Dietary supplementation with Chlorella vulgaris in broiler chickens submitted to heat-stress: effects on growth performance and meat quality

Heat stress can greatly challenge growth and meat quality of broiler chickens where research is looking for sustainable ingredients, such as microalgae, that could also alleviate its negative impacts. Thus, in the present study, 576 1-D-old chicks (Ross 308) were housed until commercial slaughtering (42 D) in 36 pens in 2 rooms of a poultry house, according to a full factorial design encompassing 2 room temperatures (standard vs. high), 2 sexes (females vs. males), and 3 dietary treatments, that is, diet C0 (control diet), diet C3, and diet C6 containing 0, 3, and 6%, respectively, of C. vulgaris meal replacing the same quantities of soybean meal. The highest inclusion level of C. vulgaris decreased feed intake (P < 0.001) and body weight (P < 0.0001) compared to the control diet; it increased yellow and red indexes (P < 0.0001) of the breast muscle, besides the proportion of n3 polyunsaturated fatty acids (PUFA) (P = 0.028). Heat stress decreased feed intake (P = 0.001), breast (P = 0.001) and p. major yields (P = 0.036), and increased meat pH (P= 0.008) and cooking losses (P < 0.001), umami (P = 0.021) and brothy flavor (P < 0.001), and the proportion of n3 PUFA rates (P = 0.027), while reducing the contents of several amino acids in the breast meat (P ≤ 0.05). Compared to females, males displayed higher feed intake and growth, and more favorable feed conversion (P < 0.001). Carcass and p. major yields were greater in females (P < 0.001) which also showed a higher occurrence of spaghetti meat compared to males (P < 0.001). In conclusion, C. vulgaris can be used to replace until 3% of soybean meal in diets for broiler chickens without negative implications, while positively affecting breast meat color according to consumers' preferences. However, the microalgae inclusion did not mitigate the negative effects of a chronic heat stress on growth performance nor reduced the occurrence of any myopathies.

Mycoproteins and their health-promoting properties: Fusarium species and beyond.

Filamentous fungal mycoproteins have gained increasing attention as sustainable alternatives to animal and plant-based proteins. This comprehensive review summarizes the nutritional characteristics, toxicological aspects, and health-promoting effects of mycoproteins, focusing on those derived from filamentous fungi, notably Fusarium venenatum. Mycoproteins are characterized by their high protein content, and they have a superior essential amino acid profile compared to soybeans indicating excellent protein quality and benefits for human nutrition. Additionally, mycoproteins offer enhanced digestibility, further highlighting their suitability as a protein source. Furthermore, mycoproteins are rich in dietary fibers, which have been associated with health benefits, including protection against metabolic diseases. Moreover, their fatty acids profile, with significant proportions of polyunsaturated fatty acids and absence of cholesterol, distinguishes them from animal-derived proteins. In conclusion, the future of mycoproteins as a health-promoting protein alternative and the development of functional foods relies on several key aspects. These include improving the acceptance of mycoproteins, conducting further research into their mechanisms of action, addressing consumer preferences and perceptions, and ensuring safety and regulatory compliance. To fully unlock the potential of mycoproteins and meet the evolving needs of a health-conscious society, continuous interdisciplinary research, collaboration among stakeholders, and proactive engagement with consumers will be vital.

Analysis of carcass traits, meat quality, amino acid and fatty acid profiles between different duck lines

