically well-recognized beneficial compounds widely used as nutrient supplements for various applications. As the human body cannot accomplish de novo synthesis of PUFA, we depend on exogenous bioresources for the supply of these essential PUFA for day-to-day activities. The PUFA global market is expected to reach about US$ 9.7 billion by 2032. The recent drop in natural fish catch and global climate change severely hamper the global PUFA supply chain. As a sustainable, promising resource, marine zoosporic microorganisms are considered to be safe and alternative primary producers of beneficial PUFAs for direct and indirect human consumption. However, by considering the limitations associated with strain efficiency, final product yield and production cost, more viable low-cost production strategies and establishment of highly efficient, stable, high production cell lines specific for marine zoosporic microbes are essential to meet global PUFA market demand. The exploration of the latest techniques for research and development could shape the dynamics better for the global market for marine zoosporic organism-based PUFA products. Therefore, this review first summarizes low-cost production and strain improvement strategies, and later highlights the advantages and commercial prospects for marine zoosporic microbe-derived PUFA.

Diet effects on longevity, heat tolerance, lipid peroxidation and mitochondrial membrane potential in Daphnia.

The dietary supply of polyunsaturated fatty acids (PUFA) crucially affects animals' performance at different temperatures. However, the underlying physiological mechanisms are still insufficiently understood. Here, we analyzed lifespan and heat tolerance of four genotypes of Daphnia magna reared on either the green alga Scenedesmus obliquus that lacks long-chain (> C18) PUFA, or the heterokont alga Nannochloropsis limnetica that contains C20 PUFA, both either at saturating and near-starvation levels. A significant genotype-by-diet interaction in lifespan was observed at saturating diets. The C20 PUFA-rich diet eliminated differences in lifespan among genotypes on the PUFA-deficient diet. Corrected for body length, acute heat tolerance was higher at low than at high food concentration, at least in the older of the two age groups analyzed. Genotypes differed significantly in heat tolerance, but there were no genotype-by-diet interactions. As predicted, the C20 PUFA-rich diet resulted in higher lipid peroxidation (LPO) and a lower mitochondrial membrane potential (ΔΨm). LPO levels averaged across clones and rearing conditions were inversely related to acute heat tolerance. Yet, heat tolerance was higher on the PUFA-rich diet than on the PUFA-deficient diet, particularly in older Daphnia, indicating that the C20 PUFA-rich diet allowed Daphnia to compensate for higher LPO. In contrast, Daphnia with intermediate levels of ΔΨm showed the lowest heat tolerance. Neither LPO nor ΔΨm explained the diet effects on lifespan. We hypothesize that antioxidants present in the PUFA-rich diet may have enabled higher heat tolerance of Daphnia despite higher LPO, which may also explain the lifespan expansion of otherwise short-lived genotypes.

A diet rich in polyunsaturated fatty acids as supportive therapy in the treatment and prevention of psychotic disorders

IntroductionProper nutrition with fats has a protective effect on the functioning of the nervous system. However, a disturbed ratio of essential polyunsaturated fatty acids supply is nowadays a common phenomenon. Deficiency of (n-6) but Not (n-3) Polyunsaturated Fatty Acids Inhibits the Secretagogue Effect of Prolactin in Lactating Rat Mammary Epithelial Cells

The repercussions of various kinds of dietary polyunsaturated fatty acid (PUFA) deficiencies on the fatty acid composition of membranes and on the secretory activity of lactating female rat mammary epithelial cells were investigated. Primiparous female rats were fed different PUFA diets from weaning: adequate (n-6) and (n-3) PUFA supply; overall PUFA deficiency; specific (n-6) PUFA deficiency or specific (n-3) PUFA deficiency. Mammary gland phospholipids contained very low amount of (n-3) PUFA in control rats, and only 1% docosahexaenoic acid. The fatty acid composition of membrane phospholipids reflected the type of diet received by the animals, i.e., the diets deficient in the (n-3) or (n-6) PUFA series resulted in lower (n-3) or (n-6) PUFA, and the (n-3) + (n-6) deficient diet caused a true overall PUFA deficiency in the membranes. The morphology of cells from overall PUFA- or (n-6) PUFA-deficient rats showed an accumulation of secretory vesicles in the cytoplasm. Basal casein secretion was independent of the diet and of the composition of membrane phospholipids. However, prolactin did not have a secretagogue effect on cells from (n-6) PUFA- or overall PUFA-deficient rats but retained this effect on cells from (n-3)-deficient rats. These results emphasize the specific role of (n-6) PUFA in the functioning of the lactating mammary epithelial cell.

