Effects of aflatoxin B1 on subacute exposure of hybrid groupers (Epinephelus fuscoguttatus♀ × Epinephelus lanceolatus♂): Growth, liver histology, and integrated liver transcriptome and metabolome analysis

With the increasing incorporation of plant-based ingredients into the grouper diet, the issue of aflatoxin B1 (AFB1) contamination in the diet has become a significant concern. In this study, the negative effects of AFB1 on the growth and liver health of hybrid grouper (Epinephelus fuscoguttatus♀ × Epinephelus lanceolatus♂) were investigated in the context of growth, liver histology, serum biochemical indices, and integrated transcriptomic and metabolomic data. A total of 540 healthy hybrid groupers, initially weighing 11.59 ± 0.03 g, were randomly divided into six groups (three replicates of 30 fish each): the Control group was fed a basal diet, and the experimental groups were supplemented with 7 (AF7), 30 (AF30), 111 (AF111), 445 (AF445) and 2230 μg/kg AFB1 (AF2230) in the basal diet respectively, for 56 days. Groups Control, AF445, and AF2230 were selected for subsequent histological, muscle fatty acid, and transcriptomic and metabolomic analyses based on the results of hybrid grouper growth and serum biochemical indices. Compared to the Control group, both whole-body crude lipid and muscle crude lipid contents showed significant decreases in the AF2230 group (P < 0.05), while only muscle crude lipid content showed a significant decrease in the AF445 group (P = 0.001). Liver damage was seen in the histology of the liver of AF445 and AF2230 groups. Muscle fatty acid results showed that the addition of 445 and 2230 μg/kg AFB1 to the ration increased saturated fatty acids and monounsaturated fatty acids and decreased polyunsaturated fatty acids and highly unsaturated fatty acids in muscle (P < 0.05). Transcriptome analyses revealed multiple metabolic pathways associated with AFB1 metabolism, and metabolomics analyses further confirmed changes in the activity of these pathways. The results of the combined transcriptomic and metabolomic analyses indicated that AFB1 causes liver injury mainly by affecting liver retinol metabolism, metabolism of xenobiotics by cytochromes P450, drug metabolism-cytochromes P450 and biosynthesis of unsaturated fatty acids. In conclusion, dietary AFB1 levels above 445 μg/kg resulted in growth inhibition, liver injury, liver AFB1 accumulation, and reduced muscle polyunsaturated fatty acid content in grouper, thereby affecting muscle quality. This study provides novel insights into the detrimental effects of AFB1 on aquatic species and contributes to the scientific basis for the health and sustainability of aquaculture practices.

The role of vitamin E in polyunsaturated fatty acid synthesis and alleviating endoplasmic reticulum stress in sub-adult grass carp (Ctenopharyngodon idella)

Vitamin E (VE) is an essential lipid-soluble vitamin that improves the fish flesh quality. However, the underlying molecular mechanisms remain unclear. This study aimed to investigate the effects of VE on growth performance and flesh quality in sub-adult grass carp (Ctenopharyngodon idella). A total of 450 fish (713.53 ± 1.50 g) were randomly divided into six treatment groups (three replicates per treatment) and fed for nine weeks with different experimental diets (dietary lipid 47.8 g/kg) that contained different levels of VE (5.44, 52.07, 96.85, 141.71, 185.66, and 230.12 mg/kg diet, supplemented as dl-α-tocopherol acetate). Notably, the treatment groups that were fed with dietary VE ranging from 52.07 to 230.12 mg/kg diet showed improvement in the percent weight gain, special growth rate, and feed efficiency of grass carp. Moreover, the treatment groups supplemented with dietary VE level of 141.71, 185.66, and 230.12 mg/kg diet showed enhancement in crude protein, lipid, and α-tocopherol contents in the muscle, and the dietary levels of VE ranging from 52.07 to 141.71 mg/kg diet improved muscle pH24h and shear force but reduced muscle cooking loss in grass carp. Furthermore, appropriate levels of VE (52.07 to 96.85 mg/kg diet) increased the muscle polyunsaturated fatty acid content in grass carp. Dietary VE also increased the mRNA levels of fatty acid synthesis-related genes, including fas, scd-1, fad, elovl, srebp1, pparγ, and lxrα, and up-regulated the expression of SREBP-1 protein. However, dietary VE decreased the expression of fatty acid decomposition-related genes, including hsl, cpt1, acox1, and pparα, and endoplasmic reticulum stress-related genes, including perk, ire1, atf6, eif2α, atf4, xbp1, chop, and grp78, and down-regulated the expression of p-PERK, p-IRE1, ATF6, and GRP78 proteins. In conclusion, dietary VE increased muscle fatty acid synthesis, which may be partly associated with the alleviation of endoplasmic reticulum stress, and ultimately improves fish flesh quality. Moreover, the VE requirements for sub-adult grass carp (713.53 to 1590.40 g) were estimated to be 124.9 and 122.73 mg/kg diet based on percentage weight gain and muscle shear force, respectively.