To investigate the effect of genetic selection on meat quality in ducks, twenty of each fast growth ducks (LCA) and slow growth ducks (LCC) selected from F6 generation of Cherry Valley ducks (♂) x Liancheng white ducks (♀) were analyzed for carcass characteristics, meat quality (physicochemical and textural characteristics), amino acid and fatty acid profiles at 7 wk. Results showed that live body weight, slaughter weight, eviscerated yield and abdominal fat percentage of LCA were significantly higher than those in LCC ducks (P < 0.01). Moreover, the average area and diameter of myofiber were larger in LCA than LCC ducks (P < 0.01). The breast and thigh muscles of LCA exhibited significantly lower water holding capacity and thermal loss compared with LCC ducks (P < 0.01). In addition, the content of nonessential amino acids (Glu, Asp, and Arg) in breast muscles and Asp, Ser, Thr, and Met in thigh muscles was higher in LCC than LCA ducks (P < 0.05). The proportion of polyunsaturated fatty acids (PUFA) in breast muscles of LCC was higher than LCA ducks (P < 0.05). However, the content of saturated fatty acids (SFA) in breast and thigh muscles of LCA was higher compared with LCC ducks (P < 0.05). The proportion of monounsaturated fatty acids (MUFA) in thigh muscles was significantly higher in LCC compared with LCA ducks (P < 0.01). Finally, multiple traits were evaluated by applying principal component analysis (PCA) and the results indicated that PUFA and SFA in breast muscles of LCA played important roles in meat quality, followed by Warner-Bratzler shear force (WBSF) and MUFA. However, water holding capacity (WHC) had a dominant effect in meat quality of thigh muscles in both LCA and LCC ducks.

Fatty acid composition of two small fish in the Gyda River (Kara Sea Basin, Northwestern Siberia, Russia)

The fatty acid composition of the flesh of rainbow smelt (RS, Osmerus mordax) and ruffe (RF, Gymnocephalus cernuus), two small fish species inhabiting the Gyda River (Siberia, Russia), was assayed using a gas-liquid chromatographic method with an ionization detection system and high efficiency capillary columns. RS was found to be richer in the total fatty acid content than RF (21.5 vs. 7.07 mg g–1). The proportion of saturated fatty acids (SFA) was similar in the fish studied (29.2 % in RS and 29.6 % in RF) whereas the flesh of RS contained a higher proportion of monounsaturated fatty acids, MUFA (42.2 vs. 13.9 %), and a lower proportion of polyunsaturated fatty acids, PUFA (28.6 vs. 56.5 %) than RF. RS is rich in essential n-3 PUFAs (2610 and 2390 μg g–1 for docosahexaenoic and eicosapentaenoic acids, respectively). In RF, the atherogenic index (0.39) and thrombogenic (0.20) index were lower than in RS (0.59 and 0.27, respectively). The fatty acid profile of RS in the study area differed from that reported for the flesh of this species from a Canadian estuary probably due to substantial differences in the diet of RS in these habitats. A comparison among fish species from the Gyda River indicated that in terms of nutrition indices, RS has the highest similarity with the muksun Coregonus muksun while RF is closely related to the northern pike Esox lucius. Based on the results from this study, both RF and RS can be recommended for human consumption. In particular, RS may be used as a replacement for muksun, an overfished species, in the traditional diet of indigenous peoples in northwestern Siberia.

Effect of genotype on performance, vitamin and carotenoid deposition, oxidative stability, fatty acid profile and sensory characteristics in cockerels housed on litter and in mobile boxes on pasture

The study evaluated the effect of housing system and genotype on performance and breast meat quality in 540 cockerels. Three genotypes of cockerels differing in growth intensity (slow-growing ISA Dual, medium-growing Hubbard JA757 and fast-growing Ross 308) and two housing systems (litter and mobile box on pasture) were compared. The significantly lowest feed conversion (p < .001) was recorded for Ross 308 cockerels housed on litter, while ISA Dual cockerels from mobile boxes had a 74% higher conversion ratio. The highest concentrations of lutein (p = .043) and α-tocopherol (p = .024) in breast meat were found in the cockerels from mobile boxes of genotypes ISA Dual and Ross 308, respectively. Meat stored for five days showed the highest oxidative stability of fat (p = .001) in slow- and medium-growing cockerels housed on pasture. The ISA Dual genotype housed both on litter and on pasture and the Hubbard JA757 genotype from mobile boxes had the highest proportion of polyunsaturated fatty acids in the meat (p < .001). The lowest n6/n3 fatty acid ratio (p = .045) and thrombogenic index (p < .001) and the highest hypocholesterolaemic/hypercholesterolaemic index (p < .001) were recorded in slow-growing chickens with access to pasture. The meat of cockerels fattened on litter was considered more fragrant (p = .013), more tender (p < .001), juicier (p < .001) and overall more acceptable (p = .001). It can be concluded that the ISA Dual genotype showed the highest willingness to graze compared to Hubbard JA757 and Ross 308 cockerels and made the best use of the benefits of pasture fattening, which was reflected in the high quality of the meat. The meat of conventionally housed cockerels was sensorially evaluated as better. Highlights The ISA Dual genotype showed the highest willingness to graze, which was reflected in the higher content of health-promoting substances in meat. The more frequent use of slow-growing cockerels for the purpose of fattening will be hindered by lower performance of the cockerels and low level of meat tenderness.