Indonesian freshwater fisheries’ oil for health and nutrition applications: a narrative review

The consumption and production of freshwater fish in Indonesia have recently increased significantly, especially in the food sector. This is for health purposes due to the dietary content, including freshwater fish protein. Fish is not only a source of protein but also a considerable supply of polyunsaturated fatty acids (PUFAs) and is essential. Using fish oils has caught a growing interest due to an increased understanding of the possible health benefits of omega-3 fatty acids in humans. This review provided thoughts on progress in the research of Indonesia’s freshwater fisheries’ oil potential. Here, fish oil's potential was addressed through its outstanding biochemical features and its potential as an antiinflammatory, cardiovascular, cancer, and autoimmune disorder agent. Generally, nutritional properties, their possible application in food production, and therapeutic and socioeconomic significance are considered. Indonesian freshwater aquaculture fisheries and inland capture fisheries have the potential for refined fish oil production. The top freshwater fisheries include aquaculture (Oreochromis niloticus, Clarias, Cyprinus carpio, Pangasius, and Osphronemus goramy) and inland capture fisheries (Channa striata, Oreochromis niloticus, Trichogaster, Hemibagrus capitulum, and Pangasius). Indonesian fish oil is rich in omega-3 fatty acids, especially eicosapentaenoic acid and docosahexaenoic acid, and can potentially be developed into drugs and food supplements

Effects of fatty acid synthase-inhibitors on polyunsaturated fatty acid production in marine diatom Fistulifera solaris JPCC DA0580

Omega-3 polyunsaturated fatty acids (PUFAs) such as eicosapentaenoic acid (EPA, C20:5ω-3) are essential for human health and fish growth especially in aquaculture sector. However, with the growing of aquaculture, the demand of PUFA supply also has been increasing. Fistulifera solaris, a marine diatom, is known for its ability to accumulate 65% of lipid content per dry cell weight, and can produce the high content of EPA. Thus, this diatom shows a great potential to be a feedstock of omega-3 PUFAs for fish feeds. In this study, in order to further understand and enhance the metabolism of PUFA biosynthesis in the diatom, the impacts of ketoacyl-ACP synthase (KAS) and ketoacyl-CoA synthase (KCS) inhibition on the PUFA production were analyzed by adding the specific inhibitors. KAS and KCS enzymes both play a role in the fatty acid elongation. As a result, the inhibition of KAS showed an increase in EPA content without arresting the cell growth. On the other hand, inhibition of KCS did not show a significant impact on the PUFA content in F.solaris. Our finding suggests that the specific suppression of KAS function can be a promising way to enhance the omega-3 PUFA production in F.solaris.

α-Linolenic acid and linoleic acid modulate the lipidome and the skin barrier of a tissue-engineered skin model