Utilisation of mealworm (Tenebrio molitor) oil in Pacific white shrimp (Litopenaeus vannamei) diet

Abstract Mealworm (Tenebrio molitor) and their derivatives are sustainable ingredients in aquaculture feeds. This study evaluated the effect of fish oil (FO) replacement with mealworm oil (MO) on growth performance, non-specific immune responses and disease resistance against Vibrio parahaemolyticus for Pacific white shrimp (Litopenaeus vannamei). FO in a control diet (Con) was replaced with MO at levels of 25%, 50%, 75% and 100% (designated as MO25, MO50, MO75 and MO100, respectively). A total of 480 shrimp (initial mean body weight, 0.884 ± 0.004 g) were randomly distributed into 20 acrylic tanks (240 L) in quadruplicates groups and fed the experimental diets for 58 days. Growth performance was significantly higher in all MO groups than in Con group. Shrimp fed MO50 and MO75 diets showed significantly improved feed utilisation efficiency than shrimp fed Con diet. Lipid metabolism related-gene expressions of fatty acid binding protein and triacylglycerol lipase in hepatopancreas were significantly downregulated by increment of MO in the diets. Digestibility of protein, lipid, energy and dry matter were not significantly affected by the dietary treatments, while digestibility of poly-unsaturated fatty acids (PUFAs) were significantly decreased by dietary MO supplementation. The levels of n-3 PUFA in muscle and hepatopancreas were significantly decreased with increasing MO levels in the diets. Cumulative survival of shrimp in a challenge test against V. parahaemolyticus was higher in all MO groups and significantly higher in MO25 group than in Con group. Non-specific immune responses were significantly enhanced in all MO groups than in Con group. Replacement of FO with MO in the diets significantly increased haemolymph cholesterol and triglyceride levels. Our findings suggest that MO could be utilised as a promising substitute for FO in the diets of L. vannamei and optimal FO replacement level would be 50% for growth and 25% for disease resistance against V. parahaemolyticus.

Effects of low protein diet with a balanced amino acid pattern on growth performance, meat quality and cecal microflora of finishing pigs.