Comparative transcriptome analysis of longissimus dorsi muscle reveal potential genes affecting meat trait in Chinese indigenous Xiang pig

In this study, we compared the transcriptome of longissimus dorsi muscle between Guizhou Xiang pigs (XP) and Western commercial Large White pigs (LW), which show diffirent meat quality between them. In terms of meat quality traits, the pH 45 min, color score, backfat thickness, and intramuscular fat (IMF) content were higher in Xiang pigs than in Large White pigs (P < 0.01), while the drip loss, lean meat percentage, shear force, and longissimus dorsi muscle area of Xiang pigs were lower than that of Large White pigs (P < 0.01). Nutrients such as monounsaturated fatty acid (MUFA), total amino acids (TAA), delicious amino acids (DAA) and essential amino acids (EAA) in Xiang pigs were higher than that in Large White pigs, and the proportion of polyunsaturated fatty acid (PUFA) of Xiang pigs was significantly lower than Large White pigs (P < 0.01). Transcriptome analysis identified 163 up-regulated genes and 88 genes down-regulated in Xiang pigs longissimus dorsi muscle. Combined with the correlation analysis and quantitative trait locis (QTLs) affecting meat quality, a total of 227 DEGs were screened to be significantly associated with meat quality values. Enrichment analysis indicated that numerous members of genes were gathered in muscle development, adipogenesis, amino acid metabolism, fatty acid metabolism and synthesis. Of those, 29 genes were identified to be hub genes that might be related with the meat quality of Xiang pig, such as MYOD1, ACTB, ASNS, FOXO1, ARG2, SLC2A4, PLIN2, and SCD. Thus, we screened and identified the potential functional genes for the formation of meat quality in Xiang pigs, which provides a corresponding theoretical basis for the study of the molecular regulatory mechanism of pork quality and the improvement of pork quality.

The Effect of Combined Exposure of Fusarium Mycotoxins on Lipid Peroxidation, Antioxidant Defense, Fatty Acid Profile, and Histopathology in Laying Hens' Liver.

Fumonisin B1, T-2 toxin, and deoxynivalenol are frequently detected in feed materials. The mycotoxins induce free radical formation and, thereby, lipid peroxidation. The effects of mycotoxin exposure at the EU recommended limit (T-2/HT-2 toxin: 0.25 mg/kg; DON = 3AcDON/15-AScDON: 5 mg/kg; fumonisin B1: 20 mg/kg) and double dose (T-2/HT-2 toxin: 0.5 mg/kg, DON/3-AcDON/15-AcDON: 10 mg, and FB1: 40 mg/kg feed) were investigated during short-term (3 days) per os exposure in the liver of laying hens. On day 1 higher while on day 3 lower MDA concentrations were found in the low-dose group compared to the control. Fatty acid composition also changed: the proportion of monounsaturated fatty acids increased (p < 0.05) and the proportion of polyunsaturated fatty acids decreased by day 3. These alterations resulted in a decrease in the index of unsaturation and average fatty acid chain length. Histopathological alterations suggested that the incidence and severity of liver lesions were higher in the mycotoxin-treated laying hens, and the symptoms correlated with the fatty acid profile of total phospholipids. Overall, the findings revealed that mycotoxin exposure, even at the EU-recommended limits, induced lipid peroxidation in the liver, which led to changes in fatty acid composition, matched with tissue damage.

Molecular phylogenetics of the Umbelopsis genus-identification of new species and evaluation of their oil application value.