Polyunsaturated fatty acids (PUFAs) play an important role in the establishment and the maintenance of the skin barrier function. However, the impact of their derived lipid mediators remains unclear. Skin substitutes were engineered according to the self-assembly method with a culture medium supplemented with 10 μM of both α-linolenic acid (ALA) and linoleic acid (LA). The supplementation with ALA and LA decreased testosterone absorption through a tissue-engineered reconstructed skin model, thus indicating an improved skin barrier function following supplementation. The exogenously provided fatty acids were incorporated into the phospholipid and triglyceride fractions of the skin substitutes. Indeed, the dual supplementation increased the levels of eicosapentaenoic acid (EPA) (15-fold), docosapentaenoic acid (DPA) (3-fold), and LA (1.5-fold) in the epidermal phospholipids while it increased the levels of ALA (>20-fold), DPA (3-fold) and LA (1.5-fold) in the epidermal triglycerides. The bioactive lipid mediator profile of the skin substitutes, including prostaglandins, hydroxy-fatty acids, N-acylethanolamines and monoacylglycerols, was next analyzed using liquid chromatography-tandem mass spectrometry. The lipid supplementation further modulated bioactive lipid mediator levels of the reconstructed skin substitutes, leading to a lipid mediator profile more representative of the one found in normal human skin. These findings show that an optimized supply of PUFAs via culture media is essential for the establishment of improved barrier function in vitro. Elevated temperature and browning increase dietary methylmercury, but decrease essential fatty acids at the base of lake food webs

Climate change scenarios predict increases in temperature and organic matter supply from land to water, which affect trophic transfer of nutrients and contaminants in aquatic food webs. How essential nutrients, such as polyunsaturated fatty acids (PUFA), and potentially toxic contaminants, such as methylmercury (MeHg), at the base of aquatic food webs will be affected under climate change scenarios, remains unclear. The objective of this outdoor mesocosm study was to examine how increased water temperature and terrestrially-derived dissolved organic matter supply (tDOM; i.e., lake browning), and the interaction of both, will influence MeHg and PUFA in organisms at the base of food webs (i.e. seston; the most edible plankton size for zooplankton) in subalpine lake ecosystems. The interaction of higher temperature and tDOM increased the burden of MeHg in seston (< 40 μm) and larger sized plankton (microplankton; 40–200 μm), while the MeHg content per unit biomass remained stable. However, PUFA decreased in seston, but increased in microplankton, consisting mainly of filamentous algae, which are less readily bioavailable to zooplankton. We revealed elevated dietary exposure to MeHg, yet decreased supply of dietary PUFA to aquatic consumers with increasing temperature and tDOM supply. This experimental study provides evidence that the overall food quality at the base of aquatic food webs deteriorates during ongoing climate change scenarios by increasing the supply of toxic MeHg and lowering the dietary access to essential nutrients of consumers at higher trophic levels.

Intrauterine Transfer of Polyunsaturated Fatty Acids in Mother-Infant Dyads as Analyzed at Time of Delivery.

Polyunsaturated fatty acids (PUFAs) are essential for fetal development, and intrauterine transfer is the only supply of PUFAs to the fetus. The prevailing theory of gestational nutrient transfer is that certain nutrients (including PUFAs) may have prioritized transport across the placenta. Numerous studies have identified correlations between maternal and infant fatty acid concentrations; however, little is known about what role maternal PUFA status may play in differential intrauterine nutrient transfer. Twenty mother–infant dyads were enrolled at delivery for collection of maternal and umbilical cord blood, and placental tissue samples. Plasma concentrations of PUFAs were assessed using gas chromatography (GC-FID). Intrauterine transfer percentages for each fatty acid were calculated as follows: ((cord blood fatty acid level/maternal blood fatty acid level) × 100). Kruskal–Wallis tests were used to compare transfer percentages between maternal fatty acid tertile groups. A p-value < 0.05 was considered significant. There were statistically significant differences in intrauterine transfer percentages of arachidonic acid (AA) (64% vs. 65% vs. 45%, p = 0.02), eicosapentaenoic acid (EPA) (41% vs. 19% vs. 17%, p = 0.03), and total fatty acids (TFA) (27% vs. 26% vs. 20%, p = 0.05) between maternal plasma fatty acid tertiles. Intrauterine transfer percentages of AA, EPA, and TFA were highest in the lowest tertile of respective maternal fatty acid concentration. These findings may indicate that fatty acid transfer to the fetus is prioritized during gestation even during periods of maternal nutritional inadequacy.