The present study aimed to investigate the effects of low protein diets balanced with four amino acids on growth performance, meat quality and cecal microflora of finishing pigs. Fifty-four healthy hybrid barrows (Duroc × Landrace × Yorkshire) with an average body weight of 70.12 ± 4.03 kg were randomly assigned to one of the three dietary treatments with six replicate pens per treatment (three barrows per pen). The three dietary treatments included a normal protein diet (NP), a low protein diet (LP) and a very low protein diet (VLP). The average daily gain, average daily feed intake and feed conversion ratio of pigs were not significantly changed with the LP and VLP diets compared to the NP diet (P > 0.05). The water holding capacity and shear force of longissimus dorsi muscle were decreased, whereas the intramuscular fat content of the longissimus dorsi muscle was increased (P < 0.05) in pigs fed with the LP and VLP diets compared to the NP diet. The contents of saturated fatty acids in muscle were decreased (P < 0.05), whereas the content of polyunsaturated fatty acids in muscle was increased (P < 0.01) with the VLP diet compared to the NP diet. The contents of histamine, spermidine, spermine and tyramine of muscle were decreased with the VLP diet compared to the NP diet (P < 0.05). The relative abundance of Turicibacter, Terrisporobacter, Clostridium_sensu_stricto_1 and UCG-005 was higher (P < 0.05), whereas the relative abundance of Lactobacillus and Streptococcus was lower (P < 0.05) in pigs fed with the LP and VLP diets compared to the NP diet. Based on the correlation of cecal microbiota and cecal biogenic amine, the contents of tyramine, spermidine and histamine were negatively correlated with the abundance of Terrisporobacter (P < 0.01) and the content of histamine was positively correlated with the abundance of Lactobacillus (P < 0.01). Balanced with four essential amino acids, the VLP diet with crude protein levels decreased by > 4% increased the intramuscular fat content, changed the fatty acid and amino acid composition of longissimus dorsi muscle and the profile of cecum microbiota, and reduced the content of cecum bioamine, with no negative effect on the growth performance of pigs. © 2022 Society of Chemical Industry.

Effects of Different Roughage Diets on Fattening Performance, Meat Quality, Fatty Acid Composition, and Rumen Microbe in Steers.

This study aimed to evaluate different roughages on fatting performance, muscle fatty acids, rumen fermentation and rumen microbes of steers. Seventy-five Simmental crossbred steers were randomly divided into wheat straw group (WG), peanut vine group (PG) and alfalfa hay group (AG), with 5 replicates of 5 steers each. The results showed a highest average daily gain and lowest feed/gain ratio in AG group (P = 0.001). Steers fed alfalfa hay had the highest muscle marbling score and n-3 polyunsaturated fatty acid (PUFA), and also the rumen NH3-N and microbial protein (MCP) concentration among the three groups (P < 0.05). Correlation analysis showed that ruminal NH3-N and MCP were negatively correlated with muscle saturated fatty acid (SFA), while ruminal MCP was positively correlated with muscle PUFA and n-3 PUFA (P < 0.05). 16S rRNA analysis indicated that fed alfalfa hay decreased the abundance of Ruminococcaceae_UCG-001(P = 0.005). More importantly, muscle SFA deposition were positively correlated to the abundance of Ruminococcaceae_UCG-001 (P < 0.05), while the muscle PUFA and n-3 PUFA deposition were negatively correlated to it (P < 0.01). Therefore, alfalfa hay provides a better fattening effect on steers. Alfalfa rich in n-3 PUFA would reduce the abundance of Ruminococcaceae_UCG-001 involved in hydrogenation, increase the rumen protective effect of C18:3 n−3, which is beneficial to the deposition of muscle n-3 PUFA and PUFA.

A parallel survey on the fatty acid composition in backfat and longissimus lumborum and comparison of their associations with growth and carcass traits in pigs

Compositions of fatty acids are critical parameters of meat nutrition and palatability. This study investigated phenotypes of 21 fatty acid composition traits in both backfat (BF) and longissimus lumborum (LL) tissues in 583 F7 mosaic pigs from an eight-breed crossed heterogeneous population, and their associations with 25 detailed measured growth and carcass traits in the same population. We showed that the LL has significantly higher content of saturated and mono-unsaturated fatty acids, and lower poly-unsaturated fatty acids than BF. Most of fatty acid composition traits in both LL and BF show significant correlations with growth and carcass traits. Notably, the correlations with the growth and carcass traits for fatty acid composition traits in BF are much stronger than that in LL, suggesting closer relationship of the growth and carcass traits with metabolism of the fatty acids in fat than in muscle. The polyunsaturated fatty acid traits including C18:2n6, PUFA, C20:2n6, C20:3n6 were among the fatty acid composition traits showing the strongest correlation with growth and carcass traits in both BF and LL, suggesting an interesting link between the metabolism of polyunsaturated fatty acids in muscle and fat tissues with the growth and carcass phenotypes. The results provide helpful insights that are helpful to the breeding program aiming to simultaneously improve the fatty acid composition, growth and carcass traits in pigs.