The aim of this study was to reconstruct the evolutionary framework of the genus Umbelopsis by using modern taxonomic strategies and evaluating the quality of oil and prospective uses of three distinct species. Three species of Umbelopsis were identified based on morphological characteristics and phylogenetic evidence obtained from three genes (ITS, LSU, and ACT). A new species of Umbelopsis was described and illustrated, and subsequently named U. ophiocordycipiticola. The characteristics of U. ophiocordycipiticola exhibited sporangia with a diameter ranging from 8 to 17µm. and sporangiospores that were oval to ellipsoidal in shape, irregularly angular, with dimensions of ∼1.9-2.9×1.7-3.0µm. Gas chromatography and mass spectrometry (GC-MS) were used to examine the composition of fatty acids. Notably, U. ophiocordycipiticola showed a significantly higher oil content of 50.89% in dry cell weight (DCW) compared to U. vinacea and U. ramanniana. The mean proportion of polyunsaturated fatty acids (PUFAs) in U. ophiocordycipiticola was 32.38%, and the maximum levels of γ-linolenic acid (GLA), arachidonic acid (ARA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA) in U. ophiocordycipiticola were found to be 14.51, 0.24, 0.54, and 0.53%, respectively. The biodiesel quality from all three species complied with applicable standards set by the American Association for Testing and Materials (ASTM 6751) and the Brazilian National Petroleum Agency (ANP 255). The establishment of a novel species, U. ophiocordycipiticola, was strongly supported by morphological and molecular evidence. Umbelopsis ophiocordycipiticola exhibited a high-value PUFA content. Additionally, three Umbelopsis species demonstrated good quality for biodiesel production.

Effects of postharvest piling up in bulk on qualities of Camellia oleifera seeds

The study investigated the effects of postharvest piling up in bulk on Camellia oleifera seed qualities, including moisture content, oil content, fatty acid composition, soluble sugar, soluble protein content, acid value, peroxide value, lipase activity, lipoxygenase activity, and mold colony, under varying relative humidity (60%, 80%, 98%) and time durations (1–8 days). Additionally, it examined the impact of piling temperature and diverse postharvest treatments. The results revealed that piling up in bulk promoted the post-ripening of Camellia oleifera seeds, leading to a gradual decrease in seed moisture content while initially increasing oil content before stabilizing. After 2 days of piling, seed oil content increased by 5.65%, 4.40%, and 1.06% under RH conditions of 60%, 80%, and 98%, respectively. However, after 4 days, the oil content declined by 8.36%, 6.06%, and 4.14%, respectively. The proportions of saturated and polyunsaturated fatty acids declined gradually, while monounsaturated fatty acids increased significantly. The acid value decreased, correlating positively with lipase activity. Mold colonies on seeds increased minimally with piling duration, while those on fruit shells increased sharply by 1–4 logarithmic units after the 4th day. When examining the effect of temperature, higher piling temperatures were found to gradually increase the seed oil content. Based on the order of oil content, the different treatment methods were ranked as follows: piling up in bulk-sun drying > piling up in bulk-drying > shelling-sun drying > direct sun drying > shelling-drying. Therefore, these findings advance our understanding of postharvest piling up in bulk of Camellia oleifera seeds and provide valuable insights for selecting treatment methods and parameters.

Assimilation of fish farm wastes by the ecosystem engineering bivalve Atrina zelandica

As feed-additive aquaculture expands to open ocean areas, there is concern that ecologically important habitats may be adversely impacted by sedimentation of farm wastes. In this study, we investigated assimilation of salmon faecal wastes by an ecosystem engineering bivalve that occurs in open ocean environments (Atrina zelandica), as well as effects on physiology and fatty acid metabolism. A. zelandica were subjected to one of 3 treatment diets (fish faeces, 1:1 mix of algae:faeces and algae) in a 51 d laboratory trial. We found a diet-related response in fatty acid composition, including increased prevalence of oleic acid (OA) in digestive tissues of A. zelandica fed on both the fish faeces diet and the mixed diet, indicating fish wastes were assimilated in both treatments. Fish waste consumption was related to a more marked reduction in fatty acid content of digestive gland, as well as lower proportions of long-chain polyunsaturated fatty acids (LC-PUFA) in digestive tissues. Fatty acid composition in gonad and muscle tissues was more strongly influenced by sex. Regardless of dietary treatment, females accumulated C18 fatty acids in gonad tissues, particularly OA, which may preclude the use of OA as a fish waste tracer in this organ. The accumulation of specific fatty acids according to sex may indicate a capacity for preferential selection and retention or biosynthesis of biologically important fatty acids. If present, these mechanisms may increase resilience of A. zelandica to stress from deficiencies in LC-PUFA when using fish wastes as a trophic subsidy.