Dietary lipid quality mediates salt tolerance of a freshwater keystone herbivore

Salinization of freshwater ecosystems is a growing hazard for organisms and ecosystem functioning worldwide. In northern latitudes, road salt that is being transported into water bodies can cause year-round increases in lake salinity levels. Exploring the environmental factors driving the susceptibility of freshwater zooplankton to road salt is crucial for assessing the impact of salinization on food web processes. We studied the role of essential lipids, i.e., sterols and long-chain polyunsaturated fatty acids (PUFAs), in mediating salt tolerance of the freshwater keystone herbivore Daphnia. Sterols and PUFAs are involved in regulating ion permeability of biological membranes and thus we hypothesized that the susceptibility to salt is affected by the dietary sterol and PUFA supply. Life history experiments revealed opposing effects of sterol and PUFA supplementation on salt tolerance, i.e., tolerance increased upon sterol supplementation but decreased upon PUFA supplementation, which is consistent with their proposed impact on membrane permeability. Our results suggest that the susceptibility of freshwater zooplankton to salinization strongly depends on the dietary lipid supply and thus the phytoplankton community composition. Hence, trophic state related differences in the phytoplankton community composition need to be considered when assessing the consequences of salinization for freshwater ecosystem functioning.

Comparison of the Cholesterol-Lowering Effect of Scallop Oil Prepared from the Internal Organs of the Japanese Giant Scallop (Patinopecten yessoensis), Fish Oil, and Krill Oil in Obese Type II Diabetic KK-A y Mice.

Due to the growing demand of n-3 polyunsaturated fatty acids (PUFA) as supplements and pharmaceutical products worldwide, there are concerns about the exhaustion of n-3 PUFA supply sources. We have successfully prepared high-quality scallop oil (SCO), containing high eicosapentaenoic acid and phospholipids contents, from the internal organs of the Japanese giant scallop (Patinopecten yessoensis), which is the largest unutilized marine resource in Japan. This study compared the cholesterol-lowering effect of SCO with fish oil (menhaden oil, MO) and krill oil (KO) in obese type II diabetic KK-A y mice. Four-week-old male KK-A y mice were divided into four groups; the control group was fed the AIN93G-modified high-fat (3 wt% soybean oil + 17 wt% lard) diet, and the other three groups (SCO, MO, and KO groups) were fed a high-fat diet, in which 7 wt% of the lard in the control diet was replaced with SCO, MO, or KO, respectively. After the mice were fed the experimental diet for 42 days, their serum, liver, and fecal lipid contents as well as their liver mRNA expression levels were evaluated. The SCO group had significantly decreased cholesterol levels in the serum and liver; this decrease was not observed in the MO and KO groups. The cholesterol-lowering effect of SCO was partly mediated by the enhancement of fecal total sterol excretion and expression of liver cholesterol 7α-hydroxylase, a rate-limiting enzyme for bile acid synthesis. These results indicate that dietary SCO exhibits serum and liver cholesterol-lowering effects that are not found in dietary MO and KO and can help prevent lifestyle-related diseases.

146 Nutritional Advances in Fetal and neonatal development: effect of fatty acid supplementation

Abstract Data from a series of experiments demonstrates that maternal supply of polyunsaturated fatty acids, particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), during late gestation affects offspring growth. The increase in growth is independent on the fatty acid supplemented during the growing or finishing phase of the offspring; but it is sex dependent. Dam PUFA supplementation increases wether growth. Supplementation with EPA and DHA to pregnant ewes and to their offspring after weaning showed a treatment interaction in mRNA concentration of hypothalamic neuropeptides associated with dry matter intake (DMI) regulation. A dose increased in EPA and DHA in pregnant ewe diets shows a linear increase in growth, but a quadratic change in DMI or feed efficiency; growth was associated with a linear increase in plasma glucose concentration and a linear decrease in plasma ghrelin concentration. In lambs born from ewes supplemented with different sources of FA during a glucose tolerance test; males’ plasma insulin concentration increased as FA unsaturation degree increased in the dam diet, the opposite happened with females’ plasma insulin concentration. Recent data from our lab showed that the supplementation with EPA and DHA during the last third of gestation to pregnant ewes increased liver and small intestine global DNA methylation and small intestine transporters for amino acids in the fetus. Despite EPA and DHA during late gestation increase growth in the offspring; when EPA and DHA were supplemented in early gestation, offspring growth was lesser that lambs born from ewes supplemented a saturated and monounsaturated lipid. The reason for the difference in results it is not clear. However, more studies focusing in some aspect of the biology will help to understand what specific fatty acid needs to be supplemented at different stages of gestation to improve offspring growth.