Interaction Between the Intestinal Microbial Community and Transcriptome Profile in Common Carp (Cyprinus carpio L.).

In a previous study, we found that the growth performance of the new strain of Huanghe carp is related to gene expression and bacterial community in the gut. In order to better understand the relationship between the gene expression level and bacterial abundance in the gut, we studied the growth performance, gut bacterial structure, and transcriptome profile in the 4th generation of the new carp strain (selection group) at harvesting time, and compared them with the control line (traditional Huanghe carp). Body weight, depth, width, and length increased 14.58, 7.14, 5.04, and 5.07%, respectively. The gut microbiome of the selection group also exhibited significantly higher species diversity parameters (Shannon, Simpson, and chao1). Both PCA and phylogenetic analyses divided all gut samples into two parts: control and selection group. Aeromonas was the dominant taxon in the control group, followed by Firmicutes and Roseomonas; in the selection group, Roseomonas was the dominant taxon, followed by Firmicutes and then Aeromonas. Among the 249 significantly differentially expressed genes, 194 were downregulated and 55 were upregulated. Functional GO annotation produced 13 terms in the biological process, 8 in the cellular component, and 7 in the molecular function categories. KEGG annotation indicated that most of these genes were associated with the immune-related pathways. A total of 2,892 pairs of genes (245) and baceterial genera (256) were analyzed using Pearson’s correlation analysis. Most of the identified associations were mapped to the immune system, bacterial community, and cell differentiation categories. The top-10 bacterial genera identified by these analyses were Methylocystis, Ohtaekwangia, Roseomonas, Shewanella, Lutispora, GpVI, Desulfovibrio, Candidatus_Berkiella, Bordetella, and Azorhizobium. Genes paired with bacteria flora were divided into four functional categories: immune, growth, adipocyte differentiation, and nerve regulation. These genes may be related to the comparatively fast growth and high muscle polyunsaturated fatty acid content of the Huanghe carp new strain. Meanwhile, nerve regulation-related genes may be a reflection of the microbiota-gut-brain axis. These results illustrate that gut bacterial community structure is associated with the growth performance and gene expression in the Huanghe carp new strain.

Comparative evaluation of nutritional value and flavor quality of muscle in triploid and diploid common carp: Application of genetic improvement in fish quality

Hybridization can combine the advantages of different strains to produce new varieties, which may also improve the quality of fish. Triploid common carp (3n = 150) were derived from the female diploid common carp (Cyprinus carpio L.) (2n = 100) crossing with the male allotetraploid (4n = 200). Nutritional value and flavor quality were investigated in triploid and diploid common carps fed with the same diet for 13 weeks. Results indicated that the triploid common carp exhibited significantly higher levels of crude protein, essential and total amino acids, eicosapentaenoic acid (EPA, 20:5 n-3), docosahexaenoic acid (DHA, 22:6 n-3), total polyunsaturated fatty acids (PUFAs) and the ratio of n-3 to n-6 PUFAs in muscle than the diploid common carps (P < 0.05). Additionally, the concentration of sour and bitter amino acid in the muscle of triploid common carp significantly decreased than that in diploid (P < 0.05). Triploid common carp had significantly higher concentration of salt amino acid (P < 0.05) and kept unchanged the content of sweet and umami amino acid than diploid (P > 0.05). Furthermore, the characteristic volatile compounds which including styrene, ethylbenzene, butylated hydroxytoluene, 1-octen-3-ol and (Z)-β-ocimene were screened in the muscle of diploid and triploid common carps, of which styrene and 1-octen-3-ol were the odor-active compounds. Triploid common carp had significantly higher concentration of styrene and 1-octen-3-ol than diploid (P < 0.05), of which 1-octen-3-ol was the highest odor-active compound in triploid. These findings demonstrated that the triploid common carp had improved nutritional and flavor quality, which provided a novel insight for the application of hybridization in fish quality improvement.

