Blood indices of omega-3 and omega-6 polyunsaturated fatty acids are altered in hyperglycemia

Polyunsaturated fatty acids (PUFAs) may favorably influence the risk and clinical course of diabetes mellitus (DM). In particular, eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and arachidonic acid (AA) alleviate oxidative injury and insulin resistance characteristic of DM. Uncertainty still remains, however, as to the composition and proportions of blood PUFAs in relation to fasting blood glucose levels. This study, thus, aims to examine the patterns of blood PUFA indices in normoglycemic (NG) and hyperglycemic (HG) Saudi subjects. Age, gender, FA profiles, and laboratory records of 143 subjects collected from September 2014 to March 2018 were retrospectively analyzed. Means, prevalence rates, associations, risk measures, and the diagnostic accuracy of PUFAs were determined. HG subjects had significantly lower AA (0.70%, 95% CI: 0.59–0.80% vs 0.46%, 95% CI: 0.38–0.53%) and higher EPA/AA ratio (0.36, 95% CI: 0.30–0.42 vs 0.69, 95% CI: 0.61–0.77). Gender-wise comparisons revealed that ώ-6/ώ-3 ratio was the only PUFA index significantly elevated in HG males (0.36, 95% CI: 0.26–0.45 vs 5.68, 95% CI: 4.98–6.38) while both DHA (2.91%, 95% CI: 2.54–3.29% vs 3.37%, 95% CI: 3.13–3.60%) and ώ-3 index (3.1%, 95% CI: 2.70–3.49% vs 3.63%, 95% CI: 3.38–3.88%) were significantly elevated in HG females. Furthermore, reduced AA and elevated EPA/AA ratio were more prevalent in HG subjects (26.53 vs 28.72 and 30.61 vs 38.29, respectively) and exhibited the highest diagnostic accuracy for HG among all PUFA indices. Altogether, our study revealed that distinct, gender-specific blood PUFA indices are differentially regulated in HG subjects which may be valuable for DM management.

Spectrum of esterified fatty acids of the omega-3 and omega-6 in the blood of persons with vibration pathology

Introduction. Vibration is one of the leading unfavourable physical factors of the working environment and leads to vibration disease (VD) development. The study of metabolic profiles of markers of fatty acids (FA) metabolism is one of the directions in the early diagnosis of vascular diseases and metabolic syndrome (MS), caused, among other things, by exposure to vibration. The aim of this work is to study the spectrum of esterified fatty acids of the omega-3 and omega-6 families in the blood of patients with vibration disease (VD) and combination with metabolic syndrome (MS). Materials and methods. The study involved 66 people, divided into two groups: Group I - 34 patients with an established diagnosis of VD; Group II - 32 patients with an established diagnosis of VD in combination with metabolic syndrome (MS). The levels of esterified forms (EF) of polyunsaturated fatty acids (PUFA) of the ω-3 and ω-6 families in blood plasma samples were determined using the method of gas chromatography-mass spectrometry on an Agilent 7890A gas chromatograph with an Agilent 5975C mass selective detector. Results. The distribution of quantitative indices of PUFA of the ω-3 and ω-6 families in the examined groups was studied. In VD patients with MS, compared with VD patients, there was a statistically significant increase in the level of eicosapentaenoic, α-linolenic acids of the ω-3 family, the sum of ω-3 PUFAs, γ-linolenic and linoleic acids of the ω-6 family. Conclusion. The study of the quantitative indices of EF of the families ω-3 and ω-6 in the blood plasma of VD patients with MS showed the levels of ω-6 to be 4.5-5.4 times statistically higher than the levels of ω-3 fatty acids.