Effect of thermal and nutritional conditions on fatty acid metabolism and oxidative stress response in juvenile European sea bass (Dicentrarchus labrax)

Coastal nursery areas are subjected to a wide range of natural and anthropogenic stressors, including global warming, which indirectly influence trophic food webs. A global rarefaction of n-3 polyunsaturated fatty acids (PUFA) in trophic networks is in progress. The aim of this study was to assess the effect of a reduction in the dietary availability of n-3 PUFA on some molecular and biochemical parameters related to lipid metabolism and oxidative stress response in juvenile European sea bass (Dicentrarchus labrax) raised at two temperatures (15 °C and 20 °C). Fish were fed for five months with a reference diet (RD; 1.65% n-3 PUFA on a dry matter basis, DM), used as a proxy of trophic networks where n-3 PUFA is plentiful, and a lower n-3 PUFA diet (LD; 0.73% n-3 PUFA DM), designed to mimic a decrease in n-3 PUFA resulting from global changes (the n-3 PUFA levels tested remained above the nutritional minimum required for this species). Results showed that diet did not affect the hepatic expression of some mRNA coding for transcriptional factors involved in regulating the metabolic pathways related to fatty acid bioconversion. Although our molecular analysis was limited to transcript expression, these data suggest the presence of a threshold in the nutritional supply of PUFA above which the activation of these molecular pathways does not occur. However, the expression for most of the transcripts tested was up-regulated at 20 °C. Despite the high peroxidation index in fish fed RD, very few modifications of the oxidative stress response were associated with diet. At 20 °C, an increase of the enzymatic antioxidant response was observed, but there was no correlation with the peroxidation index or malondialdehyde products.

Abstract LB-055: Novel therapeutic option targeting the tumor cell lysosomes

Abstract Malignant brain tumors represent a surgical and therapeutic challenge. Despite major breakthrough in the precision neurosurgery and the personalized medicine, it remains impossible to circumvent the tumor relapse. This is due to the highly infiltrative nature of the glioma cells, with single tumor-initiating cells invading far distances in the brain. However, we recently identified a major flaw in the invasive tumor cell biology: a high sensitivity to the lysosomal membrane permeabilization (LMP). We first identified the fatty acid binding-protein 3 (FABP3) specifically expressed by invasive gliomas cells. Expression of FABP3 is indispensable to the transportation of poly-unsaturated fatty acids (PUFAs), which cannot be otherwise synthetized by the cells, from the extracellular space to biological membranes. Silencing of FABP3 stopped the supply of PUFAs that are essential to the lysosomal membrane maintenance. Induction of the LMP activated a cascade of events including the release of the lysosomal cathepsins in the cytosol, leading to the tumor cell death. Interestingly, the specific killing of invasive glioma cells through the LMP can be achieved by using lysosomotropic drugs, such as anti-histamines. These cationic amphiphilic molecules accumulate in the lysosome and induce the phospholipidosis of the lysosomal membrane. We therefore tested the ability of the blood-brain-barrier permeable clemastine, an anti-histamine, to reduce the tumorigenicity several patient-derived gliomas implanted in nude mice. Although clemastine treatment did not modify the primary tumor mass growth, it completely eradicated the invasive population. These promising results obtained by therapeutic activation of the LMP could also be beneficial for other aggressive cancers models, such as brain metastases of breast cancer or metastatic pancreatic cancer. Abstract The intestine plays a crucial role in nutrient digestion and absorption, and its function is critical for animal growth and health. However, the effect of maternal nutrition on offspring’s intestine is not well documented. The objective of this study was to evaluate the effects of maternal supply of polyunsaturated fatty acids, methionine or both during late-gestation on offspring’s intestine amino acids transporters and global DNA methylation. Twenty ewes were used in a randomized block design and treatments were arranged in a 2×2 factorial design. Treatments were: 1) control diet (CTR) without polyunsaturated fatty acids (PUFA) or methionine (MET) supplementation; or supplemented with 2) Ca salts of PUFA (1% DMI; Strata G113®, Virtus Nutrition); rumen-protected MET (0.1% DMI; Smartamine®, Adisseo); and 4) PUFA and MET (PUFA+MET). Treatments were fed from day 100 to 145 of gestation. On day 145, ewes were euthanized and fetuses’ intestines were sampled. Protein quantification of amino acid transporters was measured using Simple Western System and global DNA methylation was determined using a commercial kit. No interactions between PUFA×MET were observed for the variables analyzed. PUFA increased (P ≤ 0.03) global DNA methylation and protein expression of SLC7A5, SLC38A2, and SLC38A10 compared with CTR. Furthermore, PUFA tended to increase (P = 0.07) SLC38A1 protein expression compared with CTR, whereas no difference (P &amp;gt; 0.05) was observed for SLC6A9. Compared with CTR, MET increased (P ≤ 0.05) global DNA methylation and SLC38A10 protein expression, and tented to increase (P ≤ 0.10) SLC6A19 and SLC38A2. MET did not affect SLC7A5 and SLC38A1 compared with CTR. Maternal supply of PUFA and MET led to greater protein expression of amino acid transporters and global DNA methylation on offspring’ intestine; however, the effects of these nutrients were not additive. Further research is necessary to better understand the role of DNA methylation on intestine’s amino acid transporters.