Polyunsaturated fatty acid, volatile and sensory profiles of beef from steers fed citrus pulp or grape pomace

The present study compared the effects of feeding dried grape pomace (DGP) or citrus pulp (DCP) at 150 g/kg dry matter compared to a control diet on major polyunsaturated fatty acids (PUFA), volatile and sensory profiles of beef. Feeding DGP or DCP diets to Angus steers for 90 d increased the proportions of C18:2n-6, C20:4n-6, C18:3n-3, total conjugated linoleic acid (CLA), n-3 and n-6 PUFA in muscle. Control-fed beef had greater concentrations of C18:1n-9, total aldehydes, ketones, and alcohols compared to DCP and DGP. Feeding DGP and DCP diets produced less tender beef than control. Overall, finishing steers on diets containing DGP or DCP compared to control increased proportions of total CLA, n-3 and n-6 PUFA, and reduced concentrations of aldehydes, ketones, and alcohols, but did not affect beef sensory attributes except for a slight reduction in tenderness.

The effects of high‐macronutrient (protein, fat and carbohydrate) diets on growth performance and muscular metabolic responses in grass carp

Four purified diets were fed to quadruplicate groups of grass carp (Ctenopharyngodon idella) (initial body weight 55 g; 20 fish per replicate) for 11 weeks. The gross energy of three high-macronutrient diets was 11% higher than that of the control diet. The high-protein diet led to the highest weight gain (WG) and specific growth rate (SGR), while the lowest protein efficiency ratio (PER) and protein retention efficiency (PRE) (p < .05). No significant effect on the above four indexes was observed in fish fed the high-fat diet (p > .05). Fish fed high-carbohydrate diet had the lowest WG and SGR (p < .05), while no significant difference was found in PER and PRE (p > .05). The contents of crude fat, total polyunsaturated fatty acids in muscle in high-fat and high-carbohydrate groups, but not in high-protein group, were significantly higher than those of control group (p < .05). The contents of muscular protein and most amino acids did not show any clear change in response to high-macronutrient diet (p > .05). Gene expression data in muscle indicated that g6pd, fas and acc1 in muscle were up-regulated in high-fat and high-carbohydrate groups, and fbp and pck were significantly up-regulated in high-macronutrient groups (p < .05). In conclusion, the high-fat and high-carbohydrate diets increased muscular fat deposition by up-regulation the key enzymes expressions of pentose phosphate pathway and fat synthesis, and high-macronutrient diets increased muscular glycogen content by up-regulation gluconeogenesis enzymes expressions.

Seasonal variations of the fatty acid profiles in the edible portions of two freshwater fish species (Pseudophoxinus fahrettini and Capoeta mauricii)

In this study, seasonal and sexual variations of fatty acid profiles of Pseudophoxinus fahrettini and Capoeta mauricii which are freshwater fishes in Turkey inland waters were investigated. The monounsaturated fatty acids (MUFAs) values were higher than saturated fatty acids (SFAs) and polyunsaturated fatty acids (PUFAs) values for both species. Also the ratios of MUFAs in P. fahrettini were higher than those of C. mauricii in all seasons. The results indicated that palmitic acid was the most dominant SFA for both species in all seasons. Oleic acid in the muscle of P. fahrettini and palmitoleic acid in the muscle of C. mauricii were the highest MUFA respectively in both genders in all seasons. The highest ratios of ω3 PUFAs were 23,00% in the muscle of C. mauricii in summer and 18,44% in P. fahrettini in winter. The ratios of ω3 PUFAs in muscles of both species and genders were higher than the ratios of ω6 PUFAs during all the seasons. Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) were significantly higher than the other PUFAs in both sexes of the species. The ratio of ω3 PUFAs to ω6 PUFAs was higher than 1,76 in both species and genders in all seasons.

Dietary N-Carbamylglutamate Supplementation in a Reduced Protein Diet Affects Carcass Traits and the Profile of Muscle Amino Acids and Fatty Acids in Finishing Pigs.