Fatty Acid Content of Structural and Storage Lipids in Muscles of the Daubed Shanny Postlarvae Leptoclinus maculatus (Fries, 1838) from Kongsfjord (Svalbard Archipelago)

A fatty acid content of structural (phospholipids, PLs) and storage (triacylglycerols, TAGs) lipids in muscles of daubed shanny Leptoclinus maculatus postlarvae from Kongsfjord (Svalbard archipelago waters) was studied. A high level of monounsaturated fatty acids (MUFAs), mainly due to 20:1(n-9) and 22:1(n-11), was shown in TAGs of daubed shanny muscles at the L2 stage of larval development. Polyunsaturated fatty acids (PUFAs) were found to dominate in PLs due to n-3, mainly 20:5(n-3) and 22:6(n-3), fatty acids. The ∑n-3/∑n-6 PUFA index was established to be 2-fold higher in PLs compared to TAGs (9.7 vs. 4.1). The fatty acid profile at the L1 stage of larval development was characterized by a pronounced specificity: a highest content of saturated fatty acids (SFA) due to 16:0 and a dominance of 18:1(n-9) among MUFAs. Our results demonstrate the age-related specificity of the fatty acid content of PLs and TAGs in daubed shanny muscles, which determines their functional significance and reflects both feeding features and the physiological state of the daubed shanny at different stages of its development under the Arctic conditions.

Comparison of the Fatty Acid Composition and Lipid Nutritional Indices of Meat in Two Lines of Slow-Growing Chickens and Their F1 Crosses

The study was carried out to compare the fatty acid profile and related lipid nutritional indices in the breast and thigh meat of two slow-growing lines of chickens La Belle (LB) and Bresse Gauloise (BB) and their crosses (♂LBx♀BB, ♂BBx♀LB). The birds were reared indoors in the experimental poultry farm of the Institute of Animal Science –Kostinbrod, Bulgaria. At the age of 12 weeks, 6 male chickens of each line were slaughtered and lipid analysis of the breast and thigh meat was done. The differences in the fatty acid profile and related nutritional indices of the meat between the lines were assessed by one-way ANOVA. The fatty acid composition was affected by the crossing of the lines but to a different extent in breast and thigh meat. The highest amount of monounsaturated fatty acids (MUFA) was observed in the meat of ♂BBx♀LB chickens due to the significant increase of C16:1n-7 in breast and C18:1n-9 in the thigh of these birds. On the other hand, this crossbred line exhibited the lowest levels of polyunsaturated fatty acids (PUFA). Furthermore, the lowest content of MUFA was observed in the breast of the pure lines and thigh in BB and ♂LBx♀BB, while the highest PUFA level was determined in the breast of LB and thigh of ♂LBx♀BB. The differences in the meat dietetic quality described by the lipid indices showed no consistent patterns in the pure and crossbred lines depending on the type of meat. When compared with the rest, the breast meat of the LB male chickens showed significantly higher polyunsaturated/saturated fatty acids (P/S) ratio, while lowest n-6/n-3 PUFA and thrombogenic index (TI). The values of the atherogenic index (AI) and the ration between the hypo-and hypercholesterolemic fatty acids (h/H) were also improved in this line. Thigh meat however, showed best characteristics in terms of P/S and n-6/n-3 in the ♂LBx♀BB cross.

Improvements in the conception rate, milk composition and embryo quality of rabbit does after dietary enrichment with n-3 polyunsaturated fatty acids

This work attempts to confirm the effect of an enriched diet with n-3 polyunsaturated fatty acids (PUFA) trying to mitigate the reproductive performances issues such as low conception rate of primiparous rabbits. A total of 127 does were fed ad libitum throughout their two first cycles with two diets with different fat sources: mixed fat in the control and salmon oil in the enriched one, with 3.19 g/100 g (n=63 does) and 28.77 g/100 g (n=64 does) of n-3 of the total fatty acid, respectively. Feed intake was similar between groups (P>0.05). Plasma progesterone concentration was higher in the enriched females than in control ones at 7 (30.9±2.18 v. 23.9±2.30 ng/ml, respectively; P=0.029) and 14 (38.7±2.18 v. 28.2±2.30 ng/ml, respectively; P=0.001) days of first gestation. Considering both cycles, reproductive parameters of mothers (fertility, duration of gestation and prolificacy) and litter parameters (weight at parturition and weaning, mortality and average daily gain (ADG) of kits during lactation) were similar in both groups. However, individual measurements of neonates of enriched group improved 5.87%, 7.10% and 18.01% (P<0.05) in terms of crown-rump length, biparietal and thoracic diameters, respectively, compared to control ones at first parturition. It is noteworthy that at the second insemination, critical point in rabbit, fertility rate of enriched group did not decline as sharply as in the control group (89.7% v. 76.6%, respectively; P=0.067), although ADG and littler weight were slightly lower at the second lactation after PUFA enrichment (P<0.05). Total PUFA and unsaturated index of milk of enriched does group were significantly elevated than in control one (33.3±0.02 v. 23.2±0.02 g/100 g and 1.20±0.00 v. 0.86±0.00, respectively; P<0.05). Finally, plasma progesterone, ovulation rate, fertility and embryo development at 3.5 days after the artificial insemination were similar between diets (P>0.05), but embryo apoptosis rate was higher in control group than in enriched one (31.1±4.56% v. 17.1±3.87%, respectively; P<0.05). In conclusion, dietary PUFA enrichment from the rearing and throughout two productive cycles improved plasma progesterone during pregnancy, fertility, milk fatty acid profile and neonates development of primiparous supporting the beneficial effect of n-3 PUFA supplementation in rabbit does.