Effects of Genotype on Pig Carcass, Meat Quality and Consumer Sensory Evaluation of Loins and Bellies.

The objective of the study was to compare carcass and meat quality among Lithuanian White, Lithuanian Indigenous Wattle and conventional hybrids as well as consumer sensory evaluations. The pigs were slaughtered at a live weight of approximately 95-100 kg. After 24 h of slaughter, carcasses were evaluated and sampled for the analysis of meat quality traits of loins and consumer evaluation of cooked loins and baked bellies. The pigs of both Lithuanian breeds had higher backfat thickness and depth of underbelly but lower loin area compared with hybrids. However, more fatty local breeds had higher intramuscular fat content but lower cholesterol content than hybrids. The loins from local pigs displayed higher pH and colour with lower lightness and yellowness but higher redness and also lower cooking loss, shear force and hardness than conventional hybrids. Pork from lean hybrids had a higher proportion of polyunsaturated fatty acids and more favourable lipid quality indices such as atherogenic (AI) and thrombogenic (TI) indices and the hypocholesterolemic/hypercholesterolemic (h/H) ratio; however, the peroxidisability index (PI) and iodine value (IV) were less favourable compared with local pigs. Consumers evaluated cooked loins and baked bellies. The loins from local breeds scored higher in juiciness, taste and overall acceptability compared with conventional hybrids. However, a higher overall acceptability was observed for the lean bellies of hybrids. The results can be used to increase pork consumption choices.

Effects of sex on meat quality traits, amino acid and fatty acid compositions, and plasma metabolome profiles in White King squabs

The objective of this study was to investigate the effects of sex on meat quality and the composition of amino and fatty acids in the breast muscles of White King pigeon squabs. Untargeted metabolomics was also conducted to distinguish the metabolic composition of plasma in different sexes. Compared with male squabs, female squabs had greater intramuscular fat (IMF) deposition and lower myofiber diameter and hydroxyproline content, leading to a lower shear force. Female squabs also had higher monounsaturated fatty acid and lower n-6 and n-3 polyunsaturated fatty acid proportions in the breast muscle, and had greater lipogenesis capacity via upregulation of PPARγ, FAS and LPL gene expression. Moreover, female squabs had lower inosine 5'-monophosphate, essential, free and sweet-tasting amino acid contents. Furthermore, Spearman's correlations between the differential plasma metabolites and key meat parameters were assessed, and putrescine, N-acetylglutamic acid, phophatidylcholine (18:0/P-16:0) and trimethylamine N-oxide were found to contribute to meat quality. In summary, the breast meat of male squabs may have better nutritional value than that of females, but it may inferior in terms of sensory properties, which can be attributed to the lower IMF content and higher shear force value. Our findings enhance our understanding of sex variation in squab meat quality, providing a basis for future research on pigeon breeding.

Bacteroides thetaiotaomicron ameliorates mouse hepatic steatosis through regulating gut microbial composition, gut-liver folate and unsaturated fatty acids metabolism