The aim of this study was to investigate whether dietary N-carbamylglutamate (NCG) supplementation in a reduced protein diet affected carcass traits and meat quality in finishing pigs. A total of 120 gilts were randomly assigned to one of four treatments for 40 days, including a standard protein diet (SP), a reduced protein diet supplemented with 1.7% l-alanine (RP + Ala), a reduced protein diet supplemented with 1.0% l-arginine (RP + Arg), and a reduced protein diet supplemented with 0.1% NCG and 1.7% l-alanine (RP + NCG). NCG supplementation increased the endogenous synthesis of l-arginine. The RP + NCG diet significantly increased the loin eye area (p < 0.05) and tended to decrease the 10th rib fat depth (p = 0.08). NCG supplementation in a reduced protein diet was effective to produce functional pork with a high content of leucine (p < 0.05). The composition of several ω-6 and ω-3 polyunsaturated fatty acids (PUFAs) but not the ratio of ω-6/ω-3 PUFAs in muscles was altered in finishing pigs with dietary NCG supplementation. In conclusion, the RP + NCG diet is effective to increase the longissimus dorsi muscle area, decrease back fat accretion, and produce functional pork with a high content of leucine but without a negative impact on the muscle fatty acid profile in finishing pigs.

RNA-seq based detection of differentially expressed genes in the skeletal muscle of Duroc pigs with distinct lipid profiles

We have used a RNA-seq approach to investigate differential expression in the skeletal muscle of swine (N = 52) with divergent lipid profiles i.e. HIGH (increased intramuscular fat and muscle saturated and monounsaturated fatty acid contents, higher serum lipid concentrations and fatness) and LOW pigs (leaner and with an increased muscle polyunsaturated fatty acid content). The number of mRNAs and non-coding RNAs (ncRNAs) expressed in the porcine gluteus medius muscle were 18,104 and 1,558, respectively. At the nominal level of significance (P-value ≤ 0.05), we detected 1,430 mRNA and 12 non-coding RNA (ncRNA) transcripts as differentially expressed (DE) in the gluteus medius muscle of HIGH vs LOW pigs. This smaller contribution of ncRNAs to differential expression may have biological and technical reasons. We performed a second analysis, that was more stringent (P-value ≤ 0.01 and fold-change ≥ 1.5), and only 96 and 0 mRNA-and ncRNA-encoding genes happened to be DE, respectively. The subset of DE mRNA genes was enriched in pathways related with lipid (lipogenesis and triacylglycerol degradation) and glucose metabolism. Moreover, HIGH pigs showed a more lipogenic profile than their LOW counterparts.

Fatty acids, lipid and protein oxidation, metmyoglobin reducing activity and sensory attributes of &lt;i&gt;biceps femoris&lt;/i&gt; muscle in goats fed a canola and palm oil blend

The link between the consumption of saturated fatty acids of ruminant meat and the incidence of chronic diseases in humans substantiates efforts to modify its lipid profile. However, altering the lipid profile of ruminant meat could affect its quality attributes and shelf life. This study investigated the effect of a blend of 80% canola oil and 20% palm oil (BCPO) and post mortem ageing on lipid composition, antioxidant status, colour, metmyoglobin reducing activity (MRA) and lipid and protein oxidation in biceps femoris muscle in goats. Twenty-four bucks were randomly assigned to diets containing on a dry matter basis, 0%, 4% or 8% BCPO, fed for 100 d, and slaughtered. The biceps femoris was subjected to 7 d post mortem ageing (4 oC). Neither diet nor post mortem ageing altered catalase, superoxide dismutase and glutathione peroxidase activities or sensory attributes of the biceps femoris . Dietary BCPO improved α- and γ-tocopherol, but did not affect total carotenoids and δ-tocopherol, colour, metmyoglobin content, MRA, TBARS, carbonyl content and free thiol content. Goats fed 4% BCPO had higher cis-9 trans -11 CLA, while those fed 8% BCPO had higher C18:3n-3 and C20:5n-3 compared with other treatments. The 4% and 8% BCPO meat had lower C14:0, but higher C18:1n-9 and C22:5n-3 than the control goats. Post mortem ageing decreased muscle polyunsaturated fatty acids (PUFA), antioxidant compounds, redness, MRA and oxidative stability of lipid and proteins, but increased metmyoglobin content. Dietary BCPO enhanced beneficial muscle lipids without compromising the sensory quality, and lipid and protein stability of the goat meat. Lipid composition in muscle and liver of sympatric coregonid fishes from Lake Baikal (Coregonus spp.) under common garden experiment