Annual cycle and spatial trends in fatty acid composition of suspended particulate organic matter across the Beaufort Sea shelf

Fatty acid profiles of suspended particulate organic matter (POM) were determined over an annual cycle (September 2003 to August 2004) on the Beaufort Sea shelf, Canadian Arctic. Special emphasis was placed on the nutritional quality of the fatty acid pool available to zooplankton by examining spatial and temporal patterns in the proportions of total polyunsaturated fatty acids (PUFA) and the essential fatty acids 22:6n−3 (DHA) and 20:5n−3 (EPA). EPA and DHA were the two most abundant PUFA throughout the study period. A log-ratio multivariate (LRA) analysis revealed strong structure in fatty acid profiles related to season and depth. Dominant fatty acids accounting for the observed trend included 18:5n−3, 18:4n−3, 16:1n−7, 20:5n-3, 18:0 and 20:3n−3. We observed a shift in fatty acid profiles from summer to autumn (e.g., from 16:1n−7 and EPA to 18:5n−3 and 18:4n−3) that likely corresponded to a shift in the relative importance of diatoms versus dinoflagellates, prymnesiophytes and/or prasinophytes to the POM pool. Fatty acid composition during winter was dominated by more refractory saturated fatty acids. A surprising finding was the depth and seasonal trend of 20:3n-3, which was higher in winter, aligned with 18:0 in the LRA, but behaved differently than other n−3 PUFA. We interpret fatty acid profiles during summer to be predominantly driven by phytoplankton inputs, whereas fatty acid profiles in winter were dominated by fatty acids that were left over after consumption and/or were generated by heterotrophs. The highest diatom inputs (EPA, the diatom fatty acid marker), n−3/n−6 ratios, and C16 PUFA index were located in an upwelling region off Cape Bathurst. This study is the first annual time series of fatty acid profiles of POM in Arctic seas, expanding our knowledge of the composition of POM throughout the dark season.

The Metabolic Syndrome and the Membrane Content of Polyunsaturated Fatty Acids in Hypertensive Patients.

Polyunsaturated fatty acids (PUFA) have been reported to be beneficial on some components of the metabolic syndrome (MetS). We tested the hypothesis that in hypertensive patients, presence of MetS and its related components is associated with cell membrane content of PUFA, a measure that reflects the dietary intake of these fatty acids. In 55 consecutive patients with primary hypertension referred to our university center, we measured anthropometric variables, 24-hour ambulatory blood pressure, general biochemistries including plasma lipids, and the fatty acid composition of red blood cell (RBC) membrane by gas chromatography. The prevalence of the MetS was 36.4% and in hypertensive patients with MetS, the RBC membrane content of total PUFA, PUFA of the n-6 family (n-6 PUFA), PUFA of the n-3 family (n-3 PUFA), polyunsaturated to saturated fatty acid ratio (PUFA/SFA), and omega-3 index was significantly lower than in patients without MetS. RBC membrane total PUFA, n-6 PUFA, n-3 PUFA, PUFA/SFA ratio, and omega-3 index were significantly and directly correlated with high-desity lipoprotein (HDL) cholesterol levels, a correlation that did not differ across tertiles of plasma apolipoprotein-A1. In multivariate linear regression analysis, HDL-cholesterol resulted to be directly and independently related to RBC membrane n-6 PUFA, PUFA/SFA ratio, and omega-3 index. Conversely, total cholesterol to HDL-cholesterol ratio had inverse and independent relationship with n-6 PUFA, PUFA/SFA ratio, and omega-3 index. In patients with hypertension the MetS is associated with lower cell membrane content of PUFA that is explained by a direct and independent relationship of membrane PUFA with HDL-cholesterol. This observation suggests reduced dietary intake of PUFA in these patients that might contribute to their cardiovascular risk.