ABSTRACT Gut microbiota plays an essential role in the progression of nonalcoholic fatty liver disease (NAFLD), making the gut-liver axis a potential therapeutic strategy. Bacteroides genus, the enriched gut symbionts, has shown promise in treating fatty liver. However, further investigation is needed to identify specific beneficial Bacteroides strains for metabolic disorders in NAFLD and elucidate their underlying mechanisms. In this study, we observed a positive correlation between the abundance of Bacteroides thetaiotaomicron (B. theta) and the alleviation of metabolic syndrome in the early and end stages of NAFLD. Administration of B. theta to HFD-fed mice for 12 weeks reduced body weight and fat accumulation, decreased hyperlipidemia and insulin resistance, and prevented hepatic steatohepatitis and liver injury. Notably, B. theta did not affect these indicators in low-fat diet (LFD)-fed mice and exhibited good safety. Mechanistically, B. theta regulated gut microbial composition, characterized by a decreased Firmicutes/Bacteroidetes ratio in HFD-Fed mice. It also increased gut-liver folate levels and hepatic metabolites, alleviating metabolic dysfunction. Additionally, treatment with B. theta increased the proportion of polyunsaturated fatty acid in the mouse liver, offering a widely reported benefit for NAFLD improvement. In conclusion, this study provides evidence that B. theta ameliorates NAFLD by regulating gut microbial composition, enhancing gut-liver folate and unsaturated fatty acid metabolism, highlighting the therapeutic role of B. theta as a potential probiotic for NAFLD.

Chestnut tannin extract modulates growth performance and fatty acid composition in finishing Tan lambs by regulating blood antioxidant capacity, rumen fermentation, and biohydrogenation

Tannins as plant extracts have emerged as promising and potential alternatives for antibiotics in modern livestock cultivation systems. This study investigates the effect of dietary chestnut tannin extract (CTE) in finishing Tan lambs. Twenty-seven male Tan lambs were randomly divided into three groups: (1) control group (CON; basal diet); (2) low-dose CTE group (LCTE; basal diet + 2 g/kg CTE, dry matter [DM] basis); (3) high-dose CTE group (HCTE; basal diet + 4 g/kg CTE, DM basis). The HCTE group exhibited markedly higher average daily gain (ADG) and DM intake than CON (P < 0.01). The ruminal total volatile fatty acid concentration increased linearly with increasing CTE supplementation (P < 0.01), while the opposite trend was observed for butyrate molar proportion (P < 0.01). Upon increasing CTE dosage, plasma glucose, high-density lipoprotein cholesterol, glutathione peroxidase, and superoxide dismutase content increased linearly (P < 0.05), whereas low-density lipoprotein cholesterol and urea nitrogen decreased linearly or quadratically (P < 0.05), respectively. A linear increase was also observed in ruminal t6 C18:1 and t9, c12 C18:2 proportions (P < 0.01), and plasma C18:2n-6 and n-6 polyunsaturated fatty acids proportions with increased CTE supplementation (P < 0.01). In the longissimus dorsi muscle, the atherogenic index decreased linearly (P < 0.05), while c11 C18:1 and C20:5n-3 increased linearly (P < 0.05). Moreover, c9, t11 conjugated linoleic acids proportion increased in subcutaneous fat with CTE supplementation (P < 0.01). In conclusion, Dietary CTE enhances the ADG of finishing Tan lambs in a dose-dependent manner, modulates plasma metabolites and antioxidant capacity, and improves rumen fermentation and body fatty acid composition. These results provide a reference for the rational application of CTE in ruminant production.

Utilization of Blackthorn Plums (Prunus spinosa) and Sweet Cherry (Prunus avium) Kernel Oil: Assessment of Chemical Composition, Antioxidant Activity, and Oxidative Stability

Prunus avium L. and Prunus spinosa L. are valuable fruit-bearing trees known for their bioactive compounds and medicinal properties. However, limited research exists regarding their kernel oils. This study aimed to compare the chemical composition, quality parameters, and bioactive potential of the kernel oils extracted from Prunus avium L. and Prunus spinosa L. The kernel oils’ fatty acid and tocopherol profiles were characterized, and the presence of bioactive compounds were identified and quantified. Total polyphenol content (TPC) and antioxidant activity (AAC) were also measured, indicating the presence of bioactive compounds in both oils. Additionally, the main quality parameters, including oxidative status, were evaluated. The fatty acid analysis revealed a higher proportion of polyunsaturated fatty acids compared to monounsaturated fatty acids in both kernel oil samples. Linoleic acid (57–64%) and oleic acid (18–29%) were the major fatty acids in both Prunus avium L. and Prunus spinosa L. kernel oils. α-Eleostearic acid (11.87%) was quantified only in Prunus avium kernel oil. Furthermore, the α-, β-, γ-, and δ-tocopherol content were determined, and it was found that both kernel oils contained γ-tocopherol as the major tocopherol (~204–237 mg/Kg). TPC in Prunus avium L. kernel oil was measured at 9.5 mg gallic acid equivalents (GAE)/Kg and recorded as ~316% higher TPC than Prunus spinosa L. kernel oil. However, the recorded AAC were 11.87 and 14.22 μmol Trolox equivalent (TE)/Kg oil, respectively. Both oils recorded low peroxide values (~1.50 mmol H2O2/Kg), and low TBARS value (~0.4 mmol malondialdehyde equivalents, MDAE/Kg oil), but high p-anisidine value (23–32). The results indicated that both Prunus avium L. and Prunus spinosa L. kernel oils exhibited unique chemical compositions.