The content of total lipids, total phospholipids, and fatty acids of total lipids in muscles and liver of juvenile Lake Baikal sympatric coregonid fishes have been analyzed for the first time under a common garden experiment. Baikal omul, Coregonus migratorius Georgi, is an active migrant of the pelagic zones of the lake. Baikal (lacustrine) whitefish, C. baicalensis Dybowski, is a colonizer of the bottom habitats, which are relevant to the pelagic zones of the littoral and underwater slope. Structural lipids (phospholipids and cholesterol) dominated in total lipids of the tissues of all fish under study. Spare lipids significantly prevailed in omul muscles when compared to whitefish. The highest variability of fatty acid composition was reported in muscles of coregonid fishes. Statistically significant differences were revealed in the content of the ω3 polyunsaturated fatty acids in muscles of lacustrine whitefish and omul. Associations of lipid compositions revealed in tissues of the whitefishes under the study with their respective ecotypes have been discussed.

Fatty acids and oxidative stability of meat from lambs fed carob-containing diets

Male Comisana lambs were individually stalled and, for 56days, were fed concentrates with 60% barley (n=8 lambs), or concentrates in which barley was partially replaced by 24% or 35% carob pulp (n=9 lambs in each group). The intramuscular fatty acids were analyzed and the color stability, lipid and protein oxidation were measured in fresh meat overwrapped with polyvinyl chloride film at 0, 3 or 6days of storage at 4°C in the dark. Carob pulp increased the concentration of polyunsaturated fatty acids (PUFA) in muscle, including the rumenic acid (P<0.01), and reduced the saturated fatty acids (P<0.01) and the n-6/n-3 PUFA ratio (P=0.01). The meat did not undergo extensive oxidative deterioration and the diet did not affect the oxidative stability parameters. Therefore, carob in lamb diet could increase PUFA in muscle without compromising meat oxidative stability.

The proximate composition of male and femaleLeuciscus lepidusin Beyşehir Lake was investigated. The fatty acid profiles of total lipid, phospholipid, and triacylglycerol in muscle and liver of male and femaleL. lepiduswere evaluated by gas chromatography. Proximate analyses showed that meat of male and femaleL. lepidushad 15.13 ± 0.04 and 18.75 ± 0.11% fat, 20.42 ± 0.45 and 22.21 ± 0.56% protein, 65.47 ± 1.37 and 61.28 ± 1.03% moisture, and 1.51 ± 0.05 and 1.50 ± 0.03% ash, respectively. The percentage of total saturated fatty acids was higher in liver than in muscle, whereas the total polyunsaturated fatty acid (PUFA) content was the lowest in all fatty acid profiles. The phospholipids contained more PUFAs than triacylglycerol. Analysis of variance indicated significant differences (P&lt;0.05) between male (47.51%) and female (49.98%) muscle PUFAs in total lipid. The proportion of omega 3 (ω3) to omega 6 (ω6) fatty acids of total lipid was 3.15 in male and 3.68 in female. The ratio is an important indicator for comparing the value of fish oil. Therefore, it was concluded thatL. lepiduswas considered to be a high quality product for healthy food choice. Additionally, femaleL. lepidusmay especially be used to produce fish oil supplements from freshwater fish combined with vegetable oils.