Associations of n-6 and n-3 polyunsaturated fatty acids and tocopherols with proxies of membrane stability and subcutaneous fat sites in male elite swimmers

We hypothesize that membrane stability of elite swimmers adapted to chronic intense training is dependent on polyunsaturated fatty acids (PUFAs) and tocopherols in blood pools and that the composition of PUFA in plasma nonesterified fatty acids (NEFAs) might be associated with specific subcutaneous fat sites. Our aims were to investigate in male elite swimmers the associations of n-6 and n-3 PUFA and alpha- and gamma-tocopherols with proxies of membrane stability (phase angle and erythrocyte osmotic fragility) and of PUFA in plasma NEFA with specific skinfolds. Brazilian male elite swimmers (n = 20) under regular training for an average of 4.1 h/d and 6.1 d/wk took part in the study. Blood samples were obtained once after 18-hour rest and an overnight fast. Fatty acids were determined in plasma NEFA and erythrocytes by gas chromatolography and tocopherols were determined in plasma and erythrocytes by high-performance liquid chromatography. The status of PUFA was assessed as mean melting point, PUFA index [(Sigman-6 + Sigman-3) / (Sigman-7 + Sigman-9)] and docosahexaenoic acid indices (22:5n-6/22:4n-6 and 22:6n-3/22:5n-6 ratios) calculated from erythrocyte fatty acids. Phase angle was associated with an index of docosahexaenoic acid inadequacy (22:5n-6/22:4n-6; r = -0.53, P = .019) and with 22:5n-3 in erythrocytes (r = 0.51, P = .024), and erythrocyte osmotic fragility was associated with plasma alpha-tocopherol (r = -0.51, P = .05), which is a biomarker of vitamin E status. Plasma NEFAs 18:3n-3 and 20:4n-6 were positively associated with skinfolds of the trunk and arms (r = 0.49-0.59, P = .011-.043). The data presented indicate that n-3 PUFA and vitamin E states possibly improve membrane stability in elite swimmers and that the extent of specific anatomic sites of subcutaneous adipose tissue in the upper body might contribute to the composition of NEFA in the resting state.

N‐3 Polyunsaturated Fatty Acids and Atopy in Korean Preschoolers

Atopy is a growing problem for Korean children. Since eicosapentaenoic acid is a precursor of less active inflammatory eicosanoids, n-3 polyunsaturated fatty acids (PUFA) may have a protective effect on atopy. This study was undertaken to determine whether n-3 PUFA in red blood cells (RBC) is lower in atopic than in non-atopic preschoolers. Three hundred and eight Korean children aged 4-6 years were enrolled. Total RBC fatty acid composition was measured by gas chromatography. The prevalence of atopic dermatitis, allergic rhinitis, or asthma was 29%. Total RBC n-3 PUFA were lower in preschoolers with atopy than controls (9.8 +/- 1.2 vs. 11.4 +/- 1.6%; P<0.05), while n-6 PUFA (33.0 +/- 1.4 vs. 32.2 +/- 1.0%; P<0.05) and n-6/n-3 PUFA ratio (3.4 +/- 0.6 vs. 2.8 +/- 0.5; P<0.05) were greater. The following factors were also associated with an increase in atopy: higher saturated fatty acids (39.6 +/- 1.4 vs. 40.6 +/- 1.9; P<0.05) and arachidonic acid (15.3 +/- 1.6 vs. 16.0 +/- 2.9; P<0.05), and lower total PUFA (43.8 +/- 0.7 vs. 42.8 +/- 1.4; P<0.05) and omega-3 index (EPA + DHA; 9.1 +/- 0.8 vs. 7.8 +/- 0.5; P<0.05) in RBC. Maternal history of atopy was a significant (P<0.05) risk factor, while lactation was not. The results suggest that a reduced content of n-3 PUFA in the RBC membrane could play a role in early children atopy.