Endothelial cells (ECs) play an important role in tissue homeostasis. Recently, EC lipid metabolism has emerged as a regulator of EC function. The liver X receptors (LXRs) are involved in the transcriptional regulation of genes involved in lipid metabolism and have been identified as a potential target in cardiovascular disease. We aimed to decipher the role of LXRs in the regulation of lipid metabolism in human aortic endothelial cells. Lipid composition analysis of endothelial cells treated with the LXR agonist T0901317 revealed that LXR activation increased the proportion of polyunsaturated fatty acids (PUFAs) and decreased the proportion of saturated fatty acids. The LXR agonist decreased the uptake of fatty acids (FAs) by ECs. This effect was abolished by LXRα silencing. LXR activation increased the activity and the expression of lysophosphatidylcholine acyltransferase, LPCAT3, which is involved in the turnover of FAs at the sn-2 position of phospholipids. Transcriptomic analysis also revealed that LXRs increased the expression of key genes involved in the synthesis of PUFAs, including FA desaturase one and 2, FA elongase 5 and fatty acid synthase. Subsequently, the LXR agonist increased PUFA synthesis and enhanced arachidonic acid, eicosapentaenoic acid, and docosahexaenoic acid content in the EC phospholipids. Modification of the FA composition of ECs by LXRs led to a decrease of arachidonate and linoleate derived prostaglandins synthesis and release. No change on markers of inflammation induced by plasma from sickle cell patient were observed in presence of LXR agonist. These results identify LXR as a key regulator of lipid metabolism in human aortic endothelial cells and a direct effect of LXR agonist on lysophosphatidylacyl transferase (LPCAT3).

Влияние зимовки на морфофизиологические и химические показатели судака Sander lucioperca

During the wintering period, sander from their natural habitat and grown in farm conditions undergo changes in morphophysiological and chemical status due to the expenditure of energy reserves of the body and the redistri-bution of nutrients in the muscles and liver. This is expressed in a significant decrease in the content of ab-dominal fat in the body of wild and factory individuals (by 45 and 16%, respectively), muscle fat content (by 26% in wild individuals) and liver (by 59% in wild and 16% in farm fish), vitamin C in muscles (by 56 and 23%, respectively). At the same time, the content of nitrogen–free extractives in the muscles and liver increases many times – by 47 and 31%, respectively, in wild individuals and by 31 and 29% in sander raised in cages. The proportion of monounsaturated fatty acids in the lipids of sander muscles in winter significantly increases and the proportion of polyunsaturated fatty acids (PUFA) decreases, especially eicosapentaenoic (EPA), docosahexaenoic (DHA) and arachidonic, against the background of an increase in the level of these acids in the liver. From the point of view of meat quality, estimated by the content of long-chain PUFA, during the wintering period there is a significant decrease in the nutritional value &#x0D; of caught and raised fish, which is expressed in a 2-3-fold decrease in EPA + DHA in fillets. Such changes are the adaptation of fish to cold temperature conditions during prolonged wintering and are associated with puberty.