Influence of Different Sources of Dietary Fats on Fatty Acid Profile of Striped Snakehead (Channa Striatus) Fish Carcass

Various dietary fats were used to assess their impact on the fatty acid profile of snakehead murrel, saul (Channa striatus). The saul grow-out (av. wt. 27.36 ± 0.09 to 32.54 ± 0.41 g) were fed for 12-weeks with seven experimental diets (F1, F2, F3, F4, F5, F6 and a control, F7 of natural foodstuffs, NATFO). F1 (L3HUF) contains 0.5% n-3 fatty acid & 7.5 % saturated oil; F2 (H3HUF) contains 1.0% n-3 fatty acid & 7.0% saturated oil; F3 (MUSOL) contains 8.0% mustard oil; F4 (LINOL) contains 8.0% linseed oil; F5 (MIXOL) contains 4.0% mustard oil and 4.0 % linseed oil; F6 (SATOL) contains 8.0 % saturated oil. The muscle polyunsaturated fatty acids (PUFA) contents varied with different dietary lipids level which impacted on deposition of fatty acid in flesh. The muscle unsaturated fatty acids, including Docosahexaenoic acid (22:6n3, DHA) and Eicosapentanoic Acid {C20:5n-3, EPA), levels, were comparatively higher in MIXOL diet followed by H3HUF than in the other diets, indicating selective deposition of various fatty acids in each feeding trial. It was concluded that dietary fats in the diet has role in the carcass composition of fatty acid profile in Channa striatus and the MIXOL (contains mustard oil and linseed oil: 1:1 w/w) could be safely used for better deposition of beneficial healthy fatty acids like EPA and DHA. The addition of mustard oil and linseed oil in the diet are comparative better if we look towards economizing the cost of the brood-stock feed in comparison to the addition of pure fatty acids in the fish diets.

Fish Oil Increases Muscle Protein Mass and Modulates Akt/FOXO, TLR4, and NOD Signaling in Weanling Piglets After Lipopolysaccharide Challenge1–3

Proinflammatory cytokines play a key role in the pathophysiology of muscle atrophy. In addition, n3 polyunsaturated fatty acids (PUFAs) exert an inhibitory effect on proinflammatory cytokines affecting many inflammatory diseases. We hypothesized that dietary supplementation of fish oil could attenuate lipopolysaccharide (LPS)-induced muscle atrophy. Weanling pigs were used in a 2 × 2 factorial design and the main factors included diet (5% corn oil or 5% fish oil) and immunological challenge (LPS or saline). After 21 d of treatment with either fish oil or corn oil, pigs received an i.p. injection of either saline or LPS. At 4 h postinjection, blood and muscle samples were obtained. Fish oil led to enrichment of eicosapentaenoic acid, docosahexaenoic acid, and total n3 PUFAs in muscles. Fish oil increased muscle protein mass, indicated by a higher protein:DNA ratio in gastrocnemius and longissimus dorsi (LD) muscles. In addition, fish oil increased Akt1 mRNA abundance and decreased Forkhead Box O (FOXO) 1 and FOXO4 mRNA abundance. Fish oil also increased phosphorylation of Akt and FOXO1 in gastrocnemius and LD muscles. Fish oil decreased the mRNA abundance of muscle atrophy F-box (MAFbx) and muscle RING finger 1 in gastrocnemius and LD muscles. Moreover, fish oil reduced the plasma tumor necrosis factor (TNF) α, muscle TNFα, and prostaglandin E2 concentrations, and muscle TNFα and cyclooxygenase 2 (COX2) mRNA abundance. Finally, fish oil downregulated the mRNA abundance of muscle toll-like receptor (TLR4) and its downstream signaling molecules [myeloid differentiation factor 88 (MyD88), TNFα receptor-associated factor 6 (TRAF6), and NF-κB p65], and nucleotide-binding oligomerization domain protein (NOD1), NOD2, and their adaptor molecule [receptor-interacting serine/threonine-protein kinase 2 (RIPK2)]. These results indicate fish oil may suppress muscle proinflammatory cytokine production via regulation of TLR and NOD signaling pathways and therefore improve muscle protein mass, possibly through maintenance of Akt/FOXO signaling.