FADS1 and FADS2 Polymorphisms Modulate Fatty Acid Metabolism and Dietary Impact on Health.

Variants in the FADS gene cluster modify the activity of polyunsaturated fatty acid (PUFA) desaturation and the lipid composition in human blood and tissue. FADS variants have been associated with plasma lipid concentrations, risk of cardiovascular diseases, overweight, eczema, pregnancy outcomes, and cognitive function. Studies on variations in the FADS genecluster provided some of the first examples for marked gene-diet interactions in modulating complex phenotypes, such as eczema, asthma, and cognition. Genotype distribution differs markedly among ethnicities, apparently reflecting an evolutionary advantage of genotypes enabling active long-chain PUFA synthesis when the introduction of agriculture provided diets rich in linoleic acid but with little arachidonic and eicosapentaenoic acids. Discovering differential effects of PUFA supply that depend on variation of FADS genotypes could open new opportunities for developing precision nutrition strategies based either on an individual's genotype or on genotype distributions in specific populations.

Dietary polyunsaturated fatty acid supply improves Daphnia performance at fluctuating temperatures, simulating diel vertical migration

Abstract Diel vertical migration (DVM) is a common predator avoidance strategy of zooplankton. Migration to deeper water layers during the day to escape visually hunting predators is most likely to require physiological adaptations to periodically changing temperatures. Polyunsaturated fatty acids (PUFA) play crucial roles in membrane temperature acclimation. Exposure to cold temperatures typically results in an increase in the relative abundance of PUFA in cell membranes and PUFA requirements of Daphnia increase with decreasing temperatures. To assess the role of dietary PUFA in coping with temperature fluctuations experienced during DVM, we reared Daphnia magna at either constantly warm or fluctuating temperatures, simulating DVM both with and without dietary PUFA supplementation. We show that the well‐known positive effect of dietary eicosapentaenoic acid supplementation on offspring production and population growth rates of D. magna is more pronounced at alternating temperatures than at constantly warm temperatures. Exposure to alternating temperatures caused modification in body PUFA concentrations and, consequently, increased lipid peroxidation. However, detrimental effects of lipid peroxidation were not evident. Our data demonstrate that the capacity to cope with the distinct temperature fluctuations experienced during DVM increases with dietary eicosapentaenoic acid supplementation, suggesting that an adequate dietary PUFA supply is crucial especially for migrating Daphnia populations. A dietary deficiency in long‐chain PUFA may thus severely constrain predator evasion, potentially resulting in increased mortality and cascading effects on lower trophic levels.