Association between erythrocyte membrane fatty acids and gut bacteria in obesity-related cognitive dysfunction

Obesity increases the risk of cognitive impairment and dementia, and the gut microbiota can affect brain cognitive function and obesity through a variety of pathways such as the gut-brain axis. This study aimed to discover how fatty acid affect cognitive function by regulating intestinal flora in obesity. Obese subjects were recruited for cognitive function assessment, and participants were divided into obese group with cognitive impairment (MCI, n = 49) and obese cognitively normal group (Non_MCI, n = 55). In the erythrocyte membrane, the proportion of polyunsaturated fatty acids (PUFA), linoleic acid (C18:2 n-6) and arachidonic acid (C20:4 n-6) and n-6/n-3 ratio was higher in the MCI group than in the Non_MCI group. However, the α-linolenic acid (C18:3 n-3) percentage of the erythrocyte membrane was lower in the MCI group. We found that Coriobacteriales_Incertae_Sedis was positively correlated with erythrocyte membrane C20:4 n-6 and n-6 PUFA and negatively correlated with cognitive scores in obese patients. In addition, several of the functional pathways we predicted were significantly different in the MCI and Non_MCI groups. Higher levels of n-6/n-3 polyunsaturated fatty acids ratio in the erythrocyte membranes may influence the inflammatory response in the organism causing obesity induced cognitive damage. Moreover, high levels of n-6/n-3 polyunsaturated fatty acids ratio may also affect the intestinal flora of obese patients, which in turn may affect the cognitive function of obese patients.

The fatty acid profile and the quality of breast and leg muscles in female and male pearl gray guinea fowl (Numida meleagris)

The aim of this study was to evaluate the fatty acid (FA) profile and the quality of breast and leg muscles in male and female guinea fowl. In comparison with leg muscles, breast muscles had a higher content of dry matter and total protein, and contained less fat and total collagen (P ≤ 0.05). Sex had no effect on the chemical composition of the studied muscles or the FA profile of intramuscular fat (IMF) (P > 0.05). The IMF of breast muscles was characterized by higher proportions of total saturated fatty acids (SFAs) and polyunsaturated fatty acids (PUFAs), and lower proportions of total unsaturated fatty acids (UFAs) and monounsaturated fatty acids (MUFAs) (P ≤ 0.05) than the IMF of leg muscles. The physicochemical properties of guinea fowl meat were significantly influenced by muscle type (P ≤ 0.05), but not by sex (P > 0.05). Leg muscles were characterized by a higher pH, higher cooking loss, lower lightness (L*), lower drip loss, a higher contribution of redness (a*), and a lower contribution of yellowness (b*) than breast muscles (P ≤ 0.05). The experimental factors had no effect on the sensory attributes or the shear force values of the analyzed muscles (P > 0.05). The unique sensory attributes, high protein content, and low fat content of the analyzed muscles indicate that guinea fowl meat can be a dietary supplement and an addition to other types of meat consumed, especially that most guinea fowl are reared in extensive farming systems that contribute to sustainable food production.

Comparative oil extraction from mutt (Myliobatis goodei) liver by enzymatic hydrolysis: free versus immobilized biocatalyst.

The development and fine-tuning of biotechnological processes for fish oil extraction constitute a very important focus to contribute to the development of a food industry based on fish consumption. This work lies in a comparative analysis of the oil extraction yield of Myliobatis goodei livers using free and immobilized enzymes. An immobilized biocatalyst was designed from the cell-free extract of a Bacillus sp. Mcn4. A complete factorial design was used to study the components of the bacterial culture medium and select the condition with the highest titers of extracellular enzymatic activities. Wheat bran had a significant effect on the culture medium composition for enzymatic production. The immobilized biocatalyst was designed by covalent binding of the proteins present in the cocktail retaining a percentage of different types of enzymatic activities (Mult.Enz@MgFe2 O4 ). Among the biocatalyst used, Alcalase® 2.4 L and Purazyme® AS 60 L (free commercial proteases) showed extraction yields of 87.39% and 84.25%, respectively, while Mult.Enz@MgFe2 O4 achieved a better one of 89.97%. The oils obtained did not show significant differences in their physical-chemical properties while regarding the fatty acid content, the oil extracted with Purazyme® AS 60 L showed a comparatively lower proportion of polyunsaturated fatty acids. Our results suggest that the use of by-products of M. goodei is a valid alternative and encourages the use of immobilized multienzyme biocatalysts for the treatment of complex substrates in the fishing industry. © 2023 Society of Chemical Industry.