Elevated Gestational IL-13 During Fetal Development Is Associated With Hyperactivity and Inattention in Eight-Year-Old Children.

Maternal immune activation (MIA) during fetal development leads to behavioral and psychological disorders in the offspring. Concomitantly, insufficient supply of polyunsaturated fatty acids (PUFAs) is suspected to contribute to early neuronal maldevelopment due to the immune modulatory capabilities of PUFAs. However, human data are missing considering both of these aspects and their impact on children's behavioral outcomes. In line, this study aimed to elucidate the influence of gestational cytokines and PUFA-containing lipids during late pregnancy on behavioral sequelae in childhood, particularly focusing on an immune activation shaped by a history of maternal atopic diseases instead of a pathogen-mediated immune response. Based on the prospective mother-child cohort LINA we assessed the unstimulated blood cytokine profiles and concentrations of PUFA-containing lipids of 293 mothers at the 34th week of pregnancy. Maternal history of atopic diseases was obtained from questionnaires and behavior in eight-year-old children was assessed by the standardized Strength and Difficulties Questionnaires (SDQ) generating scores for hyperactivity/inattention, emotional symptoms, conduct problems, and peer relationship problems. Elevated IL-13 increased the risk for the child to show behavioral difficulties, in particular, hyperactive/inattentive behavior [adj. OR (95% CI): 2.47 (1.51–4.02), n = 255 vs. 38] at the age of eight years. Although the presence of maternal atopic dermatitis (AD) was associated with increased gestational IL-13 concentrations [adj. MR (95% CI): 1.17 (1.04–1.32)], no effect on children's behavioral difficulties was observed. However, a decrease in the PUFA containing lipid species PC aa C38:6 was not only associated with an increased gestational IL-13 concentration but also mediated the indirect effect of low PC aa C38:6 concentrations on children's abnormal behavior independent of maternal AD. We additionally assessed whether maternal IL-13 and PC aa C38:6 concentrations translate their effect by altering children's cord blood PC aa C38:6 and IL-13. While also the children's cord blood IL-13 was related to children's behavior, no effect of children's PC aa C38:6 was observed. This is the first study demonstrating that elevated gestational IL-13 increases the risk for children to develop behavioral difficulties. Analyses suggest that a reduced supply of gestational PC aa C38:6 contributes to elevated gestational IL-13 leading to behavioral sequelae in the offspring.

Rearing Temperature and Fatty Acid Supplementation Jointly Affect Lipid Fluorescence Polarization and Heat Tolerance in Daphnia.

The homeoviscous adaptation hypothesis states that the relative abundance of polyunsaturated fatty acids (PUFAs) in membrane phospholipids of ectothermic organisms decreases with increasing temperatures to maintain vital membrane properties. We reared Daphnia magna at 15°, 20°, and 25°C and increasing dietary concentrations of the long-chain PUFA eicosapentaenoic acid (EPA) to test the hypothesis that the well-documented increase in heat tolerance of high-temperature-reared Daphnia is due to a reduction in body PUFA concentrations. Heat tolerance was assessed by measuring the time to immobility at a lethally high temperature (Timm at 37°C), and whole body lipid fluorescence polarization (FP) was used as an estimate of membrane fluidity. At all rearing temperatures, EPA supplementation resulted in an increase in the relative abundance of EPA in body tissues, but only at 15° and 25°C did this result in a decrease in heat tolerance, and only at 20°C was this associated with an increase in membrane fluidity (i.e., decrease in FP). Overall, however, the degree of tissue fatty acid unsaturation correlated well with heat tolerance and FP. Our results support the homeoviscous adaptation hypothesis by showing that cold-reared Daphnia accumulate PUFAs within their body tissues and thus are more susceptible to heat than hot-reared Daphnia accumulating fewer PUFAs. However, our data also point out that further studies are required that elucidate the complex relationships between PUFA supply, membrane fluidity, and heat tolerance in ectotherms.
